Abstract: As a concept, drainage of excess fluid volume in the cranium has been around for more than 1000 years. Starting with the original decompression-trepanation of Abulcasis to modern programmable shunt systems, to other nonshunt-based treatments such as endoscopic third ventriculostomy and choroid plexus cauterization, we have come far as a field. However, there are still fundamental limitations that shunts have yet to overcome: namely posture-induced over- and underdrainage, the continual need for valve opening pressure especially in pediatric cases, and the failure to reinstall physiologic intracranial pressure dynamics. However, there are groups worldwide, in the clinic, in industry, and in academia, that are trying to ameliorate the current state of the technology within hydrocephalus treatment. This chapter aims to provide a historical overview of hydrocephalus, current challenges in shunt design, what members of the community have done and continue to do to address these challenges, and finally, a definition of the “perfect” shunt is provided and how the authors are working toward it.

Abstract: In 1889 the German poet and novelist Theodor Fontane wrote the popular literary ballad “Herr von Ribbeck auf Ribbeck im Havelland.” The Squire von Ribbeck is described as a gentle and generous person, who often gives away pears from his pear trees to children passing by and continued donating pears after his death. Now, 135 years later the rock called Ribbeck is giving us insight into processes that happened 4.5 billion years ago. The meteorite Ribbeck (official find location: 52°37’15"N, 12°45’40"E) fell January 21, 2024, and has been classified as a brecciated aubrite. This meteoroid actually entered the Earth's atmosphere at 00:32:38 UTC over Brandenburg, west of Berlin, and the corresponding fireball was recorded by professional all sky and video cameras. More than 200 pieces (two proved by radionuclide analysis to belong to this fresh fall) were recovered totaling about 1.8 kg. Long-lived radionuclide and noble gas data are consistent with long cosmic ray exposure (55-62 Ma) and a preatmospheric radius of Ribbeck between 10 and 30 cm. The heavily brecciated aubrite consists of major (76±3 vol%) coarse-grained FeO-free enstatite (En99.1Fs<0.04Wo0.9), with a significant abundance (15.0±2.5 vol%) of albitic plagioclase (Ab95.3 An2.0Or2.7), minor forsterite (5.5±1.5 vol%; Fo99.9) and 3.5±1.0 vol% of opaque phases (mainly sulfides and metals) with traces of nearly FeO-free diopside (En53.2Wo46.8) and K-feldspar (Ab4.6Or95.4). The rock has a shock degree of S3 (U-S3), and terrestrial weathering has affected metals and sulfides, resulting in the brownish appearance of rock pieces and the partial destruction of certain sulfides already within days after the fall. The bulk chemical data confirm the feldspar-bearing aubritic composition. Ribbeck is closely related to the aubrite Bishopville. Abstract: Background: Young women with breast cancer (BC) have an increased chance of carrying germline BRCA pathogenic variants (PVs). Limited data exist on the prognostic impact of tumor histology (i.e. ductal versus lobular) in hereditary breast cancer. Methods: This multicenter retrospective cohort study included women aged ≤40 years with early-stage breast cancer diagnosed between January 2000 and December 2020 and known to carry germline PVs in BRCA1/2. Histology was locally assessed in each center. The Kaplan-Meier method and Cox regression analysis were used to assess disease-free survival and overall survival. Results: Of 4628 patients included from 78 centers worldwide, 3969 (86%) had pure ductal, 135 (3%) pure lobular, and 524 (11%) other histologies. Compared with ductal tumors, lobular tumors were more often grade 1/2 (57.7% versus 22.1%), stage III (29.6% versus 18.5%), and luminal A-like (42.2% versus 12.2%). Lobular tumors were more often associated with BRCA2 PVs (71.1% BRCA2), while ductal tumors were more often associated with BRCA1 PVs (65.7% BRCA1). Patients with lobular tumors more often had mastectomy (68.9% versus 58.3%), and less often received chemotherapy (83.7% versus 92.9%). With a median follow-up of 7.8 years, no significant differences were observed in disease-free survival (adjusted hazard ratio 1.01, 95% confidence interval 0.74-1.37) or overall survival (hazard ratio 0.96, 95% confidence interval 0.62-1.50) between patients with ductal versus lobular tumors. No significant survival differences were observed according to specific BRCA gene, breast cancer subtype, or body mass index. Conclusions: In this large global cohort of young BRCA carriers with breast cancer, the incidence of pure lobular histology was low and associated with higher disease stage at diagnosis, luminal-like disease and BRCA2 PVs. Histology did not appear to impact prognosis.

