1. Investigating the role of Lysine Specific Demethylase 1 (LSD1) in embryonic development using a gastruloid model system

Abstract: Lysine specific demethylase 1 (LSD1) is a chromatin modifying protein which specifically demethylates the permissive histone marks H3K4me1/2 and thereby acts as a transcriptional repressor. It performs this activity as part of the CoREST (co-repressor of Repressor Element1 Silencing Transcription Factor) complex, which also encompasses histone deacetylase 1/2 (HDAC1/2) activity and is required for stability of the complex. LSD1 is essential in embryonic development, with loss of LSD1 resulting in embryonic lethality at ~E6.5, a developmental stage which correlates with the onset of gastrulation. To further investigate the role of LSD1 in embryonic development, we employed a model system, gastruloids, which closely mimic early embryogenesis and aspects of gastrulation. We generated gastruloids from both induced and control Lsd1 conditional knockout (KO) mouse embryonic stem cells (ESCs) and utilised RNAsequencing (RNA-seq) analysis to identify differentially expressed genes at timepoints representative of ESCs, early gastrulation and late gastrulation. We identified dysregulated expression of genes associated with mesodermal lineages, epithelial-tomesenchymal transition (EMT) and the bone morphogenic protein (BMP) pathway. To investigate whether gene expression changes were a result of the loss of direct LSD1 demethylase activity or of broader CoREST complex activity, we performed similar RNAseq analysis on gastruloids generated from cells with wildtype (WT) or catalytically inactive LSD1 rescue constructs. We identified genes whose expression was not rescued, suggesting dependence on LSD1 demethylase activity, and genes whose expression was fully or partially rescued, including genes associated with EMT and the BMP pathway. The work in this thesis has enabled us to gain further insight into the role of LSD1 during embryonic development.

2. Modelling and managing the charging of massed electric vehicles on constrained residential power networks

Abstract: This thesis investigates how Electric Vehicles (EVs) and UK residential 230 V Low Voltage (LV) networks interact, as EVs supplant fossil-fuel cars. Home EV charging can provoke LV overloads, invisible to analyses set at higher voltages using averaged data. The Feeder Phase Balancer (FPB) simulator is presented, which models EVs on an LV network. FPB simulates various EVs driving independent trips, with charging control by a local manager and/or a 3rd party Aggregation service. Vehicle to Grid, Demand Response, Fast Frequency Response and Time of Use services are modelled. Impacts on EV and LV conditions are recorded; the EV support capability of various LV networks is explored and remedial options assessed. New knowledge is found concerning seasonal EV need to charge and of ability of networks to support Winter / Summer charging. Recommendations to DNOs and interested parties are formed. The FPB has simulated over 4.9 x 10⁹ independent EV trips, finding: a) LV networks, built to historic guidelines for gas-heated homes, cannot support EVs replacing cars 1:1 without reinforcement and / or local ICT control; b) Replacing gas-heating with Heat Pumps adds far greater load, forcing network reinforcement for the great majority of UK LV networks. c) Localised LV overloads (faults and blackouts) rise with EV charging duties; d) Network conditions, EV numbers and characteristics, ambient temperature and driven distances affect overloads, seen as EVs reach 10% ~ 30% homes e) There is seasonality: cold Winters provoke overloads (possibly simultaneous) f) Local ICT management of charging helps limit overloads, but can cause undercharging. Vehicle to Grid EVs can reduce undercharging. g) Top-down control is found not beneficial e.g. Time of Use constraints can cause undercharging when imposed and "crowd-rush" overloads when lifted; h) Destination charger use has great seasonality (x4 to x22 rise in Winter); failing to charge threatens stranding vehicles in the evenings. The above outcomes are dependant upon assumptions (e.g. battery characteristics) which are described. A repository of results data is available.

3. Vernacular numeracy practices : an exploration of the numeracy resources young people bring to their learning

Abstract: Current narrow definitions of numeracy and mathematics within the functional mathematics agenda, conceptualised as the autonomous model (Street 1984,1995; Street, Baker and Tomlin 2008) of numeracy, offer students entering further education limited and deficit subject positions and may contribute to a continued lack of success in college mathematics and numeracy. Drawing on New Literacy Studies theory (Barton and Hamilton 1998; Baker and Street 2004; Heath 1983; Gee 1996; Ivanic et al 2009; Street 1984, 1995, 1998, 2000) and a funds of knowledge perspective, (Moll, Amanti, Neff, & Gonzalez 1992), I suggest that adopting a social practice approach to numeracy teaching and learning provides affordances to reframe these deficit positions through an examination of studentsâ€TM everyday numeracy practices. A social practice theory of numeracy constructs it as numeracy embedded in social purposes, context dependent and reflective of studentsâ€TM everyday social practices. It also recognises the importance of power in structuring relations between people and ideas within practices. In an effort to avoid reifying the distinction between home/school, formal/informal, I propose the term vernacular numeracy practices as a way of conceptualising studentsâ€TM out of college numeracy practices. Using focus groups, classroom observations and interviews, the research explores studentsâ€TM vernacular numeracy practices embedded in a range of purposes and across different settings of studentsâ€TM lives. The invisibility of studentsâ€TM socially situated, vernacular numeracy practices, both to themselves and their teachers is revealed. However, students were willing to engage with the research practice which provided opportunities for them to access and share their vernacular numeracy practices. These vernacular numeracy practices are not readily understood across learning contexts and the challenge for tutors and students is how to access and use them to challenge the largely deficit subject positionings available and taken up by students. Within the research, studentsâ€TM vernacular numeracy practices were â€ ̃translatedâ€TM into the language of the adult numeracy core curriculum as a way of supporting students and tutors to â€ ̃see through to the mathsâ€TM (Coben 2003). The purpose of extracting the mathematics which was embedded in studentsâ€TM out of college practices, was to support students to engage in the process of â€ ̃repairing their mathematical identityâ€TM which Gee (2007) suggests, needs to take place before students can move on to become successful learners of mathematics. The research concludes that valuing and using studentsâ€TM vernacular numeracy practices may provide opportunities for students with low or no qualifications in mathematics to recruit â€ ̃identities of participationâ€TM (Solomon 2009) in place of identities of exclusion.

