Hypoxia, defined as dissolved oxygen (DO) < 2 mg/L, in the central basin of Lake Erie has been studied since the mid‐1900s. Even so, spatial patterns of hypoxia, and episodic hypoxia in nearshore areas where drinking water plant intakes are located, are not well characterized owing to limited observations and short‐term dynamics. We evaluated a physically based, DO model with respect to patterns of hypoxia observed in Lake Erie. The DO model used assigned rates of sediment and water column oxygen demand that were temperature dependent but otherwise spatially and temporally uniform. The DO model was linked to National Oceanic and Atmospheric Administration's (NOAA) Lake Erie Operational Forecasting System hydrodynamic model, an application of the Finite Volume Community Ocean Model (FVCOM). Model temperature and DO were compared with observations from ship‐based studies, real‐time sensor networks and an array of moored sensors that we deployed in 2017. In years with dominant southwesterly winds, persistent downwelling occurred along the south shore, which resulted in a thinner thermocline and earlier initiation of hypoxia along the south shore than the north. Occasional northeast winds temporarily reversed this pattern, causing upwelling along the south shore that brought hypoxic water to nearshore locations and water intakes. The DO model reproduced observed spatial and temporal patterns of hypoxia and revealed locations subject to episodes of hypoxia, including nearshore Ohio, north of Pelee Island, and near the Bass Islands. Model skill was limited in some respects, highlighting the importance of accurate simulation of the thermal structure and spatial patterns of oxygen demand rates. We compared model temperature and DO with measurements conducted by us and by others. In 2013, The Better Image (TBI) was contacted to examine A Play of Selves, a unique work created by artist Cindy Sherman during her graduate studies in 1975. The work is composed of 244 paper doll-like photographs of Sherman that are presented within 72 frames. During initial condition checks, it was noted that a number of individual elements had begun to show signs of chemical staining. Over the course of 2013/2014, a collaborative response was developed with the help of the artist, the artist's printer, the representing gallery, TBI staff and the owner. The main goal was to address the physical and chemical issues present and preserve this work for future exhibition and study. Determining an effective decision- making model based on a hierarchy of values was crucial before undertaking any conservation treatment, as there were several invested parties, each with a different perspective. The focus of this paper is the interplay of our responsibilities as conservators with the concerns of the artist, her gallery, her printer, and the owner to balance aesthetic vs. physical integrity of the artwork, reversibility of treatments, importance of record keeping while developing a balanced response within a world of rapidly disappearing traditional photographic papers.

Low-cost compact sensors for ultrasmall systems are a pressing need in many new applications. One potential solution is a shallow aspect ratio system using a lenslet array to form multiple undersampled subimages of a scene on a single focal plane array, where processing techniques then produce an upsampled restored image. We have investigated the optimization and theoretical limits of the performance of such arrays. We have built a hardware simulator and developed algorithms to process imagery similar to that of a full lenslet imaging sensor, which allowed us to quickly test optical components, algorithms, and complete system designs for future lenslet imaging systems.

Abstract.:  Within population variation in plant density can affect reproductive success and breeding systems. We examined such effects in Listera cordata R. Br., a north temperate terrestrial orchid, in a subalpine population in Colorado, USA. Hand pollinations showed that L. cordata was self-compatible as all pollinations produced fruits. Cross-pollinations differed significantly from self-pollinations, and had a higher number of seeds per capsule and higher % of fertilized ovules. Together they could compensate for the transmission advantage of self-pollinations. Average number of flowers per inflorescence was 4.2–4.4 and average fruit set was 20.7–23.7%. By establishing 36 plots with varying plant densities (1, 4, 8 and 16) and emasculating all flowers in target plants of a set of 20 plots, we estimated density effects on fruit and seed production. Plant density had no effect on the amount of reproduction except in reduced seed production of highly dispersed plants. Our results suggest that inbreeding depression may be an important selective factor in L. cordata. Further studies are needed to define its role in the evolution of mating systems in this species.

The transport and capture of pollen in ~20% of all angiosperm families occurs in air and water. In other words, pollination is abiotic and occurs via the fluid media, not an animal vector. Whereas some early concepts considered abiotic pollination to be largely a stochastic phenomenon, there is sufficient evidence to indicate that wind pollination (i.e. anemophily) and water pollination (i.e. hydrophily) have deterministic features and are sophisticated fluid dynamic solutions to the problem of pollen release, dispersal, and capture.

