Evaluation plays a significant role in modern natural language processing. Most modern NLP benchmarks consist of arbitrary sets of tasks that neither guarantee any generalization potential for the model once applied outside the test set nor try to minimize the resource consumption needed for model evaluation. This paper presents a theoretical instrument and a practical algorithm to calculate similarity between benchmark tasks, we call this similarity measure "Vygotsky distance". The core idea of this similarity measure is that it is based on relative performance of the "students" on a given task, rather that on the properties of the task itself. If two tasks are close to each other in terms of Vygotsky distance the models tend to have similar relative performance on them. Thus knowing Vygotsky distance between tasks one can significantly reduce the number of evaluation tasks while maintaining a high validation quality. Experiments on various benchmarks, including GLUE, SuperGLUE, CLUE, and RussianSuperGLUE, demonstrate that a vast majority of NLP benchmarks could be at least 40% smaller in terms of the tasks included. Most importantly, Vygotsky distance could also be used for the validation of new tasks thus increasing the generalization potential of the future NLP models.

In recent years, several industrial solutions for the problem of multi-token code completion have appeared, each making a great advance in the area but mostly focusing on cloud-based runtime and avoiding working on the end user's device. In this work, we describe our approach for building a multi-token code completion feature for the JetBrains' IntelliJ Platform, which we call Full Line Code Completion. The feature suggests only syntactically correct code and works fully locally, i.e., data querying and the generation of suggestions happens on the end user's machine. We share important time and memory-consumption restrictions, as well as design principles that a code completion engine should satisfy. Working entirely on the end user's device, our code completion engine enriches user experience while being not only fast and compact but also secure. We share a number of useful techniques to meet the stated development constraints and also describe offline and online evaluation pipelines that allowed us to make better decisions. Our online evaluation shows that the usage of the tool leads to 1.5 times more code in the IDE being produced by code completion. The described solution was initially started with the help of researchers and was bundled into two JetBrains' IDEs - PyCharm Pro and DataSpell - at the end of 2023, so we believe that this work is useful for bridging academia and industry, providing researchers with the knowledge of what happens when complex research-based solutions are integrated into real products., Comment: 12 pages, 4 figures

Increasing numbers of small proteins with diverse physiological roles are being identified and characterized in both prokaryotic and eukaryotic systems, but the origins and evolution of these proteins remain unclear. Recent genomic sequence analyses in several organisms suggest that new functions encoded by small open reading frames (sORFs) may emerge de novo from noncoding sequences. However, experimental data demonstrating if and how randomly generated sORFs can confer beneficial effects to cells are limited. Here, we show that by upregulating hisB expression, de novo small proteins (<= 50 amino acids in length) selected from random sequence libraries can rescue Escherichia coli cells that lack the conditionally essential SerB enzyme. The recovered small proteins are hydrophobic and confer their rescue effect by binding to the 5 ' end regulatory region of the his operon mRNA, suggesting that protein binding promotes structural rearrangements of the RNA that allow increased hisB expression. This study adds RNA regulatory elements as another interacting partner for de novo proteins isolated from random sequence libraries and provides further experimental evidence that small proteins with selective benefits can originate from the expression of nonfunctional sequences.

The CMS experiment at the CERN LHC will be upgraded to accommodate the 5-fold increase in the instantaneous luminosity expected at the High-Luminosity LHC (HL-LHC) [1]. Concomitant with this increase will be an increase in the number of interactions in each bunch crossing and a significant increase in the total ionising dose and fluence. One part of this upgrade is the replacement of the current endcap calorimeters with a high granularity sampling calorimeter equipped with silicon sensors, designed to manage the high collision rates [2]. As part of the development of this calorimeter, a series of beam tests have been conducted with different sampling configurations using prototype segmented silicon detectors. In the most recent of these tests, conducted in late 2018 at the CERN SPS, the performance of a prototype calorimeter equipped with ~12,000 channels of silicon sensors was studied with beams of high-energy electrons, pions and muons. This paper describes the custom-built scalable data acquisition system that was built with readily available FPGA mezzanines and low-cost Raspberry Pi computers.

As part of its HL-LHC upgrade program, the CMS collaboration is developing a High Granularity Calorimeter (CE) to replace the existing endcap calorimeters. The CE is a sampling calorimeter with unprecedented transverse and longitudinal readout for both electromagnetic (CE-E) and hadronic (CE-H) compartments. The calorimeter will be built with ~30,000 hexagonal silicon modules. Prototype modules have been constructed with 6-inch hexagonal silicon sensors with cell areas of 1.1 cm2, and the SKIROC2-CMS readout ASIC. Beam tests of different sampling configurations were conducted with the prototype modules at DESY and CERN in 2017 and 2018. This paper describes the construction and commissioning of the CE calorimeter prototype, the silicon modules used in the construction, their basic performance, and the methods used for their calibration.

The Arctic is warming faster than any other region on Earth. Putting this rapid warming into perspective is challenging because instrumental records are often short or incomplete in polar regions and precisely-dated temperature proxies with high temporal resolution are largely lacking. Here, we provide this long-term perspective by reconstructing past summer temperature variability at Yamal Peninsula – a hotspot of recent warming – over the past 7638 years using annually resolved tree-ring records. We demonstrate that the recent anthropogenic warming interrupted a multi-millennial cooling trend. We find the industrial-era warming to be unprecedented in rate and to have elevated the summer temperature to levels above those reconstructed for the past seven millennia (in both 30-year mean and the frequency of extreme summers). This is undoubtedly of concern for the natural and human systems that are being impacted by climatic changes that lie outside the envelope of natural climatic variations for this region. © 2022, The Author(s).

azTotMD 2.0 is a parallel molecular dynamics program which includes both conventional algorithms and a novel “radiative” thermostat and a temperature-dependent force field. The radiative thermostat is based on the black-body radiation law and acts like virtual adsorption and radiation of photons. The thermostat algorithm has complexity of O(N), it can accelerate and decelerate atoms and this action over time leads to a Maxwell-like distribution of velocities. The temperature-dependent pair potential includes atomic radii, which are functions of the thermal excitation of atoms. The combination of the radiative thermostat and the potential allows to reproduce many phenomena such as phase transitions, thermal expansion, defect formation, surface tension, vapor saturation, glass formation and devitrification. © 2022 The Author(s).