4. Social taphonomy : agency, biography and chaîne opératoire of cattle bones in a mediaeval European city

Abstract: This PhD sets out to tackle the subject of animal bone material from heterogeneous pits, especially on the edge of Mediaeval European cities. These are often the most common feature encountered by zooarchaeologists yet the analysis given them often does little to add to our understanding of the lives and actions of the people who lived and worked in the city, or of the city's relationship with its region or hinterland. Chapter 1 reviews the approaches taken to answering some of these questions by zooarchaeologists in the past as well as outlining the history of taphonomic research as it applies to the question. Chapter 2 aims to provide context to current approaches to the subject through a brief overview of relevant urban history. Chapter 3 focuses on how zooarchaeologists have studied butchery, other carcass related products and their waste. This current approach is typified by a standard, or traditional, analysis of an assemblage from Princesshay, Exeter, South West Britain (a previously unstudied assemblage), in Chapter 4. The second half of the PhD takes a different tack. Having presented the status quo, chapter 5 looks at how similar questions from other allied branches of archaeology have been investigated. These conceptual models are used, in combination with the established approaches already identified, to propose a new model based on chaîne opératoire theory for analysing the flow of Mediaeval urban fauna material that make up the final assemblages of individual contexts. It is suggested that through an understanding of the Guilds, and therefore memes, of industry in the city (recognising the raw materials and wastes from the varied processes/trades), the animal bone data can provide further insights into society and the city from the same typical heterogeneous pits and ditches that ordinarily provide so little cheer for zooarchaeologists. In a short test-case, and again in chapter 6 with a large case study, the potential of this new model (using chaîne opératoire theory to inform interpretation of routinely recorded zooarchaeological information (including representation of particular body parts e.g. horns, ribs, vertebra, skulls, feet and modifications such as butchery evidence chop/cut/fracturing)), is explored by applying it to the same dataset from Mediaeval Exeter analysed in the first half of the PhD. The additional insights provided by the new model are then discussed. Employing this model in Princesshay suggests the development of a intricate system of trade specialisation and societal complexity between the earlier and later periods of Medieval Exeter in a more nuanced way than could be understood through the earlier analysis.

Abstract: Genome-wide association studies (GWAS) have identified more than 200 common genetic variants independently associated with colorectal cancer (CRC) risk, but the causal variants and target genes are mostly unknown. We sought to fine-map all known CRC risk loci using GWAS data from 100,204 cases and 154,587 controls of East Asian and European ancestry. Our stepwise conditional analyses revealed 238 independent association signals of CRC risk, each with a set of credible causal variants (CCVs), of which 28 signals had a single CCV. Our cis-eQTL/mQTL and colocalization analyses using colorectal tissue-specific transcriptome and methylome data separately from 1299 and 321 individuals, along with functional genomic investigation, uncovered 136 putative CRC susceptibility genes, including 56 genes not previously reported. Analyses of single-cell RNA-seq data from colorectal tissues revealed 17 putative CRC susceptibility genes with distinct expression patterns in specific cell types. Analyses of whole exome sequencing data provided additional support for several target genes identified in this study as CRC susceptibility genes. Enrichment analyses of the 136 genes uncover pathways not previously linked to CRC risk. Our study substantially expanded association signals for CRC and provided additional insight into the biological mechanisms underlying CRC development.

Abstract: Sea turtles are vulnerable to climate change since their reproductive output is influenced by incubating temperatures, with warmer temperatures causing lower hatching success and increased feminization of embryos. Their ability to cope with projected increases in ambient temperatures will depend on their capacity to adapt to shifts in climatic regimes. Here, we assessed the extent to which phenological shifts could mitigate impacts from increases in ambient temperatures (from 1.5 to 3°C in air temperatures and from 1.4 to 2.3°C in sea surface temperatures by 2100 at our sites) on four species of sea turtles, under a “middle of the road” scenario (SSP2-4.5). Sand temperatures at sea turtle nesting sites are projected to increase from 0.58 to 4.17°C by 2100 and expected shifts in nesting of 26–43 days earlier will not be sufficient to maintain current incubation temperatures at 7 (29%) of our sites, hatching success rates at 10 (42%) of our sites, with current trends in hatchling sex ratio being able to be maintained at half of the sites. We also calculated the phenological shifts that would be required (both backward for an earlier shift in nesting and forward for a later shift) to keep up with present-day incubation temperatures, hatching success rates, and sex ratios. The required shifts backward in nesting for incubation temperatures ranged from −20 to −191 days, whereas the required shifts forward ranged from +54 to +180 days. However, for half of the sites, no matter the shift the median incubation temperature will always be warmer than the 75th percentile of current ranges. Given that phenological shifts will not be able to ameliorate predicted changes in temperature, hatching success and sex ratio at most sites, turtles may need to use other adaptive responses and/or there is the need to enhance sea turtle resilience to climate warming.

Abstract: 3C84 is a nearby radio source with a complex total intensity structure, showing linear polarisation and spectral patterns. A detailed investigation of the central engine region necessitates the use of VLBI above the hitherto available maximum frequency of 86GHz. Using ultrahigh resolution VLBI observations at the highest available frequency of 228GHz, we aim to directly detect compact structures and understand the physical conditions in the compact region of 3C84. We used EHT 228GHz observations and, given the limited (u,v)-coverage, applied geometric model fitting to the data. We also employed quasi-simultaneously observed, multi-frequency VLBI data for the source in order to carry out a comprehensive analysis of the core structure. We report the detection of a highly ordered, strong magnetic field around the central, SMBH of 3C84. The brightness temperature analysis suggests that the system is in equipartition. We determined a turnover frequency of $\nu_m=(113\pm4)$GHz, a corresponding synchrotron self-absorbed magnetic field of $B_{SSA}=(2.9\pm1.6)$G, and an equipartition magnetic field of $B_{eq}=(5.2\pm0.6)$G. Three components are resolved with the highest fractional polarisation detected for this object ($m_\textrm{net}=(17.0\pm3.9)$%). The positions of the components are compatible with those seen in low-frequency VLBI observations since 2017-2018. We report a steeply negative slope of the spectrum at 228GHz. We used these findings to test models of jet formation, propagation, and Faraday rotation in 3C84. The findings of our investigation into different flow geometries and black hole spins support an advection-dominated accretion flow in a magnetically arrested state around a rapidly rotating supermassive black hole as a model of the jet-launching system in the core of 3C84. 8. Consistent Validation for Predictive Methods in Spatial Settings

Abstract: Spatial prediction tasks are key to weather forecasting, studying air pollution, and other scientific endeavors. Determining how much to trust predictions made by statistical or physical methods is essential for the credibility of scientific conclusions. Unfortunately, classical approaches for validation fail to handle mismatch between locations available for validation and (test) locations where we want to make predictions. This mismatch is often not an instance of covariate shift (as commonly formalized) because the validation and test locations are fixed (e.g., on a grid or at select points) rather than i.i.d. from two distributions. In the present work, we formalize a check on validation methods: that they become arbitrarily accurate as validation data becomes arbitrarily dense. We show that classical and covariate-shift methods can fail this check. We instead propose a method that builds from existing ideas in the covariate-shift literature, but adapts them to the validation data at hand. We prove that our proposal passes our check. 9. A structural design methodology to reduce structural complexity to improve coating application and performance in water ballast tanks