Sepsis has been reported to cause mitochondrial dysfunction and inhibition of key enzymes that regulate the tricarboxylic acid (TCA) cycle. We investigated the effect of sepsis on high-energy phosphates, glycolytic and TCA cycle intermediates, and specific amino acids that are involved in regulating the size of the TCA cycle pool during changes in metabolic state of the heart. Sepsis was induced in 12 female rats by the cecal ligation and perforation technique under halothane anesthesia; seven control rats underwent cecal manipulation without ligation. At 36-42 h postsurgery, the rats were reanesthetized, the chest was opened, and the hearts were freeze-clamped. Perchloric acid extracts of the hearts were analyzed with fluorometric enzymatic methods and 31P nuclear magnetic resonance spectroscopy. There were no significant differences in the levels of the TCA cycle intermediates or high-energy phosphates between the septic and control rats. The major metabolic changes were the 28% decrease in alanine and the 31% decrease in glutamate in the septic hearts compared with control (P less than 0.05 and P less than 0.005, respectively). Phosphocholine, a component of membrane phospholipids, was increased by 91% in the septic hearts (P less than 0.01). We conclude that sepsis does not impair the TCA cycle or induce significant cellular ischemia in the heart. The increase in phosphocholine may represent significant cellular membrane disruption during sepsis.

The effects of a rust infection (Sphenosphora saphena, Basidiomycetes) on several fitness components of the orchid Tolumnia (Oncidium) variegata were quantified in a subtropical moist forest of Puerto Rico. Infected and uninfected plants of two size categories were observed and manipulated by increasing natural levels of fruit production to determine the effects of infection on short and long-term sexual reproduction, subsequent vegetative and reproductive growth, and mortality. Under artificially high levels of fruit production, infection had no effect on short or long-term reproduction through male or female function except for a modest decline in seed viability in small plants. Under natural levels of fruit production, infection was only related to a reduction in the number of leaves per shoot. At artificially high levels of fruit production, infection reduced leaf length, leaf width, and the number of live shoots. Survival was not associated with rust occurrence. The rust appears to have minor ecological impact on this orchid population.

Will increased fruit and seed production in a severely pollination-limited orchid stimulate population growth? We tested whether safe sites for germination and seedling establishment are limiting for the twig epiphyte, Tolumnia variegata, by manipulating fruit set and monitoring subsequent seedling establishment for two seasons (1991–1992, 1992–1993). In the Cambalache Forest Reserve of Puerto Rico, we established 36 plots along a transect. Each plot consisted of nine trees. A center tree was designated as the site for attaching Tolumnia and manipulating fruit set. The other eight potential host trees were 1–3 and 3–5 m from the center tree in each of the cardinal directions. A 1-m length of stem 1 m from the ground was monitored for recruits on each of the nine trees of 24 fruit-enhanced plots and 12 controls (23 and 13, respectively for the 1992–1993 season). Fruit enhancement plots were divided among two treatments: one-fruit and five-fruit additions for the 1st year and one to five and more than five fruits for the 2nd year. Availability of suitable host species was not limiting. T. variegata showed little specificity for host tree species, good host trees and shrubs were common, and there was no evidence that the orchid had a preference for small branches, despite possessing the entire suite of characteristics thought to respresent “obligate” twig epiphytes. Fruit enhancement increased seed rain and seedling establishment consistently in only the high-fruit treatment plots. Most recruitment occurred near fruiting plants. Over the 2-year period, mortality was 18% for adults and 85.5% for the 1991–1992 cohort of recruits. Net recruitment was positive for both the treatment (average = 1.74) and control plots (average = 0.67). Seedling establishment at our study site was not microsite-limited. If selection for increased pollinator attraction occurs, then an increase in seed output should result in population growth.

Iosulamide is a bis-benzoic analogue of metrizoate that shows clear advantages in animal tests over meglumine iodipamide. The intravenous toxicity of iosulamide meglumine is considerably lower than that of iodipamide (Cholografin) in the mouse and rat. The LD50in mice for iosulamide meglumine is 11,500 ± 844 mg free acid/kg and for iodipamide is 2380 ± 290 mg free acid/kg. A threefold difference in toxicity was seen in rats; the LD50for iosulamide meglumine is 13,600 ± 1710 mg free acid kg and for iodipamide is 4430 ± 310 mg free acid/kg. Iosulamide is a highly effective contrast agent for cholangiocholecystographic visualization in cats and monkeys. Speed and degree of opacification are equivalent to that of iodipamide at equimolar doses. Studies of biliary and urinary excretion patterns indicate iosulamide is rapidly excreted compared to iodipamide, while at the same time providing equal concentrations in bile on an mg/ml bile basis. A more efficient blood to bile clearance rate and a shorter blood half-life for iosulamide may account for the lower circulating blood levels and rapid total excretion compared to iodipamide. Iosulamide's rapid blood-bile clearance coupled with its extremely low toxicity may allow rapid administration of high doses, affording superior visualization and safety compared to iodipamide. It may also provide visualization of the liver parenchyma with computerized axial tomography, due to the pharmacokinetic profile that provides for high liver clearance but low blood levels. The emetic potential of iosulamide meglumine is quite low compared to iodipamide. Iosulamide meglumine also lacks hypotensive activity. Little or no effect on blood pressure was seen with iosulamide meglumine in cats or monkeys, whereas iodipamide caused marked transient, or sustained, reductions. Iosulamide meglumine did not produce significant toxic effects when administered as single daily intravenous injections to albino rats for three weeks, or in 10-minute intravenous infusions to rhesus monkeys 10 times in 14 days. Clinical trials with iosulamide are under way.

