Swine production has focused its efforts to reduce the amount and improve the quality of fat deposited on carcass. Due to the association of the lipid profile with cardiovascular diseases, researches have been developed to understand how the deposition occurs and to improve the fatty acid profile of swine fat. Among the factors that affect the fat composition, race, sex, and diet, the third one has the greatest impact. Lipid inclusion seems to be a promising way to modulate the lipid profile of fat in pig carcass, with an effect on saturated, monounsaturated and polyunsaturated fatty acids. The effect is greater in that fatty acid with the highest concentration in the source used. Soybean oil can double the value of linoleic acid, a by-product of olive oil increases oleic acid by 4%, and flaxseed oil has the ability to increase α-linolenic acid from less than 1% to over 18% in total fatty acids. The effect of copper and nickel ions on the lipid composition of freshwater mussels Anodonta cygnea was investigated using an aquarium experiment. The contents of the main lipid classes were determined using thin layer chromatography and spectrophotometry, and the fatty acid composition was analysed using gas--liquid chromatography. The results indicated that copper and nickel had different effects on the mussels' lipid composition, with copper producing less effect on the composition of lipids and fatty acids than nickel. On the first experiment day, excessive copper content in the organism caused oxidative stress and modifications in membrane lipids, which returned to their initial level by the end of the experiment. Nickel accumulated predominantly in gills; however, significant modifications in lipids and fatty acids were seen also in the digestive gland from the first day of the experiment. This indicated the activation of nickel-induced lipid peroxidation processes. Moreover, this study revealed a dose-dependent effect of nickel on lipid composition, which was likely related to the development of compensatory biochemical mechanisms in response to nickel-induced oxidative stress. A comparative study of the lipid composition alteration in mussels in response to copper and nickel action revealed the consequences of the toxic impact of metal ions and the defensive mechanisms that organisms employ to detoxify and remove xenobiotics. Chemical and physical mutagens like sodium azide, H2O2 and UV light have a significant role in the metabolism of living organisms. Effect of these mutagens was recorded during the present work on lipid content of Oscillatoria sp. and Ulothrix sp. The algal species were subjected to the mutagens by adding them in culture media. Lipids were extracted by Microwave-assisted extraction using methanol, ethanol and chloroform. During the present work generally a significant effect of algal species and mutagens was observed. All the tested mutagen significantly affected the lipid content. A significant effect of solvent on lipid content of algal species was observed when the algae were extracted with different solvents after treatment with H2O2 and UV light however this effect was non-significant when algae were subjected to sodium azide. In general, the mutagen resulted in an increase in lipid content. In Oscillatoria sp. the lipid content was generally less as compared to Ulothrix sp. The highest lipid content of 0.5433±0.002 g100g-1 was recorded in Ulothrix sp. after 3 days stress of sodium azide stress followed by H2O2 treated (50mM) samples with a content of 0.5133±0.05g100g-1. In Oscillatoria sp. 15mM treatment for 3 days gave a lipid content of 0.3733±0.0001g100g-1 after 3 days. A reduction in fresh weights of both tested algal species was recorded after UV stress. The oil content also showed a significant reduction after UV exposure. GC-MS analysis of the transesterified lipids showed that different fatty acids show variation when algal samples were treated with mutagens. Lipid membrane turnover and myelin repair play a central role in diseases and lesions of the central nervous system (CNS). The aim of the present study was to analyze lipid composition changes due to inflammatory conditions. We measured the fatty acid (FA) composition in erythrocytes (RBCs) and spinal cord tissue (gas chromatography) derived from mice affected by experimental allergic encephalomyelitis (EAE) in acute and remission phases; cholesterol membrane content (Filipin) and GM1 membrane assembly (CT-B) in EAE mouse RBCs, and in cultured neurons, oligodendroglial cells and macrophages exposed to inflammatory challenges. During the EAE acute phase, the RBC membrane showed a reduction in polyunsaturated FAs (PUFAs) and an increase in saturated FAs (SFAs) and the omega-6/omega-3 ratios, followed by a restoration to control levels in the remission phase in parallel with an increase in monounsaturated fatty acid residues. A decrease in PUFAs was also shown in the spinal cord. CT-B staining decreased and Filipin staining increased in RBCs during acute EAE, as well as in cultured macrophages, neurons and oligodendrocyte precursor cells exposed to inflammatory challenges. This regulation in lipid content suggests an increased cell membrane rigidity during the inflammatory phase of EAE and supports the investigation of peripheral cell membrane lipids as possible biomarkers for CNS lipid membrane concentration and assembly. Organic waste is generated worldwide, and its disposal and recycling are becoming a challenge. Due to its high carbon content, however, it may be converted into valuable products. Carbon neutrality is essential, and unstable international oil prices stress the increasing importance of biofuels significantly. Volatile fatty acids (VFA) derived from organic waste can be converted to microbial lipids by oleaginous yeast using it as a carbon source. When VFA is consumed by oleaginous yeast, the pH of the medium rises; hence, acidic agents have to be added to the medium to maintain the broth's pH. In this study, we enhanced microbial lipid productivity by automatic fed-batch cultivation using VFA as an acidic agent, and the modified cultivation showed 48.9% and 69.0% higher biomass and lipid productivity than manual multi-fed culture. At a VFA concentration of 5 g/L and pH 7.0, a lipid yield of 0.25 g/g alongside lipid productivity of 0.11 g/L/h was obtained from an automatic fed-batch system. Background. Patatin-like phospholipase domain-containing 3 (PNPLA3) is a major susceptibility gene for nonalcoholic fatty liver disease (NAFLD), and its rs738409 (I148M) polymorphism is associated with the occurrence and progression of NAFLD. Endoplasmic reticulum (ER) stress-related hepatocyte lipoapoptosis contributes to the progress of NAFLD. PNPLA3 is also known as a member of the calcium-independent phospholipase A2ε family, which can hydrolyze fatty acids to generate lysophosphatidylcholine (LPC) that induces ER stress-related hepatocyte lipoapoptosis. Whether the PNPLA3 risk genotype 148M/M is involved in more severe ER stress-associated lipoapoptosis is unclear. Methods. A PNPLA3148I knock-in HepG2 cell model was constructed based on HepG2 expressing PNPLA3 148M/M using the Cas9/sgRNA system. PNPLA3 148M/M, I/M, and I/I cells were treated with 0.3 mM palmitic acid (PA) for 24 h to induce lipid deposition. Cellular lipid deposition was detected by oil red staining. Apoptosis was observed by TUNEL. LPC was determined by ELISA, and the expression of PNPLA3, the ER stress marker Bip, molecules involved in the ER stress PERK/elF-2a pathway, and its downstream C/EBP homologous protein (CHOP)-mediated apoptotic pathway were detected by western blot. Results. The results showed no difference in PNPLA3 basal expression and basal hepatocyte lipid content between the three genotypes of cells. Lipid deposition and apoptosis were more severe in PNPLA3 148M/M and 148I/M cells than in I/I cells after PA treatment. PA-induced upregulation of protein expression of Bip, ER stress-responsive PERK pathway molecules p-PERK, p-eIF2α, CHOP, and CHOP-associated apoptotic molecules PUMA and Bax were more pronounced in PNPLA3 148M/M cells than in PNPLA3 148I/I cells. The basal LPC levels and the PA-treated increase of LPC levels in the cell culture supernatants did not differ between the three genotypic cells. Conclusion. PNPLA3 148M/M cells were more susceptible to PA-induced lipid deposition and ER stress-related apoptosis than 148I/I cells, and the proapoptotic susceptibility of PNPLA3 148M/M is independent of LPC. Kefalotyri cheese made in December and April using milk from sheep grazing on pastures was aerobically kept, in the dark or under fluorescent light, for 40 days at 4°C. Unsaturated fatty acids, which were higher in April than in December cheese, were significantly (P < 0.05) decreased in all cheese samples under both storage conditions. According to lipid hydroperoxides, malondialdehyde and carbonyls changes, grated cheese samples