This work explores the significance of sedimentary proxies recorded within the Paleogene fine-grained detritus of the Adriatic foredeep (Northern Italy). Such sequences represent the only sedimentary record describing the source-to-sink system delivering detritus from the growing Alpine belt to the basin during the Eocene – Oligocene boundary. The combination of x-ray powder diffractometric, petrographic, SEM-EDS and biostratigraphic analyses on more than twenty samples from the Ternate-Travedona Formation, the Chiasso Formation, and the Villa Olmo Conglomerate allowed to identify the source areas of detritus and potential fluvial paleo-drainages, possibly ascribable to two different sectors of the Southalpine domain. Comparison between these results and the petrographic composition of the Cretaceous wedge in the same area opens to the possibility that Cenozoic drainages were inherited from more limited paleo-drainages developed during the end of the Mesozoic. Results of provenance analysis also gives new insights on how detrital compositions may vary in function of grain-size in tropical-subtropical environments.

Bruton’s tyrosine kinase (BTK) is a non-receptor intracellular kinase playing a key role in the proliferation and survival of normal and malignant B-lymphocytes. Its targeting by Ibrutinib, the first specific inhibitor, represented a turning point for the therapy of certain types of B-cell leukemias/lymphomas and several more BTK inhibitors are today in the clinic or advanced clinical trials. BTK expression was successively found to occur also outside of the hematopoietic compartment. In fact, we identified p65BTK, a novel 65 kDa isoform lacking an N-term stretch of 86 amino acids (compared to the 77 kDa protein expressed in B cells) as highly expressed in colon cancer patients. We demonstrated that p65BTK is a powerful oncogene acting downstream of the RAS/MAPK pathway and necessary for RAS-mediated transformation. Notably, the kinase domain is conserved and therefore inhibited by the available BTK-targeting drugs (Ibrutinib, Spebrutinib, etc.) which we used to demonstrate that p65BTK is an actionable target in drug-resistant colorectal carcinomas. We found p65BTK expressed also in >50% non-small cell lung cancers (NSCLC) and demonstrated that it is an actionable target in KRAS-mutated/EGFR-wild type drug-resistant NSCLC models (for which no targeted therapy is available). We also reported a significant correlation between p65BTK expression and low-grade tumors and overall survival of patients with grade III gliomas and showed that its targeting induced a significant decrease in the viability of in glioma stem cells. Finally, in ovarian cancer patients, p65BTK expression levels correlate with early relapse and shorter progression-free survival, both indicators of resistance to therapy. Remarkably, Ibrutinib is more effective than standard of care (SOC) therapeutics in in vitro and ex vivo settings. BACKGROUND: Posttraumatic stress disorder (PTSD) is associated with markers of accelerated aging. Estimates of brain age, compared to chronological age, may clarify the effects of PTSD on the brain and may inform treatment approaches targeting the neurobiology of aging in the context of PTSD. METHOD: Adult subjects (N = 2229; 56.2% male) aged 18-69 years (mean = 35.6, SD = 11.0) from 21 ENIGMA-PGC PTSD sites underwent T1-weighted brain structural magnetic resonance imaging, and PTSD assessment (PTSD+, n = 884). Previously trained voxel-wise (brainageR) and region-of-interest (BARACUS and PHOTON) machine learning pipelines were compared in a subset of control subjects (n = 386). Linear mixed effects models were conducted in the full sample (those with and without PTSD) to examine the effect of PTSD on brain predicted age difference (brain PAD; brain age - chronological age) controlling for chronological age, sex, and scan site. RESULTS: BrainageR most accurately predicted brain age in a subset (n = 386) of controls (brainageR: ICC = 0.71, R = 0.72, MAE = 5.68; PHOTON: ICC = 0.61, R = 0.62, MAE = 6.37; BARACUS: ICC = 0.47, R = 0.64, MAE = 8.80). Using brainageR, a three-way interaction revealed that young males with PTSD exhibited higher brain PAD relative to male controls in young and old age groups; old males with PTSD exhibited lower brain PAD compared to male controls of all ages. DISCUSSION: Differential impact of PTSD on brain PAD in younger versus older males may indicate a critical window when PTSD impacts brain aging, followed by age-related brain changes that are consonant with individuals without PTSD. Future longitudinal research is warranted to understand how PTSD impacts brain aging across the lifespan.

