1. Differential expansion of T central memory precursor and effector subsets is regulated by division speed

Abstract

While antigen-primed T cells proliferate at speeds close to the physiologic maximum of mammalian cells, T cell memory is maintained in the absence of antigen by rare cell divisions. The transition between these distinct proliferative programs has been difficult to resolve via population-based analyses. Here, we computationally reconstruct the proliferative history of single CD8+ T cells upon vaccination and measure the division speed of emerging T cell subsets in vivo. We find that slower cycling central memory precursors, characterized by an elongated G1 phase, segregate early from the bulk of rapidly dividing effector subsets, and further slow-down their cell cycle upon premature removal of antigenic stimuli. In contrast, curtailed availability of inflammatory stimuli selectively restrains effector T cell proliferation due to reduced receptivity for interleukin-2. In line with these findings, persistence of antigenic but not inflammatory stimuli throughout clonal expansion critically determines the later size of the memory compartment.

Abstract

The flat spectrum radio quasar (FSRQ) PKS 1510-089 is known for its complex multiwavelength behavior, and is one of only a few FSRQs detected at very high energy (VHE, $E>100\,$GeV) $\gamma$-rays. VHE $\gamma$-ray observations with H.E.S.S. and MAGIC during late May and early June 2016 resulted in the detection of an unprecedented flare, which reveals for the first time VHE $\gamma$-ray intranight variability in this source. While a common variability timescale of $1.5\,$hr is found, there is a significant deviation near the end of the flare with a timescale of $\sim 20\,$min marking the cessation of the event. The peak flux is nearly two orders of magnitude above the low-level emission. For the first time, curvature is detected in the VHE $\gamma$-ray spectrum of PKS 1510-089, which is fully explained through absorption by the extragalactic background light. Optical R-band observations with ATOM reveal a counterpart of the $\gamma$-ray flare, even though the detailed flux evolution differs from the VHE ightcurve. Interestingly, a steep flux decrease is observed at the same time as the cessation of the VHE flare. In the high energy (HE, $E>100\,$MeV) $\gamma$-ray band only a moderate flux increase is observed with Fermi-LAT, while the HE $\gamma$-ray spectrum significantly hardens up to a photon index of 1.6. A search for broad-line region (BLR) absorption features in the $\gamma$-ray spectrum indicates that the emission region is located outside of the BLR. Radio VLBI observations reveal a fast moving knot interacting with a standing jet feature around the time of the flare. As the standing feature is located $\sim 50\,$pc from the black hole, the emission region of the flare may have been located at a significant distance from the black hole. If this correlation is indeed true, VHE $\gamma$ rays have been produced far down the jet where turbulent plasma crosses a standing shock., Comment: 25 pages, 17 figures. Abstract

Context: MWC 656 has recently been established as the first observationally detected high-mass X-ray binary system containing a Be star and a black hole (BH). The system has been associated with a gamma-ray flaring event detected by the AGILE satellite in July 2010. Aims: Our aim is to evaluate if the MWC 656 gamma-ray emission extends to very high energy (VHE > 100 GeV) gamma rays. Methods. We have observed MWC 656 with the MAGIC telescopes for $\sim$23 hours during two observation periods: between May and June 2012 and June 2013. During the last period, observations were performed contemporaneously with X-ray (XMM-Newton) and optical (STELLA) instruments. Results: We have not detected the MWC 656 binary system at TeV energies with the MAGIC Telescopes in either of the two campaigns carried out. Upper limits (ULs) to the integral flux above 300 GeV have been set, as well as differential ULs at a level of $\sim$5% of the Crab Nebula flux. Abstract

The notion of Multi-Agent System environment is currently considered as a mediating entity, functioning as enabler but possibly also as a manager and constrainer of agent actions, perceptions, and interactions. In this paper, we analyze how the environment could be a first class abstraction to support the building, the development and the management of Agreements in decentralized and open systems between autonomous agents. To this aim we analyze the synergies between the environment and the foundational dimensions of agreement technologies such as semantics, norms, organizations, argumentation & negotiation, trust.

