Toxoplasma gondiiinfections are common in humans and animals worldwide. Ingestion of raw or undercooked meat containing tissue cysts of T. gondiiis one major source of transmission of this parasite. It is important to guarantee the meat quality of China since our pork industry produces about half of the world's pork. In this study, a total of 746 pig samples were collected from Zhejiang and Jiangsu provinces in eastern China, and examined for T. gondiiinfection by PCR amplification targeting B1gene. In this study, we found that 57 of 746 (7.6%) pigs were positive for B1gene, with 8.5% (48/562) in Zhejiang province and 4.9% (9/184) in Jiangsu province, respectively. The positive DNA samples were further genotyped at 11 genetic markers, including SAG1, 5′-and 3′-SAG2, alternative SAG2, SAG3, BTUB, GRA6, L358, PK1, c22-8, c29-2, and an apicoplast locus Apico through PCR–restriction fragment length polymorphism (PCR-RFLP) technology. Two genotypes (ToxoDB 9 and ToxoDB 10) of T. gondiiwere identified by PCR-RFLP in Zhejiang province. However, both genotypes were not determined from Jiangsu province, which is speculated on the low DNA concentration and the small number of samples. These results indicate that T. gondiiinfection is endemic in pigs in eastern China and may raise public food safety concerns, suggesting more interventions for T. gondii-related risks are needed in the future.

Solar desalination is expected to solve the problem of global water shortage. Yet its stability is plagued by salt accumulation. Here, a paper-based thermal radiation-enabled evaporation system (TREES) is demonstrated to achieve sustainable and highly efficient salt-collecting desalination, featuring a dynamic evaporation front based on the accumulated salt layer where water serves as its own absorber via energy down-conversion. When processing 7 wt % brine, it continuously evaporates water at a high rate─2.25 L m–2h–1under 1 sun illumination─which is well beyond the input solar energy limit for over 366 h. It is revealed that such enhanced evaporation arises from the unique vertical evaporation wall of the paper-TREES, which captures the thermal energy from the heated bottom efficiently and gains extra energy from the warmer environment. These findings provide novel insights into the design of next-generation salt-harvesting solar evaporators and take a step further to advance their applications in green desalination.

How metal-reducing bacteria transfer electrons during dissimilatory energy generation under electron acceptor-limited conditions is poorly understood. Here, we incubated the iron and manganese-reducing bacterium Shewanella oneidensisMR-1 without electron acceptors. Removal of soluble extracellular organic compounds (EOCs) dramatically retarded transfer of electrons to an experimental electron acceptor, Cr(VI), by MR-1. However, the return of either high MW (>3000 Da) or low MW (<3000 Da) soluble EOCs produced by MR-1 to washed cells restored Cr(VI) reduction though Cr(VI) reduction was fastest when both size fractions were added together. Spectral and electrochemical characterization of EOCs indicated the presence of flavins and c-type cytochromes (c-Cyts). A model of the kinetics of individual elementary reactions between cells, flavins, released c-Cyts, and Cr(VI), including the direct reduction of flavins, released c-Cyts, and Cr(VI) by cells and the indirect reduction of Cr(VI) by reduced forms of flavins and released c-Cyts, was developed. Model results suggest that released c-Cyts could act as electron mediators to accelerate electron transfer from cells to Cr(VI), and the relative contribution of this pathway was higher than that mediated by flavins. Hence, extracellular c-Cyts produced by MR-1 likely play a role in extracellular electron transfer under electron acceptor-limited conditions. These findings provide new insights into extracellular electron shuttling and the metabolic strategy of metal-reducing bacteria under electron acceptor-limited conditions.

Based on a novel umpolung strategy, an efficient and highly enantioselective cascade aldol/cyclization/tautomerization of the 2-(2-oxoindolin-3-yl)malononitrile to active carbonyl compounds with excellent diastereo- and enantioselectivity has been developed. Also, various enantio-enriched multifunctional dispiro[2-amino-4,5-dihydrofuran-3-carbonitrile]bisoxindoles with adjacent spiro-stereocenters were conveniently obtained by this novel methodology. Also, the dispiro[2-amino-4,5-dihydrofuran-3-carbonitrile]bisoxindoles were easily transformed into structurally complex molecules without any effect on the diastereo- and enantioselectivity.