stored under fluorescent light showed a higher protein and lipid oxidation as compared to samples stored in the dark. Simple Summary: Insects are an important source of protein but insect fat increasingly slides into focus due to its high content of unsaturated fatty acids. Tenebrio molitor larvae were reared on several substrates with different nutritional contents to influence the fat and fatty acid contents of the larvae. The fat and fatty acid composition of the mealworm larvae were then analyzed in order to determine if a nutritional change could be detected using near-infrared reflectance spectroscopy. This is a rapid and non-destructive method for the online analysis of chemical composition. In this study, the diet used for rearing had a significant effect on the larval fat content and fatty acid composition as well as the absorbance of the near-infrared spectra and the larval growth rate and weight gain, i.e., a high fat content substrate reduced weight gain and larval growth. The most prevalent fatty acids identified and quantified were palmitic, oleic, and linoleic acid, showing a correlation between larval content and rearing diets containing high fatty acid amounts. A high dietary content of lauric acid, myristic acid, and α-linolenic acid resulted in a high content of these fatty acids in mealworm larvae. The fat and fatty acid content could be predicted accurately using near-infrared reflectance spectroscopy and were highly influenced by several diets having different proximate compositions. Several studies have shown that mealworms (Tenebrio molitor L.) could provide animals and humans with valuable nutrients. Tenebrio molitor larvae were studied to determine whether their rearing diets affected their fat and fatty acid content and to ascertain if it is possible to detect the changes in the larval fat composition using near-infrared reflectance spectroscopy (NIRS). For this reason, a standard control diet (100% wheat bran) and an experimental diet, consisting of wheat bran and the supplementation of a different substrate (coconut flour, flaxseed flour, pea protein flour, rose hip hulls, grape pomace, or hemp protein flour) were used. The results showed lesser weight gain and slower growth rates for larvae raised on diets with a high fat content. A total of eight fatty acids were identified and quantified, where palmitic, oleic, and linoleic acids were the most prevalent and showed a correlation between larval content and their content in the rearing diets. There was a high content of lauric acid (3.2–4.6%), myristic acid (11.4–12.9%), and α-linolenic acid 8.4–13.0%) in mealworm larvae as a result of the high dietary content of these fatty acids. NIR spectra were also influenced by the fat and fatty acid composition, as larval absorbance values differed greatly. The coefficient of the determination of prediction (R2P) was over 0.97, with an RPD value of 8.3 for the fat content, which indicates the high predictive accuracy of the NIR model. Furthermore, it was possible to develop calibration models with great predictive efficiency (R2P = 0.81–0.95, RPD = 2.6–5.6) for all fatty acids, except palmitoleic and stearic acids which had a low predictive power (R2P < 0.5, RPD < 2.0). Lipids synthesized by oleaginous yeasts are considered to be the best candidates for biodiesel production. Cryptococcus humicola as an oleaginous yeast accumulated lipid in cells. In order to optimize the conditions for lipid production, different carbon and nitrogen sources were used and metals were added into the medium. Ca2+ addition increased the lipid production greatly. Xylose and peptone were optimal carbon source and nitrogen source, respectively for lipid accumulation. Response surface experiment results revealed that the accumulation of lipid could be maximized when the xylose, peptone and Ca2+ concentration was 61 g/L, 4.31 g/L, 0.67 mmol/L. C16 and C18 fatty acid account for about 91% of the total fatty acids. The most abundant fatty acid was oleic acid (42.68%), followed by palmitic acid (29.7%) and stearic acid (13.87%). The addition of Ca2+ increased the content of unsaturated fatty acids (such as C16:1 and C18:1) and improved the unsaturation of fatty acids. Quantitative real time PCR analysis revealed that expression of genes related to lipid biosynthesis showed up-regulated by Ca2+ treatment. In this study, five different types of fish burgers were produced using 80% ground fish meat, 10% lipid, 6% spices mixture and 4% water. Burgers (5 mm thickness and 90 g weight) were produced using 100% fish oil (control), 75% fish oil and 25% mango kernel oil (MKO) (T1), 50% fish oil and 50% MKO (T2), 25% fish oil and 75% MKO (T3), 0% fish oil and 100% MKO (T4). Burgers were packaged in polyethylene bags and stored at −16 to −18°C for 90 days. The mangiferin, quercetin, catechin, chlorogenic and caffeic acid in MKO were 1381, 87, 512, and 973 mg/100 g. Total antioxidant capacity of control, T1, T2, T3, and T4 were 18.97, 31.47, 47.69, 65.37, and 78.82%. GC-MS analysis showed that ALA, EPA, and DHA) in T2 were 0.85, 3.62, and 4.19%. In controlled storage, loss of ALA, EPA, and DHA was 55, 14, and 11%. At T2 concentration, antioxidant compounds of MKO significantly altered the lipid oxidation in burgers. After storage in T2, loss of ALA, EPA, and DHA was 3.6, 1.98, and 1.71%. POV of 90 days stored control, T1, T2, T3, and T4 was 3.42, 1.55, 0.63, 0.41, and 0.31 (MeqO2/kg). Induction period of fish oil, MKO, in all levels was 2.45, 64.28, 7.33, 13.42, 18.39, and 62.84 hrs. Color flavor and texture score of 90 days stored T2 was 91, 90, and 86% of total score. Vitis vinifera L. is highly susceptible to the biotrophic pathogen Plasmopara viticola. To control the downy mildew disease, several phytochemicals are applied every season. Recent European Union requirements to reduce the use of chemicals in viticulture have made it crucial to use alternative and more sustainable approaches to control this disease. Our previous studies pinpoint the role of fatty acids and lipid signalling in the establishment of an incompatible interaction between grapevine and P. viticola. To further understand the mechanisms behind lipid involvement in an effective defence response we have analysed the expression of several genes related to lipid metabolism in three grapevine genotypes: Chardonnay (susceptible); Regent (tolerant), harbouring an Rpv3-1 resistance loci; and Sauvignac (resistant) that harbours a pyramid of Rpv12 and Rpv3-1 resistance loci. A highly aggressive P. viticola isolate was used (NW-10/16). Moreover, we have characterised the grapevine phospholipases C and D gene families and monitored fatty acid modulation during infection. Our results indicate that both susceptible and resistant grapevine hosts did not present wide fatty acid or gene expression modulation. The modulation of genes associated with lipid signalling and fatty acids seems to be specific to Regent, which raises the hypothesis of being specifically linked to the Rpv3 loci. In Sauvignac, the Rpv12 may be dominant concerning the defence response, and, thus, this genotype may present the activation of other pathways rather than lipid signalling. [ABSTRACT FROM AUTHOR]

We have examined the effect of gypenosides on hepatic lipid metabolism in hyperlipidemic mice. To this end, male mice were placed on a high-fat diet and then administered gypenosides. Compared to the mice on the high-fat diet, the levels of liver index, total cholesterol, and low-density lipoprotein cholesterol in the serum were significantly reduced by gypenoside treatment. Gypenosides influenced the overall profile of 22 hepatic fatty acid metabolism genes significantly. Three genes, Cyp4a10, Cyp4a14, and Cyp4a31, had their mRNA expression altered by about tenfold. As a possible mechanism, the findings suggest that gypenosides regulate lipid homeostasis by influencing fatty acid metabolism and promoting fatty acid ω-oxidation. [ABSTRACT FROM AUTHOR]

