The pituitary gonadotropins, luteinizing hormone (LH) and follicle-stimulating hormone (FSH), are the principle endocrine drivers of reproductive processes in the gonads of jawed vertebrates. Canonically, FSH recruits and maintains selected ovarian follicles for maturation and LH induces the stages of germinal vesicle breakdown and ovulation. In mammals, LH and FSH specifically activate cognate G-protein-coupled receptors that affect the proteins involved in steroidogenesis, protein hormone synthesis, and gametogenesis. This dual-gonadotropin model also exists in some fish species, but not in all. In fact, due to their diverse number of species, extended number of ecological niches, and remarkably flexible reproductive strategies, fish are appropriate as models to understand the co-evolution of gonadotropins and their receptors. In this study, we cloned and characterized the expression profile over the final stages of ovarian maturation of carp (Cyprinus carpio) LHCGR and FSHR. Expression of both gonadotropin receptors increased in the later stage of early vitellogenesis, suggesting that both LH and FSH play a role in the development of mature follicles. We additionally tested the activation of cLHCGR and cFSHR using homologous and heterologous recombinant gonadotropins in order to gain insight into an evolutionary model of permissive gonadotropin receptor function. These data suggest that carp (Cyprinus carpio) gonad development and maturation depends on a specific gonadotropin profile that does not reflect the temporally distinct dual-gonadotropin model observed in salmonids or mammals, and that permissive gonadotropin receptor activation is a specific feature of Ostariophysi, not all teleosts.

Turbulent mixing processes and species transport govern many important properties like stability and pollutant formation of reactive systems. We propose spectrally resolved absorption tomography as a viable tool to quantitatively assess species transport by imaging tracer pulses at high time resolution. Measurements on a gas-assisted solid-fuel combustor operating in air and oxy-fuel atmospheres are presented. The measurements are conducted using a scanning tomographic imaging system that has previously been validated for gas temperature measurements. As the scanning system itself is not able to achieve the necessary time resolution, a phase-locked tomographic scheme is applied, i.e. each measurement beam is acquired separately for several tracer pulses and phase-averaged. Hence, the full 5 kHz time resolution of the underlying tunable diode laser spectroscopy system can be utilized, granting insight into species transport in the recirculation zones as well as the flame brush. Despite a possible influence of the tracer injections on the inner recirculation zone, it is shown that qualitative differences between methane combustion and gas-assisted solid-fuel operating conditions can be detected, and influences of devolatilization as well as volatile combustion become visible.

A combination of Large-Eddy Simulation (LES) and Particle Image Velocimetry (PIV) was utilized to investigate the three-dimensional in-cylinder flow within an optically accessible Direct Injection Spark Ignition (DISI) engine at motored engine operation. The PIV measurements were used to guide the meshing procedure by identifying the regions were refinements and improvements were needed. From the iteratively optimized meshes LES results are shown from two selected meshes, an intermediate coarse mesh and the final optimized mesh, and compared to PIV measurements. The evolution of the intake flow and the tumble in the central tumble plane during compression are presented and discussed. Exploitation of the LES results allowed showing the influence of out-of-plane velocities along the cylinder liner impacting the formation of the tumble flow. The optimized mesh was then used to investigate the influence of the spark plug on the in-cylinder flow. For the studied engine the spark plug had a significant impact on the evolution of the tumble flow during compression. Finally 35 engine cycles were simulated using the optimized mesh with the spark plug in place. Velocity distributions in a region below the spark plug are shown and compared with PIV results. The two-sample Kolmogorov-Smirnov test revealed a strong similarity between the velocity distributions obtained by PIV and LES, thus validating the potential of LES for investigating cycle-to-cycle variability.
