1. Impact of Deep Water Formation on Antarctic Circumpolar Transport During Gateway Opening

Abstract

Ambiguity over the Eocene opening times of the Tasman Gateway and Drake Passage makes it difficult to determine the initiation time of the Antarctic Circumpolar Current. If the Tasman Gateway opened later than Drake Passage, then Australia may have prevented the proto-ACC from forming. Recent modeling results have shown that only a relatively weak circumpolar transport results under Eocene surface forcing. This leads to warm and buoyant coastal water around Antarctica, which may impede the formation of deep waters and convective processes. This suggests that a change in deep water formation might be required to increase the density contrast across the Southern Ocean and increase circumpolar transport. Here we use a simple reduced gravity model with two basins, to represent the Atlantic and the Pacific. This fixes the density difference between surface and deep water and allows us to isolate the impact of deep water formation on circumpolar transport. With no obstacle on the southern boundary the circumpolar current increases its transport from 82.3 to 270.0 Sv with deep water formation. Placing an Antipodean landmass on the southern boundary reduces this transport as the landmass increases in size. However, circumpolar flow north of this landmass remains a possibility even without deep water formation. Weak circumpolar transport continues until the basin is completely blocked by the Antipodes. When the Antipodes is instead allowed to split from the southern boundary, circumpolar transport recovers to its unobstructed value. Flow rapidly switches to south of the Antipodes when the gateway is narrow.

Abstract

The NA48/2 experiment at CERN reports the first observation of the $K^{\pm} \rightarrow \pi^{0} \pi^{0} \mu^{\pm} \nu$ decay based on a sample of 2437 candidates with 15% background contamination collected in 2003--2004. The decay branching ratio in the kinematic region of the squared dilepton mass above $0.03$~GeV$^2/c^4$ is measured to be $(0.65 \pm 0.03) \times 10^{-6}$. The extrapolation to the full kinematic space, using a specific model, is found to be $(3.45 \pm 0.16) \times 10^{-6}$, in agreement with chiral perturbation theory predictions., Comment: 16 pages, 9 figures

5. Deepening of Southern Ocean gateway leads to abrupt onset of a deep-reaching meridional overturning circulation

Abstract

During the Eocene and the Eocene-Oligocene transition, the lower cell of the meridional overturning circulation (MOC), associated with bottom water formation, underwent changes associated with the geological evolution of Southern Ocean gateways. These are important for the Cenozoic climate transition from Greenhouse to Icehouse, but their dynamics still remain elusive. We demonstrate, using an idealized eddying ocean model, that the opening of a gateway leads to the abrupt onset of a vigorous, deep-reaching, MOC. This MOC has a maximum transport for a shallow gateway, and decreases with further deepening of the gateway. This abrupt change in the MOC can be explained through the ability with which standing meanders—turbulent features located downstream of the gateway—can induce deep vertical heat transport at high latitudes where bottom waters are produced. Our results demonstrate the crucial role of turbulent processes in setting the strength of the global ocean's deep-reaching MOC.

Abstract

The NA48/2 experiment at CERN reports the first observation of the K-+/- -> pi(+/-)pi(0)e(+)e(-) decay from an exposure of 1.7 x 10(11) charged kaon decays recorded in 2003-2004. A sample of 4919 candidates with 4.9% background contamination allows the determination of the branching ratio in the full kinematic region, BR(K-+/- -> pi(+/-)pi(0)e(+)e(-)) = (4.24 +/- 0.14) x 10(-6). The study of the kinematic space shows evidence for a structure dependent contribution in agreement with predictions based on chiral perturbation theory. Several P- and CP-violating asymmetries are also evaluated.

Abstract

$K^{+}\rightarrow\pi^{+}\nu\overline{\nu}$ is one of the theoretically cleanest meson decay where to look for indirect effects of new physics complementary to LHC searches. The NA62 experiment at CERN SPS is designed to measure the branching ratio of this decay with 10\% precision. NA62 took data in pilot runs in 2014 and 2015 reaching the final designed beam intensity. Background Patient and public involvement (PPI) is now an expectation of research funders, in the UK, but there is relatively little published literature on what this means in practice – nor is there much evaluative research about implementation and outputs. Policy literature endorses the need to include PPI representation at all stages of planning, performing and research dissemination, and recommends resource allocation to these roles; but details of how to make such inputs effective in practice are less common. While literature on power and participation informs the debate, there are relatively few published case studies of how this can play out through the lived experience of PPI in research; early findings highlight key issues around access to knowledge, resources, and interpersonal respect. This article describes the findings of a case study of PPI within a study about PPI in research. Methods The aim of the study was to look at how the PPI representatives’ inputs had developed over time, key challenges and changes, and lessons learned. We used realist evaluation and normalisation process theory to frame and analyse the data, which was drawn from project documentation, minutes of meetings and workshops, field notes and observations made by PPI representatives and researchers; documented feedback after meetings and activities; and the structured feedback from two formal reflective meetings. Findings Key findings included the need for named contacts who support, integrate and work with PPI contributors and researchers, to ensure partnership working is encouraged and supported to be as effective as possible. A structure for partnership working enabled this to be enacted systematically across all settings. Abstract

