Synthetic micas are materials with interesting properties that combine high charge (high cation exchange capacity) with swelling capacity. Among the various methods for synthesizing this clay, the “NaCl-melt method” is the most widely used, producing a material with excellent crystallinity. Furthermore, synthetic micas are also susceptible to modifications with a wide range of compounds, mainly organic compounds, which further increases their reactivity. Thus, this Review addresses the properties and syntheses of this clay. The types of modifications used and the changes that occur in the properties of this clay are reported, in addition to applications in the removal of contaminating compounds that can be found in water. Research to date has demonstrated that synthetic mica-based adsorbents are versatile in the adsorption of various contaminants, such as metals, pharmaceuticals and others. Therefore, this material can be promising in the adsorption and release of drugs, bioactive compounds, enzymes, proteins, etc., thus increasing its field of application.

This article provides a comprehensive study of ecosystem rehabilitation methods widely used in the 21st century, focusing on Europe. The review covers the evolution and trends in scientific article publication, identification of European countries demonstrating high publication outputs, collaboration patterns, leading journals, and thematic areas. Additionally, it examines primary stressors in European aquatic ecosystems, and different methods and treatments commonly employed for remediation purposes. The analysis of selected articles revealed a significant increase in studies over time, driven by public awareness and financial incentives from national, European and global organizations. Italy, Portugal and Spain were the leading countries in degraded ecosystem rehabilitation studies, mainly focusing on remediating contaminated areas where metals were identified as the primary stressor (chemical pollution). Chemical remediation method emerged as the most used, closely followed by biological remediation method, which have gained prominence in recent years due to their ecological, economic, and social combined benefits. Furthermore, recent studies demonstrate a growing trend towards the combined use of more than one treatment/method to rehabilitate ecosystems, particularly with biological treatments. This combined approach has the potential for synergistic effects in achieving more effective rehabilitation and their sustainability in the long term, thus, a focus for future research.

High charge swelling micas are layered silicates used in adsorption of several inorganic and organic species. In this study, we evaluated the adsorption of chlorhexidine digluconate on highly charge mica Na-mica-4. Drug release and antibacterial action against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) using the direct contact test for mica/chlorhexidine hybrids were evaluated. The hybrids were characterized using X-ray diffraction, Fourier transform infrared spectroscopy, zeta potential measurements, CHN elemental analysis, and scanning electron microscopy. Chlorhexidine interacts successfully with the mica surface. The maximum amount of chlorhexidine incorporated was 583 mg g−1, and the characterizations indicated that electrostatic attraction between the protonated amino group of the drug and the negative surface of clay was predominant in the formation of hybrids. Release kinetics study indicated that 80% of the drug release occurred in the first 12 h. The antibacterial test proved to be dependent on the time of chlorhexidine release, reaching 100% inhibition against S. aureus and E. coli after 0.5 h and 6 h, respectively.

BackgroundDrugs taken during pregnancy can affect maternal and child health outcomes, but few studies have compared the safety and virological efficacy of different antiretroviral therapy (ART) regimens. We report the primary safety outcomes from enrolment up to 50 weeks post partum and a secondary virological efficacy outcome at 50 weeks post partum of three commonly used ART regimens for HIV-1.MethodsIn this multicentre, open-label, randomised, controlled, phase 3 trial, we enrolled pregnant women aged 18 years or older with confirmed HIV-1 infection at 14-28 weeks of gestation. Women were enrolled at 22 clinical research sites in nine countries (Botswana, Brazil, India, South Africa, Tanzania, Thailand, Uganda, the USA, and Zimbabwe). Participants were randomly assigned (1:1:1) to one of three oral regimens: dolutegravir, emtricitabine, and tenofovir alafenamide; dolutegravir, emtricitabine, and tenofovir disoproxil fumarate; or efavirenz, emtricitabine, and tenofovir disoproxil fumarate. Up to 14 days of antepartum ART before enrolment was permitted. Women with known multiple gestation, fetal anomalies, acute significant illness, transaminases more than 2·5 times the upper limit of normal, or estimated creatinine clearance of less than 60 mL/min were excluded. Primary safety analyses were pairwise comparisons between ART regimens of the proportion of maternal and infant adverse events of grade 3 or higher up to 50 weeks post partum. Secondary efficacy analyses at 50 weeks post partum included a comparison of the proportion of women with plasma HIV-1 RNA of less than 200 copies per mL in the combined dolutegravir-containing groups versus the efavirenz-containing group. Analyses were done in the intention-to-treat population, which included all randomly assigned participants with available data. We propose a scheme to dissipatively produce steady-state entanglement in a two-qubit system, via an interaction with a bosonic mode. The system is driven into a stationary entangled state, while we compensate the mode dissipation by injecting energy via a coherent pump field. We also present a scheme which allows us to adiabatically transfer all the population to the desired entangled state. The dynamics leading to the entangled state in these schemes can be understood in analogy with electromagnetically induced transparency and stimulated Raman adiabatic passage, respectively.

