We present an efficient method for modeling the single and double ionization dynamics of the H2molecule in ultrashort, intense laser fields. This method is based on a semianalytical approach to calculate the time-dependent single and double molecular ionization rates and on a numerical approach to describe the vibrational motion that takes place in the intermediate molecular ion H2+. This model allows for the prediction of the single and double ionization probabilities of the H2molecule to be made over a wide range of frequencies and laser intensities with limited computational time while providing a realistic estimate of the energy of the products of the dissociative ionization and of the Coulomb explosion of the H2molecule. The effect of vibrational dynamics on ionization yields and proton kinetic energy release spectra is demonstrated and, in the case of the latter, is discussed in terms of basic strong-field molecular fragmentation mechanisms.

Recessive and de novo mutations in the TRIOgene are associated with intellectual deficiency (ID), autism spectrum disorder (ASD) and developmental epileptic encephalopathies (DEE). TRIO is a dual guanine nucleotide exchange factor (GEF) that activates Rac1, Cdc42 and RhoA. Trio has been extensively studied in excitatory neurons, and has recently been found to regulate the switch from tangential to radial migration in GABAergic interneurons (INs) through GEFD1-Rac1-dependent SDF1α/CXCR4 signaling. Given the central role of Rho-GTPases during neuronal migration and the implication of IN pathologies in ASD and DEE, we investigated the relative roles of both Trio’s GEF domains in regulating the dynamics of INs tangential migration. In Trio–/–mice, we observed reduced numbers of tangentially migrating INs, with intact progenitor proliferation. Further, we noted increased growth cone collapse in developing INs, suggesting altered cytoskeleton dynamics. To bypass the embryonic mortality of Trio–/–mice, we generated Dlx5/6Cre;Trioc/cconditional mutant mice (TriocKO), which develop spontaneous seizures and behavioral deficits reminiscent of ASD and ID. These phenotypes are associated with reduced cortical IN density and functional cortical inhibition. Mechanistically, this reduction of cortical IN numbers reflects a premature switch to radial migration, with an aberrant early entry in the cortical plate, as well as major deficits in cytoskeletal dynamics, including enhanced leading neurite branching and slower nucleokinesis reflecting reduced actin filament condensation and turnover as well as a loss of response to the motogenic effect of EphA4/ephrin A2 reverse signaling. Further, we show that both Trio GEFD1 and GEFD2 domains are required for proper IN migration, with a dominant role of the RhoA-activating GEFD2 domain. Altogether, our data show a critical role of the DEE/ASD-associated Triogene in the establishment of cortical inhibition and the requirement of both GEF domains in regulating IN migration dynamics.

The capability to reach ultracold atomic temperatures in compact instruments has recently been extended into space1,2. Ultracold temperatures amplify quantum effects, whereas free fall allows further cooling and longer interactions time with gravity—the final force without a quantum description. On Earth, these devices have produced macroscopic quantum phenomena such as Bose–Einstein condensates (BECs), superfluidity, and strongly interacting quantum gases3. Terrestrial quantum sensors interfering the superposition of two ultracold atomic isotopes have tested the universality of free fall (UFF), a core tenet of Einstein’s classical gravitational theory, at the 10−12level4. In space, cooling the elements needed to explore the rich physics of strong interactions or perform quantum tests of the UFF has remained elusive. Here, using upgraded hardware of the multiuser Cold Atom Lab (CAL) instrument aboard the International Space Station (ISS), we report, to our knowledge, the first simultaneous production of a dual-species BEC in space (formed from 87Rb and 41K), observation of interspecies interactions, as well as the production of 39K ultracold gases. Operating a single laser at a ‘magic wavelength’ at which Rabi rates of simultaneously applied Bragg pulses are equal, we have further achieved the first spaceborne demonstration of simultaneous atom interferometry with two atomic species (87Rb and 41K). These results are an important step towards quantum tests of UFF in space and will allow scientists to investigate aspects of few-body physics, quantum chemistry and fundamental physics in new regimes without the perturbing asymmetry of gravity.

Traditional Chinese medicine (TCM), a major holistic approach to health and disease, has been used for a long time globally (>100 years) and in China (>1000 years) for various medical conditions. However, the contrast between TCM and modern medicine is remarkable, and the underlying mechanisms of TCM remain unclear. In modern medicine, immunology is a complex, holistic defense system against pathogens, preserving homeostasis and influencing disease development. Notably, TCM and immunology share core concepts, such as holism, defense mechanisms, equilibrium, balance, and circadian rhythms. This review focuses on the similarities and distinctions between these two disciplines and sheds light on ancient TCM theories by leveraging modern immunological parameters. In addition, we emphasize the potential of advanced “omics” research methods to unravel TCM's mechanisms, providing compelling biological evidence for its therapeutic principles and effects using immunological data.

