The hidden presence of opportunistic bacterial pathogens in the environment evokes concerns about emerging diseases, especially in the light of climate change. The co-evolution of bacteria and their predators (protozoa) has led to bacterial defence strategies of which some contribute to the ability of bacteria to cause disease. To increase our understanding of the interplay between bacteria, protozoa, land use, and climate scenarios in Nordic brackish and freshwater, four studies were designed. The first study explored the co-occurrence patterns between predation resistant bacteria (PRB) and bacterivorous protozoa in a coastal area in the northern Baltic Sea. The results showed higher PRB diversity in the bays and freshwater inlets, than in the offshore waters. Further, genotype specific interactions between protozoa and bacteria were identified. The second study focused on Legionella species diversity and their association with humic substances and low salinity, potentially facilitated through the promotion of the heterotrophic microbial food web or by iron availability. The third study examined the impact of intensified land use on bacterial taxa abundance and community composition in lake inflows, demonstrating indirect downstream effects on water quality. Factors such as pastures, fields, farms, aluminium, iron, and humic substances were linked to increased Legionella abundance. The fourth study exposed aquatic organisms to climate change scenarios, causing eutrophication or brownification with elevated iron levels. Pseudomonas aeruginosa were found to be especially persistent to iron, likely linked to the same mechanism that enables survival in protozoan cells. This trait was shared with other observed intracellular pathogens and uncultured species, who showed elevated resilience to brownification and ability to survive outside host cells. The upcoming Comet Interceptor mission involves a parking phase around the Sun-Earth L2 point before transferring to intercept the orbit of a long period comet, interstellar object or a back-up target in the form of a short-period comet. The target is not certain to be known before the launch in 2029. During the parking phase there may thus arise a scenario wherein a decision needs to be taken of whether to go for a particular comet or whether to discard that option in the hope that a better target will appear within a reasonable time frame later on. Context. The Rosetta spacecraft followed comet 67P/Churyumov-Gerasimenko (67P) for more than two years at a slow walking pace (similar to 1 m s(-1)) within 1500 km from the nucleus. During one of the radial movements of the spacecraft in the early phase of the mission, the radial distribution of the plasma density could be estimated, and the ionospheric density was found to be inversely proportional to the cometocentric distance r from the nucleus (a 1/r distribution). Aims. This study aims to further characterise the radial distribution of plasma around 67P throughout the mission and to expand on the initial results. We also aim to investigate how a 1/r distribution would be observed during a flyby with a fast (similar to 10's km s(-1)) spacecraft, such as the upcoming Comet Interceptor mission, when there is also an asymmetry introduced to the outgassing over the comet surface. Methods. To determine the radial distribution of the plasma, we used data from the Langmuir probe and Mutual Impedance instruments from the Rosetta Plasma Consortium during six intervals throughout the mission, for which the motion of Rosetta was approximately radial with respect to the comet. We then simulated what distribution a fast flyby mission would actually observe during its passage through a coma when there is a 1/r plasma density distribution as well as a sinusoidal variation with a phase angle (and then a sawtooth variation) multiplied to the outgassing rate. Results. The plasma density around comet 67P is found to roughly follow a 1/r dependence, although significant deviations occur in some intervals. If we normalise all data to a common outgassing rate (or heliocentric distance) and combine the intervals to a radial range of 10-1500 km, we find a 1/r(1.19) average distribution. The simulated observed density from a fast spacecraft flying through a coma with a 1/r distribution and an asymmetric outgassing can, in fact, appear anywhere in the range from a 1/r distribution to a 1

Moment analysis of ion spectrograms measured by the Ion Composition Analyser (ICA) in the coma of comet 67P typically produces an ion number density estimate markedly lower than the number density of free electrons as measured by the Mutual Impedance Probe and the dual Langmuir Probe. While there are good reasons to distrust the ion density moment estimate in these circumstances, the issue cannot yet be considered fully understood and it is of interest to see whether any natural non-instrumental cause is possible. An obvious such cause would be whether a significant fraction of the positive charge density resides in positively charged dust grains that are not measured by the ICA. Here, we show that this scenario is highly unlikely, even near perihelion where photoemission is the strongest. In our semi-analytical grain charging model, we balance the current contributions to grains of photoelectron emission and electron attachment so as to find the expected charge state for a grain of a given radius. The charge state is affected by the ambient electron number density, the electron temperature, and the heliocentric distance. While at times the bulk of the dust population around comet 67P could be charged positive, dust charging, including photoelectron emission, should have a negligible influence on the overall ionization balance in the cometary coma simply because the dust particles are not ubiquitous enough.

