Historic cultural heritage and archival manuscripts have a significant importance worldwide and efforts should be considered to preserve such property for as long as possible. The current study aimed to establish a non-toxic, and eco-friendly conservation strategy for retarding paper biodeterioration. To this end, fungal isolates associated with the biodeteriorated paper manuscripts of the historical book "DESCRIPTION DE L´EGYPTE" dating back to 1823 A.D were isolated. These isolates were identified as Asperigllus niger, Aspergillus flavus, Penicillium chrysogenum and Alternaria alternata and then evaluated for cellulolytic activities. Amongst them, Aspergillus niger A2 exhibited the highest cellulase activity. In in vitro study using filter papers as model, the suppressive effect of biosynthesized silver and zinc oxide nanoparticles (NPs) on A. niger A2 bioderteriorant activity was evaluated. Biosynthesized Ag-NPs and ZnO-NPs were characterized by UV-Vis spectroscopy, X-ray diffraction patterns (XRD), Transmission Electron Microscopy (TEM) and Fourier Transform Infrared Spectroscopy (FT-IR). Color changes, tensile strength, FT-IR-spectroscopy, and scanning electron microscopy of NPs-treated papers were examined. Treatments with 2 mM Ag-NPs and 2 mM ZnO-NPs exhibited the best conservation effect on treated papers and alleviated cellulose degradation. Furthermore, this treatments enhanced tensile strength and preserved the original morphology of paper fibers. These results introduced a potential approach for historical documents conservation against fungal biodeterioration through NPs sprays. Background: The plant pathogens significantly affect quality and marketability of several cereal crops. Of these, seed-borne phytopathogens are responsible for quality and quantity losses in crops on commercial scale. The aim of this study was to isolate and detect the seed-borne mycoflora from seed lots of various bread wheat (Triticum aestivum L.) genotypes widely grown in Bahawalpur division of south Punjab, Pakistan. Methods: Eleven commercial wheat cultivars/genotypes were investigated through standard blotter paper method. The samples were randomly collected from farmer fields and grain markets. The collected samples were brought to the laboratory and analyzed with different methods to detect the mycoflora. Results: Twenty-one fungal species were recovered from seeds lots of diversified gene pool. The most frequently isolated fungi were Alternaria alternata, Fusarium monoliformae, Aspergillus flavus, Helminthosporium spp., Curvularia spp., Bipolaris sorokiniana, Phoma spp. and Penicillium spp. The one devasting fungal specie Alternaria alternata (88.8%) was the dominating species in all wheat genotypes included in the study. Conclusion: These results divulge the presence of fungal pathogens in all wheat cultivars and could exert adverse effect on their allometric traits. So, there is a potential need to control these seed borne pathogens to minimize the crop yield losses.

Paper items from historical archives and libraries are frequently colonized by biodeteriogens, the management of which is a major concern. Essential oil Volatile Organic Compounds (VOCs) of thymol, carvacrol and eugenol, with high levels of antimicrobial and insect repellent activity, were stabilized within crystalline networks of β-cyclodextrins and phenazine-based cocrystals, as a new tool for the control of paper-degrading agents. These formulations were obtained via solvent-free methodologies and resulted as easy handling powders, suitable for the treatment of paper items by indirect contact. Their antimicrobial activity was evaluated on the following species isolated from a book depository at Forte Belvedere (Florence, IT): Alternaria alternata , Aspergillus sp., Cladosporium sp., Trichoderma orientale , Metschnikowia sp., and Bacillus sp. Both formulates displayed a significant antimicrobial activity in vitro , with cocrystals showing higher efficacy than β-cyclodextrins. The formulates were also tested against the pest Lasioderma serricorne , towards which the cocrystals entrapping carvacrol and thymol exhibited repellent activity. Overall, the phenazine-carvacrol cocrystal was the best-performing formulate, also giving favourable outcomes in terms of antifungal activity in an on-paper in vitro experiment designed to reproduce on a small-scale the critical conditions of an infested archive. These promising results pave the way towards further experimentations of VOC-based solid formulates, to shed light on such products applicability for the preservation of paper items. Microbial contamination control in indoor environments, such as libraries and archives, represents a challenge. Essential oils (EOs), well-known for their antimicrobial properties, have been applied in pharmaceutical and food industry from many years. In the present study, Thymus vulgaris and Origanum vulgare EO antimicrobial efficacy on paper-born microorganisms, Staphylococcus epidermidis, Rhodotorula