Listeria monocytogenes is a facultative Gram-positive bacterium that causes listeriosis, a severe foodborne disease. We previously discovered that ring-fused 2-pyridone compounds can decrease virulence factor expression in Listeria by binding and inactivating the PrfA virulence activator. In this study, we tested PS900, a highly substituted 2-pyridone that was recently discovered to be bactericidal to other Gram-positive pathogenic bacteria, such as Staphylococcus aureus and Enterococcus faecalis. We show that PS900 can interact with PrfA and reduce the expression of virulence factors. Unlike previous ring-fused 2-pyridones shown to inactivate PrfA, PS900 had an additional antibacterial activity and was found to potentiate sensitivity toward cholic acid. Two PS900-tolerant mutants able to grow in the presence of PS900 carried mutations in the brtA gene, encoding the BrtA repressor. In wild-type (WT) bacteria, cholic acid binds and inactivates BrtA, thereby alleviating the expression of the multidrug transporter MdrT. Interestingly, we found that PS900 also binds to BrtA and that this interaction causes BrtA to dissociate from its binding site in front of the mdrT gene. In addition, we observed that PS900 potentiated the effect of different osmolytes. We suggest that the increased potency of cholic acid and osmolytes to kill bacteria in the presence of PS900 is due to the ability of the latter to inhibit general efflux, through a yet-unknown mechanism. Our data indicate that thiazolino 2-pyridones constitute an attractive scaffold when designing new types of antibacterial agents. Upon encountering stress, a large multicomponent protein complex known as the stressosome becomes activated, ultimately leading to SigB activation. RsbX is a protein needed to reset a "stressed"stressosome and prevent unnecessary SigB activation in nonstressed conditions. Consequently, absence of RsbX leads to constitutive activation of SigB even without prevailing stress stimulus. To further examine the involvement of SigB in the virulence of this pathogen, we investigated whether a strain with constitutively active SigB would be affected in virulence factor expression and/or infectivity in cultured cells and in a chicken embryo infection model. Our results suggest that increased SigB activity does not substantially alter virulence gene expression compared with the wild-type (WT) strain at transcript and protein levels. Bacteria lacking RsbX were taken up by phagocytic and nonphagocytic cells at a similar frequency to WT bacteria, both in stressed and nonstressed conditions. Finally, the absence of RsbX only marginally affected the ability of bacteria to infect chicken embryos. Mercury's extreme toxicity and persistence in the environment justifies a thorough evaluation of its dynamics in ecosystems. Aveiro Lagoon (Portugal) was for decades subject to mercury effluent discharges. A Nature-based Solution (NbS) involving Zostera noltei re-colonization is being tested as an active ecosystem restoration measure. To study the effect of Zostera noltei on the sediment contaminant biogeochemistry, seasonal (summer/winter) sediment, interstitial water and labile mercury vertical profiles were made in vegetated (Transplanted and Natural seagrass meadows) and non-vegetated sites (Bare-bottom area). While no significant differences (p > 0.05) were observed in the sedimentary phase, Zostera noltei presence reduced the reactive/labile mercury concentrations in the top sediment layers by up to 40% when compared to non-vegetated sediment, regardless of season. No differences were found between vegetated meadows, highlighting the fast recovery of the contaminant regulation ecosystem function provided by the plants after re-colonization and its potential for the rehabilitation of historically contaminated ecosystems.

