Aims/hypothesis: The Latino population has been systematically underrepresented in large-scale genetic analyses, and previous studies have relied on the imputation of ungenotyped variants based on the 1000 Genomes (1000G) imputation panel, which results in suboptimal capture of low-frequency or Latino-enriched variants. The National Heart, Lung, and Blood Institute (NHLBI) Trans-Omics for Precision Medicine (TOPMed) released the largest multi-ancestry genotype reference panel representing a unique opportunity to analyse rare genetic variations in the Latino population. We hypothesise that a more comprehensive analysis of low/rare variation using the TOPMed panel would improve our knowledge of the genetics of type 2 diabetes in the Latino population. Methods: We evaluated the TOPMed imputation performance using genotyping array and whole-exome sequence data in six Latino cohorts. To evaluate the ability of TOPMed imputation to increase the number of identified loci, we performed a Latino type 2 diabetes genome-wide association study (GWAS) meta-analysis in 8150 individuals with type 2 diabetes and 10,735 control individuals and replicated the results in six additional cohorts including whole-genome sequence data from the All of Us cohort. Results: Compared with imputation with 1000G, the TOPMed panel improved the identification of rare and low-frequency variants. We identified 26 genome-wide significant signals including a novel variant (minor allele frequency 1.7%; OR 1.37, p=3.4 × 10−9). A Latino-tailored polygenic score constructed from our data and GWAS data from East Asian and European populations improved the prediction accuracy in a Latino target dataset, explaining up to 7.6% of the type 2 diabetes risk variance. The COVID-19 outbreak and subsequent public health interventions have depressed demand and disrupted supply chains for many fishing businesses. This paper provides an analysis of the COVID-19 impacts on the profitability of the EU fishing fleets. Nowcasting techniques were used to estimate the impact of the COVID-19 pandemic on the economic performance for the EU fishing fleet in 2020 and 2021. Our results show that the economic impact of COVID-19 on this sector was smaller than initially expected and overall profits remained positive. This was in part due to low fuel prices that reduced operating costs of fishing, and the early response from governments to support the sector. The results vary by fishing fleet, revealing that small-scale fleets and the fleets in the Mediterranean and Black seas have been more impacted than large-scale fleets and the fleets in the Northeast Atlantic.

Protein-coding genetic variants that strongly affect disease risk can yield relevant clues to disease pathogenesis. Here we report exome-sequencing analyses of 20,791 individuals with type 2 diabetes (T2D) and 24,440 non-diabetic control participants from 5 ancestries. We identify gene-level associations of rare variants (with minor allele frequencies of less than 0.5%) in 4 genes at exome-wide significance, including a series of more than 30 SLC30A8 alleles that conveys protection against T2D, and in 12 gene sets, including those corresponding to T2D drug targets (P = 6.1 × 10-3) and candidate genes from knockout mice (P = 5.2 × 10-3). Within our study, the strongest T2D gene-level signals for rare variants explain at most 25% of the heritability of the strongest common single-variant signals, and the gene-level effect sizes of the rare variants that we observed in established T2D drug targets will require 75,000-185,000 sequenced cases to achieve exome-wide significance. We propose a method to interpret these modest rare-variant associations and to incorporate these associations into future target or gene prioritization efforts.

The highly competitive international market is challenging the Australian sugar industry with volatile sugar prices particularly when prices are low. The need to be sustainable during periods of low global sugar prices is driving the industry to seek ways to reduce maintenance costs. Generally, the current maintenance decision making is based on traditional experience-based practices despite high maintenance expenditure.. This study has proposed a general framework for the implementation of condition-based maintenance in sugar mills. The structure of the framework has been discussed, including the equipment condition assessment tools, the operation and maintenance cost models, and the maintenance optimisation approach. Subsequently, a case study on the condition assessment of boiler station components in an Australian sugar mill has been presented. The main function of the condition assessment tool is to generate a set of reliable condition indicators based on multiple data sources (e.g. work orders, process data).

