Under sjökaptensutbildningen ska teoretiska studier varvas med fartygsförlagdutbildning. Detta görs för att utöver skolans teori få känna på verkligheten och att ipraktiken ta till sig de kunskaper som behövs för att efter examen vara vakthavandebefäl ombord. Under den fartygsförlagda praktiken ska eleven bli lotsad av enhandledare som guidar och hjälper för att tiden ombord skall bli så effektiv och givandesom möjligt. Vi ville undersöka hur elever upplever handledarskapet ombord med fokuspå om det eventuellt är några skillnader beroende på vilken praktikperiod eleven befannsig i. Detta gjordes med hjälp av en kvalitativ metod genom fokusgrupper med syfte attfå utförliga och ingående svar. Det framkom vissa skillnader mellan den tredjepraktikperioden kontra de andra två sett till vilka arbetsuppgifter man fick göra samt hurbesättningen såg på eleven i form av förväntade kunskaper och vilket fritt ansvar somgavs eleven. Dock blev det tydligt att en god relation till sin handledare, oavsett vilkenpraktikperiod eleven befann sig i, var av väsentlighet för en lyckad praktik. Eventuellinverkan i handledarskapet beroende på kön och ålder hos handledaren var också någotvi ville undersöka. Kvinnliga handledare visade sig vara något mer sympatiska men denstörsta skillnaden fanns beroende på vilken ålder handledaren hade. En yngrehandledare ansågs generellt som mer uppskattad då denne hade ett större engagemangoch en större vilja att lära ut., During their time at the “captain’s program”, students alternate theoretical studies with practical experience aboard ships. This combination aims to give them a taste of real-world situations alongside classroom learning, helping them develop the skills they need to be in charge on a ship after graduation. During their time at sea, students areguided by mentors who help them make the most of their experience onboard. Wewanted to investigate how students feel about this mentorship, particularly if it differsdepending on which stage of practical training, they're in. We used a qualitative methodusing focus groups to get detailed and thorough answers. We noticed some differencesbetween the third training period and the earlier ones, like the tasks students were givenand how they were viewed by the crew. But what mattered most was having a goodrelationship with their mentor, no matter at what stage they were in their teaching. Wealso investigated whether the mentor's gender and age had any impact on their teaching.Female mentors were seen as a bit more sympathetic, but the biggest difference wasage. Younger mentors were generally more appreciated because they were moreengaged and eager to teach.

3. Novel Anti-CRISPR-Assisted CRISPR Biosensor for Exclusive Detection of Single-Stranded DNA (ssDNA).

- Abstract
Nucleic acid analysis plays an important role in disease diagnosis and treatment. The discovery of CRISPR technology has provided novel and versatile approaches to the detection of nucleic acids. However, the most widely used CRISPR-Cas12a detection platforms lack the capability to distinguish single-stranded DNA (ssDNA) from double-stranded DNA (dsDNA). To overcome this limitation, we first employed an anti-CRISPR protein (AcrVA1) to develop a novel CRISPR biosensor to detect ssDNA exclusively. In this sensing strategy, AcrVA1 cut CRISPR guide RNA (crRNA) to inhibit the cleavage activity of the CRISPR-Cas12a system. Only ssDNA has the ability to recruit the cleaved crRNA fragment to recover the detection ability of the CRISPR-Cas12 biosensor, but dsDNA cannot accomplish this. By measuring the recovered cleavage activity of the CRISPR-Cas12a biosensor, our developed AcrVA1-assisted CRISPR biosensor is capable of distinguishing ssDNA from dsDNA, providing a simple and reliable method for the detection of ssDNA. Furthermore, we demonstrated our developed AcrVA1-assisted CRISPR biosensor to monitor the enzymatic activity of helicase and screen its inhibitors.

Choosing a degree and courses is an important part of every student's education, providing the foundation for future success. Many students have difficulty deciding on a profession when they enter college, while some already know what they want to do. Due to the large number of courses offered, some students find themselves switching majors. Therefore, all students must understand what they truly want to do with their lives, which will guide them in choosing the proper career path. Their choices will greatly impact how they live for the rest of their lives. The researchers aim to discover factors influencing students' decisions to take BSHM as a course. They also aim to assist students in shaping their perceptions of this course and inspire them to choose it among other courses. This study utilized the descriptive method of research to investigate and collect information about the motivational factors influencing senior high school students in choosing BSHM. The researchers used Google Forms to distribute survey questionnaires to fifty (50) specifically selected respondents at Bestlink College of the Philippines to collect data. According to the survey's findings, the majority of respondents (30%) were from the home economics strand, were 21 years old on average (60%) and were equally split between men and women (25% each). Most respondents claimed that their economic status belonged to the middle class (54%). Most respondents agreed that "Percentage of employment is high after graduation" is one of their factors in choosing BSHM. The respondents strongly agreed that one of their perceived problems in choosing a BSHM degree is needing more motivation. However, they also agreed on the advantages of choosing BSHM as a field of study, such as global exposure and boosting self-esteem. Selective Mutism is an uncommon and debilitating anxiety disorder. Because of its scarcity, finding a succinct, implementable intervention within the school setting is often difficult for school-based professionals. School may be the best place to implement an intervention as often the symptoms of Selective Mutism are most prevalent in the school setting. This project examines the Selective Mutism literature to create an intervention guide that is both usable and manipulatable. School Psychologists, Speech Language Pathologists, general and special education teachers, and other school-based professionals can use this guide to piece together an intervention that is relevant to the student and the school setting. Various templates and forms are provided for use.

