1. Christian theology emerging from the Akan single-tiered unitive perspective on reality

Abstract: My thesis proposes using the Akan single-tiered unitive perspective on reality in two ways: first, as a hermeneutical tool for reading and understanding African Christian theologies,* and second, as a means for opening up space in global conversations between African and western theologians on the relationship between the seen and unseen, the material and spiritual, and on the perceived fact-faith divide in the contemporary West. I have developed the term ‘single-tiered unitive perspective on reality’ to designate the idea, present in the literature under various other names, that the material and spiritual exist inseparably in an overlapping, permeable way that creates an interpretive landscape for the reading and writing of African Christian theologies. In considering the interpretive landscape which emerges from this single-tiered unitive perspective on reality, two terms become especially useful. The first is Andrew Walls’ ‘maps of the universe’, and the second is Kwesi Dickson’s ‘religio-culture’. Within the context of the single-tiered unitive perspective, these conceptual frames of reference provide us with secondary interpretive tools for analysing the work of the mostly Akan scholars and ideas discussed in this thesis, —particularly Kwame Bediako and his three-pronged use of identity as a new theological category. The single-tiered unitive perspective also provides the interpretive landscape—and various categories that are important in the make-up of that landscape – for what I am calling ‘analogical continuity’, which takes advantage of particular analogical ‘landing places’, or ‘bridges’ between the primal Akan and biblical ‘maps’ or conceptions of reality. I suggest that ‘analogical continuity’ offers a new way through the continuity-discontinuity debate by locating continuity at the level of the single-tiered perspective. Finally, use of the single-tiered unitive perspective sheds light on the contribution Akan theologians are making to the global theological conversation in two important areas: Christology and biblical anthropology. In these two areas I bring various Akan scholars into conversation with each other, using the interpretive lens of the single-tiered perspective to demonstrate where and why they differ even as they work from a common interpretive perspective on reality. *I use ‘African’ to refer to sub-Saharan Africa, with particular attention to Ghana.

2. National identity, propaganda, and the ethics of war in English historical literature, 1327-77

Abstract: This thesis argues against the common assumption that English writers ignored the ethical problems of war during the particularly brutal wars of Edward III, king of England, 1327–77. English historical literature in this period is typically mined for ‘facts’ to create visions of the past, or read as literature with little context, but never properly considered for its engagement with the morality of warfare. Chapter One shows that the many uncertain aspects of war, such as intention, are those that most affect how military acts are judged. Chapter Two argues that writers use theo-retical frameworks in a more nuanced and rhetorical sense than commonly believed. Chapter Three argues against the common belief that there was no concept of civilian immunity in the period, and demonstrates how writers present these civilian victims in different ways to attach moral value to those who attack them. Chapter Four examines how writers show the English to only attack enemy civilians, in their campaigns of devastation on a day-to-day basis, to force the enemy to do battle, and thereby end war. Chapter Five shows that writers avoid any of the morally unsavoury aspects of violence but revel in the suffering endured by their own men as meritorious asceticism. Chapter Six assesses how writers engage with the difficulties of ending hostilities and offering mercy, especially when martial culture encouraged bellicosity and vengeance. The thesis focuses throughout on the often nuanced and sensitive perspectives of English writers in this period before the age of Chaucer. The Appendix introduces each of the main sources used throughout this thesis and provides a detailed list of their manuscripts. The many errors and poor descriptions repeated in scholarship are corrected throughout. Several previously unidentified manuscripts, variant versions, and previously unknown texts have been described.

3. Design of Stickbug: a Six-Armed Precision Pollination Robot

Abstract: This work presents the design of Stickbug, a six-armed, multi-agent, precision pollination robot that combines the accuracy of single-agent systems with swarm parallelization in greenhouses. Precision pollination robots have often been proposed to offset the effects of a decreasing population of natural pollinators, but they frequently lack the required parallelization and scalability. Stickbug achieves this by allowing each arm and drive base to act as an individual agent, significantly reducing planning complexity. Stickbug uses a compact holonomic Kiwi drive to navigate narrow greenhouse rows, a tall mast to support multiple manipulators and reach plant heights, a detection model and classifier to identify Bramble flowers, and a felt-tipped end-effector for contact-based pollination. Initial experimental validation demonstrates that Stickbug can attempt over 1.5 pollinations per minute with a 50% success rate. 4. An investigation into the role of the hyperpolarisation-activated cyclic nucleotide-gated on channels in dorsal root ganglion neurons in rat models of chronic inflammatory and neuropathic pain