The Sunshine Mining Co. has recently started up its new silver refinery in Kellogg, ID. The new plant recovers silver and copper values from a high grade tetrahedrite concentrate using hydrometallurgical processes. The plant and process are both new and have gone through the initial startup phase during the first six months of 1985. The full scale plant was developed from laboratory test work. While the metallurgy of the process worked as expected, the material handling problems were difficult to overcome. As might be expected, those parts of the plant that represented available technology started up easily, while the parts that represented new technology presented the most difficulty. Topics covered in this paper include: 1) insufficient type and amount of agitation in the leach vessels, 2) discharge of a nonhomogeneous slurry from the leach vessels, and 3) problems associated with very low flow rates in slurry lines.

Fluorine-19-nuclear magnetic resonance (19F-NMR) spectroscopic detection of the NMR-active Ca2+ indicator 5-fluoro-1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (5F-BAPTA) is one method for measuring cytosolic free Ca2+ concentration ([Ca2+]i) and has been used previously to measure [Ca2+]i in isolated cells and perfused organs. The aim of the present investigation was to demonstrate the feasibility of determining [Ca2+]i in vivo and in situ using 19F-NMR and 5F-BAPTA. Experiments were performed on male Sprague-Dawley rats with a surface-coil antenna employed for NMR interrogation. The Ca2+ indicator, 5F-BAPTA, was infused either intravenously (kidney, spleen) or intraventricularly (brain) as a 100 mg/ml solution of the cell-permeant acetoxymethyl ester (5F-BAPTA-AM) in dimethyl sulfoxide. Rats tolerated intravenous infusion without evident change in mean arterial blood pressure. In all tissues examined, kidney, spleen, and brain, [Ca2+]i was approximately 200 nM. To our knowledge, these results represent the first in vivo and in situ determinations of [Ca2+]i employing 19F-NMR.

Soil formation is often associated with the spatial reorganization of soil components, particularly Fe oxides. However, the methods available to describe the spatial distribution of Fe oxides generally lack measurable parameters that allow comparison between different types of soils. In this work, we demonstrate the use of statistical methods (e.g., χ2goodness‐of‐fit test, mean, variance, and cluster coefficient) to describe the basic distribution pattern of Fe oxide nodules in soil thin sections. Using a computer, we mapped the spatial distribution (i.e., xand ycoordinates) of Fe oxide nodules in six soil thin sections from the B horizons of three sandy soils from Alberta, Canada. We calculated the statistical tests at a range of grid sizes (2–10 mm) with a Fortran program to (i) examine the effect of grid size on the statistical description of the distribution and (ii) determine the “appropriate scale of observation” for each thin section. We defined this scale as the grid size at which the mean number of observation is at least five, which was found to vary in size from 5 to 9 mm in our samples. We recommend the determination of the appropriate scale of observation before any statistical description of the Fe nodule distribution. In all but one of our six samples, the statistical distribution of nodules was clustered; the remaining one was uniform in distribution. The methods described in this study provide statistical methods whereby results from different studies can be compared.

1. [31P]NMR spectra were obtained from a functioning isolated perfused rat kidney with the aim of determining intrarenal pH in acute acidosis. 2. Signals from intracellular inorganic phosphate could be observed in the absence of phosphate in the perfusion medium. Under these conditions renal ATP and inorganic phosphate content fell by 30% but total adenine nucleotide and phosphorylation potential ATP/ADP x Pi were unchanged compared with kidneys perfused with phosphate‐containing medium. In addition, G.F.R., Na+ reabsorption and ammonia formation from glutamine remained normal. Ammonia production increased 93%, urine pH fell to 5.8 +/‐ 0.1 and kidney 2‐oxoglutarate content fell by 80% upon acidification of the perfusion medium from pH 7.4 to pH 6.9, findings identical with those obtained in controls (Ross & Tannen, 1979). 3. [31P]NMR spectra of the isolated perfused rat kidney showed a pattern of adenine nucleotides and a small concentration of phosphocreatine, Intra‐renal pH was measured from the resonance position of intracellular inorganic phosphate and in perfusions with pH 7.4 buffer was 7.19 +/‐ 0.10 (n = 11). 4. Acidification of the perfusion medium to pH 7.0 resulted in 0.3 pH unit fall in intrarenal pH. This fall in total intrarenal pH is insufficient to explain the fall in 2‐oxoglutarate concentration observed if the glutamate‐dehydrogenase‐equilibrium model is invoked. 5. The line‐width of the NMR signal is compatible either with heterogeneity of intra‐renal pH or the existence of a pH gradient between cytosol and mitochondria, or both.

