Progranulin is a pleiotropic growth factor with important physiological roles in embryogenesis and maintenance of adult tissue homeostasis. While-progranulin deficiency is associated with a broad range of pathological conditions affecting the brain, such as frontotemporal dementia and neuronal ceroid lipofuscinosis, progranulin upregulation characterizes many tumors, including brain tumors, multiple myeloma, leiomyosarcoma, mesothelioma and epithelial cancers such as ovarian, liver, breast, bladder, adrenal, prostate and kidney carcinomas. The increase of progranulin levels in tumors might have diagnostic and prognostic significance. In cancer, progranulin has a pro-tumorigenic role by promoting cancer cell proliferation, migration, invasiveness, anchorage-independent growth and resistance to chemotherapy. In addition, progranulin regulates the tumor microenvironment, affects the function of cancer-associated fibroblasts, and modulates tumor immune surveillance. However, the molecular mechanisms of progranulin oncogenic function are not fully elucidated. In bladder cancer, progranulin action relies on the activation of its functional signaling receptor EphA2. Notably, more recent data suggest that progranulin can also modulate a functional crosstalk between multiple receptor-tyrosine kinases, demonstrating a more complex and context-dependent role of progranulin in cancer. Here, we will review what is currently known about the function of progranulin in tumors, with a focus on its molecular mechanisms of action and regulation.

We aimed to determine the prevalence of abnormal imaging findings in the acute phase of NA. METHODS: Magnetic resonance neurography (MRN) and high-resolution ultrasound (US) examinations were performed at five sites. The investigation included 39 patients with acute NA who underwent imaging within 31 days of symptom onset. Correlation between imaging and electromyography (EMG) findings was measured. RESULTS: US was performed in 29 patients and MRN in 23; 16 patients underwent US only, 10 MRN only, and 13 had both. US and MRN showed nerve abnormalities within 1 mo from NA onset in 90% of patients. HGCs were found in 74% (29/39) of the patients: 4 within 1 wk, 8 within 2 wk, 5 within 3 wk, and 12 within 4 wk. The earliest HGC on US was found within 12 h, and on MRN within 3 days from symptom onset. MRN demonstrated a denervation edema pattern of affected muscles in 91% of the patients. The shortest time to observe an edema pattern on MRN was 8 days. EMG was performed in 30 patients and revealed fibrillation potentials in affected muscles in 22 (73%). A denervation edema pattern on MRN was significantly associated with the presence of HGCs both on MRN and US, and with fibrillation potentials on EMG. DISCUSSION: In the early phase of NA, US and MRN are useful diagnostic techniques for demonstrating nerve abnormalities.

SOXS (SOn of X-Shooter) is a medium resolution (~4500) wide-band (0.35 - 2.0 {\mu}m) spectrograph which passed the Final Design Review in 2018. The instrument is in the final integration phase and it is planned to be installed at the NTT in La Silla by next year. It is mainly composed of five different optomechanical subsystems (Common Path, NIR spectrograph, UV-VIS spectrograph, Camera, and Calibration) and other mechanical subsystems (Interface flange, Platform, cable corotator, and cooling system). A brief overview of the optomechanical subsystems is presented here as more details can be found in the specific proceedings while a more comprehensive discussion is dedicated to the other mechanical subsystems and the tools needed for the integration of the instrument. Moreover, the results obtained during the acceptance of the various mechanical elements are presented together with the experiments performed to validate the functionality of the subsystems. In this paper we present the post-FDR modifications in the mechanical design due to the various iterations with the manufacturers and the actual procurement status. The last part describes the strategy used to keep under control the mechanical interfaces between the subsystems.

