1. A semi-idealized modeling study on the long-lived eastward propagating mesoscale convective system over the Tibetan Plateau

Abstract: Based on a 16-warm-season statistical study on the mesoscale convective systems (MCSs) that were generated over the Tibetan Plateau (TP), 11 long-lived eastward propagating MCSs of the same type were selected for a composite semi-idealized simulation and a corresponding no-latent-heating sensitivity run by using the Weather Research and Forecasting (WRF) model. Common evolutionary features and associated mechanisms of this type of long-lived eastward propagating MCS were investigated. Main results are as follows: (i) This type of MCS was generated in a favorable background environment which was characterized by a notable upper-tropospheric divergence south of an upper-level jet, a strong warm advection around a middle-level shortwave trough’s central area, and an instable convective stratification below the trough. Development of the MCS featured rapid increase of cyclonic vorticity in the middle and lower troposphere. The convergence-related vertical stretching and tilting were key factors for the cyclonic-vorticity’s production, and convection-related upward cyclonic-vorticity transport contributed to the upward extending of the MCS. (ii) During the vacating stage of the MCS, it first coupled with a quasi-stationary Tibetan Plateau vortex (TPV) over the TP’s eastern section, and then decoupled from the vortex. In the former stage, the MCS contributed to maintaining ascending motions and convergence associated with the TPV, which favored its persistence; whereas, in the latter stage, decoupling weakened the TPV-associated convection significantly. This reduced the upward transport of cyclonic vorticity notably, which, together with the negative tilting effect, finally led to the vortex’s dissipation. (iii) After vacating TP, the MCS first weakened due to the disappearance of strong direct sensible heating from the TP on its bottom, and then, under the favorable conditions associated with the shortwave trough over the eastern section of the TP, the MCS redeveloped rapidly. Convergence-related cyclonic-vorticity production in the middle and lower troposphere and upward transport of cyclonic vorticity due to convection governed the MCS’s redevelopment. (iv) Sensitivity simulation shows that latent heating was a necessary condition for the formation and development of the long-lived eastward propagating MCS. On the one hand, this MCS affected the TP’s eastern section and downstream regions directly by inducing precipitation; and on the other hand, it exerted effects on the precipitation over a wider range in the downstream regions by modulating large-scale circulations over and around the TP.

2. Outcomes of Congenital Zika Disease Depend on Timing of Infection and Maternal-Fetal Interferon Action

Abstract: Zika virus (ZIKV) infection during pregnancy in humans results in intrauterine growth restriction, spontaneous abortion, and microcephaly. Here, we found that fetus-derived type I interferon (IFN-I) signaling can enhance anti-ZIKV responses and provide clinical benefits to the fetus. Because IFN-λ shares signaling cascades and antiviral functions with IFN-I, we investigated the in vivoeffects of IFN-λ in ZIKV-infected pregnant mice. IFN-λ administration during mid-pregnancy reduced ZIKV burden in maternal and fetal organs and alleviated placental injuries and fetal demise. In addition, prophylactic and therapeutictreatment of IFN-λ1 in a human trophoblast line, as well as in primary human amniotic epithelial cells, greatly reduced the ZIKV burden. Our data highlight IFN-λ1 as a potential therapeutic useful for women at risk for congenital Zika disease.

3. Critical Role of the CXCL10/C-X-C Chemokine Receptor 3 Axis in Promoting Leukocyte Recruitment and Neuronal Injury during Traumatic Optic Neuropathy Induced by Optic Nerve Crush

Abstract: Traumatic optic neuropathy (TON) is an acute injury of the optic nerve secondary to trauma. Loss of retinal ganglion cells (RGCs) is a key pathological process in TON, yet mechanisms responsible for RGC death remain unclear. In a mouse model of TON, real-time noninvasive imaging revealed a dramatic increase in leukocyte rolling and adhesion in veins near the optic nerve (ON) head at 9 hours after ON injury. Although RGC dysfunction and loss were not detected at 24 hours after injury, massive leukocyte infiltration was observed in the superficial retina. These cells were identified as T cells, microglia/monocytes, and neutrophils but not B cells. CXCL10 is a chemokine that recruits leukocytes after binding to its receptor C-X-C chemokine receptor (CXCR) 3. The levels of CXCL10 and CXCR3 were markedly elevated in TON, and up-regulation of CXCL10 was mediated by STAT1/3. Deleting CXCR3 in leukocytes significantly reduced leukocyte recruitment, and prevented RGC death at 7 days after ON injury. Treatment with CXCR3 antagonist attenuated TON-induced RGC dysfunction and cell loss. In vitroco-culture of primary RGCs with leukocytes resulted in increased RGC apoptosis, which was exaggerated in the presence of CXCL10. These results indicate that leukocyte recruitment in retinal vessels near the ON head is an early event in TON and the CXCL10/CXCR3 axis has a critical role in recruiting leukocytes and inducing RGC death.

