Chitosan oligosaccharides longer than a hexamer have higher bioactivity than polymer or shorter oligosaccharides, such as the monomer or dimer. In our previous work, we generated Paenibacillus fukuinensischitosanase-displaying yeast using yeast cell surface displaying system and demonstrated the catalytic base. Here we investigated the specific function of putative four amino acid residues Trp159, Trp228, Tyr311, and Phe406 engaged in substrate binding. Using this system, we generated chitosanase mutants in which the four amino acid residues were substituted with Ala and the chitosanase activity assay and HPLC analysis were performed. Based on these results, we demonstrated that Trp159 and Phe406 were critical for hydrolyzing both polymer and oligosaccharide, and Trp228 and Tyr311 were especially important for binding to oligosaccharide, such as the chitosan-hexamer, not to the chitosan polymer. From the results, we suggested the possibility of the effective strategy for designing useful mutants that produce chitosan oligosaccharides holding higher bioactivity.

Binding of a highly de-N-acetylated chitosan to Japanese pheasant lysozyme (JPL), which differs from hen egg white lysozyme (HEWL) by nine amino acid substitutions (including Arg114→His), was investigated by 1H-NMR spectroscopy. The profile of the one-dimensional spectrum of JPL is essentially identical to that of HEWL. Using two-dimensional spectral of JPL and HEWL, several aromatic and aliphatic proton resonances of JPL were assigned by comparison. When a highly de-N-acetylated chitosan (number-average degree of polymerization, about 18; degree of acetylation, 0.04), where the N-acetylated units are predominantly surrounded by de-N-acetylated units (a monoacetylated chitosan), was added to the JPL solution, the NMR signals were clearly affected in Trp28 C5H and Ile98 γCH, as in the case of binding to HEWL. The dissociation constant of the monoacetylated chitosan evaluated from the NMR signal responses was calculated to be 0.23±0.05 mM (−31.5 kJ/mol), which is similar to that of HEWL (0.11±0.02 mM, −33.3 kJ/mol). Thus, the Arg→His substitution of the 114th amino acid, which participates in sugar residue binding at the right-sided subsite F, did not significantly affect the chitosan binding. In addition, the C2H signal of His114 of JPL was not affected by the chitosan binding. These results suggest that the monoacetylated chitosan binds to subsites E and F through the left-sided binding mode.

Inspired by the fact that chitosan is a representative constituent of the ectocellular structure of Cryptococcus neoformansand a typical biomaterial for improving drug oral absorption, we designed an elegant and efficient C. neoformans-targeted drug delivery system via oral administration. A chitosan-binding peptide screened by phage display was used as the targeting moiety, followed by conjugation to the surface of poly(lactic-co-glycolic acid) nanoparticles as the drug carrier, which was then incubated with free chitosan. The noncovalently bound chitosan adheres to mucus layers and significantly enhances penetration of nanoparticles through the oral absorption barrier into circulation and then re-exposed the targeting ligand for later recognition of the fungal pathogen at the site of infection. After loading itraconazole as a model drug, our drug delivery system remarkably cleared lung infections of C. neoformansand increased survival of model mice. Currently, targeted drug delivery is mainly performed intravenously; however, the system described in our study may provide a universal means to facilitate drug targeting to specific tissues and disease sites by oral administration and may be especially powerful in the fight against increasingly severe fungal infections.

Chitosan is a cationic biopolymer that has many potential applications in the food industry because of its unique nutritional and physicochemical properties. Many of these properties depend on its ability to interact with anionic surface-active molecules, such as surfactants, phospholipids, and bile acids. The purpose of this study was to examine the influence of pH (3 and 7), ionic strength (0−200 mM NaCl), and temperature (10−50 °C) on the interactions between a model anionic surfactant (sodium dodecyl sulfate, SDS) and chitosan using isothermal titration calorimetry, selective surfactant electrode, and turbidity measurements. At pH 3 and 30 °C, SDS bound strongly to chitosan to form an insoluble complex that contained about 4−5 mmol of SDS/1 g of chitosan at saturation. When SDS and chitosan were mixed at pH 7 they did not interact strongly, presumably because the biopolymer had lost most of its positive charge at this pH. However, when SDS and chitosan were mixed at pH 3 and then the solution was adjusted to pH 7, the SDS remained bound to the chitosan. The presence of NaCl (0−200 mM) in the solutions decreased the critical micelle concentration (cmc) of SDS (in both the absence and the presence of chitosan) but had little influence on the amount of SDS bound to chitosan at saturation. The cmc of SDS and the amount of SDS bound to the chitosan at saturation were largely independent of the holding temperature (10−40 °C). Nevertheless, the enthalpy changes associated with micelle dissociation were highly temperature-dependent, indicating the importance of hydrophobic interactions, whereas the enthalpy changes associated with SDS−chitosan binding were almost temperature-independent, indicating the dominant contribution of electrostatic interactions. This study provides information that may lead to the rational design of chitosan-based ingredients or products with specific nutritional and functional characteristics, for example, cholesterol lowering.

