ProfileXpert is a genomic facility from the Federative Institute of Neuroscience of Lyon, specialized in the analysis of transcriptome and genotyping using the DNA microarray technology. ProfileXpert was founded in 2003 to implement these technologies into the Federative Institute of Neuroscience of Lyon and in the Rhône-Alpes area. ProfileXpert has three objectives: to provide a global DNA microarray (transcriptome and genotyping) service, to reinforce teaching and training activity in genomic and finally, to develop research and development projects, focused on innovating micro- and nanogenomic tools and on the identification of clinical molecular markers. ProfileXpert is equipped with state-of-the-art required hardware and operates by highly-qualified staff. ProfileXpert is characterized by its capacities: to ensure two complementary approaches (transcriptome and genotyping); to propose a wide range of chips (expression, exons, promoters, mapping, tiling), applications (qualitative and quantitative expression, alternative splicing, loss of heterozygoty, copy number variation analysis) and genomes (human, mouse, rat, bovine, chicken, c-elegans…); to analyse micro-amount of nucleic acids compatible with clinical samples (biopsy, isolated cells, biological fluids…) and to ensure data analysis, by using data mining and data analysis softwares.

Objectives: The objectives of this study are to identify the presence of tinnitus and classify its different forms, in terms of changes in noise.

The influence of variable durations of contralateral acoustic stimulation on the suppression of click-evoked otoacoustic emissions was investigated in order to determine whether olivocochlear efferent fibers are equally effective whatever the acoustical stimulation duration or if they show fatigue. The suppression effect was measured for contralateral stimulus durations ranging from 10 to 180 s prior to the onset of otoacoustic emission recording, and continuing throughout the recording time (60 s). No significant stimulus duration effect was found.

1. The static and dynamic sensitivities to thermal and mechanical stimuli of polymodal nociceptors in hairy skin of the anaesthetized monkey have been investigated by recording activity in their primary nerve fibres. 2. Polymodal nociceptors responded to skin pricking, pinching and heating to temperatures higher than 40 degrees C. They did not respond to touch, stretch or cold. The conduction velocity of their axons was from 0.6 to 1.1 m/sec. 3. Three types of cutaneous receptive fields have been observed: single spot‐like areas of 1‐2 mm2; multiple spot‐like areas of 1‐2 mm2; and larger areas up to 25 mm2 with heterogeneous sensitivity. 4. Polymodal nociceptors were subjected to heat stimuli that commenced from a 33 or 37 degrees C adapting temperature. A series consisted of heating their receptive fields to 43, 45, 47 and 50 degrees C from one or the other adapting temperatures at a constant rate of 0.2 degrees C/sec. Each heat stimulus intensity was maintained for 4 min after which the skin was returned to the adapting temperature. Immediately after the first series the identical series was repeated in order to determine the effect of prior heating upon the dynamic responses to re‐heating. The dynamic responses were characterized by three phases: an initiation of a discharge at a threshold level of skin temperature; a dynamic discharge during the suprathreshold change, that reached a peak frequency when the temperature reached its maximum; and an adaptation phase while the temperature remained at the high intensity. Adaptation was rapid initially, and then slowed during the final minutes at the high intensity. 5. Adapting the receptive field to either 33 degrees C or to 37 degrees C before the heat stimuli did not affect the sensitivity and the discharge pattern of the polymodal nocicpetors. 6. During the first series of stimulations, the threshold at which the individual polymodal nociceptors began to discharge to heat stimuli varied from 40 to 46.5 degrees C. The mean threshold of the population was 42.5 degrees C. 7. No change in the threshold was observed when responses to 0.2 and 1.5 degrees C/sec rates of heating were compared...

Daily food intake, masticatory activity and reticular movements were studied in sheep before and after intracerebroventricular injections of various drugs. Pour animals received nialamide (1 mg twice daily for 21 days) and four others received 6-hydroxydopamine (one injection of 4 mg). Nialamide increased food intake in all the subjects. It increased rumination time and inhibited the oral reflex of the reticulum in half of the subjects. 6-hydroxydopamine reduced food intake and rumination time and accelerated the rhythm of reticular contractions during rest and rumination. The effects of nialamide and 6-hydroxydopamine on the cerebral concentration of serotonin, noradrenaline and dopamine have been extensively documented over the past few years. Modifications of food intake, rumination and reticular motility can be explained by variations in the cerebral concentration of noradrenaline and/or dopamine. These two amines (or one of them) could be implicated in the control of rumination and could act on central mechanisms monitoring forestomach motility.

