Uncertainty exists about whether lowering systolic blood pressure to less than 120 mm Hg is superior to that of less than 140 mm Hg, particularly in patients with diabetes and patients with previous stroke.

Proton pump inhibitors (PPIs) differ in onset of action and bioavailability. This trial was conducted to investigate the pharmacokinetics and pharmacodynamics of an immediate‐release capsule formulation containing lansoprazole 30 mg and sodium bicarbonate 1100 mg (T preparation) in healthy Chinese subjects. This was an open, single‐center, randomized, single and multiple oral doses, and two‐period crossover study in 30 healthy subjects. After single‐ and multiple‐dose oral administration, blood samples were obtained and lansoprazole concentration in serum was measured for pharmacokinetic analysis. Meanwhile, the intragastric pH was monitored continuously to evaluate the pharmacodynamics of the investigational drugs. The Tmaxof the T preparation was 0.5 hours, while the Tmaxof the R preparation was 1.5 hours after multiple doses, which indicated that the absorption speed of the T preparation was significantly faster than that of the R preparation. The same characteristics also existed after single‐dose administration. The area under the curve (AUC)ssof the T preparation was bio‐equivalent to that of the R preparation under steady state. The time percentage of intragastric pH > 4.0 for the T preparation was higher than that of the R preparation after 1 hour for both single‐ and multiple‐dose. It suggested compared with R preparation, the time percentage of intragastric pH > 4.0 met the criteria for superiority after 1 hour administration for the T preparation. In addition, no serious adverse events occurred in this study. Across this study, the T preparation was better than the R preparation at improving drug absorption and increasing intragastric pH, and had a favorable safety profile.

Polysaccharide nanocrystals have led to the development of multifunctional and sustainable materials, but most are glucose-based carbohydrates derived from valuable natural sources. Here, we present a top-down strategy that enables one, for the first time, to isolate xylose-based hemicellulose nanocrystals from available industrial biowastes. By leveraging the selective oxidation of alkaline periodate, as high as 34 wt % solid yield is accessible. The hemicellulose nanocrystals exhibit platelet-like shapes (10–20 nm thickness, 30–80 nm wide), crystalline features, and superior dispersibility in water. We also showcase their successful interface applications for one-dimensional (1D) carbon nanotube (CNT) nanoinks and two-dimensional (2D) transition-metal dichalcogenide (TMD) nanozymes, which are comparable to the traditional cellulose nanocrystals. The scalable, low-cost, and sustainable hemicellulose nanocrystals can be envisioned to provide an alternative for glucose-based polysaccharide nanocrystals, as well as hold promise for the high-value utilization of biowastes.

Delayed or non-healing of diabetic wounds is a significant complication, often attributed to high glucose-induced M1 macrophage accumulation, impaired angiogenesis, and reactive oxygen species (ROS) buildup. Addressing this, we introduced a strontium polyphenol network microneedle patch (SrC-MPNs@MN-PP) for percutaneous drug delivery. This patch, formulated with polymer poly(γ-glutamic acid) (γ-PGA) and epsilon-poly-l-lysine (ε-PLL), incorporates strontium polyphenol networks (SrC-MPNs). The release of chlorogenic acid (CGA) from SrC-MPNs not only neutralizes ROS, but strontium ions also foster angiogenesis. Consequently, SrC-MPNs@MN-PP can ameliorate the diabetic wound microenvironment and expedite healing.

ABSTRACTLong non-coding RNAs (lncRNAs) are closely associated with the development of lung adenocarcinoma (LADC). The present study focused on the role of LINC00960 in LADC. miRNA and mRNA expression levels were detected using quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Cellular functions were evaluated by MTT, colony formation, and Transwell assays, respectively. LINC00960 Luciferase and RNA pull-down assays were performed to clarify the interaction between miR-124a and LINC00960 or Recombinant Sphingosine Kinase 1 (SphK1). We observed that LINC00960 was overexpressed in LADC tumor tissues and cell lines. LINC00960 knockdown suppressed the proliferation, migration, and invasion of LADC cells. Moreover, LINC00960 sponged miR-124a to inhibit the SphK1/S1P pathway in LADC cells. LINC00960 knockdown markedly reduced the rate of tumor growth. The luciferase reporter assay results demonstrated an interaction between miR-124a and LINC00960 or SphK1. This interaction was confirmed using the RNA pull-down assay. In addition, miR-124a downregulation or SphK1 upregulation reversed the inhibitory effects of LINC00960 knockdown on cellular functions of LADC cells, suggesting that LINC00960 may be a potential therapeutic biomarker for LADC via the miR-124a/SphK1 axis. Accordingly, LINC00960 may be a potential therapeutic biomarker for LADC.

