From the President of the Research Society on Alcoholism In recent years the alcohol research field has matured and is attracting a substantial number of eager and technically sophisticated researchers. There is a feeling of excitement and premonitions of breakthroughs as more and more of alcohol's actions are being detailed. I, however, have at times been sobered by the perception that the lure of parsimonious explanations and the appeal of studying easily demonstrable effects obscure the critical issues re­ garding alcohol abuse and alcoholism. Central questions regarding (1) the reinforcing properties of ethanol and other factors responsible for inappro­ priate consumption of alcohol, (2) the heterogenous characteristics (both bi­ ologic and sociologic) of the alcoholic population, and (3) the differential predisposition of individuals to alcohol-induced medical problems needed to be vigorously pursued. Researchers who used animals as models for the human condition needed to become more intimately aware of the variety of factors that are of importance in the development of alcoholism in an indi­ vidual. On the other hand, researchers studying humans needed to attempt to more clearly define and categorize diagnostic criteria and characteristics of various alcoholic populations. Such categorization and continued character­ ization of alcoholism allows not only for a framework of concepts within which proper animal models for studies of biologic mechanisms can be de­ veloped but also allows for consideration of the most apt treatment approaches.

Neurodegeneration is one of the most important subjects of the investigation now and in the coming 21st century. Alzheimer's disease is the leading cause of dementia in the elderly people and Parkinson's disease is one of the major neurologic disorders with the prevalence between 1 and 2/1 000 population in advanced countries. Many others are suffering from intractable neurodegener­ ative disorders such as amyotrophic lateral sclerosis, Huntington's disease, or spinocerebellar degeneration. No truly effective treatment is available for any of these neurodegenerative disorders except for Parkinson's disease; even in Parkinson's disease, still it is impossible to slow down the disease process with the currently available treatment. It is urgently needed to develop new effective technique to halt or slow down the disease process in each of those disorders. Recent advance in the molecular biological and molecular genetic technique has brought us great progress in the understanding of etiology and pathogenesis of these disorders, but still it is not known how neurons are going to die in these disorders. To explore the question, mutual cooperation and exchange of ideas between basic scientists and clinical peoples are of utmost importance.

High blood pressure disease is one of the most prevalent pathological conditions in modem society with potentially serious consequences. Neuronal nicotinic receptors are key molecules for signal transduction in a number of neuronal pathways. They are widely distributed in the brain and are known to be involved in cognitive tasks, including learning and memory, in smoking addiction and in several brain diseases, such as Alzheimer's and Parkinson's dementias, schizophrenia, and epilepsy. This book provides a comprehensive review of the field, starting with a historical perspective and dealing with the molecular structure of these receptors, their biophysical and pharmacological properties, their distribution in central and peripheral nervous systems, and their major involvement in brain functions. Particular emphasis is paid to drugs (both new and old) that are useful in the diagnosis and treatment of diseases involving neuronal nicotinic receptors. Finally, the relevance of these receptors in smoking addiction is carefully evaluated, together with future trends and the latest results.

The purpose of this book is to review our state of knowledge about the neurobehavioral and psychosocial processes involved in behavioral inhibitory processes and to provide an insight into how these basic research findings may be translated into the practice of drug abuse prevention interventions. Over the last decade, there has been a wealth of information indicating that substance use disorders do not simply reflect an exaggeration of reward seeking behavior, but that they also represent a dysfunction of behavioral inhibitory processes that are critical in exercising self-control. A number of studies have determined that individuals with substance use disorders have poor inhibitory control compared to non-abusing individuals. In addition, the fact that the adolescent period is often characterized by a lack of inhibitory control may be one important reason for the heightened vulnerability for the initiation of drug use during this time. Controlled experiments utilizing neuroscience techniques in laboratory animals or neuroimaging techniques in humans have revealed that individual differences in prefrontal cortical regions may underlie, at least in part, these differences in inhibitory control. Although a few excellent journal reviews have been published on the role of inhibitory deficits in drug abuse, there has been relatively little attention paid to the potential applications of this work for drug abuse prevention. The current book will provide both basic and applied researchers with an overview of this important health-relevant topic. Since translational research cuts across multiple disciplines and most readers are not familiar with all of these disciplines, the reading level will be geared to be accessible to graduate students, as well as to faculty and researchers in the field. The book will be organized around three general themes, encased within introductory and concluding chapters. The first theme will review basic neurobehavioral research findings on inhibition and drug abuse. Chapters in this theme will emphasize laboratory studies using human volunteers or laboratory animals that document the latest research implicating a relation between inhibition and drug abuse at both the neural and behavioral levels of analysis. The second theme will move the topic to at-risk populations that have impulse control problems, including children, adolescents and young adults. The third theme will concentrate on prevention science as it relates to inhibitory control. Astrocytes were the original neuroglia that Ramón y Cajal visualized in 1913 using a gold sublimate stain. This stain targeted intermediate filaments that we now know consist mainly of glial fibrillary acidic protein, a protein used today as an astrocytic marker. Cajal described the morphological diversity of these cells with some ast- cytes surrounding neurons, while the others are intimately associated with vasculature. We start the book by discussing the heterogeneity of astrocytes using contemporary tools and by calling into question the assumption by classical neuroscience that neurons and glia are derived from distinct pools of progenitor cells. Astrocytes have long been neglected as active participants in intercellular communication and information processing in the central nervous system, in part due to their lack of electrical excitability. The follow up chapters review the “nuts and bolts” of ast- cytic physiology; astrocytes possess a diverse assortment of ion channels, neu- transmitter receptors, and transport mechanisms that enable the astrocytes to respond to many of the same signals that act on neurons. Since astrocytes can detect chemical transmitters that are released from neurons and can release their own extracellular signals there is an increasing awareness that they play physiological roles in regulating neuronal activity and synaptic transmission. In addition to these physiological roles, it is becoming increasingly recognized that astrocytes play critical roles during pathophysiological states of the nervous system; these states include gliomas, Alexander disease, and epilepsy to mention a few.