The development of anticancer drug delivery systems which retain or enhance the cytotoxic properties of the drug to tumorous tissues, while reducing toxicity to other organs is of key importance. We investigated different poly(methacrylic acid)-g-poly(ethyleneglycol methacrylate) polymers as in situ coating agents for magnetite nanocrystallites. The obtained magnetic nano-assemblies were in turn thoroughly characterized for their structural, colloidal and physicochemical properties (drug loading capacity/release, magnetic field triggered drug release, cell uptake and localization) in order to select the best performing system. With the focus on in vivo validation of such magnetic drug delivery systems for first time, we selected cisplatin as the drug, since it is a potent anticancer agent which exhibits serious side effects due to lack of selectivity. In addition, cisplatin would offer facile determination of the metal content in the animal tissues for biodistribution studies. Alongside post-mortem Pt determination in the tissues, the biodistribution of the drug nanocarriers was also monitored in real time with PET-CT (positron emission tomography/computed tomography) with and without the presence of magnetic field gradients; using a novel chelator-free method, the nanoparticles were radiolabeled with 68Ga without having to alter their structure with chemical modifications for conjugation of radiochelators. The ability to be radiolabeled in such a straightforward but very robust way, along with their measured high MRI response, renders them attractive for dual imaging, which is an important functionality for translational investigations. Their anticancer properties were evaluated in vitro and in vivo, in a cisplatin resistant HT-29 human colon adenocarcinoma model, with and without the presence of magnetic field gradients. Gene transfer has therapeutic potential for treating HIV-1 infection by generating cells that are resistant to the virus. We have engineered a novel self-inactivating lentiviral vector, LVsh5/C46, using two viral-entry inhibitors to block early steps of HIV-1 cycle. The LVsh5/C46 vector encodes a short hairpin RNA (shRNA) for downregulation of CCR5, in combination with the HIV-1 fusion inhibitor, C46. We demonstrate here the effective delivery of LVsh5/C46 to human T cell lines, peripheral blood mononuclear cells, primary CD4(+) T lymphocytes, and CD34(+) hematopoietic stem/progenitor cells (HSPC). CCR5-targeted shRNA (sh5) and C46 peptide were stably expressed in the target cells and were able to effectively protect gene-modified cells against infection with CCR5- and CXCR4-tropic strains of HIV-1. LVsh5/C46 treatment was nontoxic as assessed by cell growth and viability, was noninflammatory, and had no adverse effect on HSPC differentiation. LVsh5/C46 could be produced at a scale sufficient for clinical development and resulted in active viral particles with very low mutagenic potential and the absence of replication-competent lentivirus. Based on these in vitro results, plus additional in vivo safety and efficacy data, LVsh5/C46 is now being tested in a phase 1/2 clinical trial for the treatment of HIV-1 disease.

FOR SUGAR factories with cogeneration plants major changes to the process stations have been undertaken to reduce the consumption of exhaust steam from the turbines and maximise the generated power. In many cases the process steam consumption has been reduced from greater than 52% on cane to ~40% on cane. The main changes have been to install additional evaporation area at the front of the set, operate the pan stages on vapour from No 1 or No 2 effects and undertake juice heating using vapour bleed from evaporators as far down the set as the penultimate stage. Operationally, one of the main challenges has been to develop a control system for the evaporators that addresses the objectives of juice processing rate (throughput) and steam economy, while producing syrup consistently at the required brix and providing an adequate and consistent vapour pressure for the pan stage operations. The cyclic demand for vapour by batch pans causes process disturbances through the evaporator set and these must be regulated in an effective manner to satisfy the above list of objectives for the evaporator station. The impact of the cyclic pan stage vapour demand has been modelled to define the impact on juice rate, steam economy, syrup brix and head space pressures in the evaporators. Experiences with the control schemes used at Pioneer and Rocky Point Mills are discussed. For each factory the paper provides information on (a) the control system used, the philosophy behind the control system and experiences in reaching the current system for control (b) the performance of the control system to handle the disturbances imposed by the pan stage and operate within other constraints of the factory (c) deficiencies in the current system and plans for further improvements. Other processing changes to boost the performance of the evaporators are also discussed.

