The vitamin E component δ-tocotrienol has shown impressive activities in radioprotection, neuroprotection, and cholesterol reduction. Its production is limited by the low content in plants and difficulty in separation from other tocotrienols. Fermentative production using a microbial cell factory that exclusively produces and secretes δ-tocotrienol is a promising alternative approach. Assembly of the δ-tocotrienol synthetic pathway in Saccharomyces cerevisiaefollowed by comprehensive pathway engineering led to the production of 73.45 mg/L δ-tocotrienol. Subsequent addition of 2-hydroxypropyl-β-cyclodextrin (CD) and overexpression of the transcription factor PDR1 significantly elevated δ-tocotrienol titer to 241.7 mg/L (63.65 mg/g dry cell weight) in shake flasks, with 30.4% secreted. By properly adding CD and the in situ extractant olive oil, 181.12 mg/L of δ-tocotrienol was collected as an extracellular product, accounting for 85.6% of the total δ-tocotrienol production. This process provides not only a promising δ-tocotrienol cell factory but also insights into yeast engineering toward secretory production of other terpenoids.

Premature ovarian insufficiency (POI) is a major cause of female infertility due to early loss of ovarian function. POI is a heterogeneous condition, and its molecular etiology is unclear. To identify genetic variants associated with POI, here we performed whole-exome sequencing in a cohort of 1,030 patients with POI. We detected 195 pathogenic/likely pathogenic variants in 59 known POI-causative genes, accounting for 193 (18.7%) cases. Association analyses comparing the POI cohort with a control cohort of 5,000 individuals without POI identified 20 further POI-associated genes with a significantly higher burden of loss-of-function variants. Functional annotations of these novel 20 genes indicated their involvement in ovarian development and function, including gonadogenesis (LGR4and PRDM1), meiosis (CPEB1, KASH5, MCMDC2, MEIOSIN, NUP43, RFWD3, SHOC1, SLX4and STRA8) and folliculogenesis and ovulation (ALOX12, BMP6, H1-8, HMMR, HSD17B1, MST1R, PPM1B, ZAR1and ZP3). Cumulatively, pathogenic and likely pathogenic variants in known POI-causative and novel POI-associated genes contributed to 242 (23.5%) cases. Further genotype–phenotype correlation analyses indicated that genetic contribution was higher in cases with primary amenorrhea compared to that in cases with secondary amenorrhea. This study expands understanding of the genetic landscape underlying POI and presents insights that have the potential to improve the utility of diagnostic genetic screenings.

ABSTRACTOvarian granulosa cells (GCs) proliferate and differentiate along with follicular growth, and this is indispensable for oocyte development and female fertility. Although the role of macroautophagy/autophagy in ovarian function has been reported, its contribution to the regulation of GC characteristics remains elusive. The siRNA-mediated knockdown of two key autophagy-related genes ATG5and BECN1and the autophagy inhibitor chloroquine were used to interfere with autophagy in GCs. Inhibition of autophagy both genetically and pharmacologically resulted in decreased expression of genes associated with GC differentiation, including CYP19A1/Aromataseand FSHR, as well as in reduced estradiol synthesis. Mechanistically, when autophagy was disrupted, the transcription factor WT1 accumulated in GCs due to its insufficient degradation by the autophagic pathway, and this inhibited GC differentiation. Finally, decreased expression of several autophagy-related genes, as well as reduced LC3-II:LC3-I and elevated SQSTM1/p62 protein levels, which are indications of decreased autophagy, were detected in GCs from biochemical premature ovarian insufficiency patients. 7-Dehydrocholesterol (7-DHC), a key pharmaceutical intermediate in the production of vitamin D3, has a wide range of applications. To explore fermentative synthesis of 7-DHC, a 7-DHC-producing Saccharomyces cerevisiaestrain was constructed by blocking the competitive pathway, eliminating rate-limiting steps, altering global regulation, and pathway compartmentalization. After blocking the competitive pathway by disrupting ERG5and ERG6and introducing DHCR24from Gallus gallus, S. cerevisiaeproduced 139.72 mg/L (17.04 mg/g dry cell weight, hereafter abbreviated as DCW) 7-DHC. Subsequent alteration of global regulation by deleting ROX1and overexpressing UPC2-1increased 7-DHC production to 217.68 mg/L (37.56 mg/g DCW). To remove the accumulated squalene, the post-squalene pathway was strengthened by co-overexpression of PGAL1-driven ERG11and PGAL10-driven ERG1, which improved 7-DHC titer and yield to 281.73 mg/L and 46.78 mg/g DCW, respectively, and reduced squalene content by 90.12%. We surmised that the sterol precursors in the plasma membrane and peroxisomes may not be accessible to the pathway enzymes, thus we re-localized DHCR24p and Erg2p-GGGGS-Erg3p to the plasma membrane and peroxisomes, boosting 7-DHC production to 357.53 mg/L (63.12 mg/g DCW). Iron supplementation further increased 7-DHC production to 370.68 mg/L in shake flasks and 1.56 g/L in fed-batch fermentation. This study demonstrates the power of global regulation and subcellular relocalization of key enzymes to improve 7-DHC synthesis in yeast.