Abstract: The introduction of the IMO Performance Standard for Protective Coatings (PSPC) for dedicated water ballast tanks (WBT) has increased the importance of the coatings applied in these tanks. Typical structural configurations within Water Ballast Tanks (WBTs) have a high degree of complexity; these spaces contain a large extent of edges, corner details and welds, which are commonly cited as areas most likely to suffer coating failure. However there is no quantitative measure of complexity as an indicator of how difficult a structure is to coat reliably. The concept of ‘structural complexity’ is considered with the intention of improving the in-service performance of applied coatings by proposing ships structures that include the coating process as a design consideration. As a means to try and provide a quantitative measure indicative of how easy a structure is to coat, the idea of ‘structural complexity’ is developed based on fundamental structural features. This measure is then used to understand the influence that different stiffener profiles and stiffener spacing may have on the coating process if structural configurations are sought that have reduced complexity. Investigation of the principal developments of the coating process indicates that any improvement is unlikely to be driven by coating technology or process alone. If improvements are to be made the suggestion here is that they should be driven by improving the design of the structure to be coated. The intention is to promote a ‘design for the coating process’ methodology to achieve this. The global ship and structural design process have been reviewed, where the classical approach looked at the relationship between weight and strength. This work concurrently considers the implications of different structural configures on not only weight and strength but also ease of coating. The relationship between the topology of the structure and the physical task of applying paint to it has formed the foundation of a ‘design for coating’ methodology, where the influence of structural complexity on all aspects of performance is considered with equal merit. A coating cost estimator has been developed in order to demonstrate the potential savings that could be realised by considering the coating process during the design phase. A simple optimisation routine has been used to seek solutions for minimum, complexity, weight, steelwork and coating costs. This allowed the balance to be explored between the completing aspects of the steelwork and coating processes. These alternative design solutions have been assessed using mathematical computational methods to ensure that the designs provide adequate structural performance.

10. Exploring the traumatic impact of advanced cancer

Abstract: The number of individuals living longer after a diagnosis of advanced cancer (AC) is increasing. To provide effective care for this population, a sound understanding of the psychological impact of AC is necessary. Cancer has been shown to have a traumatic impact and can precipitate both post-traumatic stress (PTS) and post-traumatic growth (PTG). In individuals with cancer, PTS is manifested through intrusive thoughts and images related to the experience of cancer, avoidance of thoughts and feelings connected with this experience, hyper arousal, and negative changes to one’s mood and thinking. PTG describes the positive changes that can occur in response to managing a challenging life event, such as cancer. While PTS and PTG have been explored extensively in response to primary cancer, the traumatic impact of AC has received less attention. A review of the literature on PTS in response to AC identified 11 studies, which were collated and critiqued. Across studies, individuals reported high rates of PTS, which were influenced by numerous factors. In completing this review, the researcher noted that studies on PTG in response to AC were lacking. There was also a paucity of qualitative research in this area, and few studies with the partners of those with AC, despite evidence of high rates of traumatisation in this group. To remedy these limitations in the evidence base, a Q-methodology study (n=11) was conducted. This study explored patients’ and partners’ responses to living with AC, to determine whether the constructs of PTS and PTG resonated with their subjective experiences. This analysis interpreted four differing accounts of the impact of AC, three of which could be understood as stories involving PTG and PTS. The final paper in this thesis provides a reflective account of the research experience.

11. Buckling of stiffened variable stiffness panels

Abstract: Over the past five decades the aerospace industry has seen the increased use and development of composite materials and structures, largely due to their superior specific strength and stiffness in comparison with metals. These two major attributes, coupled with their increased tailorability, enable the design of lighter, more efficient structures. Traditional tailoring is limited to modifying the orientation of laminate plies to achieve the desired performance. However, recent advancements in tape laying and fibre placement technologies, have led to the possibility of laminates with varying fibre orientations in the plane of a ply, thereby creating variable stiffness (VS) structures. These VS laminates greatly expand the design space and provide designers with additional degrees of freedom and tailorability. In recent years, analytical methods and finite element analysis (FEA) have shown that VS laminates can deliver a significant improvement in buckling and post-buckling performance for plates and shells. A potential application, which exploits this enhanced buckling performance, is the use of VS laminates as the skin of a stiffened panel. Here, the VS skin is designed to redistribute in-plane loads towards the stiffeners, which act as panel breakers, providing an expected increase in buckling performance, thereby facilitating the design of lighter and more efficient structures. The present work was conducted with the main aim of developing fast, robust and accurate tools for the linear buckling analysis of novel stiffened VS panels. The Ritz energy method was used to develop semi-analytical models for various plate configurations subject to uniaxial and biaxial compression; firstly, plates exhibiting discontinuously varying stiffness terms with bend-stretch coupling, a feature commonly present in the skin-flange region of stiffened panels; secondly, thick plates and sandwich panels with continuously varying stiffness terms, through fibre-steering, by including a first-order shear deformation theory; and thirdly, blade stiffened VS panels by utilising the previously developed methods to capture the important features of discontinuous and continuous stiffness variations and transverse shear flexibility. Benchmarked against a commercial FEA package, the semi-analytical models are shown to be both an accurate and computationally efficient alternative, and thus well suited for design and optimisation. To conclude, design and heuristic optimisation case studies for civil airliner blade stiffened panels, with both straight fibre and VS skins, were performed applying practical design and failure constraints. Optimal VS designs showed improvements in structural efficiency compared to standard configurations, with weight savings over 5% and 20% for VS skins satisfying and neglecting the 10 % rule, applied only for 00 and 900 plies, respectively. Furthermore, the mass reductions were shown to be achievable utilising relatively few VS plies in some cases, thus providing an avenue for application without the need for significant changes in stiffener or skin design, rates of deposition, or design rules and guidelines.