Abstract: Background: COVID-19 pandemic is thought to have changed the epidemiology of some pediatric neurosurgical disease: among them are the intracranial complications of sinusitis and otitis (ICSO). According to some studies on a limited number of cases, both streptococci-related sinusitis and ICSO would have increased immediately after the pandemic, although the reason is not clear yet (seasonal changes versus pandemic-related effects). The goal of the present survey of the European Society for Pediatric Neurosurgery (ESPN) was to collect a large number of cases from different European countries encompassing the pre-COVID (2017–2019), COVID (2020–2021), and post-COVID period (2022–June 2023) looking for possible epidemiological and/or clinical changes. Material and methods: An English language questionnaire was sent to ESPN members about year of the event, patient’s age and gender, presence of immune-deficit or other favoring risk factors, COVID infection, signs and symptoms at onset, site of primary infection, type of intracranial complication, identified germ, type and number of surgical operations, type and duration of medical treatment, clinical and radiological outcome, duration of the follow-up. Results: Two hundred fifty-four cases were collected by 30 centers coming from 14 different European countries. There was a statistically significant difference between the post-COVID period (129 children, 86 cases/year, 50.7% of the whole series) and the COVID (40 children, 20 cases/year, 15.7%) or the pre-COVID period (85 children, 28.3 cases/year, 33.5%). Other significant differences concerned the presence of predisposing factors/concurrent diseases (higher in the pre-COVID period) and previous COVID infection (higher in the post-COVID period). Abstract: Axicabtagene ciloleucel showed efficacy for relapsed/refractory large B-cell lymphomas (LBCL), including primary mediastinal B-cell lymphomas (PMBCL); however, only few PMBCLs were reported. Aim was to evaluate efficacy and safety of axicabtagene ciloleucel in patients with PMBCL compared to those with other LBCL, enrolled in the Italian prospective observational CART-SIE study. PMBCLs (n = 70) were younger, with higher percentage of bulky and refractory disease, compared to other LBCLs (n = 190). Median follow-up time for infused patients was 12.17 months (IQR 5.53,22.73). The overall (complete + partial) response rate (ORR,CR + PR) after bridging was 41% for PMBCL and 28% for other LBCL, p = 0.0102. Thirty days ORR was 78% (53/68) with 50% (34) CR in PMBCL, and 75% (141/187) with 53% (100) CR in other LBCL, p = 0.5457. Ninety days ORR was 69% (45/65) with 65% (42) CR in PMBCL, and 54% (87/162) with 47% (76) CR in other LBCL; progressive disease was 21% in PMBCL and 45% in other LBCL, p = 0.0336. Twelve months progression-free survival was 62% (95% CI: 51–75) in PMBCL versus 48% (95% CI: 41–57) in other LBCL, p = 0.0386. Twelve months overall survival was 86% (95% CI: 78–95) in PMBCL versus 71% (95% CI: 64–79) in other LBCL, p = 0.0034. All grade cytokine release syndrome was 88% (228/260); all grade neurotoxicity was 34% (88/260), with 6% of fatal events in PMBCL. Non-relapse mortality was 3%. In conclusion, PMBCLs achieved significantly better response and survival rates than other LBCLs.

7. Automated categorization of pre-trained models for software engineering: A case study with a Hugging Face dataset

Abstract: Software engineering (SE) activities have been revolutionized by the advent of pre-trained models (PTMs), defined as large machine learning (ML) models that can be fine-tuned to perform specific SE tasks. However, users with limited expertise may need help to select the appropriate model for their current task. To tackle the issue, the Hugging Face (HF) platform simplifies the use of PTMs by collecting, storing, and curating several models. Nevertheless, the platform currently lacks a comprehensive categorization of PTMs designed specifically for SE, i.e., the existing tags are more suited to generic ML categories. This paper introduces an approach to address this gap by enabling the automatic classification of PTMs for SE tasks. First, we utilize a public dump of HF to extract PTMs information, including model documentation and associated tags. Then, we employ a semi-automated method to identify SE tasks and their corresponding PTMs from existing literature. The approach involves creating an initial mapping between HF tags and specific SE tasks, using a similarity-based strategy to identify PTMs with relevant tags. The evaluation shows that model cards are informative enough to classify PTMs considering the pipeline tag. 8. A Distributed Approach for Persistent Homology Computation on a Large Scale

Abstract: Persistent homology (PH) is a powerful mathematical method to automatically extract relevant insights from images, such as those obtained by high-resolution imaging devices like electron microscopes or new-generation telescopes. However, the application of this method comes at a very high computational cost, that is bound to explode more because new imaging devices generate an ever-growing amount of data. In this paper we present PixHomology, a novel algorithm for efficiently computing $0$-dimensional PH on 2D images, optimizing memory and processing time. By leveraging the Apache Spark framework, we also present a distributed version of our algorithm with several optimized variants, able to concurrently process large batches of astronomical images. Finally, we present the results of an experimental analysis showing that our algorithm and its distributed version are efficient in terms of required memory, execution time, and scalability, consistently outperforming existing state-of-the-art PH computation tools when used to process large datasets.

Abstract: Acid sphingomyelinase deficiency (ASMD) is an ultra-rare disease, and several gaps of knowledge on various issues remain, particularly at a regional/national level. Expert opinions collected through well-defined consensus methodologies are increasingly used to make available reliable information in the context of rare/ultra-rare diseases. With the aim to provide indications on infantile neurovisceral ASMD (also formerly known as Niemann–Pick disease type A), chronic neurovisceral ASMD (formerly known as Niemann–Pick disease type A/B) and chronic visceral ASMD (formerly known as Niemann–Pick disease type B) in Italy, we conducted a Delphi consensus of experts focused on five main areas: (i) patients and disease characteristics; (ii) unmet needs and quality of life; (iii) diagnostic issues; (iv) treatment-related aspects; and (v) patient journey. Pre-specified, objective criteria were used to outline the multidisciplinary panel, based on 19 Italian experts in ASMD in paediatric and adult patients from different Italian Regions, including both clinicians (n = 16) and ASMD patients’ advocacy or payors with expertise in rare diseases (n = 3). During two Delphi rounds, a high ratio of agreement was found on several topics related to ASMD characteristics, diagnosis, management and disease burden. Abstract: Checkpoint inhibitors (CPIs) are routinely employed in relapsed/refractory classical Hodgkin lymphoma. Nonetheless, persistent long-term responses are uncommon, and one-third of patients are refractory. Several reports have suggested that treatment with CPIs may re-sensitize patients to chemotherapy, however there is no consensus on the optimal chemotherapy regimen and subsequent consolidation strategy. In this retrospective study we analysed the response to rechallenge with chemotherapy after CPI failure. Furthermore, we exploratively characterized the clonal evolution profile of a small sample of patients (n = 5) by employing the CALDER approach. Among the 28 patients included in the study, 17 (71%) were primary refractory and 26 (92%) were refractory to the last chemotherapy prior to CPIs. Following rechallenge with chemotherapy, response was recorded in 23 (82%) patients experiencing complete remission and 3 (11%) patients experiencing partial remission. The tumour evolution of the patients inferred by CALDER seemingly occurred prior to the first cycle of therapy and was characterized either by linear or branching evolution patterns. Twenty-five patients proceeded to allogeneic stem cell transplantation. At a median follow-up of 21 months, median PFS and OS were not reached. In conclusion, patients who fail CPIs can be effectively rescued by salvage chemotherapy and bridged to allo-SCT/auto-SCT.