5. Dual spike and nucleocapsid mRNA vaccination confer protection against SARS-CoV-2 Omicron and Delta variants in preclinical models

Abstract: Emergence of SARS-CoV-2 variants of concern (VOCs), including the highly transmissible Omicron and Delta strains, has posed constant challenges to the current COVID-19 vaccines that principally target the viral spike protein (S). Here, we report a nucleoside-modified messenger RNA (mRNA) vaccine that expresses the more conserved viral nucleoprotein (mRNA-N) and show that mRNA-N vaccination alone can induce modest control of SARS-CoV-2. Critically, combining mRNA-N with the clinically proven S-expressing mRNA vaccine (mRNA-S+N) induced robust protection against both Delta and Omicron variants. In the hamster models of SARS-CoV-2 VOC challenge, we demonstrated that, compared to mRNA-S alone, combination mRNA-S+N vaccination not only induced more robust control of the Delta and Omicron variants in the lungs but also provided enhanced protection in the upper respiratory tract. In vivo CD8+T cell depletion suggested a potential role for CD8+T cells in protection conferred by mRNA-S+N vaccination. Antigen-specific immune analyses indicated that N-specific immunity, as well as augmented S-specific immunity, was associated with enhanced protection elicited by the combination mRNA vaccination. Our findings suggest that combined mRNA-S+N vaccination is an effective approach for promoting broad protection against SARS-CoV-2 variants.

However, patient prognosis and the host immune response differ considerably depending on the infecting parasite species. As neutrophils are a prominent component of the inflammatory infiltrate in chronic cutaneous, diffuse cutaneous and mucocutaneous lesions, we examined neutrophil activation and microbicidal activity against amastigotes of L. amazonensis and L. braziliensis. We found that murine neutrophils internalized L. braziliensis amastigotes with greater efficiency than did L. amazonensis amastigotes. Additionally, L. braziliensis infection was a potent trigger for neutrophil activation, oxidative burst, degranulation and the production of interleukin (IL)-22 and IL-10, while L. amazonensis amastigotes poorly induced these responses. Finally, neutrophils were able to kill L. braziliensis amastigotes, especially when cells were activated with phorbol myristate acetate. L. amazonensisamastigotes, however, were highly resistant to neutrophil microbicidal mechanisms. This study reveals, for the first time, differential neutrophil responsiveness to distinct species of Leishmaniaamastigotes and highlights the complexity of neutrophil-amastigote interactions during chronic leishmaniasis.© 2015 S. Abstract: In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

8. The immunologic basis for hepatitis C infection

Abstract: The host immune system is arguably involved in nearly every step of hepatitis C virus (HCV) infection. In patients, the outcome, whether it is a natural infection or results from an interferon--based treatment, is determined by a series of complex host-virus interactions. In this review, we focus on the state of research addressing the immune mechanisms critical for viral clearance and disease resolution. Additional discussion is devoted to the evasion and blockade tactics of HCV as well as to current efforts aimed at disrupting the replication cycle of this well-evolved virus. Current theories of immune-mediated injury of hepatocytes are also discussed.