The NA48/2 Collaboration at CERN has accumulated unprecedented statistics of rare kaon decays in the Ke4 modes: Ke4(+-) ($K^\pm \to \pi^+ \pi^- e^\pm \nu$) and Ke4(00) ($K^\pm \to \pi^0 \pi^0 e^\pm \nu$) with nearly one percent background contamination. The detailed study of form factors and branching rates, based on these data, has been completed recently. The results brings new inputs to low energy strong interactions description and tests of Chiral Perturbation Theory (ChPT) and lattice QCD calculations. In particular, new data support the ChPT prediction for a cusp in the $\pi^0\pi^0$ invariant mass spectrum at the two charged pions threshold for Ke4(00) decay. New final results from an analysis of about 400 $K^\pm \to \pi^\pm \gamma \gamma$ rare decay candidates collected by the NA48/2 and NA62 experiments at CERN during low intensity runs with minimum bias trigger configurations are presented. 11. The role of ocean gateways in the dynamics and sensitivity to wind stress of the early Antarctic Circumpolar Current

Abstract

The date of inception of the Antarctic Circumpolar Current is debated due to uncertainty in the relative opening times of Drake Passage and the Tasman Seaway. Using an idealized eddy-resolving numerical ocean model, we investigate whether both ocean gateways have to be open to allow for a substantial circumpolar current. We find that overlapping continental barriers do not impede a circumpolar transport in excess of 50Sv, as long as a circumpolar path can be traced around the barriers. However, the presence of overlapping barriers does lead to an increased sensitivity of the current's volume transport to changes in wind stress. This change in sensitivity is interpreted in terms of the role of pressure drops across continental barriers and submerged bathymetry in balancing the momentum input by the surface wind stress. Specifically, when the pressure drop across continents is the main balancing sink of momentum, the zonal volume transport is sensitive to changes in wind stress. Changes in zonal volume transport take place via altering the depth-independent part of the circumpolar transport rather than that arising from thermal wind shear. In such a scenario, isopycnals continue to slope steeply across the model Southern Ocean, implying a strong connection between the deep and surface oceans. This may have consequences for the meridional overturning circulation and its sensitivity to wind stress.

Abstract

The NA62 experiment will begin taking data in 2015. Abstract

From the 2002 data taking with a neutral kaon beam extracted from the CERN-SPS, the NA48/1 experiment observed 97 $\Xi^{0}\rightarrow \Sigma^{+} \mu^{-} \bar{\nu}_{\mu}$ candidates with a background contamination of $30.8 \pm 4.2$ events. From this sample, the BR($\Xi^{0}\rightarrow \Sigma^{+} \mu^{-} \bar{\nu}_{\mu}$) is measured to be $(2.17 \pm 0.32_{\mathrm{stat}}\pm 0.17_{\mathrm{syst}})\times10^{-6}$.

Abstract

Conical convergent-divergent (CCD) nozzles represent an important category of supersonic jet-engine nozzles which require variable throat areas and variable exit areas to adapt to a range of operating conditions. CCD nozzles with design Mach numbers of 1.3, 1.5, and 1.65 are examined experimentally over a range of fully expanded Mach numbers from 1.22 to 1.71. The characteristics of the flow and acoustic fields from these nozzles are explored. Shadowgraph, Particle Image Velocimetry, far-field and near-field acoustic surveys are presented. Results of a Monotonically Integrated Large Eddy Simulation are presented for the Mach 1.5 nozzle at an underexpanded condition. The agreement between simulations and measurements is excellent. It is shown that these nozzles differ from traditional smoothly contoured method-of-characteristics nozzles in that they never achieve a shock free condition. Furthermore it is shown that these nozzles produce a double diamond pattern in which two sets of shock diamonds are generated with an axial displacement between them. The cause of this phenomenon is explored. It is further shown that as a consequence they are never free from shock-associated noise even when operated at perfect expansion. In spite of this difference, it is found that CCD nozzles behave like traditional convergent-divergent nozzles in that they produce the same shock-cell size, broadband shock-associated noise peak frequency, and screech frequency as traditional convergent-divergent nozzles. Abstract

As first observed by the NA48/2 experiment at the CERN SPS, the $\p0p0$ invariant mass (M00) distribution from $\kcnn$ decay shows a cusp-like anomaly at M00=2m+, where m+ is the charged pion mass. An analysis to extract the pi pi scattering lengths in the isospin I=0 and I=2 states, a0 and a2, respectively, has been recently reported. In the present work the Dalitz plot of this decay is fitted to a new empirical parameterization suitable for practical purposes, such as Monte Carlo simulations of K+- -> pi+- pi0 pi0 decays., Comment: 9 pages, 3 figures.