Abstract: We introduce numerical algebraic geometry methods for computing lower bounds on the reach, local feature size, and weak feature size of the real part of an equidimensional and smooth algebraic variety using the variety's defining polynomials as input. For the weak feature size, we also show that nonquadratic complete intersections generically have finitely many geometric bottlenecks, and we describe how to compute the weak feature size directly rather than a lower bound in this case. 12. 'Neighborhood fear of crime and disadvantaged areas : a comparative longitudinal study'

Abstract: The current study explores how neighborhood fear of crime ("worry about specific crimes") changes over time in neighborhoods with different levels of disadvantage. With a comparative design, 81 and 123- neighborhoods in two cities in Sweden are followed over a six-year period. Fear of crime trajectories are assessed through growth curve modeling. We consider how differences in neighborhood processes, such as social and physical disorder, might influence fear of crime levels. The results show that fear of crime increased over time in both cities and the increase was related to higher levels of physical disorder in several areas. Furthermore, the change differed depending on the neighborhood type - in highly disadvantaged neighborhoods, the fear was stable at a high level. In contrast, the largest increases in fear could be seen in the areas with the lowest disadvantage in both cities. Theoretical and practical implications are discussed.

Abstract: The short-lived radionuclide aluminium-26 (26Al) isotope is a major heat source for early planetary melting. The aluminium-26 - magnesium-26 (26Al-26Mg) decay system also serves as a high-resolution relative chronometer. In both cases, however, it is critical to establish whether 26Al was homogeneously or heterogeneously distributed throughout the solar nebula. Here we report a precise lead-207 - lead-206 (207Pb-206Pb) isotopic age of 4565.56 ± 0.12 million years (Ma) for the andesitic achondrite Erg Chech 002. Our analysis, in conjunction with published 26Al-26Mg data, reveals that the initial 26Al/27Al in the source material of this achondrite was notably higher than in various other well-preserved and precisely dated achondrites. Here we demonstrate that the current data clearly indicate spatial heterogeneity of 26Al by a factor of 3-4 in the precursor molecular cloud or the protoplanetary disk of the Solar System, likely associated with the late infall of stellar materials with freshly synthesized radionuclides.

Abstract: Preliminary analyses of asteroid Ryugu samples show kinship to aqueously altered CI (Ivuna-type) chondrites, suggesting similar origins. We report identification of C-rich, particularly primitive clasts in Ryugu samples that contain preserved presolar silicate grains and exceptional abundances of presolar SiC and isotopically anomalous organic matter. The high presolar silicate abundance (104 ppm) indicates that the clast escaped extensive alteration. The 5 to 10 times higher abundances of presolar SiC (~235 ppm), N-rich organic matter, organics with N isotopic anomalies (1.2%), and organics with C isotopic anomalies (0.2%) in the primitive clasts compared to bulk Ryugu suggest that the clasts formed in a unique part of the protoplanetary disk enriched in presolar materials. These clasts likely represent previously unsampled outer solar system material that accreted onto Ryugu after aqueous alteration ceased, consistent with Ryugus rubble pile origin.

Abstract: Classical work by Salmon and Bromwich classified singular intersections of two quadric surfaces. The basic idea of these results was already pursued by Cayley in connection with tangent intersections of conics in the plane and used by Schafli for the study of hyperde-terminants. More recently, the problem has been revisited with similar tools in the context of geometric modeling and a generalization to the case of two higher dimensional quadric hyper -surfaces was given by Ottaviani. We propose and study a generalization of this question for systems of Laurent polynomials with support on a fixed point configuration. In the non-defective case, the closure of the locus of coefficients giving a non-degenerate multiple root of the system is defined by a polynomial called the mixed discriminant. We define a related polynomial called the multivariate iterated discriminant, generalizing the classical Schafli method for hyperdeterminants. This iterated discriminant is easier to compute and we prove that it is always divisible by the mixed discriminant. We show that tangent intersections can be computed via iteration if and only if the singular locus of a corresponding dual variety has sufficiently high codimension. We also study when point configurations corresponding to Segre-Veronese varieties and to the lattice points of planar smooth polygons, have their iterated discriminant equal to their mixed discriminant., QC 20230722

Abstract: Studies of material returned from Cb asteroid Ryugu have revealed considerable mineralogical and chemical heterogeneity, stemming primarily from brecciation and aqueous alteration. Isotopic anomalies could have also been affected by delivery of exogenous clasts and aqueous mobilization of soluble elements. Here, we show that isotopic anomalies for mildly soluble Cr are highly variable in Ryugu and CI chondrites, whereas those of Ti are relatively uniform. This variation in Cr isotope ratios is most likely due to physicochemical fractionation between 54Cr-rich presolar nanoparticles and Cr-bearing secondary minerals at the millimeter-scale in the bulk samples, likely due to extensive aqueous alteration in their parent bodies that occurred [Formula: see text] after Solar System birth. In contrast, Ti isotopes were marginally affected by this process. Our results show that isotopic heterogeneities in asteroids are not all nebular or accretionary in nature but can also reflect element redistribution by water.