The hyperglycemia-induced in vivo metabolic changes produced in subcutaneous murine RIF-1 tumors, grown on female C3H/Anf mice, were examined with 31P surface-coil NMR. Serum glucose levels were elevated 4-fold by bolus intraperitoneal injection of 0.3 ml of an aqueous 50% glucose solution. Tumor pH was calculated from the chemical shift of Pi and relative phosphocreatine and ATP concentrations were determined by Simpson's rule integration of the peak areas. Tumor pH decreased by ca. 0.45 unit over 2 hr while phosphocreatine concentrations decreased by ca. 50% over the same time period (n = 9). Initial tumor pH correlated inversely with the initial peak intensity ratio of Pi:ATP (r = -0.77). In a significant number of tumors (n = 4), two pH populations were observed. In these tumors, one population was unaffected by hyperglycemia and the other showed a decrease in pH. In the other tumors (n = 5), the pH distribution broadened as the pH decreased. In these tumors, the observed decreased in phosphocreatine concentration correlated with that calculated from the effect of measured tumor pH on the intracellular creatine kinase equilibrium (n = 18, r = 0.91). This correlation and consideration of the Pi distribution in the tumor suggest that the pH measured by 31P NMR is weighted heavily by intracellular pH for the RIF-1 tumor. The presence of two distinct tumor pH populations or a broadened pH distribution likely reflects variations in tumor microcellular environment. Control experiments showed negligible changes in tumor pH and high energy phosphate concentrations after bolus intraperitoneal injection of 0.3 ml of isotonic saline. In addition, negligible changes in leg muscle pH and high energy phosphate concentrations were observed after glucose injection into mice with or without tumors. These results indicate that hyperglycemia induced by intraperitoneal glucose injection is effective in lowering the tumor pH of the murine RIF-1 tumor.

High-field 13C surface coil nuclear magnetic resonance has been employed to investigate glucose and glycogen metabolism in rat liver in vivo. Natural abundance and isotopically enriched proton-decoupled 13C NMR experiments were conducted at 90.56 MHz on a standard commercial spectrometer utilizing a laboratory-built high-sensitivity double-resonance coaxial coil probe. At variance with a previous preliminary report, natural abundance spectra of the liver in vivo from a rat fed ad libitum reveal resonances of substantial intensity from hepatic glycogen with approximately 10 min of signal averaging. The response of hepatic glycogen levels to an intravenous injection of the hormone glucagon was continuously monitored through the glycogen C-1 carbon resonance intensity; this revealed an average 60% depletion of hepatic glycogen stores in vivo within approximately 1 h. In a complementary study utilizing fasted rats, 100 mg of D-[1-13C]glucose (90% enriched) was administered via a peripheral vein injection and continuously monitored by 13C NMR with 3-min time resolution as it was incorporated into hepatic glycogen. The C-1 carbon resonances of hepatic glucose and glycogen are well-resolved in vivo enabling the time course for the relative change in concentration for both metabolites to be established simultaneously. The 13C label incorporated into the glycogen pool reaches a steady-state level in approximately 40 min.

An in situ and in vivosurface coil 13C NMR study was performed to study hepatic glycogen synthesis from [3-13C]alanine and [1-13C]glucose administered by intraduodenal infusion in 18-h fasted male Sprague-Dawley rats. Combined, equimolar amounts of alanine and glucose were given. Hepatic appearance and disappearance of substrate and concurrent glycogen synthesis was followed over 150 min, with 5-min time resolution. Active glycogen synthesis from glucose via the direct (glucose→glycogen) and indirect (glucose→lactate→glycogen) pathways and from alanine via gluconeogenesis was observed. The indirect pathway of glycogen synthesis from [1-13C]glucose accounted for 30% (±6 S.E.) of total glycogen formed from labeled glucose. This estimate does not take into account dilution of label in the hepatic oxaloacetate pool and is, therefore, somewhat uncertain. Hepatic levels of [3-13C]alanine achieved were significantly lower than levels of [1-13C]glucose in the liver, and the period of active glycogen synthesis from [3-13C]alanine was longer than from glucose. However, the overall pseudo-first-order rate constant during the period of active glycogen synthesis from [3-13C]alanine (0.075 min−1± 0.026 S.E.) was almost 3 times that from [1-13C]glucose via the direct pathway (0.025 min−1± 0.005 S.E.). The most likely reason for the small rate constant governing direct glycogen formation from duodenally administered glucose compared to that from duodenally administered alanine is a low level of glucose phosphorylating capacity in the liver.