mucilaginosa and Alternaria alternata, was investigated to protect water-damaged paper documents and to control indoor air quality for operator's health safety. T. vulgaris EO was the most effective: Minimum Inhibitory Concentration (MIC) values obtained for S. epidermidis and R. mucilaginosa, with a broth macro-dilution method, were 7.5 µg/mL and 5.63 µg/mL, respectively. T. vulgaris EO (0.75% v/v), nebulized immediately after the inoculation on agar plates or paper sheets, showed a high inhibition effect against the three biodeteriogenic microorganisms, also when lyophilized on paper sheets; in this last case, the EO has a higher efficacy when applied immediately after the freeze drying. Regarding the EO effect against A. alternata, the inhibition percentage of the mycelial growth, MGI, (81.4%), observed for nonsporulated mycelium, was higher than that for the sporulated one (51.4%). Finally, T. vulgaris EO (0.75% v/v) was effectively applied on a real contaminated book cover by means of EO impregnated contact sheets. Pea (Pisum sativum) is an economically important legume crop, cultivated worldwide due to its high nutritional value and diverse culinary applications. However, like any agricultural crop, peas are vulnerable to various biotic and abiotic stresses, including fungal infection. The present paper deals with a study to identify the mycoflora associated with pea seed. The seed borne mycoflora of pea was examined by agar plate method, blotter method and Deep freezing blotter paper method. A total 17 species of fungi viz. Alternaria alternata, Aspergillus niger, Aspergillus flavus, Aspergillus fumigatus, Aspergillus candidus, Curvularia sp., Penicillium expansum, Mucor spp., Rhizopus nigricans, Rhizophus stolnifer, Fusarium monliforme, Fusarium oxysporum, Fusarium solani, Macrophomina phaseolina, Rhizoctonia solani, Stemphylium sp., Trichoderma harzianum and some unidentified species were reported. Among the method used for detection of seed borne fungi, the standard blotter paper method is more effective followed by agar plate method and deep freeze blotter paper method. The fungal inhibitory and preservative effects of essential oils (EOs) on the dominant spoilage fungi of blue honeysuckle were investigated. First, the dominant spoilage fungi (CW1, CW3, and W53) were isolated from naturally infected blue honeysuckle and identified through morphological features, inoculation study, and rDNA-ITS analysis were matched to Alternaria alternata , Metuloidea reniformis , and Rhizopus microsporus , respectively. Next, the filter paper method determined the sensitivity of EOs to the dominant spoilage fungi, and four kinds with better antifungal effects were screened out. Two or three combinations of four EOs were evaluated using a checkerboard analysis method. The fractional inhibitory concentration index (FICI) was used to assess the interactions between the combinations. The combinations of cinnamon/oregano, cinnamon/clove, oregano/clove, and cinnamon/oregano/clove showed synergistic effects (FICI ≤0.5). The best three-oil combination had a volume ratio of 31:8:8 compared to a single oil. Finally, in vivo, experiments showed that combining triple groups of EOs could prevent the spoilage of postharvest honeysuckle. Compared with the control group, the triple groups of essential oil combination maintained the best color, quality, and flavor of blue honeysuckle berries. At the same time, the spoilage rate and the weight loss rate were reduced by 38.50% and 40.71%, respectively. Delayed spoilage time to 3–5 days and the preservation effect of blue honeysuckle was extended to 16 days. Therefore, this study provides a theoretical basis for substituting chemical preservatives and highlights the potential of using complex EOs to prolong the freshness of blue honeysuckle. Background: Alternaria alternata is a melanic fungus capable of causing a wide variety of infections, some of which have lethal potential. It is a ubiquitous fungus and a well-known plant pathogen. Cutaneous infections with Alternaria alternata most often occur in the extremities of patients who perform conventional agriculture, thus being exposed to occupational hazards leading to the disruption of the skin barrier. Methods: This paper presents the first case report from Romania of an itraconazole nonresponsive cutaneous alternariosis in a patient without any type of immunosuppression. Results: After an initial misdiagnosis regarding the etiology of the patient's skin infection, two successive punch biopsies, followed by mycologic examination, lead to the final diagnosis of cutaneous alternariosis. Treatment guided by antifungal susceptibility testing has been instituted, leading to the gradual healing of the patient's skin ulcerations. Conclusions: The ability of Alternaria alternata to infect immunocompetent human hosts and to develop resistance to antifungal drugs highlight the importance of correctly diagnosing the etiology of skin ulcerations and instituting