Objective: Computed tomographic examination of the skull of cats with craniofacial trauma. Analysis of diagnostic findings with regard to the occurrence of isolated and combined maxillary and orbital fractures. Material and methods: Prospective study (August 2006 – June 2010): Computed tomography (CT) of the skull of cats with craniofacial trauma. Results: Thirty-eight cats met the inclusion criteria. Breeds were 36 Domestic Shorthair cats, one Maine Coon and one Somali cat. Age at admission ranged from 11 to 187months. The ratio of the numbers of males to females was 22:16 (1.4). Computed tomographic examination revealed a maxillary fracture in 27 (71%) animals. Sixteen (42%) cats had multiple maxillary fractures (≥2). Twenty-eight animals (74%) displayed orbital fractures. Combined maxillary and orbital fractures occurred in 26 (68%) patients. The odds ratio of this combined occurrence was 87 (p<0.001). Sixteen (57%) of 28 cats with orbital fractures showed multiple orbital fractures (≥2). The incidence of bilateral orbital fractures was 67% (25 patients). The medial orbital wall was the most commonly fractured orbital wall (66%), and the orbital floor the second most common (61%). Conclusion: Computed tomographic examination of the skull of cats with craniofacial trauma showed that maxillary and orbital fractures are more common than previously described. Combined maxillary and orbital fractures occurred in more than half of the patients. In cats, orbital fractures mainly affect the medial orbital wall and the orbital floor. Clinical relevance: Cats with craniofacial trauma often have maxillary and orbital fractures. The additional information taken from the computed tomographic examination could lead to an optimised therapeutical concept.

Objective: Medial coronoid disease (MCD) is a very common form of elbow joint disease and it’s radiographic diagnosis can be challenging since it is frequently based on the detection of rather subtle primary or secondary changes than on a large primary lesion. We hypothesized that accuracy of radiographic diagnosis of MCD is highly dependent on training and experience level. Methods: Radiographs of 102 canine elbows were evaluated for MCD by four observers with different levels of training and experience. All elbows underwent CT scans and arthroscopy. Sensitivity and specificity of radiographic and CT interpretation was determined using arthroscopy as a gold standard. Interobserver and intraobserver agreement (reliability and repeatability) were assessed by using Cohen’s Kappa (κ) statistic. Results: The sensitivity (92.4–96.7%) of the two experienced observers was almost comparable to that of CT (100%) and significantly higher than that of the two less experienced observers (77.2–80.4%). Reliability of the radiographic diagnosis of MCD was better between observers with higher experience level (κ= 0.74) than between observers of lower or different experience levels (κ=0.07–0.42). Repeatability was better in experienced (κ= 0.73–0.88) than in less experienced observers (κ= 0.31–0.42). Conclusion: Our results confirm that training and experience play important roles in reaching high sensitivity, reliability and repeatability for the radiographic diagnosis of MCD. Clinical relevance: Although radiography is inferior to CT in imaging of the medial coronoid process itself, sensitivity of radiographic diagnosis MCD can be significantly improved with observer experience almost reaching that of CT. Therefore, it is advised that radiographic screening for MCD should be performed by specialists experienced in the radiographic evaluation of elbow joint disease.

Objective: The diagnostic value of CT and MRI regarding the diagnosis of coronoid pathology in the dog. Material and methods: Computed tomography (CT) and magnetic resonance imaging (MRI) of the elbow joint were performed in dogs with clinical and radiological signs of coronoid pathology. Afterwards, all dogs underwent arthroscopic surgery. For the computed tomographic examination, a 16-slice-CT-scanner spiral-CT (Philips Brilliance 16) was used. The MRI-examination was performed with a 1-Tesla superconducting magnet (Phillips Intera 1.0). T1 and T2 weighted images with different sequences were acquired. Results: In total, 44 elbow joints from 44 patients (total of 12 breeds, including mixed breeds) were examined. The most represented breeds were Labrador Retrievers (38.6%, n=17), mixed breed dogs (22.7%, n=10) and Golden Retrievers (11.4%, n=5) were represented most. The age of the 30 male dogs (68%) and 14 female dogs (32%) ranged from 6 to 117 months (mean 2.25 years). Using CT, the following results could be evaluated: a) fissure at the level of the Processus coronoideus medialis ulnae (PCM) in 66% (n=29); b) fragments at the level of the PCM in 55% (n=24); c) deformation at the level of the PCM in all 44 joints; d) increased opacity at the level of the base of the PCM in all 44 joints; e) heterogenous opacity at the apex of the PCM in 91% (n=41). With MRI, the following results could be evaluated: a) fissure at the level of the PCM in 59% (n=26); b) fragments at the level of the PCM in 57% (n=25); c) deformation at the level of the PCM in 86% (n=38); d) increased opacity at the level of the base of the PCM, thus making assessment impossible; e) heterogenous opacity at the apex of the PCM, thus making assessment impossible. Conclusion and clinical relevance: Both diganostic imaging modalities are appropriate for evaluating coronoid pathology in the dog.