Abstract: How do ABE/ESOL (Adult Basic Education/English for Speakers of Other Languages) programs shape adult learners, and how do adult learners, in turn, shape their programs? Beyond the acquisition of important skills (such as greater fluency in the English language) what are the bigger internal meanings for adults of participating in ABE/ESOL learning? The researchers carefully followed for a year or more the inner experiences of 41 ABE/ESOL learners from all over the world. They were enrolled in three different U.S. programs oriented to greater English language fluency and improved effectiveness in learners' roles as parents, workers, or students. This monograph is organized in the following fashion: In Chapter Two the researchers present their research method. In Chapter Three they acquaint readers with the look and feel of a number of qualitatively different ways of knowing in adulthood, generally, and as they may make sense of aspects of teaching and learning, in particular. The following four chapters explore their three research sites--first the student-oriented program at Bunker Hill Community College; then the parent-oriented program at Even Start; and finally two chapters on the work-oriented program at Polaroid. After these chapters the researchers explicitly address issues of role-related competence and their collaboration with Equipped for the Future. The last chapter sums up their findings and their implications for ABE/ESOL teaching and program planning. Two appendices then follow, reviewing, respectively, their quantitative findings and developmentally oriented competency charts identifying the way similar tasks might be performed within differing "cultures of mind." Abstract: Climate change mitigation necessitates substantial alterations to patterns of worldwide economic activity, be that reduction in demand, switches to new technology or 'end-of-pipe' abatement of greenhouse gases. There are profound political, economic and ethical questions surrounding the governance of the means, rate and location of change. Within advanced capitalist economies and internationally through the auspices of the United Nations Framework Convention on Climate Change emissions trading systems have been introduced as part of the broader neoliberal attempts to 'correct market failure' through the definition of new property rights.This thesis investigates the development, constitution and consequences of institutions for the production, exchange and consumption of credits for emissions reductions. Such credits are financial instruments awarded to organisations for putative reductions in emissions from 'business as usual'. In consumption, credits are equated with a quantity of emissions released elsewhere. The 'Instituted Economic Process' framework (Randles and Harvey, 2002) is used to distinguish the various classes of agent involved in these exchanges and identify the economic and non-economic relationships that constitute these institutions. Inspired by the economic anthropology of Karl Polanyi, this approach asks how economic activity is organised and stabilised within society without presuming that there are universal economic laws of 'the market', that there are essential properties of commodities and agents, or that all economic transfers are conducted within markets.I argue that crediting is a socially contingent process of commodification of atmospheric pollution which is both ontologically and normatively problematic. Extant institutions are shown to be precarious by appealing to neutral techno-scientific justifications but remaining reliant on subjective judgement. However, they are sufficiently consistent and credible that they persist and expand. These findings are of interest to the academic communities of political economy and environmental and economic geography, climate change policy makers and the environmental movement more broadly.

Abstract: This thesis examines the political dimension of comedy in performance through a practice-as-research project incorporating elements of stand-up comedy, relational art, and participatory performance. In the wake of the depoliticisation of live performance comedy in Britain after the incorporation into the mainstream of the agit-prop driven Alternative comedy of the 1980s, I question whether stand-up in particular can have a political efficacy greater than raising awareness or representing a political struggle. Satirical comedy, comedy of Carnival, and more recently, comedies of ‘transgression,’ are held as paradigmatic of comedy’s generic political dimension, and contemporary discourse celebrates the comedian’s ability to negotiate lines of offense or taste. Opposing this view, I argue that this ‘Canivalesque logic’ is incompatible with the ideological conditions of global capitalism. A ‘radical democratic’ comedy necessitates a focus on the relational and affective dimensions of comedy performance. Following from this theoretical framework, this thesis progresses through three phases of experimental practice. I begin by interrogating and expanding my existing practice as a ‘circuit comedian.’ Next, audience-performer relationships become the site of interrogation, and I engage in two projects influenced by participatory performance and relational aesthetics. The third phase returns to stand-up comedy, coloured by my previous experiments. This project results in a model of comic performance as embodied formalist critique of ideology. The results of this project contributes to a way of reading comedy performance, as well as to discourse about the politics of theatre and performance. It is also provides an exegesis of comic techniques and a sustained analysis of my practice as a comedian and artist. Overall, this project intends to escape the false choice faced by the politically-minded comedian today: to paraphrase a well-known Marx Brothers joke, when given the choice between commenting on the world or changing it, we should answer: ‘Yes, please!’

19. Numerical and Statistical Analysis of Long-Haul Undersea Optical Communication Systems

Abstract: Firstly, we numerically model a practical 20 Gb/s undersea configuration employing the Return-to-Zero Differential Phase Shift Keying data format. The modelling is completed using the Split-Step Fourier Method to solve the Generalised Nonlinear Schrdinger Equation. We optimise the dispersion map and per-channel launch power of these channels and investigate how the choice of pre/post compensation can influence the performance. After obtaining these optimal configurations, we investigate the Bit Error Rate estimation of these systems and we see that estimation based on Gaussian electrical current systems is appropriate for systems of this type, indicating quasi-linear behaviour. The introduction of narrower pulses due to the deployment of quasi-linear transmission decreases the tolerance to chromatic dispersion and intra-channel nonlinearity. We used tools from Mathematical Statistics to study the behaviour of these channels in order to develop new methods to estimate Bit Error Rate. In the final section, we consider the estimation of Eye Closure Penalty, a popular measure of signal distortion. Using a numerical example and assuming the symmetry of eye closure, we see that we can simply estimate Eye Closure Penalty using Gaussian statistics. We also see that the statistics of the logical ones dominates the statistics of the logical ones dominates the statistics of signal distortion in the case of Return-to-Zero On-Off Keying configurations.

Abstract: The spectrin superfamily is a diverse group of proteins variously involved in cross- linking, bundling and binding to the F-actin cytoskeleton. These proteins are modular in nature and interaction with actin occurs, at least in part, via CH domain containing ABDs. The actin binding domains of the spectrin superfamily proteins are all very similar in overall structure however the functions of the individual proteins differ greatly. Utrophin is a member of the spectrin superfamily and has been used extensively to investigate and model the association of actin-binding domains with F- actin; however, much controversy exists as to whether binding occurs when the domain is in an open or a closed conformation. The data herein specifically investigates the importance of the utrophin ABD inter- CH domain linker to the conformation of the domain and how this domain associates with F-actin. We provide evidence that this particular region of the ABD is particularly sensitive to mutation and that the conformation of the domain when in solution cannot be altered by affecting the electrostatic environment surrounding the protein. It has been assumed previously that the utrophin ABD adopts a closed and compact configuration in solution similar to the fimbrin crystal structure conformation; however we present evidence that suggests this is not the case. It has been proposed that the utrophin ABD may open from this closed conformation to bind F-actin in a more open manner, we present data that demonstrates that opening of the domain is not essential to F-actin binding and that there is very little conformation change associated with the domain upon interaction with F-actin. It appears that the utrophin ABD can bind F actin in two conformations. This supports current models of utrophin ABD binding where interaction with F-actin occurs in either an open or closed conformation. The data presented here provides an interesting insight into the utrophin ABD/F-actin interaction and raises many questions regarding the evaluation of current binding models. Future research stemming from this work will serve to further the understanding of how utrophin and related actin-binding proteins interact with F-actin.