appropriate treatment guided by antifungal susceptibility testing whenever the suspicion of a fungal skin infection is plausible. Fungal decay is one of the most significant causes of postharvest losses of blueberries, with Botrytis rot caused by Botrytis cinerea and Alternaria rot caused by Alternaria alternata being the two most destructive fungal diseases. Plant essential oil has attracted the extensive attention of scholars due to its natural antifungal and anti-corrosion effects. In this study, the effects of fumigation treatment with plant essential oils on the growth of pathogenic fungi in blueberry fruits in vitro and the activity of defense-related enzymes of fungi-inoculated blueberry were evaluated. The results showed that, of the six natural plant essential oils of cinnamon, oregano, clove, tea tree, pomelo peel, and rosemary, oregano essential oil had the most efficient inhibitory effect on Botrytis cinerea and Alternaria alternata in PDA. After fumigating inoculated blueberry fruits with concentration gradients of 0, 30, 60, and 90 μL/L of oregano essential oil, it was found that the activity of defense-related enzymes such as phenylalanine ammonia lyase (PAL), polyphenol oxidase (PPO), peroxidase (POD), chitinase (CHI), and β-1,3-glucanase (GLU) in the inoculated blueberry fruits was induced and enhanced to varying degrees throughout the entire storage period, effectively enhancing the resistance of blueberry fruits to pathogenic fungi and reducing the postharvest decay caused by Botrytis cinerea and Alternaria alternata. The optimal concentration for the fumigation treatment with oregano essential oil is 60 μL/L. This paper investigates the marine effects that affect Qait Bey Fortress in Alexandria—Egypt. It presents the results of scientific studies and examinations to evaluate the deterioration conditions that affected archaeological buildings in the marine environment. In Alexandria, many monumental sites and stone buildings have suffered from many aggressive factors of deterioration (mechanical, chemical, and biological), which have caused great harmful appearances and threatened to eradicate them. These effects include the chemical actions resulting from seawater and marine aerosol and the mechanical actions of water waves. (Qait Bey Fortress), as a case study, was periodically investigated by many scientific techniques over five years to check its decay conditions and to define the most suitable conservation approaches and non-destructive methods for preservation. Different techniques and examinations were carried out to evaluate the current deterioration state of the fortress. For example, XRD analysis and PM investigation were used to study the mineralogical compositions, lithotype, and petrographic characteristics of the stone samples. SEM was used to investigate the morphological features of the same samples. AAS was also used for studying the chemical constituents of seawater samples. In addition, microbiological investigations were conducted to evaluate the colored hard crusts that affected the stone surfaces in the fortress. Our results proved that severe deterioration factors influenced the fortress by collaborating with chemical, mechanical, and biological mechanisms. These mechanisms caused several manifestations, such as abrasion and attrition, crystallizing of salt species, mortar desegregation, pitting and minerals' honeycomb (Alveolar), color changes, in addition to the accumulation of black and colored biogenic hard crusts composed of numerous tightly adjoining pits of several centimeters. Crusts, such as yellow to bluish green, resulted from P. aeruginosa and granular appearance having brownish ting in the center resulted from P. clacis. In addition, other pigmented features resulted from Bacillus firmus and Bacillus atrophaeus. The presence of some black and dark color crusts was attributed to the growth of some fungal species, such as A. niger, A. phoenicis, Cladosporium cladosporioides, and Alternaria alternata. The objective of this paper was to study the phyto-inhibitory and antimicrobial activity of brown propolis collected from the counties of four regions in Romania. The main physico-chemical and functional properties of 16 samples of propolis from different landforms of geographical regions were determined. Their antimicrobial activities were established against 5 bacterial strains (Pseudomonas fluorescens, Bacillus subtilis, Bacillus cereus, Escherichia coli, and Proteus mirabilis) and 5 fungal strains (Alternaria alternata, Cladosporium cladosporioides, Fusarium oxysporum, Mucor racemosus, and Aspergillus niger). Simultaneously, the phyto-inhibitory effect of propolis samples on different cereals was highlighted: hexaploid bread wheat (Triticum aestivum), maize (Zea mays L.), oats (Avena sativa L.), and barley (Hordeum vulgare L.). Correlations between the antioxidant activity and total flavonoid and phenol content of the propolis samples were identified, respectively, and the statistical