Biomolecular condensates comprised of specific proteins and nucleic acids are now recognized as one of the key organizing mechanisms in eukaryotic cells. However, the specific roles played by the nucleic acid secondary structure and sequence in biomolecular phase separation are still not clear. Here, utilizing giant membrane vesicles (GMVs) as a protocell model, we found that single-stranded DNA (ssDNA) with a parallel G-quadruplex structure could functionally cooperate with a G-quadruplex-binding protein to form speckle-like puncta inside the GMVs. The clustering behavior is dependent on the structural diversity of G-quadruplexes, and the reversible clustering behavior implicated a new pathway in dynamically regulating the formation of biomolecular condensates. This finding represents a potential link between G-quadruplex-binding proteins and the resulting G-quadruplex-mediated biomolecular phase separation, which would gain insight into a wide range of biological processes associated with nucleic acid-modulated phase separation inside living cells.

Engineering cell-derived nanovesicles with active-targeting ligands is an important strategy to enhance the targeting efficiency. However, the enhanced binding capability to targeting cells also leads to the binding with nontarget cells that share the same biomarkers. DNA-based logic gate is a kind of molecular system that responds to chemical inputs by generating output signals, and the relationship between the input and the output is based on a certain logic. Thus, the DNA-based logic gate could provide a new approach to improve the delivery efficiency of the nanovesicle. In this work, we developed a DNA logic-gated module that coupled two tumor cell-targeting factors (e.g., low pH and a tumor cell biomarker) in a Boolean manner. Immobilization of this module on the surface of the nanovesicle enables the nanovesicle to sense tumor cell-targeting factors and regard these cues as inputs AND logic gate. With the guide of DNA-based logic gate, gold carbon dots (GCDs) encapsulated within nanovesicles were delivered into target cells, and then the intracellular redox status variation was reflected by fluorescence change of GCDs. Overall, we developed DNA logic-gated nanovesicles that contract different targeting factors into a unique tag for target cells. This facile functionalization strategy can pave the way for constructing smart nanovesicles and would broaden their application in the field of precision medicine and personalized treatment.

Quantitative determination of tetracycline (TC) in environment and foods is of great importance, as excessive residues might have negative effects on human health and environmental risks. Herein, a self-powered molecularly imprinted photoelectrochemical (PEC) sensor based on the ZnO/C photoanode and the Fe-doped CuBi2O4(CBFO) photocathode is developed for the sensitive detection of TC. The photocathodic current can be amplified by the efficient electron transfer caused by the Fermi energy level gap between the photoanode and photocathode. Furthermore, molecularly imprinted polymers (MIPs) at photocathode can selectivity identify the TC templates and thus improve the specificity. Under the optimal conditions, the sensor has a linear range of 10‒2-1.0 × 105nmol/L, and a limit of detection (LOD) of 0.007 nmol/L (S/N = 3). More crucially, the milk sample detection is carried out using the as-prepared sensor, and the outcome is satisfactory. The research gives us a novel sensing platform for quick and accurate antibiotic (like TC) in environment and food monitoring.

We have developed a DNA nanoprobe for adenosine triphosphate (ATP) sensing in living cells, based on the split aptamer and the DNA triangular prism (TP). In which nucleic acid aptamer was split into two fragments, the stem of the split aptamer was respectively labeled donor and acceptor fluorophores that underwent a fluorescence resonance energy transfer if two ATP molecules were bound as target molecule to the recognition module. Hence, ATP as a target induced the self-assembly of split aptamer fragments and thereby brought the dual fluorophores into close proximity for high fluorescence resonance energy transfer (FRET) efficiency. In the in vitro assay, an almost 5-fold increase in FA/FDsignal was observed, the fluorescence emission ratio was found to be linear with the concentration of ATP in the range of 0.03–2 mM, and the nanoprobe was highly selective toward ATP. For the strong protecting capability to nucleic acids from enzymatic cleavage and the excellent biocompatibility of the TP, the DNA TP nanoprobe exhibited high cellular permeability, fast response, and successfully realized “FRET-off” to “FRET-on” sensing of ATP in living cells. Moreover, the intracellular imaging experiments indicated that the DNA TP nanoprobe could effectively detect ATP and distinguish among changes of ATP levels in living cells. More importantly, using of the split aptamer and the FRET-off to FRET-on sensing mechanism could efficiently avoid false-positive signals. This design provided a strategy to develop biosensors based on the DNA nanostructures for intracellular molecules analysis.

