Graphical Abstract:

This study used Celluclast-assisted hydrolysis and DEAE-cellulose anion-exchange chromatography to extract, purify, and assess the anti-inflammatory activity of fucoidan derived from Sargassum fusiforme. The Celluclast-assisted hydrolysate was used for obtaining ethanol-precipitated S. fusiformefucoidans (SFF) which were rich in polysaccharides and sulfates. Five fractions obtained through DEAE-cellulose chromatography, notably SFF-F5, displayed high sulfate content (29.16 ± 0.15%). The anti-inflammatory potential of these fractions was tested in lipopolysaccharide-induced RAW 264.7 macrophages. SFF-F5 was found to significantly reduce NO production at non-toxic concentrations of 12.5, 25, and 50 μg/mL. Structural analysis using Matrix assisted laser desorption/ionization time-of flight mass spectrometry (MALDI-TOF MS) revealed repetitive patterns in the fucoidan fractions. And Electrospray ionization mass spectrometry (ESI-TOF MS) spectrum of SFF fractions identified various structures of glycans bound with fucose and sulfate (Fuc1(SO3)1, Fuc2(SO3)1, Hex5Fuc1(SO3)1). In-silicoevaluation based on the detected glycan structures indicated that fucoidan fractions could inhibit the dimerization of the TLR4-MD2 complex, demonstrating their potential as anti-inflammatory agents. This result confirm that the anti-inflammatory activity varies depending on the glycan composition of fucoidan. This study provides a foundation for further exploration into the relationship between the structure and anti-inflammatory activity of S. fusiforme-derived fucoidan, facilitating its effective industrial application.

Advanced glycation end-products (AGEs) are complex and heterogeneous compounds generated through various steps and are naturally produced during the glycation reaction. Excessively generated AGEs bind to long-lived proteins such as collagen and cause renal dysfunction, leading to AGE-related diabetic nephropathy (DN). In this study, we aimed to confirm the therapeutic potential of marine biological resources by evaluating the role of diphlorethohydroxycarmalol (DPHC), a major phlorotannin compound contained in edible brown algae Ishige okamurae, in the key mechanisms involved in the pathogenesis and progression of AGE-induced DN. The AGE formation inhibitory and AGE-collagen cross-link inhibiting/breaking ability assay were performed to evaluate the anti-glycation effect of DPHC. The AGE-RAGE interaction inhibitory effect was evaluated through molecular docking study. The renoprotective effect of DPHC was confirmed in mouse glomerular mesangial cells. DPHC treatment exhibited anti-glycation ability through suppression of AGE formation and AGE-collagen cross-linking generation, and disruption of cross-links formed between AGE-collagen. In addition, DPHC pre-treatment significantly suppressed reactive oxygen species (ROS) production, intracellular methylglyoxal (MGO)/AGEs accumulation, and activation of apoptosis cascade by MGO. Moreover, DPHC suppressed AGE-receptor for AGE (RAGE) interaction by competing with MG-H1 for the binding site of RAGE. Furthermore, DPHC pre-treatment regulated apoptosis-related protein expression and increased Nrf2, Glo-1, HO-1, CAT, NQO1, and SOD1. These findings prove that the chemical structural characteristics of DPHC have the potential to prevent or manage AGE-induced DN by effectively mediating the key mechanisms responsible for its pathogenesis.

Marine organisms have long been recognized as rich source of bioactive material with numerous health benefit effects. Among bioactive material, proteins and peptides derived from marine organisms have been shown to inhibit the development of neuro-degenerative diseases such as Alzheimers disease, Parkinsons disease, and multiple sclerosis. The neuro-protective action of protein and peptide seem to be mediated through direct interaction of absorb protein and peptide with a numerous of cellular and molecular targets with enzyme/ion channels. This contribution presents an overview of marine proteins and peptides neuro-protective effect and their potential application in neuro-protection.

Abstract: Acid-solubilized collagen (ASC) and pepsin-solubilized collagen (PSC) were extracted from the seaweed pipefish (Syngnathus schlegeli) and partially characterized. The amount of collagens isolated in the subsequent treatments was 5.5% of ASC and 33.2% PSC on the basis of lyophilized pipefish body weight, respectively. According to the electrophoretic pattern and CM-cellulose column chromatogram, the collagens might be classified as type I collagens, containing α1 and α2 chain. The imino acid content of collagen from pipefish was lower than those of mammalian collagens as also were the denaturation temperatures (Td) of collagens were 34.8°C and 35.1°C, respectively. This study shows that there is a possibility to use pipefish collagen as the alternative source of collagen from industrial purposes and subsequently it may evaluate the economical value of the seaweed pipefish.

Abstract: Seahorse, Hippocampus kuda (SH) a marine teleost fish, is well known not only for its special medicinal composition and used as one of the most famous and expensive materials of traditional Chinese medicine. It was extracted with water (SHW), methanol (SHM), and ethanol (SHE), respectively and evaluated by various antioxidant assays. The including reducing power, total antioxidant, DPPH radical scavenging, hydroxyl radical scavenging, superoxide anion radical scavenging, alkyl radical scavenging, and protective effect on DNA damage caused by hydroxyl radicals generated. Further, the ROS level was detected using a fluorescence probe, 2′,7′-dichlorofluorescin diacetate (DCFH-DA), which could be converted to highly fluorescent dichlorofluorescein (DCF) with the presence of intracellular ROS on mouse macrophages, RAW264.7 cell and inhibited myeloperoxidase (MPO) activity in human myeloid, HL60 cells, respectively. Those various antioxidant activities were compared to standard antioxidants such as α-tocopherol. Among SHM exhibited the highest antioxidant activity in linoleic acid system, effective reducing power, DPPH radical scavenging, hydroxyl radical scavenging, superoxide radical scavenging, alkyl radical scavenging, inhibitory intracellular ROS, and inhibited MPO activity. Furthermore, MTT assay showed no cytotoxicity on mouse macrophages cell (RAW264.7) and human cell lines (MRC-5, HL60, U937). This antioxidant property depends on concentration and increasing with increased amount of extracts. The results obtained in the present study indicated that the see horse (Hippocampus kuda Bleeker) is a potential source of natural antioxidant.