The role of teacher as a guide is very significant in teaching, especially to equip students to know God through Christian education. Teachers guide the students to understand knowledge, attitudes, skills, and spirituality based on Biblical principles. But in reality, teachers do not teach the Bible principles in learning, teachers do not guide the students, and have lack of knowledge of the Bible. Teachers should have understood the significance of guiding students based on the Bible. Hence, the purpose of writing this article is to present the significant role of teachers as a guide through the theological study based on Ephesians 4:11-16 for the context of Christian education with literature study as the research method. Teachers must guide students based on the truth of God's Word, through imitating Jesus as the Great Shepherd. Christian teachers’ role as guide do not only stop at knowledge in academic learning, but to be an impact for students to be a part of service to God and to build others up as members of the body of Christ. Christian teachers must guide students to achieve, namely: the unity of the faith in Christ as God and Savior in true knowledge, thorough maturity, and growth in accordance with the fullness of Christ and to ensure to be not tossed around by the waves of false teaching that ignore The Lord. Byggsektorn är en bransch med höga produktionskostnader, som till stor del är baserat på resursslöseri. Ett av dessa slöserier är lager, som är nödvändiga inom de flesta organisationer men som ofta kan reduceras i storlek och förbättra sin struktur. Detta kan uppnås med hjälp av filosofin Lean, vars principer både gynnar företaget samt leder till en minskad miljöpåverkan (M. Kidwell 2006; J. Maxwell, 1996). Detta genom att eliminera slöserier och effektivisera processer, vilket också kan tillämpas inom lagerhantering. Projektet har genomförts i samarbete med Skanska och NCC, som båda upplevt problem med strukturen på deras lager. Det övergripande syftet med detta projekt är att ta fram och testa en tydlig metod som kan användas vid strukturering inom en verksamhet. Däremot har inte en fullständig implementering av Lean och 5S skett, utan en lättare version som enkelt kan bidra till struktur. Målet med detta projekt är framförallt att den framtagna metoden ska kunna användas av verksamheter som behöver ordning och reda. Ytterligare ett mål är att Skanska och NCC ska få förbättringsförslag om hur de kan förbättra sina lager. Dessa lösningar samt den framtagna metoden ska också bidra till de tre hållbarhetsmålen. För att kunna nå dessa mål har projektet framförallt använt workshops, men också observationer och intervjuer. Företagen hade liknande resultat, däremot är de anpassade efter de olika verksamheterna. Ett nuläge, önskat läge och förbättringsförslag utifrån 5S kunde då tas fram. Nuläget visade dålig ordning och reda på lagren hos både Skanska och NCC. Även det önskade läget var liknande då de båda ville ha markerade platser för verktyg och material, en tydlig standard och att alla hjälps åt och engagerar varandra. Detta resulterade i att förbättringsförslag kunde tas fram. - Abstract
CRISPR-Cas9 genome editing has promising therapeutic potential for genetic diseases and cancers, but safety could be a concern. Here we use whole genomic analysis by 10x linked-read sequencing and optical genome mapping to interrogate the genome integrity after editing and in comparison to four parental cell lines. In addition to the previously reported large structural variants at on-target sites, we identify heretofore unexpected large chromosomal deletions (91.2 and 136 Kb) at atypical non-homologous off-target sites without sequence similarity to the sgRNA in two edited lines. The observed large structural variants induced by CRISPR-Cas9 editing in dividing cells may result in pathogenic consequences and thus limit the usefulness of the CRISPR-Cas9 editing system for disease modeling and gene therapy. In this work, our whole genomic analysis may provide a valuable strategy to ensure genome integrity after genomic editing to minimize the risk of unintended effects in research and clinical applications.

14. Modeling Brain Tumors In Vivo Using Electroporation-Based Delivery of Plasmid DNA Representing Patient Mutation Signatures.

- Abstract
Tumor models are critical for the preclinical testing of brain tumors in terms of exploring new, more efficacious treatments. With significant interest in immunotherapy, it is even more critical to have a consistent, clinically pertinent, immunocompetent mouse model to examine the tumor and immune cell populations in the brain and their response to treatment. While most preclinical models utilize orthotopic transplantation of established tumor cell lines, the modeling system presented here allows for a "personalized" representation of patient-specific tumor mutations in a gradual, yet effective development from DNA constructs inserted into dividing neural precursor cells (NPCs) in vivo. DNA constructs feature the mosaic analysis with the dual-recombinase-mediated cassette exchange (MADR) method, allowing for single-copy, somatic mutagenesis of driver mutations. Using newborn mouse pups between birth and 3 days old, NPCs are targeted by taking advantage of these dividing cells lining the lateral ventricles. Microinjection of DNA plasmids (e.g., MADR-derived, transposons, CRISPR-directed sgRNA) into the ventricles is followed by electroporation using paddles that surround the rostral region of the head. Upon electrical stimulation, the DNA is taken up into the dividing cells, with the potential of integrating into the genome. The use of this method has successfully been demonstrated in developing both pediatric and adult brain tumors, including the most common malignant brain tumor, glioblastoma. This article discusses and demonstrates the different steps of developing a brain tumor model using this technique, including the procedure of anesthetizing young mouse pups, to microinjection of the plasmid mix, followed by electroporation. With this autochthonous, immunocompetent mouse model, researchers will have the ability to expand preclinical modeling approaches, in efforts to improve and examine efficacious cancer treatment.

