The dual functions of TMEM16F as Ca2+-activated ion channel and lipid scramblase raise intriguing questions regarding their molecular basis. Intrigued by the ability of the FDA-approved drug niclosamide to inhibit TMEM16F-dependent syncytia formation induced by SARS-CoV-2, we examined cryo-EM structures of TMEM16F with or without bound niclosamide or 1PBC, a known blocker of TMEM16A Ca2+-activated Cl- channel. Here, we report evidence for a lipid scrambling pathway along a groove harboring a lipid trail outside the ion permeation pore. This groove contains the binding pocket for niclosamide and 1PBC. Mutations of two residues in this groove specifically affect lipid scrambling. Whereas mutations of some residues in the binding pocket of niclosamide and 1PBC reduce their inhibition of TMEM16F-mediated Ca2+ influx and PS exposure, other mutations preferentially affect the ability of niclosamide and/or 1PBC to inhibit TMEM16F-mediated PS exposure, providing further support for separate pathways for ion permeation and lipid scrambling.

Optimizing research on the developmental origins of health and disease (DOHaD) involves implementing initiatives maximizing the use of the available cohort study data; achieving sufficient statistical power to support subgroup analysis; and using participant data presenting adequate follow-up and exposure heterogeneity. It also involves being able to undertake comparison, cross-validation, or replication across data sets. To answer these requirements, cohort study data need to be findable, accessible, interoperable, and reusable (FAIR), and more particularly, it often needs to be harmonized. Harmonization is required to achieve or improve comparability of the putatively equivalent measures collected by different studies on different individuals. Although the characteristics of the research initiatives generating and using harmonized data vary extensively, all are confronted by similar issues. Having to collate, understand, process, host, and co-analyze data from individual cohort studies is particularly challenging. The scientific success and timely management of projects can be facilitated by an ensemble of factors. The current document provides an overview of the 'life course' of research projects requiring harmonization of existing data and highlights key elements to be considered from the inception to the end of the project.

The study aims were to examine the factor structure and factorial invariance across parental gender of the PRFQ-I in a community sample, to compare PRF dimensions across mothers and fathers, and to investigate the association between PRF dimensions and infant socioemotional development. We examined this within a longitudinal design in a community sample of 1243 first-time mothers and 512 fathers. The results supported the expected factor structure of PRFQ-I and invariance across parental gender. Significant differences between mothers and fathers were found on the three PRF dimensions comprised by the questionnaire. The PRF dimensions of pre-mentalizing and certainty about mental states significantly predicted infant socioemotional development. Findings show that the PRFQ-I is a valid measure in parents of infants and suggest that PRF in early parenthood is an important factor in infant socioemotional development. Identification of first-time parents with limited PRF enables early intervention and may thus prevent limited PRF from having potentially adverse effects on infant socioemotional development.

Background The MINDMAP project implemented a multinational data infrastructure to investigate the direct and interactive effects of urban environments and individual determinants of mental well-being and cognitive function in ageing populations. Using a rigorous process involving multiple teams of experts, longitudinal data from six cohort studies were harmonised to serve MINDMAP objectives. This article documents the retrospective data harmonisation process achieved based on the Maelstrom Research approach and provides a descriptive analysis of the harmonised data generated. Methods A list of core variables (the DataSchema) to be generated across cohorts was first defined, and the potential for cohort-specific data sets to generate the DataSchema variables was assessed. Where relevant, algorithms were developed to process cohort-specific data into DataSchema format, and information to be provided to data users was documented. Procedures and harmonisation decisions were thoroughly documented. Results The MINDMAP DataSchema (v2.0, April 2020) comprised a total of 2841 variables (993 on individual determinants and outcomes, 1848 on environmental exposures) distributed across up to seven data collection events. The harmonised data set included 220 621 participants from six cohorts (10 subpopulations). Harmonisation potential, participant distributions and missing values varied across data sets and variable domains. Conclusion The MINDMAP project implemented a collaborative and transparent process to generate a rich integrated data set for research in ageing, mental well-being and the urban environment. The harmonised data set supports a range of research activities and will continue to be updated to serve ongoing and future MINDMAP research needs.

