There have been significant developments in the use of siRNA in the silencing of cancer-allied target proteins with substantial apoptotic effects. Nevertheless, the challenges regarding siRNA delivery for cancer therapy remain a major concern for taking these therapies successfully from laboratory to in vivostudies. Biomaterials are defined traditionally as any material that is used for either medical or dental applications that contact the host cells in any form, such as a drug carrier a device, or as a prosthesis towards the replacement of damaged tissues. The main issues to be fulfilled by the biomaterial for clinical applications are biocompatibility, bioactivity, ability to carry drug to target site, inflammatory responses and other factors based on its application. The present study focuses on the in vitroinflammatory response to the CS nanoparticles using RAW 264.7 and bone marrow derived macrophage cells. Additionally, the in vitrorelease kinetics of siRNA with varying concentrations and pH, transfection efficacy and biocompatibility were also investigated. The results of siRNA cumulative release increased at pH 5 and 3, which may be corresponding to the protonation, and a delayed release was seen at 7, which was ascribed to unprotonated amine groups inside the CS. The results of release kinetics confirmed a sustained release of siRNA from CS NPs. Considering that CS is a biocompatible polymer, it typically has little impact/damage on cells, as numerous researchers have observed during in vitroexperiments. Inflammatory studies were carried out in vitrowith RAW 264.7 and BMC cells derived from mice. The gene and protein expression studies showed that the materials might cause some slight inflammation on exposure with both RAW 264.7 and BMC cells in vitro, which is completely negligible. However, putting together the overall data it can be concluded that CS NPs can be a promising material for in vivoapplications, which is in agreement with the results of other researchers, but the only concern being its ability to carry siRNA and protect it from nuclease and other enzymatic attacks.

Background: The call for patient-focused drug development is loud and clear, as expressed in the twenty-first Century Cures Act and in recent guidelines and initiatives of regulatory agencies. Among the factors contributing to modernized drug development and improved health-care activities are easily interpretable measures of clinical benefit. In addition, special care is needed for cancer trials with time-to-event endpoints if the treatment effect is not constant over time. Objective: To quantify the potential clinical survival benefit for a new patient, would he/she be treated with the test or control treatment. Methods: We propose the predictive individual effectwhich is a patient-centric and tangible measure of clinical benefit under a wide variety of scenarios. It can be obtained by standard predictive calculations under a rank preservation assumption that has been used previously in trials with treatment switching. Results: We discuss four recent Oncology trials that cover situations with proportional as well as non-proportional hazards (delayed treatment effect or crossing of survival curves). It is shown that the predictive individual effectoffers valuable insights beyond p-values, estimates of hazard ratios or differences in median survival. Conclusion: Compared to standard statistical measures, the predictive individual effectis a direct, easily interpretable measure of clinical benefit. It facilitates communication among clinicians, patients, and other parties and should therefore be considered in addition to standard statistical results.

They can have vascular shunting mostly arteriovenous and sometimes arterioportal or portosystemic, which improves as hemangiomas involute. In contrast, congenital portosystemic shunts are developmental vascular anomalies that may go undetected for years, with significant sequelae. We describe a child with a history of multiple cutaneous and liver hemangiomas in infancy and later diagnosis of congenital portosystemic shunt. Past experience of a similar patient and a current baby followed for liver hemangiomas with portosystemic shunts, is also shared. Literature is reviewed for known association. We suggest longer-term follow-up for babies with liver hemangiomas.