The lack of control or disagreement in the logistic processes creates barriers and disadvantages in the operations of small exporting companies. For this reason, the objective of the researched topic is to propose a traceability guide in the logistic processes of banana exports to demonstrate its incidence in the commercial and financial area and its delivery to the international market. According to its purpose, the research is applied, according to its context it is field research, according to the control of the variables it is experimental, according to its temporal orientation it is longitudinal, and according to its level of generality it studies a specific situation. The theoretical method used is the systemic method, which focuses on the elaboration of the traceability guide, and the empirical method is the study of documents and surveys. The sample is finite because the study is known and is delimited in six small exporting companies of Machala, the same that will help to obtain information to present the results of the current situation of traceability in export operations in small banana exporting companies and thus focus on the development of the guide to prevent the problems that usually occur., La falta de control o desavenencia en los procesos logísticos, crean barreras de desventajas en las operaciones que presentan las pequeñas empresas exportadoras, por tal razón el objetivo del tema investigado es proponer una guía de trazabilidad en los procesos logísticos de la exportación de banano para la demostración de su incidencia en el área comercial y financiera y su envío al mercado internacional. La investigación según su finalidad es aplicada, según su contexto es de campo, según el control de las variables es experimental, según orientación temporal es longitudinal, y según su nivel de generalidad estudia una situación específica. Se ha empleado como método teórico el método sistémico el cual se enfoca en elaborar la guía de trazabilidad, y en el método empírico es el estudio de documentos y encuestas. La muestra es finita por que el estudio es conocido y se delimita en seis pequeñas empresas exportadoras de Machala, las mismas que ayudarán a la obtención de información para presentar los resultados de la situación actual de la trazabilidad en las operaciones de exportación en las pequeñas empresas exportadoras de banano y de esta manera enfocarnos en el desarrollo de la guia que prevengan los inconvenientes que usualmente se presentan.

16. Factors Influencing Professional Identity Construction in Fourth-Year Pharmacy Students

Objectives. To explore the key factors that influence professional identity construction in fourth-year pharmacy students enrolled in a Doctor of Pharmacy program.Methods. A single-site instrumental case study of current fourth-year pharmacy students from the Leslie Dan Faculty of Pharmacy, University of Toronto, was used. Thirteen students participated in semistructured interviews. Poststructural social identity theories were used to analyze the data and identify themes that influence identity construction in pharmacy students.Results. Data analysis identified five overarching themes that influence pharmacy student professional identity construction: path to pharmacy, curriculum, environment, preceptors, and patient interactions. The Leslie Dan Faculty of Pharmacy curriculum prioritized the health care provider identity, which influenced the students desire to "become" clinicians. Based on their internalized health care provider identity, they rejected preceptors and practice environments that negatively impacted their ability to embody this identity.Conclusion. The findings of this study suggest that pharmacy students align themselves strongly with health care provider identities at the cost of other potentially relevant identities. Pharmacy education programs may benefit from curricular reforms that incorporate and legitimize multiple pharmacist identities to ensure a strong pharmacy workforce for the future.