The COVID-19 pandemic had a monumental impact on the inflammatory bowel disease (IBD) community. At the beginning of the pandemic, knowledge on the effect of SARS-CoV-2 on IBD was lacking, especially in those with medication-suppressed immune systems. Throughout the pandemic, scientific literature exponentially expanded, resulting in clinical guidance and vaccine recommendations for individuals with IBD. Crohn’s and Colitis Canada established the COVID-19 and IBD Taskforce to process and communicate rapidly transforming knowledge into guidance for individuals with IBD and their caregivers, healthcare providers, and policy makers. Recommendations at the onset of the pandemic were based on conjecture from experience of prior viruses, with a precautionary principle in mind. We now know that the risk of acquiring COVID-19 in those with IBD is the same as the general population. As with healthy populations, advanced age and comorbidities increase the risk for severe COVID-19. Individuals with IBD who are actively flaring and/or who require high doses of prednisone are susceptible to severe COVID-19 outcomes. Consequently, sustaining maintenance therapies (e.g., biologics) is recommended. A three-dose mRNA COVID-19 vaccine regimen in those with IBD produces a robust antibody response with a similar adverse event profile as the general population. Breakthrough infections following vaccine have been observed, particularly as the virus continues to evolve, which supports receiving a bivalent vaccine booster. Limited data exist on the impact of IBD and its therapies on long-term outcomes following COVID-19. Ongoing research is necessary to address new concerns manifesting in those with IBD throughout the evolving pandemic.In April 2021, Crohn’s and Colitis Canada’s COVID-19 and IBD Taskforce produced the 2021 Impact of COVID-19 & Inflammatory Bowel Disease in Canada. This report summarized the key learnings of the impact of COVID-19 on the IBD community at the one-year anniversary since the COVID-19 pandemic was declared by the World Health Organization. During the first year of the pandemic, and before the widespread dissemination of COVID-19 vaccines, the body of the scientific literature was synthesized and communicated to the IBD community through Crohn’s and Colitis Canada’s COVID-19 and IBD Taskforce using online resources and a webinar series. The scientific literature from that time period suggested that those with IBD shared a similar risk of becoming infected by SARS-CoV-2 or experiencing severe outcomes as the general population with the primary exception being those flaring and/or requiring high doses of oral systemic corticosteroids. During the first year, lockdowns and shielding populations believed to be at high risk, like the IBD community, reduced the risk of transmission. However, mental health distress was prevalent due to isolation and uncertainty of risks, which was exacerbated in immunocompromised individuals with IBD. Major breakthroughs in novel SARS-CoV-2 vaccines and antiviral therapeutics, as well as adaptation to models of healthcare delivery (e.g., virtual care) offered hope that the second year of the pandemic would realize the easing of restrictions.This article reviews what we know about COVID-19 in 2023. It is an update to the 2021 Impact of COVID-19 and Inflammatory Bowel (IBD) Disease report. IBD is a disease that causes the intestines to be inflamed. Some medicine used to treat IBD can make a person more prone to infection. COVID-19 is an infection by the SARS-CoV-2 virus. So, it was once thought that people with IBD were at greater risk of COVID-19. We now know that people with IBD have the roughly same risk of COVID-19 as people without IBD. This is unless the person is on high dose steroids (>20 mg/day) or has an active IBD flare. The Crohn’s and Colitis Canada COVID-19 and IBD Taskforce assembled quickly at the beginning of the pandemic. They provided expert advise to the IBD community about COVID-19 and IBD and will keep doing so. We recommend people with IBD get a bivalent booster vaccine against COVID-19.

The therapeutic landscape for inflammatory bowel disease (IBD) has changed considerably over the past two decades, owing to the development and widespread penetration of targeted therapies, including biologics and small molecules. While some conventional treatments continue to have an important role in the management of IBD, treatment of IBD is increasingly moving towards targeted therapies given their greater efficacy and safety in comparison to conventional agents. Early introduction of these therapies—particularly in persons with Crohn’s disease—combining targeted therapies with traditional anti-metabolite immunomodulators and targeting objective markers of disease activity (in addition to symptoms), have been shown to improve health outcomes and will be increasingly adopted over time. The substantially increased costs associated with targeted therapies has led to a ballooning of healthcare expenditure to treat IBD over the past 15 years. The introduction of less expensive biosimilar anti-tumour necrosis factor therapies may bend this cost curve downwards, potentially allowing for more widespread access to these medications. Newer therapies targeting different inflammatory pathways and complementary and alternative therapies (including novel diets) will continue to shape the IBD treatment landscape. More precise use of a growing number of targeted therapies in the right individuals at the right time will help minimize the development of expensive and disabling complications, which has the potential to further reduce costs and improve outcomes.This article reviews studies on treatment for inflammatory bowel disease (IBD). IBD is a disease that causes the bowels to become inflamed. Many treatment options exist for people with IBD in Canada. Some of these options are drugs, surgery and diet. Many drugs used to treat IBD are very expensive. New biosimilar drugs may lower costs and increase how available these drugs are. It is very important to make sure that the right drug for the right person at the right time is found. New studies must seek to understand what the right treatments are for each person. We have come a long way in treating IBD over the last 60 or so years. However, people with IBD may still need surgery at some point. New drugs being tested offer hope that more ways to treat IBD will soon be available. We hope that these new drugs will prove safe an effective for people with IBD and reduce complications of disease.