Con base en estudios previos se identificó la necesidad de aumentar la validez de la toma de decisiones estratégicas para determinar (aclarar) la estrategia de desarrollo de las capacidades de las Fuerzas Armadas en el largo plazo. Se identificó la necesidad de elaborar un método multicriterio para el análisis estratégico del desarrollo de las Fuerzas Armadas basado en datos estadísticos sobre los resultados de la reforma y revisión del sector de la defensa, minimizando el uso de juicios de expertos. Para la elaboración del método se utilizó un enfoque conceptual que combina las etapas seleccionadas de elección de áreas prioritarias para el desarrollo de las capacidades de las Fuerzas Armadas y una forma recursiva de organización del proceso de cómputo. El proceso involucró el desarrollo de un sistema de indicadores y criterios de desempeño, sus coeficientes de peso, funciones recursivas y objetivo para construir un modelo matemático que sintetice los resultados del análisis estratégico del desarrollo de las Fuerzas Armadas. El método de análisis estratégico del desarrollo de las Fuerzas Armadas mediante el método recursivo de organización del proceso computacional (SADAF-RW) permite obtener valoraciones objetivas y el efecto esperado de implementar la opción estratégica elegida para la toma de decisiones estratégicas. Está dirigido a mejorar el estado de preparación y capacidades de las Fuerzas Armadas en el largo plazo, minimizando el uso de juicios de expertos. El método puede ser utilizado en la elaboración de un aparato científico y metodológico para evaluar la eficacia de la planificación estratégica en el desarrollo de las Fuerzas Armadas, ya que se basa en los principios de objetividad de las evaluaciones, consistencia y eficacia de las decisiones estratégicas., Previous research has shown the need to increase the validity of the strategic decisions taken to determine (clarify) the strategy for developing the capabilities of the Ukrainian Armed Forces in the long term. It is necessary to elaborate a multi-criteria method for the strategic analysis of the development of the Armed Forces, based on statistical data obtained from the results of the defence reform and the defence review, minimising the use of expert judgments. To elaborate this method a conceptual approach was used, which combines choosing priority areas for developing the capabilities of the Armed Forces and a recursive way of organising the computing process. The process involved the creation of a system of indicators and performance criteria, their weight coefficients, and recursive and objective functions for constructing a mathematical model for synthesising the results of the strategic analysis of the development of the Armed Forces. The method of strategic analysis of the development of the Armed Forces using the recursive method of organising the computational process (SADAF-RW) produces objective assessments and the expected effect of implementing the chosen strategy for making strategic decisions. It is aimed at improving the state of readiness and capabilities of the Armed Forces in the long term, minimising the use of expert judgments. The method can be used in the elaboration of a scientific and methodological apparatus for assessing the effectiveness of strategic planning for the development of the Armed Forces since it is based on the principles of objectivity of assessments, consistency, and effectiveness of strategic decisions.

The Nuclotron-base Ion Collider fAcility (NICA) is under construction at the Joint Institute for Nuclear Research (JINR), with commissioning of the facility expected in late 2022. The Multi-Purpose Detector (MPD) has been designed to operate at NICA and its components are currently in production. The detector is expected to be ready for data taking with the first beams from NICA. This document provides an overview of the landscape of the investigation of the QCD phase diagram in the region of maximum baryonic density, where NICA and MPD will be able to provide significant and unique input. It also provides a detailed description of the MPD set-up, including its various subsystems as well as its support and computing infrastructures. —In recent years, the supra-long Yamal tree-ring chronology has been significantly extended and became much more reliable. This article characterizes the sample wood used to build the longest absolutely dated Siberian Larch tree-ring chronology of the Subarctic area, i.e. from 6748 BC to 2019 AD, for a total continuous period of 8768 years. The ecological value of the temporal and spatial distribution of the dated trees are presented, and their potential use for application in various field of natural sciences and humanities are discussed. © 2021, Pleiades Publishing, Ltd.

For a system of nonlinear fractional differential equations, the problem of reconstructing an unknown external impact is considered. It is complicated by the fact that only a part of system's parameters is available for measuring. An algorithm for solving this problem is proposed, which is resistant to informational noises and computational errors, and is based on regularization methods and constructions of guaranteed control theory. A numerical example illustrating the operation of the algorithm is considered. © 2020 Elsevier B.V.

Current state-of-the-art NLP systems use large neural networks that require lots of computational resources for training. Inspired by human knowledge acquisition, researchers have proposed curriculum learning, - sequencing of tasks (task-based curricula) or ordering and sampling of the datasets (data-based curricula) that facilitate training. This work investigates the benefits of data-based curriculum learning for large modern language models such as BERT and T5. We experiment with various curricula based on a range of complexity measures and different sampling strategies. Extensive experiments on different NLP tasks show that curricula based on various complexity measures rarely has any benefits while random sampling performs either as well or better than curricula.

Mobile genetic elements, such as plasmids, phages, and transposons, are important sources for evolution of novel functions. In this study, we performed a large-scale screening of metagenomic phage libraries for their ability to suppress temperature-sensitivity in Salmonella enterica serovar Typhimurium strain LT2 mutants to examine how phage DNA could confer evolutionary novelty to bacteria. We identified an insert encoding 23 amino acids from a phage that when fused with a bacterial DNA-binding repressor protein (LacI) resulted in the formation of a chimeric protein that localized to the outer membrane. This relocalization of the chimeric protein resulted in increased membrane vesicle formation and an associated suppression of the temperature sensitivity of the bacterium. Both the host LacI protein and the extracellular 23-amino acid stretch are necessary for the generation of the novel phenotype. Furthermore, mutational analysis of the chimeric protein showed that although the native repressor function of the LacI protein is maintained in this chimeric structure, it is not necessary for the new function. Thus, our study demonstrates how a gene fusion between foreign DNA and bacterial DNA can generate novelty without compromising the native function of a given gene.

The article describes a clinical case presentation of PEEP adjustment under lung ultrasound control in a newborn with respiratory distress syndrome during artificial respiration. Diagnostic value for B-line reduction and appearance of A-lines as predictors of lung atelectasis opening is demonstrated. The practice of low-volume lung recruitment which is achieved by functional residual capacity increasing rather than raising of inspiratory peak pressure can be considered safer as to the risk of ventilator-associated lung injury and the development of severe air leakage syndrome. In addition, the absence of significant fluctuations in the partial pressure of carbon dioxide is prognostically more positive in terms of reducing cerebral perfusion. Nevertheless, additional evidence based data needs to be collected before any further conclusions can be drawn., Украина)В статье описан случай из клинической практики подбора величины РЕЕР под ультразвуковым контролем легких при проведении искусственной вентиляции легких у новорожденного с респираторным дистресс-синдромом. Практика низкообъёмного легочного рекрутмента, который достигается за счет увеличения функциональной остаточной емкости, а не повышение пикового инспираторного давления, может считаться более безопасной в отношении риска вентилятор-ассоциированных повреждений легких и развития тяжелого синдрома утечки воздуха. Кроме того, отсутствие существенных колебаний парциального давления диоксида углерода прогностически более положительной по уменьшению церебральной перфузии. Крім того, відсутність істотних коливань парціального тиску діоксиду вуглецю прогностично є більш позитивною щодо зменшення церебральної перфузії. Але потрібні додаткові відкриті контрольовані дослідження достатнього доказового рівня, перш ніж можна буде зробити остаточні висновки.