Epstein-Barr virus (EBV) is a worldwide endemic gamma herpesvirus of the genus Lymphocryptovirus(LCV) that infects more than 90% of the world's population. EBV has been associated with a variety of malignancies, but it has a demonstrated role in lymphomas, especially in immunosuppressed individuals. Lymphomas of the nasal cavity, paranasal sinuses, and nasopharynx are uncommon and constitute less than 5% of all extranodal lymphomas. Sinonasal non-Hodgkin's lymphomas have been reported in patients infected with human immunodeficiency virus (HIV) at an increased frequency. Rhesus LCV (rhLCV), the rhesus viral homolog of EBV, has been cloned and is associated with B-cell lymphomas in immunosuppressed rhesus macaques. We report two cases of B-cell lymphoma within the nasal cavity from 2 simian immunodeficiency virus-infected rhesus macaques with acquired immunodeficiency syndrome. The B-cell phenotype and rhLCV association were demonstrated by immunohistochemistry and confocal microscopy. The majority of the nuclei of the neoplastic B lymphocytes were EBNA-2 positive. RhLCV type 1 sequences were verified from the neoplasms by polymerase chain reaction. Nasal lymphoma is an unusual presentation of rhLCV-associated B-cell lymphoma in immunosuppressed rhesus macaques. These tumors demonstrate comparable viral pathogenesis with EBV-induced nasal lymphomas in HIV-positive people.

This mini-review highlights the involvement of PP2C (protein phosphatase type 2C) family members α and β in apoptosis. The activity of these isoenzymes can be stimulated by unsaturated fatty acids with special structural features, e.g. oleic acid. Those fatty acids capable of activating PP2Cα and PP2Cβ in vitro induce apoptosis in various cell types as shown here for neurons and endothelial cells. Using RNA interference to reduce the amount of PP2Cα and PP2Cβ results in cells significantly less susceptible to the apoptotic effect of oleic acid. Increased endothelial cell death is considered to be an initial step of atherogenesis. Thus activation of PP2C by physiological unbound (‘free’) unsaturated fatty acids (liberated from lipoproteins) could represent a crucial mechanism in the development of atherosclerosis.

The photoabsorption of Ar is studied experimentally and theoretically for energies between 32.5 eV and 33.0 eV. Resonant excitation and autoionization of doubly excited 3p4(3P)4s2P3/2,1/2np Rydberg states are observed as dominant mechanism to populate doublet and quartet satellites of the 3p44s, 3d configurations. The perturbation of the Rydberg series by promoter states is reproduced by the calculations. Measured and calculated cross sections for the strong decay channels are in good agreement. A mirror behaviour of the 3s vacancy creation and the satellite production and the influence of the radiative decay of high-lying Rydberg states on the shape of a satellite threshold are manifested.

Predissociation of the N+2C 2?+u(v') vibrational levels with v' ? 3 was observed via dispersed C 2?+u? X 2?+gfluorescence in the spectral range of 165-208 nm after resonant 1s?1?*(vr) excitation of N2and its subsequent autoionization into the N+2C state. This range is dominated by lines in atomic nitrogen, by overlapped C 2?+u(v') ? X 2?+g(v'') vibrational band sequences with ?v= const and broad unresolved band systems (D, (2))2?g(v') ? A2?u(v'') in the N+2molecular ion. With very high fluorescence resolution of about 0.1 nm FWHM individual C 2?+u(v') ? X 2?+g(v'') vibrational bands have been resolved. Calculation of the observed fluorescence spectra by taking into account predissociation and molecular rotation describes well the shape of both individual vibrational bands C 2?+u(v') ? X 2?+g(v'') and the whole band system.

