In plants, recent studies have demonstrated links between the regulation of developmental processes and chromatin dynamics and organisation. Analysis of new mutations affecting overall plant architecture, leaf development and flowering time in Arabidopsis has allowed us to clone and characterise LHP1, the Drosophila heterochromatin protein 1 (HP1) homologue. LHP1 has the chromo and chromo shadow domains central to the function of animal proteins. Yeast two hybrid studies and in planta deletion experiments suggest similar modes of action in plants and animals via homodimer formation. In vivo localisation experiments revealed a specific subnuclear protein distribution in foci throughout the nucleus. Our data suggest that LHP1 may act as a main regulator of gene expression in plants, through formation of heterochromatin-like repressive complexes, to control developmental pathways involved in organ and cell size, and the vegetative to reproductive phase transition.

The process of organ positioning has been addressed, using the pin-formed 1 (pin1) mutant as a tool. PIN1 is a transmembrane protein involved in auxin transport in Arabidopsis. Loss of function severely affects organ initiation, and pin1 mutants are characterised by an inflorescence meristem that does not initiate any flowers, resulting in the formation of a naked inflorescence stem. This phenotype, combined with the proposed role of PIN1 in hormone transport, makes the mutant an ideal tool to study organ formation and phyllotaxis, and here we present a detailed analysis of the molecular modifications at the shoot apex caused by the mutation. We show that meristem structure and function are not severely affected in the mutant. Major alterations, however, are observed at the periphery of the pin1 meristem, where organ initiation should occur. Although two very early markers of organ initiation, LEAFY and AINTEGUMENTA, are expressed at the periphery of the mutant meristem, the cells are not recruited into distinct primordia. Instead a ring-like domain expressing those primordium specific genes is observed around the meristem. This ring-like domain also expresses a boundary marker, CUP-SHAPED COTYLEDON 2, involved in organ separation, showing that the zone at the meristem periphery has a hybrid identity. This implies that PIN1 is not only involved in organ outgrowth, but that it is also necessary for organ separation and positioning. A model is presented in which PIN1 and the local distribution of auxin control phyllotaxis.

Cytokinins are involved in plant cell proliferation leading to plant growth and morphogenesis. The amp1mutant of Arabidopsishas a five-fold higher level of cytokinin than the wild type plant. The mutant has a number of phenotypes with apparently increased cell proliferation rate similar to those seen in plants transformed with an iptgene and having higher cytokinin levels. In order to identify molecular factors responsible for this cytokinin-induced higher cell division rate in the mutant we have examined cyclin genes expression levels. While expression of the CycD1and cycCcyclins is not altered or is slightly decreased in the mutant, cyclin CycD3, is more highly expressed. The level of the CycD3expression decreases in a “revertant” of amp1, meaning that the over-expression of this particular cyclin is linked to the phenotype of the amp1mutant. Thus the expression of a specific G1 cyclin is upregulated in a mutant which has more cytokinin than the wild type.

Abstract: In this paper, we describe a novel plant mutant affected exclusively in the female mitosis-meiosis switch. The major effect of the swi1 mutation in Arabidopsis thaliana L. is to delay megasporogenesis events by inserting additional mitotic divisions of the mega- sporocyte. As a result of this delay, megagametogenesis is also affected. The absence of cellular polarity in the megasporocytes was also observed. Ovule ontogenesis is not affected by the mutation. The swi1 mutant is particularly interesting for studying sporophyt-gametophyte interactions. The swi1 mutation, obtained from a T-DNA tagging experiment, is monogenic recessive and mapped on chromosome five, at 16 cM from the yellow inflorescence marker.

Abstract:: Classical differential geometry can be encoded in spectral data, such as Connes' spectral triples, involving supersymmetry algebras. In this paper, we formulate non-commutative geometry in terms of supersymmetric spectral data. This leads to generalizations of Connes' non-commutative spin geometry encompassing non-commutative Riemannian, symplectic, complex-Hermitian and (Hyper-) K�hler geometry. A general framework for non-commutative geometry is developed from the point of view of supersymmetry and illustrated in terms of examples. In particular, the non-commutative torus and the non-commutative 3-sphere are studied in some detail.

Abstract:We reconsider differential geometry from the point of view of the quantum theory of non-relativistic spinning particles, which provides examples of supersymmetric quantum mechanics. This enables us to encode geometrical structure in algebraic data consisting of an algebra of functions on a manifold and a family of supersymmetry generators represented on a Hilbert space. We show that known types of differential geometry can be classified in terms of the supersymmetries they exhibit. Our formulation is tailor-made for a generalization to non-commutative geometry, which will be presented in a separate paper.

The Arabidopsis thaliana hypocotyl is widely used to study the effects of light and plant growth factors on cell elongation. To provide a framework for the molecular-genetic analysis of cell elongation in this organ, here we describe, at the cellular level, its morphology and growth and identify a number of characteristic, developmental differences between light-grown and dark-grown hypocotyls. First, in the light epidermal cells show a characteristic differentiation that is not observed in the dark. Second, elongation growth of this organ does not involve significant cortical or epidermal cell divisions. However, endoreduplication occurs, as revealed by the presence of 4C and 8C nuclei. In addition, 16C nuclei were found specifically in dark-grown seedlings. Third, in the dark epidermal cells elongate along a steep, acropetal spatial and temporal gradient along the hypocotyl. In contrast, in the light all epidermal cells elongated continuously during the entire growth period. These morphological and physiological differences, in combination with previously reported genetic data (T. Desnos, V. Orbovic, C. Bellini, J. Kronenberger, M. Caboche, J. Traas, H. Höfte [1996] Development 122: 683-693), illustrate that light does not simply inhibit hypocotyl growth in a cell-autonomous fashion, but that the observed growth response to light is a part of an integrated developmental change throughout the elongating organ.

Ticks of the Ornithodorus moubata Murray (Ixodoidea, Argasidae) complex, distributed through East Africa, are known as vectors of African relapsing fever1. A laboratory strain of O. moubata porcinus (originally from Tanzania and bred according to the method of Geigy and Herbig1) uses the guinea-pig (Cavia porcellus L.) as experimental host. Guinea pig blood crystallises more easily than that of other hosts2in the gut of blood-sucking arthropods3. In O. moubata crystals appear 5–10 d after the blood meal, first small (0.015–0.05 mm) and clear red, later up to 0.2 mm in all three directions and dark red. These ‘tick-grown’ crystals may still be present after even more than a year. They possibly serve as nutrient reserve4since they finally disappear after prolonged starvation. We describe here a study of the formation and structure of these crystals and the changes within these crystals with time. We show the crystals to be composed principally of guinea-pig haemoglobin.