Abstract: Retinal microglia represent a highly complex population of resident myeloid cells. Function ranges from control of neuronal growth to clearance of cellular debris. Microglia are implicated in the pathogenesis of diseases such as Multiple Sclerosis and Alzheimer's. Microglia possess a distinct immunophenotype in response to inflammatory mediators IFNγ and LPS where MHC class II, CD86 and CD40 are up regulated, conferring a putative antigen presenting function, but there is an IL-10 dependent loss of co-accessory molecule expression, suggesting that microglia may modulate immune responses within the retina and maintain the specialised immunoregulatory environment normally present. Maintenance of this environment is further mediated via retinal CD200 expression. Results suggest that CD200:CD200 receptor engagement suppresses macrophage activation and inhibition of this exacerbates tissue destruction. This thesis examined the contribution of CD200 expression on maintaining immune regulation within the retina. Novel data is presented that shows expression of CD200 on retinal vascular endothelium and neurons. CD200-/- mice have increased numbers of retinal microglia which display normal morphology, but unlike resident CD200+/+ retinal microglia express constitutive NOS2. In CD200-/- mice, onset and severity of IRBP peptide 1-20 induced EAU is accelerated but overall tissue destruction is no greater than seen in CD200+/+ mice. Despite increased levels of apoptosis within the retina throughout the course of disease, myeloid cell infiltrate is greater in CD200-/- mice at all time points, while the level of NOS2 expression during height of disease is less in CD200-/- mice. CD200-depletion does not affect either T cell proliferation or APC function. Results indicate that retinal vascular CD200 expression regulates myeloid cell numbers in the retina and trafficking during inflammation and that CD200:CD200 receptor balance regulates retinal microglial activation.

Abstract: Contemporary Black political discourse has recently engaged several differentpropositions and models seeking to actualize reparations for chattel slavery in the United States as a major political project. On September 30, 2021, the state of California established the Task Force to Study and Develop Reparation Proposals for African Americans after the ratification of Assembly Bill 3121.1 As reparations has entered mainstream political discourse, it is important to historicize the contemporary movement and the organizations leading the discussions regarding reparations. This project will begin with tracing back the reparations movement to some of the first recorded demands of financial restitution made by formerly enslaved Africans in the United States. Furthermore, this project will explore how demands of reparations for slavery and racist oppression have evolved in the contemporary social-economic and political landscape. This project will investigate the political ideologies and frameworks that influenced the demands crafted by leading theorists and advocates for the reparations movement during the 20th century.

Abstract: The vast majority of the Earth system is inaccessible to direct observation. Consequently, the structure and dynamics of the Earth can only be determined indirectly, via geophysical sensing. These methods have the mathematical form of an inverse problem, in which the data and the unknowns are linked by a physical process, such as seismic wave propagation. From the possibly noisy data, we have indirect access to the unknowns. The vast majority of geophysical inverse problems are ill-posed, and require the provision of a priori knowledge to stabilize the solution. This thesis investigates methods for designing inverse problems to better take advantage of geophysical or geological constraints, to allow better resolution or more interpretability of the solutions. Four major themes are investigated: In Chapter 2, we study the collection of a novel dataset of Rayleigh wave horizontal-to-vertical ratios to provide stronger constraints on upper-crustal structure in Southern California. In Chapters 3 and 4, we develop a method for wavefield-reconstruction of sparse seismic data, including heterogeneous networks consisting of both displacement and strain instruments. This method amounts to an inversion in data-space, and promises to unlock the potential of wavefield based methods for complex datasets. In Chapters 5 and 6, we investigate a new structural parameterization based on a combination of Gaussian processes and the level-set method, that better models discontinuous geological features such as sedimentary basins. We test our method on a variety of synthetic and real datasets, culminating in a detailed study of the northeastern Los Angeles basin, which we found to be significantly deeper and steeper than in previous models. Finally, we develop a method of model selection for noisy historical datasets, which we investigate using the case study of correcting Oldham's data misinterpretation in the 1906 paper that "discovered" Earth's core.

Abstract: In 2002, in Atkins v. Virginia, the Supreme Court abolished the death penalty for defendants with intellectual disabilities. The Court held that executing individuals with intellectual disabilities is cruel and unusual punishment, violating the Eighth Amendment. The Court afforded the states the power to define intellectual disability for the purpose of death penalty eligibility. Post-Atkins cases reveal that the states have composed superficial and oversimplified definitions of intellectual disability. State definitions lack consistency and include nonclinical standards. As a result, courts continue to sentence defendants with intellectual disabilities to death. This Note argues that states should adopt a uniform definition of intellectual disability for the purpose of death penalty eligibility and proposes a model standard in line with clinical standards.

Abstract: While the cost of sequencing genomes has decreased dramatically in recent years, this expense often remains nontrivial. Under a fixed budget, scientists face a natural trade-off between quantity and quality: spending resources to sequence a greater number of genomes or spending resources to sequence genomes with increased accuracy. Our goal is to find the optimal allocation of resources between quantity and quality. Optimizing resource allocation promises to reveal as many new variations in the genome as possible. We introduce a Bayesian nonparametric methodology to predict the number of new variants in a follow-up study based on a pilot study. When experimental conditions are kept constant between the pilot and follow-up, we find that our prediction is competitive with the best existing methods. Unlike current methods, though, our new method allows practitioners to change experimental conditions between the pilot and the follow-up. We demonstrate how this distinction allows our method to be used for more realistic predictions and for optimal allocation of a fixed budget between quality and quantity. We validate our method on cancer and human genomics data.

Abstract: Interpretation of resolved polarized images of black holes by the Event Horizon Telescope (EHT) requires predictions of the polarized emission observable by an Earth-based instrument for a particular model of the black hole accretion system. Such predictions are generated by general relativistic radiative transfer (GRRT) codes, which integrate the equations of polarized radiative transfer in curved spacetime. A selection of ray-tracing GRRT codes used within the EHT collaboration is evaluated for accuracy and consistency in producing a selection of test images, demonstrating that the various methods and implementations of radiative transfer calculations are highly consistent. When imaging an analytic accretion model, we find that all codes produce images similar within a pixel-wise normalized mean squared error (NMSE) of 0.012 in the worst case. When imaging a snapshot from a cell-based magnetohydrodynamic simulation, we find all test images to be similar within NMSEs of 0.02, 0.04, 0.04, and 0.12 in Stokes I, Q, U , and V respectively. We additionally find the values of several image metrics relevant to published EHT results to be in agreement to much better precision than measurement uncertainties.

