Principal component analysis (PCA) has been used to study the pathogenesis of diseases. To enhance the interpretability of classical PCA, various improved PCA methods have been proposed to date. Among these, a typical method is the so-called sparse PCA, which focuses on seeking sparse loadings. However, the performance of these methods is still far from satisfactory due to their limitation of using unsupervised learning methods; moreover, the class ambiguity within the sample is high. To overcome this problem, this paper developed a new PCA method, which is named the supervised discriminative sparse PCA (SDSPCA). The main innovation of this method is the incorporation of discriminative information and sparsity into the PCA model. Specifically, in contrast to the traditional sparse PCA, which imposes sparsity on the loadings, here, sparse components are obtained to represent the data. Furthermore, via the linear transformation, the sparse components approximate the given label information. On the one hand, sparse components improve interpretability over the traditional PCA, while on the other hand, they are have discriminative abilities suitable for classification purposes. A simple algorithm is developed, and its convergence proof is provided. SDSPCA has been applied to the common-characteristic gene selection and tumor classification on multiview biological data. The sparsity and classification performance of SDSPCA are empirically verified via abundant, reasonable, and effective experiments, and the obtained results demonstrate that SDSPCA outperforms other state-of-the-art methods. The presented algorithms are testified in several publicly open single cell RNA-seq data sets. 3β-hydroxy-Δ5-steroid dehydrogenases (3βHSDs) are supposed to be involved in 5β -cardenolide biosynthesis. Here, a novel 3βHSD (Dl3βHSD2) was isolated from Digitalis lanata shoot cultures and expressed in E. coli. Recombinant Dl 3βHSD1 and Dl 3βHSD2 shared 70% amino acid identity, reduced various 3-oxopregnanes and oxidised 3-hydroxypregnanes, but only rDl 3βHSD2 converted small ketones and secondary alcohols efficiently. To explain these differences in substrate specificity, we established homology models using borneol dehydrogenase of Salvia rosmarinus (6zyz) as the template. Hydrophobicity and amino acid residues in the binding pocket may explain the difference in enzyme activities and substrate preferences. Compared to Dl3βHSD1, Dl3βHSD2 is weakly expressed in D. lanata shoots. High constitutive expression of Dl3βHSDs was realised by Agrobacterium -mediated transfer of Dl3βHSD genes fused to the CaMV-35S promotor into the genome of D. lanata wild type shoot cultures. Transformed shoots (35S: Dl3βHSD1 and 35S: Dl3βHSD2) accumulated less cardenolides than controls. The levels of reduced glutathione (GSH), which is known to inhibit cardenolide formation, were higher in the 35S: Dl3βHSD1 lines than in the controls. In the 35S: Dl3βHSD1 lines cardenolide levels were restored after adding of the substrate pregnane-3,20-dione in combination with buthionine-sulfoximine (BSO), an inhibitor of GSH formation. RNAi-mediated knockdown of the Dl3βHSD1 yielded several shoot culture lines with strongly reduced cardenolide levels. In these lines, cardenolide biosynthesis was fully restored after addition of the downstream precursor pregnan-3β-ol-20-one, whereas upstream precursors such as progesterone had no effect, indicating that no shunt pathway could overcome the Dl3βHSD1 knockdown. Aiming at the problem that the current image encryption algorithms generally use dissipative chaos, which leads to the algorithms being vulnerable to reconstruction attack, and the coding rules of image encryption algorithms using biological coding are too simple, this paper proposes an image encryption algorithm based on conservative hyperchaotic and biological gene algorithms. Firstly, a new conservative hyperchaotic system is proposed, which can resist reconstruction attacks, has a larger key space, and can generate pseudorandom sequences with strong randomicity. Secondly, SHA-512 is used to calculate the hash value of plaintext image, and the initial key is generated through the key generation algorithm we devised to improve the sensitivity of the algorithm to the key and plaintext. Finally, new dynamic coding rules are designed to enrich the biological coding rules. In addition, dynamic gene uniform crossover algorithm and dynamic gene mutation algorithm are devised, and pseudorandom sequences dynamically control gene operation, further enriching coding rules and expanding key space, improving the security of the algorithm.