In this research, optimization of the production medium to enhance tannase production by Bacillus gottheiliiM2S2 in laboratory-scale packed bed reactor was studied. Amount of substrate Triphala, moisture content, aeration rate, and fermentation period was chosen for optimization study. During one variable at a time optimization, the highest tannase activity of 0.226 U/gds was shown with Triphalaas a substrate at the fermentation period of 32 h. Furthermore, the optimum conditions predicted by response surface methodology (RSM) and genetic algorithm (GA) were found to be 11.532 g of substrate Triphala, 47.071% of the moisture content, and 1.188 L/min of an aeration rate with uppermost tannase activity of 0.262 U/gds. In addition, the single hidden layer feedforward neural network (SLFNN) and the radial basis function neural network (RBFNN) of an artificial neural network (ANN) were adopted to compare the prediction performances of the RSM and GA. It revealed that the ANN models (SLFNN, R2= 0.9930; and RBFNN, R2= 0.9949) were better predictors than the RSM (R2= 0.9864). Finally, the validation experiment exhibited 0.265 U/gds of tannase activity at the optimized conditions, which is an 11-fold increase compared to unoptimized media in shake flask.

Chaperones TAPBPR and tapasin associate with class I major histocompatibility complexes (MHC-I) to promote optimization (editing) of peptide cargo. Here, we use solution NMR to investigate the mechanism of peptide exchange. We identify TAPBPR-induced conformational changes on conserved MHC-I molecular surfaces, consistent with our independently determined X-ray structure of the complex. Dynamics present in the empty MHC-I are stabilized by TAPBPR and become progressively dampened with increasing peptide occupancy. Incoming peptides are recognized according to the global stability of the final pMHC-I product and anneal in a native-like conformation to be edited by TAPBPR. Our results demonstrate an inverse relationship between MHC-I peptide occupancy and TAPBPR binding affinity, wherein the lifetime and structural features of transiently bound peptides control the regulation of a conformational switch located near the TAPBPR binding site, which triggers TAPBPR release. These results suggest a similar mechanism for the function of tapasin in the peptide-loading complex.

ABSTRACTStaphylococcal enterotoxin B (SEB), a shock-inducing exotoxin synthesized by Staphylococcus aureus, is an important cause of food poisoning and is a class B bioterrorism agent. SEB mediates antigen-independent activation of a major subset of the T-cell population by cross-linking T-cell receptors (TCRs) with class II major histocompatibility complex (MHC-II) molecules of antigen-presenting cells, resulting in the induction of antigen independent proliferation and cytokine secretion by a significant fraction of the T-cell population. Neutralizing antibodies inhibit SEB-mediated T-cell activation by blocking the toxin's interaction with the TCR or MHC-II and provide protection against the debilitating effects of this superantigen. We derived and searched a set of monoclonal mouse anti-SEB antibodies to identify neutralizing anti-SEB antibodies that bind to different sites on the toxin. A pair of non-cross-reactive, neutralizing anti-SEB monoclonal antibodies (MAbs) was found, and a combination of these antibodies inhibited SEB-induced T-cell proliferation in a synergistic rather than merely additive manner. In order to engineer antibodies more suitable than mouse MAbs for use in humans, the genes encoding the VL and VH gene segments of a synergistically acting pair of mouse MAbs were grafted, respectively, onto genes encoding the constant regions of human Igκ and human IgG1, transfected into mammalian cells, and used to generate chimeric versions of these antibodies that had affinity and neutralization profiles essentially identical to their mouse counterparts. When tested in cultures of human peripheral blood mononuclear cells or splenocytes derived from HLA-DR3 transgenic mice, the chimeric human-mouse antibodies synergistically neutralized SEB-induced T-cell activation and cytokine production.

Ly49A, an inhibitory C-type lectin-like mouse natural killer cell receptor, functions through interaction with the major histocompatibility complex class I molecule, H-2D(d). The x-ray crystal structure of the Ly49A.H-2D(d) complex revealed that homodimeric Ly49A interacts at two distinct sites of H-2D(d): Site 1, spanning one side of the alpha1 and alpha2 helices, and Site 2, involving the alpha1, alpha2, alpha3, and beta(2)m domains. Mutants of Ly49A, H-2D(d), and beta(2)-microglobulin at intermolecular contacts and the Ly49A dimer interface were examined for binding affinity and kinetics. Although mutations at Site 1 had little affect, several at Site 2 and at the dimer interface hampered the Ly49A.H-2D(d) interaction, with no effect on gross structure or T cell receptor interaction. The region surrounding the most critical residues (in H-2D(d), Asp(122); in Ly49A, Asp(229), Ser(236), Thr(238), Arg(239), and Asp(241); and in beta(2)-microglobulin, Gln(29) and Lys(58)) of the Ly49A.H-2D(d) interface at Site 2 includes a network of water molecules, suggesting a molecular basis for allelic specificity in natural killer cell recognition.