While protein conformational heterogeneity plays an important role in many aspects of biological function, including ligand binding, its impact has been difficult to quantify. Macromolecular X-ray diffraction is commonly interpreted with a static structure, but it can provide information on both the anharmonic and harmonic contributions to conformational heterogeneity. Here, through multiconformer modeling of time- and space-averaged electron density, we measure conformational heterogeneity of 743 stringently matched pairs of crystallographic datasets that reflect unbound/apo and ligand-bound/holo states. When comparing the conformational heterogeneity of side chains, we observe that when binding site residues become more rigid upon ligand binding, distant residues tend to become more flexible, especially in non-solvent-exposed regions. Among ligand properties, we observe increased protein flexibility as the number of hydrogen bonds decreases and relative hydrophobicity increases. Across a series of 13 inhibitor-bound structures of CDK2, we find that conformational heterogeneity is correlated with inhibitor features and identify how conformational changes propagate differences in conformational heterogeneity away from the binding site. Collectively, our findings agree with models emerging from nuclear magnetic resonance studies suggesting that residual side-chain entropy can modulate affinity and point to the need to integrate both static conformational changes and conformational heterogeneity in models of ligand binding.

Parrots have undergone substantial karyotype evolution compared to most other birds. Here, Huang et al. analyze chromosome-level genome assemblies for four parrot species and elucidate the complex evolutionary history of parrot chromosomes. The karyotype of most birds has remained considerably stable during more than 100 million years' evolution, except for some groups, such as parrots. The evolutionary processes and underlying genetic mechanism of chromosomal rearrangements in parrots, however, are poorly understood. Here, using chromosome-level assemblies of four parrot genomes, we uncover frequent chromosome fusions and fissions, with most of them occurring independently among lineages. The increased activities of chromosomal rearrangements in parrots are likely associated with parrot-specific loss of two genes, ALC1 and PARP3, that have known functions in the repair of double-strand breaks and maintenance of genome stability. We further find that the fusion of the ZW sex chromosomes and chromosome 11 has created a pair of neo-sex chromosomes in the ancestor of parrots, and the chromosome 25 has been further added to the sex chromosomes in monk parakeet. Together, the combination of our genomic and cytogenetic analyses characterizes the complex evolutionary history of chromosomal rearrangements and sex chromosomes in parrots.

Here, using chromosome-level assemblies of four parrot genomes, we uncover frequent chromosome fusions and fissions, with most of them occurring independently among lineages. The increased activities of chromosomal rearrangements in parrots are likely associated with parrot-specific loss of two genes, ALC1 and PARP3, that have known functions in the repair of double-strand breaks and maintenance of genome stability. We further find that the fusion of the ZW sex chromosomes and chromosome 11 has created a pair of neo-sex chromosomes in the ancestor of parrots, and the chromosome 25 has been further added to the sex chromosomes in monk parakeet. Together, the combination of our genomic and cytogenetic analyses characterizes the complex evolutionary history of chromosomal rearrangements and sex chromosomes in parrots.

While protein conformational heterogeneity plays an important role in many aspects of biological function, including ligand binding, its impact has been difficult to quantify. Macromolecular X-ray diffraction is commonly interpreted with a static structure, but it can provide information on both the anharmonic and harmonic contributions to conformational heterogeneity. Here, through multiconformer modeling of time- and space-averaged electron density, we measure conformational heterogeneity of 743 stringently matched pairs of crystallographic datasets that reflect unbound/apo and ligand-bound/holo states. When comparing the conformational heterogeneity of side chains, we observe that when binding site residues become more rigid upon ligand binding, distant residues tend to become more flexible, especially in non-solvent-exposed regions. Among ligand properties, we observe increased protein flexibility as the number of hydrogen bonds decreases and relative hydrophobicity increases. Across a series of 13 inhibitor-bound structures of CDK2, we find that conformational heterogeneity is correlated with inhibitor features and identify how conformational changes propagate differences in conformational heterogeneity away from the binding site. Collectively, our findings agree with models emerging from nuclear magnetic resonance studies suggesting that residual side-chain entropy can modulate affinity and point to the need to integrate both static conformational changes and conformational heterogeneity in models of ligand binding.