analysis highlighted that the diameter of the inhibition zone was influenced by the strain type (bacterial and fungal) and the geographical regions of propolis. Principal component analysis (PCA) indicated that out of seven principal components, only two exhibited > 0.5. Pearson's correlation coefficient showed a low and moderate positive linear relationship between the diameter of the inhibition zone and the flavonoid and phenol concentration of the propolis samples. Dandong has become the largest strawberry production and export base in China. Strawberry black spot disease is widespread and causes significant economic losses to strawberry growers in both the growing and harvest seasons. Until now, no study has reported the presence of the Alternaria species, the pathogen of strawberry black spot disease, in Dandong, Liaoning province, China. In 2020–2022, 108 isolates were obtained from strawberry leaves with typical symptoms of strawberry black spot disease from 56 major professional growing operations. Combined with morphological and molecular characteristics, the majority of isolates were identified as A. tenuissima (78 isolates, 72.2%), which had established total supremacy, followed by A. alternata (30 isolates, 27.8%). The pathogenicity results show that A. tenuissima and A. alternata are the two main pathogenic factors of strawberry black spot disease, the disease indexes of which were designated as 49.6–100.0% and 20.4–59.5%. To our knowledge, this paper is the first to identify A. tenuissima and A. alternata as causing black spot disease in strawberries in Dandong, China. [ABSTRACT FROM AUTHOR]

The present investigation “Effect of selected seed-borne fungi on seed germination and vigour index of Legume seeds” was conducted during 2006-2007 in the laboratory at Research Center for Advanced Studies in Plant Sciences, P.G. of Department Botany, Shivaji Mahavidyalaya, Udgir Dist. Latur. Legume seed mycoflora, in their order of merit as Blotter paper method and Agar plate methods, were found to be the most effective in seed health testing. Large amounts of processing tomato are grown in Xinjiang, China. Tomato black spot disease, caused by Alternaria spp., and the produced alternaria toxins in tomato products are posing risks to human health. In this study, we isolated a rhizospheric bacterium, XJ-BV2007, from tomato (Solanum lycopersicum) fields, which we identified as Bacillus amyloliquefaciens. We found that this bacterium has a strong antagonistic effect against Alternaria alternata and reduces the accumulation of alternaria toxins in tomatoes. According to the antifungal activity of the bacteria-free filtrate, we revealed that B. amyloliquefaciens XJ-BV2007 suppresses A. alternata by the production of antifungal metabolites. Combining semi-preparative high-performance liquid chromatography, we employed UPLC-QTOF-MS analysis and the Oxford cup experiment to find that fengycin plays an important role in inhibiting A. alternata. This paper firstly reported that B. amyloliquefaciens efficiently controls tomato black spot disease and mycotoxins caused by A. alternata. Fritillaria taipaiensis P. Y. Li is one of the biological sources for Fritillariae Cirrhosae Bulbus. Its bulbs are widely used for treating respiratory diseases such as pneumonia, bronchitis and influenza. Cultivated F. taipaiensis suffers from many diseases during its growing season. Leaf spot is a destructive disease that is increasingly affecting F. taipaiensis and can cause an incidence of up to 30% in severe cases. Leaf spot inhibits the growth of F. taipaiensis by causing disease spots on the surface of leaves. In severe cases, these spots can result in leaf desiccation and blackspot formation at the lesion site, leading to a decrease in photosynthesis. Leaf spot has shown little benefit, and it can even result in a reduced yield of bulbs and the death of plants. According to previous studies, Alternaria alternata has been identified as the pathogen of leaf spot in many medicinal plants, but the main pathogens of the leaf spot of F. taipeiensis remains uncertain. In this paper, five isolates from diseased leaves of F. taipaiensis were isolated and purified and the pathogenicity test showed that isolates B-5 and B-7 induced leaf spot symptoms on healthy F. taipaiensis leaves. Integrating multiple phylogenetic analyses of rDNA using Internal transcribed spacer region (ITS), Beta-tubulin (TUB2), RNA polymerase II second largest subunit (RPB2) and Translation elongation factor 1-alpha (TEF1-a) primers, strain B-5 and strain B-7 were eventually identified as Didymella segeticola and A. alternata. The paper deals with the research results of studies of antimicrobial activity of actinobacteria isolated from water column and biofilm of the lake system La Izvor (Republic of Moldova). Antimicrobial activity of 34 strains of actinobacteria (Actinomadura, Actinoplanes, Frankia, Geodermatophilus, Micromonospora, Nocardia, Rhodococcus, Streptomyces) was determined by