Extracts of plants, such as papaya, pineapple and fig, are known to be effective at killing intestinal nematodes that inhabit anterior sites in the small intestine, such as Heligmosomoides polygyrus. In this paper, we demonstrate that similar in vitroefficacy also occurs against a rodent nematode of the large intestine, Trichuris muris, and confirm that the cysteine proteinases present in the plant extracts are the active principles. The mechanism of action of these enzymes involved an attack on the structural proteins of the nematode cuticle, which was similar to that observed with H. polygyrus. However, not all plant cysteine proteinases were equally efficacious because actinidain, from the juice of kiwi fruit, had no detrimental effect on either the motility of the worms or the nematode cuticle. Papaya latex was also shown to significantly reduce both worm burden and egg output of mice infected with adult T. muris, demonstrating that enzyme activity survived passage to the caecum and was not completely inactivated by the acidity of the hosts stomach or destroyed by the gastric or pancreatic proteinases. Thus, the cysteine proteinases from plants may be a much-needed alternative to currently available anthelmintic drugs due to their efficacy and novel mode of action against different gastrointestinal nematode species.

We examined the mechanism of action and compared the anthelmintic efficacy of cysteine proteinases from papaya, pineapple, fig, kiwi fruit and Egyptian milkweed in vitro using the rodent gastrointestinal nematode Heligmosomoides polygyrus. Within a 2 h incubation period, all the cysteine proteinases, with the exception of the kiwi fruit extract, caused marked damage to the cuticle of H. polygyrus adult male and female worms, reflected in the loss of surface cuticular layers. Efficacy was comparable for both sexes of worms, was dependent on the presence of cysteine and was completely inhibited by the cysteine proteinase inhibitor, E-64. LD50 values indicated that the purified proteinases were more efficacious than the proteinases in the crude latex, with purified ficin, papain, chymopapain, Egyptian milkweed latex extract and pineapple fruit extract, containing fruit bromelain, having the most potent effect. The mechanism of action of these plant enzymes (i.e. an attack on the protective cuticle of the worm) suggests that resistance would be slow to develop in the field. The efficacy and mode of action make plant cysteine proteinases potential candidates for a novel class of anthelmintics urgently required for the treatment of humans and domestic livestock.

The release of neurotransmitter was monitored at the neuromuscular junctions of larval housefly ventrolateral muscles 6a and 7a, using intracellular recording, and a loose patch clamp technique to isolate discrete release sites. Transmitter release occurred spontaneously and could also be evoked by neural stimuli. Spontaneous discharges consisted of events which were randomly distributed in time and of bursts of temporally ordered events. Evoked and spontaneous release occurred in a quantal manner. The quantal content of evoked excitatory postsynaptic currents (EPSCs) was dependent upon the extracellular calcium concentration, increasing with a 3.8 power dependency. The relationship between the quantal content of a response and extracellular calcium concentration was offset by the presence of magnesium in the bathing saline. The rates of decay of miniature EPSCs (mEPSCs) and EPSCs were also found to increase with extracellular calcium concentration, consistent with a non-diffusion limited block of the glutamate receptor-channel complex by calcium ions (KB 2.5×104 s-1 M-1,P<0.01). The frequency of random mEPSCs (0.26±0.32 Hz,n=24 cells) was independent of the extracellular calcium concentration. Random mEPSCs were not inhibited by 1 µM tetrodotoxin which blocked mEPSC bursts and neurally evoked responses. EPSCs evoked during mEPSC bursts had a significantly lower quantal content than those EPSCs recorded from the same nerve terminal between bursting, indicating that both of these forms of release recruited quanta from a common pool of transmitter. Following a neurally evoked EPSC the mEPSC frequency was potentiated severalfold, this delayed release was influenced by EPSCs with large quantal contents evoked in saline containing elevated calcium concentrations. The alpha-cyano pyrethroid insecticide, cypermethrin, when applied at 10 nM concentration to this NMJ, caused a sustained 10-fold increase of transmitter quanta (240% of control), and blocked neurally evoked EPSCs after ca. 20 min (22°C). mEPSC frequency remained elevated for several hours after treatment with cypermethrin (ca. 3 Hz), before declining below control levels (0.26 Hz). A model based upon the frequency of mEPSC discharges, the quantal content of evoked responses, and the association constants of calcium to the four binding sites of calmodulin was used to investigate a possible role for this metalloprotein in both the spontaneous and evoked release of neurotransmitter. The probable lack of involvement of calmodulin in the mechanism of neurotransmitter release at the larval housefly neuromuscular junction is discussed in the light of this model, as are the implications of these findings for vesicular amino acid release in relation to pyrethroid insecticide action.

Nerve-ganglion preparations from rat dorsal spinal nerve roots were maintained in organotypic culture for 20 h. Free axonal sprouts formed at the cut tips. Clear and dense-core vesicles, mitochondria and smooth endoplasmic reticulum accumulated in the axons for a distance of 500 µm behind the cut, as has previously been described in dorsal roots sectioned in vivo. Sprouting did not occur in dorsal roots maintained in culture without their ganglia attached. Sprouting was also prevented by demecolcine (3 × 10-7 M) which reduced the number of microtubules in non-myelinated, small myelinated and large myelinated axons to respectively 45, 30 and 20% of control values. The sprouts contained several types of vesicle including small clear vesicles, large and small dense-core vesicles and flattened vesicles. The possible relevance of the vesicles to transmitter mechanisms in these neurones is discussed.