Abstract.: A solid state magnetic resonance imaging technique is used to measure true three-dimensional mineral density of synthetic hydroxyapatite phantoms and specimens of bone ex vivo. The phosphorus-31 free induction decay at 2.0 T magnetic field strength is sampled following application of a short, hard radiofrequency excitation pulse in the presence of a fixed amplitude magnetic field gradient. Multiple gradient directions covering the unit sphere are used in an efficient spherical polar to Cartesian interpolation and Fourier transform projection reconstruction scheme to image the three-dimensional distribution of phosphorus within the specimen. Using 3–6 Gauss/cm magnetic field gradients, a spatial resolution of 0.2 cm over a field of view of 10 cm is achieved in an imaging time of 20–35 minutes. Comparison of solid state magnetic resonance imaging with dual energy X-ray absorptiometry (DXA), gravimetric analysis, and chemical analysis of calcium and phosphorus demonstrates good quantitative accuracy. Direct measurement of bone mineral by solid state magnetic resonance opens up the possibility of imaging variations in mineral composition as well as density. Advantages of the solid state magnetic resonance technique include avoidance of ionizing radiation; direct measurement of a constituent of the mineral without reliance on assumptions about, or models of, tissue composition; the absence of shielding, beam hardening, or multiple scattering artifacts; and its three-dimensional character. Disadvantages include longer measurement times and lower spatial resolution than DXA and computed tomography, and the inability to scan large areas of the body in a single measurement, although spatial resolution is sufficient to resolve cortical from trabecular bone for the purpose of measuring bone mineral density.

During electrorefining of spent nuclear fuels, fission products, and actinides accumulate in the LiCl−KCl electrolyte salt. From the standpoint of high-level waste minimization, it is advantageous to remove these from the salt rather that discarding the salt after these have built up to certain concentrations. Laboratory experiments have shown that zeolite A has the desirable properties for selective removal of fission products by the ion-exchange process. An analytical model has been developed to assess the performance of the ion-exchange column and to scale up the data to engineering size equipment. The model employs empirically determined exchange factors to represent the equilibrium chemistry of the exchange process. The exchange kinetics is modeled by the Nernst−Planck equation applied to multicomponent diffusion in the zeolite pellet. The self-diffusion coefficients, derived from batch kinetic data, are substantially different for monovalent, divalent, and trivalent fission products. The method of characteristics is used to solve the transport equations for the salt phase. The computed breakthrough of the fission products through the experimental zeolite column is in partial but not complete agreement with the measured concentration profiles.

Severe bloodstream-borne infection--i.e., sepsis--and the resulting multiorgan failure are now the most common cause of death in many intensive care units. One of the most fundamentally important and controversial issues concerning the pathophysiology of sepsis is the role of intracellular free calcium concentration ([Ca2+]i) in this disorder. Because of the critical role of calcium as an intracellular second messenger and as a potential cellular toxin, resolution of this issue is crucial. Using 19F NMR spectroscopy and the calcium indicator 5,5'-difluoro-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetate we demonstrate in the intact perfused organ, the rat thoracic aorta, that [Ca2+]i in aortic smooth muscle is increased > 2-fold during sepsis. Furthermore, we determined that sodium dantrolene, a drug that decreases release of calcium from the sarcoplasmic reticulum and that is lifesaving in malignant hyperthermia (a disorder due to increased [Ca2+]i), is able to reduce the elevated [Ca2+]i in sepsis to control values when added in vitro or when given in vivo to the animal. These results suggest that an increase in [Ca2+]i is an early event in sepsis and that increased [Ca2+]i may be responsible for, or contribute to, cellular injury. Dantrolene may offer a therapeutic strategy in the treatment of sepsis.

beta-Very-low-density lipoproteins (beta-VLDL)O were conjugated to the 19F-containing residualizing label, NN-dilactitol-3,5-bis(trifluoromethyl)benzylamine (DLBA), to determine whether the metabolism of this lipoprotein fraction could be characterized in vivo with n.m.r. spectroscopy. Solution state 19F high-resolution n.m.r. spectroscopy of DLBA-beta-VLDL, containing either intact apoproteins or selectively enzymically digested products, demonstrated that the extent of degradation could be distinguished by differences in spin-spin relaxation times (T2 times). DLBA-beta-VLDL was injected intravenously into rabbits, and accumulation of 19F in hepatic tissue was quantified non-invasively by n.m.r. spectroscopy 5 and 30 h after injection. In addition to quantifying the accumulation of DLBA-beta-VLDL in hepatic tissue, a marked decrease (approx. 100 Hz) in the linewidth of 19F resonance from labelled lipoproteins was observed at 30 h compared with the 5 h interval in continuously monitored animals. The change in linewidth was consistent with a decrease in molecular size that occurred during protein degradation, resulting in increased T2 times. To demonstrate that T2 times can be used as an index to quantify apoprotein degradation in vivo, relaxation measurements were performed on livers excised 20 h after injection of DLBA-beta-VLDL into rabbits. Two molecular motional fractions were revealed by relaxation profiles representing either an intact or an extensively degraded form of apoprotein. The amplitudes of each component were compared with results from trichloroacetic acid precipitation of liver homogenates acquired from rabbits 20 h after injection of beta-VLDL labelled with the radioiodinated analogue of DLBA, dilactitol-125I-tyramine. The amount of degraded apoprotein determined by n.m.r. spectroscopy and acid precipitation was 68.6 +/- 7.0% and 58.7 +/- 7.5% (n = 4) respectively. The results of this study demonstrate that 19F n.m.r. spectroscopy can be used to define the temporal characteristics of the hepatic metabolism of lipoproteins in vivo by quantifying both the tissue-specific accumulation and extent of apoprotein degradation. The methodology developed offers promise for the non-invasive, sequential and longitudinal evaluation of lipoprotein metabolism in vivo.