Abstract: Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Abstract: Background: In recent years, advances in technology have led to an influx of mental health apps, in particular the development of mental health and well-being chatbots, which have already shown promise in terms of their efficacy, availability, and accessibility. The ChatPal chatbot was developed to promote positive mental well-being among citizens living in rural areas. ChatPal is a multilingual chatbot, available in English, Scottish Gaelic, Swedish, and Finnish, containing psychoeducational content and exercises such as mindfulness and breathing, mood logging, gratitude, and thought diaries. Objective: The primary objective of this study is to evaluate a multilingual mental health and well-being chatbot (ChatPal) to establish if it has an effect on mental well-being. Secondary objectives include investigating the characteristics of individuals that showed improvements in well-being along with those with worsening well-being and applying thematic analysis to user feedback. Methods: A pre-post intervention study was conducted where participants were recruited to use the intervention (ChatPal) for a 12-week period. Recruitment took place across 5 regions: Northern Ireland, Scotland, the Republic of Ireland, Sweden, and Finland. Outcome measures included the Short Warwick-Edinburgh Mental Well-Being Scale, the World Health Organization-Five Well-Being Index, and the Satisfaction with Life Scale, which were evaluated at baseline, midpoint, and end point. Written feedback was collected from participants and subjected to qualitative analysis to identify themes. Results: A total of 348 people were recruited to the study (n=254, 73% female; n=94, 27% male) aged between 18 and 73 (mean 30) years. The well-being scores of participants improved from baseline to midpoint and from baseline to end point; however, improvement in scores was not statistically significant on the Short Warwick-Edinburgh Mental Well-Being Scale (P=.42), the World Health Organization-Five Well-Being In, Validerad;2023;Nivå 2;2023-08-09 (hanlid)

Abstract: 01 september 2023, Contains fulltext : 295917.pdf (Publisher’s version ) (Open Access), BACKGROUND: The PERFORM study aimed to understand causes of febrile childhood illness by comparing molecular pathogen detection with current clinical practice. METHODS: Febrile children and controls were recruited on presentation to hospital in 9 European countries 2016-2020. Each child was assigned a standardized diagnostic category based on retrospective review of local clinical and microbiological data. Subsequently, centralised molecular tests (CMTs) for 19 respiratory and 27 blood pathogens were performed. FINDINGS: Of 4611 febrile children, 643 (14%) were classified as definite bacterial infection (DB), 491 (11%) as definite viral infection (DV), and 3477 (75%) had uncertain aetiology. 1061 controls without infection were recruited. CMTs detected blood bacteria more frequently in DB than DV cases for N. meningitidis (OR: 3.37, 95% CI: 1.92-5.99), S. pneumoniae (OR: 3.89, 95% CI: 2.07-7.59), Group A streptococcus (OR 2.73, 95% CI 1.13-6.09) and E. coli (OR 2.7, 95% CI 1.02-6.71). Respiratory viruses were more common in febrile children than controls, but only influenza A (OR 0.24, 95% CI 0.11-0.46), influenza B (OR 0.12, 95% CI 0.02-0.37) and RSV (OR 0.16, 95% CI: 0.06-0.36) were less common in DB than DV cases. Of 16 blood viruses, enterovirus (OR 0.43, 95% CI 0.23-0.72) and EBV (OR 0.71, 95% CI 0.56-0.90) were detected less often in DB than DV cases. Combined local diagnostics and CMTs respectively detected blood viruses and respiratory viruses in 360 (56%) and 161 (25%) of DB cases, and virus detection ruled-out bacterial infection poorly, with predictive values of 0.64 and 0.68 respectively. INTERPRETATION: Most febrile children cannot be conclusively defined as having bacterial or viral infection when molecular tests supplement conventional approaches. Abstract: We present an in-depth analysis of a newly proposed correlation function in visibility space, between the E and B modes of the linear polarization, hereafter the EB-correlation, for a set of time-averaged GRMHD simulations compared with the phase map from different semi-analytic models as well as the Event Horizon Telescope (EHT) 2017 data for M87* source. We demonstrate that the phase map of the time-averaged EB-correlation contains novel information that might be linked to the BH spin, accretion state and the electron temperature. A detailed comparison with a semi-analytic approach with different azimuthal expansion modes shows that to recover the morphology of the real/imaginary part of the correlation function and its phase, we require higher orders of these azimuthal modes. To extract the phase features, we propose to use the Zernike polynomial reconstruction developing an empirical metric to break degeneracies between models with different BH spins that are qualitatively similar. We use a set of different geometrical ring models with various magnetic and velocity field morphologies and show that both the image space and visibility based EB-correlation morphologies in MAD simulations can be explained with simple fluid and magnetic field geometries as used in ring models. SANEs by contrast are harder to model, demonstrating that the simple fluid and magnetic field geometries of ring models are not sufficient to describe them owing to higher Faraday Rotation depths. A qualitative comparison with the EHT data demonstrates that some of the features in the phase of EB-correlation might be well explained by the current models for BH spins as well as electron temperatures, while others may require a larger theoretical surveys., Comment: 26 pages, 21 Figures

Abstract: BackgroundThe efficacy of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) convalescent plasma (CCP) for preventing infection in exposed, uninfected individuals is unknown. CCP might prevent infection when administered before symptoms or laboratory evidence of infection.MethodsThis double-blinded, phase 2 randomized, controlled trial (RCT) compared the efficacy and safety of prophylactic high titer (≥1:320 by Euroimmun ELISA) CCP with standard plasma. Asymptomatic participants aged ≥18 years with close contact exposure to a person with confirmed coronavirus disease 2019 (COVID-19) in the previous 120 hours and negative SARS-CoV-2 test within 24 hours before transfusion were eligible. The primary outcome was new SARS-CoV-2 infection.ResultsIn total, 180 participants were enrolled; 87 were assigned to CCP and 93 to control plasma, and 170 transfused at 19 sites across the United States from June 2020 to March 2021. Two were excluded for screening SARS-CoV-2 reverse transcription polymerase chain reaction (RT-PCR) positivity. Of the remaining 168 participants, 12/81 (14.8%) CCP and 13/87 (14.9%) control recipients developed SARS-CoV-2 infection; 6 (7.4%) CCP and 7 (8%) control recipients developed COVID-19 (infection with symptoms). There were no COVID-19-related hospitalizations in CCP and 2 in control recipients. Efficacy by restricted mean infection free time (RMIFT) by 28 days for all SARS-CoV-2 infections (25.3 vs 25.2 days; P = .49) and COVID-19 (26.3 vs 25.9 days; P = .35) was similar for both groups.ConclusionsAdministration of high-titer CCP as post-exposure prophylaxis, although appearing safe, did not prevent SARS-CoV-2 infection.Clinical trials registrationNCT04323800.

