The production of biofuels and "green" chemicals from the lignocellulose of fast-growing hardwood species is hampered by extensive acetylation of xylan. Analysis of soluble cell wall polysaccharides revealed a 4% reduction in the fraction of xylose units and an 18% increase in the fraction of glucose units, whereas the contents of cellulose and lignin were not affected. Enzymatic saccharification of wood from transgenic plants resulted in 27% higher glucose yield than for wild-type plants. Brunauer-Emmett-Teller (BET) analysis and Simons' staining pointed toward larger surface area and improved cellulose accessibility for wood from transgenic plants compared to wood from wild-type plants, which could be achieved by HjAXE deacetylating xylan bound to cellulose. Background: Incisional hernia (IH) is a known complication of abdominal surgery. Its incidence remains high in spite of the great improvement in the techniques and suture materials used for closing the abdominal wall incisions. This study was undertaken to study the incidence and various risk factors leading to IH. Methods: A total of 40 patients having IH admitted in a Tertiary Care Hospital at Hyderabad from 2007 to 2009 have been studied in various aspects like age, sex, clinical presentation and nature of previous operation, site of previous scar, precipitating factors like obesity, nutrition and wound infection. Results: Incisional hernia was common in 4th, 5th and 6th decade with female predominance (Male: Female ratio = 1:9). IH was more common in patients with previous history of gynaecological operation. Most of the patients presented with IH in the infra-umbilical region. In this study, 65% of patients with IH found to be obese, among them, 76.9% had wound infection during previous surgery. The P value was extremely significant. It is showing strong association between obesity and wound infection to the occurrence of IH. Conclusions: The highest incidence of incisional hernia observed in elderly patients with female preponderance. Operations on the female pelvic organs were the most common procedure preceding the development of IH. Obesity and post operative wound infection are important predisposing factors. All these factors are inter-related. Presence of more than one risk factor in a patient shows increased predisposition to the incidence of IH.

Background: Laparoscopic umbilical hernia repair has largely replaced open method and is gaining increasing popularity due to its various advantages. Hence the present research was undertaken to study the feasibility, efficiency, and safety of the laparoscopic umbilical hernia repair technique using modified two ports, combined with Herniorraphy and intraabdominal mesh fixation by trans- abdominal non-absorbable suture technique. Methodology: In this study, total 100 patients, including non-emergency and emergency underwent laparoscopic repair by combined Herniorraphy and intraabdominal mesh were studied. Two-port technique was used and the umbilical defect was closed using trans-abdominal suture, composite polypropylene, and PTFE mesh was placed intra abdominally and fixed to abdominal wall using trans abdominal  polypropylene sutures at 4 corners of mesh and with absorbable fixation device. Result: The average operative time for laparoscopic repair was 60?min, ranged from 45-100?min. The average hospital stay was 2.5 days (range 1 to 4 days). Early complications was seen in 13 patients who developed urinary retention requiring catheterization, 04 patients developed ileus which resolved spontaneously by 3rd day, including 02 emergency patients. Late postoperative complications occurred in six patients, complaining of abdominal pain which resolved over 6 months without further treatment. No patients presented with chronic pain or recurrence over the follow-up period of more than one year. None of the patient developed seroma at umbilicus or discharge from suture site. Conclusion: The modified 2 port laparoscopic umbilical hernia repair with combined Herniorraphy and intraabdominal mesh fixation has good clinical outcomes, less post-operative morbidity and offers an efficient, safe, and effective repair for umbilical hernia.

Background: Lignocellulose from fast growing hardwood species is a preferred source of polysaccharides for advanced biofuels and "green" chemicals. However, the extensive acetylation of hardwood xylan hinders lignocellulose saccharification by obstructing enzymatic xylan hydrolysis and causing inhibitory acetic acid concentrations during microbial sugar fermentation. To optimize lignocellulose for cost-effective saccharification and biofuel production, an acetyl xylan esterase AnAXE1 from Aspergillus niger was introduced into aspen and targeted to cell walls. Results: AnAXE1-expressing plants exhibited reduced xylan acetylation and grew normally. Without pretreatment, their lignocellulose yielded over 25% more glucose per unit mass of wood (dry weight) than wild-type plants. Glucose yields were less improved (+7%) after acid pretreatment, which hydrolyses xylan. The results indicate that AnAXE1 expression also reduced the molecular weight of xylan, and xylan-lignin complexes and/or lignin co-extracted with xylan, increased cellulose crystallinity, altered the lignin composition, reducing its syringyl to guaiacyl ratio, and increased lignin solubility in dioxane and hot water. Cell wall hemicelluloses and pectins are O-acetylated at specific positions, but the significance of these substitutions is poorly understood. Using a transgenic approach, we investigated how reducing the extent of O-acetylation in xylan affects cell wall chemistry, plant performance and the recalcitrance of lignocellulose to saccharification. The Aspergillus niger acetyl xylan esterase AnAXE1 was expressed in Arabidopsis under the control of either the constitutively expressed 35S CAMV promoter or a woody-tissue-specific GT43B aspen promoter, and the protein was targeted to the apoplast by its native signal peptide, resulting in elevated acetyl esterase activity in soluble and wall-bound protein extracts and reduced xylan acetylation. No significant alterations in cell wall composition were observed in the transgenic lines, but their xylans were more easily digested by a beta-1,4-endoxylanase, and more readily extracted by hot water, acids or alkali. Enzymatic saccharification of lignocellulose after hot water and alkali pretreatments produced up to 20% more reducing sugars in several lines. Fermentation by Trametes versicolor of tissue hydrolysates from the line with a 30% reduction in acetyl content yielded similar to 70% more