- Abstract
BACKGROUND: People with a low socioeconomic position (SEP) are less likely to benefit from eHealth interventions, exacerbating social health inequalities. Professionals developing eHealth interventions for this group face numerous challenges. A comprehensive guide to support these professionals in their work could mitigate these inequalities. OBJECTIVE: We aimed to develop a web-based guide to support professionals in the development, adaptation, evaluation, and implementation of eHealth interventions for people with a low SEP. METHODS: This study consisted of 2 phases. The first phase involved a secondary analysis of 2 previous qualitative and quantitative studies. In this phase, we synthesized insights from the previous studies to develop the guide's content and information structure. In the second phase, we used a participatory design process. This process included iterative development and evaluation of the guide's design with 11 professionals who had experience with both eHealth and the target group. We used test versions (prototypes) and think-aloud testing combined with semistructured interviews and a questionnaire to identify design requirements and develop and adapt the guide accordingly. RESULTS: The secondary analysis resulted in a framework of recommendations for developing the guide, which was categorized under 5 themes: development, reach, adherence, evaluation, and implementation. The participatory design process resulted in 16 requirements on system, content, and service aspects for the design of the guide. For the system category, the guide was required to have an open navigation strategy leading to more specific information and short pages with visual elements. Content requirements included providing comprehensible information, scientific evidence, a user perspective, information on practical applications, and a personal and informal tone of voice. Army Corps of Engineers" NAVIGATION AND ECOSYSTEM SUSTAINABILITY PROGRAM ENVIRONMENTAL DESIGN TOOL KIT JULY 2023 EXECUTIVE SUMMARY The Upper Mississippi River System has a robust ecosystem restoration history, with successful projects planned, designed, and constructed through a variety of programs, including the Navigation and Ecosystem Sustainability Program (NESP), the Upper Mississippi River Restoration (UMRR) Program, and Continuing Authority Projects. These ecosystem restoration projects were completed in coordination with numerous state, federal and non-profit resources agencies to meet ecosystem goals and objectives. The U.S. Army Corps of Engineers, Rock Island District, St. Paul District, and St. Louis District have worked together to gather information from a variety of sources to develop developed key design elements to assist teams in efficiently identifying methods and measures to address ecosystem needs during the planning level of design. The purpose of the Environmental Design Tool Kits is to provide a consistent format of design considerations, design details, and parametric cost development to support plan formulation and eventual selection of the Recommended Plan. The information contained herein include features or measures that are common to ecosystem restoration projects completed within the Upper Mississippi River System. These features include shoreline stabilization, dredging, river training structures and secondary channel modifications, floodplain restoration, and islands. The financial risks that the companies of the secondary sector that entered into the dissolution and liquidation processes of zone 7 went through, are multiple, they had to be corrected at the time to avoid the dissolution and liquidation, for the development of the investigation a comparative descriptive analysis is carried out In order to determine the evolution of the economic and financial indicators, the study period covers the years 2015 to 2020, as preliminary information, the degree theses of the ladies Tamara Alvarado, Martha Montaño and Carmen Celi were reviewed, who build a database of the companies that entered into dissolution and liquidation processes in zone 7, the financial economic situations that the companies were going through are: over-indebtedness, illiquidity, low profitability, negative working capital in various periods, minimum economic returns, and in others periods without returns on the assets that the companies own and the patrimony of their shareholders, in addition they do not have a planning for the management of possible financial risks., Los riesgos financieros que atravesaron las compañías del sector secundario que entraron en procesos de disolución y liquidación de la zona 7, son múltiples debieron corregir en su momento para evitar a la disolución y liquidación, para el desarrollo de la investigación se realiza un análisis descriptivo comparativo para determinar la evolución de los indicadores económico financueros, el periodo de estudio comprende los años 2015 al 2020, como información preliminar se revisó las tesis de grado de las señoritas Tamara Alvarado, Martha Montaño y Carmen Celi, quienes construyen una base de datos de las compañías que entraron en procesos de disolución y liquidación en la zona 7, las situaciones económicas financieras por las cuales a travesaban las compañías son: sobre endeudamiento, iliquidez, baja rentabilidad, capital de trabajo negativo en varios periodos, rendimientos económicos mínimos, y en otros periodos sin rendimientos sobre los activos que poseen las compañías y el patrimonio de sus accionista, además no cuentan con una planificación para el manejo de posibles riesgos financieros.

The research is basic, its objective is to propose a methodological guide to promote the strengthening of the cultural identity of secondary education students in Tumbes; quantitative approach, descriptive-propositive type, non-experimental design, cross section. A questionnaire was applied to a sample of 130 high school students, simple random probabilistic sampling was used, the results of the Cronbach's alpha inference of the information collection instrument was 0.894. The results of the study indicate that the cultural identity of the respondents is at the SCARE level, with an average of seven points obtained from the four dimensions., La investigación es básica, su objetivo es proponer una guía metodológica para promover el fortalecimiento de la identidad cultural de los estudiantes de educación secundaria de Tumbes; de enfoque cuantitativo, tipo descriptivo- propositivo, diseño no Experimental, corte transversal. Se aplicó un cuestionario a una muestra a 130 estudiantes del nivel secundaria, se utilizó el muestreo probabilístico aleatorio simple, los resultados de la inferencia del alfa de Cronbach del intrumento de recojo de información fue de 0.894. Los resultados del estudio indican que la identidad cultural de los encuestados se encuentra en el nivel ESCASO, con un promedio de siete puntos obtenido de las cuatro dimensiones.

- Abstract
The Delegation of Well-being in the state of Veracruz, committed to the population health and well-being of the program “The School is Ours”, undertook the task of designing a guide that facilitates this aim which complies with the aforementioned article 75 of the General Law of Health Education. Objective: to describe the dietary and nutritional reality of the beneficiary children of the program “The School is Ours”. Methodology: applied, descriptive, qualitative-quantitative, not experimental and transversal research. Results: the focus groups reported malnutrition; most consumed regional foods as boiled beans with nopales, epazote, cilantro and onion, fried pumpkins with shrimp head, pasta soup with cheese, among others; soft drinks considered most consumed beverages; boiled foods the main method of cooking; and insufficient physical activity. Conclusions: malnutrition, overweight and obesity are the main problems of this population; although their breakfast consists of healthy foods, they have a high consumption of ultra-processed foods. Keywords: nutrition, well-being, children., La Delegación de Bienestar en Veracruz, se dio a la tarea de diseñar, una guía que facilite proporcionar una alimentación saludable a los escolares del Programa “La Escuela es Nuestra” (LEEN), y cumplir con el artículo 75 de la Ley General de Educación en Materia de Salud. Objetivo: Elaborar una Guía Alimentaria que contribuya a proporcionar una alimentación saludable a la población escolar en el marco de la Estrategia Nacional de Alimentación Saludable, Justa y Sostenible. Metodología: investigación aplicada, descriptiva, cuali-cuantitativa, y transversal. Resultados: Se identificaron problemas de malnutrición, alimentos regionales de mayor consumo, técnicas de preparación, y el consumo de Productos Ultraprocesados (PUP); lo cual permitió elaborar una guía alimentaria con 10 recomendaciones. Conclusiones: La guía alimentaria basada en la cultura alimentaria de la población, la disponibilidad regional de alimentos y la problemática nutricional, facilitará proporcionar una alimentación saludable a los escolares del Programa LEEN.

