Noncontact optical nanothermometers operating within the biological transparency windows are required to study temperature-sensitive biological phenomena at the nanoscale. Nanoparticles containing rare-earth ions such as Nd3+have been reported to be efficient luminescence-based ratiometric thermometers, however often limited by poor water solubility and concentration-related quenching effects. Herein, we introduce a new type of nanothermometer, obtained by employing low-dimensional carbon nanodots (CNDs) as matrices to host Nd3+ions (NdCNDs). By means of a one-pot procedure, small (∼7–12 nm), water-soluble nanoparticles were obtained, with high (15 wt %) Nd3+loading. This stable metal-CND system features temperature-dependent photoluminescence in the second biological window (BW II) upon irradiation at 808 nm, thereby allowing accurate and reversible (heating/cooling) temperature measurements with good sensitivity and thermal resolution. The system possesses remarkable biocompatibility in vitroand promising performance at a high penetration depth in tissue models.

Circular polarization-resolved Raman scattering methods include Raman optical activity (ROA) and its derivative─surface-enhanced Raman optical activity (SEROA). These spectroscopic modalities are rapidly developing due to their high information content, stand-off capabilities, and rapid development of Raman-active chiral nanostructures. These methods enable a direct readout of the vibrational energy levels of chiral molecules, crystals, and nanostructured materials, making it possible to study complex interactions and the dynamic interfaces between them. They were shown to be particularly valuable for nano- and biotechnological fields encompassing complex particles with nanoscale chirality that combine strong scattering and intense polarization rotation. This perspective dives into recent advancements in ROA and SEROA, their distinction from surface-enhanced Raman scattering, and the potential of these information-rich label-free spectroscopies for the detection of chiral biomolecules.

The interactions between gold nanoparticles, their surface ligands and the solvent critically influence the properties of these nanoparticles. Although spectroscopic and scattering techniques have been used to investigate their ensemble structure, a comprehensive understanding of these processes at the nanoscale remains challenging. Electron microscopy makes it possible to characterize the local structure and composition but is limited by insufficient contrast, electron beam sensitivity and the requirement for ultrahigh-vacuum conditions, which prevent the investigation of dynamic aspects. Here we show that, by exploiting high-quality graphene liquid cells, we can overcome these limitations and investigate the structure of the ligand shell around gold nanoparticles and at the ligand–gold interface in a liquid environment. Using this graphene liquid cell, we visualize the anisotropy, composition and dynamics of ligand distribution on gold nanorod surfaces. Our results indicate a micellar model for surfactant organization. This work provides a reliable and direct visualization of ligand distribution around colloidal nanoparticles.

The bottom-up production of chiral gold nanomaterials holds great potential for the advancement of biosensing and nano-optics, among other applications. Reproducible preparations of colloidal nanomaterials with chiral morphology have been reported, using cosurfactants or chiral inducers such as thiolated amino acids. However, the underlying growth mechanisms for these nanomaterials remain insufficiently understood. We introduce herein a purposely devised chiral inducer, a cysteine modified with a hydrophobic chain, as a versatile chiral inducer. The amphiphilic and chiral features of this molecule provide control over the chiral morphology and the chiroptical signature of the obtained nanoparticles by simply varying the concentration of chiral inducer. These results are supported by circular dichroism and electromagnetic modeling as well as electron tomography to analyze structural evolution at the facet scale. Our observations suggest complex roles for the factors involved in chiral synthesis: the chemical nature of the chiral inducers and the influence of cosurfactants.

Despite the great progress in the colloidal syntheses of plasmonic metal nanoparticles, batch processes still face a general limitation toward achieving large-scale production and reproducibility, due to insufficient control over reagent addition conditions and other reaction parameters, such as mixing rate, temperature, etc. Flow reactors provide the opportunity to counteract the limitations of traditional batch nanomaterials synthesis. We demonstrate the use of a purposely devised millifluidic flow reactor for the seeded growth of monodisperse gold nanoparticles with high process control and batch-to-batch reproducibility. This system offers versatility toward the synthesis of various gold nanoparticle shapes and dimensions, as demonstrated here for nanospheres, nanorods, and nanostars.