To study the lightning electromagnetic pulse (LEMP) coupling and protection problems of shielding enclosure with penetrating wire, we adopt the model with proper size which is close to the practical engineering and the two-step finite-difference time-domain (FDTD) method is used for calculation in this paper. It is shown that the coupling voltage on the circuit lead inside the enclosure increases about 34 dB, when add 1.0 m long penetrating wire at the aperture, comparing with the case without penetrating wire. Meanwhile, the waveform, has the same wave outline as the lightning current source, shows that the penetrating wire brings a large number of low frequency component into the enclosure. The coupling effect in the enclosure will reduce greatly when penetrating wire has electrical connection with the enclosure at the aperture and the coupling voltage increase only about 12 dB than the case without penetrating wire. Moreover, the results show that though the waveguide pipe can reduce the coupling effect brought by the penetrating wire, the exposing part of penetrating wire can increase the coupling when the penetrating wire outside the enclosure is longer than the waveguide pipe and the longer the exposing part is, the stronger the coupling is.

D-mannose, a C-2 epimer of glucose, exists naturally in many plants and fruits, and is found in human blood at concentrations less than one-fiftieth of that of glucose. However, although the roles of glucose in T cell metabolism, diabetes and obesity are well characterized, the function of D-mannose in T cell immune responses remains unknown. Here we show that supraphysiological levels of D-mannose safely achievable by drinking-water supplementation suppressed immunopathology in mouse models of autoimmune diabetes and airway inflammation, and increased the proportion of Foxp3+regulatory T cells (Tregcells) in mice. In vitro, D-mannose stimulated Tregcell differentiation in human and mouse cells by promoting TGF-β activation, which in turn was mediated by upregulation of integrin αvβ8and reactive oxygen species generated by increased fatty acid oxidation. This previously unrecognized immunoregulatory function of D-mannose may have clinical applications for immunopathology.

There is high heterogeneity of phenotype and etiology in women with overt POI. Phenotypic variabilities exist among patients, even when stratified by different etiological diagnosis.

The development of highly efficient nanomaterial-based mimetic enzymes is much needed to replace natural enzymes due to their low cost, high stability and efficient catalytic ability. Herein, multiple Mo-doped CeO2nanoparticles (Mo/CeO2NPs) with different molar ratios of Mo were synthesized using a facile microwave-assisted method. For characterizing the as-prepared Mo/CeO2NPs, XRD, TEM, HRTEM, BET and XPS have been performed systematically. The peroxidase-like activity of Mo/CeO2NPs was investigated, and they exhibited enhanced peroxidase-like activity in comparison to that of pure CeO2NPs, the best peroxidase-like activity was obtained for the Mo/CeO2NPs when the molar ratio of Mo was 9%. On the basis of the high activity of Mo/CeO2NPs, the reaction provides a simple, sensitive and selective method for colorimetric detection of glucose with the detection limit of 4 × 10−7mol L−1and in a dynamic range from 5 × 10−7to 5 × 10−5mol L−1. This work will pave a way for high throughput synthesis of other ultra-small doped metal oxide nanoparticles, which have bright prospects in biomedical analysis.