Promoting axon regeneration in the central and peripheral nervous system is of clinical importance in neural injury and neurodegenerative diseases. Both pro- and antiregeneration factors are being identified. We previously reported that the Rtca mediated RNA repair/splicing pathway restricts axon regeneration by inhibiting the nonconventional splicing of Xbp1 mRNA under cellular stress. However, the downstream effectors remain unknown. Here, through transcriptome profiling, we show that the tubulin polymerization-promoting protein (TPPP) ringmaker/ringer is dramatically increased in Rtca-deficient Drosophila sensory neurons, which is dependent on Xbp1. Ringer is expressed in sensory neurons before and after injury, and is cell-autonomously required for axon regeneration. While loss of ringer abolishes the regeneration enhancement in Rtca mutants, its overexpression is sufficient to promote regeneration both in the peripheral and central nervous system. Ringer maintains microtubule stability/dynamics with the microtubule-associated protein futsch/MAP1B, which is also required for axon regeneration. Furthermore, ringer lies downstream from and is negatively regulated by the microtubule-associated deacetylase HDAC6, which functions as a regeneration inhibitor. Taken together, our findings suggest that ringer acts as a hub for microtubule regulators that relays cellular status information, such as cellular stress, to the integrity of microtubules in order to instruct neuroregeneration.

Starting with the Q559K mutant that shows no current rundown and less outward rectification in excised patch, we found that the channel shifted its ion selectivity in response to the change of intracellular Ca2+ concentration, with an increased permeability ratio of Cl- to Na+ (PCl-/PNa+) at a higher Ca2+ level. The gradual shift of relative ion permeability did not correlate with the channel activation state. Instead, it was indicative of an alteration of electrostatic field in the permeation pathway. The dynamic change of ion selectivity suggests a charge-screening mechanism for TMEM16F ion conduction, and it provides hints to further studies of TMEM16F physiological functions.

Recorded in NaCl−based solution, the channel shifted its ion selectivity from Na+-selective to Cl−-selective when intracellular Ca2+ was increased. Shifting ion-selectivity synergistically by intracellular divalent ions and membrane potential could work as a built-in mechanism that allows TMEM16F to brake the positive feedback.

A fundamental goal of evolutionary ecology is to identify the sources underlying trait variation on which selection can act. Phenotypic variation will be determined by both genetic and environmental factors, and adaptive phenotypic plasticity is expected when organisms can adjust their phenotypes to match environmental cues. Much recent research interest has focused on the relative importance of environmental and genetic factors on the expression of behavioral traits, in particular, and how they compare with morphological and life‐history traits. Little research to date examines the effect of development on the expression of heritable variation in behavioral traits, such as boldness and activity. We tested for genotype, environment, and gen‐ otype‐by‐environment differences in body mass, development time, boldness, and activity, using developmental density treatments combined with a quantitative ge‐ netic design in the sand field cricket (Gryllus firmus). Similar to results from previous work, animals reared at high densities were generally smaller and took longer to ma‐ ture, and body mass and development time were moderately heritable. In contrast, neither boldness nor activity responded to density treatments, and they were not heritable. The only trait that showed significant genotype‐by‐environment differ‐ ences was development time. It is possible that adaptive behavioral plasticity is not evident in this species because of the highly variable social environments it naturally experiences. Our results illustrate the importance of validating the assumption that behavioral phenotype reflects genetic patterns and suggest questions about the role of environmental instability in trait variation and heritability.

Our study reveals the dependence of TMEM16F activity on phosphoinositides and provides one mechanism for TMEM16F activation to be strictly regulated in the cell membrane.

DOD spends billions of dollars each year to maintain key business functions intended to support the warfighter. While DOD maintains military forces with unparalleled capabilities, it continues to confront decades-old management weaknesses related to its business functions that support these forces. GAO designated DOD's approach to business transformation as high-risk in 2005 because DOD had not taken steps to achieve and sustain business reform on a strategic, department-wide basis. This report identifies the extent to which (1) DOD has addressed business transformation weaknesses and (2) the Office of the DCMO has effectively monitored the progress of its business transformation efforts. GAO reviewed documentation, such as DOD's Fiscal Year 2014-2015 Strategic Management Plan and performance measures established for its business functions. GAO also interviewed officials responsible for DOD's business transformation efforts. GAO recommends that the CMO and DCMO document and communicate priorities for DOD's business transformation efforts and oversee the development of a correction action plan. GAO also recommends that the DCMO develop clear and comprehensive performance measures that are aligned with strategic goals to monitor the progress of its business transformation efforts. In May 2012, OMB directed agencies, among other things, to establish senior-level review processes for all hosted or attended conferences exceeding specific cost thresholds. DOD and DOE implemented conference policies in 2012, with subsequent updates, to comply with OMB s requirements. GAO was mandated to review the effects of OMB s requirements on DOD s and DOE s S&T missions. Among other things, this report (1) examines S&T conference participation changes since policy implementation, and examines the extent to which DOD and DOE (2) face and have mitigated implementation challenges, and (3) have identified risks from conference participation changes and analyzed them for any potential effects. GAO reviewed conference policies, and interviewed and surveyed conference management and S&T officials from principal research labs. Survey results were not generalizable. GAO recommends that DOD and DOE (1) establish time frames for providing conference request decisions and (2) develop a plan to analyze and periodically reevaluate risks from conference participation changes. DOD partially concurred with the first recommendation, stating it will collect and analyze data to implement further solutions. GAO still believes the recommendation is valid, as discussed in the report. DOD concurred with the second recommendation and DOE concurred with both recommendations., Report to Congressional Committees.