the disk diffusion method, as test cultures served pathogenic bacteria and fungi which are common on territory of Republic of Moldova and cause diseases of crops. According to results of antibacterial activity, the diameter of growth inhibition zones varied from 9.0 to 16.3 mm (active strains are represented by genera Streptomyces and Micromonospora). The best results of antifungal activity were noted by strains of genera Actinomadura, Actinoplanes, Micromonospora, and Streptomyces. Diameter of growth inhibition zones of such phytopathogenic fungi as Alternaria alternata, Aspergillus niger, and Fusarium solani varied within 20.7-24.0 mm. New strains of Actinoplanes, Actinomadura, Micromonospora, and Streptomyces will replenish the National Collection of Non-pathogenic Microorganisms of the Institute of Microbiology and Biotechnology. The present study was conducted to detect seed borne mycoflora associated with seven cultivars of sunflower (Helianthus annuus L.) and five cultivars of Zinnia (Zinnia elegans L.) by using Agar and Blotter paper methods. A total 12 genera of fungi were detected including; Alternaria alternata, Aspergillus flavus, Aspergillus niger, Cladosporium sp., Stemphylium helianthi, Penicillium sp., Fusarium oxysporum, Mucor sp., Fusarium moniliforme, Fusarium solani, Rhizocotonia solani and Rhizopus sp. The detected fungi depleted germination 10-30%, 10-20% and seedling mortality 10-17%, 10-12%. in sunflower and Zinnia respectively. A comparative study was designed to evaluate three fungicides to control seed mycoflora at the rate of of 1.5g/kg, 2.0g/kg, 2.5g/kg, 3.0g/kg, Maximum germination percentage was attained in case of seed dressing with Topsin M-70 followed by Bayton and Dithane M-45 at the rate of 3.0g/kg. Alternaria species are well‐known aggressive pathogens that are widespread globally and warmer temperatures caused by climate change might increase their abundance more drastically. Early blight (EB) disease, caused mainly by Alternaria solani, and brown spot, caused by Alternaria alternata, are major concerns in potato, tomato and eggplant production. The development of EB is strongly linked to varieties, crop development stages, environmental factors, cultivation and field management. Several forecasting models for pesticide application to control EB were created in the last century and more recent scientific advances have included modern breeding technology to detect resistant genes and precision agriculture with hyperspectral sensors to pinpoint damage locations on plants. This paper presents an overview of the EB disease and provides an evaluation of recent scientific advances to control the disease. First of all, we describe the outline of this disease, encompassing biological cycles of the Alternaria genus, favorite climate and soil conditions as well as resistant plant species. Second, versatile management practices to minimize the effect of this pathogen at field level are discussed, covering their limitations and pitfalls. A better understanding of the underlying factors of this disease and the potential of novel research can contribute to implementing integrated pest management systems for an ecofriendly farming system. © 2021 The Authors. The aim of this paper was to study the microbiological stability of bio-based composite building materials, which are made using organic-rich lake sediments (further - sapropel) with lime and magnesium cement as binders and hemp shives as filler. The microbial stability properties of the obtained composite materials were investigated and compared to similar composites. Because of their high organic content, these materials are prone to biodegradation; therefore, they were coated with ALINA LIFE TM organoclay coating, which helps to extend the product life, reducing the rate of biodegradation compared to the biocides used in industry. The effect of the coating on the resistance to decay by the Aspergillus versicolor, Penicillium chrysogenum, Alternaria alternata, Cladosporium herbarum, Chaetomium sp. and Trichoderma asperellum fungi was investigated under different conditions: relative humidity modes of 75% and 99% at 20°C for 45 days and four months. The results indicated that the composites made of sapropel and lime have similar microbial stability properties as lime and magnesium cement binder composite materials. ALINA LIFE TM organoclay coating showed advanced resistance to biodegradation; sapropel-lime composites have shown several good properties that make them suitable to be considered for use in the construction material industry. We report a sensitive lateral flow assay (LFA) in which the assay colour change originated from reporter labels constructed from silica spheres (radius = 450 nm) coated with approximately 3.9 × 103 gold nanoparticles (radius = 8.5 nm). These reporter labels were modified with DNA and deposited in the conjugation area of an LFA device assembled on wax-patterned Fusion 5 paper. Test and control zones of the device were