Abstract: The Hayabusa2 spacecraft explored asteroid Ryugu and brought its surface materials to Earth. Ryugu samples resemble Ivuna-type (CI) chondrites-the most chemically primitive meteorites-and contain secondary phyllosilicates and carbonates, which are indicative of aqueous alteration. Understanding the conditions (such as temperature, redox state and fluid composition) during aqueous alteration is crucial to elucidating how Ryugu evolved to its present state, but little is known about the temporal changes in these conditions. Here we show that calcium carbonate (calcite) grains in Ryugu and Ivuna samples have variable O-18/O-16 and C-13/C-12 ratios that are, respectively, 24-46 & PTSTHOUSND; and 65-108 & PTSTHOUSND; greater than terrestrial standard values, whereas those of calcium-magnesium carbonate (dolomite) grains are much more homogeneous, ranging within 31-36 & PTSTHOUSND; for oxygen and 67-75 & PTSTHOUSND; for carbon. We infer that the calcite precipitated first over a wide range of temperatures and oxygen partial pressures, and that the proportion of gaseous CO2/CO/CH4 molecules changed temporally. By contrast, the dolomite formed later in a more oxygen-rich and thus CO2-dominated environment when the system was approaching equilibrium. The characteristic isotopic compositions of secondary carbonates in Ryugu and Ivuna are not observed for other hydrous meteorites, suggesting a unique evolutionary pathway for their parent asteroid(s). The asteroid Ryugu experienced aqueous alteration under changing temperature and redox conditions, according to an isotopic analysis of secondary calcite and dolomite grains in samples from Ryugu obtained by the Hayabusa2 spacecraft.

Abstract: European Reference Network (ERN) CRANIO is focused on optimizing care for patients with rare or complex craniofacial anomalies, including craniosynostosis and/or rare ear, nose, and throat disorders. The main goal of ERN CRANIO is to collect uniform data on treatment outcomes for multicenter comparison. We aimed to develop a reproducible and reliable suture-specific photo score that can be used for cross-center comparison of phenotypical severity of sagittal synostosis and aesthetic outcome of treatment. We conducted a retrospective study among nonsyndromic sagittal synostosis patients aged <19 years. We included preoperative and postoperative photo sets from 6 ERN CRANIO centers. Photo sets included bird's eye, lateral, and anterior-posterior views. The sagittal synostosis photo score was discussed in the working group, and consensus was obtained on its contents. Interrater agreement was assessed with weighted Fleiss' Kappa and intraclass correlation coefficients.The photo score consisted of frontal bossing, elongated skull, biparietal narrowness, temporal hollowing, vertex line depression, occipital bullet, and overall phenotype. Each item was scored as normal, mild, moderate, or severe. Results from 36 scaphocephaly patients scored by 20 raters showed kappa values ranging from 0.38 [95% bootstrap CI: 0.31, 0.45] for biparietal narrowness to 0.56 [95% bootstrap CI: 0.47, 0.64] for frontal bossing. Agreement was highest for the sum score of individual items [intraclass correlation coefficients agreement 0.69 [95% CI: 0.57, 0.82]. This is the first large-scale multicenter study in which experts investigated a photo score to assess the severity of sagittal synostosis phenotypical characteristics. Abstract: We present oxygen isotopic analyses of fragments of the near-Earth Cb-type asteroid Ryugu returned by the Hayabusa2 spacecraft that reinforce the close correspondence between Ryugu and CI chondrites. Small differences between Ryugu samples and CI chondrites in can be explained at least in part by contamination of the latter by terrestrial water. The discovery that a randomly sampled C-complex asteroid is composed of CI-chondrite-like rock, combined with thermal models for formation prior to significant decay of the short-lived radioisotope 26Al, suggests that if lithified at the time of alteration, the parent body was small (≪50 km radius). Abstract: Preliminary analyses of asteroid Ryugu samples show kinship to aqueously altered CI (Ivuna-type) chondrites, suggesting similar origins. We report identification of C-rich, particularly primitive clasts in Ryugu samples that contain preserved presolar silicate grains and exceptional abundances of presolar SiC and isotopically anomalous organic matter. The high presolar silicate abundance (104 ppm) indicates that the clast escaped extensive alteration. The 5 to 10 times higher abundances of presolar SiC (~235 ppm), N-rich organic matter, organics with N isotopic anomalies (1.2%), and organics with C isotopic anomalies (0.2%) in the primitive clasts compared to bulk Ryugu suggest that the clasts formed in a unique part of the protoplanetary disk enriched in presolar materials. These clasts likely represent previously unsampled outer solar system material that accreted onto Ryugu after aqueous alteration ceased, consistent with Ryugu's rubble pile origin.

Abstract: Carbonaceous meteorites are thought to be fragments of C-type (carbonaceous) asteroids. Samples of the C-type asteroid (162173) Ryugu were retrieved by the Hayabusa2 spacecraft. We measured the mineralogy and bulk chemical and isotopic compositions of Ryugu samples. The samples are mainly composed of materials similar to those of carbonaceous chondrite meteorites, particularly the CI (Ivuna-type) group. The samples consist predominantly of minerals formed in aqueous fluid on a parent planetesimal. The primary minerals were altered by fluids at a temperature of 37 degrees +/- 10 degrees C, about 5.2(-0.7)(+0.8) million (statistical) or 5.2(-2.1)(+1.6) million (systematic) years after the formation of the first solids in the Solar System. After aqueous alteration, the Ryugu samples were likely never heated above similar to 100 degrees C. The samples have a chemical composition that more closely resembles that of the Sun's photosphere than other natural samples do.

