Die Einrichtung von Archiven zur Archivierung und Nachnutzung qualitativer Forschungsdaten wurde vonseiten qualitativer Sozialforscher*innen mit intensiven Debatten um konzeptuelle und methodologische Fragen begleitet. Die konkrete Forschungspraxis blieb dabei jedoch meist ausgeklammert, obwohl sich Erfolg und Mehrwert der Datenarchivierung letztlich an der Praxis des Data Sharings und der Sekundärnutzung bemessen. Wir nehmen deshalb eigene praktische Erfahrungen mit der Sekundärnutzung archivierter Interviewdaten in einem methodologischen Forschungsprojekt zur qualitativen Interviewforschung zum Anlass, organisatorische, method(olog)ische und epistemologische Reflexionen zur Datenarchivierung anzustellen. Anhand verschiedener Prozesse der Beschaffung und Prüfung geeigneter Interviewtranskripte zeigen wir, wie sich die Herstellung des Data Fit in Sekundärforschungsprojekten innerhalb einer bedeutungsstiftenden kommunikativen Triade von Primärforschenden, Archivar*innen und Sekundärforschenden vollzogen hat., The creation of archives to preserve and share qualitative research data has been accompanied by intense conceptual debates and methodological questions among scholars. Although the success of archiving data can finally be measured by the practices of data sharing and secondary use, the actual practices and experiences of working with this kind of data have been neglected in discussions. Thus, we utilize our experiences in a methodological research project on qualitative interviews to reflect on organizational, methodological and epistemic aspects of archiving qualitative data. By presenting processes of how we found and reviewed archived interview transcripts in our project, we illustrate how the production of data fit in secondary analysis is performed within a communicative triad of primary researchers, secondary researchers, and archivists.

Interviews appear to be the qualitative research method of choice but have rarely been the subject of qualitative methodological research. Therefore, it is largely unclear what exactly happens during an interview and how interviews are empirically constituted as communication and interaction situations. This research gap is the focus of ROULSTON's anthology "Interactional Studies of Qualitative Research Interviews," which I discuss in this review essay. Over the course of 12 contributions, the authors present ethnomethodologically inspired (re)analyses of interactional dynamics, social actions, and conversational resources in interviews. The volume is particularly worth reading, and the contributors (once again) demonstrate the utility of an ethnomethodological and conversation analysis-based perspective for understanding interview interactions. This can inspire researchers to examine interactional dynamics in their own projects more intensively. For researchers interested in the methodological discussion on interview research, this book offers not only insights into the state of the art but also suggestions for future research in the field of "social studies of interview studies" (RAPLEY 2012, p.552). If this research field is to be consolidated, I propose more systematic studies of interview research, the consideration of different research traditions, and the intensification of the international exchange on interview research., Interviews erscheinen als die qualitative Forschungsmethode der Wahl, sind selbst jedoch bislang nur selten Gegenstand qualitativer Methodenforschung geworden. Weitgehend unklar ist daher, was in qualitativen Interviews eigentlich genau geschieht und wie Interviews als Kommunikations- und Interaktionssituationen empirisch konstituiert werden. Mit dem in diesem Review Essay besprochenen Sammelband "Interactional Studies of Qualitative Research Interviews" setzt ROULSTON an dieser Forschungslücke an. In 12 Beiträgen widmen sich die Autor*innen ethnomethodologisch inspirierten (Re-)Analysen interaktiver Dynamiken, sozialer Handlungen und konversationeller Ressourcen in vorwiegend mündlich und Face to Face durchgeführten Interviews. Insgesamt liegt ein sehr lesenswerter Band vor, mit dem (erneut) die Leistungsfähigkeit einer ethnomethodologisch und konversationsanalytisch geschulten Perspektive für das Verständnis von Interviewinteraktionen demonstriert wird und dadurch Forschende angeregt werden können, in ihren eigenen Projekten Interaktionsdynamiken intensiver zu untersuchen. Für Forschende, die an einer methodologischen Auseinandersetzung mit Interviewforschung interessiert sind, bietet der Band nicht nur Einblicke in den interaktionstheoretischen State of the Art, sondern auch Anregungen für zukünftige Untersuchungen im Bereich der "social studies of interview studies" (RAPLEY 2012, S.552). Soll die Forschung zur Interviewforschung als Feld konsolidiert werden, schlage ich vor, die bislang dominierenden reflexiven Auseinandersetzungen mit eigenen Forschungserfahrungen durch systematische Studien zur Interviewforschung zu ergänzen, die Perspektiven verschiedener Forschungstraditionen einzubeziehen und den internationalen Austausch zur Interviewforschung weiter zu stärken.