In this study, density functional theory (DFT) was used to calculate second-order polarizabilities and second-order polarizabilities densities of a series of organic substitution for Lindqvist-type polyoxometalates (POMs), and the nonlinear optical (NLO) properties was also analyzed. We found that βzzz has the main contribution to β value. The expansion of molecular structure on z-axis greatly increased second-order polarizabilities. Both the size of the organic segments and metal hybridization exert an influence on β value. The analysis on the second-order polarizabilities density is used to explain the NLO phenomenon. In the present investigation, metal hybridization and π-conjugation changed the contribution of βzzz value from different parts. The results of this work will contribute to the potential applications in high-performance NLO materials.

Increased aerobic glycolysis is a hallmark of cancer metabolism. How cancer cells coordinate glucose metabolism with extracellular glucose levels remains largely unknown. Here, we report that coactivator-associated arginine methyltransferase 1 (CARM1 or PRMT4) signals glucose availability to glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and suppresses glycolysis in liver cancer cells. CARM1 methylates GAPDH at arginine 234 (R234), inhibiting its catalytic activity. Glucose starvation leads to CARM1 upregulation, further inducing R234 hypermethylation and GAPDH inhibition. The re-expression of wild-type GAPDH, but not of its methylation-mimetic mutant, sustains glycolytic levels. CARM1 inhibition increases glycolytic flux and glycolysis. R234 methylation delays tumor cell proliferation in vitroand in vivo. Compared with normal tissues, R234 is hypomethylated in malignant clinical hepatocellular carcinoma samples. Notably, R234 methylation positively correlates with CARM1 expression in these liver cancer samples. Our findings thus reveal that CARM1-mediated GAPDH methylation is a key regulatory mechanism of glucose metabolism in liver cancer.

A simulation is carried out for the pressure fluctuation driven by the water hammer, based on a joint use of the onedimensional method of characteristics (MOC) and the three-dimensional finite volume method (FVM). The three-dimensional visualization of the cavitation induced by the water hammer is implemented, and the temporal and spatial analyses of extreme regions are made. A practical case of the water hammer, with the minimum boundary pressure higher than the saturated vapor pressure condition, is simulated. The simulation prediction that the cavitation would occur in the front of the gasket could serve some guideline for the optimization of industrial designs.

Associative memory, considered as the ability to link unrelated objects, is the most appealing behaviour found in both biological and artificial neural networks. Here we for the first time proposed a three terminal phase-change based nanoscale device where associative memory can be theoretically achieved according to the developed electro-thermal model. Compared with the conventional neural networks circuit, the designed device allows for an exemption from a large number of circuit elements, and thus exhibits an exciting potential for ultra-high integration density and super-fast response speed. The simulation results opens up a novel route to imitate the complex mechanism of the biological neural networks, thus rendering phase-change materials a promising contender for the key components in next-generation neuromorphic system.

Twenty one phytoplankton samples containing chrysophytes were collected in July 2009 and August 2011 in Daxinganling Mountains and Wudalianchi Lake Regions located in Northeast China. Forty two species and two varieties of silica-scaled chrysophytes have been identified by means of transmission electron microscopy, nine of them belonging to Paraphysomonadaceae and thirty-five to Synurales. Chrysosphaerella longispina, Spiniferomonas abei, Paraphysomonas eiffelii, Mallomonas maculata, M. lelymene, M. pumilio, M. eoa, M. torquata, Tesselaria lapponica, Synura bjoerkiiare reported for the first time in China.Chrysosphaerella longispina, Mallomonas punctifera, M. multiunca, M. torquata, Tesselaria lapponica, Synura bjoerkiiare distributed in the northern temperature region. The geographical distribution and ecological characteristics of the silica-scaled chrysophytes collected from these regions are discussed.

Memristor, as the fourth fundamental circuit element, has recently attracted considerable attention due to its widespread applications from programmable logic to neural networks. Today, the most representative memristor consists of a thin film of titanium oxide sandwiched by two electrodes. However, such a memristor gives rise to a relatively low switching speed, discouraging it from receiving more research enthusiasm. To overcome the disadvantages of the already established memristors (e.g. low switching speed and poor endurance) and thus to optimize the memristor performance, we proposed a novel memristor concept using chalcogenide alloy that has been widely used for nonvolatile memory storage. According to the developed electro-thermal model, the designed memristor using chalcogenide alloy clearly shows a variation of resistance along with the history of current as well as a fast switching speed and ultra-low energy consumption.