The negative impacts of standard pharmacologic sedative agents suggest that alternative agents should be investigated. Dexmedetomidine could be the new option for sedation in newborns with hypoxic-ischemic encephalopathy requiring mechanical ventilation. The aim – to determine the impact of dexmedetomidine and other sedatives on the cerebral blood flow and outcomes of hypoxic-ischemic encephalopathy in term neonates. Data of 205 term infants with hypoxic-ischemic encephalopathy by Sarnat scale stage II-III were collected during ≤72 hours of life. The infants were divided using a simple open randomization by pharmacological sedative agents during mechanical ventilation into dexmedetomidine group (n=46) and the control group (n=159), which included morphine, sodium oxybutyrate, and diazepam in standard recommended doses. A comparative analysis of the effect of dexmedetomidine and other drugs on cerebral perfusion and outcomes of hypoxic-ischemic encephalopathy was performed. A significant difference between groups in days of trachea extubation (p=0.022) was found; the chance for babies to be extubated before the 7th day of treatment was significantly higher in the dexmedetomidine group 68% versus 33% in the control group (p=0.018) with HR 0.48 (95% CI 0.27-0.86, p=0.011). Also, the NIRS index rScO2 differed significantly between the studied and control groups on the 1st day of treatment (65% versus 79%, p=0.012) and on the 2nd day of treatment (74% versus 81%, p=0.035). Mean arterial pressure was higher in the dexmedetomidine group compared to the control group – (58 [51-65] mm Hg versus 53 [46-60] mm Hg, p<0.001), with a lower dose of dobutamine (EV -1.87, 95% CI from -3.25 to -0.48, p=0.009). To date, there are no published results of large randomized controlled studies compared the differentiated influence of different modes of ventilation on cerebral perfusion in newborns with hypoxic-ischemic encephalopathy (HIE). New mode of ventilation named neurally adjusted ventilatory assist (NAVA) looks promising on this field, because it has already proved its advantages in premature babies. The purpose was to compare the impact of neurally adjusted ventilatory assist and other modes of ventilation on cerebral blood flow in the acute period of HIE in full-term neonates. Materials and methods. Data of 205 term infants with hypoxi­c-ischemic encephalopathy Sarnat stage II–III was collected during ≤ 72 hours of life. All the infants were randomized into the group of NAVA (n = 16) and the control group (n = 189), which included such modes of ventilation as pressure control (PC), synchronized intermittent-mandatory ventilation/pressure support ventilation (SIMV/PSV) and pressure-regulated volume control (PRVC). A multivariate dispersion analysis of the impact of NAVA and other modes of ventilation on cerebral perfusion during the acute period of neonatal hypoxi­c-ische­mic encephalopathy was performed. Results. A significant difference was found between groups on day 3 of treatment at the end of the period of therapeutic hypothermia and the rewarming beginning in terms of Doppler resistive index (RI) of cerebral blood flow (0.70 [0.67–0.74] in the NAVA group and 0.66 [0.58–0.72] in the control group; p = 0.021) and the pulsatile index (PI) (1.3 [1.2–1.5] in the NAVA group and 1.2 [1.0–1.40] in the control group; p = 0.032). Also, analysis of variance results confirmed that compared with other ventilation modes, NAVA had a statistically significant positive influence both on the RI (p = 0.009) and on the PI (p = 0.012) at days 2 and 3 of observation. В статье представлен обзор литературы по гипоксически-ишемической энцефалопатии (ГИЭ), которая развивается у младенцев после перенесенной перинатальной ишемии. Она остается существенной проблемой интенсивной терапии у новорожденных в связи с высоким уровнем смертности и неврологической инвалидности с детства, несмотря на достигнутые определенные успехи в отношении диагностики, мониторинга и методов лечения. Главной целью терапии следует считать обеспечение церебральной перфузии и предупреждение вторичных повреждений нейронов, в том числе развития апоптоза. Среди методов диагностики и мониторинга состояния центральной нервной системы у новорожденных ведущую роль играют: нейросонография с допплеровским определением паттернов мозгового кровотока, мониторинг NIRS в сочетании с амплитудно-интегрированной электроэнцефалографией. Сегодня единственным методом интенсивной терапии с доказанным нейропротективным действием остается терапевтическая гипотермия 33–35 °С в течение 72 часов. Ее эффективность пока остается недостаточной: она снижает долю комбинированного результата «смерть/тяжелое нарушение неврологического развития» с 65 до 40–50 %, поэтому продолжаются поиски методов лечения, которые могли бы улучшить последствия умеренной и тяжелой ГИЭ. Среди методов респираторной поддержки, перспективным выглядит применение Neurally Adjusted Ventilatory Assist. До сих пор не решен вопрос идеального раствора для восстановления дефицита объема циркулирующей крови у новорожденных. Кристаллоиды из-за низкого волемического коэффициента оказывают нестойкое воздействие на сердечный выброс. Использование альбумина ограничено из-за нарушенной проницаемости гематоэнцефалического барьера при ГИЭ. Рассмотрена возможность использования 6% гидроксиэтилкрахмала 130/0,42, поскольку последние исследования не продемонстрировали дополнительных рисков по сравнению с применением кристаллоидов. It remains a significant challenge for neonatal intensive care because of high mortality and neurological disabilities in children, despite some progress in diagnosis, monitoring and treatment. The processes of training air traffic controllers require the development of a quality management system for their training, which should allow to evaluate the results of their training. The main task of assessing the quality of training is to obtain the information necessary for proper conclusion on the success of their training course and assess the expected quality of their work. The article establishes that when training controllers, whose erroneous actions influence the performance of air traffic control tasks, “indirect indicators” are also used along with direct indicators – indicators of the body’s physiological “price” for the results achieved. The choice of indirect indicators is due to the nature of the activity of the controllers. Citicoline was administered basingon simple randomization in dose of 200 mg once daily intravenously for the first 5 days of treatment. Endpoints included indicators such as duration of positive pressure ventilation, total days of intensive care and the morbidity of cerebral leukomalacia, as well as frequency of the development of seizures. Seasonal migrations and herd instinct are traditionally recognized as wild reindeer (Rangifer tarandus L.) species-specific behavioral signs. These animals are forced to overcome water obstacles during the migrations. Behaviour peculiarities are considered as the result of the selection process, which has chosen among the sets of strategies, as the only evolutionarily stable one, determining the reproduction and biological survival of wild reindeer as a species. Natural processes in the Taimyr population wild reindeer are currently occurring against the background of an increase in the influence of negative factors due to the escalation of the industrial development of the Arctic. That is why the need to identify the ethological features of these animals completely arose. This paper presents the results of applying the classical methods of the theory of optimal control and differential games to the wild reindeer study of the migration patterns in overcoming water barriers, including major rivers. Based on these animals' ethological features and behavior forms, the herd is presented as a controlled dynamic system, which presents also two classes of individuals: the leader and the rest of the herd, for which their models, describing the trajectories of their movement, are constructed. The models are based on hypotheses, which are the mathematical formalization of some animal behavior patterns. This approach made it possible to find the trajectory of the important one using the methods of the optimal control theory, and in constructing the trajectories of other individuals, apply the principle of control with a guide. Approbation of the obtained results, which can be used in the formation of a common "platform" for the adaptive behavior models systematic construction and as a reserve for the cognitive evolution models fundamental development, is numerically carried out using a model example with observational data on the Werchnyaya Taimyra River. © 2019 Natalia V. Die Studie zeichnet das Gruppenportrait von acht der jüngsten Bundestagsabgeordneten von Bündnis 90/Die Grünen in der Wahlperiode von 1994 bis 1998. Für die Studie wurden biographische Interviews geführt. Das Gruppenportrait besteht aus drei Teilen. Der erste enthält die Motivationen und Wege, warum und in welchen Kontexten die Interviewten den Grünen beigetreten sind. Der zweite Teil verfolgt in acht Etappen den Weg in die Bundestagsfraktion, von der Entscheidung für die Kandidatur, die Listenparteitage ihrer Landesverbände, den Wahlkampf und den Wahltag, die Ankunft in Bonn, die konstituierende Sitzung der neuen Fraktion mit der Wahl des Fraktionsvorstandes, der Verteilung der Ausschüsse und der ersten Pressekonferenz des Vorstandes bis zum Beginn der politischen Arbeit der Fraktion. Der dritte Teil beschreibt ihre Haltung zum Bundeswehreinsatz in Bosnien. Dabei wird herausgearbeitet, wie sich ihre Position zum zentralen Konflikt der Bundesgrünen während der Wahlperiode wandelte. Zum Fazit gehört, dass die Unterschiede der Herkunft - sechs Abgeordnete kamen aus der alten Bundesrepublik, zwei aus der ehemaligen DDR - in einigen Punkten sehr ins Gewicht fallen. Auch bildeten die acht Abgeordneten keine Gruppe in dem Sinne, dass sie eine gemeinsame Haltung vertreten hätten. Wohl aber gab es eine Gemeinsamkeit hinsichtlich ihres Politikstils. Anstelle der früheren Symbolpolitik der Grünen ist ihr Politikstil pragmatischer und professioneller geworden. Insofern waren sie Protagonisten des beginnenden Generationswechsels bei den Bündnisgrünen.

