In this work, a new method named laser-heating-wax-printing (LHWP) is described to fabricate paper devices for developing sensitive, affordable, user-friendly paper-based enzyme-linked immunosorbent assays (P-ELISAs) that initially use common pen-type pH meters for portable, quantitative readout. The LHWP enables a rapid patterning of wax in paper via one step of heating the wax layer coated on the paper surface using a mini-type CO 2 laser machine. Wax-patterned paper microzones created in this way are utilized to conduct the pen-type pH meter-based P-ELISAs with enzyme-loaded SiO 2 microbeads for highly efficient signal amplification of each antibody-antigen binding event. The results show that this new P-ELISA system is quantitatively sensitive to the concentrations of a model protein analyte in buffer samples ranging from 12.5 to 200 pg mL -1 , with a limit of detection of ca. 7.5 pg mL -1 (3σ). Moreover, the satisfactory recovery results of assaying several human serum samples validate its feasibility for practical applications.

Paper-based microfluidics is an emerging analysis tool used in various applications, especially in point-of-care (PoC) diagnostic applications, due to its advantages over other types of microfluidic devices in terms of simplicity in both production and operation, cost-effectiveness, rapid response time, low sample consumption, biocompatibility, and ease of disposal. Recently, various techniques have been developed and utilized for the fabrication of paper-based microfluidics, such as photolithography, micro-embossing, wax and PDMS printing, etc. In this study, we offer a fabrication methodology for a microfluidic paper-based immunosorbent assay (μPISA) platform and the detection of Hepatitis C Virus (HCV) was carried out to validate this platform. A laser ablation technique was utilized to form hydrophobic barriers easily and rapidly, which was the major advantage of the developed fabrication methodology. The characterization of the μPISA platform was performed in terms of micro-channel properties using bright-field (BF) microscopy, and surface properties using scanning electron microscopy (SEM). At the same time, sample volume and liquid handling capacity were analyzed quantitatively. Ablation speed (S) and laser power (P) were optimized, and it was shown that one combination (10P60S) provided minimal deviation in micro-channel dimensions and prevented deterioration of hydrophobic barriers. Also, the minimum hydrophobic barrier width, which prevents cross-barrier bleeding, was determined to be 255.92 ± 10.01 μm. Furthermore, colorimetric HCV NS3 detection was implemented to optimize and validate the μPISA platform. Here, HCV NS3 in both PBS and human blood plasma was successfully detected by the naked eye at concentrations as low as 1 ng mL−1 and 10 ng mL−1, respectively. Moreover, the limit of detection (LoD) values for HCV NS3 were acquired as 0.796 ng mL−1 in PBS and 2.203 ng mL−1 in human blood plasma with a turnaround time of 90 min. In comparison with conventional ELISA, highly sensitive and rapid HCV NS3 detection was accomplished colorimetrically on the developed μPISA platform. [ABSTRACT FROM AUTHOR]