Singer PDC cutters of five different cone top diameters are designed and manufactured.On the basis of rock-breaking test with stinger PDC cutters, effects of significant factors on cutting forces are discussed, including cutting angle, cutter shape and rock hardness. The test results show that the stinger PDC cutters have the ability of drilling hard rock.The sharper the stinger PDC cutter is, the stronger the ability of drilling the formation will become.15°is an optimal front rake angle.

In order to increase calculation accuracy of CNC system reliability, this paper proposed a maximum likelihood parameter estimation method based on improved genetic algorithm. In the parameter estimation process for CNC system reliability distribution model, the maximum likelihood function value was gained by improving genetic algorithm through simulated annealing algorithm. Parameter estimation was carried out by setting Weibull distribution as an example. The result shows that the improved genetic algorithm can increase solution efficiency and convergence rate. Besides, it can effectively estimate parameters of reliability distribution model.

The wide application of Information and Communication Technologies offers great opportunities for the innovation of education, especially in universities. There are varieties of resources on the Internet, which teachers can use for their teaching, assimilating into the curriculum and into teaching methods. Based on constructivism learning theory, this paper centers on the advantages of practicing the web-based instruction by comparison to the traditional one. The media centers, a web-based learning enviroment, has been set up for students to learn as a platform for their after-class review, for their individulized instruction, for their collaborative learning and for exchanging educational resources among teachers. The notes of the Internet-assisting instruction is also discussed to emphasize that the new instruction is a useful complement not replacement to conventional teaching. So it is pointed out that the Internet should become a necessary supplementary for teaching and learning in university as the input resources and the output tools.

We go deep into the internal structure of the Boolean functions values, and study the relationship of algebraic immunity and algebraic degree of Boolean functions with the Hamming weight with the diffusion included. Then we get some theorems which relevance together algebraic immunity, annihilators and algebraic degree of H Boolean functions by the e-derivative which is a part of the H Boolean function. Besides, we also get the results that algebraic immunity and algebraic degree of Boolean functions can be linked together by the e-derivative of H Boolean functions and so on.

The objective of the study was to optimize the conditions in a culture medium for the selenium yeild enriched by Saccharomyces sp. III using Plackett-Burman design and Box-Behnken design. The Plackett-Burman multifactorial design was first employed to screen the significant factors in the fermentation for the selenium yeild, and subsequent use of the response surface methodology was further optimized for the selenium yeild by Box-Behnken design. The important factors in the culture medium, identified by the initial screening method of Placket-Burman, were sodium selenite, glucose and the liquid volume. The optimal amounts for maximum selenium yeild were: sodium selenite 15.8 mg/L; glucose 40.2 g/L; the liquid volume 120 mL in 250 mL flask. Using this statistical experimental design, the selenium yeild under optimal condition reached about 1679.32 μg selenium /g dry cell.

This study investigated the influence of broadleaf and conifer vegetation on soil microbial communities in a distinct vertical distribution belt in Northeast China. Soil samples were taken at 0–5, 5–10 and 10–20 cm depths from four vegetation types at different altitudes, which were characterized by poplar (Populus davidiana) (1250–1300 m), poplar (P. davidiana) mixed with birch (Betula platyphylla) (1370–1550 m), birch (B. platyphylla) (1550–1720 m), and larch (Larix principis-rupprechtii) (1840–1890 m). Microbial biomass and community structure were determined using the fumigation-extraction method and phospholipid fatty acid (PLFA) analysis, and soil fungal community level physiological profiles (CLPP) were characterized using Biolog FF Microplates. It was found that soil properties, especially soil organic carbon and water content, contributed significantly to the variations in soil microbes. With increasing soil depth, the soil microbial biomass, fungal biomass, and fungal catabolic ability diminished; however, the ratio of fungi to bacteria increased. The fungal ratio was higher under larch forests compared to that under poplar, birch, and their mixed forests, although the soil microbial biomass was lower. The direct contribution of vegetation types to the soil microbial community variation was 12%. If the indirect contribution through soil organic carbon was included, variations in the vegetation type had substantial influences on soil microbial composition and diversity.

In this paper, we investigated the stochastic resonance (SR) phenomenon in a laser system with correlated pump noise and quantum noise. The signal-to-noise ratio (SNR) is calculated when a square sine pulse signal is added to the system. The effects of the duty cycle of pulse signal and the correlation strength of noises on the SNR are discussed. Some valuable phenomena are investigated to improve the output SNR of laser.