Cell–cell communication plays a vital role in biological activities; in particular, membrane–protein interactions are profoundly significant. In order to explore the underlying mechanism of intercellular signaling pathways, a full range of artificial systems have been explored. However, many of them are complicated and uncontrollable. Herein we designed an artificial signal transduction system able to control the influx of environmental ions by triggering the activation of synthetic transmembrane channels immobilized on giant membrane vesicles (GMVs). A membrane protein-like stimulator from one GMV community (GMVB) stimulates a receptor on another GMV community (GMVA) to release ssDNA messengers, resulting in the activation of synthetic transmembrane channels to enable the influx of ions. This event, in turn, triggers signal responses encapsulated in the GMVAprotocell model. By mimicking natural signal transduction pathways, this novel prototype provides a workable tool for investigating cell–cell communication and expands biological signaling systems in general as well as explores useful platforms for addressing scientific problems which involve materials science, chemistry, and medicine.

ABSTRACTImmune checkpoint inhibitors have demonstrated promising efficacy and tolerable safety for advanced malignancies. However, a proportion of patients who had received immunotherapy may experience hyperprogressive disease and a resultant poor prognosis. Here, we report a patient with advanced esophageal squamous carcinoma who developed hyperprogressive disease shortly after immunotherapy. This patient received nivolumab after multiple lines of treatment, including chemotherapy, radiotherapy, and antiangiogenic therapy. Through the comprehensive analysis of NGS results, we concluded that the PI3K/AKT signaling pathway might be associated with hyperprogressive disease after immunotherapy. Additionally, potential mechanisms underlying hyperprogressive disease after immunotherapy reported in other malignant tumors were also summarized.

Metabolism is often regulated by the transcription and translation of RNA. In turn, it is likely that some metabolites regulate enzymes controlling reversible RNA modification, such as N6-methyladenosine (m6A), to modulate RNA. This hypothesis is at least partially supported by the findings that multiple metabolic diseases are highly associated with fat mass and obesity-associated protein (FTO), an m6A demethylase. However, knowledge about whether and which metabolites directly regulate m6A remains elusive. Here, we show that NADP directly binds FTO, independently increases FTO activity, and promotes RNA m6A demethylation and adipogenesis. We screened a set of metabolites using a fluorescence quenching assay and NADP was identified to remarkably bind FTO. In vitro demethylation assays indicated that NADP enhances FTO activity. Furthermore, NADP regulated mRNA m6A via FTO in vivo, and deletion of FTO blocked NADP-enhanced adipogenesis in 3T3-L1 preadipocytes. These results build a direct link between metabolism and RNA m6A demethylation.

Lignin dissolution is of great importance for the valorization of lignin, and development of novel efficient solvent systems is a crucial topic. Herein, 17 kinds of 1,5-diazabicyclo[5.4.0]-5-undecene-based eutectic molecular liquids (EMLs) were designed for dissolution of industrial lignins. The solubility of lignin in the EMLs significantly depended on the types of the EMLs and lignin. Also the relationship between lignin solubility and the solvatochromic parameters (α, β, and π*) varied significantly based on the EMLs and the types of lignin.

Membrane vesicles derived from live cells show great potential in biological applications due to their preserved cell membrane properties. Here, we demonstrate that cell-derived giant membrane vesicles can be used as vectors to deliver multiple therapeutic drugs and carry out combinational phototherapy for targeted cancer treatment. We show that therapeutic drugs can be efficiently encapsulated into giant membrane vesicles and delivered to target cells by membrane fusion, resulting in synergistic photodynamic/photothermal therapy under light irradiation. This study highlights biomimetic giant membrane vesicles for drug delivery with potential biomedical application in cancer therapeutics.