Cervical cancer is a leading cause of cancer death for women in low-resource settings. The World Health Organization recommends that cervical cancer screening programs incorporate HPV DNA testing, but available tests are expensive, require laboratory infrastructure, and cannot be performed at the point-of-care. We developed a two-dimensional paper network (2DPN), hybrid-capture, signal amplification assay and a point-of-care sample preparation protocol to detect high-risk HPV DNA from exfoliated cervical cells within an hour. The test does not require expensive equipment and has an estimated cost of <$3 per test without the need for batching. We evaluated performance of the paper HPV DNA assay with short synthetic and genomic HPV DNA targets, HPV positive and negative cellular samples, and two sets of clinical samples. The first set of clinical samples consisted of 16 biobanked, provider-collected cervical samples from a study in El Salvador previously tested with careHPV and subsequently tested in a controlled laboratory environment. The paper HPV DNA test correctly identified eight of eight HPV-negative clinical samples and seven of eight HPV-positive clinical samples. We then performed a field evaluation of the paper HPV DNA test in a hospital laboratory in Mozambique. Cellular controls generated expected results throughout field testing with fully lyophilized sample preparation and 2DPN reagents. When evaluated with 16 residual self-collected cervicovaginal samples previously tested by the GeneXpert HPV assay ("Xpert"), the accuracy of the HPV DNA paper test in the field was reduced compared to testing in the controlled laboratory environment, with positive results obtained for all eight HPV-positive samples as well as seven of eight HPV-negative samples. Further evaluation showed reduction in performance was likely due in part to increased concentration of exfoliated cells in the self-collected clinical samples from Mozambique compared with provider-collected samples from El Salvador. Finally, a formal usability assessment was conducted with users in El Salvador and Mozambique; the assay was rated as acceptable to perform after minimal training. With additional optimization for higher cell concentrations and inclusion of an internal cellular control, the paper HPV DNA assay offers promise as a low-cost, point-of-care cervical cancer screening test in low-resource settings. We report a highly accurate single-step label-free testing technology for simultaneous and independent hematocrit (Hct) and hemoglobin (Hb) level detection from a drop of whole blood by employing a disposable paper strip sensor interfaced with a portable impedimetric device. The paper strip is fabricated by in situ automated printing of a customized electrode template on the non-glossy side of a commercially available photo paper substrate followed by graphite deposition. The integrated platform device technology additionally includes a compact detection cum readout unit comprising a high precision impedance converter system that combines an on-board frequency generator with an analog-to-digital converter evaluation board, collectively interfaced with a central processor, calibration circuit, and smartphone. Employing a dispensed blood sample volume of 25 μL, the device is shown to have a sensitivity of 92 Ω/Hct and 287 Ω/Hb at an optimal frequency of 57 kHz. The respective linear response regimes appear to be wide enough to cover physiologically relevant limits, with excellent stability and reproducibility. Validation with clinical samples reveals limits of detection of Hct and Hb levels as low as 4.66% and 1.89 g dL−1, respectively, which are beyond the quantitative capability of commonly used affordable point of care test kits. The envisaged paradigm of rapid, robust, highly accurate, energy-efficient, simple, user-friendly, multiplex portable detection, obviating any possible ambiguities in interpretation due to common artefacts of colorimetric detection technologies such as optical interference with the image analytical procedure due to the inherent redness of blood samples and background illumination, renders this ideal for deployment in resource-limited settings. The pressing priority for flexible photo-responsive systems is scalability and simplistic fabrication routes at low production costs. The extraordinary properties seen in 2D materials, paired with solution-processing advantages, assert their employability for superior photoresponsive systems. Herein, we demonstrate versatile and flexible photodetectors and photo-thermal actuators by integrating 2D materials onto paper substrates. All-solution processed flexible photodetectors have been fabricated by spray coating dispersions of MoS2 nanosheets onto screen printed carbon electrodes. By employing MoSe2 as a photothermal conversion layer, actuators with paper-tape bilayers have also been demonstrated. For both photoresponsive applications, the process of calendering investigated in detail results in remarkable augmentation of the performances. Real-world thermally stimulated soft robotic applications like jacks, grippers and crawlers are realized by leveraging a configuration dependent response to illumination. Thus, a well-structured and feasible implementation of a chain of protocols that includes screen printing, spray coating, lamination and calendering with paper as the substrate led us to fabricate flexible photoresponsive systems with significantly enhanced performances. We demonstrated an instrument-free miniaturized adaptation of the laboratory gold standard methodology for the direct estimation of plasma glucose from a drop of whole blood using a low-cost single-user-step paper-strip sensor interfaced with a smartphone. Unlike a majority of the existing glucose meters that use whole blood-based indirect sensing technologies, our direct adaptation of the gold-standard laboratory benchmark could eliminate the possibilities of cross interference with other analytes present in the whole blood by facilitating an in situ plasma separation, capillary flow and colorimetric reaction occurring concomitantly, without incurring additional device complexity or embodiment. The test reagents were dispensed in lyophilized form, and the resulting paper strips were found to be stable over three months stored in a normal freezer, rendering easy adaptability commensurate with the constrained supply chains in extreme resource-poor settings. Quantitative results could be arrived at via a completely-automated mobile-app-based image analytics interface developed using dynamic machine learning, obviating manual interpretation. The tests were demonstrated to be of high efficacy, even when executed by minimally trained frontline personnel having no special skill of drawing precise volume of blood, on deployment at under-resourced community centres having no in-built or accessible healthcare infrastructure. Clinical validation using 220 numbers of human blood samples in a double-blinded manner evidenced sensitivity and specificity of 98.11% and 96.7%, respectively, as compared to the results obtained from a laboratory-benchmarked biochemistry analyser, establishing its efficacy for public health and community disease management in resource-limited settings without any quality compromise of the test outcome. Bacterial meningitis, an infection of the membranes (meninges) and cerebrospinal fluid (CSF) surrounding the brain and spinal cord, is one of the major causes of death and disability worldwide. Higher case-fatality rates and short survival times have been reported in developing countries. Hence, a quick, straightforward, and low-cost approach is in great demand for the diagnosis of meningitis. In this research, a microfluidic fully paper-based analytical device (μFPAD) integrated with loop-mediated isothermal amplification (LAMP) and ssDNA-functionalized graphene oxide (GO) nano-biosensors was developed for the first time for a simple, rapid, low-cost, and quantitative detection of the main meningitis-causing bacteria, Neisseria meningitidis (N. meningitidis). The results can be successfully read within 1 hour with the limit of detection (LOD) of 6 DNA copies per detection zone. This paper device also offers versatile functions by providing a qualitative diagnostic analysis (i.e., a yes or no answer), confirmatory testing, and quantitative analysis. These features make the presented μFPAD capable of a simple, highly sensitive, and specific diagnosis of N. meningitis. We leverage electroosmotic-flow generation in porous media in combination with a hydrophobic air gap to create a controllable valve capable of operating in either finite dosing or continuous flow mode, enabling the implementation of multi-step assays on paper-based devices. The hydrophobic air gap between two paper pads creates a barrier keeping the valve nominally closed. Electroosmotic actuation, implemented using a pair of electrodes under the upstream pad, generates sufficient pressure to overcome the barrier and connect the two pads. We present a model describing the flow and governing parameters, including the electric potentials required to open and close the valve and the threshold potential for switching between the modes of operation. We construct the air gap using a hierarchical superhydrophobic surface and study the stability of the closed valve under strenuous conditions and find good agreement between our model and experimental results, as well as stable working conditions for practical applications. We present a straightforward design for a compact and automated device based on paper pads placed on top of printed circuit boards (PCB), equipped with heating and actuation electrodes and additional power and logic capabilities. Finally, we demonstrate the use of the device for amplification of SARS-CoV-2 sequences directly from raw saliva samples, using a loop-mediated isothermal amplification (LAMP) protocol requiring sample lysis followed by enzymatic deactivation and delivery to multiple amplification sites. Since PCB costs scale favorably with mass-production, we believe that this approach could lead to a low-cost diagnostic device that offers the sensitivity of amplification methods. In this paper, we propose a generalized concept of microfluidic polydimethylsiloxane (PDMS) on paper (POP) devices, which combines well the merits of paper chips and PDMS chips. First, we optimized the conditions for accurate PDMS spatial patterning on paper, based on screen printing and a high temperature enabled superfast curing technique, which enables PDMS patterning to an accuracy of tens of microns in less than ten seconds. This, in turn, makes it available for seamless, reversible and reliable integration of the resulting paper layer with other PDMS channel structures. The integrated POP devices allow for both porous paper and smooth channels to be spatially defined on the devices, greatly extending the flexibility for designers to be able to construct powerful functional structures. To demonstrate the versatility of this design, a prototype POP device for the colorimetric analysis of liver function markers, serum protein, alkaline phosphatase (ALP) and aspartate aminotransferase (AST), was constructed. On this POP device, quantitative sample loading, mixing and multiplex analysis have all been realized.

