Abstract Accurate assessment of the mechanical properties of organic matter, clay matrix, and bulk shale during maturation remains a challenge. Here, we aim to assess the mechanical properties of organic-rich shale during maturation using a combination of nanoindentation methods and various geochemical analyses, i.e., mineral composition, mass loss rate, chemical structure of organic matter, and Rock-Eval analyses. Results show that the evolution of mechanical properties of organic matter in shale during maturation can be divided into: the main oil-generation stage, and the condensate oil and gas generation stage. The stiffening of organic matter in the shale is mainly due to increased aromaticity and condensation of aromatic groups. The clay matrix experiences a slight decrease in hardness and Young’s modulus at low maturity levels due to the generation of liquid hydrocarbons. However, overall, the clay matrix becomes stiffer as the shale matures due to shale dehydration, expulsion or cracking of liquid hydrocarbons, transformation of clay minerals, and hardening of organic matter. The Young’s modulus and hardness of bulk shale generally increase with increasing maturity. This is closely related to the hardening of organic matter and clay matrix, as well as the development of the more compact and dense microstructure in the shale.

Introduction Mild cognitive impairment (MCI) is an intermediate phase between normal cognitive ageing and dementia and poses a serious threat to public health worldwide; however, it might be reversible, representing the best opportunity for secondary prevention against serious cognitive impairment. As a non-pharmacological intervention for those patients, interventions that combine physical exercise and cognitive training, whether delivered simultaneously or sequentially, may have superior effects on various cognitive domains, including global cognition, memory, executive function and attention. The supportive evidence remains incomplete. This study aims to assess the effectiveness of a combined exercise and cognitive intervention in Chinese older adults with mild cognitive impairment (COGITO), empowered by digital therapy and guided by the Health Action Process Model and the Theory of Planned Behaviour (HAPA-TPB theory) in a home-based setting.Methods and analysis This study is a randomised controlled, assessor-blinded multi-centre study. Four parallel groups will include a total of 160 patients, receiving either a combined exercise and cognitive intervention, an isolated exercise intervention, an isolated cognitive intervention or only health education. These interventions will be conducted at least twice a week for 50 min each session, over 3 months. All interventions will be delivered at home and remotely monitored through RehabApp and Mini-programme, along with an arm-worn heart rate telemetry device. Specifically, supervisors will receive participants’ real-time training diaries, heart rates or other online monitoring data and then provide weekly telephone calls and monthly home visits to encourage participants to complete their tasks and address any difficulties based on their training information. Eligible participants are community-dwelling patients with no regular exercise habit and diagnosed with MCI. The primary outcome is cognitive function assessed by the Alzheimer’s Disease Assessment Scale-Cognitive (ADAS-Cog) and Community Screening Instrument for Dementia (CSI-D), with baseline and three follow-up assessments. Secondary outcomes include quality of life, physical fitness, sleep quality, intrinsic capacity, frailty, social support, adherence, cost-effectiveness and cost-benefit.Ethics and dissemination The study was approved by the Institutional Review Board of Peking University. Research findings will be presented to stakeholders and published in peer-reviewed journals and at provincial, national and international conferences.Trial registration number ChiCTR2300073900.

Abstract Background Rectal neuroendocrine neoplasms (NENs) are rare neoplasms with limited understanding of its genomic alterations and molecular typing. Methods The paraffin‐embedded tissue specimens of 38 patients with rectal NENs after surgery were subjected to whole gene sequencing (WGS), and mutation profilings were drawn to identify high‐frequency mutation genes, copy‐number variations (CNVs), tumor mutation burden (TMB), signal pathways, mutation signatures, DNA damage repair (DDR) genes, and molecular types. The differences of mutated genes and signaling pathways in different pathological grades and metastatic/non‐metastatic groups were compared. It helped to search for potential targets. Results C > T and T > C transitions are the most common base substitutions in rectal NENs. DNA mismatch repair deficiency, DNA base modifications, smoking and exposure to ultraviolet light might play a role in the occurrence of rectal NENs. DAXX, KMT2C, BCL2L1, LTK, MERTK, SPEN, PKN1, FAT3, and LRP2 mutations were found in only low‐grade rectal NETs, whereas APC, TP53, NF1, SOX9, and BRCA1 mutations were common in high‐grade rectal NECs/MiNENs. These genes helped in distinguishing poorly‐differentiated or well‐differentiated rectal NENs. Alterations in P53, Wnt and TGFβ signaling pathways were more pronounced in rectal NECs and MiNENs. Alterations in Wnt, MAPK and PI3K/AKT signaling pathways promoted metastases. Rectal NENs were classified into two molecular subtypes by cluster analysis based on the mutant genes and signaling pathways combined with clinicopathological features. Patients with mutations in the LRP2, DAXX, and PKN1 gene showed a trend of well‐differentiated and early‐stage tumors with less metastasis (p = 0.000). Conclusions This study evaluated risk factors for regional lymphatic and/or distant metastases, identified high‐frequency mutated genes, mutation signatures, altered signaling pathways through NGS. Rectal NENs were divided into two molecular types. This helps to evaluate the likelihood of metastasis, formulate follow‐up strategies for patients and provide a target for future research on precision treatment of rectal NENs. PARP inhibitors, MEK inhibitors, mTOR/AKT/PI3K and Wnt signaling pathway inhibitors may be effective drugs for the treatment of metastatic rectal NENs.