9. Sand fly saliva enhances Leishmania amazonensis infection by modulating interleukin-10 production.

Abstract: After transmission through the bite of female sand flies, Leishmania spp. can cause a broad spectrum of disease manifestations collectively known as leishmaniases. L. amazonensis is endemic in South America, where it causes cutaneous, diffuse cutaneous, and visceral leishmaniasis. In this study, we have provided evidence that salivary gland extracts (SGE) of Lutzomyia longipalpis enhances L. amazonensis infection. BALB/c mice infected intradermally in the ear with 10(5) metacyclic promastigotes of L. amazonensis together with SGE (equivalent to 0.5 gland) showed an early onset of disease and larger lesions that contained approximately 3-log-units more parasites than did controls. To determine the potential mechanism underlying this enhancement, we assessed cytokine production via reverse transcriptase PCR and enzyme-linked immunosorbent assay. Mice coinjected with parasites and SGE displayed higher levels of interleukin-10 (IL-10) mRNA in the ear tissues, as well as higher levels of IL-10 in supernatants of restimulated draining lymph node (LN) cells, than did controls. Flow cytometric analysis revealed high frequencies of IL-10-producing CD4(+) and CD8(+) T cells in the draining LN of mice coinjected with the parasite and SGE. In addition, we examined bone marrow derived-macrophage cultures and detected increased IL-10 but decreased nitric oxide (NO) production in cells exposed to SGE prior to infection with L. amazonensis. Together, these results imply that the sand fly saliva facilitates Leishmania evasion of the host immune system by modulating IL-10 production.

We hypothesized that deficient activation of early immune responses contributes to the pathogenesis in L. amazonensis-infected mice. To distinguish early molecular events that determine the outcome of Leishmania infections, we examined cytokine gene expression in C57BL/6 mice infected with either L. amazonensis or L. major (a healing model). After 2 to 4 weeks, L. amazonensis-infected mice had significantly delayed and depressed expression of inflammatory cytokines (interleukin-12 [IL-12], gamma interferon, IL-1 alpha, IL-1 beta), CC chemokines (CC chemokine ligand 3 [CCL3]/macrophage inflammatory protein 1 alpha [MIP-1 alpha], CCL4/MIP-1 beta, CCL5/RANTES, MIP-2), and chemokine receptors (CCR1, CCR2, CCR5) in foot tissues and draining lymph nodes compared to the expression in L. major-infected controls. These findings correlated with defective T-cell responsiveness to parasite stimulation in vivo and in vitro. Adoptive transfer of L. amazonensis-specific Th1 cells prior to infection overcame the immune defects of the animals, leading to complete control of the disease. Studies with gene knockout mice suggested that IL-10, but not IL-4, contributed partially to compromised immunity in L. amazonensis-infected hosts. The data suggest that there is impairment in multiple immune functions at early stages of infection with L. amazonensis parasites and provide a compelling rationale to explore immune augmentation as an intervention in American cutaneous leishmaniasis.

13. Hepatitis C Virus Core and Envelope Proteins Do Not Suppress the Host's Ability To Clear a Hepatic Viral Infection

Abstract: ABSTRACTSeveral hepatitis C virus (HCV) proteins have been shown in vitro to interact with host cellular components that are involved in immune regulation. However, there is a paucity of data supporting the relevance of these observations to the in vivo situation. To test the hypothesis that such an interaction suppresses immune responses, we studied a line of transgenic C57BL/6 mice that express the HCV core and envelope proteins in the liver. The potential effects of these proteins on the hepatic immune response were evaluated by challenging these mice with a hepatotropic adenovirus. Both transgenic and nontransgenic mice developed similar courses of infection and cleared the virus from the liver by 28 days postinfection. Both groups of mice mounted similar immunoglobulin G (IgG), IgG2a, interleukin-2, and tumor necrosis factor alpha responses against the virus. Additionally, BALB/c mice were able to clear infection with recombinant adenovirus that does or does not express the HCV core and envelope 1 proteins in the same manner. These data suggest that HCV core and envelope proteins do not inhibit the hepatic antiviral mechanisms in these murine experimental systems and thus favor a model in which HCV circumvents host responses through a mechanism that does not involve general suppression of intrahepatic immune responses.

14. Increased efficiency of folding and peptide loading of mutant MHC class I molecules

Abstract: Class I molecules, encoded by diverse alleles at several loci of the major histocompatibility complex (MHC) are assembled in the endoplasmic reticulum (ER) from heavy chain, β2 microglobulin and peptide in association with accessory proteins of the peptide loading complex. We show here, that mutations in the α2 domain (Q115A; D122A) of the human class I allele HLA-A2 cause a lack of apparent association with the loading complex and a faster assembly. Despite the drastically reduced association with the TAP loading complex, i. e. less than 20 % of HLA-A2 expressed in the cells can be co-precipitated with either TAP, calreticulin or tapasin, the mutant proteins are expressed on the cell surface in a stable conformation, and bind a complex set of peptides almost identical to that of wild-type HLA-A2. Furthermore, the mutant class I molecules are more rapidly exported from the ER than wild-type HLA-A2 and undergo faster maturation. The mutation Q115A does not destroy a binding site for the loading complex as this HLA-A2 mutant associates with the loading complex when peptide supply is limited. The association of class I molecules with the TAP-associated loading complex appears to be a reflection of how quickly the stable conformation is gained.