The development of a competitive immunoassay system for colorimetric detection on microfluidic paper-based analytical devices (μPADs) is reported. The μPADs were fabricated via photolithography to define hydrophilic flow channels and consisted of three main elements: the control and test zones, where target detection was performed, the sample introduction zone, and the competitive capture zone located between the sample introduction zone and the test zone. The chromogenic substrate 3,3',5,5'-tetramethylbenzidine (TMB) was deposited at the control and test zones. μPAD surface modification was performed at the capture zone first via chitosan activation, then the BSA-conjugated target compound was immobilized. The sample solution consisting of the target compound, the peroxidase-conjugated antibody, and the hydrogen peroxide oxidizing agent was introduced into the device and competition occurred at the capture zone, allowing only the target-bound peroxidase-conjugated antibody to travel past the capture zone and into the test zone via capillary action. The developed competitive immunoassay system was successfully demonstrated on the μPAD detection of biotin as a model compound with a detection limit of 0.10 μg mL -1 . The applicability of the proposed immunoassay system for point-of-need testing was further demonstrated using aflatoxin B 1 , a highly toxic foodborne substance, with a detection limit of 1.31 ng mL -1 . The μPAD with the competitive immunoassay format showed promising results for practical applications in point-of-need testing involving small molecular weight targets in food and water safety and quality monitoring, environmental analysis, and clinical diagnostics.

Culturing cells at the air-liquid interface (ALI) is essential for creating functional in vitro models of lung tissues. We present the use of direct-patterned laser-treated hydrophobic paper as an effective semi-permeable membrane, ideal for ALI cell culture. The surface properties of the paper are modified through a selective CO 2 laser-assisted treatment to create a unique porous substrate with hydrophilic regions that regulate fluid diffusion and cell attachment. To select the appropriate model, four promising hydrophobic films were compared with each other in terms of gas permeability and long-term strength in an aqueous environment (wet-strength). Among the investigated substrates, parchment paper showed the fastest rate of oxygen permeability (3 times more than conventional transwell cell culture membranes), with the least variation in its dry and wet tensile strengths (124 MPa and 58 MPa, remaining unchanged after 7 days of submersion in PBS).The final paper-based platform provides an ideal, robust, and inexpensive device for generating monolayers of lung epithelial cells on-chip in a high-throughput fashion for disease modelling and in vitro drug testing.

The quantification of blood cells provides critical information about a patient's health status. Sophisticated analytical equipment, such as hematology analyzers, have been developed to perform these measurements, but limited-resource settings often lack the infrastructure required to purchase, operate, and maintain instrumentation. To address these practical challenges, paper-based microfluidic devices have emerged as a platform to develop diagnostic assays specifically for use at the point-of-care. To date, paper-based microfluidic devices have been used broadly in diagnostic assays that apply immunoassay, clinical chemistry, and electrochemistry techniques. The analysis of cells, however, has been largely overlooked. In this communication, we demonstrate a paper-based microfluidic device that enables the controlled transport of red blood cells (RBCs) and the measurement of the hematocrit-the ratio of RBC packed cell volume to total volume of whole blood. The properties of paper, device treatment, and device geometry affect the overall extent and reproducibility of transport of RBCs. Ultimately, we developed an inexpensive (US$0.03 per device) thermometer-styled device where the distance traveled by RBCs is proportional to the hematocrit. These results provide a foundation for the design of paper-based microfluidic devices that enable the separation and detection of cells in limited-resource settings.

