The environmental fate of metal ions is influenced by their interactions with natural organic and inorganic ligands, which modify the ions’ structure and charge and thus influence their interactions with mineral phases. We investigate the impact of ubiquitous sulfate on the retention of trivalent f-element cations (M(III) = Am, Eu, Y) by muscovite. We combine ex situ alpha spectrometry and in situ surface X-ray diffraction (i.e., crystal truncation rod and resonant anomalous X-ray reflectivity) to determine M(III) coverages and interfacial structures at the molecular level. M(III) cations adsorb as two distinct outer-sphere (OS) complexes (i.e., adsorbed and extended OS complexes) whose coverages vary with increasing sulfate concentration, [SO42-]. When [SO42-] ≤ 0.4 mM, coverages increase and exceed the amounts needed for surface charge compensation of muscovite by a factor of ~3. This overcompensation is likely controlled by ion-ion correlations at the mineral/water interface rather than adsorption of MSO4+, which has a lower thermodynamic stability in the solutions and weaker electrostatic attraction to the mica surface than M3+. For higher [SO42-], MSO4+ and M(SO4)2- dominate solution speciation, leading to a strong decrease of the M(III) coverage due to their lower sorption affinity and weaker ion-ion correlations compared to M3+.

1 Introduction Transport of contaminants, e.g. radionuclides, in the environment depends strongly on their interactions with mineral phases. In a repository for radioactive waste, crystalline rock (e.g. granite) as one potential host rock in Germany and many other countries, may affect the mobility of radionuclides. Main constituents of granite are feldspars. In spent nuclear fuel, trivalent actinides (Am, Cm, but also Pu) contribute strongly to the radiotoxicity. Therefore, this work studies the retention of Am and Cm, as well as their rare earths element analogues (Eu, La, Lu, Nd, Y) on K-feldspar. By combining batch sorption experiments and time-resolved laser-induced fluorescence spectroscopy (TRLFS), a generic surface complexation model (SCM) was obtained that is valid for all investigated M3+. Thermodynamic sorption data were obtained and an understanding of sorption mechanisms on the molecular level was achieved. 2 Results Batch sorption experiments were performed over a broad range of environmental conditions (pH 4 – 10, [M3+] = 52 nM – 10 µM, 3 – 50 g/L K-feldspar (dp < 21 µm; 63 – 200 µm))[1]. Sorption is weak for pH < 5, strongly increases between pH 5 – 7 and reaches complete uptake at higher pH. By deconvolution of Cm emission spectra, an inner-sphere complex and its first two hydrolysis forms were found to be responsible for retention in this pH range. For determination of the deprotonation constant pKa of K-feldspar, as one important input parameter of the model, column titration experiments were conducted. Batch sorption results of all studied M3+ were used to develop two alternative SCMs. The experimental sorption data were used to determine surface complexation parameters by coupling the parameter estimation code UCODE with PHREEQC (SCM-A). In a second approach, spectroscopic data were also considered (SCM-B). The mobility of heavy metal contaminants and radionuclides in the environment is directly controlled by their interactions with charged mineral surfaces, hence an assessment of their potential toxicity, e.g. in the context of nuclear waste disposal sites, requires understanding of sorption processes on the molecular level. Here, we investigate the sorption of a variety of rare earth elements (REE) and trivalent actinides (Am, Cm) on K-feldspar using batch sorption and column transport experiments, time-resolved laser-induced fluorescence spectroscopy (TRLFS), and a surface complexation model. Initially, a reliable pKa for K-feldspar’s surface deprotonation reaction was determined as 2.5 ± 0.02, in excellent agreement with a measured pHIEP of 2.8. Batch sorption experiments over a broad range of experimental conditions in terms of mineral grain size, pH, [M3+], ionic radius, solid/liquid ratio, ionic strength, and equilibration procedures were carried out to quantify macroscopic immobilization. Similar pH-dependent uptake behavior was found for all investigated trivalent REE and actinides. In parallel, spectroscopic investigations provided insight into surface speciation. Cm TRLFS spectra indicate the formation of three inner-sphere sorption complexes with increasing hydrolysis. Additionally, a ternary K-feldspar/Cm/silicate complex was found for pH > 10, and batch and spectroscopic data at low pH (< 4) point to small amounts of outer sphere sorption complexes. Based on TRLFS data, batch sorption, and titration data, a generic geochemical sorption model was developed, that describes sorption edges for all investigated M3+/K-feldspar systems satisfactorily. The derived stability constants for the binary sorption complexes (logK1-4 = −3.6, −7.7, −11.5, and −17.4, respectively) are in good agreement with previous studies on similar systems, and could successfully be used to reproduce literature data. Weak retardation below pH 5, followed by a strong increase between pH 5 and 7 and complete removal from solution at pH ≥ 8 was observed for all investigated metals. The fundamental aspects of uranyl-spectroscopy (absorption, luminescence, IR), data interpretation and subsequent conclusions to interpret the U(VI)-speciation will be discussed. Furthermore, it will be explained how spectroscopy (NMR and IR) can be used to identify the binding properties of organic molecules, exemplarily explained for a polyhydroxy-carboxylic acid.

