Knowledge of the scale of dispersal and the mechanisms governing gene flow in marine environments remains fragmentary despite being essential for understanding evolution of marine biota and to design management plans. We use the limpets Patella ulyssiponensis and Patella rustica as models for identifying factors affecting gene flow in marine organisms across the North-East Atlantic and the Mediterranean Sea. A set of allozyme loci and a fragment of the mitochondrial gene cytochrome C oxidase subunit I were screened for genetic variation through starch gel electrophoresis and DNA sequencing, respectively. An approach combining clustering algorithms with clinal analyses was used to test for the existence of barriers to gene flow and estimate their geographic location and abruptness. Sharp breaks in the genetic composition of individuals were observed in the transitions between the Atlantic and the Mediterranean and across southern Italian shores. An additional break within the Atlantic cluster separates samples from the Alboran Sea and Atlantic African shores from those of the Iberian Atlantic shores. The geographic congruence of the genetic breaks detected in these two limpet species strongly supports the existence of transpecific barriers to gene flow in the Mediterranean Sea and Northeastern Atlantic. The aim of the present work was to identify cryptic species in the Anopheles maculipennis and Culex pipiens complexes and to study the genetic structure of the dominant mosquito species Ochlerotatus caspius (Diptera: Culicidae) in the Province of Alessandria close to the vast area untreated rice fields in Piedmont, NW Italy. With the help of PCR-RFLP analysis, four members of the Anopheles maculipennis complex were identified: A. messeae, A. maculipennis, A. sacharovi and A. atroparvus. Only C. pipiens f. molestus was identified in 11 habitats studied in Piedmont. Partial sequences of the cytochrome c oxidase subunit 1 (COI) mitochondrial gene and the second internal transcribed spacer (ITS2) of nuclear ribosomal RNA genes for Italian O. caspius are reported here for the first time. The results indicate that this species diverged from Iranian representatives of this species about one million years ago. The present study examines the population genetic structure of fifty-nine specimens of Glis glis (Linneaus, 1766) from thirteen localities in central Europe, sequencing a 400-bp segment of the mitochondrial cytochrome b (cyt b) gene and a 673-bp segment of the cytochrome c oxidase subunit I (COI) gene. The consensus tree obtained from Bayesian analysis revealed a robust dichotomy, showing two sister groups: one clade includes samples from a wide geographical area, extending from north-central Europe to northern Italy (major branch sensu Bilton), and the other comprises samples collected in central and southern Italy and in Sicily (Italian branch). According to the Tajima-Nei model, the two phylogroups were separated by a sequence divergence of 0.8% (cyt b) - 2.6% (COI), showing the COI gene to be more informative than cyt b. On a smaller geographical scale, the Italian clade was further substructured, displaying geographical differentiation along the Peninsula. Abstract: In this work, we present molecular barcoding results obtained in 69 processed fish products belonging to 27 teleost species traded in Italian commercial markets during 2008. DNA barcoding using direct sequencing of about 900bp of mitochondrial genes cytochrome oxidase subunit I (COI) and cytochrome b (Cyt b) revealed uncorrect labelling in 22 samples (32%). Among substituted species, 18 (26%) were serious frauds under both economic and nutritional points of view. In some cases, frauds concerned species of major conservation regard and reported in the IUCN and CITES directories. Results add further concern on the trading of processed fish products in Italy from both health and conservation points of view. [Copyright &y& Elsevier]

Pelobates fuscus is a fossorial amphibian that inhabits much of the European plain areas. To unveil traces of expansion and contraction events of the species’ range, we sequenced 702 bp of the mitochondrial cytochrome b gene. To infer the population history we applied phylogeographical methods, such as nested clade phylogeographical analysis (NCPA), and used summary statistics to analyse population structure under a neutral model of evolution. Populations were assigned to different drainage systems and we tested hypotheses of explicit refugial models using information from analysis of molecular variance, nucleotide diversity, effective population size estimation, NCPA, mismatch distribution and Bayesian dating. Coalescent simulations were used as post hoc tests for plausibility of derived or a priori assumed biogeographical hypotheses. Our combination of all approaches enabled the reconstruction of the colonization history and phylogeography of P. fuscus and confirmed a previous assumption of the existence of two major genetic lineages within P. fuscus. Using the Afro-European vicariance of Pelobates cultripes and Pelobates varaldii and applying Bayesian dating we estimated the divergence of these phylogeographical lineages to the Pliocene. We suggest the existence of three different glacial refugia: (i) the area between the Caspian and Black Seas as the origin for the expansion of the ‘eastern lineage’; (ii) the Danube system as a centre of diversity for part of the ‘western lineage’; (iii) the Po Valley, the largest centre of genetic variability. This fits the hypothesis that climatic fluctuation was a key event for differentiation processes in P. fuscus. [ABSTRACT FROM AUTHOR]