It is set to identify, compare and explore common elements in successful games. In order to validate the results, it materialized the knowledge extracted into three tangible materials: a design guide, an infographic and a one-page RPG made from scratch using the previous two, and put it to the test through different methods of playtesting.

This guide opens with an introduction followed by an overview of Altmetric and the Altmetric Attention Score, Altmetrics and Responsible Research Assessment, Output Types Tracked by Altmetric, and the Altmetric Sources of Attention, which include: News and Mainstream Media, Social Media (X (formerly Twitter), Facebook, Reddit, and historical data from Google+, Pinterest, LinkedIn, and Sina Weibo); Patents, Peer Review, Syllabi (historical data only), Multimedia, Public Policy Documents, Wikipedia, Research Highlights, Reference Managers, and Blogs; finally, there is a conclusion, a list of related resources and readings, two appendices, and references. Through the case of commercial timber production in Nepal's community forests, we uncover and explain how effective anti-corruption and harvest regulation have produced a kind of 'allowed' corruption that promotes forest conservation. An ethnographic study in four community forests and in-depth interviews of nearly 200 actors along the Sal (Shorea robusta) timber commodity chain showed that all actors participated in a highly organised form of collusive corruption. Anti-corruption officials call this practice "corruption without illegality" because it does not involve unauthorised harvest in community forests. Instead, it suppresses producer prices through legally required but rigged timber auctions that generate windfall profits, which sawmill owners share with upstream actors to ensure a steady supply of raw logs. Local-level timber brokers connect community forest user groups to sawmill owners. They also operate as stealth conveyers of unofficial payments to forestry officials and other upstream actors because they can camouflage such cash flows as transaction costs. Anti-corruption authorities enforce formal timber harvesting rules, which deters forestry officials from getting involved in overharvesting schemes. However, these same rules, plus some legal posturing, allow forestry officials to extract rents from legally harvested logs at minimum risk. This is hardly a coincidence because it enables the central administration to regulate difficult-to-control field-level forestry officials' behaviour without curtailing their access to informal incomes. Forest user groups lose out, but they could increase their timber income substantially by exercising their powers to decide whether or not to harvest timber. Elevating timber auction floor prices through state intervention is also feasible.

Family engagement in schools has been looked at and researched for many years. It has been shown that increased family engagement in schools leads to higher student achievement (Epstein and Voorhis, 2010). Family engagement literature explains engagement can be increased, the benefits, common barriers, and implications for schools (Dikkers, 2013). It continues to be researched to look at multiple ways engagement can be effective for students, families, and communities. Increased family engagement in education positively affects student performance and overall experience (Epstein, 1987). As such, it is important for schools to have a program in place to encourage and enable family involvement. A planning guide can be used to provide a step-by-step guide on creating educational and fun events for families. These events can be tailored to the needs of the school and community, as well as the interests of the families. This planning guide can provide the foundation for successful family engagement initiatives. Through these initiatives, schools can create a greater connection between the students, their families, and the school. This connection can help foster a better overall educational experience for everyone involved.

The aim of this thesis is to elucidate possible factors which influence the acceptability of assistance dogs, and make comparisons between Japan and the UK. Two main questions were asked. Firstly, what are the differences in attitudes towards and relationships with animals between Japan and the UK? Secondly, what are the differences in the current state of the assistance dog movement between the two countries? In the first study, the relationship between childhood experiences and attitudes towards animals in adulthood was investigated. The second study examined attitudes towards dogs in general. The third study, which investigated knowledge of and attitudes towards assistance dogs suggested that assistance dogs, except guide dogs for the blind, are still not widely used in either country. In the fourth study, guide dog owners were interviewed about their experiences of using a guide dog. Both Japanese and British guide dog owners shared similar problems associated with other people, in terms of prejudice towards people with disabilities and denial of access to facilities such as restaurants. They were also commonly annoyed by people's interference in the dogs' work. The worst issue for British owners seemed to be people who were selfish and inconsiderate to them and their dogs. For Japanese owners, people who criticised them when they disciplined their dogs seemed to be the worst problem. In the final study, interactions between guide dog owners and the general public were observed unobtrusively. The dog attracted people's attention and sometimes promoted social interactions between its owner and strangers both in Japan and the UK. The dogs themselves received more interactions from people in Japan than in the UK.

Summary: In recent years, the building sector has been turning towards intervening in the existing city building stock. In fact, it is generally accepted that the refurbishment of buildings and urban regeneration based on sustainability must form the axis of reformulation of the building sector. At present, achieving sustainable urban development inevitably involves improving existing buildings, thereby preventing the need for city growth, and for the emptying of established neighborhoods. Furthermore, considering the whole life cycle, the great amount of greenhouse emissions derived from the construction sector is well known; thus, in order to reach a decarbonized society, it is important to provide eco-efficient construction materials and solutions, adding the principles of the circular economy and resource efficiency. Therefore, the theme of this Special Issue is the "Decarbonization and Circular Economy in the Sustainable Development and Renovation of Buildings and Neighborhoods" in response to the objectives not only raised in the Horizon 2020 but by all the people who seek a more sustainable world. - Abstract
Understanding microbial gene functions relies on the application of experimental genetics in cultured microorganisms. However, the vast majority of bacteria and archaea remain uncultured, precluding the application of traditional genetic methods to these organisms and their interactions. Here, we characterize and validate a generalizable strategy for editing the genomes of specific organisms in microbial communities. We apply environmental transformation sequencing (ET-seq), in which nontargeted transposon insertions are mapped and quantified following delivery to a microbial community, to identify genetically tractable constituents. Next, DNA-editing all-in-one RNA-guided CRISPR-Cas transposase (DART) systems for targeted DNA insertion into organisms identified as tractable by ET-seq are used to enable organism- and locus-specific genetic manipulation in a community context. Using a combination of ET-seq and DART in soil and infant gut microbiota, we conduct species- and site-specific edits in several bacteria, measure gene fitness in a nonmodel bacterium and enrich targeted species. These tools enable editing of microbial communities for understanding and control.