The gum acacia Senegalia senegal (L.) Britton (Fabales: Fabaceae) is a drought-tolerant plant belonging to the genus Acacia of the Leguminosae family, possessing significant economic and ecological value. Despite its importance, there is a knowledge gap regarding the potential impact of climate change on the distribution of S. senegal, crucial for the conservation of plant resources and optimizing its use in introductory silviculture. In this study, we selected 23 environmental variables and utilized the optimized maximum entropy (MaxEnt) model to analyze the key environmental factors affecting the distribution of S. senegal worldwide and simulate the current and future distribution range of S. senegal in Pakistan under the SSP1-2.6, SSP2-4.5, SSP3-7.0, and SSP5-8.5 climate change scenarios. The results highlight key environmental factors influencing the distribution of S. senegal, including BIO3 (isothermally), BIO4 (coefficient of seasonal variation of temperature), BIO11 (mean temperature of the coldest season), and BIO12 (annual precipitation). Regions with higher and less fluctuating temperatures exhibit a higher potential for S. senegal distribution. Currently, suitable habitats of S. senegal are concentrated in the southern region of Pakistan, covering provinces such as Punjab, Sindh, and Balochistan, with highly suitable habitats accounting for 6.06% of the total area. Under the current climatic conditions, this study identifies the spatial patterns of suitable habitats and their concentration in specific regions. With climate change, a notable expansion of suitable habitats towards higher latitudes is observed, with the most significant expansion under the extremely severe climate change scenario (SSP5-8.5), reaching 223.45% of the current level. The results of this study enhance our understanding of the dynamics of S. senegal distribution under climate change and offer valuable insights into the long-term introduction of S. senegal for afforestation and soil conservation in Pakistan. This study provides theoretical support for the sustainable development of the local ecosystem and socio-economy, emphasizing the importance of proactive measures to adapt to changing climatic conditions.

Abstract Background MicroRNAs (miRNAs) and other epigenetic modifications play fundamental roles in all eukaryotic biological processes. DNA damage repair is a key process for maintaining the genomic integrity of different organisms exposed to diverse stresses. However, the reaction of miRNAs in the DNA damage repair process is unclear. Results In this study, we found that the simultaneous mutation of zinc finger DNA 3′-phosphoesterase (ZDP) and AP endonuclease 2 (APE2), two genes that play overlapping roles in active DNA demethylation and base excision repair (BER), led to genome-wide alteration of miRNAs. The transcripts of newly transcribed miRNA-encoding genes (MIRs) decreased significantly in zdp/ape2, indicating that the mutation of ZDP and APE2 affected the accumulation of miRNAs at the transcriptional level. In addition, the introduction of base damage with the DNA-alkylating reagent methyl methanesulfonate (MMS) accelerated the reduction of miRNAs in zdp/ape2. Further mutation of FORMAMIDOPYRIMIDINE DNA GLYCOSYLASE (FPG), a bifunctional DNA glycosylase/lyase, rescued the accumulation of miRNAs in zdp/ape2, suggesting that the accumulation of DNA damage repair intermediates induced the transcriptional repression of miRNAs. Conclusions Our investigation indicates that the accumulation of DNA damage repair intermediates inhibit miRNAs accumulation by inhibiting MIR transcriptions.

Background: Programmed cell death protein-1 inhibitors combined with lenvatinib have become a popular treatment option for patients with unresectable hepatocellular carcinoma. Transarterial chemoembolization combined with programmed cell death protein-1 inhibitors and lenvatinib has also shown preliminary efficacy in the unresectable hepatocellular carcinoma. We conducted this observational, retrospective, cohort study to compare the clinical outcomes and safety of transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib versus programmed cell death protein-1 inhibitors plus lenvatinib in patients with unresectable hepatocellular carcinoma. Methods: Between November 2019 and November 2021, patients who were diagnosed with unresectable hepatocellular carcinoma and received transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib or programmed cell death protein-1 inhibitors plus lenvatinib treatment were reviewed for eligibility. The primary endpoints included objective response rate, overall survival, and progression-free survival. The secondary endpoint was the frequency of key adverse events. Results: In total, 105 patients were eligible for the present study, and they were divided into the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group (n = 46) and the programmed cell death protein-1 inhibitors plus lenvatinib group (n = 59). The patient cohort after a one-to-one propensity score matching (n = 86) was also analyzed. The transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group had a higher objective response rate both in the patient cohort before propensity score matching (54.3% vs 25.4%, P = .002) and after propensity score matching (55.8% vs 30.2%, P = .017). The patients in the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group had prolonged overall survival (median, 20.5 vs 12.6 months, P = .015) and progression-free survival (median, 10.2 vs 7.4 months, P = .035). For patient cohort- propensity score matching, the overall survival (20.5 vs 12.8 months, P = .013) and progression-free survival (12.1 vs 7.8 months, P = .030) were also significantly better in the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group than in the programmed cell death protein-1 inhibitors plus lenvatinib group. There were no significant differences between the 2 groups concerning adverse reactions caused by immunotherapy and lenvatinib. The adverse reactions caused by transarterial chemoembolization were transient and were quickly reversed. Conclusions: Compared to programmed cell death protein-1 inhibitors plus lenvatinib, transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib may provide better treatment response and survival benefits for patients with unresectable hepatocellular carcinoma, and the adverse events were manageable.