Chiral ligands are considered a required ingredient during the synthesis of dissymmetric plasmonic metal nanocrystals. The mechanism behind the generation of chiral structures involves the formation of high Miller index chiral facets, induced by the adsorption of such chiral ligands. We found, however, that chirality can also evolve spontaneously, without the involvement of any chiral ligands, during the co-deposition of Au and Ag on Au nanorods. When using a specific Au/Ag ratio, phase segregation of the two metals leads to an interface within the obtained AuAg shell, which can be exposed by removing the Ag component via oxidative etching. Although a close-to-racemic mixture of chiral Au nanorods with right- and left-handedness is found in solution, electron tomography analysis evidences left- and right-handed helicities, both at the Au–Ag interface and at the exposed surface of Au NRs after Ag etching. The helicity profile of the NRs indicates dominating inclination angles in the range from 30° to 60°. Single-particle optical characterization also reveals random handedness in the plasmonic response of individual nanorods. We hypothesize that the origin of chirality is related to symmetry breaking during the co-deposition of Au and Ag, through an initial perturbation at a small region on the Au–Ag interface, which eventually leads to chiral segregation throughout the nanocrystal.

Morphological characterization of nanoparticle assemblies and hybrid nanomaterials is critical in determining their structure-property relationships as well as in the development of structures with desired properties. Electron tomography has become a widely utilized technique for the three-dimensional characterization of nanoparticle assemblies. However, the extraction of quantitative morphological parameters from the reconstructed volume can be a complex and labor-intensive task. In this study, we aim to overcome this challenge by automating the volumetric segmentation process applied to three-dimensional reconstructions of nanoparticle assemblies. The key to enabling automated characterization is to assess the performance of different volumetric segmentation methods in accurately extracting predefined quantitative descriptors for morphological characterization. In our methodology, we compare the quantitative descriptors obtained through manual segmentation with those obtained through automated segmentation methods, to evaluate their accuracy and effectiveness. To show generality, our study focuses on the characterization of assemblies of CdSe/CdS quantum dots, gold nanospheres and CdSe/CdS encapsulated in polymeric micelles, and silica-coated gold nanorods decorated with both CdSe/CdS or PbS quantum dots. We use two unsupervised segmentation algorithms: the watershed transform and the spherical Hough transform. Our results demonstrate that the choice of automated segmentation method is crucial for accurately extracting the predefined quantitative descriptors. Specifically, the spherical Hough transform exhibits superior performance in accurately extracting quantitative descriptors, such as particle size and interparticle distance, thereby allowing for an objective, efficient, and reliable volumetric segmentation of complex nanoparticle assemblies.

Gold nanorods (Au NRs) are an exceptionally promising tool in nanotechnology due to three key factors: (i) their strong interaction with electromagnetic radiation, stemming from their plasmonic nature, (ii) the ease with which the resonance frequency of their longitudinal plasmon mode can be tuned from the visible to the near-infrared region of the electromagnetic spectrum based on their aspect ratio, and (iii) their simple and cost-effective preparation through seed-mediated chemical growth. In this synthetic method, surfactants play a critical role in controlling the size, shape, and colloidal stability of Au NRs. For example, surfactants can stabilize specific crystallographic facets during the formation of Au NRs, leading to the formation of NRs with specific morphologies.

The mechanical properties and stability of metal nanoparticle colloids under high-pressure conditions are investigated by means of optical extinction spectroscopy and small-angle X-ray scattering (SAXS), for colloidal dispersions of gold nanorods and gold nanospheres. SAXS allows us to follow in situ the structural evolution of the nanoparticles induced by pressure, regarding both nanoparticle size and shape (form factor) and their aggregation through the interparticle correlation function S(q) (structure factor). The observed behavior changes under hydrostatic and nonhydrostatic conditions are discussed in terms of liquid solidification processes yielding nanoparticle aggregation. We show that pressure-induced diffusion and aggregation of gold nanorods take place after solidification of the solvent. The effect of nanoparticle shape on the aggregation process is additionally discussed.

Micro-and nanorobots have the potential to perform non-invasive drug delivery, sensing, and surgery in living organisms, with the aid of diverse medical imaging techniques. To perform such actions, microrobots require high spatiotemporal resolution tracking with real-time closed-loop feedback. To that end,  photoacoustic imaging has appeared as a promising technique for imaging microrobots in deep tissue with higher molecular specificity and contrast. Here, we present different strategies to track magnetically-driven micromotors with improved contrast and specificity using dedicated contrast agents (Au nanorods and nanostars). Furthermore, we discuss the possibility of improving the light absorption properties of the employed nanomaterials considering possible light scattering and coupling to the underlying metal-oxide layers on the micromotor’s surface. For that, 2D COMSOL simulation and experimental results were correlated, confirming that an increased spacing between the Au-nanostructures and the increase of thickness of the underlying oxide layer lead to enhanced light absorption and preservation of the characteristic absorption peak. These characteristics are important when visualizing the micromotors in a complex in vivo environment, to distinguish them from the light absorption properties of the surrounding natural chromophores.