Ab initiocalculations of the corresponding potential curves and transition probabilities showed that the observed irregular intensity dependence of the C2?+u(v') ? X2?+g(v'')(?v= const) fluorescence lines on the vibrational quantum number vris due to transitions between vibrational levels during the reaction N2(v0= 0)? N*2(1s?1?*(vr)) C2?+u(v') ? X2?+g(v'').

The alignment parameter A20and the orientation parameter O10for the 4p4(1D)5p2F7/2and the 4p4(1D)5p2D5/2states of Kr+populated in resonant Auger transitions from 3d95p/6p photo-excited states of Kr were measured at BESSY II for photon energies between 91 eV and 92.8 eV. The method of fluorescence polarimetry was applied. A bandwidth of the exciting photons of 10 meV allowed to study A20and O10within the natural line width. Interference effects for resonances overlapping due to lifetime broadening were predicted and observed even when separated by 15 times the natural line width. The relative contribution of the three allowed electron partial waves was derived from A20and O10as a function of the exciting-photon energy and compared with the calculated contributions. The resulting good overall agreement between experiment and prediction is considered as a noticeable improvement of the understanding of excitation and decay of the prominent Kr 3d95p resonances.

The interaction between resonances through autoionization continua and the interaction between autoionization continua were investigated theoretically and experimentally for the photoionization process of the 4p- and 4s-shells and for the population of 4p4(3P)5s 4PJ, 4p4(3P)5s 2PJand 4p4(3P)4d 4DJsatellites of KrII. Cross sections for the satellite production and the angular distribution parameter of the fluorescence radiation were measured by photon-induced fluorescence spectroscopy after excitationx with linearly polarized monochromatized synchrotron radiation at exciting-photon energies between 28.45 eV and 29.95 eV with an exciting-photon energy resolution of 10 meV (FWHM). Measured cross sections are in good agreement with the computed ones. A refined assignment of resonances in the proximity of the 4s14p65p resonance was performed. It was concluded that there is a strong influence of the core rearrangement, of the interaction between resonances through the autoionization continua and of the interaction between autoionization continua, on the investigated processes on the basis of the observed good overall agreement between the computed and measured quantities.

The decay of doubly excited helium states below the N= 2 threshold, cascading radiatively over three steps, were investigated using the photon-induced fluorescence spectroscopy (PIFS) at BESSY II. Absolute cross sections as the product of the resonance excitation cross section of the doubly excited states and their fluorescence rate to decay into the singly excited 1sms(1S) and 1smd(1D) states were measured. The experiments showed that the (sp,2n+)(1P) states decay predominantly into the 1sns(1S) states, whereas the (pd,2n)(1P) states prefer to decay into the 1snd(1D) states. For the (sp,2n?)(1P) states with n= 4, 5 and 6 we observed a broad and complex decay pattern. In addition the angular distribution of the fluorescence radiation was measured. The results are in good agreement with theoretical predictions. Furthermore, the weakening of the LS coupling scheme and the mixing between singlet and triplet states in helium was confirmed by observation of the 1s6d(3D0,1,2) ? 1s2p(3P0,1,2) transition on a doubly excited singlet state.

In a retrospective study, 51 cases of gastritis (14%) were identified from among 341 necropsies performed on simian immunodeficiency virus (SlV)-infected rhesus macaques (Macaca mulatta) at the New England Primate Research Center from 1993 to 2001. Protozoa were seen in the stomach of 13 monkeys (25%) with gastritis. Two histopathologic manifestations of gastritis were observed: seven cases of lymphoplasmacytic gastritis with trichomonad trophozoites within lumens of gastric glands and four cases of necrosuppurative gastritis containing intralesional periodic acid-Schiff-positive protozoa; two cases of gastritis had morphologic features of both types of gastritis. In instances of necrosuppurative and combined lymphoplasmacytic and necrosuppurative gastritis, protozoa were 4-35 μm in diameter and round to tear-shaped. Because of the unusual morphology of the protozoa in these latter cases, transmission electron microscopy and polymerase chain reaction (PCR) were used to further identify these organisms. The protozoa were definitively identified as Tritrichomonas in all cases on the basis of ultrastructural characteristics (flagella and undulating membranes) and amplification of a 347-bp product of the 5.8S ribosomal RNA gene of Tritrichomonas foetus, Tritrichomonas suis and Tritrichomonas mobilensis by PCR using DNA extracted from stomach tissue. On the basis of these observations, we conclude that Tritrichomonas can be a significant cofactor in the development of necrosup-purative gastritis in SIV-infected rhesus macaques.