В статье рассмотрены антенатальные и интранатальные факторы риска развития тяжелой гипоксически-ишемической энцефалопатии (ГИЭ) и их влияние на течение острого периода ГИЭ у доношенных новорожденных. Проанализированы данные 205 доношенных новорожденных, которые находились на лечении в отделении анестезиологии и интенсивной терапии новорожденных КУ «Днепропетровская областная детская клиническая больница» ДОС» в период 2009–2016 гг. Исследованы такие факторы риска, как пол, гестационный возраст, масса тела при рождении, первые или повторные роды, введение окситоцина, отслойка плаценты, кесарево сечение, дистресс плода, обвитие пуповины, наличие мекония в околоплодных водах, оценка по Апгар на 1, 5 и 20-й минуте после рождения, проведение неонатальной реанимации при рождении, а также в течение какого времени после родов дети поступали в отделение интенсивной терапии для проведения специализированного лечения (0–6 часов, 6–24 часа или 24–72 часа). В качестве конечных показателей использованы летальность, продолжительность проведения искусственной вентиляции легких и вообще респираторной поддержки (включая неинвазивную), продолжительность нахождения в отделении интенсивной терапии и в целом в больнице до выписки на амбулаторное лечение, развитие тяжелого осложнения в виде церебральной лейкомаляции., В статті розглянуто антенатальні та інтранатальні фактори ризику розвитку тяжкої гіпоксично-ішемічної енцефалопатії (ГІЕ) та їх вплив на перебіг гострого періоду ГІЕ у доношених новонароджених. Hypoxic ischemic encephalopathy (HIE), despite significant advances in monitoring technology and understanding fetal and neonatal pathology, is one of the most frequent causes of cerebral palsy and other severe neurological disorders in children. Analysis of severe HIE cases shows preconceptual and antepartum risk factors in 69 % of all newborns with moderate-to-severe neonatal encephalopathy. The purpose of the study was to investigate the antenatal and intranatal risk factors for the development of hypoxic ischemic encephalopathy in term newborns and their impact on the further course of acute period of HIE. Materials and methods. Analysis of the data of 205 term newborns was conducted. The average gestational age was 39.6 ± 1.4 (37–42) weeks, the birth weight was 3573 ± 549 (2440–5300) grams. 128 babies (62.4 %) were males and 77 (37.6 %) were females. Results. 47 (22.9 %) babies were admitted to the neonatal intensive care unit (NICU) in the first 6 hours of life, 136 (66.3 %) — in the 6–24 hours of life, 19 (9.3 %) — in 24–72 hours of life and 3 infants (1.5 %) were admitted after 72 hours. Mortality ratio on the 28th day of treatment was 3 of 205 babies (1.46 %). In 82 cases (40 %), there were first labors and in 123 (60 %) — second. The ratio of cesarean sections was 42 of 205 infants (20.5 %). From 42 births by caesarean section, 17 (40.5 %) were first deliveries and 25 (59.5 %) were second (p = 0.994). A significant difference was found on the Apgar score between the two groups on 1st minute after birth (3 (2–6) points in vaginal labors vs. 5 (3–7) points after caesa­rean section, p = 0.013), but no difference was found on the 5th minute after birth. Also, there was no significant difference in serum lactate level (6 (3.5–10.9) and 6.6 (3.9–14.9) mmol/l, respectively, p = 0.676), but in both groups, it doubled the normal range. It remains unclear if the serum concentrations of neuron-specific enolase (NSE) and protein S-100 correlate with severity and possible complications of hypoxic ischemic encephalopathy (HIE) as well as monitoring parameters of cerebral perfusion and central hemodynamics in term newborns.The aim of research. To determine diagnostic and predictable values of NSE and protein S-100 in acute period of severe hypoxic-ischemic encephalopathy in neonates.Materials and methods. Data of 89 term infants with Apgar score at birth of seven or less and Sarnat stage II-III was collected during ≤72 hours of life. The correlation between NSE, S-100 and Glasgow Coma Scale, seizures, confirmed by the aEEG, the unwanted development of neurological complications such as cerebral leukomalacia, and the short-term clinical results were analyzed.Result. In acute period of HIE the concentration of serum neuronal biomarkers S-100 and NSE proteins substantially exceeds the reference values for the first hours after labor and exceeds the normal range until the 5th day of life. The dynamics of these data significantly changes during this period. Mostly diagnostically and prognostically significant the measurement of biomarkers was on the 3rd day of treatment, immediately after the end of the therapeutic hypothermia, at the beginning of rewarming and brain reperfusion, when both indicators were most closely correlated with the assessment of the GCS: NSE (r=-0.3; p=0.002 ) and S-100 (r=-0.3; p=0.003). The value of S-100 on day 1 correlating with the following unwanted development of leukomalacia is above 1.8 μg/L (p=0.002), which is more than 3 times higher from the reference level of 0.51 μg/L given by Simon-Pimmel J. (2017) for term newborns.Conclusion. The evaluation of the serum concentrations of protein S-100 and neuron-specific enolase in the acute period of hypoxic ischemic encephalopathy in the term newborns has a significant diagnostic and predictable value and correlates with the severity, На сегодняшний день остается невыясненным соответствие абсолютных сывороточных концентраций нейрон-специфической энолазы (NSE) и протеина S-100 степени тяжести и возможным последствиям гипоксически-ишемической энцефалопатии (ГИЭ), а также мониторинговым показателям мозговой перфузии и центральной гемодинамики у новорожденных.Цель исследования. Определить диагностическую и прогностическую значимость NSE и S-100 в остром периоде ГИЭ у доношенных новорожденных.Материалы и методы. В исследование включено 89 доношенных новорожденных с оценкой по Апгар при рождении менее 7 баллов и Sarnat II-III вв. в сроке ≤72 часов после родов. Проанализирована связь показателей NSE и протеина S-100 с оценкой по шкале комы Глазго, возникновением судорог, подтвержденных по данным аЭЭГ, нежелательным развитием неврологических осложнений в виде церебральной лейкомаляции и конечными клиническими показателями эффективности лечения.Результаты. В остром периоде ГИЭ сывороточные концентрации биомаркеров поражения головного мозга протеина S-100 и NSE существенно превышают референтные значения с первых часов после рождения и остаются выше нормативных к 5м суткам жизни. Динамика данных показателей достоверно меняется в течение этого периода. Наиболее диагностически и прогностически значимыми оказались данные измерения биомаркеров на 3й день лечения, сразу после окончания лечебной гипотермии, начала согревания и реперфузии головного мозга, когда оба показателя более всего коррелировали с оценкой по ШКГ: как NSE (r=-0,3; p=0,002), так и S-100 (r=-0,3; p=0,003). Критической величиной S-100 в 1й день в отношении вероятного развития лейкомаляции является >1,8 мкг/л (р=0,002), что в 3 раза превышает контрольный уровень 0,51 мкг/л по данным Simon-Pimmel J. (2017) для доношенных новорожденных.Выводы. Досліджено 89 доношених новонароджених з оцінкою по Апгар при народженні менше 7 балів та Sarnat II-III ст. в терміні ≤72 годин після пологів. Проаналізований зв’язок показників NSE та протеїну S-100 з оцінкою за шкалою коми Глазго, виникненням судом, підтверджених за даними аЕЕГ, небажаним розвитком неврологічних ускладнень у вигляді церебральної лейкомаляції, та кінцевими клінічними показниками ефективності лікування.Результати. В гострому періоді ГІЕ сироваткові концентрації біомаркерів ураження головного мозку протеїну S-100 та NSE істотно перевищують референтні значення з перших годин після народження і залишаються вище нормативних до 5ї доби життя. Динаміка даних показників достовірно змінюється протягом цього періоду. Найбільш діагностично і прогностично значущими виявились дані вимірювання біомаркерів на 3й день лікування, одразу після закінчення лікувальної гіпотермії, початку зігрівання та реперфузії головного мозку, коли обидва показники найбільше корелювали з оцінкою за ШКГ: як NSE (r=-0,3; p=0,002), так і S-100 (r=-0,3; p=0,003). Критичною величиною S-100 у 1й день щодо вірогідного розвитку лейкомаляції є показник >1,8 мкг/л (р=0,002), що понад у 3 рази перевищує контрольний рівень 0,51 мкг/л, наведений Simon-Pimmel J. (2017) для доношених новонароджених.Висновки. Method for low-temperature (<200°C) deposition of SiCN-based coatings in a large volume chamber (0.3 m 3 ) by decomposition of organosilicon precursor in nitrogen-argon plasma of discharge with self-heated hollow cathode and remote anode (40 cm) at pressure of ∼1 mTorr was investigated. The analysis of the composition of coatings by IR spectroscopy and the analysis of plasma composition are carried out by optical emission spectroscopy. The influence of discharge current (10-40 A) and the flow of gas mixture (5-15 sccm hexamethyldisilazane and 60 sccm Ar-N 2 mixture) on plasma composition and elemental composition of coatings was investigated. It is shown that the degree of decomposition of precursor molecules is increased with growth of the current and with increase of Ar content in the gas mixture. © 2018 Web Portal IOP. In this case the image of the city is studied as an element and reason of urban cultural processes. The scientifi c fi xing of dynamic components of the image became available after the emergence of technologies of analysis like big data. Special methods are promoting transformation from qualitative data to quantitative data. Visualization of dynamic model of perception is becoming an effective tool for monitoring, prediction of changes, formation and adjustment of cultural processes.