In principle, designing and synthesizing almost any class of colloidal crystal is possible. Nonetheless, the deliberate and rational formation of colloidal quasicrystals has been difficult to achieve. Here we describe the assembly of colloidal quasicrystals by exploiting the geometry of nanoscale decahedra and the programmable bonding characteristics of DNA immobilized on their facets. This process is enthalpy-driven, works over a range of particle sizes and DNA lengths, and is made possible by the energetic preference of the system to maximize DNA duplex formation and favour facet alignment, generating local five- and six-coordinated motifs. This class of axial structures is defined by a square–triangle tiling with rhombus defects and successive on-average quasiperiodic layers exhibiting stacking disorder which provides the entropy necessary for thermodynamic stability. Taken together, these results establish an engineering milestone in the deliberate design of programmable matter.

Chiral plasmonics is a rapidly developing field where breakthroughs and unsolved problems coexist. We have recently reported binary surfactant-assisted seeded growth of chiral gold nanorods (Au NRs) with high chiroptical activity. Such a seeded-growth process involves the use of a chiral cosurfactant that induces micellar helicity, in turn driving the transition from achiral to chiral Au NRs, from both the morphological and the optical points of view. We report herein a detailed study on both transitions, which reveals intermediate states that were hidden so far. The correlation between structure and optical response is carefully analyzed, including the (linear and CD) spectral evolution over time, electron tomography, the impact of NR dimensions on their optical response, the variation of the absorption-to-scattering ratio during the evolution from achiral to chiral Au NRs, and the near-field enhancement related to chiral plasmon modes. Our findings provide further understanding of the growth process of chiral Au NRs and the associated optical changes, which will facilitate further study and applications of chiral nanomaterials.

The effects of nonhydrostatic pressure on the morphology and stability of gold nanorods (AuNRs) and nanospheres (AuNSs) in 4:1 methanol–ethanol mixtures were studied by optical absorption spectroscopy and transmission electron microscopy at pressures of up to 23 and 30 GPa, respectively. Solvent solidification and associated nonhydrostatic stresses were found to have a negligible effect on the shape and size of AuNSs. On the contrary, while AuNRs maintained their initial morphology in the hydrostatic range, the uniaxial stress component induced under nonhydrostatic conditions had a shearing effect on the AuNRs, breaking them into smaller particles. Interestingly, colloidal stability was maintained in all cases, and the particles showed no sign of aggregation, despite the severe nonhydrostatic conditions to which both AuNR and AuNS colloids were subjected.

Surface-enhanced infrared absorption (SEIRA) leads to a largely improved detection of polar molecules, compared to standard infrared absorption. The enhancement principle is based on localized surface plasmon resonances of the substrate, which match the frequency of molecular vibrations in the analyte of interest. Therefore, in practical terms, the SEIRA sensor needs to be tailored to each specific analyte. We review SEIRA sensors based on metal plasmonics for the detection of biomolecules such as DNA, proteins, and lipids. We further focus this review on chemical SEIRA sensors, with potential applications in quality control, as well as on the improvement in sensor geometry that led to the development of multiresonant SEIRA substrates as sensors for multiple analytes. Finally, we give an introduction into the integration of SEIRA sensors with surface-enhanced Raman scattering (SERS).

Anisotropic plasmonic nanoparticles have found applications in a wide range of scientific and technological fields, including medicine, energy storage and production, ultrasensitive sensing, catalysis, and photonics. These colloids owe their all-around success in such different scenarios to the development of rapid, scalable, and rational synthetic schemes. Gold nanotriangles (AuNTs), geometrically termed truncated triangular bipyramids, have attracted the attention of the scientific community because of their combination of well-defined crystallography, anisotropic plasmon spatial distribution, sharp tips that favor the generation of high electric fields, atomically flat surfaces, and a wide spectral tunability within the visible and infrared ranges combined with narrow bandwidths of their plasmon resonances. In this context, we previously reported a procedure for the production of AuNTs, based on a seed-mediated approach that guarantees batch-to-batch reproducibility in both size (within 5 nm in edge-length) and extinction spectra (down to 1 nm precision). The protocol involves numerous synthetic steps, and reproducibility requires awareness and familiarity with several details, which are usually learned through practice and repetition and may not always be intuitive on the basis of standard experimental protocols. We provide herein an enhanced protocol with full details and demonstration videos, which we expect will further foster the utilization of this fascinating type of anisotropic nanomaterials by researchers who are less experienced in the preparation and handling of gold colloids.