pre-loaded with capture probe formed by avidin-coated mesoporous silica nanoparticles attached with biotin-tagged DNA sequences. Proof-of-concept was demonstrated by the detection of a partial sequence of the actin gene of Colletotrichum truncatum. The DNA target could be detected with an LOD of 46 pM, which was 5 times lower than a comparative assay using gold nanoparticles alone. The assay showed good selectivity against the Colletotrichum species C. scovillei and C. gloeosporioides , as well as against DNA from the fungal genera Aspergillus niger and Alternaria alternata. There was negligible change in sensor response over storage for one month at room temperature. The LFA was used to detect PCR products following extraction from mycelium. [Objectives] The paper was to investigate and identify the fungal diseases of wild and red heart kiwifruit in Qiandongnan Prefecture. [Methods] The pathogenic fungi were isolated from diseased leaves and fruits of wild and red heart kiwifruit by tissue separation method. DNA sequencing was carried out by using the sequence analysis of ribosomal rDNA-ITS region, and molecular evolutionary trees were built by using MEGA 4.0 software. Finally, the pathogenic fungi were classified and identified by combining morphological observation. [Results] The main fungal diseases were anthracnose caused by Colletotrichum gloeosporioides on wild kiwifruit, fruit anthracnose caused by C. acutatum on red heart kiwifruit, leaf soft rot caused by Fusarium incarnatum on red heart kiwifruit, and brown spot caused by Alternaria alternata on red heart kiwifruit. Alternaria black spot of pomegranate (Punica granatum) was reported for the first time in Italy. In spring 2023, an outbreak of this disease was noticed in commercial pomegranate 'Wonderful' orchards of the municipality of Misterbianco (Sicily), following an unusually rainy period. A total of 30 randomly selected Alternaria isolates recovered from typical necrotic spots of leaves and fruits were characterized. Based on the colony morphology on solid agar media (PDA and MEA), isolates were separated into three distinct morphotypes (1, 2, and 3). The first two morphotypes comprised only isolates from fruits, while morphotype 3 comprised only isolates from leaves. Multigene phylogenetic analysis of four DNA regions, including internal transcribed spacer (ITS), translation elongation factor 1-α (EF-1α), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), and a SCAR marker (OPA10–2), identified the isolates of morphotypes 1 and 2 as Alternaria alternata and morphotype 3 isolates as A. arborescens. In pathogenicity tests on unwounded leaves and fruit, the isolates of all three morphotypes produced symptoms on the leaves of three pomegranate cultivars, 'Acco', 'Wonderful', and 'Etna'. The symptoms on 'Acco' leaves were the least severe. Conversely, the fruits of 'Acco' were the most susceptible. The isolates of morphotypes 2 and 3 were not pathogenic on the fruits of 'Wonderful' and 'Etna'. This is the first report of Alternaria black spot in Italy and of A. arborescens associated with Alternaria black spot of pomegranate worldwide. [ABSTRACT FROM AUTHOR]

Background: Alternaria alternata is the primary pathogen of potato leaf spot disease, resulting in significant potato yield losses globally. Endophytic microorganism-based biological control, especially using microorganisms from host plants, has emerged as a promising and eco-friendly approach for managing plant diseases. Therefore, this study aimed to isolate, identify and characterize the endophytic fungi from healthy potato leaves which had great antifungal activity to the potato leaf spot pathogen of A. alternata in vitro and in vivo. Results: An endophytic fungal strain SD1-4 was isolated from healthy potato leaves and was identified as Talaromyces muroii through morphological and sequencing analysis. The strain SD1-4 exhibited potent antifungal activity against the potato leaf spot pathogen A. alternata Lill, with a hyphal inhibition rate of 69.19%. Microscopic and scanning electron microscope observations revealed that the strain SD1-4 grew parallel to, coiled around, shrunk and deformed the mycelia of A. alternata Lill. Additionally, the enzyme activities of chitinase and β-1, 3-glucanase significantly increased in the hyphae of A. alternata Lill when co-cultured with the strain SD1-4, indicating severe impairment of the cell wall function of A. alternata Lill. Furthermore, the mycelial growth and conidial germination of A. alternata Lill were significantly suppressed by the aseptic filtrate of the strain SD1-4, with inhibition rates of 79.00% and 80.67%, respectively. Decrease of leaf spot disease index from 78.36 to 37.03 was also observed in potato plants treated with the strain SD1-4, along with the significantly increased plant growth characters including plant height, root length, fresh weight, dry weight, chlorophyll content and photosynthetic rate of potato seedlings. Conclusion: The endophyte fungus