Abstract: BackgroundThe coronavirus pandemic has caused a worldwide health crisis. Bariatric patients require extensive pre- and post-operative follow-up, which may be less feasible during public health social distancing mandates. We assessed the impact of the pandemic on the behaviors and weight loss outcomes of our pre- and post-operative bariatric patients.MethodsA retrospective review of a prospectively maintained database identified patients who underwent either sleeve gastrectomy (SG) or Roux-en-Y gastric bypass (RYGB) at a single institution between March 2018 and May 2020. A cohort undergoing surgery within 12 months before the pandemic as well as a cohort undergoing a medically supervised diet prior to surgery was surveyed regarding pre- and post-COVID-19 lifestyle habits. Excess weight loss (EWL) outcomes from a group of pre-COVID surgical patients were compared to that of a group of post-COVID surgical patients. Primary outcome was whether the lockdown changed 1-year weight loss outcomes. Secondary outcome was whether patient lifestyle behaviors were changed during the pandemic.ResultsThere was no difference in 1-year EWL between pre- and post-COVID SG patients (51.7% versus 55.9%, p = 0.35), or between pre- and post-COVID RYGB patients (88.9% versus 80.4%, p = 0.42). Pre-stay-at-home order, 91.8% endorsed physical activity compared to 80.3% post-stay-at-home order (p = 0.0025). Mean physical activity decreased from 4.2 h/week to 2.7 h/week after the stay-at-home order (p < 0.0001). Additionally, 41.3% reported worsened dietary habits post-stay-at-home order.ConclusionThe COVID-19 pandemic has greatly impacted the behaviors of bariatric surgery patients. Despite deterioration of lifestyle habits, 1-year weight loss outcomes after bariatric surgery remained the same before and after the instatement of social distancing measures. Abstract: BackgroundNo platform for objective, synchronous and on-line evaluation of both intraoperative error and surgeon physiology yet exists. Electrokardiogram (EKG) metrics have been associated with cognitive and affective features that are known to impact surgical performance but have not yet been analyzed in conjunction with real-time error signals using objective, real-time methods.MethodsEKGs and operating console point-of-views (POVs) for fifteen general surgery residents and five non-medically trained participants were captured during three simulated robotic-assisted surgery (RAS) procedures. Time and frequency-domain EKG statistics were extracted from recorded EKGs. Intraoperative errors were detected from operating console POV videos. EKG statistics were synchronized with intraoperative error signals.ResultsRelative to personalized baselines, IBI, SDNN and RMSSD decreased 0.15% (S.E. 3.603e-04; P = 3.25e-05), 3.08% (S.E. 1.603e-03; P < 2e-16) and 1.19% (S.E. 2.631e-03; P = 5.66e-06), respectively, during error. Relative LF RMS power decreased 1.44% (S.E. 2.337e-03; P = 8.38e-10), and relative HF RMS power increased 5.51% (S.E. 1.945e-03; P < 2e-16).ConclusionsUse of a novel, on-line biometric and operating room data capture and analysis platform enabled detection of distinct operator physiological changes during intraoperative errors. Monitoring operator EKG metrics during surgery may help improve patient outcomes through real-time assessments of intraoperative surgical proficiency and perceived difficulty as well as inform personalized surgical skills development.

Abstract: BackgroundMultisystem inflammatory syndrome in children (MIS-C), a hyperinflammatory condition associated with SARS-CoV-2 infection, has emerged as a serious illness in children worldwide. Immunoglobulin or glucocorticoids, or both, are currently recommended treatments.MethodsThe Best Available Treatment Study evaluated immunomodulatory treatments for MIS-C in an international observational cohort. Analysis of the first 614 patients was previously reported. In this propensity-weighted cohort study, clinical and outcome data from children with suspected or proven MIS-C were collected onto a web-based Research Electronic Data Capture database. After excluding neonates and incomplete or duplicate records, inverse probability weighting was used to compare primary treatments with intravenous immunoglobulin, intravenous immunoglobulin plus glucocorticoids, or glucocorticoids alone, using intravenous immunoglobulin as the reference treatment. Primary outcomes were a composite of inotropic or ventilator support from the second day after treatment initiation, or death, and time to improvement on an ordinal clinical severity scale. Secondary outcomes included treatment escalation, clinical deterioration, fever, and coronary artery aneurysm occurrence and resolution. This study is registered with the ISRCTN registry, ISRCTN69546370.FindingsWe enrolled 2101 children (aged 0 months to 19 years) with clinically diagnosed MIS-C from 39 countries between June 14, 2020, and April 25, 2022, and, following exclusions, 2009 patients were included for analysis (median age 8·0 years [IQR 4·2-11·4], 1191 [59·3%] male and 818 [40·7%] female, and 825 [41·1%] White). 680 (33·8%) patients received primary treatment with intravenous immunoglobulin, 698 (34·7%) with intravenous immunoglobulin plus glucocorticoids, 487 (24·2%) with glucocorticoids alone; 59 (2·9%) patients received other combinations, including biologicals, and 85 (4·2%) patients received no immunomodulators. Abstract: BackgroundMultidisciplinary approaches to weight loss have been shown to improve outcomes in bariatric patients. Few studies have been performed assessing the utility and compliance of fitness tracking devices after bariatric surgery. We aim to determine whether use of an activity tracking device assists bariatric patients in improving postoperative weight loss behaviors.MethodsA fitness wearable was offered to patients undergoing bariatric surgery from 2019 to 2022. A telephone survey was conducted to elucidate the impact of the device on the patient's postoperative weight loss efforts 6 to 12 months after surgery. Weight loss outcomes of sleeve gastrectomy (SG) patients receiving the fitness wearable (FW) were compared to those of a group of SG patients who did not receive one (non-FW).ResultsThirty-seven patients were given a fitness wearable, 20 of whom responded to our telephone survey. Five patients reported not using the device and were excluded. 88.2% reported that using the device had a positive impact on their overall lifestyle. Patients felt that using the fitness wearable to keeping track of their progress helped them both to achieve short-term fitness goals and sustain them in the long run. From the patients that utilized the device, 44.4% of those that discontinued felt like it helped them build a routine that they maintained even after they were no longer using it. Demographic data between FW and non-FW groups (age, sex, CCI, initial BMI, and surgery BMI) did not differ significantly. The FW group trended towards greater %EWL at 1 year post-operation (65.2% versus 52.4%, p = 0.066) and had significantly greater %TWL at 1 year post-operation (30.3% versus 22.3%, p = 0.02).ConclusionThe use of an activity tracking device enhances a patient's post-bariatric surgery experience, serving to keep patients informed and motivated, and leading to improved activity that may translate to better weight loss outcomes.