The aim of this study was to evaluate the growth of rape (Brassica napusL.) seedlings under different light intensities to select appropriate conditions for cultivation in an indoor system. Seedlings were grown under different light intensities of red and blue light provided by light-emitting diodes (LEDs) and their self-adjustment ability and changes in leaf microstructure were evaluated. Light was supplied by red LEDs with peak wavelengths of 630 (R1) and 660 nm (R2) and by blue LEDs (B) with a peak wavelength of 445 nm (the light intensity ratio of R1:R2:B was 3:3:2), at intensities of 400 (R1R2B400), 300 (R1R2B300), and 200 μmol m−2s−1(R1R2B200). Natural solar light served as the control (C). Plant height, stem diameter, root length, leaf area, and dry weight of rape seedlings gradually increased with increasing light intensity. The seedlings in the R1R2B400 treatment grew more vigorously, while those in the R1R2B200 treatment were weaker. The photosynthetic pigment contents did not differ significantly between the R1R2B400 treatment and C, but were significantly lower in the R1R2B300 and R1R2B200 treatments. The highest intercellular CO2concentration, stomatal conductance, and transpiration rate were in the R1R2B300 treatment. The highest photosynthetic rate was in the R1R2B400 treatment, and was related to more compact leaves, thicker and tidier palisade and spongy tissues, and well-developed chloroplasts. In contrast, the seedlings in the R1R2B200 treatment had disordered mesophyll cells, round chloroplasts, and fractured and fuzzy grana lamellae, all of which inhibited plant growth. In conclusion, the seedlings in the R1R2B400 treatment had well-developed leaves, which favored photosynthesis. Compared with the light intensities below 300 μmol m−2s−1, the light intensity of 400 μmol m−2s−1provided by a combination of red and blue LEDs was beneficial for cultivating strong and healthy rape seedlings in an artificial system.

Oocyte maturation is vital to attain full competence required for fertilization and embryogenesis. NLRP14 is preferentially expressed in mammalian oocytes and early embryos. Yet, the role and molecular mechanism of NLRP14 in oocyte maturation and early embryogenesis are poorly understood, and whether NLRP14deficiency accounts for human infertility is unknown. Here, we found that maternal loss of Nlrp14resulted in sterility with oocyte maturation defects and early embryonic arrest (EEA). Nlrp14ablation compromised oocyte competence due to impaired cytoplasmic and nuclear maturation. Importantly, we revealed that NLRP14 maintained cytoplasmic UHRF1 abundance by protecting it from proteasome-dependent degradation and anchoring it from nuclear translocation in the oocyte. Furthermore, we identified compound heterozygous NLRP14variants in women affected by infertility with EEA, which interrupted the NLRP14-UHRF1 interaction and decreased UHRF1 levels. Our data demonstrate NLRP14 as a cytoplasm-specific regulator of UHRF1 during oocyte maturation, providing insights into genetic diagnosis for female infertility.

