A series of polycarbinol-siloxane surfactants with variations of two to six hydroxyl groups in the head structure and tri- and tetrasiloxane tail structures is synthesized, characterized and tested for foaming ability and fire suppression. The synthesis is practical, involving two steps, and produces products in high yield with good purity. The trisiloxane surfactants with 5 or 6 hydroxyl groups have sufficient water solubility, critical micelle concentrations, and display foaming ability levels to be effective in fire suppression testing. The tetrasiloxane surfactant with six hydroxyl groups was less soluble by a factor of 10 and did not exhibit sufficient foaming for effective fire suppression.

Recently, we found that a new malaria vaccine, R21/Matrix-M, had over 75% efficacy against clinical malaria with seasonal administration in a phase 2b trial in Burkina Faso. Here, we report on safety and efficacy of the vaccine in a phase 3 trial enrolling over 4800 children across four countries followed for up to 18 months at seasonal sites and 12 months at standard sites.

Malaria and iron deficiency (ID) are common and interrelated public health problems in African children. Observational data suggest that interrupting malaria transmission reduces the prevalence of ID1. To test the hypothesis that malaria might cause ID, we used sickle cell trait (HbAS, rs334), a genetic variant that confers specific protection against malaria2, as an instrumental variable in Mendelian randomization analyses. HbAS was associated with a 30% reduction in ID among children living in malaria-endemic countries in Africa (n= 7,453), but not among individuals living in malaria-free areas (n= 3,818). Genetically predicted malaria risk was associated with an odds ratio of 2.65 for ID per unit increase in the log incidence rate of malaria. This suggests that an intervention that halves the risk of malaria episodes would reduce the prevalence of ID in African children by 49%.

Recent global declines in bee health have elevated the need for a more complete understanding of the cellular stress mechanisms employed by diverse bee species. We recently uncovered the biomarker lethal (2) essential for life [l(2)efl] genes as part of a shared transcriptional program in response to a number of cell stressors in the western honey bee (Apis mellifera). Here, we describe another shared stress-responsive gene, glycine N-methyltransferase (Gnmt), which is known as a key metabolic switch controlling cellular methylation reactions. We observed Gnmt induction by both abiotic and biotic stressors. We also found increased levels of the GNMT reaction product sarcosine in the midgut after stress, linking metabolic changes with the observed changes in gene regulation. Prior to this study, Gnmt upregulation had not been associated with cellular stress responses in other organisms. To determine whether this novel stress-responsive gene would behave similarly in other bee species, we first characterized the cellular response to endoplasmic reticulum (ER) stress in lab-reared adults of the solitary alfalfa leafcutting bee (Megachile rotundata) and compared this with age-matched honey bees. The novel stress gene Gnmt was induced in addition to a number of canonical gene targets induced in both bee species upon unfolded protein response (UPR) activation, suggesting that stress-induced regulation of cellular methylation reactions is a common feature of bees. Therefore, this study suggests that the honey bee can serve as an important model for bee biology more broadly, although studies on diverse bee species will be required to fully understand global declines in bee populations.

The hydrolytic susceptibility of sulfobetaine-siloxane surfactants is investigated by comparison of a homologous series in this subclass of surfactants (R-(CH2)3N+(Me)2(CH2)3SO3–; R = (Me3SiO)3Si-, (Me3SiO)2Si(Me)-, (Me2SiO)3-Si(Me)-) with an analogue series of oxyethylene-siloxane surfactants (R-(CH2)3(OCH2CH2)10.2OH; R = (Me3SiO)3Si-, (Me3SiO)2Si(Me)-, (Me2SiO)3-Si(Me)-). Nuclear magnetic resonance (NMR) monitoring of these surfactants in an aqueous solution shows that the presence of the sulfobetaine head structure greatly enhances the hydrolysis rate of the siloxane tail as compared with oxyethylene-siloxane analogue control experiments. This sulfobetaine effect is confirmed by adding a model compound, (Me)3N+(CH2)3SO3–, to the oxyethylene-siloxane surfactants and observing the large hydrolysis enhancement. Measurements of pH indicate the sulfobetaine presence greatly enhances acidity, but rigorous analysis could discover no source of acid other than the presence of the sulfobetaine structure. Titration measurements confirmed the presence of a tightly bound hydration layer of 4–7 water molecules per sulfobetaine group. It is speculated that the source of acidity may originate from an aqueous exclusion zone nucleated by the hydrated sulfobetaine at the interface of a sulfobetaine-siloxane surfactant bilayer aggregate. Hydrolysis prevention is investigated by addition of a pH 7 phosphate buffer, of an alkyl polyglycoside cosurfactant, and of a combination of both, with a finding of very significant but not complete suppression of the hydrolysis.

The RTS,S/AS01Emalaria vaccine (RTS,S) was introduced by national immunisation programmes in Ghana, Kenya, and Malawi in 2019 in large-scale pilot schemes. We aimed to address questions about feasibility and impact, and to assess safety signals that had been observed in the phase 3 trial that included an excess of meningitis and cerebral malaria cases in RTS,S recipients, and the possibility of an excess of deaths among girls who received RTS,S than in controls, to inform decisions about wider use.