BackgroundZebrafish have practical features that make them a useful model for higher-throughput tests of gene function using CRISPR/Cas9 editing to create 'knockout' models. In particular, the use of G0 mosaic mutants has potential to increase throughput of functional studies significantly but may suffer from transient effects of introducing Cas9 via microinjection. Further, a large number of computational and empirical tools exist to design CRISPR assays but often produce varied predictions across methods leaving uncertainty in choosing an optimal approach for zebrafish studies.MethodsTo systematically assess accuracy of tool predictions of on- and off-target gene editing, we subjected zebrafish embryos to CRISPR/Cas9 with 50 different guide RNAs (gRNAs) targeting 14 genes. We also investigate potential confounders of G0-based CRISPR screens by assaying control embryos for spurious mutations and altered gene expression.ResultsWe compared our experimental in vivo editing efficiencies in mosaic G0 embryos with those predicted by eight commonly used gRNA design tools and found large discrepancies between methods. Assessing off-target mutations (predicted in silico and in vitro) found that the majority of tested loci had low in vivo frequencies (< 1%). To characterize if commonly used 'mock' CRISPR controls (larvae injected with Cas9 enzyme or mRNA with no gRNA) exhibited spurious molecular features that might exacerbate studies of G0 mosaic CRISPR knockout fish, we generated an RNA-seq dataset of various control larvae at 5 days post fertilization. While we found no evidence of spontaneous somatic mutations of injected larvae, we did identify several hundred differentially-expressed genes with high variability between injection types. Network analyses of shared differentially-expressed genes in the 'mock' injected larvae implicated a number of key regulators of common metabolic pathways, and gene-ontology analysis revealed connections with response to wounding and

[EN] If the incorporation of the gender perspective in the urban fabric requires a clear prior position in favor of the equality of women and men, it also requires instruments or tools that allow it to be effectively incorporated into practice. Urban morphology studies offer a holistic view that tries to encompass the phenomenon of living together, in all its scales and in all its complexity, as well as the tool of the morphological map that tries to offer an ordered way of working, in the search for what makes urban fabrics vital, diverse and close. In this context, the Guide for the incorporation of the gender perspective prepared by Generalitat Valenciana, also tries to provide tools that help to become aware of how complex projecting a space or place is, so that in the future it to be vital, diverse and, therefore, safe. Both strategies could be linked so that the relationship between the parts and the whole can be seen from a gender perspective, what could facilitate the task of both those who plan and those who supervise., [ES] Si la incorporación de la perspectiva de género en el tejido urbano requiere una clara posición previa a favor de la igualdad de mujeres y hombres, también requiere instrumentos o herramientas que permitan incorporarla efectivamente en la práctica. Genome-wide functional genetic screens have been successful in discovering genotype-phenotype relationships and in engineering new phenotypes. While broadly applied in mammalian cell lines and in E. coli, use in non-conventional microorganisms has been limited, in part, due to the inability to accurately design high activity CRISPR guides in such species. Here, we develop an experimental-computational approach to sgRNA design that is specific to an organism of choice, in this case the oleaginous yeast Yarrowia lipolytica. A negative selection screen in the absence of non-homologous end-joining, the dominant DNA repair mechanism, was used to generate single guide RNA (sgRNA) activity profiles for both SpCas9 and LbCas12a. This genome-wide data served as input to a deep learning algorithm, DeepGuide, that is able to accurately predict guide activity. DeepGuide uses unsupervised learning to obtain a compressed representation of the genome, followed by supervised learning to map sgRNA sequence, genomic context, and epigenetic features with guide activity. Experimental validation, both genome-wide and with a subset of selected genes, confirms DeepGuide's ability to accurately predict high activity sgRNAs. DeepGuide provides an organism specific predictor of CRISPR guide activity that with retraining could be applied to other fungal species, prokaryotes, and other non-conventional organisms.

Recruiting endogenous adenosine deaminases using exogenous guide RNAs to edit cellular RNAs is a promising therapeutic strategy, but editing efficiency and durability remain low using current guide RNA designs. In this study, we engineered circular ADAR-recruiting guide RNAs (cadRNAs) to enable more efficient programmable adenosine-to-inosine RNA editing without requiring co-delivery of any exogenous proteins. Using these cadRNAs, we observed robust and durable RNA editing across multiple sites and cell lines, in both untranslated and coding regions of RNAs, and high transcriptome-wide specificity. Additionally, we increased transcript-level specificity for the target adenosine by incorporating interspersed loops in the antisense domains, reducing bystander editing. In vivo delivery of cadRNAs via adeno-associated viruses enabled 53% RNA editing of the mPCSK9 transcript in C57BL/6J mice livers and 12% UAG-to-UGG RNA correction of the amber nonsense mutation in the IDUA-W392X mouse model of mucopolysaccharidosis type I-Hurler syndrome. cadRNAs enable efficient programmable RNA editing in vivo with diverse protein modulation and gene therapeutic applications.