Some of the predation-resistant bacteria (PRB) are also pathogenic to humans and other mammals. The links between PRB and protozoa in natural aquatic systems are poorly known, but they are important in predicting outbreaks and determining the long-term consequences of a contamination event. To elucidate co-occurrence patterns between PRB (16S rRNA) and bacterivorous protozoa (18S rRNA), we performed a field study in a coastal area in the northern Baltic Sea. Interactions between bacteria and protozoa were explored by using two complementary statistical tools. We found co-occurrence patterns between specific PRB and protozoa, such as Legionella and Ciliophora, and we also found that the interactions are genotype-specific as, for example, Rickettsia. The PRB sequence diversity was larger in bays and freshwater inlets compared to offshore sites, indicating local adaptions. Considering the PRB diversity in the freshwater in combination with the large spring floods in the area, freshwater influxes should be considered a potential source of PRB in the coastal northern Baltic Sea. These findings are relevant for the knowledge of survival and dispersal of potential pathogens in the environment.

The environment of a comet is a fascinating and unique laboratory to study plasma processes and the formation of structures such as shocks and discontinuities from electron scales to ion scales and above. The European Space Agency's Rosetta mission collected data for more than two years, from the rendezvous with comet 67P/Churyumov-Gerasimenko in August 2014 until the final touch-down of the spacecraft end of September 2016. This escort phase spanned a large arc of the comet's orbit around the Sun, including its perihelion and corresponding to heliocentric distances between 3.8 AU and 1.24 AU. The length of the active mission together with this span in heliocentric and cometocentric distances make the Rosetta data set unique and much richer than sets obtained with previous cometary probes. Here, we review the results from the Rosetta mission that pertain to the plasma environment. We detail all known sources and losses of the plasma and typical processes within it. The findings from in-situ plasma measurements are complemented by remote observations of emissions from the plasma. Overviews of the methods and instruments used in the study are given as well as a short review of the Rosetta mission. The long duration of the Rosetta mission provides the opportunity to better understand how the importance of these processes changes depending on parameters like the outgassing rate and the solar wind conditions. We discuss how the shape and existence of large scale structures depend on these parameters and how the plasma within different regions of the plasma environment can be characterised. We end with a non-exhaustive list of still open questions, as well as suggestions on how to answer them in the future.

We present a semianalytical photochemical model of Saturn's near-equatorial ionosphere and adapt it to two regions (similar to 2200 and similar to 1700 km above the 1 bar level) probed during the inbound portion of Cassini's orbit 292 (2017 September 9). The model uses as input the measured concentrations of molecular hydrogen, hydrogen ion species, and free electrons, as well as the measured electron temperature. The output includes upper limits, or constraints, on the mixing ratios of two families of molecules, on ion concentrations, and on the attachment rates of electrons and ions onto dust grains. The model suggests mixing ratios of the two molecular families that, particularly near similar to 1700 km, differ notably from what independent measurements by the Ion Neutral Mass Spectrometer suggest. Possibly connected to this, the model suggests an electron-depleted plasma with a level of electron depletion of around 50%. This is in qualitative agreement with interpretations of Radio Plasma Wave Science/Langmuir Probe measurements, but an additional conundrum arises in the fact that a coherent photochemical equilibrium scenario then relies on a dust component with typical grain radii smaller than 3 angstrom.