The density and compressibility of nanoscale gold (both nanospheres and nanorods) and microscale gold (bulk) were simultaneously studied by X-ray diffraction with synchrotron radiation up to 30 GPa. Colloidal stability (aggregation state and nanoparticle shape and size) in both hydrostatic and nonhydrostatic regions was monitored by small-angle X-ray scattering. We demonstrate that nonhydrostatic effects due to solvent solidification had a negligible influence on the stability of the nanoparticles. Conversely, nonhydrostatic effects produced axial stresses on the nanoparticle up to a factor 10× higher than those on the bulk metal. Working under hydrostatic conditions (liquid solution), we determined the equation of state of individual nanoparticles. From the values of the lattice parameter and bulk modulus, we found that gold nanoparticles are slightly denser (0.3%) and stiffer (2%) than bulk gold: V0= 67.65(3) Å3, K0= 170(3)GPa, at zero pressure.

Monitoring dynamic processes in complex cellular environments requires the integration of uniformly distributed detectors within such three-dimensional (3D) networks, to an extent that the sensor could provide real-time information on nearby perturbations in a non-invasive manner. In this context, the development of 3D-printed structures that can function as both sensors and cell culture platforms emerges as a promising strategy, not only for mimicking a specific cell niche but also toward identifying its characteristic physicochemical conditions, such as concentration gradients. We present herein a 3D cancer model that incorporates a hydrogel-based scaffold containing gold nanorods. In addition to sustaining cell growth, the printed nanocomposite inks display the ability to uncover drug diffusion profiles by surface-enhanced Raman scattering, with high spatiotemporal resolution. We additionally demonstrate that the acquired information could pave the way to designing novel strategies for drug discovery in cancer therapy, through correlation of drug diffusion with cell death.

The incidence and mortality of cancer demand more innovative approaches and combination therapies to increase treatment efficacy and decrease off-target side effects. We describe a boron-rich nanoparticle composite with potential applications in both boron neutron capture therapy (BNCT) and photothermal therapy (PTT). Our strategy is based on gold nanorods (AuNRs) stabilized with polyethylene glycol and functionalized with the water-soluble complex cobalt bis(dicarbollide) ([3,3′-Co(1,2-C2B9H11)2]−), commonly known as COSAN. Radiolabeling with the positron emitter copper-64 (64Cu) enabled in vivotracking using positron emission tomography imaging. 64Cu-labeled multifunctionalized AuNRs proved to be radiochemically stable and capable of being accumulated in the tumor after intravenous administration in a mouse xenograft model of gastrointestinal cancer. The resulting multifunctional AuNRs showed high biocompatibility and the capacity to induce local heating under external stimulation and trigger cell death in heterogeneous cancer spheroids as well as the capacity to decrease cell viability under neutron irradiation in cancer cells. These results position our nanoconjugates as suitable candidates for combined BNCT/PTT therapies.

The development of optical nanothermometers operating in the near-infrared (NIR) is of high relevance toward temperature measurements in biological systems. We propose herein the use of Nd3+-doped lanthanum oxychloride nanocrystals as an efficient system with intense photoluminescence under NIR irradiation in the first biological transparency window and emission in the second biological window with excellent emission stability over time under 808 nm excitation, regardless of Nd3+concentration, which can be considered as a particular strength of our system. Additionally, surface passivation through overgrowth of an inert LaOCl shell around optically active LaOCl/Nd3+cores was found to further enhance the photoluminescence intensity and also the lifetime of the 1066 nm, 4F3/2to 4I11/2transition, without affecting its (ratiometric) sensitivity toward temperature changes. As required for biological applications, we show that the obtained (initially hydrophobic) nanocrystals can be readily transferred into aqueous solvents with high, long-term stability, through either ligand exchange or encapsulation with an amphiphilic polymer.