Abstract: Chronic pain {CP} is a major health problem that affects about 20% of adults worldwide. Unlike acute, physiological pain, which resolves promptly once the painful stimulus is removed, CP can last for months, years or even a lifetime. CP, which includes inflammatory pain {IP} that is associated with both tissue injury and the accompanying inflammation and peripheral neuropathic pain {NP} that is a direct consequence of a lesion or disease affecting the peripheral nervous system {PNS}, often is a constant burden, degrades peoples’ quality of life, and costs billions of pounds. Patients with CP usually complain of either: a) continuous or intermittent spontaneous, un-provoked pain; or b) hypersensitivity, due to either increased pain from a stimulus that normally provokes pain or pain due to a stimulus that does not normally provoke pain. Successful therapy for CP, particularly NP, remains a challenge because the currently available drugs are largely ineffective and many result in adverse side effects. Therefore, there is a pressing need to understand the pathophysiology of CP, in order to devise appropriate palliative and curative strategies. CP is believed to be due, at least partly, to increased excitability of normally quiescent dorsal root ganglion {DRG} neurons, which convey sensory information from the periphery to the central nervous system {CNS}. However, the underlying ionic and molecular mechanisms of this neuronal hyperexcitability and spontaneous activity {SA} are poorly understood. The aim of this research project was to examine the hypothesis that during peripheral CP states, hyperexcitability in DRG neurons could be due to increased expression of hyperpolarisation-activated cyclic nucleotide-gated {HCN} channels, possibly in combination with a change in their activation properties. This is because these channels, which are composed of 4 subunits {HCN1-4}, produce an excitatory inward current, termed the hyperpolarisation-activated {Ih} current in neurons, that depolarizes the membrane potential toward the threshold of action potential {AP} generation. To test this hypothesis, several integrated approaches, including behavioural pharmacology, in vivo electrophysiology, and immunofluorescent staining, were used in two rat models of CP that were compared to appropriate controls. The rat models of CP were: a) chronic inflammatory pain {CIP} model, which involved induction of hindlimb inflammation with complete Freund’s adjuvant {CFA}; and b) chronic neuropathic pain {CNP} model that involved L5 spinal nerve {SN} axotomy, in addition to loose ligation of the L4 SN with neuro-inflammation inducing chromic gut, referred to as modified SN Axotomy {mSNA}. The objectives of the current project were to: (i) evaluate, using behavioural pharmacology, the influence of modulating the HCN channels with the Ih-specific blocker, ZD7288, on pain hypersensitivity in both CIP and CNP. (ii) determine, using in vivo intracellular voltage and current recordings, the difference in AP parameters between normal and mSNA-treated L4 DRG neurons and the effect of ZD7288 on SA in the mSNA-treated L4 DRG neurons. (iii) determine, using immunofluoresence, the types of DRG neuron that express HCN1-HCN3 subunits in normal rats and whether expression of these subunits is altered in both CIP and CNP. The results showed: 1. In both CIP and CNP, peripheral administration of ZD7288 resulted in significant attenuation of mechanical hypersensitivity and a non-significant absence of spontaneous pain {SP}. 2. In the L4 DRG neurons of mSNA-treated animals with CNP, ZD7288 had no effect on the frequency of SA from low-threshold mechanoreceptors {LTM} and induced changes in hyperpolarisation-associated AP parameters in Aα/β-fibre DRG neurons. 3. In both CIP and CNP, an increased proportion of small and medium sized DRG neurons express HCN2, but not HCN1 or HCN3, channel protein. Taken together, the findings suggest that HCN channels, particularly HCN2, in specific sub-populations of DRG neurons contribute to the development!of CIP and CNP.