Стаття присвячена вивченню особливостей розвитку та перебігу різних форм бронхолегеневої дисплазії у недоношених новонароджених з синдромом дихальних розладів та відкритою артеріальною протокою, особливостям інтенсивної терапії.Вивчено вплив різних строків закриття артеріальної протоки на розвиток та ступінь тяжкості БЛД. Доведено, що при фармакологічному закритті ВАП у ранній термін (до 3 діб) ризик розвитку БЛД мінімальний. Також доведено, що при пізньому закритті ВАП, особливо при хірургічному клемування протоки, істотно зростає ризик розвитку тяжких та середньо-тяжких форм БЛД. Вперше використана та доведена безпека та ефективність ректальної форми ібупрофену для фармакологічного закриття ВАП. Визначено вплив різних об'ємів інфузійної терапії на термін закриття ВАП та на розвиток БЛД та її тяжких форм. The necessity of antenatal and intrapartum factors analysis in preterm infants due to risk of BPD formation was established. Timely analysis of BPD predictors allows you to make more precise prognosis of BPD development and start preventive therapy as early as possible. The effect of different timing of the closure of the ductus arteriosus in the development and severity of BPD was studied. It was proven that the pharmacological closure of PDA in the early stages (up to 3 days), significantly decreases the risk of BPD formation. The late closing of PDA, especially in cases of surgical clipping, increases the risk of severe and moderate-severe BPD. For the first time we have used the rectal forms of ibuprofen for pharmacological PDA closure. Its safety and efficiency have been proven. The effect of different amounts of infusion therapy for PDA closing and BPD formation was defined. Taking into account the correlations it was shown that the using of volume intake restriction increases the effectiveness of COX to PDA closure in the early stages and prevents the development of BPD or improves its course.