Background: Elevated level of interleukin-8 (IL-8) promotes hepatocellular carcinoma (HCC) development and contributes to poor prognosis. Previously, we have proved that Dicer inhibits HCC progression. In this study, we evaluated the potential interaction between IL-8 and Dicer as well as their influence on HCC. Methods: Hepatocellular carcinoma cells of SMMC-7721 were divided into 2 groups for subsequent analysis: pCMV-Dicer group for Dicer-overexpressing lentivirus transfected cells (pCMV-Dicer cells) and pCMV-NC group for empty lentivirus transfected cells (pCMV-NC cells). Cell Counting kit-8 (CCK8), wound healing, and transwell were used to evaluate the inhibitory effect of Dicer overexpression on proliferation, migration, and invasion of HCC cells. The level of IL-8 was measured by flow cytometry bead-based immunoassays. Male nude BALB/c mice injected with pCMV-Dicer or pCMV-NC cell suspensions was used for transplant of HCC tumor. Results: We found that the secretion of IL-8 was reduced in the medium of pCMV-Dicer cells ( P = .027). Recombinant human IL-8 (rhIL-8) reversed the inhibitory effect of Dicer on proliferation ( P < .01), migration ( P = .003), and invasion ( P = .001), whereas IL-8 inhibitor of reparixin enhanced inhibitory effect of Dicer on proliferation ( P < .05), migration ( P = .008), and invasion ( P = .000). Lenvatinib downregulated the IL-8 level of HCC cells ( P = .000) as well as promote Dicer-induced inhibition for HCC cells referring to proliferation ( P < .05), migration ( P = .000), and invasion ( P = .000). Animal experiments also demonstrated that Dicer cooperated with lenvatinib to inhibit the growth of HCC tumors ( P < .05). Conclusions: Dicer cooperated with lenvatinib to inhibit HCC growth via downregulating IL-8, and Dicer displayed its potential capability to enhance the anti-tumor effect of lenvatinib.

Heavy metal pollution in soils caused by mining has led to major environmental problems around the globe and seriously threatens the ecological environment. The assessment of heavy metal pollution and the local phytoremediation potential of contaminated sites is an important prerequisite for phytoremediation. Therefore, the purpose of this study was to understand the characteristics of heavy metal pollution around a copper-nickel mine tailings pond and screen local plant species that could be potentially suitable for phytoremediation. The results showed that Cd, Cu, Ni, and Cr in the soil around the tailings pond were at the heavy pollution level, Mn and Pb pollution was moderate, and Zn and As pollution was light; The positive matrix factorization (PMF) model results showed that the contributions made by industrial pollution to Cu and Ni were 62.5% and 66.5%, respectively, atmospheric sedimentation and agricultural pollution contributions to Cr and Cd were 44.6% and 42.8%, respectively, the traffic pollution contribution to Pb was 41.2%, and the contributions made by natural pollution sources to Mn, Zn, and As were 54.5%, 47.9%, and 40.0% respectively. The maximum accumulation values for Cu, Ni, Cr, Cd, and As in 10 plants were 53.77, 102.67, 91.10, 1.16 and 7.23 mg/kg, respectively, which exceeded the normal content of heavy metals in plants. Ammophila breviligulata Fernald had the highest comprehensive extraction coefficient (CEI) and comprehensive stability coefficient (CSI) at 0.81 and 0.83, respectively. These results indicate that the heavy metal pollution in the soil around the copper nickel mine tailings pond investigated in this study is serious and may affect the normal growth of plants. Ammophila breviligulata Fernald has a strong comprehensive remediation capacity and can be used as a remediation plant species for multiple metal compound pollution sites.