Context. The Rosetta spacecraft followed comet 67P/Churyumov-Gerasimenko (67P) for more than two years at a slow walking pace (similar to 1 m s(-1)) within 1500 km from the nucleus. During one of the radial movements of the spacecraft in the early phase of the mission, the radial distribution of the plasma density could be estimated, and the ionospheric density was found to be inversely proportional to the cometocentric distance r from the nucleus (a 1/r distribution). Aims. This study aims to further characterise the radial distribution of plasma around 67P throughout the mission and to expand on the initial results. We also aim to investigate how a 1/r distribution would be observed during a flyby with a fast (similar to 10's km s(-1)) spacecraft, such as the upcoming Comet Interceptor mission, when there is also an asymmetry introduced to the outgassing over the comet surface. Methods. To determine the radial distribution of the plasma, we used data from the Langmuir probe and Mutual Impedance instruments from the Rosetta Plasma Consortium during six intervals throughout the mission, for which the motion of Rosetta was approximately radial with respect to the comet. We then simulated what distribution a fast flyby mission would actually observe during its passage through a coma when there is a 1/r plasma density distribution as well as a sinusoidal variation with a phase angle (and then a sawtooth variation) multiplied to the outgassing rate. Results. The plasma density around comet 67P is found to roughly follow a 1/r dependence, although significant deviations occur in some intervals. Some of the predation-resistant bacteria (PRB) are also pathogenic to humans and other mammals. The links between PRB and protozoa in natural aquatic systems are poorly known, but they are important in predicting outbreaks and determining the long-term consequences of a contamination event. To elucidate co-occurrence patterns between PRB (16S rRNA) and bacterivorous protozoa (18S rRNA), we performed a field study in a coastal area in the northern Baltic Sea. Interactions between bacteria and protozoa were explored by using two complementary statistical tools. We found co-occurrence patterns between specific PRB and protozoa, such as Legionella and Ciliophora, and we also found that the interactions are genotype-specific as, for example, Rickettsia. The PRB sequence diversity was larger in bays and freshwater inlets compared to offshore sites, indicating local adaptions. Considering the PRB diversity in the freshwater in combination with the large spring floods in the area, freshwater influxes should be considered a potential source of PRB in the coastal northern Baltic Sea. These findings are relevant for the knowledge of survival and dispersal of potential pathogens in the environment.

Background: This is the study plan of the Karolinska NeuroCOVID study, a study of neurocognitive impairment after severe COVID-19, relating post-intensive care unit (ICU) cognitive and neurological deficits to biofluid markers and MRI. The COVID-19 pandemic has posed enormous health challenges to individuals and health- systems worldwide. An emerging feature of severe COVID-19 is that of temporary and extended neurocognitive impairment, exhibiting a myriad of symptoms and signs. The causes of this symptomatology have not yet been fully elucidated. Methods: In this study, we aim to investigate patients treated for severe COVID-19 in the ICU, as to describe and relate serum-, plasma- and cerebrospinal fluid-borne molecular and cellular biomarkers of immune activity, coagulopathy, cerebral damage, neuronal inflammation, and degeneration, to the temporal development of structural and functional changes within the brain as evident by serial MRI and extensive cognitive assessments at 3-12 months after ICU discharge. Results: To date, we have performed 51 3-month follow-up MRIs in the ICU survivors. Of these, two patients (similar to 4%) have had incidental findings on brain MRI findings requiring activation of the Incidental Findings Management Plan. Furthermore, the neuropsychological and neurological examinations have so far revealed varying and mixed patterns. Several patients expressed cognitive and/or mental concerns and fatigue, complaints closely related to brain fog. Conclusion: The study goal is to gain a better understanding of the pathological mechanisms and neurological consequences of this new disease, with a special emphasis on neurodegenerative and neuroinflammatory processes, in order to identify targets of intervention and rehabilitation.

Background: Postoperative neurocognitive decline is a frequent complication in adult patients undergoing major surgery with increased risk for morbidity and mortality. The mechanisms behind cognitive decline after anaesthesia and surgery are not known. We studied the association between CSF and blood biomarkers of neuronal injury or brain amyloidosis and long-term changes in neurocognitive function. Methods: In patients undergoing major orthopaedic surgery (knee or hip replacement), blood and CSF samples were obtained before surgery and then at 4, 8, 24, 32, and 48 h after skin incision through an indwelling spinal catheter. CSF and blood concentrations of total tau (T-tau), neurofilament light, neurone-specific enolase and amyloid β (Aβ1-42) were measured. Neurocognitive function was assessed using the International Study of Postoperative Cognitive Dysfunction (ISPOCD) test battery 1–2 weeks before surgery, at discharge from the hospital (2–5 days after surgery), and at 3 months after surgery. Results: CSF and blood concentrations of T-tau, neurone-specific enolase, and Aβ1-42 increased after surgery. A similar increase in serum neurofilament light was seen with no overall changes in CSF concentrations. There were no differences between patients having a poor or good late postoperative neurocognitive outcome with respect to these biomarkers of neuronal injury and Aβ1-42. Conclusions: The findings of the present explorative study showed that major orthopaedic surgery causes a release of CSF markers of neural injury and brain amyloidosis, suggesting neuronal damage or stress. We were unable to detect an association between the magnitude of biomarker changes and long-term postoperative neurocognitive dysfunction.