This review shows the recent advances and state of the art in paper-based analytical devices (PADs) through the analysis of their integration with microfluidics and LOC micro- and nanotechnologies, electrochemical/optical detection and electronic devices as the convergence of various knowledge areas. The important role of the paper design/architecture in the improvement of the performance of sensor devices is discussed. The discussion is fundamentally based on μPADs as the new generation of paper-based (bio)sensors. Data about the scientific publication ranking of PADs, illustrating their increase as an experimental research topic in the past years, are supplied. In addition, an analysis of the simultaneous evolution of PADs in academic lab research and industrial commercialization highlighting the parallelism of the technological transfer from academia to industry is displayed. A general overview of the market behaviour, the leading industries in the sector and their commercialized devices is given. Finally, personal opinions of the authors about future perspectives and tendencies in the design and fabrication technology of PADs are disclosed.

This paper describes the modification of microfluidic paper-based analytical devices (μPADs) with chitosan to improve the analytical performance of colorimetric measurements associated with enzymatic bioassays. Chitosan is a natural biopolymer extensively used to modify biosensing surfaces due to its capability of providing a suitable microenvironment for the direct electron transfer between an enzyme and a reactive surface. This hypothesis was investigated using glucose and uric acid (UA) colorimetric assays as model systems. The best colorimetric sensitivity for glucose and UA was achieved using a chromogenic solution composed of 4-aminoantipyrine and sodium 3,5-dichloro-2-hydroxy-benzenesulfonate (4-AAP/DHBS), which provided a linear response for a concentration range between 0.1 and 1.0 mM. Glucose and UA were successfully determined in artificial serum samples with accuracies between 87 and 114%. The limits of detection (LODs) found for glucose and UA assays were 23 and 37 μM, respectively. The enhanced analytical performance of chitosan-modified μPADs allowed the colorimetric detection of glucose in tear samples from four nondiabetic patients. The achieved concentration levels ranged from 130 to 380 μM. The modified μPADs offered analytical reliability and accuracy as well as no statistical difference from the values achieved through a reference method. Based on the presented results, the proposed μPAD can be a powerful alternative tool for non-invasive glucose analysis.

In this report, we describe the methodology for 96 parallel organic syntheses of peptides on Teflon-patterned paper assisted by heating with an infra-red lamp. SPOT synthesis is an important technology for production of peptide arrays on a paper-based support for rapid identification of peptide ligands, epitope mapping, and identification of bio-conjugation reactions. The major drawback of the SPOT synthesis methodology published to-date is suboptimal reaction conversion due to mass transport limitations in the unmixed reaction spot. The technology developed in this report overcomes these problems by changing the environment of the reaction from static to dynamic (flow-through), and further accelerating the reaction by selective heating of the reaction support in contact with activated amino acids. Patterning paper with Teflon allows for droplets of organic solvents to be confined in a zone on the paper array and flow through the paper at a well-defined rate and provide a convenient, power-free setup for flow-through solid-phase synthesis and efficient assembly of peptide arrays. We employed an infra-red (IR) lamp to locally heat the cellulosic support during the flow-through delivery of the reagents to each zone of the paper-based array. We demonstrate that IR-heating in solid phase peptide synthesis shortened the reaction time necessary for amide bond formation down to 3 minutes; in some couplings of alpha amino acids, conversion rates increased up to fifteen folds. The IR-heating improved the assembly of difficult sequences, such as homo-oligomers of all 20 natural amino acids.

Hydrogel-based flexible electronics have received increasing attention owing to the requirement in a variety of applications. However, the design and development of underwater and self-adaptive high-temperature strain sensors remain a significant challenge because of the swelling and dehydration of hydrogels in aquatic environments and extreme climate conditions. Herein, a versatile gel with superior self-adhesiveness, long-lasting underwater stability, excellent high-temperature tolerance and long-term non-drying properties is successfully prepared, which can serve as a wearable sensor for precisely and reliably detecting diverse body movements and the direction of human movements in air and aquatic environments. Meanwhile, based on the Morse code expression, the as-fabricated gel sensor can also be utilized as a wearable communication device for delivering various messages to the receiver. Furthermore, a wireless sensor with high-temperature adaptability is constructed, which can transmit in real time the motion signals at high temperature to the specific APP of a smartphone at room temperature via wireless communication. This strategy provides a new perspective and great convenience for motion monitoring under harsh conditions. In particular, the as-prepared gel demonstrates an obvious transparency-shifting behavior in different polar solvents, thus inspiring us to engineer it as a novel water-writing paper for achieving repeatable "writing–erasing" applications. The SARS-CoV-2 virus is known as the causal agent for the current COVID-19 global pandemic. The majority of COVID-19 patients develop acute respiratory distress syndrome (ARDS), while some experience a cytokine storm effect, which is considered as one of the leading causes of patient mortality. Lipids are known to be involved in the various stages of the lifecycle of a virus functioning as receptors or co-receptors that controls viral propagation inside the host cell. Therefore, lipid-related metabolomics aims to provide insight into the immune response of the novel coronavirus. Our study has focused on determination of the potential metabolomic biomarkers utilizing a Teslin® Substrate in paper spray mass spectrometry (PS-MS) for the development of a rapid detection test within 60 seconds of analysis time. In this study, results were correlated with PCR tests to reflect that the systemic responses of the cells were affected by the COVID-19 virus.

