Graphene oxide (GO) humidity sensors based on poly(vinyl alcohol) (PVA) nanofibers have been proposed. The PVA nanofiber layers of different densities were obtained by adjusting the electrospinning time. Then, GO films were deposited on PVA nanofibers by a spin-coating method. The electrical properties of GO films are improved due to the increased distribution of PVA nanofibers in the GO films. The humidity sensors exhibit good sensitivity under a high relative humidity range of 40–80%. The response of sensors has reached 98.44% at a humidity level of 80% RH. The GO/PVA sensors have good stability at various humidity levels for 1 week. Furthermore, the GO/PVA sensors were used for respiration monitoring under different statuses. These sensors have good application prospects in the respiratory detection and analysis of diseases.

Windows are commonly the primary way for noise to enter the building environment. Most windows achieve higher sound insulation targets by simply increasing glass thickness or laminate. However, the glass thickness cannot be increased freely due to the window frame’s limitation. This research selects eleven kinds of asymmetric insulating laminated glass (glass + polyvinyl butyral + glass + air + glass) within 27 mm, which are the typical thicknesses of window frames. The three pieces of glass have equal thicknesses. The half-space acoustics method investigated the acoustic properties and modal behavior of the glass with finite thickness. The results show that when PVB or three glass increases simultaneously, and the sound insulation of each glass can be improved by 0.9∼3.4 dB or 3.7∼21.8 dB at first resonant frequencies, respectively. The glass with the best sound insulation is 7 mm glass + 0.76 mm polyvinyl butyral + 7 mm glass + 5 mm air + 7 mm glass (7 + 0.76 + 7 + 5A + 7 for short). Its sound transmission loss (TL) is 38.5 dB from 125 to 4000 Hz and 16.8 dB in the first resonant frequency (200 Hz). In addition, its first resonant frequency is the highest among eleven kinds of glass, and the number of natural frequencies in the low frequency range is only 9. These findings can provide a basis for designing asymmetric insulating laminated glass to improve the low frequency noise reduction effect effectively.

Telomerase is a basic reverse transcriptase that maintains the telomere length in cells, and accurate and specific sensing of telomerase in living cells is critical for medical diagnostics and disease therapeutics. Herein, we demonstrate for the first time the construction of an enzymatically controlled DNA nanomachine with endogenous apurinic/apyrimidinic endonuclease 1 (APE1) as a driving force for one-step imaging of telomerase in living cells. The DNA nanomachine is designed by rational engineering of substrate probes and reporter probes embedded with an enzyme-activatable site (i.e., AP site) and their subsequent assembly on a gold nanoparticle (AuNP). Upon recognition and cleavage of the AP site in the substrate probe by APE1, the loop of the substrate probe unfolds, exposing telomeric primer (TP) with the 3′–OH end. Subsequently, the TP is elongated by telomerase at the 3′–OH end to generate a long telomeric product. The resultant telomeric product acts as a swing arm that can hybridize with a reporter probe to initiate the APE1-powered walking reaction, ultimately generating a significantly enhanced fluorescence signal. Notably, endogenous APE1 is used as the driving force of the DNA nanomachine, avoiding the introduction of exogenous auxiliary cofactors into the cellular microenvironment. Owing to the high kinetics and high amplification efficiency of the APE1-powered DNA nanomachine, this strategy enables one-step sensitive sensing of telomerase in vitro and in vivo. It can successfully discriminate telomerase activity between cancer cells and normal cells, screen telomerase inhibitors, and monitor the variations of telomerase activity in living cells, offering a prospective platform for molecular diagnostics and drug discovery.

Amuc_1100 is a membrane protein from Akkermansia muciniphila, which has been found to play a role in host immunological homeostasis in the gastrointestinal tract by activating TLR2 and TLR4. In this study we investigated the effects and underlying mechanisms of Amuc_1100 on acute pancreatitis (AP) induced in mice by intraperitoneal injection of caerulein and lipopolysaccharide (LPS). The mice were treated with the protein Amuc_1100 (3 μg, i.g.) for 20 days before caerulein injection. Cecal contents of the mice were collected for 16S rRNA sequencing. We found that pretreatment with Amuc_1100 significantly alleviated AP-associated pancreatic injury, reduced serum amylase and lipase. Amuc_1100 pretreatment significantly inhibited the expression of proinflammatory cytokines (TNF-α, IL-1β, IFN-γ and IL-6) in spleen and pancreas through inhibiting NF-κB signaling pathway. Moreover, Amuc_1100 pretreatment significantly decreased the inflammatory infiltration, accompanied by the reduction of Ly6C+macrophages and neutrophils in the spleen of AP mice. Gut microbiome analysis showed that the abundance of Bacteroidetes, Proteobacteria, Desulfobacterotaand Campilobacterotawas decreased, while the proportion of Firmicutesand Actinobacteriotawas increased in AP mice pretreated with Amuc_1100. We further demonstrated that Amuc_1100 pretreatment restored the enrichment of tryptophan metabolism, which was mediated by intestinal flora. These results provide new evidence that Amuc_1100 lessens the severity of AP through its anti-inflammatory properties with a reduction of macrophages and neutrophil infiltration, as well as its regulation of the composition of intestinal flora and tryptophan metabolism.

N6-Methyladenine (6mdA) and N4-methylcytosine (4mdC) are the two most dominant DNA modifications in both prokaryotes and eukaryotes, but standard hybridization-based techniques cannot be applied for the 6mdA/4mdC assay. Herein, we demonstrate the silver-coordinated Watson–Crick pairing-driven three-dimensional (3D) DNA walker for locus-specific detection of genomic 6mdA/4mdC at the single-molecule level. 6mdA-DNA and 4mdC-DNA can selectively hybridize with the binding probes (BP1 and BP2) to form 6mdA-DNA-BP1 and 4mdC-DNA-BP2 duplexes. The 6mdA-C/4mdC-A mismatches cannot be stabilized by AgI, and thus, 18-nt BP1/BP2 cannot be extended by the catalysis of KF exonuclease. Through toehold-mediated strand displacement (TMSD), the signal probe (SP1/SP2) functionalized on the gold nanoparticles (AuNPs) can competitively bind to BP1/BP2 in 6mdA-DNA-BP1/4mdC-DNA-BP2 duplex to obtain SP1–18-nt BP1 and SP2–18-nt BP2 duplexes. The resulting DNA duplexes can act as the substrates of lambda exonuclease, leading to the cleavage of SP1/SP2 and the release of Cy3/Cy5 and 18-nt BP1/BP2. The released 18-nt BP1/BP2 can subsequently serve as the walker DNA, moving along the surface of the AuNP to activate dynamic 3D DNA walking and releasing abundant Cy3/Cy5. The released Cy3/Cy5 can be quantified by single-molecule imaging. This nanosensor exhibits high sensitivity with a limit of detection (LOD) of 9.80 × 10–15M for 6mdA-DNA and 9.97 × 10–15M for 4mdC-DNA. It can discriminate 6mdA-/4mdC-DNA from unmodified genomic DNAs, distinguish 0.01% 6mdA-/4mdC-DNA from excess unmethylated DNAs, and quantify 6mdA-/4mdC-DNA at specific sites in genomic DNAs of liver cancer cells and Escherichia coliplasmid cloning vector, providing a new platform for locus-specific analysis of 6mdA/4mdC in genomic DNAs.