Protein residualizing labels facilitate localization of tissue sites of protein catabolism and the quantification of protein accumulation because of their prolonged intracellular retention of protein accumulation because of their prolonged intracellular retention times. Radioiodinated residualizing labels have been used to define the metabolism of a wide variety of proteins, but this has necessitated destructive analysis. Here we describe the implementation and validation of a novel 19F-containing residualizing label for protein, NN-dilactitol-3,5-bis(trifluoromethyl)benzylamine (DLBA), that permits the non-invasive assessment of protein accumulation and catabolism by n.m.r. spectroscopy in vivo. DLBA comprises a reporter molecule containing six equivalent 19F atoms. 19F is strongly n.m.r.-active, has 100% natural abundance, and is present in minimal background concentrations in soft tissues. We validated the use of DLBA as a protein-labelling compound by coupling to asialofetuin (ASF), a protein that is recognized exclusively by hepatic tissue via a saturable receptor-mediated process. Coupling of DLBA to ASF by reductive amination had no effect on the physiological receptor-mediated uptake of the protein in rat liver in vivo. The 19F-n.m.r. spectrum of DLBA exhibited a single peak that was subject to a small chemical-shift change and broadening after coupling to ASF. Pronase digestion of DLBA-ASF was performed to simulate intracellular degradation products, and resulted in a narrower set of resonances, with chemical shifts intermediate between those of uncoupled DLBA and DLBA-ASF. Intravenous administration of DLBA-ASF to rats followed by quantification of 19F in homogenates of liver tissue indicated that the half-life of residence time of degradation products from DLBA-ASF in liver was approx. 2 days. This intracellular half-life was comparable with that described for similar residualizing labels that contain radioiodide as a reporter. Similar results for the half-life of retention were obtained non-destructively and non-invasively in situ with the use of a whole-body radio-frequency antenna to acquire sequential spectra over 80 h after intravenous administration of DLBA-ASF. Quantification of these spectra demonstrated an initial accumulation of DLBA-ASF in liver followed by an expected gradual loss of 19F-labelled degradation products. The approach developed offers promise for the sequential and longitudinal characterization of metabolism of specific proteins in individual experimental animals and ultimately in human subjects.

Abundance patterns during 6–7 years and orchid visitation were determined for 51 species of the 57 local euglossine bees. Male bees were counted at 3 chemical attractants presented in the same manner each month. Sites were separated by 75 km but included wet Atlantic forest at 500 m elevation, moist forest at 180 m near Barro Colorado Island, and cloud forest at 900 m near the Pacific ocean. 1. From 15 to 30 euglossine species of 4 genera were active in each month and site; monthly species number and general bee abundance were positively correlated. Many species had 3 annual abundance peaks (range 1–4) and were active throughout the year, but peak annual abundances rarely occurred during late wet or early dry seasons. In contrast, Eufriesea generally were present as adults only 1–2 months in a year. 2. Euglossine populations were exceptionally stable. Species at each site were more stable than any known insect population, and stability and abundance were positively associated. However, year-to-year population stability and the degree of seasonality were not correlated. Among the three sites, the more diverse (species rich) bee assemblages displayed lower stability; these were the wetter and upland sites. 3. The most abundant bees visited more orchid species. Eg. and El. each visited and average of 4 orchid species (range 0–13); Ex. and Ef. visited 0–3. Stable populations did not visit more or fewer orchid species than did unstable populations. 4. Less than 68% of species at each site visited orchid flowers; less than a few dozen of the 100–800 bees counted in a day carried orchid pollinaria. Over 20% of the euglossine species never were seen with pollinaria at any site and probably seldom visit orchids in central Panama. 5. Most bee species visited 1 or no fragrance orchids in a given habitat. Orchids tended to utilize common pollinators that seldom included more than 1 species, and they utilized stable or unstable, seasonal or aseasonal bees. However, the most stable and abundant bee, Eg. imperialis, rarely pollinated orchids; fewer than 10 of ca. 20000 bees carried pollinaria. 6. Orchids may interact primarily with discrete seasonal bee population peaks-probably the emerging adults. Although specialized orchid preferences are implicated for species that visit few or no local orchids but pollinate other species and carry pollinaria in other areas, euglossine bees do not need orchids to survive or reproduce.

Abstract: Electrorefining uranium in a molten salt bath is the key step in the electrometallurgical treatment of spent nuclear fuels. The versatility of the electrometallurgical treatment suggests its use for a variety of spent fuel types, as well as for nonnuclear metal-separation applications.

