Abstract

We study the transverse mode instability (TMI) in the limit where a single higher-order mode (HOM) is present. We demonstrate that when the beat length between the fundamental mode and the HOM is small compared to the length scales on which the pump amplitude and the optical mode amplitudes vary, TMI is a three-wave mixing process in which the two optical modes beat with the phase-matched component of the index of refraction that is induced by the thermal grating. This limit is the usual limit in applications, and in this limit TMI is identified as a stimulated thermal Rayleigh scattering (STRS) process. We demonstrate that a phase-matched model that is based on the three-wave mixing equations can have a large computational advantage over current coupled mode methods that must use longitudinal step sizes that are small compared to the beat length.

In this article we investigate the existence of (2,3)-cordial labelings of oriented hypercubes. In this investigation, we determine that there exists a (2,3)-cordial oriented hypercube for any dimension divisible by 3. Next, we provide examples of (2,3)-cordial oriented hypercubes of dimension not divisible by 3 and state a conjecture on existence for dimension 3k + 1. Abstract

Recently L. B. Beasley introduced $(2,3)$-cordial labelings of directed graphs in [1]. He made two conjectures which we resolve in this article. He conjectured that every orientation of a path of length at least five is $(2,3)$ cordial, and that every tree of max degree $n =3$ has a cordial orientation. We show these two conjectures to be false. We also discuss the $(2,3)$ cordiality of orientations of the Petersen graph, and establish an upper bound for the number of edges a graph can have and still be $(2,3)$ cordial. An application of $(2,3)$ cordial labelings is also presented.

Abstract

Pathogenic germline mutations in the RAD51 paralog genes RAD51C and RAD51D, are known to confer susceptibility to ovarian and triple-negative breast cancer. Here, we investigated whether germline loss-of-function variants affecting another RAD51 paralog gene, RAD51B, are also associated with breast and ovarian cancer. Among 3422 consecutively accrued breast and ovarian cancer patients consented to tumor/germline sequencing, the observed carrier frequency of loss-of-function germline RAD51B variants was significantly higher than control cases from the gnomAD population database (0.26% vs 0.09%), with an odds ratio of 2.69 (95% CI: 1.4-5.3). Furthermore, we demonstrate that tumors harboring biallelic RAD51B alteration are deficient in homologous recombination DNA repair deficiency (HRD), as evidenced by analysis of sequencing data and in vitro functional assays. Our findings suggest that RAD51B should be considered as an addition to clinical germline testing panels for breast and ovarian cancer susceptibility.

Abstract

ObjectivesClinical trials evaluating the safety and effectiveness of sedative medication use in critically ill adults undergoing mechanical ventilation differ considerably in their methodological approach. This heterogeneity impedes the ability to compare results across studies. The Sedation Consortium on Endpoints and Procedures for Treatment, Education, and Research Recommendations convened a meeting of multidisciplinary experts to develop recommendations for key methodologic elements of sedation trials in the ICU to help guide academic and industry clinical investigators.DesignA 2-day in-person meeting was held in Washington, DC, on March 28-29, 2019, followed by a three-round, online modified Delphi consensus process.ParticipantsThirty-six participants from academia, industry, and the Food and Drug Administration with expertise in relevant content areas, including two former ICU patients attended the in-person meeting, and the majority completed an online follow-up survey and participated in the modified Delphi process.Measurements and main resultsThe final recommendations were iteratively refined based on the survey results, participants' reactions to those results, summaries written by panel moderators, and a review of the meeting transcripts made from audio recordings. Fifteen recommendations were developed for study design and conduct, subject enrollment, outcomes, and measurement instruments. Consensus recommendations included obtaining input from ICU survivors and/or their families, ensuring adequate training for personnel using validated instruments for assessments of sedation, pain, and delirium in the ICU environment, and the need for methodological standardization.ConclusionsThese recommendations are intended to assist researchers in the design, conduct, selection of endpoints, and reporting of clinical trials involving sedative medications and/or sedation protocols for adult ICU patients who require mechanical ventilation. These recommendations should be viewed as a starting point to improve clinical trials and help reduce methodological heterogeneity in future clinical trials.