We investigate an ill-posed Cauchy problem for a linear functional differential equation of retarded type on the negative half-line. Using a step-by-step procedure this problem is replaced by an inverse problem for an operator equation of the first kind in a functional space. The Tikhonov’s method is then used for finding solutions. We also construct special boundary value problems for the functional differential equations. The physical nature of quasi-static and transient anomalies in the geomagnetic field induced by underground explosions or earthquakes is reviewed. New theoretical results obtained recently and so far little known to general circles of geophysicists are presented. The physical nature of residual magnetic and electrotelluric fields at the explosion point are considered. The seismic waves from explosions or far earthquakes are suggested to be used as a tool for the preliminary probing of the Earth's crust sensitivity to various seismo-electromagnetic effects. The use of induction effects for the tsunami detection and for the crust sounding is outlined. Translation initiation factor 1 (IF1) is an essential protein in prokaryotes. The nature of IF1 interactions with the mRNA during translation initiation on the ribosome remains unclear, even though the factor has several known functions, one of them being RNA chaperone activity. In this study, we analyzed translational gene expression mutant variants of IF1 with amino acid substitutions, R40D and R69L, using two different reporter gene systems. The strains with them mutant IF1 gave higher reporter gene expression than the control strain. The extent of this effect was dependent on the composition of the translation initiation region. The Shine–Dalgarno (SD) sequence, AU-rich elements upstream of the SD sequence and the region between the SD sequence and the initiation codon are important for the magnitude of this effect. The data suggest that the wild-type form of IF1 has a translation initiation region-dependent inhibitory effect on translation initiation. Kasugamycin is an antibiotic that blocks translation initiation. Addition of kasugamycin to growing wild-type cells increases reporter gene expression in a very similar way to the altered IF1, suggesting that the and kasugamycin affect some related step in translation initiation. Genetic knockout of three proteins (YggJ, BipA, and CspA) that are known to interact with RNA causes partial suppression of the IF1-dependent cold sensitivity.

Translation initiation factor 1 (IF1) is an essential bacterial protein, without any clearly defined function in translation initiation. Same point mutants in this protein exhibit cold-sensitivity. In addition, a sequence specific increase in test protein expression has been observed for these mutants. Here in Paper I we analyze the parts of mRNA translation initiation region (TIR) involved in this effect, showing that Shine-Dalgarno sequence (SD), upstream enhancer region as well as linker between the initiation codon and SD are important. The similarity in action between the mutated IF1 and the antibiotic kasugamycin leads us to the suggestion that it occurs during the recently discovered proof-reading stage after the subunit joining step in the translation initiation. Deletion of yggJ and cspA genes involved in mRNA translation partially suppresses the cold-sensitivity phenotype of the R40D IF1 mutant. Deletion of the bipA gene confers even higher suppression confirming the previously assigned function of this protein in initiation of translation. In Paper II of this thesis proteome analysis of the IF1 mutation and kasugamycin action reveals some interesting features, such as increase in S6 polyglutamylation in the IF1 mutant cells or high level of GroEL-GroES protein in the cells treated with kasugamycin. A subset of genes having a similar TIR-specific increase in the expression under both conditions confirms previously made observations. Finally, in the Paper III we use cryo-electron tomography as well as chemical probing and computational modeling to address the question, how tmRNA moves on the ribosome during trans-translation. Trans-translation is a process which switches the synthesis of a polypeptide chain encoded by a nonstop messenger RNA to the mRNA-like domain of a transfer-messenger RNA (tmRNA). It is used in bacterial cells for rescuing the ribosomes arrested during translation of damaged mRNA and directing this mRNA and the product polypeptide for degradation. The molecular basis of this process is not well understood. Earlier, we developed an approach that allowed isolation of tmRNA-ribosomal complexes arrested at a desired step of tmRNA passage through the ribosome. We have here exploited it to examine the tmRNA structure using chemical probing and cryo-electron microscopy tomography. During the last five years there has been considerable doubt over the age of the continental uppermost Permian Russian stages, the Kazanian and Tatarian. Traditionally they have been regarded as Late Permian but were re-dated as Middle Permian in the 2004 international time scale, despite fossil evidence that the Tatarian, at least, is Late Permian. These debated ages are tested by magnetostratigraphic study of five sections spanning the Permian Triassic Boundary (PTB) of the SE Urals in the Orenburg region of Russia. The Upper Permian and Lower Triassic of this region have a well documented vertebrate fauna whose evolution has a significant bearing on our understanding of the PTB mass extinction event. If the Tatarian is viewed as Mid Permian, then the Late Permian in Russia is marked by a 9–10 Ma stratigraphic gap. The palaeomagnetic data yield a distinct series of polarity zones that provide clear local and regional correlation and are readily tied to a recently compiled global magnetostratigraphic record. On the basis of this correlation the sampled sections span the upper Guadalupian to Induan stages without any obvious break, so confirming the traditional view that the Tatarian is Late Permian in age. Anomalies in the magnetic inclination are consistent with sediment compaction (inclination shallowing, a common phenomenon of red beds) but declination anomalies between these sites and elsewhere in Russia may suggest localised vertical axis rotation.