The use of deuterium oxide (2H2O) is proposed as a freely diffusible nuclear magnetic resonance (NMR) blood flow and tissue perfusion tracer of potential clinical utility. Deuterium is a stable, nonradiative isotope commercially available as 2H2O at enrichment levels of essentially 100%--i.e., 110 molar equivalent deuterium. This high concentration, together with the short relaxation time of the spin 1 (quadrupole) deuterium nuclide, provides substantial sensitivity for NMR spectroscopy. As a result, when 2H2O is administered in a bolus fashion to a specific tissue or organ in vivo, the deuterium NMR intensity time course can be analyzed, using mathematical models developed by others for radiolabeled tracers, to measure the rate of blood flow and tissue perfusion. Such an application is demonstrated herein at a static magnetic field of 8.5 tesla. Using single-compartment flow modeling, hepatic blood flow and tissue perfusion in fasted (18 hr) male Sprague-Dawley rats was determined to be 61 +/- 17 (mean +/- SD) ml/100 g per min (n = 5).

High-resolution 31P nuclear magnetic resonance spectra at 73.83 MHz are reported for rat heart in vivo. In live rats, it was possible to observe the cardiac content of ATP, phosphocreatine, and Pi. Only a small amount of whole-blood 2,3-diphosphoglycerate was observed in the spectra, precluding the possibility that blood phosphate compounds were masking the spectra of cardiac phosphate compounds. The 31P nuclear magnetic resonance spectra of in vivo and perfused rat hearts were similar and support the utilization of the perfused rat heart as a model system for studying high-energy phosphate metabolism of the heart in vivo. The dynamic flux of high-energy phosphate compounds was investigated by subjecting the rat to respiratory arrest. In this experiment, the heart followed the classic metabolic pattern known to occur during cardiac arrest; phosphocreatine and then ATP decreased in concentration while Pi increased in concentration. The 31P nuclear magnetic resonance analysis of rat heart in vivo is demonstrated to be a practical and feasible method for studying cardiac high-energy phosphate metabolism.

Effects of peripheral venous injection of glucagon and insulin on [1-13C]glucose incorporation into hepatic glycogen of rats were studied by 13C NMR in vivo. Each animal was given a continuous somatostatin infusion and a 100-mg intravenous injection of [1-13C] glucose in NMR experiments or unlabeled glucose in parallel experiments for determination of serum glucose. Insulin administration caused serum glucose to fall below basal levels and accelerated the loss of hepatic [1-13C]glucose; these effects were counteracted by the addition of glucagon. Glucagon administration alone did not affect serum glucose or hepatic [1-13C] glucose but caused the loss of [1-13C]glucose from glycogen and inhibited [1-13C]glucose incorporation into glycogen. Insulin did not alter [1-13C]glucose incorporation into glycogen when given alone or in combination with glucagon. The data are consistent with a model in which liver glycogen synthesis increases linearly with hepatic glucose concentration above a threshold glucose concentration. Insulin did not alter the rate constant or the threshold for synthesis.

Abstract The object of this retrospective study was to determine the sites of abdominal aortic bifurcation and inferior vena cava confluence in relation to age and sex. The study group comprised 180 subjects (90 males and 90 females) divided into 9 groups by age (in decades). The positions of the aortic bifurcation and the inferior vena cava confluence were evaluated by CT, and linear regression models were fitted to the data. The positions of the aortic bifurcation and venous confluence showed a highly significant downward shift with increasing age (p=0.0001). The shift was more pronounced in women. The mean site of the aortic bifurcation for the whole group was at lower L4 (range, upper L3 to upper S1); in males, it was at upper L4 (range, upper L3 to upper L5), and in females at lower L4 (range, upper L3 to upper S1). The mean site of the venous confluence for the whole group was at disc L4-L5 (range, lower L3 to upper S1); in males, it was at disc L4-L5 (range, upper L4 to disc L5-S1), and in females at disc L4-L5 (range, lower L3 to upper S1). Thus, the aorta and the inferior vena cava can extend as low as the level of S1. These data are of relevance in laparoscopic procedures, especially in laparoscopic lumbar discectomy.

Depletion of CD4+ cells using anti-CD4 monoclonal antibodies leads to allograft tolerance. Here we show that anti-CD4-mediated tolerance to pancreatic islets of Langerhans transplanted from an A/J (IEk) donor to a diabetic C57B1/6 (B6) (IE-) recipient occurs in the absence of clonal deletion of the potentially IE-reactive V beta 11+ T cells. Instead, a state of clonal anergy is induced in both the CD4+V beta 11+ and CD8+V beta 11+ T cell subsets. This clonal anergy can be partially overcome in vitro by the addition of recombinant interleukin 2.