Aims: The study was designed to explore the role of apolipoprotein E (ApoE) deficiency concomitant with dietary docosahexaenoic acid (DHA) treatment on brain β-amyloid (Aβ) and lipid levels. Method: A 5-month dietary DHA intervention was conducted in ApoE-deficient (ApoE-/-) mice and wild-type C57BL/6J (C57 wt) mice. The Morris water maze test was performed to assess the behaviour of the animals. The cortical contents of soluble Aβ1-40 and Aβ1-42 were detected by enzyme-linked immunosorbent assay (ELISA). Cortical fatty acid levels were detected by gas chromatography. Gene and protein expression of molecules associated with cerebral Aβ and lipid metabolism were measured using real-time polymerase chain reaction (PCR), Western blot and histological methods. Results: DHA treatment increased the content of cortical DHA and n-3 polyunsaturated fatty acids (n-3 PUFAs) but decreased the ratio of n-6/n-3 PUFAs in ApoE-/- mice; whereas the content of cortical DHA and n-3 PUFAs in C57 wt mice remained unchanged after DHA treatment. Cerebral Fabp5 and Cd36 gene expression were significantly downregulated in DHA-fed C57 wt mice; cerebral Cd36 and Scarb1 gene expression were significantly upregulated, whereas Fabp5 gene expression was downregulated in DHA-fed ApoE-/- mice. In comparison with C57 wt mice, the content of cortical soluble Aβ1-42, total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) increased, whereas the level of high-density lipoprotein cholesterol (HDL-C) decreased in ApoE-/- mice. Interestingly, these differences were significantly reversed by DHA dietary treatment. Conclusion: DHA intervention has discrepant impacts on cerebral lipids, fatty acid transporter expression and soluble Aβ levels in ApoE-/- and C57 wt mice, suggesting the modifying role of ApoE status on the responses of cerebral lipids and Aβ metabolism to DHA treatment. Hepatitis C virus (HCV) is a major human pathogen that requires a better understanding of its interaction with host cells. There is a close association of HCV life cycle with host lipid metabolism. Lipid droplets (LDs) have been found to be crucial organelles that support HCV replication and virion assembly. In addition to their role in replication, LDs also have protein-mediated antiviral properties that are activated during HCV infection. Studies have shown that HCV replicates well in cholesterol and sphingolipid-rich membranes, but the ways in which HCV alters host cell lipid dynamics are not yet known. In this study, we performed a kinetic study to check the enrichment of LDs at different time points of HCV infection. Based on the LD enrichment results, we selected early and later time points of HCV infection for global lipidomic study. Early infection represents the window period for HCV sensing and host immune response while later infection represents the establishment of viral RNA replication, virion assembly, and egress. We identified the dynamic profile of lipid species at early and later time points of HCV infection by global lipidomic study using mass spectrometry. At early HCV infection, phosphatidylinositol phospholipids (PIPs), lysophosphatidic acid (LPA), triacyl glycerols (TAG), phosphatidylcholine (PC), and trihexosylceramides (Hex3Cer) were observed to be enriched. Similarly, free fatty acids (FFA), phosphatidylethanolamine (PE), N-acylphosphatidylethanolamines (NAPE), and tri acylglycerols were enriched at later time points of HCV infection. Lipids enriched at early time of infection may have role in HCV sensing, viral attachment, and immune response as LPA and PIPs are important for immune response and viral attachment, respectively. Moreover, lipid species observed at later infection may contribute to HCV replication and virion assembly as PE, FFA, and triacylglycerols are known for the similar function. In conclusion, we identified lipid species that exhibited dynamic profile across early and later time points of HCV infection compared to mock cells, which could be therapeutically relevant in the design of more specific and effective anti-viral therapies. New data on the fatty acid compositions of the muscle tissues of the two most widespread families of the mesopelagic zone—the Myctophidae (Notoscopelus kroyeri and Symbolophorus veranyi) and Stomiidae (Chauliodus sloani, Stomias boa, Borostomias antarcticus, and Malacosteus niger) families—were obtained from the Irminger Sea (North East Atlantic). The fatty acids (FAs) in the total lipids (TLs), phospholipids (PLs), triacylglycerols (TAGs), and cholesterol esters and waxes were analyzed using gas–liquid chromatography with a mass-selective detector and flame-ionized detector (GC-MS and GC-FID, respectively). Species-specific differences in the FA/alcohol profiles of the studied fishes were revealed. A directed deep-vise trend in the changes in the content and performance of certain FAs for the studied species was found. Along with this, a general character of dominance for monounsaturated fatty acids (MUFAs), which were discussed as food tracers, was revealed. MUFAs in the muscle tissues included dietary markers of zooplankton (copepods)—20:1(n-9) and 22:1(n-11), the content of which varied in association with the species—such that the biomarker Calanus glacialis predominated in muscles of B. antarcticus and C. hyperboreus prevailed in other studied species. Lipids are key compounds in the transfer of energy and nutrients in marine food webs. Specifically, neutral lipids are the main reservoir of dietary energy in most marine animals and are often used as indicators of nutritional condition. In contrast, polar lipids, including acetone-mobile polar lipids and phospholipids, are key structural components of cell membranes. Here, we explored the fatty acid (FA) composition across the neutral and polar lipid fractions in benthic organisms belonging to five major phyla (Arthropoda, Cnidaria, Echinodermata, Mollusca, and Porifera), which were collected from a deep-water region in the Northwest Atlantic. Overall, the FA composition of acetone-mobile polar lipids (e.g., glycolipids) and phospholipids was more similar to each other and characterized by higher proportions of saturated FA (e.g., 16:0) compared to that of neutral lipids. This result reflected the different functions (structural vs storage) of these lipid fractions. Furthermore, independently of the taxon, monounsaturated FA (e.g., 16:1ω7, 18:1ω9) were dominant in neutral lipids, suggesting their preferential utilization for energy production. Whereas several species, including the gastropod Buccinum sp., retained the essential polyunsaturated FA (i.e., ω3 and ω6 FA) in their neutral lipids, others, such as the crustacean Pasiphaea tarda and the echinoderm Phormosoma placenta incorporated them into their polar lipids, pointing to differences in the metabolism of essential FA across species. These findings highlight the value of quantifying differences among lipid fractions. Objetive. To determine the proximal composition and profile of fatty acids (FA) in Cotinis columbica Burmeister larva collected in the municipalities of Mogotes and Garzón, Bogotá, Colombia. Materials and methods. Six samples of C. columbica Burmeister larvae were collected during three months, from the municipalities of Mogotes and Garzón, Bogotá, Colombia (3 months x 2 = 6). The samples were subjected to proximal chemical analysis and AG analysis by gas chromatography with a flame ionization detector and capillary column. Results. In mogotes, 21.2 and 23.3% of fat and protein were found, respectively, while in Garzón values of 31.1 and 25.9% were found, there was no statistical difference (p≥0.05). Chromatographic analyzes determined 18 AG, from C4:0 to C22:2, c13.16. The t-student test showed significance (p<0.05) only in C10:0; through time (May, June and July) the values were higher in Garzón larvae. The contents of saturated, monounsaturated and polyunsaturated AG groups were between 30.20 and 36.92 (% w/w). Conclusions. The proximal composition and fatty acid profile were similar, except for C10:0, in the fat of Cotinis columbica Burmeister larvae from Mogotes and Garzón, Colombia. [ABSTRACT FROM AUTHOR]

Mycelial fungi grow as colonies consisting of polar growing hyphae, developing radially from spore or inoculum. Over time, the colony develops, hyphae are subject to various exogenous or endogenous stimuli, and mycelium becomes heterogeneous in growth, gene expression, biosynthesis, and secretion of proteins and metabolites. Although the biochemical and molecular mechanisms of mycelium heterogeneity have been the subject of many studies, the role of lipids in colony development and zonality is still not understood. This work was undertaken to extend our knowledge of mycelium heterogeneity and to answer the question of how different lipid molecular species are distributed in the surface colony of the basidial fungus Flammulina velutipes and how this distribution correlates with its morphology. The heterogeneity in the lipid metabolism and lipid composition of the fungal mycelium was demonstrated. According to the real-time PCR and LC-MS/MS results, the expression of genes of PC metabolism, accumulation of phospholipid classes, and degree of unsaturation of PC and PE increased in the direction from the center to the periphery of the colony. The peripheral zone of the colony was characterized by a higher value of the PC/PE ratio and a higher level of phospholipids esterified by linolenic acid. Considering that the synthesis of phospholipids in fungi occurs in different ways, we also conducted experiments with deuterium-labeled phospholipid precursors and found out that the Kennedy pathway is the predominant route for PC biosynthesis in F. velutipes. Sinapis alba L. (white mustard), an annual member of Brassicaceae, is an important oilseed crop with significant agronomic traits including tolerance to abiotic stress. The present