Switchable materials have gained significant attention due to their potential applications in data storage, sensors, and switching devices. Two-dimensional (2D) hybrid perovskites have demonstrated promising prospects for designing switchable materials, where the dynamic motion of the organic components coupled with the distortion of the inorganic framework provides the driving force for triggering multifunctional switchable properties. Herein, through the H/F substitution strategy, we report a polar 2D hybrid lead-based perovskite, (4,4-DCA)2PbBr4(4,4-DCA = 4,4-difluorocyclohexylammonium) (1), which exhibits dual-stable behavior in a dielectric and second harmonic generation (SHG) response during the reversible phase transition process near the high Curie temperature Tc∼ 409 K. The phase transition temperature is significantly increased by 41 K compared to the corresponding non-fluorinated (CHA)2PbBr4(CHA = cyclohexylammonium). Remarkably, the material shows rare broad-band yellow emission under UV excitation, attributed to the induction of self-trapped exciton emission by the distortion of the [PbBr6]4–octahedra, as confirmed by the first-principles analysis. 1also exhibited ferroelectricity with a saturation polarization value and a small coercive field. This study provides a new insight into the modification of multifunctional switchable materials through the H/F substitution strategy.

Bacterial infections remain a major cause of morbidity worldwide due to drug resistance of pathogenic bacteria. Photodynamic therapy (PDT) has emerged as a promising approach to overcome this drug resistance. However, existing photosensitizers (PSs) are broad-spectrum antibacterial agents that dysregulate the microflora balance resulting in undesirable side effects. Herein, we synthesized a series of aggregation-induced emission (AIE)-active PSs with a lipophilic cationic AIE core with varying charges, named TBTCP and its derivatives. The association of the difference in their molecular charge with the antibacterial effects was systemically investigated. Among the derivatives presented, TBTCP-SF with the electronegative sulfonate group nulled its ability to bind to and ablate Gram-positive (G+) or Gram-negative (G−) bacteria. TBTCP-QY modified by electropositive quaternary ammonium facilitated binding and augmented the photodynamic antibacterial activity for both G+ and G– bacteria. TBTCP-PEG with hydrophilic neutral ligands selectively bound and inactivated G+ bacteria. Under white-light illumination, TBTCP-PEG ablated 99.9% methicillin-resistant Staphylococcus aureus(MRSA) and promoted wound healing in MRSA-infected mice, eliminating MRSA infection both in vitroand in vivo. Our work provides unprecedented insight into the utility of AIE-active PSs for highly targeted and efficient photodynamic ablation of either G+ or G– bacteria that can be translated to next-generation antibacterial materials.

Herein, a series of face-capped (Tr2M3)4L4(Tr = cycloheptatrienyl cationic ring; M = metal; L = organosulfur ligand) tetrahedral cages 1–3functionalized with 12 appended crown ether moieties were designed and synthesized. The reversible binding of ammonium cations with peripheral crown ether moieties to adjust internal guest-binding was realized. Combination of a bisammonium linker and cage 3led to the formation of a supramolecular gel SPN1via host–guest interactions between the crown ether moieties and ammonium salts. The obtained supramolecular gel exhibited multiple-stimuli responsiveness, injectability, and excellent self-healing properties and could be further developed to a SPN1-based drug delivery system. In addition, the storage modulus of SPN1was 20 times higher than that of the model gel without Pd–Pd bonded blocks, and SPN1had better self-healing properties compared with the latter, demonstrating the importance of such cages in improving mechanical strength without losing the dynamic properties of the material. The cytotoxicity in vitro of the drug-loaded (doxorubicin or methotrexate) SPN1was significantly improved compared to that of free drugs.

Nucleobase oxidation and alkylation can destroy Watson–Crick base-pairing to challenge the genomic integrity. Human 8-oxoguanine glycosylase 1 (hOGG1) and alkyladenine glycosylase (hAAG) are evolved to counter these two cytotoxic lesions through base-excision repair, and their deregulations are implicated with multifactorial diseases and cancers. Herein, we demonstrate activatable self-dissociation of Watson–Crick structures with fluorescent nucleotides for sensing multiple human glycosylases at single-cell level. The presence of hOGG1 and hAAG catalyzes 8-oxoG and deoxyinosine removal in functional probe 1 to release two trigger probes (1 and 2). Then, trigger probes hybridize with functional probe 2 to activate the autocatalytic degradation of functional probes 2 (Cycle I) and 3 (Cycle II), replicating abundant trigger probes (1–4) and releasing two fluorophores (2-aminopurine (2-AP) and pyrrolo-dC (P-dC)). New trigger probes (1, 2) and (3, 4), in turn, hybridize with free functional probes 2 and 3, repeating Cycles I and II turnovers. Through multicycle self-dissociation of Watson–Crick structures, 2-AP and P-dC are exponentially accumulated for the simultaneous quantification of hOGG1 and hAAG. This nanodevice exhibits high sensitivity with a detection limit of 2.9 × 10–3U/mL for hOOG1 and 1.5 × 10–3U/mL for hAAG, and it can measure enzymatic kinetics, identify potential inhibitors, discriminate glycosylases between cancer and normal cell lines, and even quantify glycosylase activities in a single HeLa cell. Moreover, this assay may be rapidly and isothermally performed in one tube with only one tool enzyme in a quencher-free manner, promising a simple and powerful platform for multiple human glycosylase detection.

The selective separation of structurally similar aliphatic/aromatic hydrocarbons is an essential goal in industrial processes. In this study, we report the synthesis of a water-soluble (Tr2M3)4L4(Tr = cycloheptatrienyl ring; M = metal; L = organosulfur ligand) molecular cage (1) via self-assembly of the water-soluble acceptor tripalladium sandwich species [(Tr2Pd3)(CH3CN)][NO3]2and the attachment onto L of solubilizing methoxyethoxy appendants to be utilized in an energy-friendly alternative approach to the separation of structurally similar molecules under ambient conditions. Cage 1, comprising a hydrophobic inner cavity, exhibited good solubility and stability in aqueous media. It also demonstrated excellent performance in the sequential separation of alkanes (C6–C9), xylene, and other disubstituted benzene isomers and cis/trans-decalin.