Zinc iodide-osmium (ZIO) impregnation of rat dorsal root ganglia differentially stained various elements in the neuronal cells, particularly their Golgi bodies. On the basis of this differential ZIO staining dorsal root ganglion neurones have been classified into seven types. The ultrastructure of these is described and the numbers of each type in the L4 dorsal root ganglion have been determined. Prolonged nerve stimulation did not change the relative numbers of the different cell types suggesting that none of the differences between cell types represents differences in their state of activity. The possibility is discussed that differences in morphology may reflect differences in neurotransmitter function.

Rat dorsal spinal nerve roots were cut; 20 h later the axons in the vicinity of the cut were examined by light and electron microscopy. The changes in the cut tip distant from the ganglion were largely degenerative. On the ganglionic side of the cut a cap of free unmyelinated sprouts was formed. These sprouts contained clear and dense-core vesicles 40–150 nm in diameter, smooth endoplasmic reticulum and mitochondria. Some of the unmyelinated sprouts were extensions of myelinated axons, others arose from myelinated axons by lateral budding. In both myelinated and non-myelinated axons there was an accumulation of mitochondria, tubulo-vesicular smooth endoplasmic reticulum and large and small dense-core vesicles for a distance of approximately 500 µm behind the tip. Dense-core vesicles were more common in nonmyelinated axons than in their myelinated counterparts. In areas of intense accumulation the non-myelinated fibres were grossly swollen and distorted. The myelinated axons and some of the sprouts contained an unusual type of mitochondrion. The similarity between these sprouts and pre-synaptic terminals is discussed.

Rat dorsal spinal nerve roots were cut and the tip on the ganglionic side of the cut was examined by scanning electron microscopy at 0, 7, 20 and 48 h after operation. Seven hours after cutting, free axonal sprouts had started to protrude from the cut end of the nerve. After 20 h the free sprouts were more profuse than at 7 h but were smaller and had a rougher surface. At both 7 and 20 h many of the sprouts consisted of a stalk 2–7 µm in diameter with a bulbous end 5–20 µm in diameter. A few branching sprouts were seen. At 48 h the sprouts were shrunken with a deeply furrowed surface. The significance of the surface structure of the sprouts is discussed.

Corrugated areas of sarcolemma were observed in serial transverse sections. Complexes of intramembranous particles that appear after freeze-fracture replication were concluded to represent the same specialisation. These specialisations, up to 14 µm x 2 µm, are orientated parallel to the long axis of the muscle. Intramembranous particles are concentrated along the peaks of the corrugations, and are associated with the P-face. Corresponding pits are found in the E-face. Fourteen and thirty days after sectioning the excitatory motor-nerve supply to the muscle, corrugated areas 0.5–1 µm x 0.5–1 µm are found. Occurring singly or in groups, their orientation with respect to the long axis of the muscle is more variable than those of control muscles. Thin sections reveal no complementary areas on adjacent fibres or intracellular submembrane attachments or specialisations. A structural role is therefore unlikely. Mitochondria are frequently found in close association with these specialisations. Their possible role as receptors or transmembrane transport systems is discussed.

The ultrastructure of locust (Schistocerca gregaria) neuromuscular junctions as observed in freeze-fracture replicas is described and correlated with their appearance in thin sections of material prepared by various fixation regimes and by use of the sectioned replica technique. Two main specialisations are found at the junction. Postsynaptically there is a concentration of large intramembranous particles partitioning on fracturing with the E-face of the sarcolemma. These are associated with smaller, less defined subparticles thought to be proteinaceous projections seen in thin section. Presynaptically a raised row of particles is present on an elliptical plateau of the P-face of the nerve membrane, corresponding to a densely staining bar present in thin sections prepared using tannic acid and methylamine tungstate. These specialisations are found in both red and white fibres, indicating that the junctions are fast excitatory rather than inhibitory junctions. The bar-shaped particle array is not present 14 days after denervation. These specialisations are compared with those at synapses previously described and their possible function at a glutamatergic neuromuscular junction is discussed.

Freeze-fracture replicas of the neuromuscular junction were prepared from untreated retractor unguis muscles of the locust Schistocerca gregaria that were rapidly frozen by contact with a copper block cooled by liquid helium. These replicas were compared with others prepared from tissue following fixation with glutaraldehyde and cryoprotection in glycerol. Freeze-fracture of rapidly frozen tissue produced replicas of high quality with little evidence of tissue damage by ice crystals in the superficial layers. The gross fracturing characteristics of the neuromuscular junction were consistent with replicas from fixed and cryoprotected tissue; all of the membrane specializations were recognisable but with some alterations in infrastructure. In tissue replicas prepared using either method intramembranous particles in the presynaptic membrane were arranged in a bar-like array. The intramembranous particles of this presynaptic bar array of the rapidly frozen material were large and found on the E-face of the cleaved membrane. This contrasts with the P-face distribution of the comparable particles in muscles fixed in glutaraldehyde and cryoprotected in glycerol, in which they are also smaller and more numerous. This difference in partitioning between rapidly frozen, and fixed, cryoprotected nerve terminals is not found at cholinergic synapses and thus may reflect functional differences between the two types of junction.