Inspired by this elegant system of cellular adaptivity, we herein report the rational design of a dynamic artificial adaptive system able to sense and respond to environmental stresses in a unique sense-and-respond mode. Utilizing DNA nanotechnology, we constructed an artificial signal feedback network and anchored it to the surface membrane of a model giant membrane vesicle (GMV) protocell. Such a system would need to both senses incoming stimuli and emit a feedback response to eliminate the stimuli. To accomplish this mechanistically, our DNA-based artificial signal system, hereinafter termed DASsys, was equipped with a DNA trigger-induced DNA polymer formation and dissociation machinery. Thus, through a sequential cascade of stimulus-induced DNA strand displacement, DASsys could effectively sense and respond to incoming stimuli. Then, by eliminating the stimulus, the membrane surface would return to its initial state, realizing the formation of a cyclical feedback mechanism. Overall, our strategy opens up a route to the construction of artificial signaling system capable of maintaining homeostasis in the cellular micromilieu, and addresses important emerging challenges in bioinspired engineering.

In the recruitment domain, knowing the employer industry of jobs is important to get an insight about the demand in each industry. The existing system at CareerBuilder uses an employer name normalization system and an employer knowledge base (KB) to infer the employer industry of a job. However, errors may occur during the computation of the job employer and in the construction of the employer KB with the industry attributes. Since the KB is huge, it is not possible to manually detect the errors. Therefore, in this paper we use machine learning techniques to automatically detect the errors. With the observation that the main jobs posted by an employer often relate to the employer industry, e.g., truck driver jobs often correspond to employers in the transportation industry, we develop a system that classifies the industry of an employer using job posting data. We aggregate job postings from an employer and derive features from employer names, employer descriptions, job titles, and job descriptions to predict the industry of the employer. Two models are used for classification: (1) support vector machine and (2) random forest. Our experiments show that random forest is more effective than SVM in identifying the errors in the existing industry classification system, which achieves precision 0.69, recall 0.78, and f-score 0.73. It especially better handles mixed feature vectors when normalization errors occur. We also observe that generally our models perform better in detecting errors for industries that have higher error rates.

Among the vast number of recognition molecules, DNA aptamers generated from cell-SELEX exhibit unique properties for identifying cell membrane biomarkers, in particular protein receptors on cancer cells. To integrate all recognition and computing modules within a single structure, a three-dimensional (3D) DNA-based logic gate nanomachine was constructed to target overexpressed cancer cell biomarkers with bispecific recognition. Thus, when the Boolean operator “AND” returns a true value, it is followed by an “ON” signal when the specific cell type is presented. Compared with freely dispersed double-stranded DNA (dsDNA)-based molecular circuits, this 3D DNA nanostructure, termed DNA-logic gate triangular prism (TP), showed better identification performance, enabling, in turn, better molecular targeting and fabrication of recognition nanorobotics.

A series of N-(1,3-diphenylallyl)benzenamine derivatives (M) were investigated by electrospray ionization mass spectrometry in the positive-ion mode. Both the anomalous [M−H]+and the regular [M+H]+were observed in the ESI mass spectra. The occurrence of [M−H]+has been supported by accurate mass spectrometry, liquid chromatography mass spectrometry, and tandem mass spectrometry analysis. Calculation results indicated that formation of [M−H]+is attributed to the ion-molecule reaction of M with the protonated ESI solvent molecule (e.g. CH3OH2+) via hydride abstraction from a tertiary Csp3−H. The competing ionization processes leading to [M−H]+or [M+H]+were significantly affected by the concentration of formic acid in the electrospray ionization solvent and the proton affinity of the Natom.

Cells interact with the extracellular environment through molecules expressed on the membrane. Disruption of these membrane-bound interactions (or encounters) can result in disease progression. Advances in super-resolution microscopy have allowed membrane encounters to be examined, however, these methods cannot image entire membranes and cannot provide information on the dynamic interactions between membrane-bound molecules. Here, we show a novel DNA probe that can transduce transient membrane encounter events into readable cumulative fluorescence signals. The probe, which translocates from one anchor site to another, mimicking motor proteins, is realized through a toehold-mediated DNA strand displacement reaction. Using this probe, we successfully monitored rapid encounter events of membrane lipid domains using flow cytometry and fluorescence microscopy. Our results show a preference for encounters within the same lipid domains.

Multifunctional synergistic therapy holds promise in biomedical studies and clinical practice. However, strategies aimed at easily integrating the components of such multimodal therapies are needed. Therefore, we herein report a smart drug release nanosystem able to perform photodynamic therapy, photothermal therapy and chemotherapy in a photocontrollable manner. Doxorubicin (DOX), a chemotherapy drug, and 5, 10, 15, 20-tetrakis (1-methylpyridinium-4-yl) porphyrin (TMPyP4), a photosensitizer, were physically intercalated into a DNA assembly immobilized on gold nanorods. The drugs were efficiently delivered to target cells and released under light irradiation, resulting in a synergism that combined phototherapy and chemotherapy for cancer treatment. This smart, photocontrollable drug release nanosystem promises precisely controlled drug release for multifunctional synergistic cancer therapy.