The Russian Academy of Sciences and Federal Space Agency, together with the participation of many international organizations, worked toward the launch of the RadioAstron orbiting space observatory with its onboard 10-m reflector radio telescope from the Baikonur cosmodrome on July 18, 2011. Together with some of the largest ground-based radio telescopes and a set of stations for tracking, collecting, and reducing the data obtained, this space radio telescope forms a multi-antenna ground-space radio interferometer with extremely long baselines, making it possible for the first time to study various objects in the Universe with angular resolutions a million times better than is possible with the human eye. The project is targeted at systematic studies of compact radio-emitting sources and their dynamics. Objects to be studied include supermassive black holes, accretion disks, and relativistic jets in active galactic nuclei, stellar-mass black holes, neutron stars and hypothetical quark stars, regions of formation of stars and planetary systems in our and other galaxies, interplanetary and interstellar plasma, and the gravitational field of the Earth. The results of ground-based and inflight tests of the space radio telescope carried out in both autonomous and ground-space interferometric regimes are reported. This phase-referenced mode is the basis of the Palomar High-precision Astrometric Search for Exoplanet Systems (PHASES), a search for giant planets orbiting either the primary or secondary star in fifty binary systems. We present the first science results from the PHASES search. The properties of the stars comprising binary systems are determined to high precision. The mutual inclinations of several hierarchical triple star systems have been determined. Objective: Evaluate spectral scaling properties of scalp electroencephalogram (EEG) and electromyogram (EMG), optimal spacing of electrodes, and strategies for mitigating EMG. Methods: EEG was recorded referentially from 9 subjects with a 64 channel linear array (electrodes 3 mm apart) placed parasagittally or transversely on forehead or occiput, at rest with eyes open or closed, or with deliberate EMG. Temporal (PSDt) and spatial (PSDx) power spectral densities were calculated with one-dimensional fast Fourier transform (FFT) for comparison with earlier analyses of intracranial EEG. Results: Scaling of PSDt from scalp resembled that from pia: near-linear decrease in log power with increasing log frequency (1/f(alpha)). Scalp PSDx decreased non-linearly and more rapidly than PSDx from pia. Peaks in PSDt (especially 4-12 Hz) and PSDx (especially 0.1-0.4 cycles/cm) revealed departures from 1/f(alpha). EMG power in PSDt was more "white" than 1/f(alpha). Conclusions: Smearing by dura-skull-scalp distorts PSDx more than PSDt of scalp EEG from 1/f(alpha) scaling at the pia. Spatial spectral peaks suggest that optimal scalp electrode spacing might be similar to I cm to capture non-local EEG components having the texture of gyri. Mitigation of EMG by filtering is unsatisfactory. A criterion for measuring EMG may support biofeedback for training subjects to reduce their EMG. Significance: High-density recording and log-log spectral display of EEG provide a foundation for holist studies of global human brain function, as an alternative to network approaches that decompose EEG into localized, modular signals for correlation and coherence. We report progress made over the past two years on a visible low-light-level 640 x 480- pixel CCD imager and the camera electronics developed for night vision applications. The CCD camera, operating at 30 frames per second, has demonstrated good performance to below starlight illuminance (<1 mLux) conditions. The high sensitivity of the CCD imager is the result of back-illumination and low-noise readout amplifiers. To improve the CCD bright-light-source performance, antiblooming drains have been integrated into the CCD architecture that inhibit pixel blooming for signal levels up to at least 105 times pixel full well. A model named “KIII” of the olfactory system con-tains an array of 64 coupled oscillators simulating the olfactorybulb (OB), with negative and positive feedback through low-passfilter lines from single oscillators simulating the anterior olfactorynucleus (AON) and prepyriform cortex (PC). It is implementedin C to run on Macintosh, IBM, or UNIX platforms. The outputcan be set by parameter optimization to point, limit cycle,quasi-periodic, or aperiodic (presumably chaotic) attractors. Thefirst three classes of solutions are stable under variations ofparameters and perturbations by input, but they are biologicallyunrealistic. Chaotic solutions simulate the properties of time-dependent densities of olfactory action potentials and EEG’s,but they transit into the basins of point, limit cycle, or quasi-periodic attractors after only a few seconds of simulated run time.Despite use of double precision arithmetic giving 64-bit words, theKIII model is exquisitely sensitive to changes in the terminal bitof parameters and inputs. The global stability decreases as thenumber of coupled oscillators in the OB is increased, indicatingthat attractor crowding reduces the size of basins in the model to???the size of the digitizing step (??? ). Chaotic solutions havingbiological verisimilitude are robustly stabilized by introducing low-level, additive noise from a random number generator at two biologically determined points: rectified, spatially incoherent noise on each receptor input line, and spatially coherent noise to the AON, a global control point receiving centrifugal inputs from various parts of the forebrain. Methods are presented for evaluating global stability in the high dimensional system from measurements of multiple chaotic outputs. Ranges of stability are shown for variations of connection weights (gains) in the KIII model. The system is devised for pattern classification.