Programmable double-strand DNA breaks (DSBs) can be harnessed for precision genome editing through manipulation of the homology-directed repair (HDR) pathway. However, end-joining repair pathways often outcompete HDR and introduce insertions and deletions of bases (indels) at the DSB site, decreasing precision outcomes. It has been shown that indel sequences for a given DSB site are reproducible and can even be predicted. Here, we report a general strategy (the "double tap" method) to improve HDR-mediated precision genome editing efficiencies that takes advantage of the reproducible nature of indel sequences. The method simply involves the use of multiple gRNAs: a primary gRNA that targets the wild-type genomic sequence, and one or more secondary gRNAs that target the most common indel sequence(s), which in effect provides a "second chance" at HDR-mediated editing. This proof-of-principle study presents the double tap method as a simple yet effective option for enhancing precision editing in mammalian cells.

Homing CRISPR gene drives could aid in curbing the spread of vector-borne diseases and controlling crop pest and invasive species populations due to an inheritance rate that surpasses Mendelian laws. However, this technology suffers from resistance alleles formed when the drive-induced DNA break is repaired by error-prone pathways, which creates mutations that disrupt the gRNA recognition sequence and prevent further gene-drive propagation. Here, we attempt to counteract this by encoding additional gRNAs that target the most commonly generated resistance alleles into the gene drive, allowing a second opportunity at gene-drive conversion. Our presented "double-tap" strategy improved drive efficiency by recycling resistance alleles. The double-tap drive also efficiently spreads in caged populations, outperforming the control drive. Overall, this double-tap strategy can be readily implemented in any CRISPR-based gene drive to improve performance, and similar approaches could benefit other systems suffering from low HDR frequencies, such as mammalian cells or mouse germline transformations.

In bacterial defense and genome editing applications, the CRISPR-associated protein Cas9 searches millions of DNA base pairs to locate a 20-nucleotide, guide RNA-complementary target sequence that abuts a protospacer-adjacent motif (PAM). Target capture requires Cas9 to unwind DNA at candidate sequences using an unknown ATP-independent mechanism. Here we show that Cas9 sharply bends and undertwists DNA on PAM binding, thereby flipping DNA nucleotides out of the duplex and toward the guide RNA for sequence interrogation. Cryogenic-electron microscopy (cryo-EM) structures of Cas9-RNA-DNA complexes trapped at different states of the interrogation pathway, together with solution conformational probing, reveal that global protein rearrangement accompanies formation of an unstacked DNA hinge. Bend-induced base flipping explains how Cas9 'reads' snippets of DNA to locate target sites within a vast excess of nontarget DNA, a process crucial to both bacterial antiviral immunity and genome editing. This mechanism establishes a physical solution to the problem of complementarity-guided DNA search and shows how interrogation speed and local DNA geometry may influence genome editing efficiency.

Often, our common ground and imagination of urban landscapes doesn’t go further than a bench, a lawn and, if lucky, a bin. This project explores how public spaces can be governed and organised by the people around it by empowering them to be in the centre of the planning and being able to make decisions and shape their habitat according to their needs, wishes and imagination. As part of this project a guide was created on how to claim and shape one’s local public space together with their community.

- Abstract
Gene drives are selfish genetic elements that are transmitted to progeny at super-Mendelian (>50%) frequencies. Recently developed CRISPR-Cas9-based gene-drive systems are highly efficient in laboratory settings, offering the potential to reduce the prevalence of vector-borne diseases, crop pests and non-native invasive species. However, concerns have been raised regarding the potential unintended impacts of gene-drive systems. This Review summarizes the phenomenal progress in this field, focusing on optimal design features for full-drive elements (drives with linked Cas9 and guide RNA components) that either suppress target mosquito populations or modify them to prevent pathogen transmission, allelic drives for updating genetic elements, mitigating strategies including trans-complementing split-drives and genetic neutralizing elements, and the adaptation of drive technology to other organisms. These scientific advances, combined with ethical and social considerations, will facilitate the transparent and responsible advancement of these technologies towards field implementation.

As the focus for CRISPR/Cas-edited plants moves from proof-of-concept to real-world applications, precise gene manipulation will increasingly require concurrent multiplex editing for polygenic traits. A common approach for editing across multiple sites is to design one guide RNA (gRNA) per target; however, this complicates construct assembly and increases the possibility of off-target mutations. In this study, we utilized one gRNA to target MYB186, a known positive trichome regulator, as well as its paralogs MYB138 and MYB38 at a consensus site for mutagenesis in hybrid poplar (Populus tremula × P. alba INRA 717-1B4). Unexpected duplications of MYB186 and MYB138 resulted in eight alleles for the three targeted genes in the hybrid poplar. Deep sequencing and polymerase chain reaction analyses confirmed editing across all eight targets in nearly all of the resultant glabrous mutants, ranging from small indels to large genomic dropouts, with no off-target activity detected at four potential sites. This highlights the effectiveness of a single gRNA targeting conserved exonic regions for multiplex editing. Additionally, cuticular wax and whole-leaf analyses showed a complete absence of triterpenes in the trichomeless mutants, hinting at a previously undescribed role for the nonglandular trichomes of poplar.