Despite the social normalisation and moral de-problematisation of separation and divorce, they are usually anything but normal or unproblematic events for those affected. The events have to be ordered, the separation needs to be legitimised and one's own identity requires rehabilitation; this can result in striking narrative dynamics in qualitative interviews dealing with separation. Previous research has revealed the connection between the form, content, and function of separation talk as well as social differentiation: role, gender, and socio-structural factors. To the functional analyses of the narratives, of 46 interviews (23 ex-couples) in our study "Couples after separation", we add, as a further and superordinate differentiation, the category of social milieu. We exemplify our argument that separation talk varies depending on this dimension by exploring the contrasting individualised and traditional milieus. They differ in a fundamental way in their relationship models and corresponding separation legitimations, and in their plausibility strategies and in their conception of what constitutes a desirable identity. Beyond the concrete subject of separation and separation talk, we also touch on two important aspects of qualitative research: the differentiation of milieus and the relationship between performance and representation in interviews., Trotz der gesellschaftlichen Normalisierung und moralischen Entproblematisierung von Trennung und Scheidung stellen diese für die Betroffenen für gewöhnlich alles andere als normale, unproblematische Ereignisse dar. Das Geschehen muss geordnet, die Trennung legitimiert und die eigene Identität rehabilitiert werden, was zu auffälligen Erzähldynamiken in Interviews der qualitativen Trennungsforschung führen kann. Neben der Analyse des Zusammenhangs von Form, Inhalt und Funktion von Trennungserzählungen konnte die bisherige Forschung auch einige soziale Differenzierungen aufzeigen: nach Rolle, Geschlecht und sozialstrukturellen Faktoren. Auf Basis einer narrativen, funktionalen Analyse von 46 Interviews (23 Ex-Paare) aus unserer Studie "Paare nach der Trennung" fügen wir als weitere, übergeordnete Differenzierung die Milieuunterscheidung hinzu, die in der Forschung zu Trennungsnarrativen bislang unbeachtet war und die die bisherigen Differenzierungen integriert. Unser Argument, dass Trennungsnarrative milieuspezifisch gestaltet sind, exemplifizieren wir anhand zweier kontrastiver Milieus: dem individualisierten und dem traditionalen Milieu. Diese unterscheiden sich in fundamentaler Weise in ihren Beziehungsleitbildern und entsprechenden Trennungslegitimationen, in ihren Plausibilisierungsstrategien und in ihrer Vorstellung dessen, was als erstrebenswerte Identität gilt. Über den konkreten Gegenstand von Trennungen und Trennungserzählungen hinaus weisen wir in diesem Beitrag auf zwei für die qualitative Forschung wichtige Aspekte hin: die Milieudifferenzierung und das Verhältnis von Performanz und Repräsentanz in Interviews.

Periconceptional environment may influence embryo development, ultimately affecting adult health. Here, we review the rodent model of maternal low-protein diet specifically during the preimplantation period (Emb-LPD) with normal nutrition during subsequent gestation and postnatally. This model, studied mainly in the mouse, leads to cardiovascular, metabolic and behavioural disease in adult offspring, with females more susceptible. We evaluate the sequence of events from diet administration that may lead to adult disease. Emb-LPD changes maternal serum and/or uterine fluid metabolite composition, notably with reduced insulin and branched-chain amino acids. This is sensed by blastocysts through reduced mammalian target of rapamycin complex 1 signalling. Embryos respond by permanently changing the pattern of development of their extra-embryonic lineages, trophectoderm and primitive endoderm, to enhance maternal nutrient retrieval during subsequent gestation. These compensatory changes include stimulation in proliferation, endocytosis and cellular motility, and epigenetic mechanisms underlying them are being identified. Collectively, these responses act to protect fetal growth and likely contribute to offspring competitive fitness. However, the resulting growth adversely affects long-term health because perinatal weight positively correlates with adult disease risk. We argue that periconception environmental responses reflect developmental plasticity and 'decisions' made by embryos to optimise their own development, but with lasting consequences.