We have produced deep radio maps of the double quasar 0957+561 from multiple-epoch VLA observations. To achieve high sensitivity to extended structure we have re-reduced the best available 1.6 GHz observations and have combined 5 GHz data from multiple array configurations. Regions of faint emission approximately 15 arcsec north and south of the radio source G are probably lobes associated with the lensing galaxy. An arc 5 arcsec to the east of G may be a stretched image of emission in the background quasar's environment. 1.4 arcsec southwest of G we detect a source that we interpret as an image of emission from the quasar's western lobe, which could provide a constraint on the slope of the gravitational potential in the central region of the lens. We report the discovery of a new gravitationally lensed radio source. Radio maps of MG0751+2716 show four lensed images, which, at higher resolution, are resolved into long arcs of emission. A group of galaxies is present in optical images, including the principal lensing galaxy, with a much brighter galaxy just a few arcseconds away. We have measured the redshift of this brighter galaxy. No optical counterpart to the background source has been detected. Lens models that can readily reproduce the lensed image positions all require a substantial shear component. However, neither the very elongated lens nor the bright nearby galaxy are correctly positioned to explain the shear. Lens models which associate the mass with the light of galaxies in the group can produce an acceptable fit, but only with an extreme mass-to-light ratio in one of the minor group members., Comment: 1 Latex aaspp4 manuscript, 17 pages with 5 postscript figures. This report covers in detail the solid state research work of the Solid State Division at Lincoln Laboratory for the period 1 August through 31 October 1980. The topics covered are Solid State Device Research, Quantum Electronics, Materials Research, Microelectronics, and Analog Device Technology. Funding is primarily provided by the Air Force, with additional support provided by the Army, DARPA, Navy, NASA, and DOE.

A new family of energetic solid propellants based on an energetic binder plus NH4ClO4 and Al is being developed. The binder consists of carboxy- terminated nitropolyesters synthesized from nitroacids and glycerol plasticized by bis(2fluoro-2,2-dinitroethyl) formal. These propellants are characterized by excellent thermal stability and insensitivity to impact and shock. Initial extrapolated values of tensile strength and elongation are 4.92 Kg/sq cm (70 psi) and 25-30%, respectively. The burning rate at 70.31 Kg/sq cm (1000 psi) is 0.914 cm/sec (0.36 in/sec) and the theoretical Isp is 268 seconds.

Details are given of the advanced vibration management system developed by Orica Mining Services which is used to measure, analyse and predict blast-induced ground vibrations based on a proprietary model. The new plant, which is integrated into the existing Goldstrike facility in Nevada, was developed to process 12 000 tons/d of refractory gold ore from open-pit and underground sources. The flowsheet comprises crushing, dry grinding, roasting, extensive gas purification, calcine quenching and leaching in the presence of activated carbon. Design, procurement, construction and commissioning of the roaster facility was carried out successfully over a three-and-a-half-year period by peaks of 400 engineering and 950 construction personnel. Este trabajo pretende ser la unión entre la teoría y la práctica de la gamificación aplicada a la propuesta de diseño, análisis y evaluación de materiales educativos, para la asignatura de Geografía e Historia, en el curso de 2o de ESO D del Centro Educativo Zola. El estudio sigue dos vertientes claramente definidas, la primera hace referencia a una estructura teórica en la que se dará una visión general y amplia sobre los juegos en relación con la educación y el aprendizaje desde la óptica de distintas disciplinas, lo que servirá de base para introducir y desarrollar el concepto de gamificación aplicado a la Pedagogía. La segunda, se refiere al trabajo práctico realizado con el curso mencionado, a partir de las premisas obtenidas sobre la aplicación de dinámicas de gamificación. Así pues, este trabajo quiere dar una visión global y completa del desarrollo de los juegos y de sus elementos, para la elaboración de herramientas (de gamificación) que permitan el aprendizaje a partir de una óptica distinta. Se parte de la premisa de que en la actualidad cada vez hay más adolescentes que dedican gran cantidad de su tiempo de ocio a distintas actividades lúdicas como los videojuegos y que, sin embargo, esto no se ve reflejado en las aulas en las que pasan, en torno a seis horas al día., This essay pretend to be a connection between theory and praxis about gamification used as a propose of design, assay and review of the educational materials for the Geography and History subject, in the second grade of Secondary, specifically to the D class in the Centro Educativo Zola. This research has two different strands. The first one is referred to a theoretical structure in which I can have a general and large view about games, from different scientific disciplines views. It will comes useful to introduce and develops the concept of gamification applied to Pedagogy. The second one, is referred to the practical work realized. Consequently, this study wants to give a global perspective about the games and their resources to elaborate tools with which could work in Pedagogy in different ways. However, the assumption is made of the knowledge of the situation about teenagers and the time they invest playing games, though, we are not including gamification on lectures.
