The common grape (Vitis viniferaL.) has been cultivated for thousands of years. Nowadays, it is cultivated using a variety of tillage practices that affect the structure of the soil microbial communities and thus the health of the vine. The aim of this study was to explore and compare the effects of tillage (shallow tillage with bare soil) and no-tillage (perennial grass cover) practices on soil physical and chemical properties and soil bacterial community diversities in a small catchment. Soil samples were taken in July and October 2020 at different slope positions of two vineyards exposed to erosion. The two sampling sites were separated by the agricultural inter-row management type: tilled and no-tilled slopes. The taxonomic diversity of bacterial communities was determined using 16S rRNA gene-based amplicon sequencing method on Illumina MiSeq platform. Based on the examined soil properties, the sampling areas were separated from each other according to the positions of the upper and lower slopes and the sampling times. Both the tilled and no-tilled soil samples were dominated by sequences assigned to phyla Pseudomonadota, Acidobacteriota, Bacteroidota, Verrucomicrobiota, Actinobacteriota, and Gemmatimonadota. The results showed that tillage had no significant effect compared to the no-tilled samples in the studied area. Water runoff and seasonally changed soil physical and chemical properties affected mainly the bacterial community structures.

The taxonomic and metabolic diversity of prokaryotes and their adaptability to extreme environmental parameters have allowed extremophiles to find their optimal living conditions under extreme conditions for one or more environmental parameters. Natural habitats abundant in extremophilic microorganisms are relatively rare in Hungary. Nevertheless, alkaliphiles and halophiles can flourish in shallow alkaline lakes (soda pans) and saline (solonetz) soils, where extreme weather conditions favor the development of unique bacterial communities. In addition, the hot springs and thermal wells that supply spas and thermal baths and provide water for energy use are suitable colonization sites for thermophiles and hyperthermophiles. Polyextremophiles, adapted to multiple extreme circumstances, can be found in the aphotic, nutrient-poor and radioactive hypogenic caves of the Buda Thermal Karst, among others. The present article reviews the organization, taxonomic composition, and potential role of different extremophilic bacterial communities in local biogeochemical cycles, based on the most recent studies on extremophiles in Hungary.

Background: Leishmaniasis is a neglected tropical disease caused by protozoa of the genus Leishmania. Current treatments are restricted to a small number of drugs that display both severe side effects and a potential for parasites to develop resistance. A new N-(3,4-methylenedioxyphenyl)-N'- (2-phenethyl) thiourea compound (thiourea 1) has shown promising in vitro activity against Leishmania amazonensis with an IC50 of 54.14 μM for promastigotes and an IC50 of 70 μM for amastigotes. Objective: To develop a formulation of thiourea 1 as an oral treatment for leishmaniasis, it was incorporated into Nanoparticles (NPs), a proven approach to provide long-acting drug delivery systems. Methods: Poly (D,L-Lactic-co-Glycolic Acid) (PLGA) polymeric NPs containing thiourea 1 were obtained through a nanoprecipitation methodology associated with solvent evaporation. The NPs containing thiourea 1 were characterized for Encapsulation Efficiency (EE%), reaction yield (% w/w), surface charge, particle size and morphology by Transmission Electron Microscopy (TEM). Results: NPs with thiourea 1 showed an improved in vitro leishmanicidal activity with a reduction in its cytotoxicity against macrophages (CC50>100 μg/mL) while preserving its IC50 against intracellular amastigotes (1.46 ± 0.09 μg/mL). This represents a parasite Selectivity Index (SI) of 68.49, which is a marked advancement from the reference drug pentamidine (SI = 30.14). Conclusion: The results suggest that the incorporation into NPs potentiated the therapeutic effect of thiourea 1, most likely by improving the selective delivery of the drug to the phagocytic cells that are targeted for infection by L. amazonensis. This work reinforces the importance of nanotechnology in the acquisition of new therapeutic alternatives for oral treatments.