The execution of process models is usually presented through a graphical user interface, especially when users' input is required. Existing mechanisms, such as Electronic Institutions (EIs), provide means to easily specify and automatically execute process models. However, every time the specification is modified, the graphical user interface (GUI) needed during the execution stage should be manually modified accordingly. This article proposes a system that helps maintain such GUIs in an efficient and automated manner. Abstract

The notion of Multi-Agent System (MAS) environment, as remarked by recent literature, has gained a key role, becoming a mediating entity, functioning as enabler but possibly also as a manager and constrainer of agent actions, perceptions, and interactions1 while addressing the requirements of openness and scalability. According to such a perspective, the environment is not a merely passive source of agent perceptions and target of agent actions which is, actually, the dominant perspective in agency, but a first-class abstraction that can be suitably designed to encapsulate some fundamental functionalities and services, such as coordination and organization, besides agent mobility, communications, security, etc [2]. Then, the environment dimension appears to intersect with all the dimensions that should be addressed to define an agreement between autonomous agents, that is, all the di erent Agreement Technologies giving support to the building, development and management of agreements in decentralized and open systems between autonomous agents. Those dimensions are the ones related to the development of technologies dealing with: Semantics, Norms, Organizations, Argumentation & Negotiation, and Trust. Though some works have already been done on the connections between environment and organizations or norms, the links and interactions of the environment with the other dimensions of these agreement technologies are still to be explored.

Abstract

The simulation of pedestrian dynamics is a consolidated area of application for agent-based models: successful case studies can be found in the literature and off-the-shelf simulators are commonly employed by end-users, decision makers and consultancy companies. These models, however, generally neglect or treat in a simplistic way aspects like (i) the impact of cultural heterogeneity among individuals and (ii) the effects of the presence of groups and particular relationships among pedestrians. This work is aimed, on one hand, at introducing some fundamental anthropological considerations on which most pedestrian models are based, and in particular Edward T. Hall's work on proxemics. On the other hand, the paper describes an agent-based model encapsulating in the pedestrian's behavioural model effects representing both proxemics and a simplified account of influences related to the presence of groups in the crowd. The model is tested in a simple scenario to evaluate the implications of some modeling choices and the presence of groups in the simulated scenario. Results are discussed and compared to experimental observations and to data available in the literature.

Abstract

El trabajo reportado en este artículo forma parte de un proyecto de investigación en el cual se tratan de desarrollar aceros de herramientas para conformado en caliente con prestaciones optimizadas. Para ello se comenzó por caracterizar el comportamiento mecánico de diversos aceros comerciales de gran utilización para tener una referencia clara. En este contexto, el artículo presenta los resultados relativos a la resistencia a fatiga de dos aceros martensíticos evaluada a 450 ºC, por ser ésta una probable temperatura de trabajo de los mismos. Se trata de aceros desarrollados para operaciones de conformado a alta temperatura, tales como moldes para la inyección de aleaciones ligeras, correspondiendo a los grados DIN W.Nr. 1.2344 (también conocido como AISI H13) y 1.2367, aleados con entorno a 5% de Cr y diferentes porcentajes de Mo y V. Para el primero de ellos se consideraron dos condiciones de tratamiento térmico, una que condujo a unas durezas de 52 HRc y otra que dejó al acero a un nivel inferior de dureza (44 HRc). Se efectuó en primer lugar una caracterización microestructural, así como ensayos de tracción a la temperatura seleccionada de 450 ºC. Los ensayos de fatiga se realizaron en control de carga, con una relación de esfuerzos de R = 0.1 y en atmósfera inerte para eliminar los efectos de la oxidación superficial. El criterio de vida infinita se fijó en 2x105 ciclos, por ser éste número acorde con el número de ciclos de inyección que puede llegar a alcanzar un molde de inyección de aluminio. Un completo estudio fractográfico permitió localizar los lugares de iniciación de las roturas por fatiga. Mientras en algunos casos la nucleación se produjo a partir de carburos más o menos próximos a la superficie, en otros casos se observó que el origen del proceso de fractura estaba en inclusiones no metálicas., Postprint (published version)