Abstract: Background: This study aims to quantitatively assess use of the NSQIP surgical risk calculator (NSRC) in contemporary surgical practice and to identify barriers to use and potential interventions that might increase use. Materials and methods: We performed a cross-sectional study of surgeons at seven institutions. The primary outcomes were self-reported application of the calculator in general clinical practice and specific clinical scenarios as well as reported barriers to use. Results: In our sample of 99 surgeons (49.7% response rate), 73.7% reported use of the NSRC in the past month. Approximately half (51.9%) of respondents reported infrequent NSRC use (<20% of preoperative discussions), while 14.3% used it in ≥40% of preoperative assessments. Reported use was higher in nonelective cases (30.2% vs 11.1%) and in patients who were ≥65 years old (37.1% vs 13.0%), functionally dependent (41.2% vs 6.6%), or with surrogate consent (39.9% vs 20.4%). NSRC use was not associated with training status or years in practice. Respondents identified a lack of influence on the decision to pursue surgery as well as concerns regarding the calculator's accuracy as barriers to use. Surgeons suggested improving integration to workflow and better education as strategies to increase NSRC use. Conclusions: Many surgeons reported use of the NSRC, but few used it frequently. Surgeons reported more frequent use in nonelective cases and frail patients, suggesting the calculator is of greater utility for high-risk patients. Surgeons raised concerns about perceived accuracy and suggested additional education as well as integration of the calculator into the electronic health record.

Abstract: Human mitochondrial (mt) protein assemblies are vital for neuronal and brain function, and their alteration contributes to many human disorders, e.g., neurodegenerative diseases resulting from abnormal protein–protein interactions (PPIs). Knowledge of the composition of mt protein complexes is, however, still limited. Affinity purification mass spectrometry (MS) and proximity-dependent biotinylation MS have defined protein partners of some mt proteins, but are too technically challenging and laborious to be practical for analyzing large numbers of samples at the proteome level, e.g., for the study of neuronal or brain-specific mt assemblies, as well as altered mtPPIs on a proteome-wide scale for a disease of interest in brain regions, disease tissues or neurons derived from patients. To address this challenge, we adapted a co-fractionation–MS platform to survey native mt assemblies in adult mouse brain and in human NTERA-2 embryonal carcinoma stem cells or differentiated neuronal-like cells. The workflow consists of orthogonal separations of mt extracts isolated from chemically cross-linked samples to stabilize PPIs, data-dependent acquisition MS to identify co-eluted mt protein profiles from collected fractions and a computational scoring pipeline to predict mtPPIs, followed by network partitioning to define complexes linked to mt functions as well as those essential for neuronal and brain physiological homeostasis. We developed an R/CRAN software package, Macromolecular Assemblies from Co-elution Profiles for automated scoring of co-fractionation–MS data to define complexes from mtPPI networks. Presently, the co-fractionation–MS procedure takes 1.5–3.5 d of proteomic sample preparation, 31 d of MS data acquisition and 8.5 d of data analyses to produce meaningful biological insights.

Abstract: Drew Kahuʻāina Broderick (Kanaka ‘Ōiwi) joins Lauren Wetmore in conversation about Māhealani Dudoit’s fundamental text, “Carving a Hawaiian Aesthetic,” published in the first issue of ‘Ōiwi: A Native Hawaiian Journal – He ‘oia mau nō kākou’, which Dudoit co-founded in 1998. Abstract: Sea turtles are vulnerable to climate change since their reproductive output is influenced by incubating temperatures, with warmer temperatures causing lower hatching success and increased feminization of embryos. Their ability to cope with projected increases in ambient temperatures will depend on their capacity to adapt to shifts in climatic regimes. Here, we assessed the extent to which phenological shifts could mitigate impacts from increases in ambient temperatures (from 1.5 to 3°C in air temperatures and from 1.4 to 2.3°C in sea surface temperatures by 2100 at our sites) on four species of sea turtles, under a “middle of the road” scenario (SSP2‐4.5). Sand temperatures at sea turtle nesting sites are projected to increase from 0.58 to 4.17°C by 2100 and expected shifts in nesting of 26–43 days earlier will not be sufficient to maintain current incubation temperatures at 7 (29%) of our sites, hatching success rates at 10 (42%) of our sites, with current trends in hatchling sex ratio being able to be maintained at half of the sites. We also calculated the phenological shifts that would be required (both backward for an earlier shift in nesting and forward for a later shift) to keep up with present‐day incubation temperatures, hatching success rates, and sex ratios. The required shifts backward in nesting for incubation temperatures ranged from −20 to −191 days, whereas the required shifts forward ranged from +54 to +180 days. However, for half of the sites, no matter the shift the median incubation temperature will always be warmer than the 75th percentile of current ranges. Given that phenological shifts will not be able to ameliorate predicted changes in temperature, hatching success and sex ratio at most sites, turtles may need to use other adaptive responses and/or there is the need to enhance sea turtle resilience to climate warming.

Abstract: The Event Horizon Telescope (EHT) is a millimeter very long baseline interferometry (VLBI) array that has imaged the apparent shadows of the supermassive black holes M87* and Sagittarius A*. Polarimetric data from these observations contain a wealth of information on the black hole and accretion flow properties. In this work, we develop polarimetric geometric modeling methods for mm-VLBI data, focusing on approaches that fit data products with differing degrees of invariance to broad classes of calibration errors. We establish a fitting procedure using a polarimetric “m-ring” model to approximate the image structure near a black hole. By fitting this model to synthetic EHT data from general relativistic magnetohydrodynamic models, we show that the linear and circular polarization structure can be successfully approximated with relatively few model parameters. We then fit this model to EHT observations of M87* taken in 2017. In total intensity and linear polarization, the m-ring fits are consistent with previous results from imaging methods. In circular polarization, the m-ring fits indicate the presence of event-horizon-scale circular polarization structure, with a persistent dipolar asymmetry and orientation across several days. The same structure was recovered independently of observing band, used data products, and model assumptions. Despite this broad agreement, imaging methods do not produce similarly consistent results. Our circular polarization results, which imposed additional assumptions on the source structure, should thus be interpreted with some caution. Polarimetric geometric modeling provides a useful and powerful method to constrain the properties of horizon-scale polarized emission, particularly for sparse arrays like the EHT.

Abstract: We describe the process to design, architect, and implement a transformative enhancement of the Event Horizon Telescope (EHT). This program—the next-generation Event Horizon Telescope (ngEHT)—will form a networked global array of radio dishes capable of making high-fidelity real-time movies of supermassive black holes (SMBH) and their emanating jets. This builds upon the EHT principally by deploying additional modest-diameter dishes to optimized geographic locations to enhance the current global mm/submm wavelength Very Long Baseline Interferometric (VLBI) array, which has, to date, utilized mostly pre-existing radio telescopes. The ngEHT program further focuses on observing at three frequencies simultaneously for increased sensitivity and Fourier spatial frequency coverage. Here, the concept, science goals, design considerations, station siting, and instrument prototyping are discussed, and a preliminary reference array to be implemented in phases is described.