Coordinating multiple artificial cellular compartments into a well-organized artificial multicellular system (AMS) is of great interest in bottom-up synthetic biology. However, developing a facile strategy for fabricating an AMS with a controlled arrangement remains a challenge. Herein, utilizing in situDNA hybridization chain reaction on the membrane surface, we developed a DNA patch-based strategy to direct the interconnection of vesicles. By tuning the DNA patch that generates heterotrophic adhesion for the attachment of vesicles, we could produce an AMS with higher-order structures straightforwardly and effectively. Furthermore, a hybrid AMS comprising live cells and vesicles was fabricated, and we found the hybrid AMS with higher-order structures arouses efficient molecular transportation from vesicles to living cells. In brief, our work provides a versatile strategy for modulating the self-assembly of AMSs, which could expand our capability to engineer synthetic biological systems and benefit synthetic cell research in programmable manipulation of intercellular communications.

We describe a comprehensive protocol for the preparation of multifunctional DNA nanostructures termed nanoflowers (NFs), which are self-assembled from long DNA building blocks generated via rolling-circle replication (RCR) of a designed template. NF assembly is driven by liquid crystallization and dense packaging of building blocks, which eliminates the need for conventional Watson-Crick base pairing. As a result of dense DNA packaging, NFs are resistant to nuclease degradation, denaturation or dissociation at extremely low concentrations. By manually changing the template sequence, many different functional moieties including aptamers, bioimaging agents and drug-loading sites could be easily integrated into NF particles, making NFs ideal candidates for a variety of applications in biomedicine. In this protocol, the preparation of multifunctional DNA NFs with highly tunable sizes is described for applications in cell targeting, intracellular imaging and drug delivery. Preparation and characterization of functional DNA NFs takes ∼5 d; the following biomedical applications take ∼10 d.

Currently, relatively large errors are found in numerical results in some low-specific-speed centrifugal pumps with unshrouded impeller because the effect of clearances and holes are not accurately modeled. Establishing an accurate analytical model to improve performance prediction accuracy is therefore necessary. In this paper, a three-dimensional numerical simulation is conducted to predict the performance of a low-specific-speed centrifugal pump, and the modeling, numerical scheme, and turbulent selection methods are discussed. The pump performance is tested in a model pump test bench, and flow rate, head, power and efficiency of the pump are obtained. The effect of taking into consideration the back-out vane passage, clearance, and balance holes is analyzed by comparing it with experimental results, and the performance prediction methods are validated by experiments. The analysis results show that the pump performance can be accurately predicted by the improved method. Ignoring the back-out vane passage in the calculation model of unshrouded impeller is found to generate better numerical results. Further, the calculation model with the clearances and balance holes can obviously enhance the numerical accuracy. The application of disconnect interface can reduce meshing difficulty but increase the calculation error at the off-design operating point at the same time. Compared with the standard k-ɛ, renormalization group k-ɛ, and Spalart-Allmars models, the Realizable k-ɛmodel demonstrates the fastest convergent speed and the highest precision for the unshrouded impeller flow simulation. The proposed modeling and numerical simulation methods can improve the performance prediction accuracy of the low-specific-speed centrifugal pumps, and the modeling method is especially suitable for the centrifugal pump with unshrouded impeller.

Oxidative stress induced by excessive production of reactive oxygen species (ROS) has been implicated in the etiology of many human diseases. Acquiring a highly efficient antioxidant with good biocompatibility is of significance in eliminating the deleterious effect induced by the oxidative stress. Herein, we address our efforts on investigating the cytoprotective effect of carboxyfullerenes on H2O2-injured cells. Meanwhile, the uptake and intracellular location of carboxyfullerenes were studied. The results show that C70-carboxyfullerenes (dimalonic acid C70fullerene (DF70) and trimalonic acid C70fullerene (TF70)) exhibit an obviously protective effect against oxidative stress on C2C12 cells at concentrations as low as 2.5 μmol L–1, whereas C60-carboxyfullerenes (dimalonic acid C60fullerene (DF60) and quadri-malonic acid C60fullerene (QF60)) show a protective effect at relatively higher concentration (40 μmol L–1). The molecular structure of carboxyfullerenes and the physiological state of cells play an important role in the different cytoprotective capability. Further study reveals that DF70and TF70could enter into cells and mainly localize into the lysosome, which possibly involves the protective mechanism by stabilizing lysosome. The use of a significantly low concentration of C70-carboxyfullerene as the antioxidative agent will benefit the therapeutic approaches aiming at alleviating ROS-induced injuries such as muscle disorder and arthritis.