Acetylcholine plays an essential role in the regulation of detrusor muscle contractions and antimuscarinics are widely used in the management of overactive bladder syndrome. However, several adverse effects limit their application and patients' compliance. Thus, this study aims to further analyze the signal transduction of M2and M3receptors in murine urinary bladder to eventually find more specific therapeutic targets. Experiments were performed on adult, male, wild-type, M2-, M3-, M2/M3- or Gαq/11-knockout (KO) and pertussis toxin (PTX)-treated mice. Contraction force and RhoA activity were measured in the urinary bladder smooth muscle (UBSM). Our results indicate that carbamoylcholine (CCh)-induced contractions are associated with increased activity of RhoA and are reduced in the presence of the Rho-associated kinase (ROCK) inhibitor Y-27632 in UBSM. The CCh-evoked contractile responses and RhoA activation were markedly reduced in detrusor strips lacking either M2or M3 receptors and abolished in M2/M3-KO mice. Inhibiting Gαi-coupled signaling by PTX-treatment shifted the concentration-response curve of CCh to the right and diminished RhoA activation. CCh-induced contractile responses were markedly decreased in Gαq/11-KO mice, however, RhoA activation was unaffected. In conclusion, cholinergic detrusor contraction and RhoA activation are mediated by both M2and M3receptors. Furthermore, whereas both Gαiand Gαq/11proteins mediate UBSM contraction, the activation at the RhoA-ROCK pathway appears to be linked specifically to Gαi. These findings may aid the identification of more specific therapeutic targets for bladder dysfunctions.

There is an increasing number of evidence showing analgesic properties of the kynurenic acid (KYNA), and also some studies demonstrate that kynurenine might interact with the opioid system. Therefore in this study, for the first time we investigated the direct binding affinity of KYNA and its structural analog KYNA-1 towards mu, kappa and delta opioid receptor in competition binding experiments applying opioid receptor specific radioligands. The binding affinity measurements were performed in Chinese hamster ovary cell lines overexpressing the corresponding opioid receptor (mu and kappa opioid receptor were rat, delta opioid receptor were mouse sequence). Additionally we also examined the chronic effect of these compounds on mu, kappa and delta opioid receptor and also nociceptin peptide receptor mediated G-protein activity in [35S]GTPγS binding assays performed in mouse cortex and striatum membranes. Our results showed that KYNA and KYNA-1 had no affinity towards any of the three classic opioid receptors. On the other hand the compounds significantly decreased opioid and nociceptin receptor mediated G-protein activity or in some cases enhanced the potency of the activating ligand. Moreover, the alterations were receptor and brain region specific. Accordingly, we conclude that KYNA and KYNA-1 do not interact directly with the opioid receptors, but more likely alter the receptor functions intracellularly.

The aims of this study were to synthesize 14-O-Methylmorphine-6-O-sulfate (14-O-MeM6SU) and examine its opioid properties (potency, affinity, efficacy) in receptor ligand binding and isolated tissues (mouse vas deferens, MVD and rat vas deferens, RVD bioassays). The results were then compared to the parent compounds morphine-6-O-sulfate (M6SU) and morphine, as well as the µ- opioid receptor (MOR) selective agonist peptide [D-Ala2,N-Me-Phe4,Gly-ol5]enkephalin (DAMGO). An additional objective was to compare the effect of subcutaneously (s.c.) or intracerebroventricularly (i.c.v.) administered 14-O-MeM6SU, M6SU and morphine in thermal nociception, rat tail-flick (RTF) test. In MVD, the EC50 (nM) value was 4.38 for 14-O-MeM6SU, 102.81 for M6SU, 346.63 for morphine and 238.47 for DAMGO. The effect of 14-O-MeM6SU and DAMGO was antagonized by naloxone (NAL) with Ke value 1-2.00 nM. The Emax values () were 99.10, 36.87, 42.51 and 96.99 for 14-O-MeM6SU, M6SU, morphine and DAMGO, respectively. In RVD 14-O-MeM6SU and DAMGO but not M6SU or morphine showed agonist activity. In binding experiments the affinity of 14-OMeM6SU, M6SU, morphine and DAMGO for MOR was 1.12, 11.48, 4.37 and 3.24 nM, respectively. The selectivity of 14-O-MeM6SU was κ/μ 269 and δ/μ 9. In G-protein activation experiments, 14-O-MeM6SU and DAMGO showed higher Emax values than M6SU or morphine. S.c. or i.c.v-injected 14-O-MeM6SU, M6SU and morphine produced a dose and time-dependent increase in RTF response latency. 14-O-MeM6SU was the most potent. Our results showed that introduction of 14-O-Me in M6SU increased the binding affinity, agonist potency, and most importantly, the intrinsic efficacy (Emax).