Colorectal cancer (CRC) is the fourth cause of death from cancer worldwide mainly due to the high incidence of drug-resistance. During a screen for new actionable targets in drug-resistant tumours we recently identified p65BTK – a novel oncogenic isoform of Bruton's tyrosine kinase. Studying three different cohorts of patients here we show that p65BTK expression correlates with histotype and cancer progression. Using drug-resistant TP53-null colon cancer cells as a model we demonstrated that p65BTK silencing or chemical inhibition overcame the 5-fluorouracil resistance of CRC cell lines and patient-derived organoids and significantly reduced the growth of xenografted tumours. Mechanistically, we show that blocking p65BTK in drug-resistant cells abolished a 5-FU-elicited TGFB1 protective response and triggered E2F-dependent apoptosis. Taken together, our data demonstrated that targeting p65BTK restores the apoptotic response to chemotherapy of drug-resistant CRCs and gives a proof-of-concept for suggesting the use of BTK inhibitors in combination with 5-FU as a novel therapeutic approach in CRC patients. © 2019 Pathological Society of Great Britain and Ireland. Background: Lung cancer is still the main cause of cancer death worldwide despite the availability of targeted therapies and immune-checkpoint inhibitors combined with chemotherapy. Cancer cell heterogeneity and primary or acquired resistance mechanisms cause the elusive behaviour of this cancer and new biomarkers and active drugs are urgently needed to overcome these limitations. p65BTK, a novel isoform of the Bruton Tyrosine Kinase may represent a new actionable target in non-small cell lung cancer (NSCLC). Methods: p65BTK expression was evaluated by immunohistochemistry in 382 NSCLC patients with complete clinico-pathological records including smoking habit, ALK and EGFR status, and in metastatic lymph nodes of 30 NSCLC patients. NSCLC cell lines mutated for p53 and/or a component of the RAS/MAPK pathway and primary lung cancer-derived cells from Kras/Trp53 null mice were used as a preclinical model. The effects of p65BTK inhibition by BTK Tyrosine Kinase Inhibitors (TKIs) (Ibrutinib, AVL-292, RN486) and first-generation EGFR-TKIs (Gefitinib, Erlotinib) on cell viability were evaluated by MTT. The effects of BTK-TKIs on cell growth and clonogenicity were assessed by crystal violet and colony assays, respectively. Cell toxicity assays were performed to study the effect of the combination of non-toxic concentrations of BTK-TKIs with EGFR-TKIs and standard-of-care (SOC) chemotherapy (Cisplatin, Gemcitabine, Pemetrexed). Results: p65BTK was significantly over-expressed in EGFR-wild type (wt) adenocarcinomas (AdC) from non-smoker patients and its expression was also preserved at the metastatic site. p65BTK was also over-expressed in cell lines mutated for KRAS or for a component of the RAS/MAPK pathway and in tumors from Kras/Trp53 null mice. BTK-TKIs were more effective than EGFR-TKIs in decreasing cancer cell viability and significantly impaired cell proliferation and clonogenicity. Triple Negative Breast Cancer (TNBC) is an aggressive neoplasia with median Overall Survival (OS) less than two years. Despite the availability of new drugs, the chance of survival of these patients did not increase. The combination of low doses of drugs in a metronomic schedule showed efficacy in clinical trials, exhibiting an anti-proliferative and anti-tumour activity. In Victor-2 study we recently evaluated a new metronomic combination (mCHT) of Capecitabine (CAPE) and Vinorelbine (VNR) in breast cancer patients showing a disease control rate with a median Progression- Free Survival (PFS) of 4.7 months in 28 TNBC patients. Here in Victor-0 study, we examined the effect of mCHT vs standard (STD) schedule of administration of different combinations of 5-Fluorouracil (5FU), the active metabolite of CAPE, and VNR in TNBC cell lines MDA-MB-231 and BT-549. A significant antiproliferative activity was observed in cells treated with metronomic vs STD administration of 5FU or VNR alone. Combination of the two drugs showed an additive inhibitor effect on cell growth in both cell lines . Moreover, after exposure of cells to 5FU and VNR under mCHT or conventional schedule of administration we also observed a downregulation of chemoresistance factor Bcl-2, changes in pro-apoptotic protein Bax and in cleaved effector caspase-3 and increased expression of LC3A/B autophagy protein. Our results therefore suggest that molecular mechanisms implicated in apoptosis and autophagy as well as the cross-talk between these two forms of cell death in MDA-MB-231 and BT-549 cells treated with 5FU and VNR is dose- and scheduledependent and provide some insights about the roles of autophagy and senescence in 5FU/VNR-induced cell death