We present here the development status of the NIR spectrograph of the Son Of X-Shooter (SOXS) instrument, for the ESO/NTT telescope at La Silla (Chile). SOXS is a R~4,500 mean resolution spectrograph, with a simultaneously coverage from about 0.35 to 2.00 micron. It will be mounted at the Nasmyth focus of the NTT. The two UV-VIS-NIR wavelength ranges will be covered by two separated arms. The NIR spectrograph is a fully cryogenic echelle-dispersed spectrograph, working in the range 0.80-2.00 micron, equipped with a Hawaii H2RG IR array from Teledyne. The whole spectrograph will be cooled down to about 150 K (but the array at 40 K), to lower the thermal background, and equipped with a thermal filter to block any thermal radiation above 2.0 micron. The 'chromosome map' diagram (ChM) proved a successful tool to identify and characterize multiple populations (MPs) in 59 Galactic Globular Clusters (GCs). Here, we construct ChMs for 11 GCs of both Magellanic Clouds (MCs) and with different ages to compare MPs in Galactic and extra-Galactic environments, and explore whether this phenomenon is universal through 'place' and 'time'. MPs are detected in five clusters. The fractions of 1G stars, ranging from about 50% to more than 80%, are significantly higher than those observed in Galactic GCs with similar present-day masses. By considering both Galactic and MC clusters, the fraction of 1G stars exhibits: (i) a strong anti-correlation with the present-day mass, and (ii) with the present-day mass of 2G stars; (iii) a mild anti-correlation with 1G present-day mass. All Galactic clusters without MPs have initial masses smaller than ~1.5 10^5 solar masses but a mass threshold governing the occurrence of MPs seems challenged by massive simple-population MC GCs; (iv) Milky Way clusters with large perigalactic distances typically host larger fractions of 1G stars, but the difference disappears when we use initial cluster masses. These facts are consistent with a scenario where the stars lost by GCs mostly belong to the 1G. By exploiting recent work based on Gaia, half of the known Type II GCs appear clustered in a distinct region of the integral of motions space, thus suggesting a common progenitor galaxy. We solve the twisted conjugacy problem on Thompson’s group F. We also exhibit orbit undecidable subgroups of Aut(F), and give a proof that Aut(F) and Aut+(F) are orbit decidable provided a certain conjecture on Thompson’s group T is true. By using general criteria introduced by Bogopolski, Martino and Ventura in [5], we construct a family of free extensions of F where the conjugacy problem is unsolvable. INTRODUCTION: African-American children have a greater acute insulin response to iv glucose (AIR) compared with Latino children despite a similar degree of insulin resistance and body composition. It is unclear whether African-Americans demonstrate an exaggerated insulin response to an oral glucose challenge and whether any differences are seen in more obese children in advanced pubertal development. PURPOSE: Our objective was to compare glucose and insulin indices derived from an oral glucose tolerance test (OGTT) and iv glucose tolerance test (IVGTT) in sedentary, obese African-American (n=59) and Latino (n=83) adolescents. METHODS: Glucose and insulin incremental area under the curve was measured during an OGTT, and AIR, insulin sensitivity, disposition index, and glucose effectiveness were assessed during an IVGTT. Body composition was assessed via dual-energy x-ray absorptiometry and magnetic resonance imaging. RESULTS: From the OGTT, glucose and insulin IAUC were 29.1 and 22.5% lower (P=0.01) in African-Americans compared with Latino adolescents. From the IVGTT, insulin sensitivity and glucose effectiveness were 41.7% (P<0.01) and 50.0% (P=0.02) lower in African-Americans compared to Latinos. AIR (P=0.001) and disposition index (P=0.02) were 63.0 and 48.8% higher in African-Americans, respectively, compared with Latinos. These findings persisted after controlling for body composition and fat distribution. CONCLUSIONS: There were marked differences in glucose and insulin indices derived from the OGTT and IVGTT. African-Americans were more insulin resistant as measured by the IVGTT compared with the Latino adolescents. However, the well-described hyperinsulinemia in response to iv glucose was not observed after oral glucose in African-American adolescents.

BACKGROUND: To date, no studies have assessed the longitudinal changes of dietary intake on metabolic risk factors in Latino youth. OBJECTIVE: We assessed the relation between changes in dietary intake, specifically sugar and fiber intakes, with changes in adiposity and risk factors for type 2 diabetes in a longitudinal analysis of overweight Latino youth. DESIGN: Overweight Latino youth (n = 85; aged 11-17 y) underwent the following measures over 2 y [mean (+/-SD) time difference was 1.5 +/- 0.5 y]: dietary intake by 2-d diet recalls, body composition by dual-energy X-ray absorptiometry and magnetic resonance imaging, and glucose and insulin indexes by oral- and intravenous-glucose-tolerance tests. Partial correlations and repeated-measures analysis of covariance assessed the relation between changes in dietary intake with changes in adiposity and glucose and insulin indexes, independent of the following a priori covariates: sex, Tanner stage, time between visits, and baseline dietary and metabolic variables of interest. RESULTS: Increases in total dietary fiber (g/1000 kcal) and insoluble fiber (g/1000 kcal) were associated with decreases in visceral adipose tissue (VAT) (r = -0.29, P = 0.02, and r = -0.27, P = 0.03, for total dietary and insoluble fiber, respectively), independent of baseline covariates and change in subcutaneous abdominal adipose tissue. Participants who had decreased total dietary fiber (mean decrease of 3 g . 1000 kcal(-1) x d(-1)) had significant increases in VAT compared with participants who had increased total dietary fiber (21% compared with -4%; P = 0.02). No other changes in dietary variables were related to changes in adiposity or metabolic variables. CONCLUSION: Small reductions in dietary fiber intake over 1-2 y can have profound effects on increasing visceral adiposity in a high-risk Latino youth population.

We investigate the presence of topological twistons in crystalline polyethylene. We describe crystalline polyethylene with a model that couples the torsional and longitudinal degrees of freedom of the polymeric chain by means of a system of two real scalar fields. This model supports topological twistons, which are described by exact and stable topological solutions that appear when the interaction between torsional and longitudinal fields is polynomial, containing up to the sixth power in the fields. We calculate the energy of the topological twiston, and the result is in very good agreement with the value obtained via molecular simulation., Comment: Latex, 10 pages. To appear in Chem. Phys. We introduce an alternate model to describe twistons in crystalline polyethylene. The model couples torsional and longitudinal degrees of freedom and appears as an extension of a model that describes only the torsional motion.