Supplemental Digital Content is available in the text.Confirming the tumor origin is often a diagnostic challenge in pathology and carries significant therapeutic impacts. Cytokeratin 7, estrogen receptor, and GATA binding protein 3 (GATA3) are well-established diagnostic markers frequently used to support a tumor’s breast origin. However, their specificities still have room to improve. Many nonbreast tumors express cytokeratin 7 and estrogen receptor, and urothelial tumors frequently express GATA3. There is a practical need for a new breast lineage marker that is sensitive and specific. Wnt family member proteins play critical roles in embryo development, tissue homeostasis and tumor development through β-catenin dependent and independent pathways. The current study evaluated Wnt9b and GATA3 expression in 163 primary breast cancers, 63 metastatic breast cancers, and 525 nonbreast epithelial tumors. The positive rates of Wnt9b and GATA3 in primary breast cancer were both 98.7%. The positive rates in metastatic breast cancer were 87.3% for Wnt9b and 96.8% for GATA3. For nonbreast tumors, including 64 cases of urothelial carcinoma, Wnt9b was negative in all except salivary gland carcinomas. The study demonstrated that Wnt9b is a breast cancer marker with similar sensitivity as GATA3 but with greater specificity than GATA3 and may ultimately become a useful diagnostic tool in routine surgical pathology practice.

The Badain Jaran Desert is the third largest desert in China, covering an area of 50000 km2. It lies in Northwest China, where the arid and rainless natural environment has a great impact on the climate, environment, and human living conditions. Based on the results of 1∶250000 regional hydrogeological surveys and previous researches, this study systematically investigates the circulation characteristics and resource properties of the groundwater as well as the evolution of the climate and ecological environment since the Quaternary in the Badain Jaran Desert by means of geophysical exploration, hydrogeological drilling, hydrogeochemistry, and isotopic tracing. The results are as follows. (1) The groundwater in the Badain Jaran Desert is mainly recharged through the infiltration of local precipitation and has poor renewability. The groundwater recharge in the desert was calculated to be 1.8684×108m3/a using the water balance method. (2) The Badain Jaran Desert has experienced four humid stages since the Quaternary, namely MIS 13-15, MIS 5, MIS 3, and the Early–Middle Holocene, but the climate in the desert has shown a trend towards aridity overall. The average annual temperature in the Badain Jaran Desert has significantly increased in the past 50 years. In detail, it has increased by about 2.5°C, with a higher rate in the south than in the north. Meanwhile, the precipitation amount has shown high spatial variability and the climate has shown a warming-drying trend in the past 50 years. (3) The lakes in the hinterland of the Badain Jaran Desert continuously shrank during 1973–2015. However, the vegetation communities maintained a highly natural distribution during 2000–2016, with the vegetation cover has increased overall. Accordingly, the Badain Jaran Desert did not show any notable expansion in that period. This study deepens the understanding of groundwater circulation and the climate and ecological evolution in the Badain Jaran Desert. It will provide a scientific basis for the rational exploitation of the groundwater resources and the ecological protection and restoration in the Badain Jaran Desert.

Alkaline phosphatase (ALP) is an important hydrolase with crucial roles in biological processes, and the dysregulation of ALP may cause various human diseases. The conventional ALP assays usually involve cumbersome procedures with poor sensitivity. Herein, taking advantage of intrinsic superiorities of molecular beacons (MBs) and unique features of terminal deoxynucleotidyl transferase (TdT), we demonstrate for the first time the 3′-terminal repair-powered dendritic nanoassembly of polyadenine (A) MBs for one-step quantification of ALP in human serum. When ALP is present, it catalyzes 3′-terminal dephosphorylation of poly-A MBs to induce TdT-mediated template-free polymerization, generating long chains of polythymidine (T) sequences. The long poly-T chains can function as the anchoring templates to hybridize with many poly-A MBs, leading to the unfolding of loop structures and the dissociation of FAM/BHQ1 pairs (the 1st amplification stage). Subsequently, all 3′-hydroxylated poly-A MBs can be extended with the assistance of TdT to generate the branched long poly-T chains, leading to the hybridization of more poly-A MBs and the dissociation of more FAM/BHQ1 pairs (the 2nd amplification stage). Through multiple rounds of extension, assembly, and activation of poly-A MBs, dendritic DNA nanostructures are automatically formed, resulting in the dissociation of abundant fluorophores from the FAM/BHQ1 pairs to generate an exponentially amplified fluorescence signal (the nth amplification stage). This strategy possesses high sensitivity and excellent specificity, and the detection limit can reach 1 cell. Moreover, it can evaluate kinetic parameters, screen inhibitors, estimate cellular inhibition effects, and measure ALP in human serums.

Phototheranostics represents a promising direction for modern precision medicine, which has recently received considerable attention for cancer research. The ingenious integration of all phototheranostic modalities in a single molecule with precise spatial colocalization is a tremendously challenging task, which mainly arises from the complexity of molecular design and energy dissipation. Reports on a single molecular one-for-all theranostic agent are still very rare. Herein, we designed two novel aggregation-induced emission (AIE)-active fluorogens (AIEgens, named DPMD and TPMD) with a cross-shaped donor–acceptor structure via a facile synthetic method and constructed versatile nanoparticles (NPs) by encapsulating AIEgen with an amphiphilic polymer. The AIEgen TPMD with a twisted structure, high donor–acceptor (D–A) strength, small singlet–triplet energy gap, and abundant intramolecular rotators and vibrators was selected as an ideal candidate for balancing and utilizing the radiative and nonradiative energy dissipations. Notably, TPMD NPs simultaneously possess adequate near-infrared (NIR) fluorescence emission at 821 nm for fluorescence imaging, effective reactive oxygen species generation for photodynamic therapy (PDT), and outstanding photothermal effect for photoacoustic imaging, photothermal imaging, and photothermal therapy (PTT), which demonstrates the superior potential of AIE NPs in multimodal imaging-guided synergistic PDT/PTT therapy.

Two-dimensional double perovskites have experienced rapid development due to their outstanding optoelectronic properties and diverse structural characteristics. However, the synthesis of high-performance multifunctional compounds and the regulation of their properties still lack relevant examples. Herein, we synthesized two multifunctional compounds, (C6H14N)4AgSbBr8(1) and (F2–C6H12N)4AgSbBr8(2), which exhibit high solid-state phase transition temperature, bistable dielectric constant switching, second harmonic generation (SHG), and bright emission. Through H/F substitution, the transition temperature increases and achieves a smaller band gap attributed to reduced interlayer spacing. Furthermore, we investigated the broad emission mechanism of the compounds through first-principles calculation and variable-temperature fluorescence, confirming the presence of the STE1 emission. Our work provides insight into the further development of multifunctional compounds and chemical modification that enhances compound properties.