Infertility noted in two congenic strains of mice was associated with abortion, metritis, and stillbirths. Pasteurella pneumotropica was recovered from affected uterine tracts of ATL mice. Tetracycline treatment for 6 weeks was instituted in an attempt to minimize Pasteurella-associated infertility. Nine ATL mice and 12 ATH mice were examined 4 and 12 months after cessation of tetracycline therapy, respectively. Histopathologically, all animals appeared normal. P. pneumotropica was recovered from six of nine prepuces of both ATH and ATL mice; similarly, two of three vaginal culture of the ATL mice yielded this bacterium. Pasteurella ureae was isolated from three of eight reproductive tracts of the female ATH mice, Negative indole and ornithine decarboxylase reactions and positive acid production for mannitol differentiated P. ureae from P. pneumotropica. This report represents the first confirmed isolation of P. urea from animals. Biochemical profiles of atypical strains of P. pneumotropica isolated from animals are discussed and compared with those of P. ureae. The need for careful biochemical characterization of all Pasteurella isolated from rodents is emphasized.

The dopamine (DA) hypothesis of schizophrenia proposes hyperactivity of the mesocorticolimbic DA system, originating within the A10 DA cells of the ventral tegmental area (VTA), as a pathophysiological mechanism. Thus, reduction of activity in this system, including that produced by putative "autoreceptor-selective" DA agonists, may be of clinical utility. The present studies compared the ability of eight D2 DA receptor agonists to inhibit the firing of rat A10 DA neurons after i.v. administration. Both N-n-propyl-N-phenylethyl-p(3-hydorxyphenyl)ethylamine hydrochloride (RU 24213) and 2-amino-6-allyl-5,6,7,8-tetrahydro-4H-thiazolo-[4,5-d]- azepine dihydrochloride (B-HT 920) were potent, high-efficacy agonists which completely inhibited the firing of A10 DA cells. The putative autoreceptor-selective DA agonists 3-(4-(4-phenyl-1,2,3,6-tetrahydropyridyl-(1)-butyl)-indole hydrochloride (EMD 23,448) and (+)-3-(3-hydroxy-phenyl)-N-n-propylpiperidine [(+)-3-PPP] were considerably weaker than RU 24213 and B-HT 920, but also exhibited "full" efficacy (i.e., they completely suppressed cell firing). The putative autoreceptor agonist preclamol [(-)-3-PPP] and its trans-fused congener (-)-HW 165 were weak partial agonists that failed to completely inhibit A10 DA cells. The new putative autoreceptor agonist N-[(8-alpha)-2-chloro-6-methylergoline-8-yl]-2,3]dimethylopropa namide (SDZ 208-911) was also a weak partial agonist that exhibited partial antagonist effects (reversed inhibition produced by the D2 agonist quinpirole), whereas its structural analog N-[(8-alpha)-2-chloro-6-methylergoline-8-yl]-2,2-dimethylopropa namide (SDZ 208-912) was nearly inactive as an agonist, but was an effective antagonist.(ABSTRACT TRUNCATED AT 250 WORDS)

Investigations of heat transfer to supercritical pressure fluids have been going on for some time, and correlations have been developed for both free and forced-convection conditions. In these investigations, unpredictable heat transfer performance has sometimes been observed when the pseudocritical temperature of the fluid is between the temperature of the bulk fluid and that of the heated surface. The unusual performance has been attributed to many causes, but one for which more evidence is being collected is that of a pseudofilm-boiling process similar to film boiling which occurs at subcritical pressures. This paper, which is an extension of work reported earlier on forced-convection heat transfer to supercritical pressure water, presents experimental evidence which suggests that a pseudofilm-boiling phenomenon can occur in smooth-bore tubes. During the period from 1963–1966, tubes with ID’s from 0.37 to 0.96 in. were tested at pressures from 3300–6000 psia and at heat fluxes and mass velocities in the range of interest in steam-generator design. The effects of heat flux, mass velocity, tube diameter, pressure, and bulk fluid temperature on both the occurrence and characteristics of pseudofilm boiling are discussed. Results of a second series of tests conducted in 1967, which show that ribbed tubes suppress pseudofilm boiling at supercritical pressure much like they do film boiling at subcritical pressures, are also discussed.

New heat transfer and draft loss data are presented for one extended surface geometry which was investigated in five staggered and one in-line bank arrangements. In these tests, 1-7/8-inch OD tubes with 1-in. long elliptical studs were investigated. The tube banks, which were in a clean condition, were either 8 or 10 rows deep and covered ratios of transverse spacing to diameter (ST/D) from 2.1 to 3.2 and longitudinal spacing to diameter (ST/D) from 1.7 to 2.4. Heat transfer was from air to the tube banks, with Reynolds numbers ranging from 13,700 to 46,400. Correlations with suitable arrangement factors are shown to adequately describe the performance of the surface investigated. The relative heat transfer, draft loss performance of all arrangements is shown to depend upon the basis of comparison used.

One approach to flat sensor design is to use a lenslet array to form multiple subimages of a scene and then combine the subimages to recover a fully sampled image by using a superresolution algorithm. Previously, superresolution image assembly has been based on information derived from the observed scene. For lenslet arrays, we propose a new scene-independent approach based only on known imager properties in which relative subimage shifts are accurately estimated with a calibration procedure using point source imaging. Thus, the relative resolution enhancement provided by the scene-independent superresolution algorithm is impervious to changes in subimage content, contrast, sharpness, and noise.