Abstract

Electrochemical plating of Li metal on the graphite electrode is the key limitation behind slow charging times of Li-ion batteries (LIBs) in electric vehicles (EVs). Currently, electrochemical methods to detect the onset of Li plating while a battery is fast charging are sparse. In this study, we use operando electrochemical impedance spectroscopy to reliably detect the onset of Li plating on graphite electrodes in three-electrode LIBs. An increase in the graphite solid-electrolyte interface (SEI) resistance indicates that Li plating has occurred. Abstract

PurposeCyclin-dependent kinase 4 (CDK4) and CDK6 inhibitors (CDK4/6i) are highly effective against estrogen receptor-positive (ER+)/HER2- breast cancer; however, intrinsic and acquired resistance is common. Elucidating the molecular features of sensitivity and resistance to CDK4/6i may lead to identification of predictive biomarkers and novel therapeutic targets, paving the way toward improving patient outcomes.Experimental designParental breast cancer cells and their endocrine-resistant derivatives (EndoR) were used. Derivatives with acquired resistance to palbociclib (PalboR) were generated from parental and estrogen deprivation-resistant MCF7 and T47D cells. Transcriptomic and proteomic analyses were performed in palbociclib-sensitive and PalboR lines. Gene expression data from CDK4/6i neoadjuvant trials and publicly available datasets were interrogated for correlations of gene signatures and patient outcomes.ResultsParental and EndoR breast cancer lines showed varying degrees of sensitivity to palbociclib. Transcriptomic analysis of these cell lines identified an association between high IFN signaling and reduced CDK4/6i sensitivity; thus an "IFN-related palbociclib-resistance Signature" (IRPS) was derived. In two neoadjuvant trials of CDK4/6i plus endocrine therapy, IRPS and other IFN-related signatures were highly enriched in patients with tumors exhibiting intrinsic resistance to CDK4/6i. PalboR derivatives displayed dramatic activation of IFN/STAT1 signaling compared with their short-term treated or untreated counterparts. In primary ER+/HER2- tumors, the IRPS score was significantly higher in lumB than lumA subtype and correlated with increased gene expression of immune checkpoints, endocrine resistance, and poor prognosis.ConclusionsAberrant IFN signaling is associated with intrinsic resistance to CDK4/6i. Experimentally, acquired resistance to palbociclib is associated with activation of the IFN pathway, warranting additional studies to clarify its involvement in resistance to CDK4/6i.

Abstract

Suppliers and large buyers of electricity often sign load-following forward contracts (LFFCs). A LFFC obligates an electricity supplier to deliver at a pre-specified unit price a fraction of the buyer's ultimate demand for electricity. We show that relative to more standard ('swap') forward contracts, LFFCs can reduce the variation in the wholesale price of electricity. However, LFFCs also can increase the expected wholesale price and thereby reduce expected consumer surplus and total surplus. Abstract

$Kepler$ revealed that roughly one-third of Sun-like stars host planets orbiting within 100 days and between the size of Earth and Neptune. How do these planets form, what are they made of, and do they represent a continuous population or multiple populations? To help address these questions, we began the Magellan-TESS Survey (MTS), which uses Magellan II/PFS to obtain radial velocity (RV) masses of 30 TESS-detected exoplanets and develops an analysis framework that connects observed planet distributions to underlying populations. In the past, small planet RV measurements have been challenging to obtain due to host star faintness and low RV semi-amplitudes, and challenging to interpret due to the potential biases in target selection and observation planning decisions. The MTS attempts to minimize these biases by focusing on bright TESS targets and employing a quantitative selection function and observing strategy. In this paper, we (1) describe our motivation and survey strategy, (2) present our first catalog of planet density constraints for 27 TESS Objects of Interest (TOIs; 22 in our population analysis sample, 12 that are members of the same systems), and (3) employ a hierarchical Bayesian model to produce preliminary constraints on the mass-radius (M-R) relation. We find that the biases causing previous M-R relations to predict fairly high masses at $1~R_\oplus$ have been reduced. Abstract

Let $K$ be a number field, and let $E/K$ be an elliptic curve over $K$. The Mordell--Weil theorem asserts that the $K$-rational points $E(K)$ of $E$ form a finitely generated abelian group. In this work, we complete the classification of the finite groups which appear as the torsion subgroup of $E(K)$ for $K$ a cubic number field. To do so, we determine the cubic points on the modular curves $X_1(N)$ for \[N = 21, 22, 24, 25, 26, 28, 30, 32, 33, 35, 36, 39, 45, 65, 121.\] As part of our analysis, we determine the complete list of $N$ for which $J_0(N)$ (resp., $J_1(N)$, resp., $J_1(2,2N)$) has rank 0. We also provide evidence to a generalized version of a conjecture of Conrad, Edixhoven, and Stein by proving that the torsion on $J_1(N)(\mathbb{Q})$ is generated by $\text{Gal}(\bar{\mathbb{Q}}/\mathbb{Q})$-orbits of cusps of $X_1(N)_{\bar{\mathbb{Q}}}$ for $N\leq 55$, $N \neq 54$., Comment: 24 pages. v2: we corrected an error in Theorem 3.1