Acetate (AcO-) can be a biotic or abiotic degradation product of organic material present in nuclear waste. Additionally it is often declared as model compound for more complex organic structures. A large number of studies is available which investigated the speciation of uranium(VI) in the presence of acetate under acidic conditions. From potentiometric titrations it seems to be very clear that three uranyl-acetate species are formed under acidic conditions: UO2AcO+ (1:1), UO2(AcO)2 (1:2) and UO2(AcO)3- (1:3). Spectroscopic methods (e.g. IR and EXAFS) provide no certainty whether the 1:3 complex exists or not. TRLFS and UV-vis spectroscopy were mainly applied to characterize the formation of the 1:1 complex. In this study TRLFS, ATR-FT-IR and UV-vis spectroscopy were applied to determine the speciation of uranium(VI) in the presence of acetate and to fill the gap of missing spectroscopic data for the other formed species. Experimental conditions were chosen so that all three previous identified species should be formed. Data evaluation was performed on the basis of factor analysis resulting in single component spectra and stability constants. The findings led us to the conclusion that the 1:3 complex was not formed. In addition the detailed evaluation of ATR-FT-IR spectra show that an approximation used in potentiometric studies should be treated with caution. All these indications reinforce the doubts with respect to the existence of the UO2(AcO)3- species.

The progress and results of these investigations as part of the MIND project (work package 1) will be presented.

The interaction of acetic acid as the smallest carboxylic acid with a side chain with uranium (VI) is of importance in two respects: firstly, it occurs often as a degradation product of organic components of radioactive waste (e.g. cellulose, bitumen, PVC), and secondly it can serve as model for more complex organic compounds like humic acids. Three complexes can be formed in aqueous solutions: UO2(AcO)n2-n, where n is 1,2, or 3. These complexes are well characterized under various experimental conditions in terms of stability and structure, reported in a number of publications. Recently published articles focused also on spectroscopic properties of uranium (VI) acetate complexes using on the one hand time-resolved laser-induced fluorescence spectroscopy (TRLFS) and on the other hand UV-vis spectroscopy. The uranium concentrations used (between 0.1 and 0.2 M) are comparatively high. However, both works revealed only data for the 1:1 complex UO2(AcO)+. Therefore, the aim of this study was to provide spectroscopic data for all three UO2(AcO)n2-n complexes to fill in the gaps in the uranium-acetate system. Both the TRLFS and UV-vis experiments were performed with solutions containing only 50 µM uranium (VI). The parameters of the test series were varied in a way that the speciation changes from the free uranyl-ion, over the 1:1 and 1:2, to the 1:3 complex UO2(AcO)3-. Thereby the pH was fixed and the acetate concentration was varied or vice versa. In contrast to conventional UV-vis set ups, we used a Liquid Waveguide Capillary Cell with a path length of 250 cm to obtain spectra of high quality at this very low uranium concentration. The absorption spectra were evaluated on the basis of factor analysis and it was possible to calculate reproducible single component spectra for all the uranyl-acetate species (see Fig. 1). Organic polymers (e.g. cellulose, PVC, bitumen) present in low and intermediate level wastes (LILW) are exposed to ionizing radiation, alkaline pH, and organic degrading microorganisms. This may lead to the formation of smaller, water soluble organic compounds, affecting amongst others the chemical behavior and mobility of radionuclides (RN). In the worst case complexation will lead to an increased mobility and a decreased retention of RN in the barriers of a nuclear waste disposal. Therefore, the characterization of RN complexes with degradation products is necessary for the assessment of the safety and the long-term performance of a nuclear waste repository. The presentation will give an overview of our planned activities in cooperation with our partners (UNIMAN, SCK·CEN, and UGR) to contribute to task 1.2 of WP 1. Here our focus and expertise lies on the application of different modern spectroscopic tools to directly characterize the speciation of Rn/Ln with organic degradation products and/or with selected microbes, to underpin the findings from UNIMAN and SCK·CEN. Current results obtained by TRLFS, cryo-TRLFS, and UV-vis giving new insights into the uranyl-acetate system will be presented.