During its two-year mission at comet 67P, Rosetta nearly continuously monitored the inner coma plasma environment for gas production rates varying over three orders of magnitude, at distances to the nucleus from a few to a few hundred km. To achieve the best possible measurements, cross-calibration of the plasma instruments is needed. We construct with two different physical models to cross-calibrate the electron density as measured by the Mutual Impedance Probe (MIP) to the ion current and spacecraft potential as measured by the Rosetta Langmuir Probe (LAP), the latter validated with the Ion Composition Analyser (ICA). We retrieve a continuous plasma density dataset for the entire cometary mission with a much improved dynamical range compared to any plasma instrument alone and, at times, improve the temporal resolution from 0.24-0.74~Hz to 57.8~Hz. The new density dataset is consistent with the existing MIP density dataset and covers long time periods where densities were too low to be measured by MIP. The physical model also yields, at 3~hour time resolution, ion flow speeds as well as a proxy for the solar EUV flux from the photoemission from the Langmuir Probes. We report on two independent mission-wide estimates of the ion flow speed which are consistent with the bulk H$_2$O$^+$ ion velocities as measured by ICA. We find the ion flow to consistently be much faster than the neutral gas over the entire mission, lending further evidence that the ions are collisionally decoupled from the neutrals in the coma. RPC measurements of ion speeds are therefore not consistent with the assumptions made in previously published plasma density models of the comet ionosphere at the start and end of the mission. Also, the measured EUV flux is perfectly consistent with independently derived values previously published from Johansson et al. In particular, we investigate the possibility of using Solar Orbiter’s DC electric and magnetic field data to estimate the solar wind speed. Methods. We used a deHoffmann-Teller (HT) analysis, based on measurements of the electric and magnetic fields, to find the velocity of solar wind current sheets, which minimises a single component of the electric field. By comparing the HT velocity to the proton velocity measured by the Proton and Alpha particle Sensor (PAS), we have developed a simple model for the effective antenna length, Leff of the E-field probes. We then used the HT method to estimate the speed of the solar wind. Results. Using the HT method, we find that the observed variations in Ey are often in excellent agreement with the variations in the magnetic field. The magnitude of Ey, however, is uncertain due to the fact that the Leff depends on the plasma environment. Here, we derive an empirical model relating Leff to the Debye length, which we can use to improve the estimate of Ey and, consequently, the estimated solar wind speed. Conclusions. The low-frequency electric field provided by RPW is of high quality. Using the deHoffmann-Teller analysis, Solar Orbiter’s magnetic and electric field measurements can be used to estimate the solar wind speed when plasma data are unavailable.

Context. The recently released spacecraft potential measured by the RPW instrument on board Solar Orbiter has been used to estimate the solar wind electron density in the inner heliosphere. Aims. The measurement of the solar wind’s electron density, taken in June 2020, has been analysed to obtain a thorough characterization of the turbulence and intermittency properties of the fluctuations. Magnetic field data have been used to describe the presence of ion-scale waves. Methods. To study and quantify the properties of turbulence, we extracted selected intervals. We used empirical mode decomposition to obtain the generalized marginal Hilbert spectrum, equivalent to the structure functions analysis, which additionally reduced issues typical of non-stationary, short time series. The presence of waves was quantitatively determined by introducing a parameter describing the time-dependent, frequency-filtered wave power. Results. A well-defined inertial range with power-law scalng was found almost everywhere in the sample studied. However, the Kolmogorov scaling and the typical intermittency effects are only present in fraction of the samples. Other intervals have shallower spectra and more irregular intermittency, which are not described by models of turbulence. These are observed predominantly during intervals of enhanced ion frequency wave activity. Comparisons with compressible magnetic field intermittency (from the MAG instrument) and with an estimate of the solar wind velocity (using electric and magnetic field) are also provided to give general context and help determine the cause of these anomalous fluctuations.

Wakes behind spacecraft caused by supersonic drifting positive ions are common in plasmas and disturb in situ measurements. We review the impact of wakes on observations by the Electric Field and Wave double-probe instruments on the Cluster satellites. In the solar wind, the equivalent spacecraft charging is small compared to the ion drift energy and the wake effects are caused by the spacecraft body and can be compensated for. We present statistics of the direction, width, and electrostatic potential of wakes, and we compare with an analytical model. In the low-density magnetospheric lobes, the equivalent positive spacecraft charging is large compared to the ion drift energy and an enhanced wake forms. In this case observations of the geophysical electric field with the double-probe technique becomes extremely challenging. Rather, the wake can be used to estimate the flux of cold (eV) positive ions. For an intermediate range of parameters, when the equivalent charging of the spacecraft is similar to the drift energy of the ions, also the charged wire booms of a double-probe instrument must be taken into account. We discuss an example of these effects from the MMS spacecraft near the magnetopause. We find that many observed wake characteristics provide information that can be used for scientific studies. An important example is the enhanced wakes used to estimate the outflow of ionospheric origin in the magnetospheric lobes to about 10 26 cold (eV) ions/s, constituting a large fraction of the mass outflow from planet Earth.