The development of mouse models of human organ-specific autoimmune diseases has been hampered by the need to immunize mice with autoantigens in potent adjuvants. Even autoantigen-specific T cell receptor transgenic models of autoimmunity have proven to be complex as the transgenic mice frequently fail to develop disease spontaneously. We have isolated a CD4⁺ T cell clone (TxA23)that recognizes the gastric parietal cell antigen, H/K ATPase α-chain630–641, from a mouse with autoimmune gastritis that developed after thymectomy on day 3 of life. The T cell receptor α and β genes from this clone were used to generate A23 transgenic mice. All A23 transgenic animals spontaneously developed severe autoimmune gastritis, and evidence of disease was detected as early as day 10 of life. Gastritis could be transferred to immunocompromised mice with a limited number of transgenic thymocytes (10³), but as many as 10⁷ induced only mild disease in wild-type animals. Due to the complete penetrance of spontaneous disease, identity of the auto-antigen, susceptibility to immunoregulation, and close relation to autoimmune gastritis in man, A23 transgenic mice represent a unique CD4⁺ T cell-mediated disease model for understanding the multiple factors regulating organ-specific autoimmunity.

Thymectomy at day 3 of life (d3Tx) results in the development of organ-specific autoimmunity. We have recently shown that d3Tx BALB/c mice which develop autoimmune gastritis contain CD4⁺ T cells specific for the gastric parietal cell proton pump, H/K ATPase. Here, we demonstrate that freshly explanted gastric lymph node (LN) cells from d3Tx mice react significantly to the H/K ATPase α chain, but only marginally to the β chain. Two H/K ATPase-reactive T cell lines were derived from the gastric LN of d3Tx mice. Both are CD4⁺ , TCR α / β⁺ , and I-A^d restricted, and recognize distinct peptides from the H/K ATPase α chain. One cell line secretes Th1 and the other Th2 cytokines, but both are equally potent in inducing gastritis with distinct profiles of cellular infiltration in nu/nu recipient animals. Neither of the cell lines induced disease in normal BALB/c recipients and transfer of disease to nu/nu recipients was blocked by co-transfer of normal BALB/c spleen cells containing CD4⁺ CD25⁺ cells. Although CD4⁺ CD25⁺ T cells are thought to emigrate from the thymus after day 3 of life, they could be identified in LN of 2-day-old animals. The capacity of CD4⁺ CD25⁺ T cells to abrogate the pathogenic activity in vivo of both activated Th1/Th2 lines strongly suggests that this suppressor T cell population may have a therapeutic role in other models of established autoimmunity. The availability of well-characterized lines of autoantigen-specific T cells should greatly facilitate the analysis of the mechanism of action and target of the CD4⁺ CD25⁺ immunoregulatory cells.