study was aimed to assess the changes in biochemical attributes of S. alba, mainly the composition of lipids and fatty acids, in response to osmotic stress. Seedlings were subjected to stress condition by growing them on media supplemented with three different concentrations of polyethylene glycol (PEG 6000) solutions, respectively with -0.04, -0.16 and -0.64 MPa osmotic potential. Compatible solute content, lipid content, and lipid peroxidation of the seedlings were measured. Total fatty acid composition was analysed by gas chromatography mass spectrometry. Overall negative impact of PEG-mediated osmotic stress was apparent from seedling morphology. Total lipid content decreased in stressed plants together with increased level of triacylglycerol. Increase of lipid peroxidation in stressed condition indicated suppression of membrane integrity. The key outcome was the significant increase of unsaturation level of fatty acids due to higher accumulation of linoleic acid, α-linolenic acid and erucic acid in the seedlings exposed to higher osmotic stress. The results indicate that, in white mustard, the biochemical changes induced by osmotic stress mainly depend on changes in lipid composition and fatty acid unsaturation level, which maintain membrane stability during the early stage of seedling development. Macro- and microalgae are currently recognized sources of lipids with great nutritional quality and attractive bioactivities for human health promotion and disease prevention. Due to the lipidomic diversity observed among algae species, giving rise to different nutritional and functional characteristics, the mixture of macro- and microalgae has the potential to present important synergistic effects resulting from the complementarity among algae. The aim of this work was to characterize for the first time the lipidome of a blend of macro- and microalgae and evaluate the antioxidant capacity of its lipid fraction. Fatty acids were profiled by GC-MS, the polar lipidome was identified by high resolution LC-MS, and ABTS+• and DPPH• assays were used to assess the antioxidant potential. The most abundant fatty acids were oleic (18:1 n-9), α-linolenic (18:3 n-3), and linoleic (18:2 n-6) acids. The lipid extract presented a beneficial n-6/n-3 ratio (0.98) and low values of atherogenic (0.41) and thrombogenic indices (0.27). The polar lipidome revealed 462 lipid species distributed by glycolipids, phospholipids, and betaine lipids, including some species bearing PUFA and a few with reported bioactivities. The lipid extract also showed antioxidant activity. Overall, the results are promising for the valorization of this blend for food, nutraceutical, and biotechnological applications. Obesity has become one of the most prevalent chronic diseases worldwide, which affects people's health and daily lives. Therefore, this study aimed to investigate the anti-obesity effects of perilla seed oil (PSO), sunflower oil (SFO), and tea seed oil (TSO) and their potential mechanisms in mice fed a high-fat diet (HFD). Mice were divided into five groups: ND, mice fed a normal diet; HFD, mice fed a high-fat diet; PSO, SFO, and TSO, mice fed a high-fat diet supplemented with PSO, SFO, and TSO at 2 g/kg body weight per day, respectively. Our findings showed that oral supplementation with all three oils for 8 weeks significantly reduced body weight, tissue weight, insulin resistance index, serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and free fatty acids (FFA), and markedly alleviated hyperglycemia, hyperlipidemia, and hepatic steatosis in obese mice. It also decreased leptin, pro-inflammatory cytokines, such as tumor necrosis factor-alpha (TNF-α), interleukin (IL)-6, and (IL)-1beta (IL-1β), and increased anti-inflammatory adipokine adiponectin at both secretion and mRNA expression levels in the epididymal adipose tissue (EAT). Moreover, PSO, SFO, and TSO administration increased the expression levels of fatty acid β-oxidation-related genes, including peroxisome proliferator-activated receptor-alpha (PPAR-α), carnitine palmitoyltransferase 1a (CPT1a) and CPT1b, and thermogenesis-related genes such as uncoupling protein 1 (UCP1), and decreased the expression levels of lipid synthesis-related genes, including fatty acid synthase (FAS) and PPAR-γ in EAT. This study investigated the effect of five in-house cooking methods (grilling, oven-cooking, frying, microwaving, and boiling) on lipid content and fatty acid composition in red mullet (Mullus barbatus). Moreover, the nutritional quality of the lipid fraction was evaluated by determining a number of lipid nutritional quality indices (LNQI). Moisture content of red mullet decreased after cooking, with the exception of boiled fish, while the lipid content significantly increased after microwave treatment and frying. After the latter, a ten-fold increase in lipid content was noted compared to fresh fish (from 2.1 to 21.1 g/100 g wet matter). All cooking methods caused significant changes in the fatty acid (FA) profile of M. barbatus. The content of saturated fatty acids (SFAs) varied significantly between raw and cooked fish with the lowest values determined for fried sample. Monounsaturated fatty acid (MUFA) content increased after oven cooking and frying, while polyunsaturated fatty acid (PUFA) content showed an opposite trend. The eicosapentaenoic acid (EPA) and docosahexaenoic (DHA) were the dominant fatty acids among n3 acids in all samples. EPA+DHA contribution in total FAs was significantly lower in the samples after oven cooking (19.6% total FAs) and frying (4.99% total FAs) than in the raw (25.5% of total FAs) fish. The n3/n6 ratio (range 1.25-3.65) decreased significantly after cooking; however, it remained above the recommended values for a healthy human nutrition (1:5). The PUFA/SFA ratio was between 0.69 in fried and 1.02 in boiled fish, both being greater than those recommended by the WHO/FAO [2003]. Atherogenicity index value decreased significantly after the cooking particularly in fried fish. The polyene index (PI), as a measure of PUFA damage, was in the range of 0.42-1.18. Although all cooking methods affected the FA profile, based on LNQI, it can be concluded that barbecue-grilling and boiling were the best cooking methods for healthy eating, due to their lowest effect on essential FAs. Aim. The purpose of the work was to study the nutritional value of siberian sturgeon caviar Acipenser baerii, grown in aquaculture conditions in the Dnieper reservoirs of Ukraine, and to compare its quality indicators with this product produced abroad. Methods: There are identified organoleptic properties of siberian sturgeon caviar, and its energetic value, composition of amino acids in albumens and content of fatty acids in lipids of the product. The data obtained in the research were compared with the same indices of quality of caviar of sturgeons bred abroad. Results. The organoleptic properties of siberian sturgeon caviar bred in Ukraine (appearance, color, consistency, taste and aroma) conformed to its standardized indices of quality. The caviar contained all eight essential amino acids and belonged to category of products rich by albumen (21.54±2.13%), and fats eicosapentaenoic and docosahexaenoic ω-3 acids (3.46 % and 11.2 %, respectively) (13.20±0.93%). The big content of fat, especially of polyunsaturated fatty acids and ω-3 acids (the eicosapentaenoic and docosahexaenoic ones — 3.46 % and 11.2 %, respectively) is one more factor, which enables to identify the siberian sturgeon caviar as the product of high biological value. It was shown that the caviar of siberian sturgeon produced in Ukraine is in close coincidence with those that were produced in other countries. Conclusions. The totality of studied characteristics of caviar of siberian sturgeon produced in Ukraine witnesses its high nutritional value. Background: Obesity is the underlying risk factor for major metabolism complications, including non-alcoholic- fatty liver disease, atherosclerosis, and cardiovascular disease. The adipose tissue is a vital endocrine organ that plays a role in the synthesis and storage of lipid and, therefore, is a contributory factor to the development and progression of obesity. A growing interest in nutraceuticals suggests that natural products can alleviate the risk factors and may be effective in mitigating obesity. Aim: The objective of this study was to examine the underlying mechanisms of immature ginger on adipocyte differentiation and lipogenesis in a 3T3-L1 cellular model. Methods: Ginger samples, extracted in 80% methanol, were dried and resuspended in DMSO at 50 µg/mL as stock solution. For analysis, the extracted samples were further diluted in media. Effects on adipogenesis were evaluated by determining lipid droplet and triglyceride accumulation, whereas effects on lipogenesis were determined by measuring triglyceride contents and fatty acid profile. The expression of key regulatory