of T. muroii SD1-4 isolated from healthy potato leaves in the present study showed high biocontrol potential against potato leaf spot disease caused by A. alternata via direct parasitism or antifungal metabolites, and had positive roles in promoting potato plant growth. Two freshwater rivers, the Amu Darya and Syr Darya, flow into the Aral Sea, but they began to diminish in the early 1960s, and by the 1980s, the lake had nearly ceased to exist due to excessive water consumption for agriculture and the unsustainable management of water resources from rivers, which transformed the Aral Sea into a hypersaline lake. Despite this, the flora and fauna of the region began to evolve in the high-salinity seabed soil, which has received little attention in studies. In this study, we isolated approximately 1400 bacterial strains from the rhizosphere and phyllosphere of plant species of distinct families. Bacterial isolates were examined for antifungal activities against a range of pathogenic fungi such as Rhizoctonia gossypii, Trichothecium ovalisporum, Fusarium annulatum, F. oxysporum, F. culmorum, F. brachygibbosum, F. tricinctum, F. verticillioides, Alternaria alternata, A. terreus, Aspergillus niger, and As. flavus. Eighty-eight bacterial isolates exhibited varying antagonistic ability against pathogenic fungi. Furthermore, DNA barcoding of isolates using the 16S rRNA gene indicated that most antagonistic bacteria belonged to the Bacillus and Pseudomonas genera. The study also explored the activity of hydrolytic and cell-wall-degrading enzymes produced by antagonistic bacteria. The findings revealed that antagonistic bacteria can be utilized to widely protect seabed plants and plants growing in saline areas against pathogenic fungi, as well as agricultural crops. [Objectives] The paper was to identify Alternaría alternata causing leaf spot disease on Huangdi banana in China. [Methods] Fungal isolates were isolated and identified by morphological features, molecular identification and pathogenicity test. [Results] There were light to dark brown, tiny oval spots on leaves. The causal agent isolated from affected leaves was identified as A. alternata based on the morphological properties, coupled with sequence analyses of the internal transcribed spacer (IIS) region, large subunit ribosomal DNA (LSU rD-NA) and the translation elongation factor 1-alpha (TEF-la) gene. Koch's postulates were fuEilled by successful re-isolation of pathogen from the artificial inoculated leaves. [ Conclusions] To our knowledge, this is the first report of leaf spot caused by A. alternata on Huangdi banana in China. The identification of A. alternata as the causal agent of the observed leaf spot disease on Huangdi banana is critical to the prevention and control of this disease in the future. In the paper are presented the peculiarities of reaction of the new created tomato lines to the treatment with the culture filtrates (CF) of some important phytopathogens - Alternaria alternata, Fusarium oxysporum, F. solani, based on germination, stem length and embryonic roots. The clusterian analysis (k-mean method) proved that, in comparison to A. alternata, the F. oxysporum and F. solani fungus species exhibited a higher discriminative capacity of the tomato clusters based on the root and stem length, which reveals the specificity of greater interaction with these pathogens. Clusters of tomato genotypes with diminished reaction to Fusarium spp., A. alternata pathogens have been identified, which is important for their involvement in the breeding programs. By factorial analysis of the variance, it was found that for the embryonic root of the tomatoes, the major contribution to the source of variation in character had the genotype of the plant, its contribution constituting 46.7%, and for the growth of the stem - factor of isolate: 62.5%. Abstract—: Antimicrobial, phytotoxic, and insecticidal activity of 20 isolates of 16 Alternaria species isolated from above-ground organs of weeds and wild herbaceous plants was determined. Assessment of the antibacterial activity by the agar blocks and paper disks methods revealed antibacterial activity against Bacillus subtilis and/or Pseudomonassyringae in over 70% of the isolates. Antifungal activity against Candida tropicalis was found in 30% of the isolates. The reporter system used in the work revealed no effect of the studied extracts on the ribosomes, although some of them inhibited topoisomerase. Extracts from the culture liquid of all Alternaria spp. isolates were toxic to isolated leaves of Arabidopsis thaliana and Elytrigia repens. Insecticidal activity of the extracts against vetch aphid (Megoura viciae) larvae was revealed in 40% of the isolates. Thus, fungi from the phyllosphere of weeds and wild herbaceous plants may exhibit a broad range of biological activity. This feature may probably be of practical importance for toxicological assessment of those weed pathogens that are evaluated as potential bioherbicides. Our results