Clinical guidelines state that neoadjuvant chemotherapy should be administered before surgery in muscle invasive urinary bladder small cell neuroendocrine carcinoma. Recently described marker insulinoma-associated protein 1 (INSM1) has been reported to be sensitive and specific for neuroendocrine differentiation, however, its efficacy in urinary tract small cell carcinoma is not well established. This study examines immunohistochemical expression of INSM1 on whole tissue sections of urinary tract small cell neuroendocrine carcinoma and compares INSM1 expression with established neuroendocrine markers. Immunohistochemical stains for CD56, INSM1, synaptophysin, and chromogranin were performed on 32 cases of small cell neuroendocrine carcinoma of the bladder. Staining was scored for intensity (0: no staining; 1: weak; 2: moderate; 3: strong) and proportion of cells stained (0: 0%; 1: >0% to ≤25%; 2: >25% to ≤50%; 3: >50% to ≤75%; 4: >75% to 100%). INSM1 was positive (intensity 1 to 3 or proportion 1 to 4) in 87% (28/32) of cases (20 with intensity 2 to 3, 17 with proportion 3 to 4). CD56, synaptophysin, and chromogranin were positive in 75% (24/32), 60% (19/32), and 44% (14/32) of cases, respectively. INSM1 was negative (n=4) or only showed weak intensity staining (n=7) in 34% (11/32) of cases. INSM1 is a sensitive marker of small cell neuroendocrine differentiation of the urinary tract. However, this study suggests that optimal utilization of INSM1 would be inclusion in a limited panel of stains rather than as a stand-alone screening marker given that it is negative or only shows weak intensity staining in a significant proportion of cases.

The aim of this study was to reveal the effect of let‐7b on osteoporosis (OP). Synthetic let‐7b mimics or inhibitors were transfected into MC3T3‐E1 cells. The expression of let‐7b in MC3T3‐E1 and its effect on cell viability, apoptosis, and the apoptosis‐related proteins (Bcl‐2, Bax, and cleaved caspase‐9) were tested by CCK‐8 assay, flow cytometry and Western blot, severally. The osteogenic differentiation markers (Runx2 and Osterix) and Wnt/β‐catenin pathway related markers (β‐catenin and C‐myc) were detected by qRT‐PCR and Western blot. The relationships between let‐7b and cyclin D1 (CCND1) were confirmed by luciferase reporter assay. The differentiation and mineralization of MC3T3‐E1 cells were analyzed by alkaline phosphatase (ALP) activity assay and alizarin red staining. The outcomes indicated that overexpression/ablation of let‐7b repressed/facilitated MC3T3‐E1 cell viability and accelerated/suppressed MC3T3‐E1 cell apoptosis. Besides, a remarkable decrease/augment of Bcl‐2 protein expression and the distinct fortify/reduction of Bax and cleaved caspase‐9 expression levels were observed in let‐7b mimics/inhibitors group in MC3T3‐E1 cells. Moreover, we discovered that let‐7b overexpression/ablation retrained/facilitated the mRNA and protein expression of Runx2 and Osterix. It was confirmed that CCND1 was a downstream target of let‐7b and was negatively modulated by let‐7b. In addition, high‐expression/deficiency of let‐7b inhibited/increased the expression levels of β‐catenin and C‐myc in MC3T3‐E1 cells. Taken together, our study revealed that let‐7b overexpression/depletion repressed/accelerated MC3T3‐E1 cell proliferation, differentiation, and mineralization while promoted/suppressed MC3T3‐E1 cell apoptosis through targeting CCND1, which might be adjusted by Wnt/β‐catenin pathway. Our findings might offer a basis for developing novel targets for OP treatment.

Introduction: Physical activity is a key component of a healthy lifestyle for youths with visual impairments (i.e., blindness or low vision). This study aims to examine the habitual physical activity and sedentary time of Chinese children and adolescents with visual impairments and to determine whether age, gender, and body composition significantly influence their behavior.Methods: A purposeful sample method was utilized to recruit 137 Chinese youths with visual impairments from a special school for individuals with visual and hearing impairments. A total of 72 students with visual impairments (aged 7–17 years; Mage = 13.4) participated in this study. ActiGraph GT3X accelerometers were used to measure the habitual physical activity and sedentary time.Results: Youths with visual impairments in this study spent an average of 287.86 (standard deviation [SD] = 45.67) minutes of light physical activity, 146.79 (SD= 56.95) minutes of moderate physical activity, 18.47 (SD= 15.15) minutes of vigorous physical activity, 165.26 (SD= 69.83) minutes of moderate-to-vigorous-intensity physical activity, and 979.51 (SD= 109.10) minutes of sedentary time per day. Children with visual impairments in elementary school engaged in more habitual physical activity and had less sedentary time than did students in middle and high school. Differences in gender and body composition with regard to time spent being active or sedentary were insignificant.Discussion: Findings demonstrated that Chinese youths with visual impairments in this study had sufficient health-enhancing physical activity and were habitually active. Future research is needed to identify the factors that account for the high habitual physical activity levels of Chinese youths with visual impairments.Implications for practitioners: Transition planning for students with visual impairments to facilitate active habitual physical activity participation when children grow up is warranted.

Reliable multiple single nucleotide polymorphisms (SNPs) detection at low abundance is of great significance for disease diagnosis and biomedical research. Herein, we have developed a novel and simple method for multiple SNPs detection combining solid-phase capture by specific hybridization with online preconcentration of capillary gel electrophoresis-laser-induced fluorescence (CGE-LIF). The method presents an excellent performance due to its favorable traits: the solid-phase short-chain hybridization ensures the high specificity of SNP detection; the effective separation ability of CGE can easily achieve multiplex detection; the simple online preconcentration significantly improves the detection sensitivity of fluorescent probe by nearly 100-fold. For a single SNP target, the assay achieves a limit of detection as low as 0.01–0.02% for three different NRASmutations in the same codon. For multiple SNP targets, as low as 0.05% abundance can be easily realized. Our method is simple, efficient, ultrasensitive, and universal for multiple SNPs detection without complex enzymatic or chemical ligation reaction, which shows great potential in early clinical diagnosis.

Circulating miRNAs are a newly emerging class of noninvasive biomarkers, and the accurate quantification of their expression is essential to the biological research and early clinic diagnosis. Herein, we demonstrate the construction of a quencher-free cascade amplification system for highly sensitive detection of serum circulating miRNAs. The target miRNA can hybridize with the linear probe to induce the cyclic strand displacement amplification (SDA) (cycle I) for the production of the binding probes. The binding probe can subsequently react with the 2-aminopurine (2-AP)-hairpin probe to induce the recycling exonuclease cleavage of 2-AP-hairpin probes (cycle II), releasing the triggers and 2-AP molecules simultaneously. The released trigger can hybridize with the free linear probe to start new cycles I and II amplifications. Through multiple rounds of cascade amplifications, a large number of 2-AP molecules are released, generating an enhanced fluorescence signal. This method exhibits a large dynamic range of 8 orders of magnitude and a detection limit of 0.16 aM. It can differentiate a single-base mismatch in miR-486-5p, quantify miR-486-5p in lung cancer cells at various stages, and even discriminate the expressions of serum circulating miR-486-5p in healthy persons from that in nonsmall-cell lung carcinoma (NSCLC) patients. Moreover, this assay can be rapidly carried out in one step under isothermal condition in a label-free manner, holding promising applications in point-of-care diagnosis and prognosis of lung cancers.