This study describes a microfluidic paper-based analytical device (μPAD) for separating plasma from whole blood and measuring glucose concentration. A two-dimensional μPAD was fabricated by wax printing, using chromatographic paper and was functionalized by the incorporation of chitosan into the plasma separation zone. Red blood cells were isolated from whole blood in the plasma separation zone consisting of a chitosan polymer structure and a wax barrier. The separated plasma, with buffer, was injected into the test zone to cause a color change. The colorimetric results were digitized with a scanner, and the concentration was calculated using a calibration curve. This μPAD, which separated the plasma from whole blood without a separation membrane, will be useful for constructing point-of-care testing devices that detect analytes in small sample volumes.

Paper-based diagnostic devices have been widely applied to assess the presence and status of a variety of clinical diseases by analyzing samples such as urine or blood. Due to their low cost, user-friendliness, and convenience, they have been used as point-of-care (POC) devices in countries lacking resources or energy. Despite wide-ranging research and implementation, paper-based devices have not previously been developed for wound analysis. Here, we discuss the successful development of such a tool to facilitate simple and rapid wound status assessment. The purpose of this study was to develop a paper-based elastase detection device (PEDD) for clinical wound assessment that specifically examines human neutrophil elastase (HNE), one of the most abundant serine proteases found in chronic wounds. The first step in this study was an examination of different paper substrate types (i.e., chromatography paper and filter paper) to determine which provided the best protease immobilization and colorimetric response. We then used a wax printing approach to create hydrophobic and hydrophilic regions and designated test zones created on both chromatography and filter papers. This allowed us to physically immobilize both substrate and protease within the desired test zone regions. This PEDD which demonstrated good sensitivity (0.631 μg mL−1, in a wound fluid system) can be used to monitor protease activity expressed in wounds. After developing this device, we examined samples from 9 patients presenting a total of 7 acute and 4 chronic wounds to determine wound HNE concentration. In 2022, the outbreak of the monkeypox virus occurred in many non-endemic countries, and the World Health Organization (WHO) assessed that this outbreak was "atypical". The establishment of a rapid and effective assay that can be used for the early diagnosis of monkeypox virus infection is crucial for outbreak prevention and control. In this study, the monkeypox virus A29 protein and the homologous vaccinia virus A27 protein and cowpox virus 162 protein were expressed in Escherichia coli BL21 for screening. We synthesized the monkeypox virus A29 17-49 peptide as the immunogen and obtained 25 monoclonal antibodies (mAbs) against the A29 protein using mouse hybridoma techniques. Then an immunochromatographic test strip method for detecting A29 was established. The strips utilizing mAb-7C5 and 5D8 showed the best sensitivity and lowest limit of detection: 50 pg mL -1 for purified A29 and specificity tests showed that the strips did not cross-react with other orthopox viruses (vaccinia virus or cowpox virus) as well as common respiratory pathogens (SARS-CoV-2, influenza A and influenza B). Therefore, this method can be used for early and rapid diagnosis of monkeypox virus infection by antigen detection.

Exposure to low concentrations of heavy metal cations seriously harms living organisms, hence they are considered environmental toxins. Portable simple detection systems are required for field monitoring of multiple metal ions. In this report, paper-based chemosensors (PBCs) were prepared by adsorbing 1-(pyridin-2-yl diazenyl) naphthalen-2-ol (chromophore), which recognizes heavy metals, onto filter papers coated with mesoporous silica nano spheres (MSNs). The high density of the chromophore probe on the surface of PBCs resulted in ultra-sensitive optical detection of heavy metal ions and short response time. The concentration of metal ions was determined using digital image-based colorimetric analysis (DICA) and compared to spectrophotometry under optimal sensing conditions. The PBCs exhibited stability and short recovery times. The detection limits determined using DICA of Cd 2+ , Co 2+ , Ni 2+ and Fe 3+ were 0.22, 0.28, 0.44, and 0.54 μM; respectively. Additionally, the linear ranges for monitoring Cd 2+ , Co 2+ , Ni 2+ and Fe 3+ were 0.44-4.4, 0.16-4.2, 0.8-8.5, and 0.002-5.2 μM; respectively. A scalable, and cost-effective method was employed to prepare self-supported CuSn bimetallic catalyst on carbon paper. The obtained CuSn catalyst demonstrates high faradaic efficiency of CO around or above 90% at a broad potential range from -0.7 to -1.8 V vs. reversible hydrogen electrode, greatly surpassing Cu or Sn counterparts.

Enzyme-linked immunosorbent assay (ELISA) is widely employed for detecting target molecules in bioassays including the serological assays that measure specific antibody titers. However, ELISA tests are inherently limited to centralized laboratories staffed with trained personnel as the assay workflow requires multiple steps to be performed in a specific sequence. Here, we report a dipstick ELISA test that automates this otherwise laborious process and reports the titer of a target molecule in a digital manner without the need for an external instrument or operator. Our assay measures titer by gradually immuno-depleting the target analyte from a flowing sample effectively diluting the residual target - a process conventionally achieved through serially diluting the whole sample in numerous, time-consuming pipetting steps performed manually. Furthermore, the execution of the depletion ELISA process is automated by a built-in flow controller which sequentially delivers different reagents with preset delays. We apply the technology to develop assays measuring (1) severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody titers (IgM/IgG antibodies to nucleocapsid and spike protein) and (2) troponin I, a cardiac biomarker.