Abstract: Over the years, Software Quality Engineering has increased interest, demonstrated by significant research papers published in this area. Determining when a software artifact is qualitatively valid is tricky, given the impossibility of providing an objective definition valid for any perspective, context, or stakeholder. Many quality model solutions have been proposed that reference specific quality attributes in this context. However, these approaches do not consider the context in which the artifacts will operate and the stakeholder's perspective who evaluate its validity. Furthermore, these solutions suffer from the limitations of being artifact-specific and not extensible. In this paper, we provide a generic and extensible mechanism that makes it possible to aggregate and prioritize quality attributes. The user, taking into account his perspective and the context in which the software artifact will operate, is guided in defining all the criteria for his quality model. The management of these criteria is then facilitated through Multi-Criteria Decision Making (MCDM). Abstract: Initial analyses showed that asteroid Ryugu’s composition is close to CI (Ivuna-like) carbonaceous chondrites (CCs) – the chemically most primitive meteorites, characterized by near-solar abundances for most elements. However, some isotopic signatures (for example, Ti, Cr) overlap with other CC groups, so the details of the link between Ryugu and the CI chondrites are not yet fully clear. Here we show that Ryugu and CI chondrites have the same zinc and copper isotopic composition. As the various chondrite groups have very distinct Zn and Cu isotopic signatures, our results point at a common genetic heritage between Ryugu and CI chondrites, ruling out any affinity with other CC groups. Since Ryugu’s pristine samples match the solar elemental composition for many elements, their Zn and Cu isotopic compositions likely represent the best estimates of the solar composition. Earth’s mass-independent Zn isotopic composition is intermediate between Ryugu/CC and non-carbonaceous chondrites (NCs), suggesting a contribution of Ryugu-like material to Earth’s budgets of Zn and other moderately volatile elements.

Abstract: Recommender systems for software engineering (RSSEs) assist software engineers in dealing with a growing information overload when discerning alternative development solutions. While RSSEs are becoming more and more effective in suggesting handy recommendations, they tend to suffer from popularity bias, i.e., favoring items that are relevant mainly because several developers are using them. While this rewards artifacts that are likely more reliable and well-documented, it would also mean that missing artifacts are rarely used because they are very specific or more recent. This paper studies popularity bias in Third-Party Library (TPL) RSSEs. First, we investigate whether state-of-the-art research in RSSEs has already tackled the issue of popularity bias. Then, we quantitatively assess four existing TPL RSSEs, exploring their capability to deal with the recommendation of popular items. Finally, we propose a mechanism to defuse popularity bias in the recommendation list. The empirical study reveals that the issue of dealing with popularity in TPL RSSEs has not received adequate attention from the software engineering community. Abstract: BackgroundGaucher disease (GD) diagnosis can be delayed due to non-specific symptoms and lack of awareness, leading to unnecessary procedures and irreversible complications. GAU-PED study aims to assess GD prevalence in a high-risk pediatric population and the presence, if any, of novel clinical or biochemical markers associated with GD.Materials and methodsDBS samples were collected and tested for beta-glucocerebrosidase enzyme activity for 154 patients selected through the algorithm proposed by Di Rocco et al. Patients showing beta-glucocerebrosidase activity below normal values were recalled to confirm the enzyme deficiency with the gold standard essay on cellular homogenate. Patients tested positive at the gold standard analysis were evaluated through GBA1 gene sequencing.Results14 out of 154 patients were diagnosed with GD, with a prevalence of 9.09% (5.06-14.78%, CI 95%). Hepatomegaly, thrombocytopenia, anemia, growth delay/deceleration, elevated serum ferritin, elevated Lyso-Gb1 and chitotriosidase were significantly associated with GD.ConclusionsGD prevalence in a pediatric population at high-risk appeared to be higher compared to high-risk adults. Lyso-Gb1 was associated with GD diagnosis. The algorithm proposed by Di Rocco et al. can potentially improve the diagnostic accuracy of pediatric GD, allowing the prompt start of therapy, aiming to reduce irreversible complications.

29. ResyDuo: Combining data models and CF-based recommender systems to develop Arduino projects

Abstract: While specifying an IoT-based system, software developers have to face a set of challenges, spanning from selecting the hardware components to writing the actual source code. Even though dedicated development environments are in place, a nonexpert user might struggle with the over-choice problem in selecting the proper component. By combining MDE and recommender systems, this paper proposes an initial prototype, called ResyDuo, to assist Arduino developers by providing two different artifacts, i. e. , hardware components and software libraries. In particular, we make use of a widely adopted collaborative filtering algorithm by collecting relevant information by means of a dedicated data model. ResyDuo can retrieve hardware components by using tags or existing Arduino projects stored on the ProjectHub repository. Then, the system can eventually retrieve corresponding software libraries based on the identified hardware devices. ResyDuo is equipped with a web-based interface that allows users to easily select and configure the under-developing Arduino project. To assess ResyDuos performances, we run the ten-fold crossvalidation by adopting the grid search strategy to optimize the hyperparameters of the CF-based algorithm. The conducted evaluation shows encouraging results even though there is still room for improvement in terms of the examined metrics.

Abstract: Since its launch in November 2022, ChatGPT has gained popularity among users, especially programmers who use it as a tool to solve development problems. However, while offering a practical solution to programming problems, ChatGPT should be mainly used as a supporting tool (e.g., in software education) rather than as a replacement for the human being. Thus, detecting automatically generated source code by ChatGPT is necessary, and tools for identifying AI-generated content may need to be adapted to work effectively with source code. This paper presents an empirical study to investigate the feasibility of automated identification of AI-generated code snippets, and the factors that influence this ability. To this end, we propose a novel approach called GPTSniffer, which builds on top of CodeBERT to detect source code written by AI. The results show that GPTSniffer can accurately classify whether code is human-written or AI-generated, and outperforms two baselines, GPTZero and OpenAI Text Classifier. Also, the study shows how similar training data or a classification context with paired snippets helps to boost classification performances.