"Warum sollte ein Mann, der eine Diktatur erlebt und bekämpft hat, virtuell den alles bestimmenden Herrscher spielen, warum ein Mann, dessen Leben von schwierigen Neuanfängen geprägt ist, im Computerspiel stündlich wieder ganz 'bei null anfangen', warum eine junge Frau, die als Au-pair Überstunden macht, zusätzlich acht Stunden täglich virtuell Hausarbeit, Hauseinrichtung und Familiengründung 'spielen'? In medizinisch-psychologischen Erklärungsansätzen für Computerspielabhängigkeit wird das hohe Abhängigkeitsrisiko bei bestimmten Spielgenres durch überindividuelle neurophysiologische Vorgänge im Gehirn erklärt. Die Autorinnen haben dagegen aus insgesamt 22 biographischen, teilnarrativen Interviews mit ehemaligen Computerspielabhängigen bewusst drei individuelle, ungewöhnliche Passungen zwischen Computerspiel und Spielerbiographie ausgewählt (s.o.), für die sie zunächst ausführliche Analyseergebnisse darstellen. Dabei konnten sie eine Einordnung in drei Hauptmotive des Spielverhaltens rekonstruieren: erstens die versuchte Kompensation realweltlicher Mängel durch ein Computerspiel ('Vitamin'), zweitens die Ablenkung und Verdrängung von realweltlichen Problemen durch als aufregend und fesselnd erlebte Computerspiele ('Betäubungsmittel') und drittens, zunächst überraschender, eine hier im Fokus stehende, schwerer zu erklärende und erstmals beschriebene Funktion: das wiederholte Nachspielen biographischer Problemkonstellationen ('virtuelles Re-Enactment'), wie es sich in den eingangs angesprochenen Fällen zeigt. Gerade die biographische Perspektive ermöglicht dabei die Abgrenzung zwischen langfristigem sowie realweltlichem 'Gelingen', was auch von den Interviewten selbst rückblickend verneint wird, und dem in der Tradition der 'Game Studies' beschriebenen positiven In-Game-Erleben, also dem kurzfristigen und virtuellen 'Funktionieren'." Previously, we have shown that a maternal low protein diet, fed exclusively during the preimplantation period of mouse development (Emb-LPD), is sufficient to induce by the blastocyst stage a compensatory growth phenotype in late gestation and postnatally, correlating with increased risk of adult onset cardiovascular disease and behavioural dysfunction. Here, we examine mechanisms of induction of maternal Emb-LPD programming and early compensatory responses by the embryo. Emb-LPD induced changes in maternal serum metabolites at the time of blastocyst formation (E3.5), notably reduced insulin and increased glucose, together with reduced levels of free amino acids (AAs) including branched chain AAs leucine, isoleucine and valine. Emb-LPD also caused reduction in the branched chain AAs within uterine fluid at the blastocyst stage. These maternal changes coincided with an altered content of blastocyst AAs and reduced mTORC1 signalling within blastocysts evident in reduced phosphorylation of effector S6 ribosomal protein and its ratio to total S6 protein but no change in effector 4E-BP1 phosphorylated and total pools. These changes were accompanied by increased proliferation of blastocyst trophectoderm and total cells and subsequent increased spreading of trophoblast cells in blastocyst outgrowths. We propose that induction of metabolic programming following Emb-LPD is achieved through mTORC1signalling which acts as a sensor for preimplantation embryos to detect maternal nutrient levels via branched chain AAs and/or insulin availability. Moreover, this induction step associates with changes in extra-embryonic trophectoderm behaviour occurring as early compensatory responses leading to later nutrient recovery.

Early embryonic development is known to be susceptible to maternal undernutrition, leading to a disease-related postnatal phenotype. To determine whether this sensitivity extended into oocyte development, we examined the effect of maternal normal protein diet (18% casein; NPD) or isocaloric low protein diet (9% casein; LPD) restricted to one ovulatory cycle (3.5 days) prior to natural mating in female MF-1 mice. After mating, all females received NPD for the remainder of gestation and all offspring were litter size adjusted and fed standard chow. No difference in gestation length, litter size, sex ratio or postnatal growth was observed between treatments. Maternal LPD did, however, induce abnormal anxiety-related behaviour in open field activities in male and female offspring (P <0.05). Maternal LPD offspring also exhibited elevated systolic blood pressure (SBP) in males at 9 and 15 weeks and in both sexes at 21 weeks (P <0.05). Male LPD offspring hypertension was accompanied by attenuated arterial responsiveness in vitro to vasodilators acetylcholine and isoprenaline (P <0.05). LPD female offspring adult kidneys were also smaller, but had increased nephron numbers (P <0.05). Moreover, the relationship between SBP and kidney or heart size or nephron number was altered by diet treatment (P <0.05). These data demonstrate the sensitivity of mouse maturing oocytes in vivo to maternal protein undernutrition and identify both behavioural and cardiovascular postnatal outcomes, indicative of adult disease. These outcomes probably derive from a direct effect of protein restriction, although indirect stress mechanisms may also be contributory. Similar and distinct postnatal outcomes were observed here compared with maternal LPD treatment during post-fertilization preimplantation development which may reflect the relative contribution of the paternal genome.

Udgivelsesdato: 2008-May, NANOG, POU5F1, and SOX2 are required by the inner cell mass of the blastocyst and act cooperatively to maintain pluripotency in both mouse and human embryonic stem cells. Inadequacy of any one of them causes loss of the undifferentiated state. Mouse primordial germ cells (PGCs), from which pluripotent embryonic germ cells (EGCs) are derived, also express POU5F1, NANOG, and SOX2. Thus, a similar expression profile has been predicted for human PGCs. Here we show by RT-PCR, immunoblotting, and immunohistochemistry that human PGCs express POU5F1 and NANOG but not SOX2, with no evidence of redundancy within the group B family of human SOX genes. Although lacking SOX2, proliferative human germ cells can still be identified in situ during early development and are capable of culture in vitro. Surprisingly, with the exception of FGF4, many stem cell-restricted SOX2 target genes remained detected within the human SOX2-negative germ cell lineage. These studies demonstrate an unexpected difference in gene expression between human and mouse. The human PGC is the first primary cell type described to express POU5F1 and NANOG but not SOX2. The data also provide a new reference point for studies attempting to turn human stem cells into gametes by normal developmental pathways for the treatment of infertility.