The development of continuous monitoring systems requires in situsensors that are capable of screening multiple chemical species and providing real-time information. Such in situmeasurements, in which the sample is analyzed at the point of interest, are hindered by underlying problems derived from the recording of successive measurements within complex environments. In this context, surface-enhanced Raman scattering (SERS) spectroscopy appears as a noninvasive technology with the ability of identifying low concentrations of chemical species as well as resolving dynamic processes under different conditions. To this aim, the technique requires the use of a plasmonic substrate, typically made of nanostructured metals such as gold or silver, to enhance the Raman signal of adsorbed molecules (the analyte). However, a common source of uncertainty in real-time SERS measurements originates from the irreversible adsorption of (analyte) molecules onto the plasmonic substrate, which may interfere in subsequent measurements. This so-called “SERS memory effect” leads to measurements that do not accurately reflect varying conditions of the sample over time. We introduce herein the design of plasmonic substrates involving a nonpermeable poly(lactic-co-glycolic acid) (PLGA) thin layer on top of the plasmonic nanostructure, toward controlling the adsorption of molecules at different times. The polymeric layer can be locally degraded by irradiation with the same laser used for SERS measurements (albeit at a higher fluence), thereby creating a micrometer-sized window on the plasmonic substrate available to molecules present in solution at a selected measurement time. Using SERS substrates coated with such thermolabile polymer layers, we demonstrate the possibility of performing over 10,000 consecutive measurements per substrate as well as accurate continuous monitoring of analytes in microfluidic channels and biological systems.

Regulation of enzymes is highly relevant toward orchestrating cell-free and stepwise biotransformations, thereby maximizing their overall performance. Plasmonic nanomaterials offer a great opportunity to tune the functionality of enzymes through their remarkable optical properties. Localized surface plasmon resonances (LSPR) can be used to modify chemical transformations at the nanomaterial’s surface, upon light irradiation. Incident light can promote energetic processes, which may be related to an increase of local temperature (photothermal effects) but also to effects triggered by generated hotspots or hot electrons (photoelectronic effects). As a consequence, light irradiation of the protein–nanomaterial interface affects enzyme functionality. To harness these effects to finely and remotely regulate enzyme activity, the physicochemical features of the nanomaterial, properties of the incident light, and parameters governing molecular interactions must be optimized. In this Perspective, we discuss relevant examples that illustrate the use of plasmonic nanoparticles to control enzyme function through LSPR excitation. Finally, we also highlight the importance of expanding the use of plasmonic nanomaterials to the immobilization of multienzyme systems for light-driven regulation of cell-free biosynthetic pathways. Although this concept is living its infancy, we encourage the scientific community to advance in the development of novel light-controlled biocatalytic plasmonic nanoconjugates and explore their application in biosensing, applied biocatalysis, and biomedicine.

Laser trapping at an interface provides a unique platform for assembling novel multiparticle-based optical matter that extends well beyond the irradiated area. Optical binding, resulting from the resonantly scattered photons by gold nanoparticles through the dipolar scattering mode, serves as the primary force for supporting the cohesion of the particles in these optically induced assemblies, which is interpreted in view of the formation of an optical binding network. Unfortunately, the dipole scattering mode is restricted to a narrow range of experimental conditions, limiting its use to specific trapping laser wavelengths and particle sizes. To address this limitation, exploring higher multipole scattering modes, such as the quadrupole, could provide a broader range of experimental conditions. In this work, we will describe how the higher quadrupole scattering mode influences the formation of optical binding and the possibility of extending the optical potential outside the irradiated area for constructing large optically induced assemblies.

Breast cancer stem cells (CSCs) play a pivotal role in therapy resistance and tumor relapse, emphasizing the need for reliable in vitromodels that recapitulate the complexity of the CSC tumor microenvironment to accelerate drug discovery. We present a bioprinted breast CSC tumor-stroma model incorporating triple-negative breast CSCs (TNB-CSCs) and stromal cells (human breast fibroblasts), within a breast-derived decellularized extracellular matrix bioink. Comparison of molecular signatures in this model with different clinical subtypes of bioprinted tumor-stroma models unveils a unique molecular profile for artificial CSC tumor models. We additionally demonstrate that the model can recapitulate the invasive potential of TNB-CSC. Surface-enhanced Raman scattering imaging allowed us to monitor the invasive potential of tumor cells in deep z-axis planes, thereby overcoming the depth-imaging limitations of confocal fluorescence microscopy. As a proof-of-concept application, we conducted high-throughput drug testing analysis to assess the efficacy of CSC-targeted therapy in combination with conventional chemotherapeutic compounds. The results highlight the usefulness of tumor-stroma models as a promising drug-screening platform, providing insights into therapeutic efficacy against CSC populations resistant to conventional therapies.