American Indian and Alaska Natives (AI/ANs) are disproportionately impacted by gestational diabetes mellitus (GDM), subsequent type 2 diabetes, and food insecurity. It is prudent to decrease risk of GDM prior to pregnancy to decrease the intergenerational cycle of diabetes in AI/AN communities. The purpose of this project is to describe and examine food insecurity, healthy eating self-efficacy, and healthy eating behaviors among AI/AN females (12–24 years old) as related to GDM risk reduction. Methods included: secondary analysis of healthy eating self-efficacy and behaviors, and household-level food insecurity measures from an randomized controlled trial that tested the effect of engagement in a GDM risk reduction educational intervention on knowledge, behavior, and self-efficacy for GDM risk reduction from baseline to 3-month follow-up. Participants were AI/AN daughters (12–24 years old) and their mothers (N= 149 dyads). Researchers found that more than one-third (38.1%) reported food insecurity. At baseline food insecurity was associated with higher levels of eating vegetables and fruit for the full sample (p= .045) and cohabitating dyads (p= .002). By 3 months healthy eating self-efficacy (p= .048) and limiting snacking between meals (p= .031) improved more in the control group than the intervention group only for cohabitating dyads. For the full sample, the intervention group had increases in times eating vegetables (p= .022) and fruit (p= .015), whereas the control group had declines. In the full sample, food insecurity did not moderate the group by time interaction for self-efficacy for healthy eating (p≥ .05) but did moderate the group by time interaction for times drinking soda (p= .004) and days eating breakfast (p= .013). For cohabitating dyads, food insecurity did moderate self-efficacy for eating 3 meals a day (p= .024) and days eating breakfast (p= .012). These results suggest food insecurity is an important factor regarding the efficacy of interventions designed to reduce GDM risk and offer unique insight on “upstream causes” of GDM health disparities among AI/AN communities.Limited access to healthful food (e.g., food insecurity) is a social determinant of health that impacts gestational diabetes risk and related disparities for American Indian and Alaska Native females.American Indian and Alaska Native (AI/AN) women are disproportionately impacted by gestational diabetes mellitus (GDM). Living in a food insecure household during preconception and pregnancy may increase risk of greater weight gain. In this paper, we examined the relationship between food insecurity with healthy eating self-efficacy and behaviors among AI/AN adolescents and young adults (AYAs) (n= 149) through secondary analysis of an existing randomized controlled trial dataset. This study offers unique insight regarding “upstream causes” of GDM health disparities among AI/AN communities. Food security had some moderating effects on individual eating behaviors. Additionally, both healthy eating behavior and self-efficacy for healthy eating improved more among the AYA who experienced food insecurity at baseline. Given the intergenerational implications of GDM, it is prudent that the healthcare sector works with AI/AN communities to support healthful eating behaviors and environments to decrease GDM disparities.

Purpose: The purpose of the study was to describe, compare, and examine associations at baseline of reproductive health awareness, knowledge, health beliefs, communication and behaviors related to gestational diabetes (GDM) and GDM risk reduction in a vulnerable population of both American Indian/Alaska Native (AIAN) adolescent girls and their mothers.Methods: Descriptive/comparative/correlational analyses examined multitribal baseline data on 149 mother-daughter (M-D) dyads (N = 298; daughter age = 12-24 years) enrolled in a longitudinal study to adapt and evaluate a culturally relevant diabetes preconception counseling (PC) program (Stopping-GDM). The associations between GDM risk reduction awareness, knowledge, health beliefs, and behaviors (eg, daughters’ eating, physical activity, reproductive-health [RH] choices/planning, M-D communication, daughters’ discussions on PC) were examined. Data collected online from 5 national sites.Results: Many M-D lacked awareness/knowledge of GDM and risk reduction. Both M-D were unaware of the girl’s risk for GDM. Mothers’ knowledge and beliefs on GDM prevention/RH were significantly higher than daughters. Younger daughters had greater self-efficacy healthy living. Overall sample reported low to moderate scores for both M-D communication and daughters’ GDM and RH risk-reduction behaviors.Conclusions: Knowledge, communication, and behaviors to prevent GDM were low in AIAN M-D, especially daughters. More than daughters, mothers perceive greater risk of GDM for daughters. Early culturally responsive dyadic PC programs could help decrease risk of developing GDM. Implications for M-D communication is compelling.