Immune dysregulation has long been proposed to be associated with adenomyosis, but the underlying mediators and mechanisms remain largely unexplored. Here, we used flow cytometry to investigate the alterations in immune cell subsets in adenomyotic uteri and analyze the phenotype and function of abnormal immune cells. We found that an increase in CD8+T-cell number was the predominant alteration in ectopic lesions in patients with adenomyosis and was significantly associated with the severity of adenomyosis. Importantly, we identified an exhausted natural killer group protein 2A (NKG2A)+CD8+T-cell subset that was associated with the severity of adenomyosis and found that the number of these cells was significantly increased in the eutopic endometrium and ectopic lesions. In addition, increases in NKG2A ligand histocompatibility leucocyte antigen E (HLA-E) and interleukin (IL)-15 expression in glandular epithelial cells in the adenomyotic microenvironment might contribute to CD8+T-cell exhaustion by promoting NKG2A expression on CD8+T cells or inhibiting the effector function of these cells. In conclusion, our data revealed a previously unrecognized role for NKG2A+CD8+T-cell exhaustion in the pathogenesis of adenomyosis, indicating that therapeutic interventions designed to target and reinvigorate exhausted CD8+T cells may be beneficial for patients with adenomyosis.

The kinetics of the hydrogen oxidation reaction (HOR) on platinum nanoparticles (PtNPs) is explored. The steady-state current for HOR on individual PtNPs is measured via the nanoimpact approach. Adopting the Tafel-Volmer mechanism, the adsorption rate constant for HOR on PtNPs is calculated as 0.020 ± 0.008 m s–1. The HOR is also studied on electrodes where PtNPs are immobilized via drop-casting on the supporting substrate prior to experimentation and the kinetic parameters contrasted with those from the nanoimpact experiments. The distinction in the kinetic parameters between the nanoimpact and the drop-casting approaches reveals that the measurement on an individual nanoparticle is required in order to correctly understand the electrochemical reaction catalyzed by nanoparticles, because the aggregation or loss of the nanoparticles drop-casted on the electrode leads to unavoidable errors in the measurement.

A novel NADP+-dependent aldehyde dehydrogenase (LeADH) was identified from Lodderomyces elongisporusand engineered for bio-oxidation of (3R,5S)-6-chloro-2,4,6-trideoxy-erythro-hexapyranose, a key chiral intermediate, for synthesis of statin side-chains. The catalytic activity of LeADH was improved by 12-fold through the combinatorial active-site saturation test (CAST) strategy. The resultant variant, LeADHI87F/N235H/P236H, was co-expressed with an NADPH oxidase SmNOXV193R/V194Hin Escherichia coli. A green-by-design system was then constructed for efficient oxidation of the unnatural hexapyranose into the key statin side chain, (3R,5S)-6-chloro-2,4,6-trideoxy-erythro-hexonolactone, with oxygen as the co-substrate and water as the sole by-product.

The hydrogen oxidation reaction was studied at bright polycrystalline platinum microelectrodes. A smaller steady-state current was observed in experiment as compared to that anticipated for a diffusion limited process. To facilitate physical insight into this system, a simulation model based on the Tafel–Volmer mechanism for the hydrogen oxidation reaction was developed. Under conditions of reversible electron transfer, the adsorption kinetics kaand kdare found to have distinctly different influences upon the voltammetry responses. Correspondence between the simulated and the experimental voltammograms is found, confirming the decrease of the steady-state current is caused by the slow adsorption process. The combined adsorption parameter kaΓmax2on the Tafel–Volmer mechanism was approximately 5.0 × 10–4m s–1, where Γmax(mol m–2) is the maximum surface coverage of adsorption hydrogen atoms.

The proton/hydrogen redox couple underpins the electrochemical sciences; however, the nonunity stoichiometry of the reaction leads to distinct voltammetric complications. This Article provides a joint analytical, numerical, and experimental investigation into the reversible hydrogen evolution reaction at a platinum microelectrode. Literature obscurities and nuances are highlighted and corrected, allowing the presentation of an holistic overview of the electrochemical reaction at the reversible limit. Under such conditions, it is demonstrated, first, how the reaction may be misinterpreted as being irreversibleand, second, that the transfer coefficient for the reversible (Nernstian) hydrogen evolution reaction is equal to 2. Importantly, the use of the reversible hydrogen electrode (RHE) as a reference potential in voltammetric experiments is critically evaluated.