Amitrole is a non-selective triazole herbicide that is widespread used to control a variety of weeds in agriculture, but it may pollute the environment and do harm to organisms. Thus, it is of critical significance to enlist a low-cost, sensitive, stable and renewable method to detect amitrole. In this paper, electrochemical experiments were carried out using carbon fibers/reduced graphene oxide/cellulose paper electrodes, which demonstrated good electrocatalytic performance for amitrole detection. The electrochemical process of amitrole on the surface of the reduced paper electrode was a quasi-reversible reaction controlled by diffusion. Cyclic voltammetry and the amperometric i - t curve method were used for amitrole determination at a micro molar level and higher-concentration range with the following characteristics: linear range 5 × 10 -6 mol L -1 to 3 × 10 -5 mol L -1 , detection limit 2.44 × 10 -7 mol L -1 . In addition, the relative standard deviation of repeatability is 3.74% and of stability is 4.68%. Plasmonic paper has received attention as a surface-enhanced Raman scattering (SERS) substrate due to several advantages. These include low cost, high rates of sample collection, high physical flexibility and relative ease of fabrication. Here, we report the development of a plasmonic paper based on a combination of graphene oxide (GO) and gold nanorods (AuNRs) on cellulose substrates using a simple layer-by-layer immersion method. Rhodamine 6G (R6G) was selected as the probe molecule to determine the SERS efficiency. The GO–AuNR plasmonic paper composite demonstrated superior signal intensity compared to that of the paper substrate coated only with AuNRs. The signal enhancement was due to synergism between the electromagnetic enhancement (EM) originating from the AuNRs and the chemical enhancement effect (CE) originating from GO. To optimize the fabrication process, the first step was accomplished by varying the GO concentration during the paper pretreatment step. This was followed by determining the number of immersion cycles required for optimal absorption of AuNRs. The highest SERS signals derived from R6G molecules indicated that the 2 mg mL−1 GO pretreatment condition followed by two immersion cycles into the AuNR solution proved to be the best condition for plasmonic paper fabrication. The enhancement factor (EF) of the plasmonic paper was calculated to be 4.56 × 107 and the limit of detection (LOD) for R6G analysis was as low as 0.1 nM. Moreover, the plasmonic paper was tested in the detection of Mitoxantrone (MTX), an anti-cancer drug. We found that the LOD for detection of MTX was on the order of 5 μM. Here, we introduce the synthesis and deposition of organic/inorganic composite ink on cellulose paper using a rapid ultrasonic spray deposition approach that can be incorporated as a counter electrode (CE) in flexible dye-sensitized solar cells (FDSSCs). The composite ink comprised a copper indium sulfide (CuInS 2 ) nanostructure ink and dispersion of poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS) in water. Fabricated counter electrodes are biodegradable, environment-friendly, flexible, and economical and meet the requirements for sustainable green energy. To evaluate the catalytic activities and power conversion efficiencies of DSSCs, the produced CuInS 2 /PEDOT:PSS composite ink-based CEs were compared with PEDOT:PSS counter electrodes. Cyclic voltammetry studies found that CuInS 2 /PEDOT:PSS had a greater cathodic charge transfer current density ( J c ) (-1.23 mA cm -2 ). Moreover, it was found that the potential separation values are small, which indicate a stronger catalytic activity than PEDOT:PSS counter electrodes. The observed exchange current density ( J 0 ) was 3.98 mA cm -2 , while the limiting current density ( J lim ) increased to 45.7 mA cm -2 /PEDOT:PSS FDSSC composed of cellulose paper was 1.06%.2 /PEDOT:PSS CE. The photovoltaic performances of CuInS 2 /PEDOT:PSS and PEDOT: PSS-based DSSC's were measured and determined to be 5.66% and 4.41%, respectively, while the performance of CuInS 2 /PEDOT:PSS FDSSC composed of cellulose paper was 1.06%.