Dispersed Kr VUV-fluorescence excitation spectra and photoion yields were recorded in the exciting photon energy range between 28.4 eV and 28.7 eV with extremely narrow bandwidth (1.7 meV at 28.55 eV) of the monochromatized synchrotron radiation used as the exciting photon source. Two series of doubly excited KrI states were observed and identified as the 4s²4p⁴ (³P) 5s ⁴P_1/2 np and the 4s²4p⁴(³P)5s ²P_3/2 np series.

A 4-year-old female pigtailed macaque (Macaca nemestrina), experimentally coinfected with simian immunodeficiency virus (SIVmac251) and Mycobacterium bovis (bacillus Calmette-Guerin), was euthanatized 1 year after infection because of weight loss and labored breathing. On gross examination, both kidneys were found to be markedly enlarged (right: 54.7 g and left: 51.7 g; normal < 20 g). Renal lesions were evaluated by histopathologic, immunohistochemical, and ultrastructural methods. Light microscopy revealed that the glomeruli were diffusely hypercellular with expansion of the mesangial matrix, and crescent formation affected approximately 60% of the glomeruli. By immunohistochemical evaluation, it was found that the crescents were composed principally of macrophages, as seen by CD68 (KP1), MRP8, MAC387, and HAM56 expression. Electron microscopic examination of the glomeruli revealed extensive intramembranous, subendothelial, and mesangial electron-dense deposits and multifocal fusion of the visceral epithelial foot processes. Immunofluorescence, used to determine the composition of the electron-dense deposits, revealed diffuse granular mesangial and capillary staining for immunoglobulin A (IgA). The renal changes described in this case report are most consistent with the findings of crescentic gloerulonephritis with IgA immune complex deposition in the glomerular basement membrane and mesangium as described in humans with IgA nephropathy.

Backward swimming is a stereotypic behavioural response of Paramecium. It is triggered by depolarizing stimuli, which open calcium channels in the excitable ciliary membrane. The influx of Ca2+ causes the reversal of ciliary beat and initiates backward swimming. Here, we demonstrate that the protein phosphatase inhibitor okadaic acid does not affect the normal forward swimming pattern of Paramecium, but greatly extends the duration of backward swimming as initiated by depolarization caused by a rise in extracellular K+. Chelation of external Ca2+ results in an immediate resumption of forward swimming. The results suggest that the voltage‐operated calcium channel is inactivated by a dephosphorylation event, and that okadaic acid blocks this dephosphorylation without any effect on the motile apparatus of the cilia. In addition, Paramecium is unique among eukaryotic cells, in that okadaic acid inhibits just one protein phosphatase, namely a type 1 enzyme, 75% of which is tightly associated with the excitable ciliary membrane. The type 2A protein phosphatases in Paramecium are unaffected by okadaic acid. The results indicate that protein phosphatase 1 is the enzyme responsible for the dephosphorylation and closure of the calcium channel in Paramecium.

We localized protein phosphatase Type 1 (PP1) in Paramecium cells using antibodies (specified on Western blots) against recombinant protein, amino- or carboxy-terminal peptides, or peptide segments containing both terminals and an intermediate segment. Cell fractionation and ELISA revealed high PP1 concentrations in cilia, corresponding to observations by immunofluorescence and immunogold labeling analyses. We compared ELISA results obtained with MnCl2- or detergent-mediated deciliation and immunolocalizations obtained with digitonin and saponin- or detergent-mediated permeabilization. We observed that detergents at too high concentrations can displace the antigen from its original position. Quantitative evaluation of immunogold labeling revealed a predominant localization of PP1 in cilia, notably in the narrow space between the membrane and the outer microtubule doublets, as ascertained by immunogold labeling of Lowicryl sections obtained after rapid freezing and freeze-substitution. This localization to the periphery of cilia is compatible with previous suggestions of PP1 involvement in ciliary beat regulation, notably of cilia on the free cell surface. Immunolabeling occurs along the entire length of surface cilia. Despite much higher PP1 concentrations in cilia, ELISA values for absolute PP1 content were considerably higher in deciliated cells. This may indicate still other functional aspects of PP1. Along these lines, we also discuss the differences observed when immunochemical and enzymatic data are compared.