The apparent failure of global health security to prevent or prepare for the COVID-19 pandemic has highlighted the need for closer cooperation between human, animal (domestic and wildlife), and environmental health sectors. However, the many institutions, processes, regulatory frameworks, and legal instruments with direct and indirect roles in the global governance of One Health have led to a fragmented, global, multilateral health security architecture. We explore four challenges: first, the sectoral, professional, and institutional silos and tensions existing between human, animal, and environmental health; second, the challenge that the international legal system, state sovereignty, and existing legal instruments pose for the governance of One Health; third, the power dynamics and asymmetry in power between countries represented in multilateral institutions and their impact on priority setting; and finally, the current financing mechanisms that predominantly focus on response to crises, and the chronic underinvestment for epidemic and emergency prevention, mitigation, and preparedness activities. We illustrate the global and regional dimensions to these four challenges and how they relate to national needs and priorities through three case studies on compulsory licensing, the governance of water resources in the Lake Chad Basin, and the desert locust infestation in east Africa. Finally, we propose 12 recommendations for the global community to address these challenges. Despite its broad and holistic agenda, One Health continues to be dominated by human and domestic animal health experts. Substantial efforts should be made to address the social–ecological drivers of health emergencies including outbreaks of emerging, re-emerging, and endemic infectious diseases. These drivers include climate change, biodiversity loss, and land-use change, and therefore require effective and enforceable legislation, investment, capacity building, and integration of other sectors and professionals beyond health.

In this article, we address the problem of simultaneous localization and mapping (SLAM)-centric maritime infrastructure inspection [using unmanned surface vehicles (USVs)] via novel approaches in tightly-coupled, graph-based DVL/IMU fusion and decoupled mapping. As our first contribution, we formalize the preintegration of linear velocity measurements, obtained by a Doppler velocity log (DVL), in combination with angular velocity measurements, obtained by an inertial measurement unit (IMU), as binary factors encoding relative position. To evaluate state estimation improvements imparted by DVL/IMU fusion, we implement our proposed factor within a state-of-the-art, graph-based lidar-visual-inertial (LVI) SLAM system as our second contribution. Accuracy and robustness improvements are demonstrated in simulation by comparing maximum a posteriori pose estimates with and without DVL/IMU fusion against ground truth poses. As our third contribution, we propose a map generation framework for downstream inspection applications decoupled from SLAM. In our framework, volumetric data (captured by sonar, lidar, etc.) is transformed into a common world coordinate frame using extrinsic calibrations and SLAM pose estimates as input. Our framework operates over the complete set of raw volumetric data, whereas SLAM systems (both online and offline) typically operate over a subset of down-sampled volumetric data. To address the processing of additional volumetric data, we present innovations in refined pose correction and staged filtering for user-controlled denoising. We experimentally evaluate our map generation framework against the LVI SLAM system adopted for this study using real-world data and demonstrate improvements to map quality metrics important to inspection.

Purpose: Music accompanies all phases of our lives, and when we reach old age, music becomes a direct symbol of nostalgia. Autobiographical memories are essential to an individual’s sense of identity, continuity, and meaning. But some pathologies, such as dementia, can interrupt the memory storage process. Music can help recall and evoke memories and can be used in alternative treatments for dementia. Methods: This work aims to propose an architecture for a music recommendation system capable of recommending music according to musical genre, with the aim of helping music therapists in therapies aimed at elderly people without dementia or in an initial state of dementia. The public music database Emotify, composed of 400 songs labeled by 1595 participants in 7975 sessions, was used. The songs were organized into two channels of 20 s windows with 5 s overlapping. These windows were represented by 34 time and frequency characteristics. Classifiers based on support vector machines and Random Forests were investigated. Results: The most suitable architecture in this experimental study was the Random Forest with 300 trees, with an accuracy of 72% ± 4%, AUC of 0.89 ± 0.03, sensitivity of 0.68 ± 0.08, and specificity of 0.88 ± 0.03. Conclusions: This exploratory study showed that it is possible to build recommendation systems to support music therapy capable of supporting music therapists based on the automatic classification of songs according to the most appropriate musical genre for the patient, according to previous evaluations carried out by the therapist.