Social interactions among conspecifics are a fundamental and adaptively significant component of the biology of numerous species. Such interactions give rise to group living as well as many of the complex forms of cooperation and conflict that occur within animal groups. Although previous conceptual models have focused on the ecological causes and fitness consequences of variation in social interactions, recent developments in endocrinology, neuroscience, and molecular genetics offer exciting opportunities to develop more integrated research programs that will facilitate new insights into the physiological causes and consequences of social variation. Here, we propose an integrative framework of social behavior that emphasizes relationships between ultimate-level function and proximate-level mechanism, thereby providing a foundation for exploring the full diversity of factors that underlie variation in social interactions, and ultimately sociality. In addition to identifying new model systems for the study of human psychopathologies, this framework provides a mechanistic basis for predicting how social behavior will change in response to environmental variation. We argue that the study of non-model organisms is essential for implementing this integrative model of social behavior because such species can be studied simultaneously in the lab and field, thereby allowing integration of rigorously controlled experimental manipulations with detailed observations of the ecological contexts in which interactions among conspecifics occur.

The tightly adherent monolayer of epithelial cells that line the luminal side of the intestine provides a dynamic and highly regulated barrier to the passive and active transport of ions, water and macromolecules. Cathecol-O-methyl transferase inhibition prevents tissue degradation of catecholaminesincluding dopamine and intracellular carbonic anhydrase activity is an important regulator of ion and water transport in the small intestine. Intestinal anaphylaxis trigger multiple effector cells and constitute adriving force for changes in active ion and water transport. Nitric oxide protect and damage the gastrointestinal mucosa through a variety of mechanisms and intestinal obstruction is caused at least in part by the activation of nerves and participation of the enteric nervous system. Studies were made in humans with the LOCI-GUT technique with isolation of a duodenal or jejunal segment between balloons. Duodenal mucosal bicarbonate secretion increased in response to the COMT inhibitor nitecapone, similar in magnitude to that obtained by a prostaglandin E1 analogue. Acetazolamide pretreatment significantly decreased mean basal and PGE2-stimulated HCO3- secretion as well as PGE2 stimulated secretion of Cl-, Na+, K+, and water. Challenge with different intestinal antigens induced a mucosal release of several inflammatory mediators. The parallel increase in albumin and hyaluronan illustrated the local anaphylactic reaction and is probably related to a mucosal lymphoedema. Basal net absorption of Cl- and Na+ ions and water was also changed into a net secretion. Further studies regarding mucosal transport in the human small intestine revealed that nitric oxide is continuously synthesized and released into the intestinal lumen in a concentration of 1/100 of that in the stomach. Cephalic-vagal stimulation with sham feeding did not increase NO synthesis, but intestinal obstruction dose-dependently and significantly increased this synthesis. Inhibition of nerve stimulation

We tested for genotype, environment, and gen‐ otype‐by‐environment differences in body mass, development time, boldness, and activity, using developmental density treatments combined with a quantitative ge‐ netic design in the sand field cricket (Gryllus firmus). Similar to results from previous work, animals reared at high densities were generally smaller and took longer to ma‐ ture, and body mass and development time were moderately heritable. In contrast, neither boldness nor activity responded to density treatments, and they were not heritable. The only trait that showed significant genotype‐by‐environment differ‐ ences was development time. It is possible that adaptive behavioral plasticity is not evident in this species because of the highly variable social environments it naturally experiences. Our results illustrate the importance of validating the assumption that behavioral phenotype reflects genetic patterns and suggest questions about the role of environmental instability in trait variation and heritability.