genes involved in adipogenesis and lipogenesis was also determined. Results: Our data indicate that the intracellular lipid accumulation decreased significantly by 15 or 25% on treatment with 25 or 50 µg/mL of ginger extract. Consistent with these data, significantly reduced triglyceride levels by 30 or 50% were observed on 25 or 50 µg/mL treatment with ginger extracts, respectively. In addition, ginger treatment significantly inhibited the differentiation-induced de novo lipogenesis and Δ9 desaturase activity. Furthermore, ginger treatment reduced adipogenesis genes, C/ebpβ and C/ebpδ, expression by 47 or 64%, respectively, but significantly increased Pparγ expression by 60% and adiponectin by 75%. Ginger extracts had no effect on Fas genes but reduced lipogenesis genes, acyl CoA carboxylase (Acc) expression by two-fold, and phosphoenolpyruvate carboxy kinase 1 (Pepck1) expression by 50%. Conclusion: Our findings suggest immature ginger can potentially inhibit lipogenesis pathways by limiting the channeling of glucose carbon in fatty acid synthesis by inhibiting the expression of ACC and glycerol production via inhibiting the expression of PEPCK, which consequently inhibits triglyceride formation. The ability of Yarrowia lipolytica to assimilate volatile fatty acids (VFAs) generated during the acidogenic fermentation of food waste (FW) and to accumulate intracellular lipids was investigated in single and two-stage batch (TSC) cultures. The TSC strategy, where the growth phase in FW-derived VFAs was followed by the lipogenic phase in crude glycerol resulted in a 10-fold improvement of intracellular lipids accumulation comparatively to the single batch culture. The biotechnological process proposed herein is a reliable and sustainable option for the combined valorization of food waste - via production of VFAs by acidogenesis - and crude glycerol. The integration of an anaerobic and aerobic process generates a value chain from wastes and by-products to microbial lipids production. Furthermore, microbial lipids produced by Y. lipolytica NCYC 2904 from FW-derived VFAs and crude glycerol have a composition similar to vegetable oils (C16–C18 fatty acids), making them suitable feedstocks for the biodiesel industry. Aims It is known that Thecaphora frezii produces peanut smut that generates numerous economic losses. For this reason, it is a priority to search for control strategies. In this sense, we investigated the lipid profile of this pathogen, as possible antifungal targets, regarding polar lipid composition, fatty acid profile, and transcriptional regulation of genes involved in each stage of the development. Method and Results Lipids from T. frezii teliospores, basidiospores, and hyphae were analyzed by HPLC/CAD and CG/FID. We found differences in the unsaturation levels as well as in the long-chain fatty acids along the stages. Phosphatidylcholine was the main component in the three development stages, followed by cardiolipins. Phosphatidylinositol, phosphatidylethanolamine, and lyso-phosphatidylethanolamine were found in similar amounts in all stages. Although ergosterol was not detected, we found two unsaponifiable lipids. In addition, we found transcripts that encode 28 enzymes involved in the biosynthesis of three lipids by RNA-Seq. Conclusions Thecaphora frezii shows changes in the composition of membrane lipids in different ontogenetic stages as well as in the expression of transcripts for enzymes involved in lipid biosynthesis. This review focuses on an understanding of lipid distribution and deposition dynamics in Atlantic salmon and explores the extent to which these can be predicted through multi-dimensional modeling. The methodology used to measure lipids can have an impact on their assessment and considerable work has been done to standardize and develop robust rapid assessment measures. The distribution of lipids in salmon fillets is spatially variable, with levels consistently highest in the belly region and lowest in the tail section. The level of lipids in the whole-body, fillet and Norwegian Quality Cut (NQC) of Atlantic salmon increases with fish size but plateaus as size increases. Lipid levels are predictable based on fish size data although there are various other factors that influence lipid deposition in the fillet. The relationship between these factors and the deposition of n-3 LC-PUFA is mathematically complex but does lend itself to multidimensional modeling. Effects are strongly influenced by dietary fatty acid composition and for the most part a "parity" exists between levels of certain fatty acids in the diet and what ends up in the body tissues. This parity results in a "dilution effect" that generally explains the relationship between diet and whole-body fatty acid levels. Some specific fatty acids demonstrate traits of selective retention in Atlantic salmon muscle tissue. Notably, docosahexaenoic acid is preferentially retained compared to other fatty acids. Which is explained by the understanding that not all fatty acids dilute at the same rate and biologically active fatty acids, like EPA, are essential for mediating inflammatory responses and have a critical role to play in biosynthesis. Industrial crops are becoming increasingly relevant as raw materials for different goods and services. Although there are many oilseed crops with industrial purposes, data about their production in Chile are scarce because the definition used by the Office of Agricultural Studies and Policies places agro‐industrial and industrial crops in the same category. Here, we review the available information about 12 industrial oleaginous crops at different levels of development (four native, two adventive and six introduced crops). Most of these crops have been evaluated for or are related to biodiesel production, despite the lack of commercial biodiesel production in Chile. Jojoba was the first industrial oilseed produced commercially in Chile; however, every year its production area decreases, and currently it does not exceed 100 ha. Sunflower and rapeseed, which are the first generation of raw materials for biodiesel production, have been poorly studied in Chile despite the large number of studies available on them worldwide. Camelina and castor bean have recently been studied; the former is produced in the south, while the latter is produced on the coastline in northern Chile. Further research is needed to improve the production and adaptation of these oilseed crops to climatic conditions in Chile; moreover, a new market must be opened to produce and commercialize their oils. Objectives: Kaempferol (KMF), has beneficial effects against hepatic lipid accumulation. In this study, we aimed to investigate molecular mechanism underlying the protective effect of KMF on lipid accumulation. Methods: HepG2 cells were treated with different concentrations of KMF and 0.5 mM palmitate (PA) for 24 h. The mRNA and protein levels of genes involved in lipid metabolism were evaluated using real-time PCR and western blot. The expression of Nrf2 was silenced using siRNA. Results: Data indicated that KMF (20 μM) reversed PA-induced increased triglyceride (TG) levels and total lipid content. These effects were accompanied by down-regulation of the mRNA and protein levels of lipogenic genes (FAS, ACC and SREBP1), and up-regulation of genes related to fatty acid oxidation (CPT-1, HADHα and PPARα). Kaempferol significantly decreased the levels of the oxidative stress markers (ROS and MDA) and enhanced the activities of antioxidant enzymes SOD and GPx in PA-challenged cells. Luciferase analysis showed that KMF increased the transactivation of Nrf2 in hepatocytes. The results also revealed that KMF-mediated activation of Nrf2 target genes was suppressed by Nrf2 siRNA. Furthermore, Nrf2 siRNA abolished the KMF-induced reduction in ROS and MDA levels in PA treated cells. In addition, the inhibitory effect of KMF on TG levels and the mRNA and protein levels of FAS, ACC and SREPB-1 were significantly abolished by Nrf2 inhibition. Nrf2 inhibition also suppressed the KMF-induced activation of genes involved in β oxidation (CPT-1 and PPAR-α). Background: Disordered lipid metabolism plays an essential role in both the initiation and progression of alcoholic fatty liver disease (AFLD), and fatty acid β-oxidation is increasingly considered as a crucial factor for controlling lipid metabolism. Hif-2α is a member of the Hif family of nuclear receptors, which take part in regulating hepatic fatty acid β-oxidation. However, its functional role in AFLD and the underlying mechanisms remain unclear. Results: Hif-2α was upregulated in EtOH-fed mice and EtOH-treated AML-12 cells. Inhibition or silencing of Hif-2α led to increased fatty acid β-oxidation and BNIP3-dependent mitophagy. Downregulation of Hif-2α activates the PPAR-α/PGC-1α signaling pathway, which is involved in hepatic fatty acid β-oxidation, by mediating BNIP3-dependent mitophagy, ultimately