Context. During its two-year mission at comet 67P, Rosetta nearly continuously monitored the inner coma plasma environment for gas production rates varying over three orders of magnitude, at distances to the nucleus ranging from a few to a few hundred kilometres. To achieve the best possible measurements, cross-calibration of the plasma instruments is needed. Aims. Our goal is to provide a consistent plasma density dataset for the full mission, while in the process providing a statistical characterisation of the plasma in the inner coma and its evolution. Methods. We constructed physical models for two different methods to cross-calibrate the spacecraft potential and the ion current as measured by the Rosetta Langmuir probes (LAP) to the electron density as measured by the Mutual Impedance Probe (MIP). We also described the methods used to estimate spacecraft potential, and validated the results with the Ion Composition Analyser (ICA). Results. We retrieve a continuous plasma density dataset for the entire cometary mission with a much improved dynamical range compared to any plasma instrument alone and, at times, improve the temporal resolution from 0.24-0.74 Hz to 57.8 Hz. The physical model also yields, at a three-hour time resolution, ion flow speeds and a proxy for the solar EUV flux from the photoemission from the Langmuir probes. Conclusions. We report on two independent mission-wide estimates of the ion flow speed that are consistent with the bulk H2O+ ion velocities as measured by the ICA. We find the ion flow to consistently be much faster than the neutral gas over the entire mission, lending further evidence that the ions are collisionally decoupled from the neutrals in the coma. Measurements of ion speeds from Rosetta are therefore not consistent with the assumptions made in previously published plasma density models of the comet 67P's ionosphere at the start and end of the mission. The molecular mechanisms of obstructive sleep apnea (OSA), in particular the gene expression patterns in whole blood of patients with OSA, can shed more light on the underlying pathophysiology of OSA and suggest potential biomarkers. In the current study, we have enrolled thirty patients with untreated moderate-severe OSA together with 20 BMI, age, and sex-matched controls and 15 normal-weight controls. RNA-sequencing of whole blood and home sleep apnea testing were performed in the untreated state and after three and twelve months of continuous positive airway pressure (CPAP) treatment. Analysis of the whole blood transcriptome of the patients with OSA revealed a unique pattern of differential expression with a significant number of downregulated immune-related genes including many heavy and light chain immunoglobulins and interferon-inducible genes. This was confirmed by the gene ontology analysis demonstrating enrichment with the biological processes associated with various immune functions. Expression of these genes was recovered after three months of CPAP treatment. After 12 months of CPAP treatment, the overall gene expression profile returns to the initial, untreated level. In addition, we have confirmed the importance of choosing BMI-matched controls as a reference group as opposed to normal-weight healthy individuals based on the significantly different gene expression signatures between these two groups.

A magnetosphere may form around an object in a stellar wind either due to the intrinsic magnetic field of theobject or stellar wind interaction with the ionosphere of the object. Comets represent the most variable magnetospheres in our solar system, and through the Rosetta mission we have had the chance to study the birth and evolution of a comet magnetosphere as the comet nucleus approached the Sun. We review the birth of the cometmagnetosphere as observed at comet 67P Churyumov–Gerasimenko, the formation of plasma boundariesand how the solar wind–atmosphere interaction changes character as the cometary gas cloud and magnetospheregrow in size. Mass loading of the solar wind leads to an asymmetric deflection of the solar wind for low outgassingrates. With increasing activity a solar wind ion cavity forms. Intermittent shock-like features were also observed.For intermediate outgassing rate a diamagnetic cavity is formed inside the solar wind ion cavity, thus well separated from the solar wind. The cometary plasma was typically very structured and variable. The region of thecoma dense enough to have significant collisions forms a special region with different ion chemistry and plasmadynamics as compared to the outer collision-free region.

Context. Solar Orbiter was launched on 10 February 2020 with the purpose of investigating solar and heliospheric physics using a payload of instruments designed for both remote and in situ studies. Similar to the recently launched Parker Solar Probe, and unlike earlier missions, Solar Orbiter carries instruments designed to measure low-frequency DC electric fields. Aims.