Abstract: The recent 20 years (2000–2019) ozonesonde profiles recorded at the Hong Kong Observatory are analyzed to study the Tropospheric Ozone (TropO3) variability over Hong Kong, South China. The TropO3profiles are described without the constraints of traditional monthly averages; they are clustered based on a Self‐Organizing Map (SOM) to explore the short‐term variability, and separated into two 10‐year periods to investigate the long‐term variability based on pooled statistical analysis. The roles of precursor emissions, dynamic transport, and photochemical formation in multiple‐timescale TropO3variability are further evaluated with the aid of meteorological and satellite data, surface observation, emission inventory, and backward trajectory analysis. The results suggest that TropO3over Hong Kong has a seasonal and vertical distribution featuring a widely recognized spring maximum. The SOM‐based clustering analysis emphasizes a strong dependence of short‐term variability in TropO3profile patterns on cross‐latitude transport conditions, including rich‐O3transport from midlatitude region and poor‐O3transport from tropics. However, high O3levels throughout the troposphere (usually in early spring) are largely due to the combination of stratospheric intrusion and biomass burning. During the whole study period, the O3concentrations have increased substantially in the lower troposphere. Springtime wildfire‐related emissions over the upwind Indochina Peninsula are responsible for the lower free‐tropospheric O3increases. In contrast, the boundary‐layer O3uptrends with a larger increasing rate near the surface are mostly attributed to the reduced NO titration effect due to the sharp reduction of NOxemissions. 16. IFN-α/β and autophagy

Abstract: Hepatitis C virus (HCV) infects approximately 130 million people worldwide. The clinical sequelae of this chronic disease include cirrhosis, functional failure and carcinoma of the liver. HCV induces autophagy, a fundamental cellular process for maintaining homeostasis and mediating innate immune response, and also inhibits autophagic protein degradation and suppresses antiviral immunity. In addition to this ploy, the HCV serine protease composed of the viral non-structural proteins 3/4A (NS3/4A) can enzymatically digest two cellular proteins, mitochondria-associated anti-viral signaling protein (MAVS) and Toll/interleukin-1 receptor domain containing adaptor inducing IFN-β (TRIF). Since these two proteins are the adaptor molecules in the retinoic acid-inducible gene I (RIG-I) and TLR3 pathways, respectively, their cleavage has been suggested as a pivotal mechanism by which HCV blunts the IFN-α/β signaling and antiviral responses. Thus far, how HCV perturbs autophagy and copes with IFN-α/β in the liver remains unclear.

17. Modulation of the ENSO on Winter Aerosol Pollution in the Eastern Region of China

Abstract: The modulation of the El Niño/Southern Oscillation (ENSO) on winter aerosol pollution (WAP) in the eastern region of China (ERC) was investigated from a climatological perspective. The results show that the weakened East Asian winter monsoon during El Niño events has an important influence on increased WAP in ERC, especially over northern China. The enhancement of the East Asian winter monsoon related to La Niña events reduces WAP in northeast, east, and south China but increases it in central and western China. Compared to northern China, anomalies in dynamic and thermodynamic conditions related to ENSO lead to more uncertainties in its effect on aerosol pollution over southern China. El Niño (La Niña) events tend to cause southwesterly (northeasterly) wind anomalies in winter southern China, along with the blocking effect of Nanling Mountains on southward (northward) air pollution, which is not (is) beneficial for the transport outside or dilution in local aerosols over southern China. Meanwhile, positive (negative) water vapor convergence and rainfall anomalies caused by El Niño (La Niña) are (are not) beneficial for the local removal of aerosols. The dominance of both factors determines the ENSO modulation on aerosols over southern China. An atmospheric global climate model largely reproduces the observed findings. Consistent with the observation, El Niño events exacerbate WAP in most of ERC, whereas La Niña ones reduce it except for western China. As a source of large‐scale ocean‐atmosphere interactions originating from the tropical Pacific, ENSO not only leads to anomalies in atmospheric circulation locally but also arouses global climatological anomalies. In this study, from a climatological perspective, the modulation of ENSO on winter aerosol pollution over Eastern China was investigated by diagnosis analysis and CAM5 simulation.