Growth hormone (GH) has been used for over 35 years, and its safety and efficacy has been studied extensively. Experimental studies showing the permissive role of GH/insulin-like growth factor 1 (IGF-I) in carcinogenesis have raised concerns regarding the safety of GH replacement in children and adults who have received treatment for cancer and those with intracranial and pituitary tumours. A consensus statement was produced to guide decision-making on GH replacement in children and adult survivors of cancer, in those treated for intracranial and pituitary tumours and in patients with increased cancer risk. With the support of the European Society of Endocrinology, the Growth Hormone Research Society convened a Workshop, where 55 international key opinion leaders representing 10 professional societies were invited to participate. This consensus statement utilized: (1) a critical review paper produced before the Workshop, (2) five plenary talks, (3) evidence-based comments from four breakout groups, and (4) discussions during report-back sessions. Current evidence reviewed from the proceedings from the Workshop does not support an association between GH replacement and primary tumour or cancer recurrence. The effect of GH replacement on secondary neoplasia risk is minor compared to host- and tumour treatment-related factors. There is no evidence for an association between GH replacement and increased mortality from cancer amongst GH-deficient childhood cancer survivors. Patients with pituitary tumour or craniopharyngioma remnants receiving GH replacement do not need to be treated or monitored differently than those not receiving GH. GH replacement might be considered in GH-deficient adult cancer survivors in remission after careful individual risk/benefit analysis. In children with cancer predisposition syndromes, GH treatment is generally contraindicated but may be considered cautiously in select patients.

In the wider context of heathland restoration, we investigated how field scale experimental acidification with sulphur (sulfur)affected soil biodiversity, fertility and function over a period of 17 years. A field experiment was conducted in the Isle of Purbeck, England, using ferrous sulphate and elemental sulphur as acidifying agents. We tested the effects of acidification on soil fertility, plant communities, litter decomposition, microbiology (including fungi bacteria and actinomycetes), arbuscular and ericoid mycorrhizal colonisation, and soil fauna (including earthworms, nematodes, rotifers and tardigrades). We found that elemental sulphur had a considerable and persistent effect on soil pH, lowering it to levels found in the surrounding reference acid grassland and heathland sites. A newly adapted heathland restoration index based on soil chemistry, found that elemental sulphur was by far the most successful treatment leading to soil conditions similar to the heathlands. Overall, acidification caused a loss of base cations and an increase in toxic aluminium compounds. Consequently the more mesotrophic components of soil biology were reduced by acidification during the course of the experiment. This transformed the soil biological system into one typical of acid grasslands and heathlands. Concomitant litter decomposition was similarly inhibited by acidification, with the microbiota more strongly hindered in acidified soil than the macroscopic fauna. Acidification resulted in a reduction in nematode and rotifer abundance and earthworm biomass. The vegetation community was also strongly modified by the elemental sulphur treatments and, where grazing was restricted, soil acidification allowed a restored heathland community to endure. Essential and non-essential trace metals are capable of causing toxicity to organisms above a threshold concentration. Extensive research has assessed the behaviour of trace metals in biological and ecological systems, but has typically focused on single organisms within a trophic level and not on multi-trophic transfer through terrestrial food chains. This reinforces the notion of metal toxicity