Abstract

We report the results from a study of ~23 Million K+- ==> pi+- pizero pizero decays recorded by the NA48/2 experiment at the CERN SPS, showing an anomaly in the pizero pizero invariant mass distribution in the region around 2m+, where m+ is the charged pion mass. This anomaly, never observed in previous experiments, can be interpreted as an effect due mainly to the final state charge exchange scattering process pi+ pi- ==> pizero pizero in K+- ==> pi+- pi+ pi- decay. Abstract

Prolonged therapy required to effectively treat mycobacterial infection frequently results in severe dose-limiting side-effects and drug resistance due to patient non-compliance with protracted dosage regimens. Biodegradable poly-a-hydroxy acid microspheres and microcapsules containing rifampicin (RIF) and isoniazid (INH) respectively have been prepared with the intention of providing high sustained site-specific concentrations to overcome some of the shortcomings of existing oral treatments. Due to the high dose, hydrophilicity and instability of both drugs, formulation strategies to attain high drug loading and methodologies to characterize in vitro drug release during ongoing decomposition were required. Stability indicating HPLC assays to quantify drug release have been developed, validated and applied to monitor drug release based on cumulative quantification of drug and degradates. A mathematical correction for serial decompositions associated with RIF was made based on the terminal pseudo equilibrium observed during stability studies. An isocratic HPLC assay was prospectively developed for the quantification of both drugs and their major metabolites in biological samples. Further preformulation studies confirmed the absence of significant polymorphs for both drugs when recrystallized from solvents later used in formulation development. Furthermore, thermal analysis revealed only modest interaction between the drugs and Resomer®. The high and moderate water solubilities of INH (145 mgmL-1) and RIF (1 mgmL-1) determined the selection of spray-drying (SD) and emulsion solvent evaporation (ESE) for RIF, whereas preparation of INH microcapsules relied solely on the former technique. Examination of the effects of varying RIF: polymer ratio, phase volumes and continuum presaturation with selected poly(L-lactide) and poly(D, L-lactide-co-glycolide) (PDLGA) Resomer® identified optimum conditions to maximize drug loading during a comparison of aqueous ESE with spray-drying with a range of nine further amorphous Resomer® polymers. Although yields were generally higher with ESE (85-90 %), SD (45- 75 %) was considered a superior preparation technique on the basis of the rapid production of microspheres of high and predictable drug loading (100 % of that attempted), with monodisperse granulometry and superior morphology. Release profiles were typically asymptotic characterized by a rapid `burst' of release followed by a slow release of residual entrapped RIF, irrespective of the preparative technique or polymer used. Poor yields (7.2 %) when SD low molecular weight (MW) PDLGA (8 kD) were greatly enhanced (74.8 %) by reduction in drying temperature and substitution of chloroform: dichloromethane (CFM: DCM) (1: 1) cosolvent with DCM. These conditions were adopted as the optimum parameters for further studies of blends of low (2 kD, R104) and moderate (11 kD, R202H) MW poly(D, L-lactide) (PDLLA); materials which demonstrated excellent sprayability and dramatically modulated the release of drug when combined compared to their use alone. Drug release showed a remarkable dependence on blend, dramatic acceleration being observed between 44 and 48 %w/w R104. Release over this range showed a marked dependence on medium temperature and led to the proposal of an autohydration mechanism linked to the hydrophilicity and glass transition (T9) of the blend which accounted for the sigmoidal profiles observed. First order dependence of release allowed calculation of Arrhenius derived activation energies of drug release in glassy anhydrous and rubbery plastic matrices of 630 and 320 J mol-1. Hydration and thermal studies supported the postulated diffusion mechanism, whereas granulometric and morphological examinations demonstrated that erosion did not contribute significantly. The criticality of matrix composition was further highlighted when interchange with nominally identical polymer, R202H, shifted the critical composition to 30 %w/w R104. Moreover, this observation contested the batch-to-batch reproducibility of commercial polymer. Substitution of DCM with halothane (HAL) and acetone (ACT) had a profound influence on the properties of compositionally identical ('R104: R202'H, 30: 70) microspheres, particularly release kinetics. This was attributed to the more rapid drying kinetics with the poor solvent, ACT, and the generation of a porous matrix. Consequently, drug was largely released during the 'burst' phase. Superior solvents, HAL and DCM resulted in enhanced matrix coherence at the expense of considerable residual solvent burdens (6 - 12.5 %), which allowed extensive matrix relaxation as solvent was lost with first order kinetics. This ageing process was followed by the development of an endotherm associated with the Tg as the matrix stabilized with a resultant increase in the induction period and a general retardation of drug release. Extension of the concept of blending R104 as release 'initiator' to a range of MW PDLGA of 50: 50 and 75: 25 comonomer ratio as release 'modulator' was of limited success generating release profiles reminiscent of each polymer when used alone. The magnitude of the 'burst' correlated to the precipitation kinetics of the predominant complementary PDLGA polymer as determined by cloud-point titration. Due to the more hydrophilic nature of the copolymers, at the critical concentration uncontrolled hydration resulted in a single rapid release phase. Spray-dried biodegradable INH microcapsules were prepared by a two stage process whereby SD cores of drug or in combination with biodegradable albumin or casein were subsequently coated with PDLGA by SD. The highly crystalline, aggregated and irregular morphology of SD drug resulted in poor coating efficiency and a rapid release of encapsulated drug. Protein microspheres of superior sphericity allowed more effective coating and hence slower INH release. It is concluded that SD has excellent industrial potential for the preparation of biodegradable poly-a-hydroxy acid microspheres for high dose drugs to be delivered directly to their site of action, e. g., intra-pulmonary. Indeed, the granulometry of these particles and, in particular, the hydrophilic character of blends of PDLLA described have considerable potential for the sustained delivery of drugs in the low volumes of fluid that prevails in the lung. These formulations might offset some of the limitations of current oral antimycobacterial therapy.