Effective chimeric antigen receptor-modified T-cell (CAR-T) therapy for liver metastases (LM) will require innovative solutions to ensure efficient delivery and minimization of systemic toxicity. We previously demonstrated the safety of CAR-T hepatic artery infusions (HAI). We subsequently conducted the phase 1b HITM-SIR trial, in which six patients (pts) with CEA+LM received anti-CEA CAR-T HAIs and selective internal radiation therapy (SIRT). The primary endpoint was safety with secondary assessments of biologic activity. Enrolled pts had a mean LM size of 6.4 cm, 4 pts had >10 LM, and pts received an average of two lines of prior systemic therapy. No grade 4 or 5 toxicities were observed, and there were no instances of severe cytokine-release syndrome (CRS) or neurotoxicity. The mean transduction efficiency was 60.4%. Following CAR-T HAI, reduced levels of GM-CSF-R, IDO, and PD-L1 were detected in LM, and serum CEA levels were stable or decreased in all subjects. Median survival time was 8 months (mean 11, range 4–31). Anti-CEA CAR-T HAI with subsequent SIRT was well tolerated, and biologic responses were demonstrated following failure of conventional therapy. HAI of CAR-T was once again confirmed not to be associated with severe CRS or neurotoxicity.

The properties of supramolecular structures are highly dependent on their metal-centered building blocks and organic linkers, thus the search for novel systems will lead to new functions and applications for these unique assemblies. Here, two discrete triangular trimetallic sandwich building blocks were developed to construct supramolecular assemblies through coordination-driven self-assembly with organosulfur ligands. A series of tubelike (Tr2Pd3)4L6assemblies (Tr = cycloheptatrienyl ring) were obtained from a discrete triangular tripalladium sandwich complex with bifunctional organosulfur ligands. By replacing the metal centers of the platinum analogue, the self-assembly process resulted in the clean formation of (Tr2Pt3)2L3triple helicates instead of tubelike species. The trimetallic sandwich building blocks were also shown to form face-capped (Tr2M3)4L4(M = Pd or Pt) tetrahedral cages when trifunctional organosulfur ligands were used. The supramolecular assemblies were comprehensively analyzed by X-ray crystallography. A metal-cluster-induced structural transformation between (Tr2Pd3)4L4tubes and (Tr2Pt3)2L3triple helicates was observed. Furthermore, the face-capped (Tr2Pd3)4L4cage possesses a tetrahedral cavity allowing the encapsulation of a series of guests.

Burgeoning evidence points to glycogen synthase kinase (GSK)3β as a key player in diverse kidney diseases. However, as a pivotal transducer of the insulin signaling pathway, the role of GSK3β in diabetic kidney disease remains uncertain. In db/dbmice, renal expression of total and activated GSK3β was increasingly elevated. This preceded the development of diabetic kidney disease, and correlated with the progression of signs of diabetic kidney injury, including albuminuria and extracellular matrix accumulation in glomeruli and tubulointerstitia. In vitro, exposure of glomerular podocytes, mesangial cells, and renal tubular cells to a diabetic milieu induced GSK3β overexpression and hyperactivity, which seem essential and sufficient for eliciting diabetic cellular damages in kidney cells, because the cytopathic effect of the diabetic milieu was mitigated by GSK3β knockdown, but was mimicked by ectopic expression of constitutively active GSK3β even in the normal milieu. In consistency, kidney biopsy specimens procured from patients with varying stages of diabetic nephropathy revealed an amplified expression of total and activated GSK3β in glomeruli and renal tubules, associated with the severity of diabetic nephropathy. Moreover, in retrospective cohorts of type 2 diabetic patients that were followed for over five years, the relative activity of GSK3β in banked urinary exfoliated cells represented an independent risk factor for development or progression of renal impairment. Furthermore, receiver operating characteristic curve analysis demonstrated that GSK3β activity in urinary exfoliated cells provided much better power than albuminuria in discriminating diabetic patients with progressive renal impairment from those with stable kidney function. Thus, renal expression and activity of GSK3β are amplified in experimental and clinical diabetic nephropathy. Hence, GSK3β in urinary exfoliated cells may serve as a novel biomarker for predicting diabetic kidney disease progression.

Background: Considering the limitations of cisplatin in clinical application, there is ongoing research to fabricate new platinum-containing prodrug which are highly effective to tumor cells and have low toxicity to normal cells. Methods: In this study, a cyclodextrin-based supramolecular platinum prodrug that is 6,6’-ophenylenediseleno- bridged bis (β-cyclodextrin)s (CD) and its potassium tetrachloroplatinate(II) complex was reported. The cytotoxicity experiments were performed to evaluate the anticancer activities of supramolecular prodrug in vitro by means of MTT assay. The practical application of supramolecular prodrug in tumor treatment in vivo were evaluated using BALB/c nude mice model bearing Hela cancer cells. Results: Compared with commercial anticancer drug cisplatin, the resultant cyclodextrin-based platinum prodrug exhibited comparative anticancer effect but with much lower toxicity side effects in vitro and in vivo. Conclusion: The cyclodextrin-based supramolecular platinum prodrug displayed antitumor activity comparable to the commercial antitumor drug cisplatin but with lower side effects both in vitro and in vivo, implying that the two adjacent cyclodextrin cavities not merely act as desired solubilizer, but also endowed the prodrug with cell permeability through the interaction of cyclodextrin with phospholipids and cholesterol on cell membrane.

The physiological uptake of 68Ga-FAPI-04 due to the change of the internal environment is little known. We report the case of a 45-year-old woman who was highly suspected to have advanced nasopharyngeal carcinoma. 18F-FDG and 68Ga-FAPI-04 PET/CT was performed for evaluating the disease. Both PET and CT with different tracers detected the primary nasopharyngeal carcinoma and metastases in right neck lymph nodes, liver, and bones. To our surprise, intense diffuse uptake of 68Ga-FAPI-04 was found in both breasts, which might be due to the hormone stimulation because the patient received 68Ga-FAPi-04 PET/CT just at the period of ovulation.

Carbon-nanotube shaped reinforcement (CSR) and traditional latitude and longitude reinforcement (LLR) made of tough resin were 3D printed and applied to concrete specimens. The element numbers of 10, 12, and 14 per layer were selected to investigate the reinforcement by CSR and LLR separately. The uniaxial compressive behaviors of the CSR and LLR reinforced concrete specimens were studied by a series of laboratory tests. The experimental results indicate that the strength of a CSR reinforced specimen with 10, 12, and 14 elements per layer increases by 59.77%, 85.94%, and 108.98%, respectively, compared with the unreinforced specimen. The strength of the LLR reinforced specimen with 10, 12, and 14 elements per layer increases by 24.22%, 46.88%, and 68.75%, respectively, compared with the unreinforced specimen. CSR thus demonstrates higher efficiency in compressive strength improvement than LLR does. The results also show that the failure pattern changes from global failure to partial failure as the element number per layer of CSR increases. The present research provides a potential innovative reinforcing technology for civil engineering applications.

