Chemical probes that covalently modify cysteine residues in a protein-specific manner are valuable tools for biological investigations. Covalent fragments are increasingly implemented as probe starting points, but the complex relationship between fragment structure and binding kinetics makes covalent fragment optimization uniquely challenging. We describe a new technique in covalent probe discovery that enables data-driven optimization of covalent fragment potency and selectivity. This platform extends beyond the existing repertoire of methods for identifying covalent fragment hits by facilitating rapid multiparameter kinetic analysis of covalent structure-activity relationships through the simultaneous determination of Ki , kinact and intrinsic reactivity. By applying this approach to develop novel probes against electrophile-sensitive kinases, we showcase the utility of the platform in hit identification and highlight how multiparameter kinetic analysis enabled a successful fragment-merging strategy.

Soteria is a user right management system designed to safeguard user-data privacy in a transparent and provable manner in compliance to regulations such as GDPR and CCPA. Soteria represents user data rights as formal executable sharing agreements, which can automatically be translated into a human readable form and enforced as data are queried. To support revocation and to prove compliance, an indelible, audited trail of the hash of data access and sharing agreements are stored on a two-layer distributed ledger. The main chain ensures partition tolerance and availability (PA) properties while side chains ensure consistency and availability (CA), thus providing the three properties of the CAP (consistency, availability, and partition tolerance) theorem. The neural architecture search (NAS) algorithm with reinforcement learning can be a powerful and novel framework for the automatic discovering process of neural architectures. However, its application is restricted by noncontinuous and high-dimensional search spaces, which result in difficulty in optimization. To resolve these problems, we proposed NAS in embedding space (NASES), which is a novel framework. Unlike other NAS with reinforcement learning approaches that search over a discrete and high-dimensional architecture space, this approach enables reinforcement learning to search in an embedding space by using architecture encoders and decoders. The current experiment demonstrated that the performance of the final architecture network using the NASES procedure is comparable with that of other popular NAS approaches for the image classification task on CIFAR-10. The results of the experiment were efficient and indicated that NASES was highly efficient to discover final architecture only in $<$3.5 GPU hours. Multi-output is essential in machine learning that it might suffer from nonconforming residual distributions, i.e., the multi-output residual distributions are not conforming to the expected distribution. In this paper, we propose "Wrapped Loss Function" to wrap the original loss function to alleviate the problem. This wrapped loss function acts just like the original loss function that its gradient can be used for backpropagation optimization. Empirical evaluations show wrapped loss function has advanced properties of faster convergence, better accuracy, and improving imbalanced data., Comment: 10 pages

Atherosclerosis-related diseases are the leading cause of morbidity or mortality in the world. They result in serious outcomes such as sudden cardiac death, unstable angina pectoris, acute myocardial infarction, stroke, or intermittent claudication due to vessel obliteration or plaque rupture with subsequent thrombosis. There are some limitations with standard treatments such as antiplatelet drugs, angiotensin-converting enzyme inhibitors, beta-blockers, coronary artery bypass surgery, and percutaneous transluminal coronary angioplasty. Therefore, complementary and alternative medicine is necessary for medication. Traditional Chinese medicine is the main complementary therapy used in the Chinese community. This article aims to explore complementary therapy with traditional Chinese medication for atherosclerosis-related diseases. There is some scientific evidence to support that traditional Chinese medicine could treat atherosclerosis and its associated conditions. Acupuncture through needling on ST36, ST40, PC6, or BL15 could alleviate atherosclerosis-related cardiovascular diseases. Tai chi and meditation have beneficial effects for mental and physical health. In addition, extracts or compounds of single Chinese herbs such as Salvia miltiorrhiza, Panax notoginseng, Ginkgo biloba, Curcuma longa, Crataegus pinnatifida, Paeonia lactiflora, Prunella vulgaris, Polygonum multiflorum, Coptis chinensis, and red yeast rice also could treat atherosclerosis-related diseases through their endothelial protective, antioxidative, anti-inflammatory, inhibiting of smooth muscle cells proliferation, and lipid-lowering effects. In accordance with evidence-based medicine, well-designed and conducted clinical studies such as randomized control clinical trials will be necessary in the future.