20. Preparation of novel modified-release dosage forms of diclofenac sodium and ibuprofen

Abstract

Mini-matrix multiple unit dosage forms (MUDFs) of diclofenac sodium and S(+) ibuprofen have been prepared. Normal tabletting techniques were used to form the mini-matrices prior to their enclosure in hard gelatin capsules. Four natural hydrophilic gums, namely xanthan, karaya, locust bean and carrageenan gums as well as hydroxypropyl methylcellulose (HPMC) were used as the principle release-retarding agents. Various excipients - lactose, Encompress®, cellulose acetate phthalate (CAP), Veegum F® and Avicel PH101® - were added in different proportions to further modify drug release. The diclofenac sodium mini-matrices (4.5 mm in diameter) were produced by the wet granulation method. The release profiles from several encapsulated minimatrices in phosphate buffer solution (pH 7.0) showed that xanthan, karaya and locust bean gums could sustain the release of diclofenac sodium while the carrageenan gum did not produce a satisfactory sustaining effect. The rank order of decreasing swelling rate in both axial and radial dimensions was xanthan > karaya > locust bean gum and each of these gums showed almost Fickian swelling behaviour. The solvent penetration rates were consistent with the swelling rates. However, the order of decreasing drug release and erosion rates was locust bean> xanthan > karaya gum. For each of these gums, the release behaviour was anomalous indicating that both Fickian drug diffusion and polymer relaxation were involved in the release process. The dominant mechanism depended on the nature and content of the gum, as well as the stage in the dissolution period. The study involving xanthan gum showed that the diclofenac sodium release rate declined linearly with a progressive increase in the gumcontent, without changing the release behaviour. However, for high drug: xanthan gum ratio (2:1), the release kinetics changed to Super Case II. Solubility differences between the excipients did not affect the release rate, but increasing proportions of each excipient produced a faster release rate with the release mechanism changing from anomalous to Case II and then to Super Case II transport. Mini-matrices containing HPMC produced faster drug release than those containing the three natural gums. There was no synergistic effect between xanthan and locust bean gums on the release of diclofenac sodium from mini-matrices. Variation in the stirring speed (used in the dissolution apparatus) and matrix volume had little effect on drug release, whereas the pH of the dissolution medium greatly affected the release of diclofenac sodium. Following on from the studies involving diclofenac sodium, xanthan and karaya gums were used to produce mini-matrices of S(+) ibuprofen. Excipients with good compressibility characteristics such as lactose, Encompress® and Avicel PH101® were needed in the formulations. At pH 7, higher drug release rates were obtained with karaya gum (Super Case II mechanism) compared with xanthan gum (anomalous behaviour). Solubility differences between the excipients slightly affected the release rate. Compression forces (11 - 26 kN) slightly affected the crushing strength. The minimatrices were relatively stable to variation in temperature (5 - 37°C) and relative humidity (10 - 75%) over a 2 month time period. These studies have shown that near zero-order release of diclofenac sodium and S(+) ibuprofen can be achieved using encapsulated mini-matrices formulations. The release mechanisms and release rates can be adjusted by variation of the type and content of gums and/or excipients.