A 47-year-old woman suffered worsening pain in the waist and numbness in the right thigh for 1 month. MRI was performed to determine the cause, which detected an osteolytic lesion in the T12 vertebral body, suggestive of possible bone metastasis. FDG PET/CT scan was undertaken to detect the primary tumor, which only showed the same isolated lesion in the T12 without any other abnormal hypermetabolic lesion. The pathology following vertebrectomy revealed granulomatous infection. The diagnosis of osseous syphilis was eventually made following a subsequent positive Treponema pallidumserological test.

A 21-year-old man complained of cough, fever, and hemoptysis for 15 days. Peripheral neutrophil cell (33.8 × 109/L) was markedly increased, and a mass in the left lung was detected by chest radiography. 18F-FDG PET/CT was referred for characterizing the lesion and found a large mass with multiple cavities in the left lung, which had markedly high uptake of 18F-FDG, mimicking pulmonary abscess. Surprisingly, the lesion was eventually proved to be neutrophil-rich anaplastic large cell lymphoma. After 4 cycles' chemotherapy, the lesion shrank significantly.

The current study aimed to identify whether ultrasonographic measurements of optic nerve sheath diameter (ONSD) could dynamically and sensitively evaluate real-time intracranial pressure (ICP). ONSD measurements were performed approximately 5  min prior to and after a lumbar puncture (LP). A total of 84 patients (mean±SD age, 43.5±14.7 years; 41 (49%) men; 18 patients with elevated ICP) were included in the study. The Spearman correlation coefficients between the two observers were 0.779 and 0.703 in the transverse section and 0.751 and 0.788 in the vertical section for the left and right eyes, respectively. The median (IQR) change in ONSD (ΔONSD) and change in ICP (ΔICP) were 0.11 (0.05–0.21) mm and 30 (20–40) mmH2O, respectively, for all participants. With a reduction in cerebrospinal fluid pressure, 80 subjects (95%) showed an immediate drop in ONSD; the median (IQR) decreased from 4.13 (4.02–4.38) mm to 4.02 (3.90–4.23) mm (p<0.001). Significant correlations were found between ONSD and ICP before LPs (r=0.482, p<0.01) and between ΔONSD and ΔICP (r=0.451, p<0.01). Ultrasonic measurement of ONSD can reflect the relative real-time changes in ICP.

The prediction skill of Arctic Oscillation (AO) in the decadal experiments with the Beijing Climate Center Climate System Model version 1.1 (BCC_CSM1.1) is assessed. As compared with the observations and historical experiments, the contribution of initialization for climate model to predict the seasonal scale AO and its interannual variations is estimated. Results show that the spatial correlation coefficient of AO mode simulated by the decadal experiment is higher than that in the historical experiment. The two groups of experiments reasonably reproduce the characteristics that AO indices are the strongest in winter and the weakest in summer. Compared with historical experiments, the correlation coefficient of the monthly and winter AO indices are higher in the decadal experiments. In particular, the correlation coefficient of monthly AO index between decadal hindcast and observation reached 0.1 significant level. Furthermore, the periodicity of the monthly and spring AO indices are achieved only in the decadal experiments. Therefore, the initial state of model is initialized by using sea temperature data may help to improve the prediction skill of AO in the decadal prediction experiments to some extent.

Background:Organ transplantation is considered the ultimate therapy for end-stage organ disease. While pharmacologic immunosuppression is the mainstay of therapeutic strategies to prolong the survival of the graft, long-term use of immunosuppressive medications carries the risk of organ toxicity, malignancies, serious opportunistic infections, and diabetes. Therapies that promote recipient tolerance in solid organ transplantation are able to improve patient outcomes by eliminating the need for long-term immunosuppression. Summary:Establishing tolerance to an allograft has become an area of intense study and would be the ideal therapy in clinical practice. The discovery of a subset of T cells naturally committed to perform immunoregulation has led to further investigation into their role in the immunopathogenesis of transplantation. Evidence suggests that regulatory T cells (Tregs) are fundamentally involved in promoting allograft tolerance. Efforts to characterize specific markers for Tregs, while challenging, have identified Foxp3 gene expression as a crucial step in promoting the tolerance-inducing features of Tregs. A number of approaches, including those based on targeting the glycogen synthase kinase 3β signaling pathway or activating the melanocortinergic pathway, have been tested as a way to promote Treg lineage commitment and maintenance as well as to facilitate immune tolerance. In order to be effective in clinical practice, Tregs must be allospecific and possess a specific phenotype to avoid suppression of other aspects of the immune system or increasing the risk of malignancy or infections. Multiple experimental and clinical studies have demonstrated the impact of currently used immunosuppressants on the immunoregulatory activities of Tregs and their Foxp3 expression status. Pharmacological induction of tolerogenic Tregs for inducing transplant tolerance, including epigenetic therapies, is in the ascendant. Key Messages:Therapies that promote Treg function and survival may represent a novel strategy for achieving immune tolerance in transplant patients.

A series of acridines were designed and synthesized for the development of effective inhibitors for mild steel corrosion in 1 M HCl solution, in which the halogen-substituted acridines showed better inhibitive performance than the nonhalogen-substituted acridines. The corrosion protection properties of the halogen-substituted acridines, including 2-chloro-9-phenylacridine (CPA), 2-chloro-9-(2-fluorophenyl)acridine (CFPA), and 2-bromo-9-(2-fluorophenyl)acridine (BFPA), were further investigated using weight loss test and electrochemical techniques. The results indicated the halogen-substituted acridines have excellent inhibitiion performance, and these acridines act as mixed-type inhibitors with predominant cathodic effectiveness. Adsorption of acridines on a mild steel surface obeyed the Langmuir adsorption isotherm. The adsorption of the inhibitor molecules on the steel surface was further supported by scanning electron microscopy (SEM), scanning electrochemical microscopy (SECM), and FTIR spectroscopy. The inhibition mechanism of the investigated halogen-substituted acridines was derived using DFT-based quantum chemical calculations for their neutral as well as protonated forms. Both experimental and DFT studies suggested that the inhibition efficiency of three halogen-substituted acridines followed the order of η(BFPA) > η(CFPA) > η(CPA).

Synapse dysfunction is an early hallmark of Alzheimer’s disease (AD), and was considered to be closely related to memory loss. The molecular mechanisms that trigger synapse loss and dysfunction remain poorly understood. Increasing evidence shows a link between Rho GTPases and synapse plasticity. Rho GTPases play a role in controlling synapse function by regulating actin cytoskeleton and dendritic spines. Observations have suggested that phytochemicals, such as flavonoids, alleviate cognition impairment in AD. However, to date, the link between the protective effect of flavonoids on AD and the activity of Rho GTPases remains uninvestigated. In this study, APP/PS1 mice were used as an AD model, and we found that synapse loss occurred in AD mice brain. Flavonoids extracted from leaves of Diospyros kaki(FLDK) were used to investigate whether its protective effects on synapse were related to Rho GTPases activity in AD mice. The Rho GTPases Activation Kit showed that Ras homologous member A (RhoA)-GTP was significantly higher and Ras-related C3 botulinum toxin substrate 1 (Rac1)-GTP was significantly lower in APP/PS1 mice than in normal mice, and RhoA-GTP activity was significantly inhibited by FLDK. We also found that FLDK improved learning and memory function, and antagonized the downregulation expressions of synapse-related proteins such as synaptophysin and drebrin. These findings suggest that FLDK is a potential therapeutic agent for AD, and modulation of Rho GTPases activity might contribute toward its protective effect.