Rapid and accurate identification of bacteria is essential to ensure food safety and prevent pathogenic bacterial infection. In this study, a highly efficient method was established for accurately identifying bacterial species by applying Ag NP-filter paper based Surface enhanced Raman spectroscopy (SERS) analysis and Partial Least Squares-Discriminant Analysis (PLS-DA) statistical methods. The flexible Ag NP filter paper substrate with high sensitivity and uniformity was prepared by a facile and low-cost silver mirror reaction at room temperature, which exhibited desirable SERS activity in bacteria detection. Furthermore, PLS-DA was successfully employed to distinguish SERS spectra from S. aureus CMCC 26003, E. faecalis ATCC29212 and L. monocytogenes ATCC 19115 with a sensitivity of 93.3-100%, specificity of 96.7-97%, and overall predicting accuracy of 95.8%. Identification of specific antibodies in patient plasma is an essential part of many diagnostic procedures and is critical for safe blood transfusion. Current techniques require laboratory infrastructure and long turnaround times which limits access to those nearby tertiary healthcare providers. Addressing this challenge, a novel and rapid paper-based antibody test is reported. We validate antibody detection with reverse blood typing using IgM antibodies and then generalise the validity by adapting to detect SARS CoV-2 (COVID-19) antibodies in patient serum samples. Reagent red blood cells (RBC) are first combined with the patient plasma containing the screened antibody and a droplet of the mixture is then deposited onto paper. The light intensity profile is analyzed to identify test results, which can be detected by eye and/or with image processing to allow full automation. The efficacy of this test to perform reverse blood typing is demonstrated and the performance and sensitivity of this test using different paper types and RBC reagents was investigated using clinical samples. As an example of the flexibility of this approach, we labeled the RBC reagent with an antibody–peptide conjugate to detect SARS CoV-2 (COVID-19) antibodies in patient serum samples. The design and preparation of a reduced graphene oxide (rGO) paper electrode with a reinforced concrete structure is an effective strategy to improve the mechanical properties of flexible electrodes. Therefore, PEDOT:PSS is added as a binder, toughening and conductive agent. CNTs, with good mechanical properties, are added to enhance the strength. Fe2O3 nanoparticles are added to increase the energy storage. According to these assumptions, we successfully prepared a flexible hydrophilic paper electrode with rGO nanosheets, PEDOT:PSS, CNTs and Fe2O3 nanoparticles by a constant temperature vacuum drying method. In addition, the assembled symmetrical supercapacitor exhibits a high capacitance of 1924 mF cm−3 at 1 mA cm−3, an excellent energy density of 35.6 W h kg−1 (at 166 W kg−1) and a remarkable cycle stability of 98.2% retention over 10 000 cycles at 1 mA cm−2. The design of the three-dimensional (3D) porous and defective molecular structure of nanocarbons can potentially enhance their functionalities for advanced electronic applications by providing a large working volume, rapid transport of electrolytes and reactants, and active sites for electrochemical reactions. Here, we report successful fabrication of 3D porous and defective nanocarbons by direct pyrolysis of crab shell-derived chitin nanofiber paper. The pyrolyzed chitin nanofiber paper has tunable electrical resistivity (1013 to 10−2 Ω cm), 3D porous structures with layered nanofiber networks, and defective nitrogen-doped carbon molecular structures. The pyrolyzed chitin nanofiber paper shows good performance as a photosensor and an energy-storage supercapacitor electrode, proving that the tailored 3D porous and defective carbon structures play key roles in the drastic improvement of the performance. This pioneering approach can be used for fabrication of chitin nanofiber-derived nanocarbons with excellent multiple functions for future sustainable electronics. [ABSTRACT FROM AUTHOR]

The analytical performance for paper spray (PS) using a new insert sample approach based on paper with paraffin barriers (PS-PB) is presented. The paraffin barrier is made using a simple, fast and cheap method based on the stamping of paraffin onto a paper surface. Typical operation conditions of paper spray such as the solvent volume applied on the paper surface, and the paper substrate type are evaluated. A paper substrate with paraffin barriers shows better performance on analysis of a range of typical analytes when compared to the conventional PS-MS using normal paper (PS-NP) and PS-MS using paper with two rounded corners (PS-RC). PS-PB was applied to detect sugars and their inhibitors in sugarcane bagasse liquors from a second generation ethanol process. Moreover, the PS-PB proved to be excellent, showing results for the quantification of glucose in hydrolysis liquors with excellent linearity (R(2) = 0.99), limits of detection (2.77 mmol L(-1)) and quantification (9.27 mmol L(-1)). The results are better than for PS-NP and PS-RC. The PS-PB was also excellent in performance when compared with the HPLC-UV method for glucose quantification on hydrolysis of liquor samples.

This work demonstrated a quantitative determination of Cu and Zn in tree rings using laser ablation-ICP-MS and the use of quantitative filter paper as a solid support for constructing the calibration curve using aqueous standards. After cleaning the filter paper using 10% v/v HNO3 for 30 min and drying it, 4 μL of standards is delivered on the filter surface and the results are attained with no normalized data. The analytical curves ranged from 0.5 to 10.0 μg g−1 and 5.0 to 100.0 μg g−1 for Cu and Zn, respectively, and the accuracy of the results was checked not only through Certified Reference Materials (NIST 1573a, tomato leaves and 1575a, pine needles) but also using ICP-MS (Inductively Coupled Plasma Mass Spectrometry) as a reference technique. Then, using such minimalist strategies, this method was applied for evaluating the mass fractions of Zn and Cu directly in tree rings. Three parallel ablation lines were drawn with a 220 μm distance between each one, with the same size of the tree ring samples. The average mass fractions for each tree ring and between the 3 ablated lines were at most ca. 18 and 13 μg g−1 for Cu and Zn, respectively, in Hymenaea courbaril, ca. 4 and 9 μg g−1 for Cu and Zn, respectively, in Tipuana tipu, and ca. 160 μg g−1 and