N6-methyladenosine (m6A) modification plays a critical role in mammalian development. However, the role of m6A in the skeletal muscle development remains largely unknown. Here, we report a global m6A modification pattern of goat skeletal muscle at two key development stages and identified that the m6A modification regulated the expression of the growth arrest and DNA damage-inducible 45B (GADD45B) gene, which is involved in myogenic differentiation. We showed that GADD45B expression increased during myoblast differentiation, whereas the downregulation of GADD45B inhibits myogenic differentiation and mitochondrial biogenesis. Moreover, the expression of GADD45B regulates the expression of myogenic regulatory factors and peroxisome proliferator-activated receptor gamma coactivator 1 alpha by activating the p38 mitogen-activated protein kinase (MAPK) pathway. Conversely, the inactivation of p38 MAPK abolished the GADD45B-mediated myogenic differentiation. Furthermore, we found that the knockdown of fat mass and obesity-associated protein (FTO) increases GADD45B m6A modification and decreases the stability of GADD45B mRNA, which impairs myogenic differentiation. Our results indicate that the FTO-mediated m6A modification in GADD45B mRNA drives skeletal muscle differentiation by activating the p38 MAPK pathway, which provides a molecular mechanism for the regulation of myogenesis via RNA methylation.

BackgroundTo date, the repeated breakout of the novel coronavirus disease 2019 (COVID-19) pandemic across many regions in China has caused continuous physical and mental harm to health care workers. This study investigates the psychological burden of the pandemic and its associated risk factors among Chinese healthcare workers (HCWs) during a single wave of COVID-19.MethodsFor this cross-sectional web-based survey conducted from January 16, 2022 to February 5, 2022, a total of 412 HCWs from Northwestern China were recruited. Their socio-demographic data and COVID-19 related survey variables were then collected using online self-rating questionnaires. In addition, the Chinese versions of well-validated instruments, including the 12-item General Health Questionnaire for psychiatric morbidity, the Generalized Anxiety Disorder Scale-7 for anxiety, the Patient Health Questionnaire-9 for depression and the Insomnia Severity Index-7 for insomnia, were used to assess the participants' mental health status. Multivariate logistic regression analysis was eventually performed to identify the risk factors associated with the psychological outcomes.ResultsOf the 388 participants who were included in the final study (94.17% response rate), the prevalence of anxiety, depression, and insomnia symptoms were 25.3% (95% CI: 20.9-29.6%), 40.7% (95% CI: 35.8-45.6%), and 30.9% (95% CI: 26.3-35.5%), respectively. Multivariate logistic regression analysis revealed that being a woman and having a perceived need for psychological support were risk factors for all psychological outcomes, while poor disease cognition and perceived susceptibility were risk factors for anxiety. Poor disease cognition and being unvaccinated against COVID-19 were risk factors for depression, with the latter also being an independent risk factor for insomnia.ConclusionThis study has identified a relatively lower prevalence rate of psychological disorders among Chinese HCWs during a single wave, deeper into the COVID-19 pandemic. Female HCWs, and those who had a perceived need for psychological support, had poor disease cognition, were perceived as susceptible to COVID-19 and had not been vaccinated against COVID-19 deserve more attention.

Background The association between inflammation and major depressive disorder (MDD) remains poorly understood, given the heterogeneity of patients with MDD.Aims We investigated inflammatory markers, such as interleukin (IL)-6, high-sensitivity C reactive protein (hsCRP) and tumour necrosis factor-α (TNF-α) in melancholic, atypical and anxious depression and explored whether baseline inflammatory protein levels could indicate prognosis.Methods The sample consisted of participants (aged 18–55 years) from a previously reported multicentre randomised controlled trial with a parallel-group design registered with ClinicalTrials.gov, including melancholic (n=44), atypical (n=37) and anxious (n=44) patients with depression and healthy controls (HCs) (n=33). Subtypes of MDD were classified according to the 30-item Inventory of Depressive Symptomatology, Self-Rated Version and the 17-item Hamilton Depression Rating Scale. Blood levels of TNF-α, IL-6 and hsCRP were assessed using antibody array analysis.Results Patients with MDD, classified according to melancholic, atypical and anxious depression subtypes, and HCs did not differ significantly in baseline TNF-α, IL-6 and hsCRP levels after adjustment. In patients with anxious depression, hsCRP levels increased significantly if they experienced no pain (adjusted (adj.) p=0.010) or mild to moderate pain (adj. p=0.038) compared with those with severe pain. However, the patients with anxious depression and severe pain showed a lower trend in hsCRP levels than patients with atypical depression who experienced severe pain (p=0.022; adj. p=0.155). Baseline TNF-α (adj. p=0.038) and IL-6 (adj. p=0.006) levels in patients in remission were significantly lower than those in patients with no remission among the participants with the atypical depression subtype at the eighth-week follow-up.Conclusions This study provides evidence of differences in inflammatory proteins in patients with varied symptoms among melancholic, atypical and anxious depression subtypes. Further studies on the immunoinflammatory mechanism underlying different subtypes of depression are expected for improved individualised therapy.Trial registration number NCT03219008.