A novel 2-deoxyribose-5-phosphate aldolase (LbDERA) was identified from Lactobacillus brevis, with high activity, excellent thermostability and high tolerance against aldehyde substrates. The half-lives of LbDERA incubated in 300 mM acetaldehyde and chloroacetaldehyde were 37.3 and 198 min, respectively, which are 2- and 7-fold higher than those of EcDERA from Escherichia coli. The crystal structure of LbDERA determined at 1.95 Å resolution revealed a stable quaternary structure which might account for its excellent aldehyde tolerance. A single mutation, E78K, was introduced to LbDERA through a consensus sequence approach, resulting in significant improvements of both thermostability and aldehyde tolerance. According to the crystal structure of LbDERAE78K, two additional hydrogen bonds and one salt bridge were introduced compared with wild-type LbDERA. As a result of its high substrate tolerance, LbDERAE78Kcould efficiently catalyze a sequential aldol condensation with 0.7 M chloroacetaldehyde and 1.4 M acetaldehyde, affording a key chiral precursor of statins, (3R,5S)-6-chloro-2,4,6-trideoxyhexapyranoside, with an unprecedented space-time yield of 792.5 g L−1d−1and only 2.5 g L−1of catalyst loading.

The inspection of lightning protection system is developing gradually presently,while it is still in a relatively backward stage, data acquisition method is old, measuring conclusion is not real-time, data measurement is out of line with the standardization of lightning business. The service area and toll station of highway prone to lightning strikes for the isolated location. Meantime, there is no lightning protection facility or facilities are not perfect, lightning accidents happen sometime which caused heavy losses to the highway with a lot of sensitive electronic equipment. The inspection of highway lightning protection should be strengthened, the mobile data acquisition technology is introduced to the highway lightning inspection in order to make the lightning protection devices play an effective role. The data acquisition process and application are described, it can ensure the diversification of data, the real-time transmission of field testing data, further standardization of the lightning inspection process and the quality of inspection to ensure safe and effective operation of building and electromechanical systems.

As a most grievous meteorological disaster in the world, lighting calamity is destructive and could be inducement of fire explosion accidents. Lightning protection business mainly consists of lightning protection devices (LPD) inspection, lightning disasters authentication and lightning risk evaluation, for which on-site data gathering is indispensible, and the status in quo has common defects. Based on tablet-PC and mobile information technology, we developed a platform including such seven modules as system configuration, LPD inspection of existing buildings, LPD inspection of under-construction buildings, identification of lightning disasters, lightning risk assessment, location of inspected clients and location of inspectors. Mobil GIS technology and fast-developing method of forms were taken into use as key technologies. With its help, an promotion of facilitation, standardization and intellectualization of lightning protection business is achieved.

Thunderstorm days are used to show the lightning frequency happened somewhere. Through comparative analysis on the thunderstorm days data of 66 weather stations in Jiangsu province and lightning data of ADTD lightning monitoring, it presents that the correlation coefficient r became the largest at 11 km range, and the longest distance of thunderstorm artificial observation is 11-14 km. According to the study of lightning activities in Jiangsu province to get the statistical fitting function curve equation by thunderstorm days and ground flash density data. Test the actual application effect of fitting equation by calculating the average relative error of fitting equation and standard equation, then make sure the actual thunderstorm days in Jiangsu province and the cloud to ground flash density formula is Ng=0.0224 Td1.48.

Global optimal path planning is always an important issue in mobile robot navigation. To avoid the limitation of local optimum and accelerate the convergence of the algorithm, a new robot global optimal path planning method is proposed in the paper. It adopts a new transition probability function which combines with the angle factor function and visibility function, and at the same time, sets penalty function by a new pheromone updating model to improve the accuracy of the route searching. The results of computer emulating experiments prove that the method presented is correct and effective, and it is better than the genetic algorithm and traditional ant colony algorithm for global path planning problem.