Solar-driven evaporators (SDEs) have attracted increasing attention in producing potable freshwater. Developing a multitasking SDE with stable self-floating performance, long-term salt resistance, effective heat management, pollutant degradation capability and good scalability for industrial production is significant but challenging. This work demonstrates the fabrication of a cellulose paper-based SDE with rough photothermal coating, controlled Janus wettability and a hierarchical Fenton catalyst for advanced solar desalination. Through the surface deposition of an urchin-like nanocatalyst and polypyrrole coating, a rough photothermal top layer for enhanced light trapping (98.6%) and a Fenton catalytic bottom layer for in situ pollutant degradation (98.1%) are constructed. Benzophenone-terminated polydimethylsiloxane is designed to realize facile asymmetric photo-grafting on porous paper, which further endows the evaporator with controlled Janus wettability, stable self-floating performance, long-term salt resistance, efficient heat management and rapid water transfer. Benefiting from such advantages, high solar-to-vapor efficiency (95.3%) and evaporation rate (1.88 kg m−2 h−1) are obtained. This cellulose paper-based evaporator also exhibits mechanical flexibility, portability and cost-effectiveness. Furthermore, the preparation process, including simple deposition and cleaner photo-grafting, is feasible and scalable for industrial production. The use of synthetic stimulants, including designer cathinones, remains a significant concern worldwide. Thus, the detection and identification of synthetic cathinones in biological matrices is of paramount importance for clinical and forensic laboratories. In this study, distribution of mephedrone and its metabolites was investigated in fingerprints. Following a controlled human mephedrone administration (100 mg nasally insufflated), two mass spectrometry-based methods for fingerprint analysis have been evaluated. The samples deposited on triangular pieces of chromatography paper were directly analysed under ambient conditions by paper spray-mass spectrometry (PS-MS) while those deposited on glass cover slips were extracted and analysed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The LC-MS/MS method was 5-6 times more sensitive than PS-MS but required sample preparation and longer analysis time. Mephedrone was detected in 62% and in 38% of all post-administration samples analysed by LC-MS/MS and PS-MS, respectively. Nor-mephedrone was the only metabolite detected in 3.8% of all samples analysed by LC-MS/MS. A large inter- and intra-subject variation was observed for mephedrone which may be due to several factors, such as the applied finger pressure, angle and duration of contact with the deposition surface and inability to control the 'amount' of collected fingerprint deposits. Until these limitations are addressed, we suggest that the sole use of fingerprints can be a useful diagnostic tool in qualitative rather than quantitative analysis, and requires a confirmatory analysis in a different biological matrix.

A bio-inspired cellulose paper-poly(amidoxime) composite hydrogel is explored via UV-polymerization. This hydrogel has a highly efficient uranium capture capacity of up to 6.21 mg g -1 for W U /W dry gel and 12.9 mg g -1 for W U /W poly(amidoxime) in seawater for 6 weeks, due to its enhanced hydrophilicity, good hydraulic/ionic conductivity and broad-spectrum antibacterial performance.

Microorganisms such as bacteria, fungi, algae and moulds are highly proficient at colonizing artistic and architectural heritage. The irreparable damage they cause to unique artefacts results in immeasurable cultural and societal losses to our shared cultural heritage, which represent an important social and economic resource for Europe. With the overall aim of preventing fungal deterioration of paper artefacts, we report the use of magnesium oxide nanoparticles (MgO NPs) of average diameter 12 nm as potent antifungal agents against fungi commonly found colonising paper heritage: A. niger, C. cladosporioides and T. reesei. Dispersions of MgO NPs on original 18th century paper samples from the Archives of the Spanish Royal Botanic Garden were effective at preventing fungal colonisation without altering the appearance of the paper artefacts. Importantly, MgO NPs also inhibit cellulase activity in the filamentous fungi T. resei and A. niger, two of the principle biodeteriogens of cellulosic materials. In addition, our report provides three simple new procedures for studying the fungal colonisation prevention properties of nanomaterials on paper samples. Overall this opens the door to the use of colourless, low-cost, and scalable nanomaterials for preventing biodeterioration in cellulose-based artefacts.

SCN- is one of the most important anions in metabolic processes. However, the investigation of SCN- in living systems is restricted by the lack of stable functional molecular tools. Herein, the first fluorene-based polymer fluorescent probe V1 was synthesized through rational design. Compared with small molecule fluorescent probes, V1 exhibited excellent fluorescence stability in bovine serum albumin (BSA) solution. Furthermore, the V1-based paper sensor was highly selective toward SCN- in aqueous solution. Significantly, these merits of the probe V1 enable the detection of SCN- in different living cell lines and zebrafish.

Sodium thiocyanate (NaSCN) is a naturally antibacterial component in milk, but excessive consumption of thiocyanate may cause potential risks to human health. Currently available methods for the detection of thiocyanate have some disadvantages such as poor sensitivity and high price. Here, we report a robust and cost-effective method to detect NaSCN based on paper substrates deposited with in situ reduced Ag nanoparticles by surface-enhanced Raman spectroscopy (SERS). Densely packed multilayer AgNPs provide uniform narrow nanogaps, which exponentially enhance the Raman signals. Moreover, these homogeneous narrow hotspots ensure that this method has high sensitivity (the limit of detection is 10−12 mol L−1), a wide linear range (from 10−9 mol L−1 to 10−4 mol L−1), and remarkable reproducibility (the coefficient of variation within a SERS sensor is 6.5%). Spiked milk samples were detected and the recovery rates of NaSCN were in the range of 95.1%–108.0%.