As decision-making tools helping to improve the understanding of soil quality, soil quality assessment and heavy metal pollution assessment are very important for the remediation of heavy metal soil pollution. In the past, soil quality and heavy metal pollution have been studied separately, and few studies have combined them. The desert steppe in the Northwest Arid Region is an important pasture resource in China, and its soil safety has always been the focus of attention. Therefore, to understand the impact of tailing stockpiles on the soil quality of desert steppe, this study analyzed 18 indicators in the sample and analyzed the soil quality status of desert steppe based on the soil quality index (SQI) and Nemerow pollution index (Pcom). The main conclusions are as follows. (1) The evaluation results of heavy metal soil pollution show that the heavy metals Cu, Ni, Cr and Cd are significant polluters, Mn is a moderate polluter and Zn is a slight polluter. The results of the positive matrix factorization model show that Cu and Ni come from industrial sources; Cr, Cd and Zn come from industrial and traffic sources; and Mn comes from natural sources. (2) Regarding the study area, the generated minimum data set contains clay, pH, soil organic matter, available phosphorus, urease and neutral phosphatase. (3) The results of the SQI show that the soil in the study area is grade V (SQI-TDSave (total data set) = 0.42; SQI-MDSave (minimum data set) = 0.39), and the soil condition is very poor. 4) The linear fitting results show that the SQI-MDS was positively correlated with the SQI-TDS (R2 = 0.79), and SQI-MDS and SQI-TDS were negatively correlated with the Pcom (R2 > 0.6). Therefore, the leakage of acid mine drainage from tailings pond accumulation has led to a significant decline in the soil quality of this desert steppe, and effective ecological restoration measures are urgently needed to ensure the sustainable stability of the steppe ecosystem.

BackgroundSystemic therapies, including immune checkpoint inhibitors (ICIs) and tyrosine kinase inhibitors (TKIs), have challenged the use of conventional therapies for hepatocellular carcinoma (HCC). It is crucial to determine which patients could benefit most from combination therapy. This study aims to examine the associations of sarcopenia and systemic inflammation response index (SIRI) with the treatment responses and efficacies in patients with HCC treated with ICIs and tyrosine kinase inhibitors TKIs, as well as investigate the correlation between sarcopenia and inflammatory or immune states.MethodsWe reviewed 160 patients with HCC treated with TKIs and ICIs. The patients’ psoas muscle size was measured on axial computed tomography scans and normalized for the patients’ height squared. This value was referred to as the psoas muscle index (PMI). Sarcopenia was determined from PMI and their relationships with patients’ clinicopathological characteristics, inflammation indexes, peripheral blood T-cell subsets and survival were evaluated.ResultsSarcopenia and systemic inflammation response index (SIRI) were independent predictors for overall survival and progression-free survival. Patients with high PMI and low SIRI demonstrated significantly better median overall survival and progression-free survival (36.0 months and 9.6 months, respectively) than those with either low PMI or high SIRI (20.8 months and 6.0 months, respectively) and those with both high SIRI and low PMI (18.6 months and 3.0 months, respectively). Portal vein tumor thrombus (P=0.003), eastern cooperative oncology group performance status score of 1 (P=0.048), high alkaline phosphatase (P=0.037), high neutrophil-to-lymphocyte ratio (NLR) (P=0.012), low lymphocyte-to-monocyte ratio (LMR) (P=0.031), high platelet-to-lymphocyte ratio (PLR) (P=0.022) and high SIRI (P=0.012) were closely associated with an increased incidence of sarcopenia. PMI was negatively correlated with SIRI (r = -0.175, P=0.003), NLR (r = -0.169, P=0.036), and PLR (r = -0.328, P=0.000) and was significantly positively correlated with LMR (r = 0.232, P=0.004). The CD3+ and CD4+ T-cell counts of the high PMI group were significantly higher than those of the low PMI group.ConclusionSarcopenia and high SIRI were associated with reduced survival in patients with HCC treated with ICIs and TKIs. Sarcopenia could affect inflammatory states and the immune microenvironment.

Background: CDC25 is a dual-specificity phosphatase that was first identified in the yeast Schizosaccharomyces pombe as a cell cycle-defective mutant. Although CDC25 is involved in the cell cycle of ovarian granulosa cells, the CDC25 signaling pathway has not been clarified fully. To explore the role of CDC25C in the cell cycle of goat ovarian granulosa cells, a CDC25C-overexpressing vector, pCMV-HA-CDC25C, was constructed and transfected into granulosa cells from adult and young white goats from Jiangsu Nantong. RT-PCR was used to measure CDC25C, CDK1, and WEE1 gene expression levels, and flow cytometry was used to distinguish ovarian granulosa cells in different phases of the cell cycle. Progesterone and estradiol levels in transfected ovarian granulosa cells were also measured. Results: In adult goat follicular granulosa cells transfected with pCMV-HA-CDC25C, CDC25C expression increased significantly, which greatly increased the relative gene expression levels of both CDK1 and WEE1. Additionally, progesterone and estradiol levels were increased in goat follicular granulosa cells overexpressing CDC25C. And the cell cycle results showed that transfection of pCMV-HA-CDC25C leads to a higher proportion of cells in S phase compared to the no vector-transfected groups. Conclusions: The results of this study indicated that the overexpression of CDC25C may increase the gene expression levels of both WEE1 and CDK1 in S phase and accelerate the transition of cells from G1 phase to S phase.