Chitosan has emerged as a promising material for biomedical applications. However, the effect of chitosan adsorption on the structure of model biomembrane is not known. In this study, atomic force microscopy (AFM) is employed to investigate the interaction between chitosan and mica-supported dipalmitoylphosphocholine (DPPC) bilayer. First, in situ AFM measurement indicates that nucleation of chitosan occurs around the membrane defects at the initial stage of chitosan incubation. Eventually, DPPC−chitosan binding and chitosan intermolecular association lead to chitosan aggregation on the membrane surface which is quantified by average height measurement and RMS roughness analysis. Lateral force microscopy (LFM) confirms that the adsorbed chitosan has distinct material properties. Furthermore, the trend of surface pressure−area isotherms supports the condensation of DPPC monolayer induced by chitosan in the aqueous subphase. Surface coverage and surface roughness analysis show that the extent of chitosan aggregation on the supported membrane is affected by the incubation time during long-term chitosan incubation.

Chitosan, a deacetylated chitin derivative, behaves like a general elicitor, inducing a non-host resistance and priming a systemic acquired immunity. The defence responses elicited by chitosan include rising of cytosolic H* and Ca2+, activation of MAP-kinases, callose apposition, oxidative burst, hypersensitive response (HR), synthesis of abscissic acid (ABA), jasmonate, phytoalexins and pathogenesis related (PR) proteins. Putative receptors for chitosan are a chitosan-binding protein, recently isolated, and possibly the chitin elicitor-binding protein (CEBiP). Nevertheless, it must be pointed out that biological activity of chitosan, besides the plant model, strictly depends on its physicochemical properties (deacetylation degree, molecular weight and viscosity), and that there is a threshold for chitosan concentration able to switch the induction of a cell death programme into necrotic cell death (cytotoxicity).

This work develops a simple and innovative test-strip to monitor antibiotics in aquaculture facilities by an equipment-free approach. It consists of a low-cost disposable cellulose paper that was chemically modified to produce a colour change when in contact with a given antibiotic. In brief, the cellulose substrate was subject to oxidation with periodate, followed by amination with chitosan binding and modification with Cu(II). The test strip was then dipped in the target solution and the intensity of the colour generated therein revealed the concentration of antibiotic present for concentrations higher than 0.5 mM. The higher the concentration in sulfadiazine (SDZ), the more intense the pink colour formed in the final solution, which was also turbid due to the insolubility of the formed product. This colour intensity also varied linearly with the logarithm of the SDZ concentration (from 0.5 to 5 mM), when plotted against the sum of the RGB coordinates extracted from digital pictures. The linear equation of this response was represented by (R + G + B) = − 256.1 log(SDZ, mol/L) − 362.0, with an R-squared of 0.9913. The test-strip was stable for at least 15 days and was selective in the presence of tetracycline and difloxacin, while the response to other members of the sulfadiazine family requires prior evaluation. Overall, the test-strips developed herein are inexpensive and provide valuable (semi-) quantitative data for monitoring SDZ in waters, a most valuable approach to control and reduce the level of antibiotics in fish tanks, which in turn may reduce the costs of fish production and the environmental concerns linked to this practice. Moreover, the test strip uses a cellulose substrate that has little environmental impact upon discard.

Chitinase J (ChiJ) from alkaliphilic Bacillus sp. strain J813 has a multidomain structure containing a catalytic domain (CatD), a fibronectin type III like domain (FnIIID) and a chitin-binding domain (ChBD). It has been shown that the ChBD binds to an insoluble chitin and enhances its degradation by the CatD. Further binding study of the ChBD was performed with a glutathione-S-transferase fusion protein. This fusion protein showed binding abilities to insoluble chitin and chitosan. Two surface-exposed aromatic residues (Trp541 and Trp542) were found in the tertiary-structure model of ChBD and targeted for mutational analysis. Single and double mutations of the two aromatic residues decreased the chitin- and chitosan-binding abilities. It was revealed that these residues would be important for substrate-binding of the ChBD.