To address the issue that how the EEG-EMG signals change according to different motion modes, an experiment was conducted on ten subjects with three tasks performing the voluntary, stimulated and imaginary finger flexion activities. The experiment was set two programs including 10s and 2s time intervals. Electroencephalogram (EEG) from C3/C4 channels and electromyogram (EMG) from flexor digitorum superficialis were recorded simultaneously. Besides the threshold detection of wave peaks between two points, morlet wavlet-based time-frequency analysis was adopted to study the independent variation mechanisms between EEG and EMG under different motion modes. The results indicated that EMG signals of 2s intervals exhibited a similar trend with 10s intervals. The EMG energy increased over 50 Hz when actions occurred. On the contrary, there were no significant changes in imaginary task. In addition, EEG signals performed obviously different. No pronounced similar changes were found in 10s and 2s intervals. Finally, the results demonstrated that EEG-EMG causality was high during 2s intervals in stimulation task.

Fabrication of self-assembled film used 792(silane coupling agent), citric acid, boric acid and triethanolamine as the main materials. Through orthogonal experiment and the single factor analysis experiment, the optimum technological conditions are 792 10mL·L-1, citric acid 24g·L-1, boric acid 8g·L-1, triethanolamine 7.5mL·L-1, temperature 20~21°C, time 10min. Simulated sea water (3% Sodium chloride) is applied to measure the effects of the film. The results show that the processing is simple, tank liquor is stable, and film is uniform, dense and colorless.

Zinc Borate with low water is one of excellent inorganic flame retardant additives. Among Zinc Borate, ZB-2335 (2ZnO·3B2O3·3.5H2O) is widely used. The paper selects borax and Zinc sulfate as raw materials. Through orthogonal experiment and the single factor analysis experiment, the optimum technological conditions are determined: temperature is 338K, ZnSO4·7H2O and Na2B2O7·10H2O mixture ratio is 1:1.26, liquid and solid ratio is 6.5:1, reaction time is 6h. The product prepared under these conditions is in good agreement with theoretical value.

In order to improve vehicle performance, an electromagnetic damping vibration absorber was designed. The parameter formula of an electromagnetic damping vibration absorber was derived from the utilization of the electromagnetic induction principle. At the same time, the electromagnetic force which was generated by induction current replaced the damping force. According to the experimental data, the damping parameter was calculated by means of the response surface method. It showed that the application of the vibration absorber was feasible.

Well-redispersed ceria nanoparticles (CeO2NPs) were synthesized by a simple hydrothermal method. The prepared CeO2NPs exhibited excellent catalytic activity towards classical peroxidase substrate 3,3,5,5-tetramethylbiphenyl dihydrochloride (TMB·2HCl) in the presence of H2O2, based on which a colorimetric method that is highly sensitive and selective was developed for glucose detection. The composition, structure, morphology and peroxidase-like catalytic activity of CeO2NPs are investigated in detail by using X-ray diffraction (XRD), transmission electron microscopy (TEM), high resolution transmission electron microscopy (HRTEM), selected area electron diffraction (SAED), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectrometer (FT-IR), thermal analysis (TG) and UV-vis absorption spectroscopy. According to this method, the detection of H2O2and glucose are in linear range from 6.0 × 10−7to 1.5 × 10−6mol L−1and 6.6 × 10−6to 1.3 × 10−4mol L−1, with the detection limit down to 5.0 × 10−7mol L−1H2O2and 3.0 × 10−6mol L−1glucose, respectively. Further, this simple, cheap, highly sensitive and selective colorimetric method for glucose detection was successfully applied for the determination of glucose in human serum samples.

Study the properties of GFRP confining concrete column through eight groups columns subjected to axial loading, and receive the stress-strain curves; various design parameters, such as amounts of GFRP sheets, width of straps and spacing of straps, have been considered. The results are: The axial compressive strength and ductility of concrete prism wrapped by GFRP sheets or GFRP straps have all increased to a certain degree, the process of destruction of concrete prism wrapped by GFRP become slower than common concrete prism. Based on the test results, deducing and validating the stress-strain relationship model of GFRP confining concrete prism.