Carbon dots (CDs) with intriguing fluorescent property, good biocompatibility, high stability, easy interaction with substrates, are burgeoning carbon nanoparticles with large potential in various applications. Incorporating CDs into the polymer matrix is becoming a popular strategy to endow the complex with new functions. Herein, the green-synthesized CDs was integrated into the mixture of gelatin (derived from waste fish scale) and chitosan, and a multifunctional bio-nanocomposite (defined as Gelatin/Chitosan/CDs) film was developed, which showed the excellent antibacterial, antioxidant, pH-sensitivity, UV shielding, and blue-emission properties. The effects of different concentrations of CDs on the physical, mechanical, structural, and functional activity of bio-nanocomposite film were tested. Compared with the Gelatin/Chitosan film, the Gelatin/Chitosan/CDs film with an optimum addition of 20% CDs showed the enhanced antibacterial, antioxidant as well as UV shielding activities. More importantly, it was used as an effective packaging material for fish meat preservation, reducing the loss of nutritional quality consumption, extending the shelf life of food. Besides, the bio-nanocomposite films also possessed the anti-counterfeiting and pH-responsive properties due to the strong fluorescent emission of CDs, and had the great potential in developing the intelligent packaging materials. Our work shed new light on the new application of CDs and the synthesis of bio-nanocomposite film in food industry.

Satellite muscle cells play an important role in regeneration of skeletal muscle. However, they are particularly vulnerable to oxidative stress. Herein, we address our efforts on the cytoprotective activities of carboxyfullerenes with different cage size (C60vs C70) and adduct number on postmitotic muscle cell (C2C12 cell). The correlation of the structural effect on the cytoprotective capability of carboxyfullerenes was evaluated. We find that quadri-malonic acid C70fullerene (QF70) exhibits higher capability on protecting cells from oxidative-induced stress among these tested carboxyfullerenes. The accumulation of intracellular superoxide dismutase (SOD) is proposed to play an important role in their diverse antioxidative ability. Moreover, the pretreatment of QF70could also obviously enhance the viability of myotubes originated from oxidative-stressed C2C12 cells, which facilitates the future application of carboxyfullerenes in tissue engineering and nanomedicine.

According to the characteristic of low visibility of night image, we proposed an improved image enhancement method based on multi-scale Retinex. In our method, we replace the Gaussian filter as Blur filter, and replace logarithm gain as linear gain to reduce the computational complexity. The proposed algorithm first segment color image into three independent R, G, B channels, and then employ our improved multi-scale Retinex algorithm to enhance contrast of each channel. Finally, the enhanced image is given by recombining the three R, G, B enhanced results. A large number of experiments show that the proposed algorithm can quickly and efficiently improve the contrast of the night image and has a better visual effect than that of the similar method.

The effect of water-soluble fullerene C70(C(COOH)2)4−8on plant growth was investigated, using the transgenic seedling lines expressing fluorescent makers. The retarded roots with shortened length and loss of root gravitropism were observed for seedlings grown in the fullerene-containing medium. Fluorescence imaging revealed the abnormalities of root tips in hormone distribution, cell division, microtubule organization, and mitochondrial activity. The study of the inhibitory effects at the cellular level provides new information on the phytotoxicity mechanism of fullerene.

Abstract: This paper presents a reliability-based sensitivity method for mechanical components with arbitrary distribution parameters. Techniques from the perturbation method, the Edgeworth series, the reliability-based design theory, and the sensitivity analysis approach were employed directly to calculate the reliability-based sensitivity of mechanical components on the condition that the first four moments of the original random variables are known. The reliability-based sensitivity information of the mechanical components can be accurately and quickly obtained using a practical computer program. The effects of the design parameters on the reliability of mechanical components were studied. The method presented in this paper provides the theoretic basis for the reliability-based design of mechanical components.