31. Functional characterisation of a HOX/PBX transcriptional complex

Abstract: Hox genes control cell fates and specify regional identities during vertebrate development. Hox proteins relaxed DNA-binding selectivity in vitro, suggests that Hox specificity of action is achieved in vivo through the action of transcriptional cofactors. Members of the EXD/PBX family of homeoproteins have been proposed for such a role on the basis of genetic and biochemical evidence. In this work we show that the human Pbxl and HOXBl proteins can cooperatively activate transcription through a genetically charaterized Hox responsive element, the autoregulatory enhancer of the mouse Hoxb-T gene (b-lARE), which directs the spatially restricted expression of Hoxb-L in the fourth rhombomere during hindbrain development. On the b-lARE, only a restricted subset of HOX proteins is able to bind cooperatively and activate transcription. Selective recognition of the b-lARE is mediated by the N-terminal region of the HOX homeodomain. The DNA binding and protein-protein interaction functions of HOXBl and Pbxl are all necessary for the assembly of a transcriptionally active complex on the b-lARE. Functional dissection of the complex allowed the localization of the main activation domain in the HOXBl N-terminal region, and of an additional one in the C-terminal region of Pbxl, which is absent in one of its two alternative splicing isoforms. The transcriptional activity of HOX/Pbxl complexes on the b-lARE element is maximal and further restricted in embryonal carcinoma (EC) cells compared to other cell lines. Mutational analysis shows that an octamer-binding protein consensus site on the b-lARE contributes to the activity of the enhancer in transfected cells and it is bound in vitro by octamer-like proteins from both EC cells and embryonic extracts.

Abstract: Niemann-Pick type C (NPC) disease is an autosomal recessive lysosomal storage disorder caused by mutations in NPC1 or NPC2 genes. In 2009, the molecular characterization of 44 NPC Italian patients has been published. Here, we present an update of the genetic findings in 105 Italian NPC patients belonging to 83 unrelated families (77 NPC1 and 6 NPC2). NPC1 and NPC2 genes were studied following an algorithm recently published. Eighty-four different NPC1 and five NPC2 alleles were identified. Only two NPC1 alleles remained non detected. Sixty-two percent of NPC1 alleles were due to missense variants. The most frequent NPC1 mutation was the p.F284Lfs*26 (5.8% of the alleles). All NPC2 mutations were found in the homozygous state, and all but one was severe. Among newly diagnosed patients, 18 novel NPC1 mutations were identified. The pathogenic nature of 7/9 missense alleles and 3/4 intronic variants was confirmed by filipin staining and NPC1 protein analysis or mRNA expression in patient’s fibroblasts. Taken together, our previous published data and new results provide an overall picture of the molecular characteristics of NPC patients diagnosed so far in Italy.

Abstract: Introduction: Chimeric antigen receptor (CAR) T-cell therapies are novel immunotherapies for the treatment of hematologic malignancies. They are administered in specialized centers by a multidisciplinary team and require the careful coordination of all steps involved in manufacturing and using cellular therapies. The Maturity Model (MM) is a tool developed and used for assessing the effectiveness of a variety of activities. In healthcare, it may assist clinicians in the gradual improvement of patient management with CAR T-cell therapy and other complex treatments. Methods: The START CAR-T project was initiated to investigate the potential of a MM in the setting of CAR T-cell therapy. Four Italian clinics participated in the creation of a dedicated MM. Following the development and test of this MM, its validity and generalizability were further tested with a questionnaire submitted to 18 Italian centers. Results: The START CAR-T MM assessed the maturity level of clinical sites, with a focus on organization, process, and digital support. For each area, the model defined four maturity steps, and indicated the actions required to evolve from a basic to an advanced status. The application of the MM to 18 clinical sites provided a description of the maturity level of Italian centers with regard to the introduction of CAR T-cell therapy. Conclusion: The START CAR-T MM appears to be a useful and widely applicable tool. It may help centers optimize many aspects of CAR T-cell therapy and improve patient access to this novel treatment option.

Abstract: On July 15, 2021, a huge fireball was visible over Poland. After the possible strewn field was calculated, the first and so far only sample, with a mass of 350 g, was discovered 18 days after the fireball event. The Antonin meteorite was found August 3, 2021, on the edge of a forest close to a dirt road near Helenow, a small suburb of the city of Mikstat. The rock is an ordinary chondrite breccia and consists of equilibrated and recrystallized lithologies. The boundaries between different fragments are difficult to detect, and the lithologies are of petrologic type 5 and type 4. The rock is moderately shocked (S4) and contains local impact melt areas and thin shock veins. The low-Ca pyroxene and olivine are equilibrated (Fs20.6 and Fa24.0, respectively), typical of L chondrites. The L chondrite classification is also supported by O isotope data and the results of bulk chemical analysis. The Ti isotope characteristics confirm that Antonin is related to the noncarbonaceous (NC) meteorites. One of the studied thin sections shows an unusual metal–chondrule assemblage, perhaps indicating that the metal in the chondrite is heterogeneously distributed, which is, however, not clearly visible in the element abundances.

Abstract: The extraterrestrial materials returned from asteroid (162173) Ryugu consist predominantly of low-temperature aqueously formed secondary minerals and are chemically and mineralogically similar to CI (Ivuna-type) carbonaceous chondrites. Here, we show that high-temperature anhydrous primary minerals in Ryugu and CI chondrites exhibit a bimodal distribution of oxygen isotopic compositions: 16O-rich (associated with refractory inclusions) and 16O-poor (associated with chondrules). Both the 16O-rich and 16O-poor minerals probably formed in the inner solar protoplanetary disk and were subsequently transported outward. The abundance ratios of the 16O-rich to 16O-poor minerals in Ryugu and CI chondrites are higher than in other carbonaceous chondrite groups but are similar to that of comet 81P/Wild2, suggesting that Ryugu and CI chondrites accreted in the outer Solar System closer to the accretion region of comets.