- Abstract
The recent development of prime editing (PE) genome engineering technologies has the potential to significantly simplify the generation of human pluripotent stem cell (hPSC)-based disease models. PE is a multicomponent editing system that uses a Cas9-nickase fused to a reverse transcriptase (nCas9-RT) and an extended PE guide RNA (pegRNA). Once reverse transcribed, the pegRNA extension functions as a repair template to introduce precise designer mutations at the target site. Here, we systematically compared the editing efficiencies of PE to conventional gene editing methods in hPSCs. This analysis revealed that PE is overall more efficient and precise than homology-directed repair of site-specific nuclease-induced double-strand breaks. Specifically, PE is more effective in generating heterozygous editing events to create autosomal dominant disease-associated mutations. By stably integrating the nCas9-RT into hPSCs we achieved editing efficiencies equal to those reported for cancer cells, suggesting that the expression of the PE components, rather than cell-intrinsic features, limit PE in hPSCs. To improve the efficiency of PE in hPSCs, we optimized the delivery modalities for the PE components. Delivery of the nCas9-RT as mRNA combined with synthetically generated, chemically-modified pegRNAs and nicking guide RNAs improved editing efficiencies up to 13-fold compared with transfecting the PE components as plasmids or ribonucleoprotein particles. Finally, we demonstrated that this mRNA-based delivery approach can be used repeatedly to yield editing efficiencies exceeding 60% and to correct or introduce familial mutations causing Parkinson's disease in hPSCs.

Chemical cross-linking enables rapid identification of RNA-protein and RNA-nucleic acid inter- and intramolecular interactions. However, no method exists to site-specifically and covalently cross-link two user-defined sites within an RNA. Here, we develop RNA-CLAMP, which enables site-specific and enzymatic cross-linking (clamping) of two selected guanine residues within an RNA. Intramolecular clamping can disrupt normal RNA function, whereas subsequent photocleavage of the cross-linker restores activity. We used RNA-CLAMP to clamp two stem loops within the single-guide RNA (sgRNA) of the CRISPR-Cas9 gene editing system via a photocleavable cross-linker, completely inhibiting gene editing. Visible light irradiation cleaved the cross-linker and restored gene editing with high spatiotemporal resolution. Design of two photocleavable linkers responsive to different wavelengths of light allowed multiplexed photoactivation of gene editing in mammalian cells. This photoactivated CRISPR-Cas9 gene editing platform benefits from undetectable background activity, provides a choice of activation wavelengths, and has multiplexing capabilities.

A compact protein with a size of <1,000 amino acids, the CRISPR-associated protein CasX is a fundamentally distinct RNA-guided nuclease when compared to Cas9 and Cas12a. Although it can induce RNA-guided genome editing in mammalian cells, the activity of CasX is less robust than that of the widely used S. pyogenes Cas9. Here, we show that structural features of two CasX homologs and their guide RNAs affect the R-loop complex assembly and DNA cleavage activity. Cryo-EM-based structural engineering of either the CasX protein or the guide RNA produced two new CasX genome editors (DpbCasX-R3-v2 and PlmCasX-R1-v2) with significantly improved DNA manipulation efficacy. These results advance both the mechanistic understanding of CasX and its application as a genome-editing tool.

Many novel CRISPR-based genome-editing tools, with a wide variety of applications, have been developed in the past few years. The original CRISPR-Cas9 system was developed as a tool to alter genomic sequences in living organisms in a simple way. However, the functions of new CRISPR tools are not limited to conventional genome editing mediated by non-homologous end-joining or homology-directed repair but expand into gene-expression control, epigenome editing, single-nucleotide editing, RNA editing and live-cell imaging. Furthermore, genetic perturbation screening by multiplexing guide RNAs is gaining popularity as a method to identify causative genes and pathways in an unbiased manner. New CRISPR tools can also be applied to ex vivo or in vivo therapeutic genome editing for the treatment of conditions such as hyperlipidaemia. In this Review, we first provide an overview of the diverse new CRISPR tools that have been developed to date. Second, we summarize how these new CRISPR tools are being used to study biological processes and disease mechanisms in cardiovascular research and medicine. Finally, we discuss the prospect of therapeutic genome editing by CRISPR tools to cure genetic cardiovascular diseases.

Giardia intestinalis, a protist parasite that infects approximately 280 million people around the world each year, has been eluding the use of CRISPR/Cas9 to generate knockout cell lines due to its tetraploid genome. In this work, we show the ability of the in vitro assembled CRISPR/Cas9 components to successfully edit the genome of G. intestinalis. The cell line that stably expresses Cas9 in both nuclei of G. intestinalis showed effective recombination of the cassette containing the transcription units for the gRNA and the resistance marker. This highly efficient process led to the removal of all gene copies at once for three independent experimental genes, mem, cwp1 and mlf1. The method was also applicable to incomplete disruption of the essential gene, as evidenced by significantly reduced expression of tom40. Finally, testing the efficiency of Cas9-induced recombination revealed that homologous arms as short as 150 bp can be sufficient to establish a complete knockout cell line in G. intestinalis.
