This book constitutes the refereed proceedings from the Second International Workshop on Foundation Models for General Medical AI, MedAGI 2024, held in conjunction with the 27th International conference on Medical Image Computing and Computer Assisted Intervention, MICCAI 2024, in Marrakesh, Morocco in October 2024. The 17 papers included in this book were carefully reviewed and selected from 26 submissions. These papers provide insights into the current landscape of medical AI and foundation models, that can pave the way for the evolution of task-specific medical AI systems into more generalized frameworks capable of tackling a diverse range of tasks, datasets, and domains.

This section is arranged in three parts. Part one begins with an overview of the retina and its relationship to the pigment epithelium, choroid, and vitreous, plus discusses the major retinal disorders, diagnostic methods, and treatment. Part two covers specific diseases and trauma to the posterior segment and provides appropriate diagnostics and reviews, manages, and provides therapies for vitreoretinal diseases. Part three reviews cryotherapy for posterior segment diseases and includes select therapeutic topics, plus laser therapy and vitreoretinal surgery. Gene therapy covers the common types of DNA therapy: gene augmentation, optogenetics, and genome editing are included.

For Augustine, that the Word became flesh transformed a merely human understanding of the virtues and grounds all virtue in humility. The Way of Humility: Augustine's Theology of Preaching explores how this truth became a new paradigm for understanding the scriptures and thus, how Augustine embodied the virtue in the preaching of the scriptures. One of Augustine's most devoted students, Possidius, said that anyone can learn from reading Augustine, but “those were able to profit still more who could hear him speak in church and see him with their own eyes. Truly, he was indeed one of those of whom it is written, ‘speak this way and act the same way.'” The Way of Humility searches for evidence of the virtue of humility in action through the preaching of the humble Word in the sermons of Augustine. Many know of Augustine through his more famous treatises but few have encountered the Doctor of Grace where he had his most immediate impact, preaching. The Way of Humility follows the sermons through several traditional theological loci, ecclesiology, Christology, soteriology to uncover what can be learned about Augustine's theology through the way he preached to a mixed audience of urbanites and rustics, many of whom did not have the benefit of a formal education. Throughout the book, we see the interplay between Augustine's action in speech and Augustine's more direct statements on his theology of Preaching. Through handing over Christ in his sermons, he became himself an example of humility for the congregation on their journey toward the final end for all people, the Beatific Vision.

Four diverse women entrepreneurs launched their ventures in a thriving entrepreneurial ecosystem that was part of a shift to a creative technology-driven economy for Miami. Although Miami was rated the #1 U.S. city for startups in 2017, the region contained structural barriers and cultural biases unfriendly to women and people of color, including lack of access to capital and relationships. The case highlights women founders'backgrounds and experiences with an ed-tech startup, a coding school and events for Black entrepreneurs; an incubator for green businesses with a Black leadership focus; and an accelerator for social impact ventures that also runs social media campaigns for problems such as climate change. The women CEOs reveal the barriers they faced, how they overcame them, and how they attempt to enrich the ecosystem for other women and people of color. This case raises the question of what must be in place for cities to take advantage of the innovation and job-creating potential of a wider population of entrepreneurs and gain the benefits of diversity, and for women founders to thrive.

Two sigma receptor subtypes have been proposed, sigma1 and 2. Much of our understanding of this system is based on biochemical and pharmacological characterization of the cloned sigma1 receptor subtype (Sigma1). It has become clear that sigma receptors are not canonical receptors. Sigma1 is highly conserved among mammalian species, however, it does not share significant homology with any other mammalian protein. Although a range of structurally diverse small molecules bind Sigma1 with high affinity, and it has been associated with a broad range of signaling systems, Sigma1 itself has no known signaling or enzymatic activity. The evolution of this field over nearly four decades has more recently led to a fundamental shift in the concept of “sigma receptors” to what may more accurately and generally be called sigma proteins. Largely based on traditional pharmacologic approaches, the Sigma1 protein has been associated with a broad range of signaling systems, including G-protein coupled receptors, NMDA receptors, and ion channels. Sigma proteins have been linked to a range of physiological processes, including intracellular calcium signaling, neuroprotection, learning, memory, and cognition. Emerging genetic, clinical, and mechanism focused molecular pharmacology data demonstrate the involvement of proteins in a range of pathophysiologies and disorders including neurodegenerative disease, pain, addiction, psychomotor stimulant abuse, and cancer. However, an understanding of the physiological role of sigma proteins has remained elusive. Emerging data associate Sigma1 with chaperone-like activities or molecular scaffold functions. This book aims to provide an updated perspective on this rapidly evolving field undergoing changes in fundamental concepts of key importance to the discipline of pharmacology. It focusses on the reported roles of sigma proteins in pathophysiology and on emergent therapeutic initiatives.