delaying the progression of AFLD. Conclusions: Hif-2α induces liver steatosis, which promotes the progression of AFLD. Here, we have described a novel Hif-2α-BNIP3-dependent mitophagy regulatory pathway interconnected with EtOH-induced lipid accumulation, which could be a potential therapeutic target for the prevention and treatment of AFLD. Simple Summary: The choice of lipid emulsions (LEs) used in parenteral nutrition (PN) is based on fatty acid composition and phytosterol content. Phytosterols are believed to be detrimental in patients receiving PN. Data from this observational study suggest that the adverse effect of phytosterols delivered to home PN patients is mitigated by long-chain omega-3 fatty acids. Background: the effect on liver function markers and inflammation of the different content of phytosterols in lipid emulsions (LEs) used in the parenteral nutrition (PN) regimen of adult home PN (HPN) patients is not clear. Methods: plasma phytosterol and cytokine concentrations, fatty acid composition, liver function markers, and triglycerides were measured in 58 adult HPN patients receiving one of three different LEs (soybean oil-based: Intralipid; olive oil-based: ClinOleic; containing fish oil: SMOFLipid). Results: patients receiving Intralipid had higher plasma campesterol and stigmasterol concentrations than those receiving ClinOleic or SMOFLipid. Plasma sterol concentrations were not different between patients receiving ClinOleic and SMOFLipid. Differences in plasma fatty acids reflected the fatty acid composition of the LEs. Markers of liver function did not differ among the three groups. Blood triglycerides were higher with ClinOleic than with Intralipid or SMOFLipid. Total bilirubin correlated positively with the plasma concentrations of two of the phytosterols, ALT correlated positively with one, AST with one, and GGT with three. Conclusions: liver function markers correlate with plasma plant sterol concentrations in adult HPN patients. Adult HPN patients receiving SMOFLipid are more likely to have liver function markers and triglycerides within the normal range than those receiving ClinOleic or Intralipid. We evaluated whether linseed oil (LO) modulates the effects of a high-carbohydrate diet (HCD) on liver inflammation, fatty acid (FA) accumulation, and lipid distribution in periportal and perivenous hepatocytes. The control group (control high-carbohydrate diet [HCD-C]) received an HCD with lard and soybean oil as the lipid source. The L10 and L100 groups received the HCD with 10% and 100% of LO as the lipid source, respectively. The animals were killed by decapitation before (day 0) and after receiving the diets. Liver FA composition, inflammation, and fibrogenesis gene expression were evaluated. Also, the percentage of lipid-occupied area in periportal end perivenous hepatocytes were measured. The L100 group exhibited a higher (P < .05) liver amount of omega-3 polyunsaturated FA (n-3 PUFA) and lower (P < .05) amounts of saturated FA (SFA), monounsaturated FA (MUFA), and omega-6 polyunsaturated FA (n-6 PUFA) compared with L10 or HCD-C mice. On day 56, interleukin 10 and type IV collagen gene expression were significantly upregulated and downregulated, respectively in L100. Also, the L100 group showed lower (P < .05) FA accumulation (i.e., total FA, SFA, MUFA, and n-6 PUFA). Also, L10 and L100 presented lower (P < .05) percentage of high lipid-containing portion in periportal and perivenous hepatocytes. We concluded that LO attenuation of liver inflammation promoted by an HCD is associated with increased liver n-3 PUFA levels, so modulating FA composition, deposition, and distribution in periportal and perivenous hepatocytes. Cardiovascular diseases (CVDs) are a major cause of death and morbidity in many parts of the world, and many dietary guidelines limit the intake of saturated fatty acids (SFA) as they are regarded as an important risk factor for CVDs due to their association with increased blood cholesterol. Dairy foods are often a major contributor to dietary intake of SFA, and since many dietary guidelines contain restrictions on SFA intake, this can lead to a moderation of dairy food intake despite meta‐analyses generally showing dairy to have a neutral or negative association with CVDs. Many prospective studies and randomised controlled trials do not support a simple positive association between SFA intake and the risk of atherosclerotic CVD and its components although some early studies had a number of methodological weakness. Studies that included blood cholesterol data do broadly support the positive relationship between SFA and blood low‐density lipoprotein cholesterol (LDL‐C) but without increased CVD risk resulting, despite LDL being a causal factor in atherosclerotic CVD. These data suggest that LDL‐C alone is not a consistently good predictor or cause of CVD risk, perhaps particularly in relation to dairy food consumption although some non‐dairy food studies have also shown LDL‐C reduction was not reflected in reduced CVD risk. This narrative review examines some reasons for these findings. Overall, restrictions on dairy food intake do not seem warranted, although there remains a need to further understand the association of different dairy food types with chronic diseases, perhaps particularly for type 2 diabetes. Herein, we report a gram‐scale synthesis of perdeuterated linoleic acid‐d31. The starting materials for the synthesis are two saturated fatty acids, azelaic acid‐d14 and pentanoic acid‐d9, which can be obtained by metal catalysed hydrothermal hydrogen‐deuterium exchange. The synthesis utilises the fatty acids directly via decarboxylative coupling. Copper catalysed coupling of a terminal alkyne intermediate with a propargyl bromide derivative affords a skipped diyne, which can be reduced using P‐2 nickel to obtain the desired cis,cis‐diene geometry. The subsequent synthesis of the tail‐deuterated phospholipid, 1‐palmitoyl‐2‐linoleoyl‐sn‐glycero‐3‐phosphocholine‐d62 (PLPC‐d62) is also described. Lipids, defined by low solubility in water and high solubility in nonpolar solvents, can be classified into fatty acids, glycerolipids, glycerophospholipids, sphingolipids, and sterols. Lipids not only regulate integrity and fluidity of biological membranes, but also serve as energy storage and bioactive molecules for signaling. Causal mutations in SPTLC1 (serine palmitoyltransferase long chain subunit 1) gene within the lipogenic pathway have been identified in amyotrophic lateral sclerosis (ALS), a paralytic and fatal motor neuron disease. Furthermore, lipid dysmetabolism within the central nervous system and circulation is associated with ALS. Here, we aim to delineate the diverse roles of different lipid classes and understand how lipid dysmetabolism may contribute to ALS pathogenesis. Among the different lipids, accumulation of ceramides, arachidonic acid, and lysophosphatidylcholine is commonly emerging as detrimental to motor neurons. The present study was to characterize the lipid profile of five black soybean (BS) produced in Indo‐Himalayan regions. Among seven fatty acids (FAs), methyl linoleate and cis‐9‐oleic acid methyl ester were maximum. Unsaturated/saturated fatty acid ratio was highest (6.10%) in BSALB (big spherical shape seed). Unsaponifiable matters and tocopherols were also higher (3.09% and 1305 μg/kg, respectively) in BSALB, while β‐carotene was improved in BSALS (small oval‐shaped seed). Flavor analysis reported 46 compounds in the oil, which were maximum in BSALS. There was significant difference (p < 0.02) in the flavor components. Crop cycle temperature is inversely correlated with major lipid constituents. The results suggest that the BS seeds produced comparatively in high‐altitude regions retain the maximum lipid antioxidants and flavor compounds. Penaeus vannamei were reared at six different water salinities (3, 10, 20, 30, 40 and 50‰) for 45 days to explore its effect on lipid class and fatty acid composition. The results revealed that the total lipid content was high (p < 0.05) at 30‰ (3.14%), and low (p < 0.05) at 20‰ (2.15%). Both phospholipids and cholesterol were found to be high (p < 0.05) at 40‰ (1.30% and 0.38%, respectively), while it was at 30‰ for triacylglycerides (0.71%). A higher correlation coefficient was observed for phospholipids (R2 = 0.5975) with rearing salinity and triacylglycerides (R2 = 0.8487) with total lipid. A total of saturated fatty acids (SFA), mono unsaturated (MUFA) and polyunsaturated (PUFA) fatty acids was increased in all treatments compared to ambient salinity (20‰), and the increase was quite high with 30‰ (38.83,% 37.07% and 48.76%, respectively), whereas it was 17.47%–24.94% in SFA, 7.85%–26.05% in MUFA and 20.69%–33.82% in PUFA at hypo (3–10‰) and hyper (40–50‰) saline conditions. The correlation coefficient determinant was high for MUFA (R2 = 0.8682) followed by SFA (R2 = 0.7370) and PUFA (R2 = 