ethanol compared with wild type. Plants expressing 35S: AnAXE1 and pGT43B:AnAXE1 developed normally and showed increased resistance to the biotrophic pathogen Hyaloperonospora arabidopsidis, probably due to constitutive activation of defence pathways. However, unintended changes in xyloglucan and pectin acetylation were only observed in 35S: AnAXE1-expressing plants. RT-qPCR and histochemical (GUS) analyses of expression patterns of GT43 members in hybrid aspen, reported here, revealed that three clades of the family have markedly differing specificity towards secondary wall-forming cells (wood and extraxylary fibres). Intriguingly, GT43A and B genes (corresponding to the Arabidopsis IRX9 clade) showed higher specificity for secondary-walled cells than GT43C and D genes (IRX14 clade), although both IRX9 and IRX14 are required for xylosyltransferase activity. The remaining genes, GT43E, F and G (IRX9-L clade), showed broad expression patterns. Transient transactivation analyses of GT43A and B reporters demonstrated that they are activated by PtxtMYB021 and PNAC085 (master secondary wall switches), mediated in PtxtMYB021 activation by an AC element. The high observed secondary cell wall specificity of GT43B expression prompted tests of the efficiency of its promoter (pGT43B), relative to the CaMV 35S (35S) promoter, for overexpressing a xylan acetyl esterase (CE5) or downregulating REDUCED WALL ACETYLATION (RWA) family genes and thus engineering wood acetylation. Helicopter rotor system operates in a highly dynamic and unsteady aerodynamic environment leading to severe vibratory loads on the rotor system. Repeated exposure to these severe loading conditions can induce damage in the composite rotor blade which may lead to a catastrophic failure. Therefore, an interest in the structural health monitoring (SHM) of the composite rotor blades has grown markedly in recent years. Two important issues are addressed in this thesis; (1) structural modeling and aeroelastic analysis of the damaged rotor blade and (2) development of a model based rotor health monitoring system. The effect of matrix cracking, the first failure mode in composites, is studied in detail for a circular section beam, box-beam and two-cell airfoil section beam. Later, the effects of further progressive damages such as debonding/delamination and fiber breakage are considered for a two-cell airfoil section beam representing a stiff-inplane helicopter rotor blade. It is found that the stiffness decreases rapidly in the initial phase of matrix cracking but becomes almost constant later as matrix crack saturation is reached. Due to matrix cracking, the bending and torsion stiffness losses at the point of matrix crack saturation are about 6-12 percent and about 25-30 percent, respectively. Due to debonding/delamination, the bending and torsion stiffness losses are about 6-8 percent and about 40-45 percent after matrix crack saturation, respectively. The stiffness loss due to fiber breakage is very rapid and leads to the final failure of the blade. An aeroelastic analysis is performed for the damaged composite rotor in forward flight and the numerically simulated results are used to develop an online health monitoring system. For fault detection, the variations in rotating frequencies, tip bending and torsion response, blade root loads and strains along the blade due to damage are investigated. The resulting structures are more efficient, economical and reliable. In traditional optimization techniques, the most important factor affecting on optimization process is the search direction which is the derivative of change in respose of structure due to change in design variables. This derivative is called as sensitivity derivative.Accuracy of sensitivity analysis is very much dependent on the method of structural analysis, technique of sensitivity calculation, computational efficiency etc. In this work, accuracy of sensitivity derivatives in elastic and plastic analyses are investigated on the basis of small strain theory. Combined with the Finite element method, which provides an excellent tool for the analysis of complex structures, the different techniques used for sensitivity calculations are finite difference method, semianalytical method and analytical method. Detail discussion of formulation and implementation of these methods are presented. Comparative study shows the relative error, cost of computation and efficiency of the above methods.From the results obtained, it can be stated that the finite difference method is the simplest technique that does not require access to the finite elemen analysis code and hence requires less efforts. However, this method is inefficient and less accurate. Analytical method is the most accurate method but its formulation and implementation is difficult as compared to other two metods. Semianalytical method is found to be a compromise of the two that results in more accurate solutions than from the finite diference method and is easy to implement as compared to the analytical method. In traditional optimization techniques, the most important factor affecting on optimization process is the search direction which is the derivative of change in respose of structure due to change in design variables. This derivative is called as sensitivity derivative.Accuracy of sensitivity analysis is very much dependent on the method of structural analysis, technique of sensitivity calculation, computational efficiency etc. In this work, accuracy of sensitivity derivatives in elastic and plastic analyses are investigated on the basis of small strain theory. Combined with the Finite element method, which provides an excellent tool for the analysis of complex structures, the different techniques used for sensitivity calculations are finite difference method, semianalytical method and analytical method. Detail discussion of formulation and implementation of these methods are presented. Comparative study shows the relative error, cost of computation and efficiency of the above methods.From the results obtained, it can be stated that the finite difference method is the simplest technique that does not require access to the finite elemen analysis code and hence requires less efforts. However, this method is inefficient and less accurate. Analytical method is the most accurate method but its formulation and implementation is difficult as compared to other two metods. Semianalytical method is found to be a compromise of the two that results in more accurate solutions than from the finite diference method and is easy to implement as compared to the analytical method.