Alumina ceramics sintered at 1350, 1450 and 1550 °C with different grain sizes were tested under dynamic load using a small-scale compressed air gun with sharpened tungsten carbide bullets impacting at a sub-ballistic velocity of 360 km/h. The deformation behavior and sub-surface cracks were recorded by visual examination. Cr3+ fluorescence was used to measure the residual stress distribution on and beneath the impact site. The results show the alumina sintered at 1350 °C with a fine grain size of 1.7 μm and high hardness of 18.3 GPa is good at bulletproof due to the cone cracks can spread the dynamic stress effectively and consume much impact energy.

IMPORTANCE: The crtierion standard method for monitoring intracranial pressure (ICP) can result in complications and pain. Hence, noninvasive, repeatable methods would be valuable. OBJECTIVE: To examine how ultrasonographic optic nerve sheath diameter (ONSD) correlated with noninvasive and dynamically monitored ICP changes. DESIGN, SETTING, AND PARTICIPANTS: The ONSD was measured before the lumbar puncture (LP) in 60 patients on admission. Patients with elevated ICP were divided into group 1 (200 &lt; LP ≤ 300 mm H2O) and group 2 (LP > 300 mm H2O). Patients underwent follow-up ONSD and LP measurements within 1 month. We analyzed the correlations between the ONSD and ICP on admission and between the changes in ONSD and ICP, which were the respective changes in ONSD and ICP from admission to follow-up. MAIN OUTCOMES AND MEASURES: The ultrasonographic ONSD and ICP were measured on admission and follow-up. The correlations between the ONSD and ICP on admission and between the changes in ONSD and ICP were analyzed using Pearson correlation analyses. RESULTS: For 60 patients (Han nationality; mean [SD] age, 36.2 [12.04] years; 29 [48%] female) on admission, the ONSD and ICP values were strongly correlated, with an r of 0.798 (95% CI, 0.709-0.867; P &lt; .001). Twenty-five patients with elevated ICP who completed the follow-up were included. The mean (SD) ONSD and ICP on admission were 4.50 (0.54) mm and 302.40 (54.26) mm H2O, respectively. The ONSD and ICP values obtained on admission were strongly correlated , with an r of 0.724 (95% CI, 0.470-0.876; P &lt; .001). The mean (SD, range) changes in ICP and ONSD were 126.64 (52.51 mm H2O, 20-210 mm H2O) (95% CI, 106.24-146.07) and 1.00 (0.512 mm, 0.418-2.37 mm) (95% CI, 0.83–1.20), respectively. The change in ONSD was strongly correlated with the change in ICP, with an r of 0.702 (95% CI, 0.425–0.870; P &lt; .001). The follow-up evaluations revealed that the elevated ICP and dilated ONSD had returned to normal, and no evidence of difference was found in the mean ONSDs between group 1 (3.49 mm; 95% CI, 3.34–3.62 mm) and group 2 (3.51 mm; 95% CI, 3.44–3.59 mm) (P = .778) at follow-up. CONCLUSIONS AND RELEVANCE: The dilated ONSDs decreased along with the elevated ICP reduction. Ultrasonographic ONSD measurements may be a useful, noninvasive tool for dynamically evaluating ICP.

Methionine is the only one of the essential amino acids that contain sulfur, widely used as a feed additive in agriculture. In this study, the availability of 5-methyl-tetrahydrofolate was confirmed as the main limitation in the complex multibranched biosynthetic pathway of L-methionine. The cycle of one-carbon units was thoroughly investigated and modified to supply 5-methyl-tetrahydrofolate for L-methionine production, such as enhancing the supply of precursor, expediting the conversion rate of the cycle, introducing exogenous serine hydroxymethyltransferase and increasing pool size of one-carbon units carrier. The final strain MYA/pAmFA-4was able to produce 20.89 g/L L-methionine by fed-batch fermentation, which was the highest titer reported in the literatures. This study is instructive for other metabolites biosynthesized needing one-carbon units or having a complex multibranched biosynthetic pathway.

Routine histopathologic examination of hemorrhoidectomy specimens is controversial having been described as not useful and expensive with few of these common cases demonstrating incidental lesions. However, unexpected premalignant and malignant lesions have been detected on excised hemorrhoids. The high-risk human papillomavirus (HR-HPV) types associated with these incidentally identified high-grade lesions are presently unknown. We aimed to identify cases of incidental high-grade anal intraepithelial neoplasia (HG-AIN) and anal squamous cell carcinoma incidentally discovered on hemorrhoidectomy specimens, genotype HR-HPVs from these lesions, and assess p53 and p16 expression by immunohistochemistry to identify risk factors for their development. With institutional approval, cases with associated demographics from 1995 to 2015 were reviewed to identify and confirm incidental HG-AIN or squamous cell carcinoma in hemorrhoidectomy specimens. Genotyping for HR-HPV types and immunohistochemical staining for p53 and p16 was performed. Statistical analysis comparing HPV genotypes, p53 and p16 staining, and potential risk factors by the Fisher exact test was performed. In the largest series of incidental high-grade lesions on hemorrhoidectomy, HPV 16 was the most common HR-HPV detected though multiple-type infections were common including some HPV 16/18-negative cases. By genotyping, HPV 39 was significantly associated with IV-drug abuse history (P=0.0015) and HIV-positive status (P=0.037), whereas HPV 58 detection correlated with chemotherapy-induced immunosuppression (P=0.029). There was frequent overlap between p53 staining and HPV positivity, particularly when HPV 31 was detected. We also identified several mimickers of HG-AIN that may present diagnostic challenges in these specimens. Our data support continued routine examination of hemorrhoidectomy specimens and suggest that adjunctive studies such as immunohistochemistry for challenging cases may be useful.

Silicon carbide ceramics with different grain sizes were tested at high strain rate using a small-scale gas gun with sharpened tungsten carbide projectiles impacting at a sub-ballistic velocity of 100 m/s. The resistance of ceramics to fracture was recorded by visual examination of the cracking on the impacted surface and the damage in the sub-surface region. The crack pattern and impact crater region size observed in silicon carbide specimens are generally due to their mechanical properties. Silicon carbide samples had good capacity to defeat the projectiles but the low toughness led to early failure. Raman SiC mapping was used to examine the residual stress and plastic deformation induced in each material. The silicon carbide samples showed different dynamic behaviours due to the extent of stress relaxation.