Abstract: Little is known about the origin of the spectral diversity of asteroids and what it says about conditions in the protoplanetary disk. Here, we show that samples returned from Cb-type asteroid Ryugu have Fe isotopic anomalies indistinguishable from Ivuna-type (CI) chondrites, which are distinct from all other carbonaceous chondrites. Iron isotopes, therefore, demonstrate that Ryugu and CI chondrites formed in a reservoir that was different from the source regions of other carbonaceous asteroids. Growth and migration of the giant planets destabilized nearby planetesimals and ejected some inward to be implanted into the Main Belt. In this framework, most carbonaceous chondrites may have originated from regions around the birthplaces of Jupiter and Saturn, while the distinct isotopic composition of CI chondrites and Ryugu may reflect their formation further away in the disk, owing their presence in the inner Solar System to excitation by Uranus and Neptune.

Abstract: The Losev–Manin moduli space parametrizes pointed chains of projective lines. In this paper we study a possible generalization to families of pointed degenerate toric varieties. Geometric properties of these families, such as flatness and reducedness of the fibers, are explored via a combinatorial characterization. We show that such families are described by a specific type of polytope fibration which generalizes the twisted Cayley sums, originally introduced to characterize elementary extremal contractions of fiber type associated to projective Q-factorial toric varieties with positive dual defect. Abstract: CO CV CM CI Ryugu A Ryugu C 0.0 0.5 1.0 1.5 44/40CaSRM915a (age corrected) The Hayabusa2 spacecraft has returned samples from the Cb-type asteroid (162173) Ryugu to Earth. Previous petrological and chemical analyses support a close link between Ryugu and CI chondrites that are presumed to be chemically the most primitive meteorites with a solar-like composition. However, Ryugu samples are highly enriched in Ca compared to typical CI chondrites. To identify the cause of this discrepancy, here we report stable Ca isotopic data (expressed as delta 44/40CaSRM915a) for returned Ryugu samples collected from two sites. We found that samples from both sites have similar delta 44/40CaSRM915a (0.58 +/- 0.03 parts per thousand and 0.55 +/- 0.08 parts per thousand, 2 s.d.) that fall within the range defined by CIs. This isotopic similarity suggests that the Ca budget of CIs and Ryugu samples is dominated by carbonates, and the variably higher Ca contents in Ryugu samples are due to the abundant carbonates. Precipitation of carbonates on Ryugu likely coincided with a major episode of aqueous activity dated to have occurred similar to 5 Myr after Solar System formation. Based on the pristine Ryugu samples, the average delta 44/40CaSRM915a of the Solar System is defined to be 0.57 +/- 0.04 parts per thousand (2 s.d.).

Abstract: The extraterrestrial materials returned from asteroid (162173) Ryugu consist predominantly of low-temperature aqueously formed secondary minerals and are chemically and mineralogically similar to CI (Ivuna-type) carbonaceous chondrites. Here, we show that high-temperature anhydrous primary minerals in Ryugu and CI chondrites exhibit a bimodal distribution of oxygen isotopic compositions: 16O-rich (associated with refractory inclusions) and 16O-poor (associated with chondrules). Both the 16O-rich and 16O-poor minerals probably formed in the inner solar protoplanetary disk and were subsequently transported outward. The abundance ratios of the 16O-rich to 16O-poor minerals in Ryugu and CI chondrites are higher than in other carbonaceous chondrite groups but are similar to that of comet 81P/Wild2, suggesting that Ryugu and CI chondrites accreted in the outer Solar System closer to the accretion region of comets.

Abstract: In this paper we present an efficient algorithm to produce a provably dense sample of a smooth compact affine variety. The procedure is partly based on computing bottlenecks of the variety. Using geometric information such as the bottlenecks and the local reach we also provide bounds on the density of the sample needed in order to guarantee that the homology of the variety can be recovered from the sample. An implementation of the algorithm is provided together with numerical experiments and a computational comparison to the algorithm by Dufresne et al. 43. Computing Seshadri Constants on Smooth Toric Surfaces

Abstract: In this paper we consider the problem of computing Seshadri constants at a general point on a smooth polarized toric surface. We consider the case when the degree of jet separation is small or the core of the associated polygon is a line segment. Our main result is that under these hypothesis the Seshadri constant at the general point can often be determined in terms of easily computable invariants of the surfaces at hand. Abstract: Initial analyses showed that asteroid Ryugu's composition is close to CI (Ivuna-like) carbonaceous chondrites (CCs) - the chemically most primitive meteorites, characterized by near-solar abundances for most elements. However, some isotopic signatures (for example, Ti, Cr) overlap with other CC groups, so the details of the link between Ryugu and the CI chondrites are not yet fully clear. Here we show that Ryugu and CI chondrites have the same zinc and copper isotopic composition. As the various chondrite groups have very distinct Zn and Cu isotopic signatures, our results point at a common genetic heritage between Ryugu and CI chondrites, ruling out any affinity with other CC groups. Since Ryugu's pristine samples match the solar elemental composition for many elements, their Zn and Cu isotopic compositions likely represent the best estimates of the solar composition. Earth's mass-independent Zn isotopic composition is intermediate between Ryugu/CC and non-carbonaceous chondrites (NCs), suggesting a contribution of Ryugu-like material to Earth's budgets of Zn and other moderately volatile elements.