Abstract: Flare frequency distributions represent a key approach to addressing one of the largest problems in solar and stellar physics: determining the mechanism that counter-intuitively heats coronae to temperatures that are orders of magnitude hotter than the corresponding photospheres. It is widely accepted that the magnetic field is responsible for the heating, but there are two competing mechanisms that could explain it: nanoflares or Alfv\'en waves. To date, neither can be directly observed. Nanoflares are, by definition, extremely small, but their aggregate energy release could represent a substantial heating mechanism, presuming they are sufficiently abundant. One way to test this presumption is via the flare frequency distribution, which describes how often flares of various energies occur. If the slope of the power law fitting the flare frequency distribution is above a critical threshold, $\alpha=2$ as established in prior literature, then there should be a sufficient abundance of nanoflares to explain coronal heating. We performed $>$600 case studies of solar flares, made possible by an unprecedented number of data analysts via three semesters of an undergraduate physics laboratory course. This allowed us to include two crucial, but nontrivial, analysis methods: pre-flare baseline subtraction and computation of the flare energy, which requires determining flare start and stop times. We aggregated the results of these analyses into a statistical study to determine that $\alpha = 1.63 \pm 0.03$. Abstract: This paper presents the PAVICS-Hydro platform, the result of long-term strategic considerations and compromises between functionality and ease-of-use, which is developed to provide users with direct access to a comprehensive hydroclimatic modeling and analysis framework over North America. This includes a Python hydrological modelling package, efficient access to meteorological data, a large array of post-processed hydroclimatic data, numerical weather forecasts and bias-corrected CMIP6 climate projections at the scale of 5797 catchments, thus providing the basic components necessary for hydrological modeling and forecasting, as well as climate change impact studies. This paper presents the principles on which the PAVICS-Hydro platform was developed while explaining the choices behind its key features, with a specific focus on lessons learned for creators of the next generations’ scientific research platforms. A case study using PAVICS-Hydro is given to illustrate how this platform can be used to support different needs in the field of water resources.

Abstract: xclim is a Python library that enables computation of climate indicators over large, hetero- geneous data sets. It is built using xarray objects and operations, can seamlessly benefit from the parallelization handling provided by dask, and relies on community conventions for data formatting and metadata attributes. xclim is meant as a tool to facilitate both climate science research and the delivery of operational climate services and products. In addition to climate indicator calculations, xclim also includes utilities for bias correction and statistical adjustment, ensemble analytics, model diagnostics, data quality assurance, and metadata standards compliance.

9. Swarm of One: Bottom-up Emergence of Stable Robot Bodies from Identical Cells

Abstract: Unlike most human-engineered systems, biological systems are emergent from low-level interactions, allowing much broader diversity and superior adaptation to the complex environments. Inspired by the process of morphogenesis in nature, a bottom-up design approach for robot morphology is proposed to treat a robot's body as an emergent response to underlying processes rather than a predefined shape. This paper presents Loopy, a "Swarm-of-One" polymorphic robot testbed that can be viewed simultaneously as a robotic swarm and a single robot. Loopy's shape is determined jointly by self-organization and morphological computing using physically linked homogeneous cells. Experimental results show that Loopy can form symmetric shapes consisting of lobes. Using the the same set of parameters, even small amounts of initial noise can change the number of lobes formed. However, once in a stable configuration, Loopy has an "inertia" to transfiguring in response to dynamic parameters. Abstract: Mapping the entire frequency bandwidth of brain electrophysiological signals is of paramount importance for understanding physiological and pathological states. The ability to record simultaneously DC-shifts, infraslow oscillations (<0.1 Hz), typical local field potentials (0.1–80 Hz) and higher frequencies (80–600 Hz) using the same recording site would particularly benefit preclinical epilepsy research and could provide clinical biomarkers for improved seizure onset zone delineation. However, commonly used metal microelectrode technology suffers from instabilities that hamper the high fidelity of DC-coupled recordings, which are needed to access signals of very low frequency. In this study we used flexible graphene depth neural probes (gDNPs), consisting of a linear array of graphene microtransistors, to concurrently record DC-shifts and high-frequency neuronal activity in awake rodents. We show here that gDNPs can reliably record and map with high spatial resolution seizures, pre-ictal DC-shifts and seizure-associated spreading depolarizations together with higher frequencies through the cortical laminae to the hippocampus in a mouse model of chemically induced seizures. Moreover, we demonstrate the functionality of chronically implanted devices over 10 weeks by recording with high fidelity spontaneous spike-wave discharges and associated infraslow oscillations in a rat model of absence epilepsy. Altogether, our work highlights the suitability of this technology for in vivo electrophysiology research, and in particular epilepsy research, by allowing stable and chronic DC-coupled recordings.