Integrating cavity-enhanced colloidal quantum dots (QDs) into photonic chip devices would be transformative for advancing room-temperature optoelectronic and quantum photonic technologies. However, issues with efficiency, stability, and cost remain formidable challenges to reach the single antenna limit. Here, we present a bottom-up approach that delivers single QD-plasmonic nanoantennas with electrical addressability. These QD nanojunctions exhibit robust photoresponse characteristics, with plasmonically enhanced photocurrent spectra matching the QD solution absorption. We demonstrate electroluminescence from individual plasmonic nanoantennas, extending the device lifetime beyond 40 min by utilizing a 3 nm electron-blocking polymer layer. In addition, we reveal a giant voltage-dependent redshift of up to 62 meV due to the quantum-confined Stark effect and determine the exciton polarizability of the CdSe QD monolayer to be 4 × 10–5meV/(kV/cm)2. These developments provide a foundation for accessing scalable quantum light sources and high-speed, tunable optoelectronic systems operating under ambient conditions.

Nanocomposites comprising hydrogels and plasmonic nanoparticles are attractive materials for tissue engineering, bioimaging, and biosensing. These materials are usually fabricated by adding colloidal nanoparticles to the uncured polymer mixture and thus require time-consuming presynthesis, purification, and ligand-exchange steps. Herein, we introduce approaches for rapid synthesis of gold nanostars (AuNSt) in situon hydrogel substrates, including those with complex three-dimensional (3D) features. These methods enable selective AuNSt growth at the surface of the substrate, and the growth conditions can be tuned to tailor the nanoparticle size and density (coverage). We additionally demonstrate proof-of-concept applications of these nanocomposites for SERS sensing and imaging. High surface coverage with AuNSt enabled 1–2 orders of magnitude higher SERS signals compared to plasmonic hydrogels loaded with premade colloids. Importantly, AuNSt can be prepared without the addition of any potentially cytotoxic surfactants, thereby ensuring a high biocompatibility. Overall, in situgrowth becomes a versatile and straightforward approach for the fabrication of plasmonic biomaterials.

Silver, king among plasmonic materials, features low inelastic absorption in the visible-infrared (vis-IR) spectral region compared to other metals. In contrast, copper is commonly regarded as too lossy for actual applications. Here, we demonstrate vis-IR plasmons with quality factors >60 in long copper nanowires (NWs), as determined by electron energy-loss spectroscopy. We explain this result by noticing that most of the electromagnetic energy in these plasmons lies outside the metal, thus becoming less sensitive to inelastic absorption. Measurements for silver and copper NWs of different diameters allow us to elucidate the relative importance of radiative and nonradiative losses in plasmons spanning a wide spectral range down to <20 meV. Thermal population of such low-energy modes becomes significant and generates electron energy gains associated with plasmon absorption, rendering an experimental determination of the NW temperature. Copper is therefore emerging as an attractive, cheap, abundant material platform for high-quality plasmonics in elongated nanostructures.

X-ray-based analytics are routinely applied in many fields, including physics, chemistry, materials science, and engineering. The full potential of such techniques in the life sciences and medicine, however, has not yet been fully exploited. We highlight current and upcoming advances in this direction. We describe different X-ray-based methodologies (including those performed at synchrotron light sources and X-ray free-electron lasers) and their potentials for application to investigate the nano–bio interface. The discussion is predominantly guided by asking how such methods could better help to understand and to improve nanoparticle-based drug delivery, though the concepts also apply to nano–bio interactions in general. We discuss current limitations and how they might be overcome, particularly for future use in vivo.

The detection of cancer biomarkers at an early stage of tumor development is vital for effective diagnosis and treatment of cancer. Current diagnostic tools can often detect cancer only when the biomarker levels are already too high, so that the tumors have spread and treatments are less effective. It is urgent therefore to develop highly sensitive assays for the detection of such biomarkers at the lowest possible concentration. In this context, we developed a sandwich immunoassay based on surface-enhanced Raman scattering (SERS) for the ultrasensitive detection of α-fetoprotein (AFP), which is typically present in human serum as a biomarker indicative of early stages of hepatocellular carcinoma. In the immunoassay design, molybdenum disulfide (MoS2) modified with a monoclonal antibody was used as a capture probe for AFP. A secondary antibody linked to an SERS-encoded nanoparticle was employed as the Raman signal reporter, that is, the transducer for AFP detection. The sandwich immunocomplex “capture probe/target/SERS tag” was deposited on a silicon wafer and decorated with silver-coated gold nanocubes to increase the density of “hot spots” on the surface of the immunosensor. The developed SERS immunosensor exhibits a wide linear detection range (1 pg mL–1to 10 ng mL–1) with a limit of detection as low as 0.03 pg mL–1toward AFP with good reproducibility (RSD < 6%) and stability. These parameters demonstrate that the proposed immunosensor has the potential to be used as an analytical platform for the detection of early-stage cancer biomarkers in clinical applications.