The effects of killing Chlamydomonas, Microcystis Aeruginosa, Cylindrospermopsis, Chlorella and Tetraedron by chlorine dioxide were studied. The reaction orders and rate constants of algae oxidated by chlorine dioxide were determined. The results show that these algae can be killed effectively by chlorine dioxide in raw water. the efficiencies of killing Microcystis Aeruginosa, Tetraedron and Cylindrospermopsis are 89.44%～92.48% during 10 to 15 Minutes. The efficiency of killing Chlamydomonas is high to 98.26%, but the efficiency of killing Chlorella is low, only 60.90%. The reaction orders for various algae are different. The reaction orders of Chlamydomonas, Microcystis Aeruginosa, Cylindrospermopsis, Chlorella and Tetraedron are 1.3, 2, 2.5, 6 and 2, respectively. The apparent rate constants are 2.1×10-3 μg-0.3L 0.3s-1, 5.6×10-3μg -1Ls -1, 8.3×10-4μg -1.5L 1.5s -1, 1.0×10-5μg -5 L 5s -1 and 3.8×10-3 μg -1Ls -1 , respectively.

In order to hydrolyze corn gluten meal efficiently, combinations of two or three out of three hydrolysis enzymes (protex6L, protex7L and papain) were used and the process parameters were optimized in this paper. The degree of hydrolysis of corn gluten meal was assayed by using pH-stat method. The ratio of enzymes was optimized and the effect of multi-enzyme hydrolysis was compared with that of single-enzyme hydrolysis. The result showed that the optimal ratio of protex6L and protex7L was 3:1, protex6L and papain was 6:5, protex7L and papain was 1:1, and the optimal ratio of protex6L, protex7L and papain was 4:1:1.The degree of hydrolysis and soluble protein content of multi-enzyme hydrolysis and single-enzyme hydrolysis were approximately equal, but multi-enzyme hydrolysis was timesaving.

Corn residue is used as raw material, and through the study of corn juice processing, we can optimize the machining process, reduce processing costs, increase economic efficiency. It is found through the test that the optimum corn processing time for the precooking is 1h, liquefaction enzyme dosage is 30 activity unit every gram, reaction time is 1.5h, glucoamylase ratio is 120 activity unit every gram, saccharification time is 1.5h, 40MPa homogeneous for two times, sterilization at 135 ℃ ultra high temperature for 10s. Under these conditions, the production of maize corn residue has high sugar content with a small amount of enzyme dosage, the production cost is reduced, the product stability is greatly enhanced, and the shelf life is effectively extended.

For successful conservation and restoration of biodiversity, it is important to understand how diversity is regulated. In the ecological research community, a current topic of interest is how much of the variation in plant species richness and composition is explained by environmental variation (niche-based model), relative to spatial processes (neutral theory). The Yellow River Estuary (YRE) is a newly formed and fragile wetland ecosystem influenced by both the Yellow River and Bohai Bay. Here, we applied variance partitioning techniques to assess the relative effects of spatial and environmental variables on species richness and composition in the YRE. We also conducted a species indicator analysis to identify characteristic species for three subestuaries within the YRE. Partial redundancy analysis showed that the variations in species richness and composition were explained by both environmental and spatial factors. The majority of explained variation in species richness and composition was attributable to local environmental factors. Among the environmental variables, soil salinity made the greatest contribution to species abundance and composition. Soil salinity was the most important factor in the Diaokou subestuary, while soil moisture was the most important factor influencing species richness in the Qingshui and Chahe subestuaries. The combined effects of soil salinity and moisture determined species richness and composition in the wetlands. These results increase our understanding of the organization and assembly of estuarine plant communities.

In this study, flavor nucleotides were extracted from beer yeast paste as flavor enhancer for soy sauce. RNA was extracted by concentrated saline method with single factor experiments and L9(34) matrix experiments, and then hydrolyzed by 5’-phosphodiesterase which was obtained from malt rootlets to gain 5’-nucleotides as flavor enhancer. Results showed that the optimal extraction conditions were as follows：12% yeast concentration, 100℃ of extraction temperature, 10% of NaCl concentration and 4h of extraction time and the yield rate of RNA was 6.83%. The yield of 5’-nucleotide was 7.528mg/mL which was higher than other methods. When 5’-nucleotide was added to soy fermentation solution at the ratio of 2.5:1(v:v), taste of the new soy sauce products was much excellent.