In this paper, a composite ZSM-5/MCM-48 material prepared by two step crystallization method has been characterized by XRD, nitrogen sorption, SEM and TEM. The activity and selectivity for the aromatization of methanol on ZSM-5/MCM-48 catalyst were much higher than these on H-ZSM-5, which may attribute to enhance the mass transfer properties and reduced diffusional limitations.

In this study, the microcapsule was prepared by emulsion crosslinking method, using trisodium trimetaphosphate (TSTP) as crosslinking agent. The microcapsule was named OMA-MPs because the mixture of Octenyl succinic starch ester(OSA-starch)，maltodextrin starch and arabic gum were used as raw material. Orthogonal experiment was used in this work to find the relationship between the small diameter of OMA - MPs and preparation factors. The better condition is that the small diameter of OMA-MPs technique was that the stirring rate 400 rpm, the materials concentration 15%, the crosslinking agent concentration 1%, and the volume ratios(w2/w1) 120:100. Due to the processing route without the use of toxic organic solvents, it is expected that these microparticles can be used as delivery vehicles for controlled release of food and drug.

Worm cam mechanism is a common indexing mechanism which is widely used in automatic machinery. Because of its high transmission precision requirements, the mechanism has high demands on the cylinder cam. The machining methods of cylinder cam are unique and the methods of special machine tool and NC machining are the two methods that general adopted in the machining of it. In this paper, the method of CNC Programming which is used for machining cylinder cam in 5-axeses high speed DMU 70eVolution Machining Center is introduced. The machining principle of cylinder cam has been studied, and the mathematical model for CNC Programming has also been worked out. The automated compensation of cutting tool’s radius is used in this field for the first time.

The myelodysplastic syndrome (MDS) is a clonal hematologic disorder with extremely heterogeneous cell component. Diagnosis is currently depending on the dysplastic morphology of bone marrow cells and the presence of specific cytogenetic abnormalities. Since no specific marker could be identified in MDS, it is sometimes hard to distinguish from other anemias, such as aplastic anemia (AA). The cell immunotype may be helpful in differential diagnosis and predicting the disease progress. To evaluate the clinical usage of immunotyping in MDS patients, we have compared the immunotypes between 36 MDS patients, 18 patients with AA and 11 healthy controls by using flow cytometry. Moreover, in combination with karyotype analysis and prognosis indicator IPSS, the value of immunotyping was further analyzed and discussed. Our results demonstrated: In MDS-RA patients, the distribution of surface antigen on BM cells had no preferential difference between lymphoid and myeloid lineages. In RAEB patients, expressions of CD13, CD33 and CD34 were prevailing, significantly higher than others(P<0.05). In contrast, in AA patients, expressions of CD2, CD7, CD19, CD20 were significantly higher than other surface antigens(P<0.05). In high risk MDS (RAEB/RAEB-t) patients, the cells expressing B cell lineage antigens (CD19,CD20) are markedly less than that in healthy people(P<0.05)while percentage of myeloid lineage antigen (CD13,CD33) are much higher than that in health control(P<0.05), However, in low risk MDS patients (RA) the frequency of expression of all myeloid, T and B lymphoid lineage antigens were not different from that in health controls. The distribution pattern of these three lineage antigens in high risk MDS patients was remarkably different with those of AA patients(P<0.05). Comparing with low-risk MDS, high-risk MDS expressed more myeloid lineage antigens (P<0.05)while expressed less B lineage antigens(P<0.05). In MDS patients, expression of CD14 was correlated with special chromosomal abnormalities defined by IPSS Our results suggested: The pattern of immunotype distribution in MDS cells are highly heterogenous. Expressions of myeloid antigens was predominant in MDS patients. Immunotyping can be helpful for discriminating MDS from AA and for monitoring the disease progression from low risk to high risk MDS. Expressions of certain immunotype are correlated with prognosis of MDS.