Approach. CSDs were reliably induced in awake nontransgenic mice using optogenetic methods. High-fidelity DC-coupled electrophysiological mapping of propagating CSDs was obtained using flexible arrays of graphene soultion-gated field-effect transistors (gSGFETs). Main results. Viral vectors targetted channelrhopsin expression in neurons of the motor cortex resulting in a transduction volume 1 mm3. 5-10 s of continous blue light stimulation induced CSD that propagated across the cortex at a velocity of 3.0 0.1 mm min-1. Graphene micro-transistor arrays enabled high-density mapping of infraslow activity correlated with neuronal activity suppression across multiple frequency bands during both CSD initiation and propagation. Localized differences in the CSD waveform could be detected and categorized into distinct clusters demonstrating the spatial resolution advantages of DC-coupled recordings. We exploited the reliable and repeatable induction of CSDs using this preparation to perform proof-of-principle pharmacological interrogation studies using NMDA antagonists. MK801 (3 mg kg-1) suppressed CSD induction and propagation, an effect mirrored, albeit transiently, by ketamine (15 mg kg-1), thus demonstrating this models' applicability as a preclinical drug screening platform. Finally, we report that CSDs could be detected through the skull using graphene micro-transistors, highlighting additional advantages and future applications of this technology. Significance. CSD is thought to contribute to the pathophysiology of several neurological diseases. 14. A catalogue of the Cybocephalidae (Coleoptera: Cucujoidea) of the world

Abstract: A world catalogue and bibliography of the family Cybocephalidae (Coleoptera) is presented. The catalogue contains 207 species within 16 genera. Each taxon is accompanied by a complete taxonomic history, including a full annotated synonymy with original references cited. The reversal of the recent synonymy of Cybocephalus brevis Grouvelle over Cybocephalus minimus Grouvelle is proposed. Cybocephalus aeneus Reiche is removed from synonymy with Cybocephalus micans Reitter and considered incertae sedis. The Cybocephalidae are a family of small predatory beetles found throughout the world (Smith 2020). While cybocephalids are typically extremely convex, shiny and dark in coloration, the defining characteristic of this group is their ability to contract their bodies to a greater or lesser degree, with many species capable of conglobulation (Fig. 1–10). These beetles are rarely collected due to their small size and the infrequency with which they are collected using traditional trapping techniques (i.e. MV/UV-light, malaise, pitfall, etc.). In most cases, cybocephalids are collected by sweeping, beating or simply gleaned by hand (Smith and Cave 2006). Edmund Reitter (1874) published the first catalogue, or at least a comprehensive list, of the cybocephalids of the world. At that time, there were 25 species known to science confined to one genus. At the time of this publication, there are a total of 207 species in 16 genera in the family worldwide, with 201 extant species in 15 genera and six extinct species in two genera.

21. Consolidation and other geotechnical properties of shales with respect to age and composition

Abstract: Studies of a wide selection of over consolidated, weak argillaceous rocks from major formations in the United Kingdom and North America have shown that the compaction history, coupled with the mineralogical composition have a decisive bearing on the nature of the material, both at depth and in the near surface zone. Current evidence indicates that maximum depths of burial of North American sediments are generally much greater than their British counterparts; infearing that overburden does not increase systematically with age. Furthermore major differences have also been observed in the mineralogy and geochemistry of these two groups. In particular, recalculated smectite formulae indicate the onset of the montmorillonite to illite transformation in the former sediments. Preferred orientation studies and electron microscopy have been used/to elucidate the clay microstructure, whereas exchangeable cations and pore water chemistry indicate possible interactions between clay, minerals. Consolidation studies to a pressure of 35000KN/m(^2) on both undisturbed and remoulded materials have led to a new interpretation of the stress-strain response, and in addition these tests have indicated the presence of diagenetic bonding in unweathered materials the strength of which is dependent upon the maximum depth of burial and mineral species present. Furthermore, since slaking and suction experiments frequently only detect this bonding in the Caroniferous materials, it has been inferred that mineral-mineral welding is present in these and that cat ionic bonding predominates in younger sediments. Consequently to avoid unforeseen engineering complications in the field caused by the subsequent destruction of the latter bond type by weathering agents it has been suggested that a combination of suction and consolidation tests should be performed on the shales in question.

Abstract: Decoding the functional connectivity of the nervous system is facilitated by transgenic methods that express a genetically encoded reporter or effector in specific neurons; however, most transgenic lines show broad spatiotemporal and cell-type expression. Increased specificity can be achieved using intersectional genetic methods which restrict reporter expression to cells that co-express multiple drivers, such as Gal4 and Cre. To facilitate intersectional targeting in zebrafish, we have generated more than 50 new Cre lines, and co-registered brain expression images with the Zebrafish Brain Browser, a cellular resolution atlas of 264 transgenic lines. Lines labeling neurons of interest can be identified using a web-browser to perform a 3D spatial search (zbbrowser.com). This resource facilitates the design of intersectional genetic experiments and will advance a wide range of precision circuit-mapping studies.