0.6542) against total lipid, whereas it was much less (R2 = 0.0402–0.0588) when performed against water salinity. The daily value of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) was >100%, and was ranged between 2.81% and 3.24% for α‐linolenic acid (ALA). The results concluded that the rearing salinity had a significant effect on lipid class and fatty acid composition in P. vannamei and their deposition was quite high in the group reared at 30‰. Background: Surgical wound excision is a necessary procedure for burn patients that require the removal of eschar. The extent of excision is currently guided by clinical judgement, with excessinto healthy tissue potentially leading to excessive scar, or inadequate debridement increasing risk of infection. Thus, an objective real-time measure to facilitate accurate excision could support clinical judgement and improve this surgical procedure. This study was designed to investigate the potential use of Rapid evaporative ionisation mass spectrometry (REIMS) as a tool to support data-driven objective tissue excision.Methods: Data were acquired using a multi-platform approach that consisted of both Rapid Evaporative Ionisation Mass Spectrometry (REIMS) performed on intact skin, and comprehensive liquid chromatography-mass spectrometry (LC-MS/MS) lipidomics performed on homogenised skin tissue extracts. Data were analysed using principal components analysis (PCA) and multivariate orthogonal projections to latent squares discriminant analysis (OPLS-DA) and logistic regression to determine the predictability of the models.Results: PCA and OPLS-DA models of the REIMS and LC-MS/MS lipidomics data reported separation of excised and healthy tissue. Molecular fingerprints generated from REIMS analysis of healthy skin tissue revealed a high degree of heterogeneity, however, intra-individual variance was smaller than inter-individual variance. Both platforms indicated high levels of skin classification accuracy. In addition, OPLS-DA of the LC-MS/MS lipidomic data revealed significant differences in specific lipid classes between healthy control and excised skin samples; including lower free fatty acids (FFA), monoacylglycerols (MAG), lysophosphatidylglycerol (LPG) and lysophosphatidylethanolamines (LPE) in excised tissue and higher lactosylceramides (LCER) and cholesterol esters (CE) compared to healthy control tissue.Conclusions: Having established the heterogeneity in the biochemical composition of healthy skin using REIMS and LC-MS/MS, our data show that REIMS has the potential to distinguish between excied and healthy skin tissue samples. Supplementation of omega-3 fatty acids is considered a valuable strategy to supply the low intake of these fatty acids. Thus, the safety of the supplements is an important milestone. Because of that, we analyzed 20 unflavored supplements sold in the French market for fatty acid and triglyceride composition, for EPA and DHA, and for tocol content, as well as for oxidative status. This study found that only 2.5% of the supplements did not meet their label claims for omega-3 content. TAG analysis showed high variability among the triglyceride distribution, and the same trend was also noticed for the tocol content; in fact, a high variability of the distribution of the six tocols (four tocopherols and two tocotrienols) was found among the samples. Of the tested products, all of them complied with peroxide value, p-anisidine value, and Totox values established by the Global Organization for EPA and DHA Omega-3s (GOED) and were not oxidized. The following study determined whether the effects of the combined addition of zinc amino acid complex (ZA) and selenomethionine (SM) was superior to their single addition in controlling the oxidative stress induced by dietary oxidised fat in laying hens. 2. Two hundred and forty 32-week-old laying hens were divided into the following dietary treatments (each consisting of six replicates of eight birds): 1) a fresh soy oil (FSO) diet; 2) an oxidised soy oil (OSO) diet; 3) an OSO diet plus 20 mg zinc as ZA/kg (OSO+ZA); 4) an OSO diet plus 0.2 mg selenium as SM/kg (OSO+SM); and 5) an OSO diet plus ZA and SM (OSO+ZA+SM). 3. After 10 weeks of feeding hens, feed intake, egg production, and egg mass in the OSO+ZA+SM group were similar to the FSO group but better (P < 0.05) than those in the OSO group. Shell thickness and shell breaking strength were significantly improved by the OSO+ZA and OSO+ZA+SM treatments. 4. Increases in the yolk concentrations of palmitic acid and total saturated fatty acids (SFA), and decreases in yolk linoleic acid, n-6 polyunsaturated fatty acids (PUFA), total PUFA, and PUFA/SFA ratio were induced by dietary oxidised fat which were normalised (P < 0.05) by OSO+SM and OSO+ZA+SM. 5. An increase (P < 0.05) in malondialdehyde and a decrease in 2,2-diphenyl-picrylhydrazyl radical scavenging activity in the yolk, induced by dietary oxidised fat, was significantly improved by all dietary supplementations, but only birds fed the OSO+ZA+SM diet exhibited similar values to those fed FSO. 6. In conclusion, the simultaneous inclusion of organic zinc plus selenium in the oxidised fat diets was beneficial for improving egg-laying performance, yolk fatty acid profile, and oxidative stability, but not for internal egg quality, compared with either zinc or selenium alone in laying hens. Summary: Provocative studies have proposed that the common 9 carbon lipid oxidation product, 4‐hydroxy‐2‐nonenal (HNE), increases myoglobin (Mb) autoxidation. Oxidised Mb (Fe3+ Mb or metMb) formation leads to an unappealing meat discoloration. Since Mb also facilitates lipid oxidation, an HNE induced increase in Mb autoxidation would constitute a positive feedback loop. However, NMR studies have noted that only fatty acids with carbon chain length exceeding 10 can significantly bind to Mb. They predict that HNE should not interact with Mb. The present study has used optical and NMR spectroscopy methods to determine if HNE interacts with Mb and if that interaction leads to an increased oxidation of Mb. The study finds no convincing evidence that HNE interacts with Mb and accelerates Mb autoxidation or oxidation by electron transfer. To assess the lipid requirements of O. macrolepis broodstock, five iso-nitrogenous diets (39 g kg−1) with five lipid levels, 50 (5 L), 70 (7 L), 90 (9 L),110 (11 L), and 130 (13 L; g kg−1), were made. A total of 105 three-year-old individuals (50.11 ± 2.86 g per fish) were divided into five groups (triplicate per group) and were fed with the diets, respectively, for eight weeks. Then, the fish were sampled, and items were determined. The results showed that growth rate and feed efficiency ratio were not significantly affected by diets (p > 0.05). A clear dose–response effect of dietary lipid was observed on somatic indexes of gonad indexes of the O. macrolepis brookstock, with the highest values corresponding to fish fed 9 and 11 g kg−1 lipids, in contrast, gonad indexes were reduced as dietary lipid moved away from this level. The other somatic indexes, such as viscerosomatic index, perivisceral fat index, etc., were not significantly affected by diets (p > 0.05). The content of crude lipid and crude protein in carcass, hepatopancreas, and gonad were not significantly affected by dietary lipid levels (p > 0.05). The gonad fatty acids of 16:0 and 22:6n-3 decreased and 18:2n-6 increased with the increasing lipid level, being significantly altered by diets (p < 0.05). The histological features of the gonad showed no significant difference among the five diets (p > 0.05). The relative expression of sex steroid-synthesizing proteins (fshr, 3β-hsd, 17β-hsd, aro., and star.) in the gonad of fish was most significantly highly expressed in the 9 L and 11 L groups (p < 0.05). Time-of-flight secondary ion mass spectrometry (TOF-SIMS) with the Bi 3 + liquid metal ion gun was used to investigate the content of lipids and amino acids (AAs) in extracellular vesicles (EVs). We induced metabolic changes in human pancreatic β -cells by stimulation with high glucose concentrations (35 mM) and tested the hypothesis of hyperglycemia (HG) has a detrimental effect on lipids and AAs in released EV subpopulations: ectosomes and exosomes. As a result of HG treatment, selected fatty acids (FAs) such as arachidonic, myristic and palmitic acids, changed their abundance in ectosomes and exosomes. Also, intensities of the characteristic peaks for cholesterol (m/z 95.09; 147.07; 161.11; 369.45) along with the molecular ion m/z 386.37 [C 27 H 46 O+] under HG conditions, both for ectosomes and exosomes, have changed significantly. Comparative analysis of HG EVs and normoglycemic (NG) ones showed statistically significant differences in the signal intensities of four AAs: valine (m/z 72.08 and 83.05), isoleucine (m/z 86.10), phenylalanine (m/z 120.08 and 132.05) and tyrosine (m/z 107.05 and 136.09). We confirmed that ToF-SIMS is a