This first survey in 2009 and 2010 on silica-scaled Chrysophytes in Zhenbao Island Wetland Preserve, Northeast China, recorded 32 taxa (5 in Chrysophyceae and 27 in Synurophyceae), of which 29 were previously found in other parts of China, three species, Spiniferomonas serrata, Mallomonas transsylvanica and Synura punctulosa are new records to China. In terms of biogeography, 1 taxon is endemic, 2 bipolar, 1 northern temperate, 9 cosmopolitan, and 18 of wide distribution. The endemism of Synura punctulosa, appears due to its poor dispersal ability; lack of sampling data may also contribute its known distribution range. Spiniferomonas serrata and Mallomonas transsylvanica, two bipolar taxa, are the first report in China. More research efforts are needed for adequately understanding the distributions of this group of algae.

Influence of frequency on the magnetic induction effect of Fe73.5Cu1Nb3Si13.5B9 amorphous ribbons by mutual induction was studied.The results showed that the magnetic induction effect increased with the higher frequency; with the increased of frequency,after a bigger rangeability of magnetic induction effect,there was a smaller one.

Objective to establish a system to evaluate the mental workload on visual display interface in airplane cockpit after synthesizing three kinds of mental workload assessment techniques, i.e. main task measure method, physiology measure method and subjective evaluate method. Methods The evaluation indices of every kind of assessment techniques was defined, and the amount of each index through experiment and subjective inquiry was gained, weighted average was calculated and a quantification evaluation value of each kind of evaluation methods was got. At last, with weighted calculation of above three evaluation values, the evaluation value of mental workload on visual display interface in airplane cockpit was obtained Results in M kinds of different designs of visual display interface in airplane cockpit, the synthetic evaluation value of one design was the smallest and the mental workload of this design was the smallest. The design of the smallest mental workload was chosen prior as the final scheme of visual display interface in airplane cockpit .Conclusion The evaluation system is combined with present main evaluation methods of mental workload and utilizes their advantages. The evaluating results can be quantified more directly and clearly. With this evaluation system, the mental workload can be evaluated during the design of visual display interface of airplane cockpit and the best scheme can be confirmed.

Biosorptions of Cr (VI) by the floc-type biosorbent AR (with glycocalyx) and AR (without glycocalyx) were compared. The initial pH was important for biosorption, and the optimum effect on Cr (VI) biosorption was acquired within initial pH 0.5-1.0. With the pH increasing, the effect on Cr(VI) biosorption was reduced, and when the initial pH exceeded 3.0, the effect on Cr (VI) biosorption was in an equilibrium of low levels. Biosorbing Cr (VI) of different concentrations, the maximal absorbing capacity of AR (with glycocalyx) and AR (without glycocalyx) was 187.36 mg•g-1, 56.1 mg•g-1 respectively, and the glycocalyx adhering on the cell improved the absorption obviously. The absorbed Cr (VI) could be recovered by desorption, and the desorption rate of AR (with glycocalyx) and AR (without glycocalyx) reached 67.9%and 62.3% separately. The two biosorption processes conformed Langmuir and Freundlich isotherm adsorption equations, so we could presume that the progress of biosorptions was reversible adsorption and irreversible adsorption simultaneously, however the reversible adsorption was more important in the biosorption and the adsorption belonged to physical adsorption. Besides Agrobacterium sp. fermentation broth could be used as bioflocculant, Agrobacterium sp. could be applied as biosorbent, and the glycocalyx improved the absorption obviously, then both the bioflocculant and biosorbent were promising biological products

Objective To study expert and novice eye movement pattern during simulated landing flight for providing references to evaluate flight performance and training of pilots. Methods The subjects were divided in to two group s of expert and novice according to their flight simulation experience. Eye movement data were recorded when they were performing landing task. Comparison of expert and novice flight performance data and eye movement data was made. Results It was found that the differences between expert and novice lay not only in flight performance but also in eye movement pattern. Performance of expert was better than novice. Expert had shorter fixation time, more fixation points, faster scan velocity, greater scan frequency and wider scan area than novice. It was also found that eye movement pattern of expert bring lower mental workload than novice. Conclusion Flight performance is related to eye movement pattern. Effective eye movement pattern is related to good flight performance. The analysis of eye movement indices can evaluate pilots’ flight performance and provide reference for flight training.

