Digital twin is a modern technology for many advanced applications. To construct a digital twin of a thermal system, it is required to make online estimations of unknown time-varying boundary conditions from sensor measured data, which needs to solve inverse heat transfer problems (IHTPs). However, a fast and accurate solution is challenging since the measured data is normally contaminated with noise and the traditional method to solve IHTP involves significant amount of calculations. Therefore, in this work, a rapid yet robust inversion algorithm called ANN-based extended Kalman smoothing algorithm is developed to realize the online prediction of desired parameter based on the measurements. The fast prediction is realized by replacing the conventional CFD-based state transfer models in extended Kalman smoothing algorithm with pre-trained ANN. Then, a two-dimensional internal convective heat transfer problem was employed as the case study to test the algorithm. The results have proved that the proposed algorithm is a computational-light and robust approach for solving IHTPs. The proposed algorithm can achieve estimation of unknown boundary conditions with a dimensionless average error of 0.0580 under noisy temperature measurement with a standard deviation of 10 K with a drastic reduction of computational cost compared to the conventional approach. Moreover, the effects of training data, location of sensor, future time step selection on the performance of prediction are investigated.

Strategies boosting both innate and adaptive immunity have great application prospects in cancer immunotherapy. Antibodies dual blocking the innate checkpoint CD47 and adaptive checkpoint PD-L1 or TIGIT could achieve durable anti-tumor effects. However, a small molecule dual blockade of CD47/SIRPα and TIGIT/PVR pathways has not been investigated. Here, an elevated expression of CD47 and PVR was observed in tumor tissues and cell lines analyzed with the GEO datasets and by flow cytometry, respectively. Compounds approved by the FDA were screened with the software MOE by docking to the potential binding pockets of SIRPα and PVR identified with the corresponding structural analysis. The candidate compounds were screened by blocking and MST binding assays. Azelnidipine was found to dual block CD47/SIRPα and TIGIT/PVR pathways by co-targeting SIRPα and PVR. In vitro, azelnidipine could enhance the macrophage phagocytosis when co-cultured with tumor cells. In vivo, azelnidipine alone or combined with irradiation could significantly inhibit the growth of MC38 tumors. Azelnidipine also significantly inhibits the growth of CT26 tumors, by enhancing the infiltration and function of CD8+ T cell in tumor and systematic immune response in the tumor-draining lymph node and spleen in a CD8+ T cell dependent manner. Our research suggests that the anti-hypertensive drug azelnidipine could be repositioned for cancer immunotherapy. © 2021 by the authors. Licensee MDPI, Basel, Switzerland.

Hollow graphitic carbon nanofibers (HGCNFs) have great promise for many important applications, such as catalysis, sensors, energy conversion, gas storage, and electronic devices. Here, we report a catalytic etching-wetting-dewetting mechanism in synthesizing HGCNFs with both hollow spherical-like and tunnel-like pores. With in situ transmission electron microscope imaging, we show that the spherical pores are formed by the evaporation of encapsulated Ni on the surface of amorphous carbon nanofiber and that hollow tunnels are developed through continuous etching of the amorphous carbon under catalysis of Ni nanoparticles. Theoretical calculations and simulations reveal that continuous tunnel etching is driven by the wetting-to-dewetting transition of the Ni-tunnel wall interaction during the catalytic graphitization of the amorphous carbon wall. In a typical application, we demonstrate that the synthesized HGCNFs, with a capacity about three times higher than that of their non-hollow counterparts, are excellent potential candidates for CO2 capture.