An adult male rhesus monkey (Macaca mulatto) developed clinical signs of severe osteomyelitis of the left femur 42 days after onset of enteritis. Salmonella sp. were cultured from feces, blood, and femoral lesions. Response to antibiotic therapy was poor, and the animal was euthanized. The left femur, with pathologic fracture and involucrum, and the right femur, tibia and fibula were most severely affected. Additionally hepatic microgranulomas, mild tubulointcrstitial nephritis, medullary histiocytosis and erythrophagocytosisin the lymph nodes, and a mild colitis were diagnosed microscopically. The severity of the disease and serum electrophoresis findings were suggestive of lowered resistance to the organism, possibly due to anemia or polychlorinated biphenyl toxicosis.

A cDNA of a type 7 adenylyl cyclase isoform was cloned from a bovine retinal pigment epithelium cDNA library using oligonucleotides developed to conserved regions common to mammalian adenylyl cyclases. A 6.7 kb mRNA of very high abundance was uniquely present on Northern blots containing mRNA or total RNA from the pigment epithelium. This transcript was undetectable in all other tissues examined. The cDNA encoded a protein of 1,097 amino acids and exhibited the known doublet of 6 transmembrane-spanning regions in a hydrophobicity plot. The novel member of the type 7 adenylyl cyclase isoform was expressed in COS-1 cells. It was stimulated 10- and 20-fold by 10 μM GTPγS and 100 μM forskolin, respectively. The high expression rate exclusively in the retinal pigment epithelium suggests that this adenylyl cyclase isoform is involved in processes specific to this functionally exceedingly important subretinal cell layer.

Simian immunodeficiency virus (SIV) induces an immunodeficiency syndrome similar to human AIDS. Although the disease course of SIV-induced immunodeficiency is generally measured in months to years, a disease syndrome that results in death in 5 to 14 days has been described in pig-tailed macaques infected with the SIVsmmPBj (PBj) strain. The purpose of this study was to derive an acutely lethal PBj molecular clone in order to study viral genes involved in pathogenesis. Six infectious molecular clones were generated; acutely fatal disease was induced by experimental inoculation of pig-tailed macaques with virus stocks derived from either of two clones, PBj6.6 or PBj14.6. Molecular chimeras were constructed by exchange of regions of the genome of PBj6.6 and a nonlethal, related clone, SIVsmH4. Only a chimera expressing the PBj genome under the control of a SIVsmH4 long terminal repeat induced death soon after inoculation. These studies suggest that multiple viral genes of PBj are critical for development of acute disease. More specifically, the env gene but not the long terminal repeat PBj was required for acute disease induction; however env must act in concert with another gene(s) of the PBj genome.

The kinetics of the ionic regulation of an adenylate cyclase associated with the excitable ciliary membrane from Paramecium tetraurelia was examined. Glycerol (30%, v/v) stabilized the enzyme, and activated by an increase in Vmax. (3-fold) and a decrease in the apparent Km for MgATP (6-fold). Kinetic analysis of Mg2+ effects showed a stimulation via a single metal-binding site separate from the substrate site, with a dissociation constant, Ks, of 0.27 mM. Analysis of Ca2+ effects showed (i) an uncompetitive inhibition with respect to substrate MgATP, and (ii) dependence of the extent of inhibition on the free Mg2+ concentration. Ki values ranged from 4 to 130 microM-Ca2+ in the presence of 0.55-2 mM-Mg2+ respectively. This indicates competition between Mg2+ and Ca2+ at the metal-binding site. The Ca2+ effect was specific; Sr2+ and Ba2+ were almost without effect, and 100 microM-Ba2+ did not interfere with the Ca2+ inhibition. The actions of Ca2+ were readily reversible after addition of EGTA. K+ activated the adenylate cyclase at concentrations around 20 mM. The stimulatory potency of K+ was dependent on the free Mg2+ concentration. At 1 mM free Mg2+, 20 mM-K+ doubled the adenylate cyclase activity. The inhibitory Ca2+ and stimulatory K+ inputs were independent of each other.