In order to evaluate pilot performance objectively, back propagation (BP) neural network model of 621423 form in topology with eye movement data was established. Data source of BP neural networks that came from former experiment and random interpolation was divided into training set and test set and normalized. Based on neural networks toolbox in Matlab, hidden layer nodes of BP networks were determined with empirical formula and experimental comparison ; BP algorithms in the toolbox were optimized; The training set data and test data were input into model for training and simulation; Pilot performance of the three skill levels was predicated and evaluated. The research shows that pilot performance can be accurately evaluated by setting up BP neural networks model with eye movement data and the evaluation method can provide a reference for flight training.

An AC servo driver speed adjusting system based on space vector pulse width modulation (SVPWM) is put forward. The principle of SVPWM and its software algorithm based on digital signal processor (DSP) are introduced in detail, and the structure of the system is designed. The simulation model of the system is built in Matlab, the simulation research provides theoretical basis for AC servo driver control performance.

Through collecting and analyzing thermal data from on-site testing of more than 30 cement plants, combining with design indicators of production line and control parameters in practical operation, the paper has summarized 4 first level, 5 second level and 133 third level energy efficiency evaluation indicators of cement enterprises after an in-depth study on the main factors influencing coal and power consumption of cement plants. On this basis, an energy efficiency evaluation system of new suspension preheater cement enterprise is to be established in China. It is expected to provide cement enterprise the basis of mastering the level of energy management and checking questions of energy utilization, to play guiding functions in settlement of energy-saving measures, development of energy-saving potential and reduction of energy consumption.

With the development of aviation equipment automation, the research on situation awareness (SA) of pilots has become an active direction in the field of aviation ergonomics. Based on the research of the development of SA history, the concept, the impacting factors and the analysis of qualitative model, and combined with previous research results, the paper has established a new definition of SA and the corresponding multi-level trigger qualitative analysis model, which has laid a foundation for further design of virtual experiments and the establishment of quantitative simulation model.

Memory depth of processing theory suggests that the factors affecting memory has nothing to do with the time but the processing level. Network label is the emerging Web 2.0 in the navigation mode. In this paper, memory processing of in-depth perspective of a comparative test was designed to explore the network of label design, and give the theoretical proof to the direction of its development.

Objective: The aim of this paper is to study the system problem to help pilots get the information which is useful among large amounts of data; Method: A new experiment on the limit of data strobe signal was built; Result: On the basis of theoretical consideration and analysis of experimental results, the effect on pilots' cognition and the design of display/control system was initially explained; Conclusion: The results showed that the increase of limit of data strobe signal can be accepted by objects; limit of data strobe signal will increase quickly by using digital or simple characters instead of words, but this will slightly increase performance errors; interruption of information doesn’t affect too much on limit of data strobe signal.

A two-step fermentation procedure for maize straw hydrolysis by Phanerochaete chrysosporium, Trichoderma and Aspergillus niger were investigated in this study. 2 mL of P. chrysosporium (107spores/mL) was added to 50 mL medium for the first fermentation stage. The optimal culture conditions were 28°C of culture temperature, 4.0 of pH, 8 days of culture time, 2 mL of Trichoderma suspensions (107spores/mL) and 1 mL of A. niger suspensions (107spores/mL) of the inoculums size on the second fermentation stage. Under the optimal conditions, the crude degradation rate achieved 48.2%. Via sulfonation, oxidate-lignosulfonate alkali lignin (OLAL) was achieved from maize straw hydrolysate(MSH).OLAL and MSH can be partial substitution for common water reducers and OLAL was more efficient compared to that of MSH.

For the weaknesses in the precision machining to leaves, the overall structure model of TX-6 CNC abrasive belt grinding machine was established by the soft of Inventor, and the grinding trajectory was planned. The finite element model, static stiffness and modal analysis of the machine were established. The design of TX-6 CNC abrasive belt grinding machine have been approved.