useful technique to study selected AAs and lipid profiles in various EV subpopulations. Our study is the first demonstration of changes in FAs and AAs in exosomes and ectosomes derived from β -cells under the influence of HG. To allow the study of unsaturated free fatty acids in live cells, we here report the use of sterculic acid, a 1,2‐cyclopropene‐containing oleic acid analogue, as a bioorthogonal probe. We show that this lipid can be readily taken up by dendritic cells without toxic side effects, and that it can subsequently be visualised using an inverse electron‐demand Diels–Alder reaction with quenched tetrazine‐fluorophore conjugates. In addition, the lipid can be used to identify changes in protein oleoylation after immune cell activation. Finally, this reaction can be integrated into a multiplexed bioorthogonal reaction workflow by combining it with two sequential copper‐catalysed Huisgen ligation reactions. Fragile X Syndrome (FXS) is the most prevalent monogenic cause of Autism Spectrum Disorders (ASDs). Despite a common genetic etiology, the affected individuals display heterogenous metabolic abnormalities including hypocholesterolemia. Although changes in the metabolism of fatty acids (FAs) have been reported in various neuropsychiatric disorders, it has not been explored in humans with FXS. In this study, we investigated the FA profiles of two different groups: (1) an Argentinian group, including FXS individuals and age- and sex-matched controls, and (2) a French-Canadian group, including FXS individuals and their age- and sex-matched controls. Since phospholipid FAs are an indicator of medium-term diet and endogenous metabolism, we quantified the FA profile in plasma phospholipids using gas chromatography. Our results showed significantly lower levels in various plasma FAs including saturated, monosaturated, ω-6 polyunsaturated, and ω-3 polyunsaturated FAs in FXS individuals compared to the controls. A decrease in the EPA/ALA (eicosapentaenoic acid/alpha linoleic acid) ratio and an increase in the DPA/EPA (docosapentaenoic acid/eicosapentaenoic acid) ratio suggest an alteration associated with desaturase and elongase activity, respectively. We conclude that FXS individuals present an abnormal profile of FAs, specifically FAs belonging to the ω-3 family, that might open new avenues of treatment to improve core symptoms of the disorder. Background: Cardiovascular mortality in dialysis population remains very high. Saturated fatty acids (SFA) contribute to atherosclerosis and to cardiovascular risk. Aim: The aim of this study was to evaluate the relationship between mortality in dialysis patients and the serum SFA content. Methods: Survival of 54 patients on dialysis was assessed. A total of 21 SFA from patients' sera were measured by gas chromatography-mass spectrometry (GC-MS). Diet was assessed by food frequency questionnaire FFQ-6. The SFA content is presented as fatty acid proportion (%). Results: During the observation time (median 66 months) 22 patients died. There was a significant relationship between elevated SFA (above SFA mean) and mortality (log-rank 3.13; p = 0.0017). Moreover, patients who ingested foods rich in SFA, according to FFQ-6, had a higher mortality risk (log-rank 2.24; p = 0.03). The hazard ratio for mortality associated with increased SFA content equalled 4.47 (1.63–12.26). Addition of age and inflammation (hsCRP > 5 mg/L) into the Cox model did not modify this relationship. However, SFA content turned out to be significantly higher in patients with diabetes mellitus and cardiovascular disease, as compared to patients free from these co-morbidities. Their addition to the model attenuated the relationship between SFA and mortality, making it statistically insignificant. Conclusion: The serum content of SFA turned out to be a strong predictor of mortality in dialysis patients. However, given the significant associations between SFA, DM, and CVD, interventional studies with controlled SFA intake are needed to evaluate the causal links between SFA, co-morbidities and survival. Objective: Clinical heterogeneity in major depressive disorder likely reflects the range of etiology and contributing factors in the disorder, such as genetic risk. Identification of more refined subgroups based on biomarkers such as white matter integrity and lipid-related metabolites could facilitate precision medicine in major depressive disorder. Methods: A total of 148 participants (15 genetic high-risk participants, 57 patients with first-episode major depressive disorder and 76 healthy controls) underwent diffusion tensor imaging and plasma lipid profiling. Alterations in white matter integrity and lipid metabolites were identified in genetic high-risk participants and patients with first-episode major depressive disorder. Then, shared alterations between genetic high-risk and first-episode major depressive disorder were used to develop an imaging x metabolite diagnostic panel for genetically based major depressive disorder via factor analysis and logistic regression. A fivefold cross-validation test was performed to evaluate the diagnostic panel. Results: Alterations of white matter integrity in corona radiata, superior longitudinal fasciculus and the body of corpus callosum and dysregulated unsaturated fatty acid metabolism were identified in both genetic high-risk participants and patients with first-episode major depressive disorder. An imaging x metabolite diagnostic panel, consisting of measures for white matter integrity and unsaturated fatty acid metabolism, was identified that achieved an area under the receiver operating characteristic curve of 0.86 and had a significantly higher diagnostic performance than that using either measure alone. And cross-validation confirmed the adequate reliability and accuracy of the diagnostic panel. Conclusion: Combining white matter integrity in corpus callosum, superior longitudinal fasciculus and corona radiata, and unsaturated fatty acid profile may improve the identification of genetically based endophenotypes in major depressive disorder to advance precision medicine strategies. Hidradenitis suppurativa (HS) is an inflammatory skin condition that is often considered a systemic disease due to its association with metabolic comorbidity. In this study, we aimed to identify differences in plasma lipidomic profiles between HS patients and control subjects. HS patients were recruited from a tertiary dermatological centre and demographic and comorbidity matched controls from the general population. A targeted lipidomic approach was performed to characterize over 700 lipid species representing 35 lipid classes/sub-classes. Linear regression models adjusted for confounding factors were used to compare the plasma lipidomic profiles of HS patients to controls. Ordinal regression models were used to study the association of lipids with disease activity and severity scores. 60 HS patients and 73 control subjects were recruited. Differential levels (p < 0.05) of 32 lipid species in HS patients compared to controls were observed, including a decrease in the long chain base d19:1 containing ceramides, and elevation of hydroxyeicosatetraenoic acid (HETE) and dihydroxyeicosatrienoic acid (DHET) oxylipins. These lipids along with several other molecules showed associations with Hurley, HS-PGA and disease activity scores. This study found mild changes in plasma lipidomic profiles, consistent with previous studies showing attenuated metabolomic changes in plasma as opposed to lesional skin. However, a number of lipid species were associated with increasing activity and severity of the disease. Background: Altered lipid profiles are frequently present in cancer, and it is necessary to elucidate the role of changed lipid profiles in hepatocellular carcinoma (HCC). We conducted this study to investigate the changed lipid profile in HCC tissues and discover some remarkably changed lipid components, and to explore the function of changed lipid components in HCC development. Methods: Gas chromatography/mass spectrometer (GC/MS analysis) was employed to measure the abundance of fatty acids between HCC tissues and adjacent noncancerous tissues. The proliferative ability of HCC cells was determined by Cell Counting Kit-8 and EdU assays. Transwell and wound healing assays were employed to determine the migratory ability of HCC cells. Protein expression was assessed by western blot assay. Results: GC/MS analysis revealed that alpha-linolenic acid was present at lower levels in HCC tissues than that in the adjacent noncancerous tissues. Alpha-linolenic acid inhibited the proliferation, migration and invasion of HCC cells in vitro. Western blotting showed that alpha-linolenic acid treatment increased Farnesoid X receptor expression and decreased β-catenin and cyclinD1 expression. Conclusions: Alpha-linolenic acid suppresses HCC progression through the FXR/Wnt/β-catenin signaling pathway.