Substantial advances have been made on the Mega AmpÚre Spherical Tokamak (MAST). The parameter range of the MAST confinement database has been extended and it now also includes pellet-fuelled discharges. Good pellet retention has been observed in H-mode discharges without triggering an ELM or an H/L transition during peripheral ablation of low speed pellets. Co-ordinated studies on MAST and DIII-D demonstrate a strong link between the aspect ratio and the beta scaling of H-mode energy confinement, consistent with that obtained when MAST data were merged with a subset of the ITPA database. Electron and ion ITBs are readily formed and their evolution has been investigated. Electron and ion thermal diffusivities have been reduced to values close to the ion neoclassical level. Error field correction coils have been used to determine the locked mode threshold scaling which is comparable to that in conventional aspect ratio tokamaks. The impact of plasma rotation on sawteeth has been investigated and the results have been well-modelled using the MISHKA-F code. Alfvén cascades have been observed in discharges with reversed magnetic shear. Measurements during off-axis NBCD and heating are consistent with classical fast ion modelling and indicate efficient heating and significant driven current. Central electron Bernstein wave heating has been observed via the O-X-B mode conversion process in special magnetically compressed plasmas. Plasmas with low pedestal collisionality have been established and further insight has been gained into the characteristics of filamentary structures at the plasma edge. Keywords: quarantine pest, plant health policy, optimization, import phytosanitary inspection, ‘reduced checks’, optimal allocation of resources, multinomial logistic regression, the Netherlands World trade is a major vector of spread of quarantine plant pests. Border phytosanitary inspection is a major barrier against introductions of quarantine pests through imported commodities, although the inspection resources are limited. This thesis provides conceptual and empirical insights that may help optimise import inspection under limited resources. The developed conceptual models analysed inspection policies under capacity constraints and in the absence of capacity constraints. The empirical models focused on finding the optimal inspection policies of propagating materials imported in the Netherlands. The results indicate that inspection effort should focus on commodities, whose inspection yields ceteris paribus greater marginal reduction in the expected costs of pest introduction. The results show that under binding capacity constraints, inspection of chrysanthemum cuttings in the Netherlands has a high marginal benefit, ranging from 8 to 49 euros for every marginal euro of inspection cost. The results further indicate that in the presence of fixed inspection costs, attaining the unconstrained allocation of inspection effort from the current, capacity constrained levels, is relatively inexpensive and greatly reduces costs to society. To expand current inspection capacities, the costs and likelihoods of pest introduction should be carefully estimated. Using the developed models for optimal allocation of inspection resources, the efficacy of the ‘reduced checks’ import inspection system in the EU was analysed. The results indicate that the expected costs of pest introduction in the EU under reduced checks could further be reduced if the economic impacts of pest introduction through various commodities are accounted for when calculating the frequencies of reduced checks

Optimal allocation of available resources to minimize quarantine risks related to international trade is a problem facing plant protection agencies worldwide. In this paper a model of budget allocation to minimize quarantine risks is developed. Theoretical conditions that budget allocation should satisfy are derived. These conditions imply that optimal allocation of resources is achieved when the marginal pest risks are equalized across risky pathways. Furthermore, an empirical model of budget distribution is developed. In the empirical model, the protecting agency wants to minimize the expected number of infested ornamental plants imported in a given country. The model is parameterized using data on import of ornamental commodities, the associated quarantine risks and costs of import phytosanitary inspections pertaining to the Netherlands. The results of the empirical model suggest that under specific assumptions (such as constant risk) greater risk reduction can be achieved by allocating larger funds to inspection of riskier pathways, and less or no funds to less risky pathways. It is well known that the Black-Scholes-Merton model suffers from several deficiencies. Jump-diffusion and Levy models have been widely used to partially alleviate some of the biases inherent in this classical model. Unfortunately, the resulting pricing problem requires solving a more difficult partial-integro differential equation (PIDE) and although several approaches for solving the PIDE have been suggested in the literature, none are entirely satisfactory. All treat the integral and diffusive terms asymmetrically and are difficult to extend to higher dimensions. We present a new, efficient algorithm, based on transform methods, which symmetrically treats the diffusive and integrals terms, is applicable to a wide class of path-dependent options (such as Bermudan, barrier, and shout options) and options on multiple assets, and naturally extends to regime-switching Levy models. We present a concise study of the precision and convergence properties of our algorithm for several classes of options and Levy models and demonstrate that the algorithm is second-order in space and first-order in time for path-dependent options.

The distribution of radioactive contamination at three island sites downstream from the Krasnoyarsk Mining and Chemical Combine (KMCC) was studied with the objectives of mapping contamination levels, interpreting radionuclide distributions through consideration of alluvial processes and determining radionuclide inventories. Contamination was measured using in situ gamma spectrometry and landforms characterised using topographic surveying methods. Maximum 137Cs contamination densities (700 kBq m−2) were found on low- and middle-level floodplains and low-lying interconnecting areas of Beriozovy Island (16 km from the KMCC). On Mikhin Island (180 km from the KMCC) maximum total 60Co, 152Eu and 154Eu activity concentrations (30–40 kBq m−2) occur in low-lying areas inundated during flooding. Maximum 137Cs and total 60Co, 152Eu and 154Eu contamination densities on Cheriomukhov Island (250 km from the KMCC) were 390 and 50 kBq m−2, respectively. Estimated 137Cs inventories were 145, 148 and 16 GBq for Beriozovy, Mikhin and Cheriomukhov Islands, respectively.

The thickness of coastal landfast ice in the Sea of Okhotsk has been examined based on field data and thermodynamic modelling. The study sites were Saroma-ko Lagoon, Hokkaido and Kleye Strait, Sakhalin. The ice sheet has a two-layer structure: a granular snow-ice layer on top and a columnar ice layer below. In Saroma-ko Lagoon the ice grows to 40-50 cm, with snow-ice portion of 10-100%. In Kleye Strait the ice grows to about 100 cm, with a remarkable addition (on average 24 cm) during mid-March to mid-April due to snow-ice formation. A one-dimensional thermodynamic ice-snow model was calibrated with observed data and used to examine the thickness climatology; the snow component takes into account snow compaction, slush formation due to flooding, melting or rain, and snow-ice growth. The model outcome showed reasonably good agreement for both sites. In Saroma-ko Lagoon the calibration was based on four winters. The maximum annual ice thickness was in the model on average 3 cm below the observed one, 16 cm in the worst case; the model snow thickness was within 10 cm from the observed ones in February; and the date of ice breakup was on average biased late by 5 days and 11 days in the worst case. The model simulations predicted formation of slush layers and their persistency for 1-4 weeks in different winters. Climatological simulation resulted in mean maximum annual ice thickness of 32 cm, of which 15 cm was snow-ice. In Kleye Strait the calibration was based on one ice season. The maximum annual ice thickness was 7 cm biased down, and the model snow thickness was within 10 cm from the observed level. Climatological simulation resulted in mean maximum annual ice thickness of 108 cm, of which 70 cm was congelation ice and 38 cm was snow-ice, and the ice season lasted from 5 November to 5 June. Thus slush formation and its freezing are crucial in the study basin.