Abstract: We have conducted a NanoSIMS-based search for presolar material in samples recently returned from C-type asteroid Ryugu as part of JAXA's Hayabusa2 mission. We report the detection of all major presolar grain types with O- and C-anomalous isotopic compositions typically identified in carbonaceous chondrite meteorites: 1 silicate, 1 oxide, 1 O-anomalous supernova grain of ambiguous phase, 38 SiC, and 16 carbonaceous grains. At least two of the carbonaceous grains are presolar graphites, whereas several grains with moderate C isotopic anomalies are probably organics. The presolar silicate was located in a clast with a less altered lithology than the typical extensively aqueously altered Ryugu matrix. The matrix- normalized presolar grain abundances in Ryugu are 4.8(-2.6)(+4.7) ppm for O-anomalous grains, 25(-)(5)(+)(6) ppm for SiC grains, and 11(-3)(+5) ppm for carbonaceous grains. Ryugu is isotopically and petrologically similar to carbonaceous Ivuna-type (CI) chondrites. To compare the in situ presolar grain abundances of Ryugu with CI chondrites, we also mapped Ivuna and Orgueil samples and found a total of 15 SiC grains and 6 carbonaceous grains. No O-anomalous grains were detected. The matrix-normalized presolar grain abundances in the CI chondrites are similar to those in Ryugu: 23(-6)(+7) ppm SiC and 9.0 (+5.4)(-3)(.6) ppm carbonaceous grains. Thus, our results provide further evidence in support of the Ryugu-CI connection. They also reveal intriguing hints of small-scale heterogeneities in the Ryugu samples, such as locally distinct degrees of alteration that allowed the preservation of delicate presolar material.

Abstract: We introduce numerical algebraic geometry methods for computing lower bounds on the reach, local feature size, and the weak feature size of the real part of an equidimensional and smooth algebraic variety using the variety's defining polynomials as input. For the weak feature size, we also show that non-quadratic complete intersections generically have finitely many geometric bottlenecks, and describe how to compute the weak feature size directly rather than a lower bound in this case. In all other cases, we describe additional computations that can be used to determine feature size values rather than lower bounds. We also present homology inference experiments that combine persistent homology computations with implemented versions of our feature size algorithms, both with globally dense samples and samples that are adaptively dense with respect to the local feature size.

Abstract: Fibrodysplasia ossificans progressiva (FOP) is a rare and devastating genetic disease, in which soft connective tissue is converted into heterotopic bone through an endochondral ossification process. Patients succumb early as they gradually become trapped in a second skeleton of heterotopic bone. Although the underlying genetic defect is long known, the inherent complexity of the disease has hindered the discovery of effective preventions and treatments. New developments in the gene therapy field have motivated its consideration as an attractive therapeutic option for FOP. However, the immune system's role in FOP activation and the as-yet unknown primary causative cell, are crucial issues which must be taken into account in the therapy design. While gene therapy offers a potential therapeutic solution, more knowledge about FOP is needed to enable its optimal and safe application.

48. GitRanking: A Ranking of GitHub Topics for Software Classification using Active Sampling

Abstract: GitHub is the world's largest host of source code, with more than 150M repositories. However, most of these repositories are not labeled or inadequately so, making it harder for users to find relevant projects. There have been various proposals for software application domain classification over the past years. However, these approaches lack a well-defined taxonomy that is hierarchical, grounded in a knowledge base, and free of irrelevant terms. This work proposes GitRanking, a framework for creating a classification ranked into discrete levels based on how general or specific their meaning is. We collected 121K topics from GitHub and considered $60\%$ of the most frequent ones for the ranking. GitRanking 1) uses active sampling to ensure a minimal number of required annotations; and 2) links each topic to Wikidata, reducing ambiguities and improving the reusability of the taxonomy. Our results show that developers, when annotating their projects, avoid using terms with a high degree of specificity. This makes the finding and discovery of their projects more challenging for other users. Furthermore, we show that GitRanking can effectively rank terms according to their general or specific meaning. This ranking would be an essential asset for developers to build upon, allowing them to complement their annotations with more precise topics. Finally, we show that GitRanking is a dynamically extensible method: it can currently accept further terms to be ranked with a minimum number of annotations ($\sim$ 15). 49. MemoRec: A Recommender System for Assisting Modelers in Specifying Metamodels

Abstract: Model Driven Engineering (MDE) has been widely applied in software development, aiming to facilitate the coordination among various stakeholders. Such a methodology allows for a more efficient and effective development process. Nevertheless, modeling is a strenuous activity that requires proper knowledge of components, attributes, and logic to reach the level of abstraction required by the application domain. In particular, metamodels play an important role in several paradigms, and specifying wrong entities or attributes in metamodels can negatively impact on the quality of the produced artifacts as well as other elements of the whole process. During the metamodeling phase, modelers can benefit from assistance to avoid mistakes, e.g., getting recommendations like meta-classes and structural features relevant to the metamodel being defined. However, suitable machinery is needed to mine data from repositories of existing modeling artifacts and compute recommendations. In this work, we propose MemoRec, a novel approach that makes use of a collaborative filtering strategy to recommend valuable entities related to the metamodel under construction. Our approach can provide suggestions related to both metaclasses and structured features that should be added in the metamodel under definition. We assess the quality of the work with respect to different metrics, i.e., success rate, precision, and recall. 50. Providing Upgrade Plans for Third-party Libraries: A Recommender System using Migration Graphs

Abstract: During the development of a software project, developers often need to upgrade third-party libraries (TPLs), aiming to keep their code up-to-date with the newest functionalities offered by the used libraries. In most cases, upgrading used TPLs is a complex and error-prone activity that must be carefully carried out to limit the ripple effects on the software project that depends on the libraries being upgraded. In this paper, we propose EvoPlan as a novel approach to the recommendation of different upgrade plans given a pair of library-version as input. In particular, among the different paths that can be possibly followed to upgrade the current library version to the desired updated one, EvoPlan is able to suggest the plan that can potentially minimize the efforts being needed to migrate the code of the clients from the library's current release to the target one. The approach has been evaluated on a curated dataset using conventional metrics used in Information Retrieval, i.e., precision, recall, and F-measure. The experimental results show that EvoPlan obtains an encouraging prediction performance considering two different criteria in the plan specification, i.e., the popularity of migration paths and the number of open and closed issues in GitHub for those projects that have already followed the recommended migration paths.