The reliability sensitivity analysis of uncertain multi-degree-of-freedom nonlinear vibration systems with independent failure modes subjected to random excitation is examined. An earlier version of the statistical fourthmoment method is extended to deal with vector-valued and matrix-valued functions and is developed to determine the first four moments of the system response and state function. Random variables and system derivatives are conveniently arranged into 2D matrices by means of Kronecker algebra. The distribution function of the system state function is approximately determined by the standard normal distribution functions using Edgeworth series technique, and its reliability and reliability sensitivity are presented and discussed.

Techniques from the perturbation method, the reliability-based design theory and optimisation techniques are employed to present a practical and effective method for the reliability-based optimisation of automobile components. The theoretical formulae for reliability-based optimisation are obtained. The most probable point perturbation method (MPPPM) and reliability-based optimisation theory are employed to develop the reliability-based optimal design of automobile components. Two examples are used to illustrate the reliability-based optimisation formulation and demonstrate the method. The accuracy of the approach proposed is evaluated by comparing their results to that obtained from Monte Carlo simulations (MCS).

The results derived are easily amenable to computational procedures.

Light-absorbing hydrogels provide a means for rapidly evaporating water by using solar energy. However, to achieve light-absorbing hydrogels with both durable mechanical properties and efficient energy utilization remains challenging due to the weak interface interactions between solar absorbers and a hydrogel matrix and difficultly controlled surface topography of swollen hydrogel-based evaporators. Herein, we demonstrate an effective nanoconfinement strategy to assemble a spongy poly(vinyl alcohol)/Janus-like graphene oxide hybrid hydrogel (SPJH) viastrong interfacial interactions of hydrogen bonding and hydrophobic interaction. The resultant SPJHs with an intriguing hierarchical microstructure templated by air bubbles and ice crystals showed a high toughness (∼231 kJ m–2) and ultimate strain (∼310%) that were more than three times as high as those of light-absorbing hydrogels and a high evaporation rate of 4.18 kg m–2h–1with an efficiency up to 95% under 1 sun irradiation (relative humidity = 20%; temperature = 25 °C), achieved by synergistic mechanical and energy nanoconfinement and tailored surface topography within the designed hybrid hydrogels. This hybrid hydrogel-based solar evaporator with an ingenious design principle provides a pathway for scalable and processable solar water purification devices.

The aim of this study was to investigate the predictive value of peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 (Pin1) with C-reactive protein (CRP) and white blood cell (WBC) count for community-acquired pneumonia (CAP) in infants. A total of 84 hospitalized infants with CAP and 69 healthy infants were included in this study. The clinical manifestations and laboratory assay results of infants were recorded. Serum Pin1 level was estimated by enzyme-linked immunosorbent assay. The median serum Pin1 concentration in infants with CAP was significantly higher than that in controls (1.44 vs. 0.21 ng/mL, P< 0.0001). Receiver operating characteristic (ROC) curve analysis showed that the area under the ROC curve (AUC) of the combination Pin1, CRP and WBC (Pin1 + CRP + WBC, 0.943) was higher than Pin1, CRP, WBC alone or the combination of Pin1 and CRP (P< 0.05). The sensitivity of Pin1 + CRP + WBC (94.0%) was higher than that of Pin1, CRP, WBC alone, or any two combined (P< 0.05). Pin1 + CRP + WBC also had a high negative predictive value (91.4%). Moreover, serum Pin1 alone had a high specificity (97.0%) and excellent positive predictive value (96.6%) for infants with CAP, which were higher than WBC, Pin1 and WBC in combination, CRP and WBC in combination, and Pin1 + CRP + WBC (P< 0.05). Therefore, serum Pin 1 was highly expressed in infants with CAP and can singly or in combination with CRP and WBC represent promising novel predictors for infants with CAP.

Based on their unique advantages, increasing interest has been shown in the use of aptamers as target ligands for specific cancer cell recognition and targeted cancer therapy. Recently, the development of aptamer-conjugated nanomaterials has offered new therapeutic opportunities for cancer treatment with better efficacy and lower toxicity. We highlight some of the promising classes of aptamer-conjugated nanomaterials for the specific recognition of cancer cells and targeted cancer therapy. Recent developments in the use of novel strategies that enable sensitive and selective cancer cell recognition are introduced. In addition to targeted drug delivery for chemotherapy, we also review how aptamer-conjugated nanomaterials are being incorporated into emerging technologies with significant improvement in efficiency and selectivity in cancer treatment.