Abstract: When proteostasis becomes unbalanced, unfolded proteins can accumulate and aggregate. Here we report that the dye, tetraphenylethene maleimide (TPE-MI) can be used to measure cellular unfolded protein load. TPE-MI fluorescence is activated upon labelling free cysteine thiols, normally buried in the core of globular proteins that are exposed upon unfolding. Crucially TPE-MI does not become fluorescent when conjugated to soluble glutathione. We find that TPE-MI fluorescence is enhanced upon reaction with cellular proteomes under conditions promoting accumulation of unfolded proteins. TPE-MI reactivity can be used to track which proteins expose more cysteine residues under stress through proteomic analysis. We show that TPE-MI can report imbalances in proteostasis in induced pluripotent stem cell models of Huntington disease, as well as cells transfected with mutant Huntington exon 1 before the formation of visible aggregates. TPE-MI also detects protein damage following dihydroartemisinin treatment of the malaria parasites Plasmodium falciparum. TPE-MI therefore holds promise as a tool to probe proteostasis mechanisms in disease. © 2017 The Author(s).

Abstract: Accurate forecasting of precipitation phase and intensity was critical information for many of the Olympic venue managers during the Vancouver 2010 Olympic and Paralympic Winter Games. Precipitation forecasting was complicated because of the complex terrain and warm coastal weather conditions in the Whistler area of British Columbia, Canada. The goal of this study is to analyze the processes impacting precipitation phase and intensity during a winter weather storm associated with rain and snow over complex terrain. The storm occurred during the second day of the Olympics when the downhill ski event was scheduled. At 0000 UTC 14 February, 2 h after the onset of precipitation, a rapid cooling was observed at the surface instrumentation sites. Precipitation was reported for 8 h, which coincided with the creation of a nearly 0°C isothermal layer, as well as a shift of the valley flow from up valley to down valley. Widespread snow was reported on Whistler Mountain with periods of rain at the mountain base despite the expectation derived from synoptic-scale models (15-km grid spacing) that the strong warm advection would maintain temperatures above freezing. Various model predictions are compared with observations, and the processes influencing the temperature, wind, and precipitation types are discussed. Overall, this case study provided a well-observed scenario of winter storms associated with rain and snow over complex terrain.

Abstract: Background & aims: Information relating to the provision of HPN in New Zealand and Scotland has been published. 1 This paper expands that benchmarking exercise to examine the organisation, prevalence and referral pathways across several countries of the world. Methods: The ESPEN Home Artificial Nutrition Chronic Intestinal Failure working group agreed a pro forma and circulated a network of international colleagues. Results: Responses were received from 16 countries treating an estimated 9200 patients. Period prevalence figures ranged from 3.25 to 66 patients per million of the population. Eight countries (53%) had published HPN guidelines; of the 7 who did not, 4 (27%) had adopted guidelines such as ESPEN. Eleven (73%) had an HPN educational programme in place. The most common underlying disease for HPN in adults were ischaemia, Crohns' disease, motility disorders and cancer; and in children enteropathy, motility and inflammatory bowel disease (unknown in 4 countries). Conclusions: There is a wide range in HPN prevalence figures and the existence of organised care varies across the countries studied. Most countries either had developed their own clinical guidelines or standards or had adopted an international society guideline to improve patient care. It is recognised that several countries have under reported the HPN prevalence as registries are not fully available or used. Abstract: Background Many peptide-based cancer vaccines have been tested in clinical trials with a limited success, mostly due to difficulties associated with peptide stability and delivery, resulting in inefficient antigen presentation. Therefore, the development of suitable and efficient vaccine carrier systems remains a major challenge. Methods To address this issue, we have engineered polylactic-co-glycolic acid (PLGA) nanoparticles incorporating: (i) two MHC class I-restricted clinically-relevant peptides, (ii) a MHC class II-binding peptide, and (iii) a non-classical MHC class I-binding peptide. We formulated the nanoparticles utilizing a double emulsion-solvent evaporation technique and characterized their surface morphology, size, zeta potential and peptide content. We also loaded human and murine dendritic cells (DC) with the peptide-containing nanoparticles and determined their ability to present the encapsulated peptide antigens and to induce tumor-specific cytotoxic T lymphocytes (CTL) in vitro. Results We confirmed that the nanoparticles are not toxic to either mouse or human dendritic cells, and do not have any effect on the DC maturation. We also demonstrated a significantly enhanced presentation of the encapsulated peptides upon internalization of the nanoparticles by DC, and confirmed that the improved peptide presentation is actually associated with more efficient generation of peptide-specific CTL and T helper cell responses. Conclusion Encapsulating antigens in PLGA nanoparticles offers unique advantages such as higher efficiency of antigen loading, prolonged presentation of the antigens, prevention of peptide degradation, specific targeting of antigens to antigen presenting cells, improved shelf life of the antigens, and easy scale up for pharmaceutical production. Therefore, these findings are highly significant to the development of synthetic vaccines, and the induction of CTL for adoptive immunotherapy.