as a closed system, failing to consider one trophic level as a pollution source to another; therefore, obscuring the full extent of ecosystem effects. Given the relatively few studies on trophic transfer of metals, this review has taken a compartment-based approach, where transfer of metals through trophic pathways is considered as a series of linked compartments (soil-plant-arthropod herbivore-arthropod predator). In particular, we consider the mechanisms by which trace metals are taken up by organisms, the forms and transformations that can occur within the organism and the consequences for trace metal availability to the next trophic level. The review focuses on four of the most prevalent metal cations in soil which are labile in terrestrial food chains: Cd, Cu, Zn and Ni. Current knowledge of the processes and mechanisms by which these metals are transformed and moved within and between trophic levels in the soil-plant-arthropod system are evaluated. We demonstrate that the key factors controlling the transfer of trace metals through the soil-plant-arthropod system are the form and location in which the metal occurs in the lower trophic level and the physiological mechanisms of each organism in regulating uptake, transformation, detoxification and transfer. The magnitude of transfer varies considerably depending on the trace metal concerned, as does its toxicity, and we conclude that biomagnification is not a general property of plant-arthropod and arthropod-arthropod systems. In this work, deep learning techniques for brain age prediction from magnetic resonance images are investigated, aiming to assist in the identification of biomarkers of the natural aging process. The identification of biomarkers is useful for detecting an early-stage neurodegenerative process, as well as for predicting age-related or non-age-related cognitive decline. Two techniques are implemented and compared in this work: a 3D Convolutional Neural Network applied to the volumetric image and a 2D Convolutional Neural Network applied to slices from the axial plane, with subsequent fusion of individual predictions. Introduction: Parkinson’s disease is most prevalent among elderly people, 65 years and over, and leads to an alteration in motor control associated with postural instability. Current evidence shows that postural control decreases with the aging process. In addition, postural control is more altered in healthy aged men than in women. Until today, few studies have evaluated the combined impact of Parkinson’s disease and sex on postural control. This review has allowed to evaluate the impact of Parkinson’s disease and sex on postural control measurements in elderly people. Methodology: Studies have been selected from two main databases: PubMed and EBSCO using the keywords “Parkinson”, “postural control OR balance” and “sex”. Articles related to the evaluation of postural control, including men and women with Parkinson’s aged over 65 years old, regardless of stage, were included (n = 179). Articles were excluded if not written in French or English or not presenting original content. Results: Ten (10) studies out of 179 that fulfilled inclusion and exclusion criteria were reported in the final analysis, which cumulates a total of 944 individuals with Parkinson’s (410 women). In general, results show greater postural instability among people with Parkinson’s compared to healthy subjects, and this according to different objective measurements using stabilographic parameters from force platforms. Only two studies out of ten evaluated postural control while briefly considering distinctions between sex, but without showing a significant difference between men and women with Parkinson’s. Parkinson’s severity, length of time of Parkinson’s disease and cognitive state of the person are the three variables with a negative impact on postural control. Conclusion: Older people with Parkinson’s disease have greater postural instability. Sex does not seem to influence the postural control of elderly people with Parkinson’s, although more studies are necessary.