The condition of a chimpanzee (C499) infected with three different isolates of human immunodeficiency virus type 1 (HIV-1) for over 10 years progressed to AIDS. Disease development in this animal was characterized by (i) a decline in CD4+ cells over the last 3 years; (ii) an increase in viral loads in plasma; (iii) the presence of a virus, termed HIV-1JC, which is cytopathic for chimpanzee peripheral blood mononuclear cells; and (iv) the presence of an opportunistic infection and blood dyscrasias. Genetic analysis of the V1-V2 region of the envelope gene of HIV-1JC showed that the virus present in C499 was significantly divergent from all inoculating viruses (> or = 16% divergent at the amino acid level) and was suggestive of a large quasispecies. Blood from C499 transfused into an uninfected chimpanzee (C455) induced a rapid and sustained CD4+-cell decline in the latter animal, concomitant with high plasma viral loads. These results show that HIV-1 can induce AIDS in chimpanzees and suggest that long-term passage of HIV-1 in chimpanzees can result in the development of a more pathogenic virus.

In isolated rat pancreatic islets exogenous glutathione which is not able to penetrate into cells, augmented glucose (11.1 mmol/l)-mediated insulin release. In the presence of a non-stimulatory glucose concentration (2.8 mmol/l) glutathione had no effect. The half-maximal synergistic action of glutathione on insulin secretion was observed at approximately 8.0 μmol/l. This concentration of glutathione is similar to that found in the plasma of non-fasted anaesthetised rats (6.5 μmol/l). Oral ingestion of glucose increased the arterial plasma glutathione in rats. Our data provide for the first time indirect evidence for a modulating effect of plasma glutathione in postprandial glucose-mediated insulin secretion which appears to be located at the extracellular site of islet cells.

Ca2+-regulated guanylate cyclase in ciliary membranes from Paramecium contained tightly bound calmodulin. Antisera against calmodulin from Tetrahymena and soybean inhibited enzyme activity. EGTA did not easily release calmodulin; however, La3+ inhibited guanylate cyclase by dissociation of calmodulin. While La could not replace Ca in the activation of guanylate cyclase, it substituted for Ca2+ in the activation of calmodulin-dependent phosphodiesterase from pig brain independently of whether homologous or Paramecium calmodulin was used. After removal of endogenous calmodulin from guanylate cyclase, reconstitution was achieved with calmodulin from Paramecium, Tetrahymena, pig brain, and soybean. Ca2+-binding proteins lacking trimethyllysine like calmodulin from Dictyostelium, parvalbumin, and troponin C failed to restore enzyme activity. The properties of the native and reconstituted guanylate cyclase/calmodulin complex were compared. Reassociation of calmodulin with its target enzyme was weak since all calmodulin remained in the supernatant after a single centrifugation. While most enzyme characteristics remained unchanged in the reconstituted complex, the inhibition by Ca greater than 100 microM was of a mixed-type compared to noncompetitive inhibition in the native enzyme. The regulation of the enzyme by cations was also altered. Whereas Ca was the most potent and specific activator of the native enzyme, in the reconstituted system Sr was far more effective.

Relative cross sections for photoionization of endohedral fullerene ions Lu3N@C80q+ (q = 1,2,3) have been measured employing the photon-ion merged-beam technique. The investigations include various ionization channels Lu3N@C80+q→+p (q = 1,2,3 and p = 2,3,4,5,6), in some cases accompanied by fragmentation of the carbon cage. Prominent structures related to the carbon K-shell ionization threshold were observed in the energy range 280-330 eV. These resonance structures have been analyzed in seven product channels and are compared with previously known absorption spectra of several fullerene species. Within the statistical significance of the data, we do not observe any signatures of the N 1s and Lu 3d thresholds in the energy ranges 390-435 eV and 1500-1700 eV, respectively. By comparing the energies for double ionization of Lu3N@C80+ and Lu3N@C802+ at the carbon 1s K-shell threshold, we derive a value of 5.0±0.4Å for the radius of the carbon cage. [ABSTRACT FROM AUTHOR]