Abstract: En este libro se sostiene que nuestro ambiente construido se encuentra sometido al mito del "hombre-padrón" que se ha venido proyectando por siglos. Mientras tanto, más de la mitad de la población no corresponde a ese tipo ideal: las personas adultas mayores, aquellas con peso excesivo, las de estatura muy alta o muy baja (incluyendo a los niños), las que tienen discapacidades físicas y las mujeres en los últimos dos meses del embarazo. La arquitectura debe ser empleada como vehículo de integración que, democráticamente, permita a todos los ciudadanos usufructuar los espacios sin ningún tipo de restricciones. Deberíamos utilizar el poder de la arquitectura como elemento facilitador de la inserción social. En Latinoamérica, región con grandes problemas sociales y económicos, hablar de "diseño universal" puede sonar como novedad revolucionaria. Sin embargo, el proceso de accesibilidad, de proyectar un medio ambiente cada vez más amplio y menos restrictivo es una tendencia mundial, irreversible, como todo proceso que mejora la calidad de vida del ser humano. Diversos aspectos contribuyen a la instalación de un entorno más adecuado. La situación sociopolítica de cada región y la conciencia de que todos tienen "derecho a la ciudad" son factores que pueden facilitar la acción en pro de la accesibilidad. Es fundamental el apoyo de la legislación y de las normas, el desarrollo y la búsqueda de consenso en esas áreas, para fortalecer los avances y el cumplimiento de las directrices establecidas.

Abstract: CD4+ T cells with immune regulatory function can be either FOXP3+ or FOXP3-. We have previously shown that priming of naturally occurring TCR-peptide-reactive CD4+FOXP3- Treg specifically controls VÎ28.2+CD4+ T cells mediating EAE. However, the mechanism by which these Treg are primed to recognize their cognate antigenic determinant, which is derived from the TCRVÎ28.2-chain, is not known. In this study we show that APC derived from splenocytes of naÃ ̄ve mice are able to stimulate cloned CD4+ Treg in the absence of exogenous antigen, and their stimulation capacity is augmented during EAE. Among the APC populations, DC were the most efficient in stimulating the Treg. Stimulation of CD4+ Treg was dependent upon processing and presentation of TCR peptides from ingested VÎ28.2TCR+CD4 + T cells. Additionally, DC pulsed with TCR peptide or apoptotic VÎ28.2+ T cells were able to prime Treg in vivo and mediate protection from disease in a CD8-dependent fashion. These data highlight a novel mechanism for the priming of CD4+ Treg by CD8Î±+ DC and suggest a pathway that can be exploited to prime antigen-specific regulation of T-cell-mediated inflammatory disease. Abstract: The −1p/−19q genotype predicts chemosensitivity in oligodendroglial neoplasms, but some with intact 1p/19q also respond and not all with 1p/19q loss derive durable benefit from chemotherapy. We have evaluated the predictive and prognostic significance of pretherapy 201Tl and 18F-FDG SPECT and genotype in 38 primary and 10 recurrent oligodendroglial neoplasms following PCV chemotherapy. 1p/19q loss was seen in 8/15 OII, 6/15 OAII, 7/7 OIII, 3/11 OAIII and was associated with response (Fisher-Exact: P=0.000) and prolonged progression-free (log-rank: P=0.002) and overall survival (OS) (log-rank: P=0.0048). Response was unrelated to metabolism, with tumours with high or low metabolism showing response. Increased 18F-FDG or 201Tl uptake predicted shorter progression-free survival (PFS) in the series (log-rank: 201Tl P=0.0097, 18F-FDG P=0.0170) and in cases with or without the −1p/−19q genotype. Elevated metabolism was associated with shorter OS in cases with intact 1p/19q (log-rank: 18F-FDG P=0.0077; 201Tl P=0.0004) and shorter PFS in responders (log-rank: 18F-FDG P=0.005; 201Tl P=0.0132). 201Tl uptake and 1p/19q loss were independent predictors of survival in multivariate analysis. In this initial study, 201Tl and 18F-FDG uptake did not predict response to PCV, but may be associated with poor survival following therapy irrespective of genotype. This may be clinically useful warranting further study.