Lindane is a broad-spectrum persistent organochlorine pesticide that has been used to control pests for many years. In this study, its toxic mechanisms in adult zebrafish were investigated using targeted metabolomics with GC-MS/MS and non-targeted metabolomics with LC-Orbitrap-MS/MS. Zebrafish was exposed to lindane in water for 48 h in three groups: control, low exposure (1/10 LC50) and high exposure (LC50). In the zebrafish exposed to low concentration of lindane, 2.24–3.98 mg/kg of lindane were determined, while 35.67–56.46 mg/kg were observed in the zebrafish exposed to high concentration. A total of 118 metabolites were identified from 394 metabolites on GC-MS/MS and 45 metabolites were selected as biomarkers. A total of 62 metabolites were identified on LC-Orbitrap-MS/MS and 7 metabolites were selected as biomarkers. Three groups were well separated on partial least squares-discriminant analysis (PLS-DA), and a total of 52 metabolites in both the targeted and non-targeted metabolites were selected as biomarkers through VIP and ANOVA tests to construct a heatmap. Five metabolic pathways such as the pentose phosphate pathway (PPP), histidine metabolism, phenylalanine metabolism, alanine/aspartate/glutamate metabolism, and phenylalanine/tyrosine/tryptophan biosynthesis, were observed to show toxicologically significant alterations. Oxidative stress was also confirmed through MDA and ROS assays. Such perturbations of the metabolic pathways of zebrafish caused by the exposure to lindane resulted in significant toxicological effects.

The dissipation of ethoprophos and tricyclazole in soil and their translocation tendency to spinach were investigated. Prior to field trials, the analytical method for the determination of these pesticide residues was optimized and validated on soil and spinach. The field trial was conducted under greenhouse conditions for two different pretreatment periods with the pesticides. After treating with pesticides 30 (PBI-30) and 60 days (PBI-60) before seeding, soil samples were collected on different days for the dissipation study of soil. Spinach samples were harvested from the soil, and 50% and 100% mature spinach samples were collected. The initial amounts of ethoprophos residue in the PBI-60 and PBI-30 soils were 0.21 and 2.74 mg/kg, respectively, and these both decreased to less than 0.01 mg/kg on the day of spinach harvest. Similar initial residues of tricyclazole were observed in the PBI-60 (0.87 mg/kg) and PBI-30 soils (0.84 mg/kg), and these decreased to 0.44 and 0.34 mg/kg, respectively. The half-lives of ethoprophos in the soils were calculated as 7.6 and 4.8 days, respectively, while relatively long half-lives of 36.5 and 77.0 days were calculated for tricyclazole. According to the pesticide residue amounts in the spinach, the translocation rate from the soil to the spinach was determined. In the case of ethoprophos, the residual amount was already rapidly degraded in the soil, and the translocation rate could not be confirmed. On the other hand, for tricyclazole, it was confirmed that 1.19 to 1.61% of the residual amount in soil was transferred to spinach. According to these results, safe management guidelines for tricyclazole in soil were suggested considering the maximum residue limit on spinach.

On the basis of Markov chain simulation, an efficient method is presented to analyze reliability sensitivity of structure. In the presented method, Markov chain is employed to draw the samples distributed in the failure region, and these samples are fitted in a form of hyperplane by the weighted regression. By use of the regressed hyperplane, it is convenient to complete the sensitivities of the failure probability with respect to the distribution parameters of basic random variables by the available method. The presented method is applied to some examples to validate its accuracy and efficiency. The obtained results show that the presented reliability sensitivity analysis method is far more efficient than Monte Carlo based method.

PTEN is a lipid phosphatase responsible for down-regulating the phosphoinositide 3-kinase product phosphatidylinositol 3,4,5-triphosphate. Phosphatidylinositol 3,4,5-triphosphate is involved in the activation of the anti-apoptotic effector target, Akt. Although the Akt pathway has been implicated in regulating NF-κB activity, it is controversial as to whether Akt activates NF-κB predominantly through mechanisms that regulate nuclear translocation or transactivation potential. In this report, we utilized PTEN as a natural biological inhibitor of Akt activity to study the effects on tumor necrosis factor (TNF)-induced activation of NF-κB. We found that the reintroduction of PTEN into prostate cells inhibited TNF-stimulated NF-κB transcriptional activity. PTEN failed to block TNF-induced IKK activation, IκBα degradation, p105 processing, p65 (RelA) nuclear translocation, and DNA binding of NF-κB. However, PTEN inhibited NF-κB-dependent transcription by blocking the ability of TNF to stimulate the transactivation domain of the p65 subunit. PTEN also inhibited the transactivation potential of the cyclic AMP-response element-binding protein, but this was not observed for c-Jun. The transactivation potential of p65 following TNF stimulation could be rescued from PTEN-dependent repression by re-introducing expression constructs encoding activated forms of phosphoinositide 3-kinase, Akt, or Akt and IKK. The ability of PTEN to inhibit the TNF-induced transactivation function of p65 is important, because expression of PTEN blocked TNF-stimulated NF-κB-dependent gene expression, thus sensitizing cells to TNF-induced apoptosis. Maintenance of the PTEN tumor suppressor protein is therefore required to modulate Akt activity and to concomitantly control the transcriptional activity of the anti-apoptotic transcription factor NF-κB.