Biochemical methane potential (BMP) of anaerobic co-digestion (co-AD) feedstocks is an essential basis for optimizing ratios of materials. Given the time-consuming shortage of conventional BMP tests, a rapid estimated method was proposed for BMP of co-AD—with straw and feces as feedstocks—based on near infrared spectroscopy (NIRS) combined with chemometrics. Partial least squares with several variable selection algorithms were used for establishing calibration models. Variable selection methods were constructed by the genetic simulated annealing algorithm (GSA) combined with interval partial least squares (iPLS), synergy iPLS, backward iPLS, and competitive adaptive reweighted sampling (CARS), respectively. By comparing the modeling performances of characteristic wavelengths selected by different algorithms, it was found that the model constructed using 57 characteristic wavelengths selected by CARS-GSA had the best prediction accuracy. For the validation set, the determination coefficient, root mean square error and relative root mean square error of the CARS-GSA model were 0.984, 6.293 and 2.600, respectively. The result shows that the NIRS regression model—constructed with characteristic wavelengths, selected by CARS-GSA—can meet actual detection requirements. Based on a large number of samples collected, the method proposed in this study can realize the rapid and accurate determination of the BMP for co-AD raw materials in biogas engineering.

A new method for producing biomass seedling trays with cow manure was developed based on heating to above the glass transition point of the lignin in cow dung. The maize seedling tray, manufactured through heat compression molding, could potentially meet the demand for transplanting maize seedlings. A scanning electron microscope and universal testing machine were used to compare the changes in the internal structure and mechanical properties of the seedling tray before and after the seeding period through a compression molding technique at normal temperature before seeding and a heat compression molding technique after seeding. The results showed that the strength of the seedling tray was mainly derived from the mechanical setting force of the fiber laminated in the seedling tray. Meanwhile, the moisture in the seedling tray hindered the lignin from filling in the gaps between the stem fibers in cow manure, as well as it reduced the protective effect of lignin on the laminated inlay structure of the stem fibers in cow manure. Therefore, under the premise that the material could be completely filled in the mold, the study concluded that a lower moisture content resulted in better strength and water resistance of the seedling tray. Background: Gastric cancer (GC) remains one of the most common malignancies worldwide. Increasing evidence has demonstrated that circRNAs serve as critical roles in human cancer, including GC. In the present study, we focused on the detailed function and mechanism of circ 0000144 on GC progression. Methods: The levels of circ 0000144, miR-623 and G-protein-coupled receptor, family C, group 5, member A (GPRC5A) were determined by quantitative real-time polymerase chain reaction (qRT-PCR). Targeted relationships among circ 0000144, miR-623 and GPRC5A were confirmed using dual-luciferase reporter and RNA immunoprecipitation (RIP) assays. Cell proliferation, colony formation, apoptosis, migration and invasion were evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), colony formation, flow cytometry and transwell assays. Measurement of glutamine and α-ketoglutarate (α-KG) levels was performed using a corresponding assay kit. GPRC5A protein expression was detected using Western blot. In vivo assays were used to explore the impact of circ 0000144 on tumor growth. Results: Our data indicated that circ 0000144 was up-regulated and miR-623 was down-regulated in GC tissues and cells. Circ 0000144 interacted with miR-623 through directly binding to miR-623. Moreover, the knockdown of circ 0000144 weakened GC cell proliferation, colony formation, migration, invasion and glutaminolysis and accelerated cell apoptosis by up-regulating miR-623. GPRC5A was a direct target of miR-623 and circ 0000144 protected against GPRC5A repression through sponging miR-623. Furthermore, miR-623-mediated regulation on GC cell progression was reversed by the stored expression of GPRC5A. Additionally, circ 0000144 depletion inhibited tumor growth in vivo. Conclusion: Our study indicated that circ-0000144 knockdown repressed GC progression at least partly by regulating GPRC5A expression via sponging miR-623, illumining a novel therapeutic target for GC treatment. Background: Significant anxiety symptoms are associated with poor clinical course and outcome in major depressive disorder (MDD). This single-arm, open-label study aimed to evaluate the efficacy and tolerability of escitalopram treatment in patients with MDD and anxiety symptoms. Methods: Adult patients with MDD and anxiety symptoms (Montgomery-Asberg Depression Rating Scale [MADRS] ≥22 and Hamilton Anxiety Rating Scale [HAM-A] ≥14) were enrolled and received escitalopram (10-20 mg/day) treatment for 24 weeks. Symptom status was assessed by MADRS, 17-item-Hamilton Depression Rating Scale, HAM-A, and Clinical Global Impression Scale at baseline and the following visits. Quality of life was assessed by Short Form-12, and safety was evaluated by adverse events, laboratory investigations, vital signs, and physical findings. Results: Overall, 200 of 318 (66.2%) enrolled patients completed the 24-week treatment. The remission (MADRS ≤10 and HAM-A ≤7) rate in the full analysis set (N=285) was 73.3% (95% confidence interval: 67.80, 78.38) at week 24. Mean (± standard deviation) MADRS total score was 33.4 (±7.13) and HAM-A score was 27.6 (±7.26) at baseline, which reduced to 6.6 (±10.18) and 6.0 (±8.39), respectively, at week 24. Patients with higher baseline depression and anxiety level took longer to achieve similar remission rates. Overall, 80 of the 302 (26.5%) patients included in the safety set reported at least 1 treatment-emergent adverse event (TEAE). Most frequently reported TEAEs (.2%) were headache (4.0%), nasopharyngitis (3.6%), nausea (3.0%), and dizziness (2.6%). Serious TEAEs were reported by 1.3% patients; no deaths were reported. Conclusion: Escitalopram 10-20 mg/day was effective and well-tolerated in the long-term treatment of MDD with anxiety symptoms in adult Chinese population. [ABSTRACT FROM AUTHOR]

