The prevalence of neuroendocrine prostate cancer (NEPC) arising from adenocarcinoma (AC) upon potent androgen receptor (AR) pathway inhibition is increasing. Deeper understanding of NEPC biology and development of novel therapeutic agents are needed. However, research is hindered by the paucity of research models, especially cell lines developed from NEPC patients. We established a novel NEPC cell line, KUCaP13, from tissue of a patient initially diagnosed with AC which later recurred as NEPC. The cell line has been maintained permanently in vitro under regular cell culture conditions and is amenable to gene engineering with lentivirus. KUCaP13 cells lack the expression of AR and overexpress NEPC-associated genes, including SOX2, EZH2, AURKA, PEG10, POU3F2, ENO2, and FOXA2. Importantly, the cell line maintains the homozygous deletion of CHD1, which was confirmed in the primary AC of the index patient. Loss of heterozygosity of TP53 and PTEN, and an allelic loss of RB1 with a transcriptomic signature compatible with Rb pathway aberration were revealed. Knockdown of PEG10 using shRNA significantly suppressed growth in vivo. Introduction of luciferase allowed serial monitoring of cells implanted orthotopically or in the renal subcapsule. Although H3K27me was reduced by EZH2 inhibition, reversion to AC was not observed. KUCaP13 is the first patient-derived, treatment-related NEPC cell line with triple loss of tumor suppressors critical for NEPC development through lineage plasticity. It could be valuable in research to deepen the understanding of NEPC.

In GC, cisplatin was cross-resistant to oxaliplatin and 5-fluorouracil which, on the other hand, was not cross-resistant to the other cytotoxic drugs. The activity of sunitinib did not obviously correlate to that of the standard drugs. AIM: To investigate the in vitro anti-hepatoma activity of taxol against SMMC-7721 human hepatoma cells. METHODS: The hepatoma cell growth was measured by MTT-microculture tetrazolium assay. Cell-cycle kinetics and apoptosis were analyzed by flow cytometry and microscopic examination. RESULTS: Taxol inhibited the hepatoma cell growth in concentration- and time-dependent manners with IC50 Of 18.96 nmol.L-1. Marked cell accumulation in G(2)/M phase and multinucleated cells were also observed after treatment with taxol 10 nmol.L-1. In addition, taxol at 10 nmol.L-1 could induce the apoptosis of hepatoma cells. CONCLUSION: Taxol suppresses the growth of SMMC-7721 human hepatoma cells in vitro by causing cell-cycle arrest, aberrant mitosis, and apoptosis of the human hepatoma cells.

Prostate cancer is the most common solid tumor among men in Western industrialized countries. A major problem in prostate cancer treatment is the development of androgen-independence, as androgen-deprivation therapy is the basic therapy for the disease. Molecular mechanisms behind prostate cancer and androgen-independent growth development are poorly known. In this study, subtractive hybridization was used for the generation of a cDNA library specific for prostate cancer. Analysis of the cDNA library revealed over-expression of several ribosomal proteins namely L4, L5, L7a, L23a, L30, L37, S14 and S18, in prostate cancer cell lines. Over-expression of L7a and L37 was also confirmed in prostate cancer tissue samples. Further, cDNA array was used in order to examine differentially expressed genes in androgen-dependent and androgen-independent prostate cancer cell line LNCaP. Monoamine oxidase A, an Expressed Sequence Tag (EST) similar to rat P044, and EST AA412049 were highly over-expressed in androgen-dependent LNCaP cells. Tissue-type plasminogen activator, interferon-inducible protein p78 (MxB), an EST similar to galectin-1, follistatin, fatty acid-binding protein 5, EST AA609749, annexin I, the interferon-inducible gene 1-8U and phospholipase D1 were highly over-expressed in androgen-independent LNCaP cells. The EST similar to rat P044, the EST similar to galectin-1, follistatin, annexin I and the interferon-inducible gene 1-8U were also expressed in benign prostatic hyperplasia tissue. The Y-linked ribosomal protein S4, Mat-8, and EST AA307912 were highly expressed in benign prostatic hyperplasia tissue.