Soil salinization is one of the major land degradation types and has greatly influenced sustainable agricultural development. Zonation of saline-alkali land is the precondition for effective amelioration. The present situation of saline-alkali land is monitored by remote sensing image processing. Causes for land salinization are analyzed, especially the two key factors, ground water depth and its mineralization degree, are analyzed by using long-term observation data. Previously, zonation of saline-alkali soil was made descriptively and artificially. Based on the present situation of saline-alkali land, ground water depth and ground water mineralization degree, the zonation of saline-alkali land for amelioration in the Yellow River Delta was completed quantitatively. Four different types of saline-alkali land amelioration zones are delineated, namely, easy ameliorated zone, relatively difficult ameliorated zone, difficult ameliorated zone and unfavorable ameliorated zone. Countermeasures for ameliorating saline-alkali soils are put forward according to ecological conditions of different saline-alkali land zones.

PROBLEM: To study the mechanism of action of major histocompatibility complex (MHC)‐linked genes affecting reproduction, growth, and susceptibility to chemical carcinogens.

The segment of rat chromosome 20 (RNO20p12) that contains the classical loci of the major histocompatibility complex (MHC; RT1.A – RT1.E) also contains genes affecting growth, reproduction and susceptibility to chemical carcinogens (the Grc) and multiple genes encoding class I MHC antigens (the EC region). The relative positions of the MHC, Grc, and EC region have not been demonstrated explicitly, although they have been postulated from genetic mapping studies. The present study was undertaken to map these regions cytogenetically by several different approaches using cosmids specific for the Rps 18, Hspcil and Batl genes. The order was shown to be: centromere –Rps 18 – Hspal – Butl – EC -Grc.

PROBLEM: The nature of major histocompatibility complex (MHC) antigen expression on rat placentas, trophoblast cell lines, and tumors derived from trophoblast cells was explored.

The purpose of this study was to determine the role of gene methylation on major histocompatibility complex (MHC) antigen expression in the rat placenta, specifically: 1) the constitutive suppression of labyrinthine class I genes and of all class II genes, 2) genomic imprinting of class I genes, and 3) the amount of fetal loss in relationship to gene methylation. Placentas from all four mating combinations, or a subset thereof, of the inbred DA and WF strains of rats were studied simultaneously through 1) molecular assessment of their levels of methylation at various stages of pregnancy and changes in methylation after treatment with 5-azacytidine and 2) immunohistochemical determination of their MHC class I and class II antigen expression. Treatment with 5-azacytidine caused demethylation of both class I and class II genes, the level depending upon the method of administration of the drug. Treatment with 5-azacytidine did not, however, alter the expression of either the class I or class II antigens. Hence, demethylation is not involved in the constitutive suppression of labyrinthine class I genes or that of all placental class II genes, and it is not involved in the genomic imprinting of placental class I genes. The effect of 5-azacytidine on fetal loss is due to its direct cellular effects and not to an immunological mechanism.

Abstract: Alignment of class I-hybridizing cosmids from an R21 (A l B l D l E u grc +) genomic DNA library gave two contigs: one [150 kilobases (kb)] encompassed theE/C region, or a large part thereof, and the other (110 kb) contained thegrc region which has genes influencing resistance to chemical carcinogens (rcc), fertility (ft), and growth (dw-3). Amplification of gene sequences in the four cosmids in theE/C region usingE u -specific andLW2 (RT1.C)-specific primers showed that each cosmid contained bothE u -like andC-like genes. They are clearly different but closely associated, and they show some variation from the prototypicE (E u ) andC (LW2) genes, respectively. Comparison of DNA fromgrc + andgrc − strains of rats showed that the deletion in thegrc − strains was approximately 50 kb, and that it was located on two of the three cosmids in thegrc-region contig. The use of specific class I probes showed that thegrc region contained tandemly duplicatedRT1.O-RT1.N genes and that theRT.BM1 loci lay outside of thegrc region. Neither contig reacted with probes specific for class II,TNFA, Hsp70, orRT1.M genes. The data presented here and the previous data in the literature (summarized in Gill et al. 1995) suggest that the gene order in the major histocompatibility complex (MHC) andMHC-linked region of the rat is:A-E/C-grc-M.