Accurate assessment of the mechanical properties of organic matter, clay matrix, and bulk shale during maturation remains a challenge. Here, we aim to assess the mechanical properties of organic-rich shale during maturation using a combination of nanoindentation methods and various geochemical analyses, i.e., mineral composition, mass loss rate, chemical structure of organic matter, and Rock-Eval analyses. Results show that the evolution of mechanical properties of organic matter in shale during maturation can be divided into: the main oil-generation stage, and the condensate oil and gas generation stage. The stiffening of organic matter in the shale is mainly due to increased aromaticity and condensation of aromatic groups. The clay matrix experiences a slight decrease in hardness and Young's modulus at low maturity levels due to the generation of liquid hydrocarbons. However, overall, the clay matrix becomes stiffer as the shale matures due to shale dehydration, expulsion or cracking of liquid hydrocarbons, transformation of clay minerals, and hardening of organic matter. The Young's modulus and hardness of bulk shale generally increase with increasing maturity. The gum acacia Senegalia senegal (L.) Britton (Fabales: Fabaceae) is a drought-tolerant plant belonging to the genus Acacia of the Leguminosae family, possessing significant economic and ecological value. Despite its importance, there is a knowledge gap regarding the potential impact of climate change on the distribution of S. senegal, crucial for the conservation of plant resources and optimizing its use in introductory silviculture. In this study, we selected 23 environmental variables and utilized the optimized maximum entropy (MaxEnt) model to analyze the key environmental factors affecting the distribution of S. senegal worldwide and simulate the current and future distribution range of S. senegal in Pakistan under the SSP1-2.6, SSP2-4.5, SSP3-7.0, and SSP5-8.5 climate change scenarios. The results highlight key environmental factors influencing the distribution of S. senegal, including BIO3 (isothermally), BIO4 (coefficient of seasonal variation of temperature), BIO11 (mean temperature of the coldest season), and BIO12 (annual precipitation). Regions with higher and less fluctuating temperatures exhibit a higher potential for S. senegal distribution. Currently, suitable habitats of S. senegal are concentrated in the southern region of Pakistan, covering provinces such as Punjab, Sindh, and Balochistan, with highly suitable habitats accounting for 6.06% of the total area. Under the current climatic conditions, this study identifies the spatial patterns of suitable habitats and their concentration in specific regions. With climate change, a notable expansion of suitable habitats towards higher latitudes is observed, with the most significant expansion under the extremely severe climate change scenario (SSP5-8.5), reaching 223.45% of the current level. The results of this study enhance our understanding of the dynamics of S. senegal distribution under climate change and offer valuable insights into the long-term introduction of S. senegal for afforestation and soil conservation in Pakistan. This study provides theoretical support for the sustainable development of the local ecosystem and socio-economy, emphasizing the importance of proactive measures to adapt to changing climatic conditions. Background: Rectal neuroendocrine neoplasms (NENs) are rare neoplasms with limited understanding of its genomic alterations and molecular typing. Methods: The paraffin‐embedded tissue specimens of 38 patients with rectal NENs after surgery were subjected to whole gene sequencing (WGS), and mutation profilings were drawn to identify high‐frequency mutation genes, copy‐number variations (CNVs), tumor mutation burden (TMB), signal pathways, mutation signatures, DNA damage repair (DDR) genes, and molecular types. The differences of mutated genes and signaling pathways in different pathological grades and metastatic/non‐metastatic groups were compared. It helped to search for potential targets. Results: C > T and T > C transitions are the most common base substitutions in rectal NENs. DNA mismatch repair deficiency, DNA base modifications, smoking and exposure to ultraviolet light might play a role in the occurrence of rectal NENs. DAXX, KMT2C, BCL2L1, LTK, MERTK, SPEN, PKN1, FAT3, and LRP2 mutations were found in only low‐grade rectal NETs, whereas APC, TP53, NF1, SOX9, and BRCA1 mutations were common in high‐grade rectal NECs/MiNENs. These genes helped in distinguishing poorly‐differentiated or well‐differentiated rectal NENs. Alterations in P53, Wnt and TGFβ signaling pathways were more pronounced in rectal NECs and MiNENs. Alterations in Wnt, MAPK and PI3K/AKT signaling pathways promoted metastases. Rectal NENs were classified into two molecular subtypes by cluster analysis based on the mutant genes and signaling pathways combined with clinicopathological features. Patients with mutations in the LRP2, DAXX, and PKN1 gene showed a trend of well‐differentiated and early‐stage tumors with less metastasis (p = 0.000). Conclusions: This study evaluated risk factors for regional lymphatic and/or distant metastases, identified high‐frequency mutated genes, mutation signatures, altered signaling pathways through NGS. Rectal NENs were divided into two molecular types. This helps to evaluate the likelihood of metastasis, formulate follow‐up strategies for patients and provide a target for future research on precision treatment of rectal NENs. Background: Programmed cell death protein-1 inhibitors combined with lenvatinib have become a popular treatment option for patients with unresectable hepatocellular carcinoma. Transarterial chemoembolization combined with programmed cell death protein-1 inhibitors and lenvatinib has also shown preliminary efficacy in the unresectable hepatocellular carcinoma. We conducted this observational, retrospective, cohort study to compare the clinical outcomes and safety of transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib versus programmed cell death protein-1 inhibitors plus lenvatinib in patients with unresectable hepatocellular carcinoma. Methods: Between November 2019 and November 2021, patients who were diagnosed with unresectable hepatocellular carcinoma and received transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib or programmed cell death protein-1 inhibitors plus lenvatinib treatment were reviewed for eligibility. The primary endpoints included objective response rate, overall survival, and progression-free survival. The secondary endpoint was the frequency of key adverse events. Results: In total, 105 patients were eligible for the present study, and they were divided into the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group (n = 46) and the programmed cell death protein-1 inhibitors plus lenvatinib group (n = 59). The patient cohort after a one-to-one propensity score matching (n = 86) was also analyzed. The transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group had a higher objective response rate both in the patient cohort before propensity score matching (54.3% vs 25.4%, P =.002) and after propensity score matching (55.8% vs 30.2%, P =.017). The patients in the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group had prolonged overall survival (median, 20.5 vs 12.6 months, P =.015) and progression-free survival (median, 10.2 vs 7.4 months, P =.035). For patient cohort- propensity score matching, the overall survival (20.5 vs 12.8 months, P =.013) and progression-free survival (12.1 vs 7.8 months, P =.030) were also significantly better in the transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib group than in the programmed cell death protein-1 inhibitors plus lenvatinib group. There were no significant differences between the 2 groups concerning adverse reactions caused by immunotherapy and lenvatinib. The adverse reactions caused by transarterial chemoembolization were transient and were quickly reversed. Conclusions: Compared to programmed cell death protein-1 inhibitors plus lenvatinib, transarterial chemoembolization combined with programmed cell death protein-1 inhibitors plus lenvatinib may provide better treatment response and survival benefits for patients with unresectable hepatocellular carcinoma, and the adverse events were manageable. Background: Elevated level of interleukin-8 (IL-8) promotes hepatocellular carcinoma (HCC) development and contributes to poor prognosis. Previously, we have proved that Dicer inhibits HCC progression. In this study, we evaluated the potential interaction between IL-8 and Dicer as well as their influence on HCC. Methods: Hepatocellular carcinoma cells of SMMC-7721 were divided into 2 groups for subsequent analysis: pCMV-Dicer group for Dicer-overexpressing lentivirus transfected cells (pCMV-Dicer cells) and pCMV-NC group for empty lentivirus transfected cells (pCMV-NC cells). Cell Counting kit-8 (CCK8), wound healing, and transwell were used to evaluate the inhibitory effect of Dicer overexpression on proliferation, migration, and invasion of HCC cells. The level of IL-8 was measured by flow cytometry bead-based immunoassays. Male nude BALB/c mice injected with pCMV-Dicer or pCMV-NC cell suspensions was used for transplant of HCC tumor. Results: We found that the secretion of IL-8 was reduced in the medium of pCMV-Dicer cells (P =.027). Recombinant human IL-8 (rhIL-8) reversed the inhibitory effect of Dicer on proliferation (P <.01), migration (P =.003), and invasion (P =.001), whereas IL-8 inhibitor of reparixin enhanced inhibitory effect of Dicer on proliferation (P <.05), migration (P =.008), and invasion (P =.000). Lenvatinib downregulated the IL-8 level of HCC cells (P =.000) as well as promote Dicer-induced inhibition for HCC cells referring to proliferation (P <.05), migration (P =.000), and invasion (P =.000). Animal experiments also demonstrated that Dicer cooperated with lenvatinib to inhibit the growth of HCC tumors (P <.05). Conclusions: Dicer cooperated with lenvatinib to inhibit HCC growth via downregulating IL-8, and Dicer displayed its potential capability to enhance the anti-tumor effect of lenvatinib.