Nervous and endocrine peptidergic structures in human Brunner's glands were studied by immunofluorescence. Endocrine cells storing immunoreactive components respectively similar to somatostatin 14, the amino-terminal portion (1–14) of somatostatin 28, gastrin-cholecystokinin, and peptide YY were distributed throughout the acini. Peptidergic nerve structures contained materials immunologically related to vasoactive intestinal peptide, peptide histidine methionine, substance P, neuropeptide Y, and gastrin-releasing peptide. The latter peptide was detected in discrete fibers running into the acini but within no cell body in the submucosa. All other neuropeptides were stored in fibers, isolated or grouped in bundles, and in perikarya of submucosal ganglia close to the acini. No immunoreactive structures were detected using antisera directed against pancreatic polypeptide, secretin, motilin, neurotensin, or calcitonin gene-related peptide. The results suggest that several regulatory peptides may be involved in the control of Brunner's glands in humans.

Reproducible cerebral evoked responses have been recorded from the vertex in two men and one woman to endorectal electrical stimulation. The first component had a 36 ms latency for the woman, and an average latency of 90 ms for the men.

WE selectively imaged the neural correlates of tinnitus, by contrasting a condition with no phantom auditory sensation with a condition during which tinnitus is present, using a rare form of tinnitus elicited by eye movements. Using positron emission tomography (PET), we demonstrate that phantom auditory sensation increases regional cerebral blood flow bilaterally in temporo-parietal association auditory areas but not in the primary auditory cortex. These results confirm that conscious perception does not necessarily require activation in primary areas and suggest that the perceptual qualities of tinnitus, e.g. intensity, frequency and spatial localization, are represented in temporo-parietal regions. Activation in these regions is compatible with cortical processing of ascending auditory messages generated at subcortical levels.

A possible role of the efferent system in the mechanisms of tinnitus generation has been put forward by several authors. A simple method for studying the functioning of this system is to compare the amplitudes of otoemissions with and without mild contralateral stimulation. In a recent communication, Veuillet et al. (1991) reported that, on the side of tinnitus, the efferent system of patients suffering from unilateral tinnitus seems to be less efficient than on the other side. The results presented here correspond to those obtained in the very first tinnitus patients submitted to a research protocol exploring systematically the efferent system. When possible, the effectiveness was tested in two ways: globally, using evoked otoacoustic emissions (EOAEs) and, very precisely in the frequency zone of the tinnitus, using cubic distortion product 2f1-f2. Preliminary results obtained in bilateral and unilateral tinnitus sufferers show that a majority of them exhibits, at least in the proximity of the tinnitus, a lack of effectiveness of the efferent system. In some of them, instead of the suppressive effect, an increasing one was even observed.

Adult frogs (Rana esculenta) were given subcutaneous injections of 10, 20, 30, 50 and 100 mg/kg capsaicin in sequential order over 5 days, or the vehicle only. The nociceptive thresholds to electrical, thermal and chemical stimuli were measured before, and 1, 5 and 24 h after each injection. Capsaicin was followed by a dose-related reduction of nociceptive responses to all stimuli, but these effects lasted for only 1–5 h after the given injection. Water/acetic extracts of undivided brains and spinal cords were prepared at the corresponding time periods for the radioimmunoassay of peptides. Spinal cord concentrations of immunoreactive substance P were essentially unaffected by capsaicin, while those of immunoreactive somatostatin were significantly increased after the second for fourth injections (20, 30 and 50 mg/kg) of capsaicin. Brain extracts showed an increase of somatostatin and substance P concentrations after the dose of 50 mg/kg. In an additional experiment, immunoreactive substance P, somatostatin and cholecystokinin were measured in tissue samples taken at 2 and 10 min, and 1, 5 and 24 h after a single dose of either 50 mg/kg capsaicin or the vehicle. The only signficant effect of capsaicin was an increase of immunoreactive somatostatin concentration in brain homogenates at 5 h, while the vehicle in itself elicited major variations of all three peptides in spinal cord and/or brain. These results indicate that capsaicin reduces the nociceptive responses to cutaneous stimuli in adult frogs. This effect is transient, and bears no clear relationship to the variations of spinal cord nor of brain concentrations of immunoreactive substance P, somatostatin and cholecystokinin. In the present experimental conditions, the effects of the vehicle injection to neuropeptides far exceeded those of capsaicin itself.