Plasmonically enhanced optical dichroism has attracted substantial interest for its application in optical sensing, where the interplay between chirality emanating from both molecules and plasmon-supporting structures has been regarded as a critical ingredient. Here, we experimentally demonstrate that suitably self-assembled achiral plasmonic nanostructures produce a high degree of enhancement in the optical dichroism observed from chiral molecules placed in their vicinity. Specifically, we identify a near-field enhancement associated with plasmonic hotpots as the mechanism enabling our observation of visible-NIR circular dichroism emanating from small amounts of chiral molecules. Our structures consist of linear arrays of gold nanorods obtained by introducing chiral anionic surfactants, such as modified bile salts, which lead to selective destabilization of a cetyltrimethylammonium bromide coating layer on Au nanorods, thereby promoting a tip-to-tip oriented assembly. The proposed mechanism of plasmonically-enhanced circular dichroism is supported by deriving a simple, yet general theoretical formalism that confirms the observed results, revealing the role of optical hotspots at the gaps of linear tip-to-tip nanorod assemblies as the origin of enhancement in the dichroism from chiral molecules. Importantly, it is the refractive rather than the absorption-mediated chiral response of the molecules that produces dichroism in the visible-NIR plasmonic regime, far from their UV absorption resonances. The observed self-assembly mechanism suggests that chiral analytes not directly interacting with the nanorod surfaces, but just able to induce tip-to-tip aggregation, can be revealed by a CD signature in the plasmonic region, thereby supporting potential applications in ultrasensitive analysis.

Raman-encoded gold nanoparticles (NPs) have been widely employed as photostable multifunctional probes for sensing, bioimaging, multiplex diagnostics, and surface-enhanced Raman scattering (SERS)-guided tumor therapy. We report a strategy toward obtaining a particularly large library of Au nanocapsules encoded with Raman codes defined by the combination of different thiol-free Raman reporters, encapsulated at defined molar ratios. The fabrication of SERS tags with tailored size and predefined codes is based on the in situincorporation of Raman reporter molecules inside Au nanocapsules during their formation viagalvanic replacement coupled to seeded growth on Ag NPs. The hole-free closed-shell structure of the nanocapsules is confirmed by electron tomography. The unusually wide encoding possibilities of the obtained SERS tags are investigated by means of either wavenumber-based encoding or Raman frequency combined with signal intensity, leading to an outstanding performance as exemplified by 26 and 54 different codes, respectively. We additionally demonstrate that encoded nanocapsules can be readily bioconjugated with antibodies for applications such as SERS-based targeted cell imaging and phenotyping.

Solution-phase self-assembly of anisotropic nanoparticles into complex 2D and 3D assemblies is one of the most promising strategies toward obtaining nanoparticle-based materials and devices with unique optical properties at the macroscale. However, controlling this process with single-particle precision is highly demanding, mostly due to insufficient understanding of the self-assembly process at the nanoscale. We report the use of in situ environmental scanning transmission electron microscopy (WetSTEM), combined with UV/vis spectroscopy, small-angle X-ray diffraction (SAXRD) and multiscale modeling, to draw a detailed picture of the dynamics of vertically aligned assemblies of gold nanorods. Detailed understanding of the self-assembly/disassembly mechanisms is obtained from real-time observations, which provide direct evidence of the colloidal stability of side-to-side nanorod clusters. Structural details and the forces governing the disassembly process are revealed with single particle resolution as well as in bulk samples, by combined experimental and theoretical modeling. In particular, this study provides unique information on the evolution of the orientational order of nanorods within side-to-side 2D assemblies and shows that both electrostatic (at the nanoscale) and thermal (in bulk) stimuli can be used to drive the process. These results not only give insight into the interactions between nanorods and the stability of their assemblies, thereby assisting the design of ordered, anisotropic nanomaterials but also broaden the available toolbox for in situ tracking of nanoparticle behavior at the single-particle level.