41. Close contacts with leprosy in newly diagnosed leprosy patients in a high and low endemic area:Comparison between Bangladesh and Thailand

Abstract: Background: As part of a larger study of the role of close contacts in the transmission of M. leprae, we explored whether the proportion of newly detected cases with a family history of leprosy differs with different incidence rates of leprosy in a population. Methods: Retrospective analysis was performed of contacts of all new leprosy patients diagnosed during a 10-yr period in well-established leprosy control programs in Thailand and Bangladesh. By our definition, a contact group consisted of the new case and of past and present cases who were relatives and in-laws of the new case. For a new case, the nearest index case was defined on the basis of time of onset of symptoms for the cases in the contact group, in combination with the level of closeness of contact between these cases and the new case. Three contact levels were distinguished. In Bangladesh these levels were defined as 'kitchen contact'; 'house contact'; and 'non-house contact'. In Thailand comparable levels were defined as 'house contact'; 'compound contact'; and 'neighbor contact'. Results: In Bangladesh 1333, and in Thailand 129 new patients were included. The average new case detection rate over 10 yrs was 50 per 100,000 general population per year in Bangladesh, and 1.5 per 100,000 in Thailand. In the high endemic area 25% of newly detected cases were known to belong to a contact group and were not the index case of this group, whereas in the low endemic area 62% of newly detected cases had these characteristics. The distribution of the nearest index cases over the three contact levels was comparable in both areas. Just over half of the nearest index cases were found within the immediate family unit ('kitchen' in Bangladesh; 'house' in Thailand). Conclusion: The results indicate that in a low endemic area a higher proportion of newly detected leprosy cases have a family history of leprosy compared to a high endemic area. 44. Protein engineering of cytochrome b562 for quinone binding and light-induced electron transfer

Abstract: The central photochemical reaction in photosystem II of green algae and plants and the reaction center of some photosynthetic bacteria involves a one-electron transfer from a light-activated chlorin complex to a bound quinone molecule. Through protein engineering, we have been able to modify a protein to mimic this reaction. A unique quinone-binding site was engineered into the Escherichia coli cytochrome 6552 by introducing a cysteine within the hydrophobic interior of the protein. Various quinones, such as p-benzoquinone and 2,3-dimethoxy-5-methyl-1,4-benzoquinone, were then covalently attached to the protein through a cysteine sulfur addition reaction to the quinone ring. The cysteine placement was designed to bind the quinone ≈ 10 Å from the edge of the bound porphyrin. Fluorescence measurements confirmed that the bound hydroquinone is incorporated toward the protein's hydrophobic interior and is partially solvent-shielded. The bound quinones remain redox-active and can be oxidized and rereduced in a two-electron process at neutral pH. The semiquinone can be generated at high pH by a one-electron reduction, and the midpoint potential of this can be adjusted by ≈500 mV by binding different quinones to the protein. The heme-binding site of the modified cytochrome was then reconstituted with the chlorophyll analogue zinc chlorin e6. By using EPR and fast optical techniques, we show that, in the various chlorin-protein-quinone complexes, light-induced electron transfer can occur from the chlorin to the bound oxidized quinone but not the hydroquinone, with electron transfer rates in the order of 108 s-1.

Aunque, se ha especulado que las aminas biogénicas son las responsables de estos resultados; con la presente investigación se descarta a las aminas biogénicas como las responsables de tales efectos y que por el contrario, a niveles moderados de histamina, espermidina o espermina se mejora el crecimiento. Al mismo tiempo, demostramos que el camarón es capaz de metabolizar las aminas, como lo hacen los vertebrados superiores, excepto la cadaverina.