Objective: In this study we analysed the relationship between mother and child using the Neuropsychomotor Video Analysis of parent and child interaction (NVA) (Moioli et al., 2010). This instrument was created in order to analyse and decode videotaped sequences of mother-child interactions in the first three years after birth. Two populations with atypical development were considered, with a first group consisting of children with Down syndrome and the second group of children born preterm. Video feedback meetings were arranged only for the second group and the effect of this (additional) approach was verified with a second videotape recording. Methodology: The sample included 29 children younger than 30 months divided into two subgroups: 10 children with the Down syndrome (5 females=50% and 5 males=50%; average age=16.40 months, sd=9.00) and 19 premature children (G.A.< 36 weeks) (10 females=53% and 9 males=47%; average age=11.53 months, sd=6.88) and a control group of 22 children of the same age (4 males=18% and 18 females=82%). Using the ten-minute tape-recording we filled in the report card: the program was computerized thus allowing the data processing and the visualization of the bend correlating the behaviour of the child with the mothers’. Socio-demographic data were collected with the Hollingshead’s Four Factor Index (1975). Results: The NVA allowed us to highlight specific relational modalities in both the analysed groups: in case of prematurity the bends usually showed a mirror trend, with a score decrease in participatory/sensitive categories associated with higher concentration of children’s scores in removal categories and mothers’ scores in approach categories. In the Down syndrome group we found lower scores in participatory/sensitive categories; the development retardation emerges from the score increase in the removal categories, with a maternal response balanced between children’s and mothers’ behaviour. We recently identified a role for GSK3B in drug resistance by demonstrating that its inhibition enables necroptosis in response to chemotherapy in p53-null drug-resistant colon carcinoma cells. We report here that, similarly to GSK3B, also GSK3A silencing/inhibition does not affect cell proliferation or cell cycle but only abolishes growth after treatment with DNA-damaging chemotherapy. In particular, blocking GSK3A impairs DNA repair upon exposure to DNA-damaging drugs. As a consequence, p53-null cells overcome their inability to undergo apoptosis and mount a necroptotic response, characterized by absence of caspase activation and RIP1-independent, PARP-dependent AIF nuclear re-localization. We therefore conclude that GSK3A is redundant with GSK3B in regulating drug-resistance and chemotherapy-induced necroptosis and suggest that inhibition of only one isoform, or rather partial inhibition of overall cellular GSK3 activity, is enough to re-sensitize drug-resistant cells to chemotherapy.

The Gulf of Sirte is a largely unexplored area in the southernmost Mediterranean Sea.We are presenting here 2008 summer data on the distribution pattern of living coccolithophores, a main phytoplankton calcifying group, in 105 samples from 20 stations. The survey includes coastal and offshore stations, enabling us to provide indications on the dynamics of phytoplankton productivity in relation to oceanographic processes. The total coccosphere concentrations show higher values in the offshore stations and lower ones for coastal sites. Umbellosphaera tenuis, Emiliania huxleyi, Florisphaera profunda, Syracosphaera pulchra HOL (Calyptrosphaera oblonga) and Rhabdosphaera spp. dominate the assemblages. The coccolithophore community shows the typical vertical zonation, with K-strategist taxa (among others U. tenuis, Rhabdosphaera spp. and Discosphaera tubifera) in the upper 75 meters depth and Lower Photic Zone taxa, dominated by F. profunda, below. The latter species shows its maximum development in the Deep Chlorophyll Maximum layer, which occurs in the upper part of the Levantine Intermediate Water, where the nutrient content is higher than in the overlying layers. The mesoscale oceanographic circulation significantly affects the spatial and vertical distribution of coccolithophores, with the thermocline and halocline depth shaping the vertical zonation of coccolithophore taxa and resulting in a strong lateral gradient within the gulf: in the eastern sector, under the influence of the Gulf of Sirte anticyclone, the DCM is deeper and so is the occurrence of K-strategist taxa and F. profunda