In the quest to reconstruct the Tree of Life, researchers have increasingly turned to phylogenomics, the inference of phylogenetic relationships using genome-scale data (Box 1). Mesmerized by the sustained increase in sequencing throughput, many phylogeneticists entertained the hope that the incongruence frequently observed in studies using single or a few genes [1] would come to an end with the generation of large multigene datasets. Yet, as so often happens, reality has turned out to be far more complex, as three recent large-scale analyses, one published in PLoS Biology [2–4], make clear. The studies, which deal with the early diversification of animals, produced highly incongruent (Box 2) findings despite the use of considerable sequence data (see Figure 1). Clearly, merely adding more sequences is not enough to resolve the inconsistencies. Here, taking these three studies as a case in point, we discuss pitfalls that the simple addition of sequences cannot avoid, and show how the observed incongruence can be largely overcome and how improved bioinformatics methods can help reveal the full potential of phylogenomics.

METHODS: Two-round postal Delphi questionnaire in the setting of general practice in Belgium, France, Germany, The Netherlands, Switzerland and the United Kingdom. Six national expert panels, each consisting of 10 members, primarily primary care practitioners and experts in the field of quality in primary care participated in the study. The main outcome measures were: (a) a European framework with indicators for the organization of primary care; and (b) ratings of the face validity of the usefulness of the indicators by expert panels in six countries. RESULTS: Agreement was reached about a definition of practice management across five domains (infrastructure, staff, information, finance, and quality and safety), and a common set of indicators for the organization of general practice. The panellist response rate was 95%. Sixty-two indicators (37%) were rated face valid by all six panels. Examples include out of hours service, accessibility, the content of doctors' bags and staff involvement in quality improvement. No indicators were rated invalid by all six panels. CONCLUSIONS: It proved to be possible to develop a European set of indicators for assessing the quality of practice management, despite the differences in health care systems and cultures in the six different countries. These indicators will now be used in a quality assessment procedure of practice management in nine European countries. While organizational indicators are part of the new GMS contract in the UK, this research shows that many practice management issues within primary care are also of relevance in other European countries.

METHODS: Two-round postal Delphi questionnaire in the setting of general practice in Belgium, France, Germany, The Netherlands, Switzerland and the United Kingdom. Six national expert panels, each consisting of 10 members, primarily primary care practitioners and experts in the field of quality in primary care participated in the study. The main outcome measures were: (a) a European framework with indicators for the organization of primary care; and (b) ratings of the face validity of the usefulness of the indicators by expert panels in six countries. RESULTS: Agreement was reached about a definition of practice management across five domains (infrastructure, staff, information, finance, and quality and safety), and a common set of indicators for the organization of general practice. The panellist response rate was 95%. Sixty-two indicators (37%) were rated face valid by all six panels. Examples include out of hours service, accessibility, the content of doctors' bags and staff involvement in quality improvement. No indicators were rated invalid by all six panels. CONCLUSIONS: It proved to be possible to develop a European set of indicators for assessing the quality of practice management, despite the differences in health care systems and cultures in the six different countries. These indicators will now be used in a quality assessment procedure of practice management in nine European countries. While organizational indicators are part of the new GMS contract in the UK, this research shows that many practice management issues within primary care are also of relevance in other European countries.

Genomic or cDNA clones for the glycolytic enzyme enolase were isolated from the amitochondriate pelobiont MASTIGAMOEBA: balamuthi, from the kinetoplastid TRYPANOSOMA: brucei, and from the euglenid EUGLENA: gracilis. Clones for the cytosolic enzyme were found in all three organisms, whereas EUGLENA: was found to also express mRNA for a second isoenzyme that possesses a putative N-terminal plastid-targeting peptide and is probably targeted to the chloroplast. Database searching revealed that ARABIDOPSIS: also possesses a second enolase gene that encodes an N-terminal extension and is likely targeted to the chloroplast. A phylogeny of enolase amino acid sequences from 6 archaebacteria, 24 eubacteria, and 32 eukaryotes showed that the MASTIGAMOEBA: enolase tended to branch with its homologs from TRYPANOSOMA: and from the amitochondriate protist Entamoeba histolytica. The compartment-specific isoenzymes in EUGLENA: arose through a gene duplication independent of that which gave rise to the compartment-specific isoenzymes in Arabidopsis, as evidenced by the finding that the EUGLENA: enolases are more similar to the homolog from the eubacterium Treponema pallidum than they are to homologs from any other organism sampled. In marked contrast to all other glycolytic enzymes studied to date, enolases from all eukaryotes surveyed here (except EUGLENA:) are not markedly more similar to eubacterial than to archaebacterial homologs. An intriguing indel shared by enolase from eukaryotes, from the archaebacterium Methanococcus jannaschii, and from the eubacterium Campylobacter jejuni maps to the surface of the three-dimensional structure of the enzyme and appears to have occurred at the same position in parallel in independent lineages.