show that Alternaria fungi may play a significant regulatory role in formation of the communities of phyllosphere organisms and are promising producers of antimicrobial compounds with potentially new mechanisms of action. Endophytic strains were isolated from different parts of a healthy “Dongzao” jujube (Ziziphus jujuba Mill. ‘Dongzao’) to find biocontrol agents against jujube shrunken-fruit disease caused by Alternaria alternata. The strains were screened using A. alternata strain CN193 as the target pathogen. The nutrient competition for all isolates was studied using the dual culture, and their inhibitive capability was tested by measuring the inhibition width of filter paper disks with filtrate. Influence of filtrate from the selected strains with strong inhibition of mycelial growth on spore germination was studied with hanging drop method on concavity slides. Colonization in the jujube leaves was assayed using a rifampicin-resistant mutant of strain St-zn-34 as the screening marker. Strains were identified based on their morphological, physiological, and biochemical characteristics, 16S rDNA sequencing, and phylogenetic analysis. A total of 81 endophytic strains were isolated from the stems, leaves, flowers, and fruits of winter jujube. Among these isolates, 14 strains showed strong antagonism against A. alternata. Further study showed that the filtrate of strains St-zn-9 and St-zn-34 could inhibit the mycelial growth of A. alternata, and the widths of their inhibition zone reached 6.14±0.03 mm and 8.27±0.09 mm, respectively. However, strain St-zn-34 showed stronger inhibition on spore germination than strain St-zn-9. St-zn-34 could significantly reduce the spore germination rate of A. alternata, and the spore did not germinate at all or the germ tube was very short. A rifampicin resistant-derivative of wild-type strain St-zn-34, which was designated as St-zn-34r, was obtained by transferring the strains to media with stepwise-increased rifampicin. Colonization assays indicated that St-zn-34r could colonize in jujube leaves, and the population of St-zn-34r was 1.2×103 CFU/g FW after inoculation for 30 days. Except for its salt tolerance, St-zn-34 was the closest to those of Bacillus subtilis. Thus, the strain was identified as B. subtilis. [ABSTRACT FROM AUTHOR]

Lignocellulosic plant biomass is the most abundant carbon source in the planet, which makes it a potential substrate for biorefinery. It consists of polysaccharides and other molecules with applications in pharmaceutical, food and feed, cosmetics, paper and textile industries. The exploitation of these resources requires the hydrolysis of the plant cell wall, which is a complex process. Aiming to discover novel fungal natural isolates with lignocellulolytic capacities, a screening for feruloyl esterase activity was performed in samples taken from different metal surfaces. An extracellular enzyme extract from the most promising candidate, the natural isolate Alternaria alternata PDA1, was analyzed. The feruloyl esterase activity of the enzyme extract was characterized, determining the pH and temperature optima (pH 5.0 and 55–60 °C, respectively), thermal stability and kinetic parameters, among others. Proteomic analyses derived from two-dimensional gels allowed the identification and classification of 97 protein spots from the extracellular proteome. Most of the identified proteins belonged to the carbohydrates metabolism group, particularly plant cell wall degradation. Enzymatic activities of the identified proteins (β-glucosidase, cellobiohydrolase, endoglucanase, β-xylosidase and xylanase) of the extract were also measured. These findings confirm A. alternata PDA1 as a promising lignocellulolytic enzyme producer. Significance Although plant biomass is an abundant material that can be potentially utilized by several industries, the effective hydrolysis of the recalcitrant plant cell wall is not a straightforward process. As this hydrolysis occurs in nature relying almost solely on microbial enzymatic systems, it is reasonable to infer that further studies on lignocellulolytic enzymes will discover new sustainable industrial solutions. The results included in this paper provide a promising fungal candidate for biotechnological processes to obtain added value from plant byproducts and analogous substrates. Moreover, the proteomic analysis of the secretome of a natural isolate of Alternaria sp. grown in the presence of one of the most used vegetal substrates on the biofuels industry (sugar beet pulp) sheds light on the extracellular enzymatic machinery of this fungal plant pathogen, and can be potentially applied to developing new industrial enzymatic tools. This work is, to our knowledge, the first to analyze in depth the secreted enzyme extract of the plant pathogen Alternaria when grown on a lignocellulosic substrate, identifying its proteins by means of MALDI-TOF/TOF mass spectrometry and characterizing its feruloyl esterase, cellulase and xylanolytic activities.