The crystal structure of the mouse major histocompatibility complex (MHC) class I molecule H-2Ddwith an immunodominant peptide, designated P18-I10 (RGPGRAFVTI), from human immunodeficiency virus envelope glycoprotein 120 was determined at 3.2 A˚ resolution. A novel orientation of the α3 domain of Ddrelative to the α1/α2 domains results in significantly fewer contacts between α3 and β2-microglobulin compared with other MHC class I proteins. Four out of ten peptide residues (P2 Gly, P3 Pro, P5 Arg and P10 Ile) are nearly completely buried in the Ddbinding groove. This is consistent with previous findings that Ddexploits a four-residue binding motif comprising a glycine at P2, a proline at P3, a positively charged residue at P5, and a C-terminal hydrophobic residue at P9 or P10. The side-chain of P5 Arg is directed toward the floor of the predominantly hydrophobic binding groove where it forms two salt bridges and one hydrogen bond with Ddresidue Asp77. The selection of glycine at P2 appears to be due to a narrowing of the B pocket, relative to that of other class I molecules, caused by Arg66 whose side-chain folds down into the binding cleft. Residue P3 Pro of P18-I10 occupies part of pocket D, which in Ddis partially split by a prominent hydrophobic ridge in the floor of the binding groove formed by Trp97 and Trp114. Residues P6 through P9 form a solvent-exposed bulge, with P7 Phe protruding the most from the binding groove and thereby probably constituting a major site of interaction with T cell receptors. A comparison of H-2Dd/P18-I10 with other MHC class I/peptide complexes of known structure provides insights into the possible basis for the specificity of the natural killer cell receptor Ly-49A for several related class I molecules.

Quality Measurement Plan (QMP) as developed by Hoadley (1981) is a statistical method for analyzing discrete quality audit data which consist of the expected number of defects given the standard quality. The QMP is based on an empirical Bayes (EB) model of the audit sampling process. Despite its wide publicity, Hoadley's method has often been described as heuristic. In this paper we offer an hierarchical Bayes (HB) alternative to Hoadley's EB model, and overcome much of the criticism against this model. Gibbs sampling is used to implement the HB model proposed in this paper. Also, the convergence of the Gibbs sampler is monitored via the algorithm of Gelman and Rubin (1992).

Abstract:  The influence of β-chain diversity on the expressed T-cell receptor (TCR) α-chain repertoire was investigated using transgenic mice which exclusively express a single rearranged TCR β-chain gene. Analysis of these mice using α-chain-specific recombinant cDNA libraries showed that expression of the transgene-encoded β chain results in significant skewing in Tcra-V gene segment usage vs nontransgenic mice. Skewing was most pronounced towards α chains using TCRA-V segments. Sequence analysis of Tcra-V8-containing genes from transgenic T cells revealed predominant use of a single Tcra-J segment (Tcra-J24), which was not detected in Tcra-V8 containing genes isolated from nontransgenic T cells. Further analysis revealed that co-expression of Tcra-V8 with Tcra-J24 in β-transgenic mice is exhibited almost exclusively by CD4+ T cells, and is associated with a limited number of closely related N-regions. Analysis of transgenic CD8+ T cells demonstrated predominant co-expression of Tcra-V8 with another Tcra-J (Tcra-J30), together with a different, limited N-region sequence. We conclude that the composition of expressed β chains can profoundly influence the selection of companion α chains expressed in the periphery, and that α-chain N and J regions play a crucial role in discriminating between class I vs class II major histocompatibility complex (MHC)-restricted recognition. Further, these results are in agreement with recent data concerning the crystal structure of the TCR, and most consistent with a model for TCR structure in which the complementarity determining region (CDR)3α domain participates in direct contact with the MHC.

Various congenital cardiac malformations have been described in patietns with Beckwith-Wiedemann (BW) syndrome, including reversible obstructive subaortic stenosis in one patient. We herein present a case of a 2.5-year-old black boy with BW syndrome and discrete subvalvular aortic stenosis of the membraneous type. Such association of these two entities has previously not been documented.

Lactobacillus plantarum MTCC 1407 represents a valuable source of an economically attractive stable long-life tannase with potential for application in various industries. The effect of fermentation parameters such as pH, temperature and agitation speed on the growth of biomass and production of tannase using liquid medium were determined at the end of fermentation period. The optimum values of pH, reaction temperature and agitation speed for tannase activity were 6.0, 30 °C and 125 rpm respectively. The maximum tannase activity was found to be 9.29 U/mL.