The shallow depth (<2 km) of well-documented SSEs at the Hikurangi subduction zone offshore New Zealand offers a unique opportunity to link geophysical imaging of the subduction zone with direct access to incoming material that represents the megathrust fault rocks hosting slow slip. Two recent International Ocean Discovery Program Expeditions sampled this incoming material before it is entrained immediately down-dip along the shallow plate interface. Drilling results, tied to regional seismic reflection images, reveal heterogeneous lithologies with highly variable physical properties entering the SSE source region. These observations suggest that SSEs and associated slow earthquake phenomena are promoted by lithological, mechanical, and frictional heterogeneity within the fault zone, enhanced by geometric complexity associated with subduction of rough crust.

Single particle, cryogenic electron microscopy (cryo-EM) experiments now routinely produce high-resolution data for large proteins and their complexes. Building an atomic model into a cryo-EM density map is challenging, particularly when no structure for the target protein is known a priori. Existing protocols for this type of task often rely on significant human intervention and can take hours to many days to produce an output. Here, we present a fully automated, template-free model building approach that is based entirely on neural networks. We use a graph convolutional network (GCN) to generate an embedding from a set of rotamer-based amino acid identities and candidate 3-dimensional C$\alpha$ locations. Starting from this embedding, we use a bidirectional long short-term memory (LSTM) module to order and label the candidate identities and atomic locations consistent with the input protein sequence to obtain a structural model. Lack of data and data quality issues are among the main bottlenecks that prevent further artificial intelligence adoption within many organizations, pushing data scientists to spend most of their time cleaning data before being able to answer analytical questions. Hence, there is a need for more effective and efficient data cleaning solutions, which, not surprisingly, is rife with theoretical and engineering problems. This report addresses the problem of performing holistic data cleaning incrementally, given a fixed rule set and an evolving categorical relational dataset acquired in sequential batches. To the best of our knowledge, our contributions compose the first incremental framework that cleans data (i) independently of user interventions, (ii) without requiring knowledge about the incoming dataset, such as the number of classes per attribute, and (iii) holistically, enabling multiple error types to be repaired simultaneously, and thus avoiding conflicting repairs. O presente artigo propõe-se analisar o sistema de espaços livres (SEL) do Distrito de Jundiapeba, localizado na Cidade de Mogi das Cruzes, Estado de São Paulo, bem como indicar ferramentas capazes de mitigar os problemas de infraestrutura urbana enfrentado pela população local. Para isso, utilizar-se-á, como ferramenta de estudo o mapeamento das áreas livres; hidrografia; a ocupação do solo e legislação urbana a fim de entender a configuração urbana do distrito. Assim, a partir da abordagem dos sistemas de espaços livres, que contribuem para a drenagem, enumerou-se questões aptas a demonstrar a importância da preservação e adequação do meio urbano. Em síntese, este estudo objetiva promover uma reflexão capaz de contribuir com o desenvolvimento urbano-sustentável do Distrito Jundiapeba e compilar informações que possam auxiliar futuras pesquisas., This article analysis the open space systems of the district of Jundiapeba, located in Mogi das Cruzes, São Paulo, as well as tools to mitigate the problems about urban infraestructure faced by the locals. Therefore, it is used as a study instrument, the mapping of open areas; hydrography; the funcional land uses and urban legislation to understand the urban configuration of the district. Thus, based on the approach of open space systems and their contribution with urban drainage, it was numbered some points which are able to demonstrate the importance of conservation and adequacy of the urban environment. Slow slip events (SSEs) at the northern Hikurangi subduction margin, New Zealand, are among the best-documented shallow SSEs on Earth. International Ocean Discovery Program Expeditions 372 and 375 were undertaken to investigate the processes and in situ conditions that underlie subduction zone SSEs at the northern Hikurangi Trough. We accomplished this goal by (1) coring and geophysical logging at four sites, including penetration of an active thrust fault (the Pāpaku fault) near the deformation front, the upper plate above the SSE source region, and the incoming sedimentary succession in the Hikurangi Trough and atop the Tūranganui Knoll seamount; and (2) installing borehole observatories in the Pāpaku fault and in the upper plate overlying the slow slip source region. Logging-while-drilling (LWD) data for this project were acquired as part of Expedition 372, and coring, wireline logging, and observatory installations were conducted during Expedition 375. Northern Hikurangi subduction margin SSEs recur every 1–2 y and thus provide an ideal opportunity to monitor deformation and associated changes in chemical and physical properties throughout the slow slip cycle. In situ measurements and sampling of material from the sedimentary section and oceanic basement of the subducting plate reveal the rock properties, composition, lithology, and structural character of material that is transported downdip into the SSE source region. A recent seafloor geodetic experiment raises the possibility that SSEs at northern Hikurangi may propagate to the trench, indicating that the shallow thrust fault (the Pāpaku fault) targeted during Expeditions 372 and 375 may also lie in the SSE rupture area and host a portion of the slip in these events. Hence, sampling and logging at this location provides insights into the composition, physical properties, and architecture of a shallow fault that may host slow slip. Avian genomes have perplexed researchers by being conservative in both size and rearrangements, while simultaneously holding the blueprints for a massive species radiation during the last 65 million years (My). Transposable elements (TEs) in bird genomes are relatively scarce but have been implicated as important hotspots for chromosomal inversions. In zebra finch (Taeniopygia guttata), long terminal repeat (LTR) retrotransposons have proliferated and are positively associated with chromosomal breakpoint regions. Here, we present the genome, karyotype and transposons of blue-capped cordon-bleu (Uraeginthus cyanocephalus), an African songbird that diverged from zebra finch at the root of estrildid finches 10 million years ago (Mya). This constitutes the third linked-read sequenced genome assembly and fourth in-depth curated TE library of any bird. Exploration of TE diversity on this brief evolutionary timescale constitutes a considerable increase in resolution for avian TE biology and allowed us to uncover 4.5 Mb more LTR retrotransposons in the zebra finch genome. In blue-capped cordon-bleu, we likewise observed a recent LTR accumulation indicating that this is a shared feature of Estrildidae. Curiously, we discovered 25 new endogenous retrovirus-like LTR retrotransposon families of which at least 21 are present in zebra finch but were previously undiscovered. This highlights the importance of studying close relatives of model organisms.