Mirror symmetry breaking in materials is a fascinating phenomenon that has practical implications for various optoelectronic technologies. Chiral plasmonic materials are particularly appealing due to their strong and specific interactions with light. In this work we broaden the portfolio of available strategies toward the preparation of chiral plasmonic assemblies, by applying the principles of chirality synchronization—a phenomenon known for small molecules, which results in the formation of chiral domains from transiently chiral molecules. We report the controlled cocrystallization of 23 nm gold nanoparticles and liquid crystal molecules yielding domains made of highly ordered, helical nanofibers, preferentially twisted to the right or to the left within each domain. We confirmed that such micrometer sized domains exhibit strong, far-field circular dichroism (CD) signals, even though the bulk material is racemic. We further highlight the potential of the proposed approach to realize chiral plasmonic thin films by using a mechanical chirality discrimination method. Toward this end, we developed a rapid CD imaging technique based on the use of polarized light optical microscopy (POM), which enabled probing the CD signal with micrometer-scale resolution, despite of linear dichroism and birefringence in the sample. The developed methodology allows us to extend intrinsically local effects of chiral synchronization to the macroscopic scale, thereby broadening the available tools for chirality manipulation in chiral plasmonic systems.

Ultrafast laser irradiation can induce morphological and structural changes in plasmonic nanoparticles. Gold nanorods (Au NRs), in particular, can be welded together upon irradiation with femtosecond laser pulses, leading to dimers and trimers through the formation of necks between individual nanorods. We used electron tomography to determine the 3D (atomic) structure at such necks for representative welding geometries and to characterize the induced defects. The spatial distribution of localized surface plasmon modes for different welding configurations was assessed by electron energy loss spectroscopy. Additionally, we were able to directly compare the plasmon line width of single-crystalline and welded Au NRs with single defects at the same resonance energy, thus making a direct link between the structural and plasmonic properties. In this manner, we show that the occurrence of (single) defects results in significant plasmon broadening.

Monitoring chemical reactions that occur in small spaces or confined environments is challenging. Surface-enhanced Raman scattering (SERS) spectroscopy offers the unique possibility to monitor spectral changes with high sensitivity and time resolution. Herein, we report the application of composite mesoporous TiO2films loaded with Ag nanoparticles (NPs) to track in situchemical processes in real time. In particular, the AgNPs@TiO2system was employed to monitor two chemical reactions: one occurring on the Ag NPs surface and another taking place in the surrounding solution. In the first case, we monitored the decarboxylation reaction of 4-mercaptobenzoic acid on Ag NPs, which allowed us to identify the conditions that favor it. In the second case, we studied the pH evolution in the nanocavities during a homogeneous alkalization process driven by chloride-assisted glycidol rupture (the Epoxide Route) and compared it with pH measurements by conventional techniques. We therefore demonstrated that the proposed nanodevice provides an excellent performance to monitor dynamic processes occurring either inside the material or in the solution in which it is immersed.

Silver is arguably the best plasmonic material in terms of optical performance. However, wide application of Ag and Ag-containing nanoparticles is usually hindered by two major drawbacks, namely, chemical degradation and cytotoxicity. We report herein a synthetic method for highly monodisperse polymer-coated Ag nanorods, which are thereby protected against external stimuli (oxidation, light, heat) and are noncytotoxic to various cell lines. The monodispersity of Ag nanorods endows them with narrow plasmon bands, which are tunable into the near-infrared biological transparency window, thus facilitating application in bioanalytical and therapeutic techniques. We demonstrate intracellular surface-enhanced Raman scattering (SERS) imaging using Ag nanorods encoded with five different Raman reporter molecules. Encoded Ag nanorods display long-term stability in terms of size, shape, optical response, and SERS signal. Our results help eliminate concerns of instability and cytotoxicity in the application of Ag-containing nanoparticles with enhanced optical response, toward the development of bioapplications.

When nanoparticles (NPs) are exposed to biological media, proteins are adsorbed, forming a so-called protein corona (PC). This cloud of protein aggregates hampers the targeting and transport capabilities of the NPs, thereby compromising their biomedical applications. Therefore, there is a high interest in the development of technologies that allow control over PC formation, as this would provide a handle to manipulate NPs in biological fluids. We present a strategy that enables the reversible disruption of the PC using external stimuli, thereby allowing a precise regulation of NP cellular uptake. The approach, demonstrated for gold nanoparticles (AuNPs), is based on a biorthogonal, supramolecular host–guest interactions between an anionic dye bound to the AuNP surface and a positively charged macromolecular cage. This supramolecular complex effectively behaves as a zwitterionic NP ligand, which is able not only to prevent PC formation but also to disrupt a previously formed hard corona. With this supramolecular stimulus, the cellular internalization of AuNPs can be enhanced by up to 30-fold in some cases, and even NP cellular uptake in phagocytic cells can be regulated. Additionally, we demonstrate that the conditional cell uptake of purposely designed gold nanorods can be used to selectively enhance photothermal cell death.