A simple colorimetric sensing method for the detection of Hg2+was developed by combining enzymatic catalysis with DNA-based hybridization chain reactions (HCRs). Firstly, thymine (T)-rich capture DNA (cDNA) was immobilized on a gold electrode viaAu–S bonding. In the presence of Hg2+, thymine (T)-rich probe DNA (pDNA) hybridized with cDNA viaT–Hg2+–T base pairs. Then the HCRs were realized using pDNA as an initiator and two biotin-labeled hairpin DNAs (H1 and H2) as fuel strands. Finally, numerous avidin-labeled horseradish peroxidase (HRP) enzymes were immobilized on long nicked ds-DNA strands, which can catalyze the H2O2-mediated oxidation of 3,3,5,5-tetramethylbenzidine dihydrochloride hydrate (TMB) to cause a dramatic color change. Under optimal conditions, the absorbance of TMB was linear with the logarithm of Hg2+concentrations in the range of 1 fM to 1 pM, with a detection limit of 0.33 fM. This strategy exhibited good selectivity and high sensitivity, which might be a potential tool for the practical detection of Hg2+in environmental monitoring.

Cadmium-based compounds have emerged as outstanding multifunctional materials due to their excellent optical and multistate switching properties. However, the hybrid compound often faces challenges such as a low transition temperature and poor fluorescence. It is necessary to understand the structure–property relationships to enhance compound performance. Herein, through H/F substitution strategy, we synthesized a multifunctional compound, (4,4-DCA)2CdCl4(4,4-DCA = 4,4-difluorocyclohexylammonium) (1), which exhibits reversible solid-state phase transition, a wide optical band gap, and blue fluorescence emission. The transition temperature is significantly increased by 42 K after fluorinating the previously reported compound because of the higher rotational energy barrier of the fluorinated cation. Moreover, through the Sb doping strategy, the quantum yields significantly improved by ∼20%, and the average fluorescence lifetime also increased. First-principles calculations revealed that Sb doping altered the frontier orbital, and the exciton carrier can return to the ground state more rapidly via Sb orbitals. This work demonstrates the enhancement of chemical properties through H/F substitution and a metal-doping strategy, providing insights into the scientific modification-induced improvement in compound performances.

Finding new cost-effective and environmentally friendly anti-corrosion materials is a never-ending task. Herein, the main objective is to prepare a new formulation based on piroxicam-cyclodextrin inclusion complex (PX@β-CD) as an efficient green inhibitor to protect mild steel against corrosion in 1 M HCl. The inhibition performance of PX@β-CD was determined by electrochemical tests coupled with theoretical study like as DFT calculation to assess the reactivity and interaction mechanisms between PX@β-CD and Fe. The obtained results revealed that PX@β-CD performs excellently inhibition performance where its inhibition efficiency reached 98.65% at 0.4 mM, and it acted as a mixed type inhibitor with anodic predominance. SEM and contact angle analysis showed the formation of compact PX@β-CD film on steel surface. The adsorption of PX@β-CD inhibitor followed the Langmuir adsorption model according to the gravimetric corrosion measurement. Thermodynamic analysis and DFT calculations revealed the chemisorption of the inhibitors on the steel surface.

In this paper, weak water cooling technology by wiper is introduced for the casting of 520×1650 mm 7A55 alloy slabs. By analyzing from experiment comprehensively, it indicates that the position of wiper will exert big influence on slab quality. Under the conditions of the experiment, if wiper height is below the bottom of sump, there will be more than 70% possibility for slab to have cracks. If it is excessively higher than 80 mm, the top of the sump, porosity will incur and macro-segregation will be enlarged. The experiment determines the correct height of wiper.

In view of the shortcoming of traditional image fusion based on discrete wavelet transform (DWT) with unclear textural information, an effective visible light and infrared image fusion algorithm via feature residual and statistical matching is proposed in this study. First, the source images are decomposed into low-frequency coefficients and high-frequency coefficients by DWT. Second, two different fusion schemes are designed for the low-frequency coefficients and high frequency ones, respectively. The low-frequency coefficients are fused by a local feature residual-based scheme to achieve adaptive fusion; the high-frequency coefficients are accomplished by a local statistical matching-based scheme to extract the edge information effectively. Finally, the fused image is obtained by inverse DWT. Experimental results demonstrate that the proposed method can produce a more accurate fused image, leading to an improved performance compared with existing methods.

Flush and chemical cleaning were applied to remove foulants on UF membrane fouled by lactic acid fermentation of food waste. Flushing methods, flushing time and agents for chemical cleaning were studied. The obtained results indicated that backwash at 0.1 MPa for 10s and chemical cleaning with NaClO could gain the best cleaning effect.

The thermal stability of constituent particles in both as-rolled and solution treated in AA7A55 has been investigated by means of scanning electron microscopy (SEM) and differential scanning calorimetry (DSC). Three types of solution treatments were used to dissolve particles constituents. Whereas the degree of recrystallization aggravates which deteriorates the properties with increasing the solution temperature, a significant amount of recrystallization exists after utilizing 480°C solution treatment whereas a much smaller extent from 480°C to 490°C. And electrochemical and exfoliation corrosion matched with the degree of recrystallization. With increasing the recrystallization the corrosion become worse.

This paper introduces a solar controller based on SCM, system uses STC15F2K61S2 microcontroller as the core device of the control circuit. This design uses PWM (pulse width modulation) control technology and MOSFET tube is opened and closed to control the battery charging and discharging. The experimental results show that, the controller performance is reliable, can monitor the state of implementation of solar battery and storage battery, battery charge and discharge control optimal, to prolong the service life of the battery to.

Multiple myeloma (MM) is a malignant plasma cells proliferative disease. The intricate cross-talk of myeloma cells with bone marrow microenvironment plays an important role in facilitating growth and survival of myeloma cells. Bone marrow mesenchymal stem cells (BMMSCs) are important cells in MM microenvironment. In solid tumors, BMMSCs can be educated by tumor cells to become cancer-associated fibroblasts (CAFs) with high expression of fibroblast activation protein (FAP). FAP was reported to be involved in drug resistance, tumorigenesis, neoplastic progression, angiogenesis, invasion, and metastasis of tumor cells. However, the expression and the role of FAP in MM bone marrow microenvironment are still less known. The present study is aimed to investigate the expression of FAP, the role of FAP, and its relevant signaling pathway in regulating apoptosis induced by bortezomib in MM cells. In this study, our data illustrated that the expression levels of FAP were not different between the cultured BMMSCs isolated from MM patients and normal donors. The expression levels of FAP can be increased by tumor cells conditioned medium (TCCM) stimulation or coculture with RPMI8226 cells. FAP has important role in BMMSCs mediated protecting MM cell lines from apoptosis induced by bortezomib. Further study showed that this process may likely through β-catenin signaling pathway in vitro. The activation of β-catenin in MM cell lines was dependent on direct contact with BMMSCs other than separated by transwell or additional condition medium from BMMSCs and cytokines.