Quarantine pests and diseases represent a significant threat to agricultural and horticultural production worldwide. Currently, it is recognised that international trade in agricultural products, and especially in plants and plant materials, is a major vector facilitating the spread of quarantine organisms. To provide the adequate level of phytosanitary protection, the responsible agencies should allocate their limited resources in the best way, so that phytosanitary risks associated with imported products are minimised. This is however a challenging task given the constantly growing volumes of imported products, their broad assortment and large number of exporting countries. This paper suggests an optimization framework that can be used for designing an optimal import inspection policy. A theoretical part of the paper considers the optimisation problem of the quarantine agency which is constructed as minimisation of the economic damage from pest introduction under given constraints. The latter are budget constraints and the acceptable level of risk (ALOR) of a given country. The results of the theoretical part of the paper present the conditions for the optimal allocation of the Agency¿s resources under constraints. In the second part of the paper a quantitative model for optimal allocation of resources for inspecting ornamental plants¿ imports to the Netherlands is presented. The model shows the optimal allocation of inspection resources given the expected phytosanitary risks associated with different commodities. More specifically, the model calculates the optimal inspection time and achieved level of quarantine protection under the varying levels of ALOR representing different risk attitudes of the protecting agencies. To the best of authors¿ knowledge both the setup and the results of the presented model represent a novel contribution in the field of economics of quarantine protection.

In Kleye Strait the ice grows to about 100 cm, with a remarkable addition (on average 24 cm) during mid-March to mid-April due to snow-ice formation. A one-dimensional thermodynamic ice-snow model was calibrated with observed data and used to examine the thickness climatology; the snow component takes into account snow compaction, slush formation due to flooding, melting or rain, and snow-ice growth. The model outcome showed reasonably good agreement for both sites. In Saroma-ko Lagoon the calibration was based on four winters. The maximum annual ice thickness was in the model on average 3 cm below the observed one, 16 cm in the worst case; the model snow thickness was within 10 cm from the observed ones in February; and the date of ice breakup was on average biased late by 5 days and 11 days in the worst case. The model simulations predicted formation of slush layers and their persistency for 1-4 weeks in different winters. Climatological simulation resulted in mean maximum annual ice thickness of 32 cm, of which 15 cm was snow-ice. In Kleye Strait the calibration was based on one ice season. The maximum annual ice thickness was 7 cm biased down, and the model snow thickness was within 10 cm from the observed level. Climatological simulation resulted in mean maximum annual ice thickness of 108 cm, of which 70 cm was congelation ice and 38 cm was snow-ice, and the ice season lasted from 5 November to 5 June. Thus slush formation and its freezing are crucial in the study basin.

We present optical integral field observations of the H II region containing the ultraluminous X-ray source Holmberg II X-1. We confirm the existence of an X-ray ionized nebula as the counterpart of the source due to the detection of an extended He II (4686A) region (21 x 47 pc) at the Chandra ACIS-S position. An extended blue objects with a size of 11 x 14 pc is coincident with the X-ray/He II region, which could indicate either a young stellar complex or a cluster. We have derived an X-ray to optical luminosity ratio of Lx/Lb>170, and presumable it is Lx/Lb~300-400 using the recent HST ACS data. We find a complex velocity dispersion at the position of the ULX. In addition, there is a radial velocity variation in the X-ray ionized region found in the He II emission of +-50 km/s on spatial scales of 2-3 arcsec. We believe that the putative black hole not only ionizes the surrounding HII gas, but also perturbs it dynamically (via jets or the accretion disk wind). The spatial analysis of the public Chandra ACIS-S data reveals a point-like X-ray source and marginal indication of an extended component (<<15 % of the total flux). The XMM-Newton EPIC-PN spectrum of HoII X-1 is best fitted with an absorbed power-law in addition to either a thermal thick plasma or a thermal thin plasma or a multi-colour disk black body (MCD). In all cases, the thermal component shows relatively low temperatures (kT~0.14-0.22 keV). Application of the digital modeling method in the design of digital computer circuits is proposed for calculation of the probabilities of reductions and multiplicity factors of errors with parallel data input. The use of the method for constant perturbation is explained by way of example of a trigger on semiconductor triodes. The advantages of the method in comparison with analysis of the performance of circuits under the influence of a set of interference sources are examined. In the overall digital computer reliability estimates the repairability factors such as the detection, removal, and prevention of failures must be included in the analysis. These in turn do not depend on the computer system organization above, but also on the capability of the maintenance personnel. The design of a computer system with a specific repairability figure is difficult since the statistical data for the new system is not available a priori. The authors make an attempt to correlate certain experience gained during the operation of a Ural-2 computer with the repairability design parameters for inclusion in future designs. Thus, the computer availability time is related to the mean restorability time which in turn is shown to depend on a number of factors: computer functional organization (i.e. whether provisions are made for executing test programs), amount of equipment redundancy and built in control circuitry, location of fault indicators, structural organization (i.e. whether the structure is modular and access to measuring and test equipment is easy), organization of maintenance routines and schedules, and the qualification of maintenance personnel. © 2016 The Author(s)Previously, we showed that CD206-targeted liposomal delivery of co-encapsulated immunodominant myelin basic protein (MBP) sequences MBP46–62, MBP124–139 and MBP147–170 (Xemys) suppressed experimental autoimmune encephalomyelitis in dark Agouti rats. The objective of this study was to assess the safety of Xemys in the treatment of patients with relapsing-remitting multiple sclerosis (MS) and secondary progressive MS, who failed to achieve a sustained response to first-line disease-modifying therapies. In this phase I, open-label, dose-escalating, proof-of-concept study, 20 patients with relapsing-remitting or secondary progressive MS received weekly subcutaneously injections with ascending doses of Xemys up to a total dose of 2.675 mg. Clinical examinations, including Expanded Disability Status Scale score, magnetic resonance imaging results, and serum cytokine concentrations, were assessed before the first injection and for up to 17 weeks after the final injection. Xemys was safe and well tolerated when administered for 6 weeks to a maximum single dose of 900 μg. Expanded Disability Status Scale scores and numbers of T2-weighted and new gadolinium-enhancing lesions on magnetic resonance imaging were statistically unchanged at study exit compared with baseline; nonetheless, the increase of number of active gadolinium-enhancing lesions on weeks 7 and 10 in comparison with baseline was statistically significant. During treatment, the serum concentrations of the cytokines monocyte chemoattractant protein-1, macrophage inflammatory protein-1β, and interleukin-7 decreased, whereas the level of tumor necrosis factor-α increased. These results provide evidence for the further development of Xemys as an antigen-specific, disease-modifying therapy for patients with MS.
