Abstract

The first half of this thesis begins from what for me are the four most significant innovations of twentieth century music: systematic composition methods, electronic and digital technology, free improvisation, and awareness of the historical, geographical and social dimensions of music. Possibilities for radicalising these areas are discussed, in the course of which several concepts central to my musical thinking, such as "radically idiomatic instrumentalism" and "seeded improvisation" are introduced. The second half focuses on the works submitted in the composition portfolio, both as practical explorations of the ideas discussed in the first half and as the environment in which those ideas originate and continue to evolve. These are world-line for electric lap steel guitar with trumpet, percussion and electronics, close-up for electroacoustic sextet, urlicht for percussion trio, eiszeiten for brass trio and electronics, and wake for three instrumental trios and electronics, all of which address from different directions the central issues of this research: the application to notated composition of concepts emerging from free improvisation, including highly systematic approaches, and an attempt to fuse the aforementioned innovations into a unified and coherent creative practice, in order to widen the way to future possibilities, and perhaps not only musical ones.

Abstract

This thesis is divided into three parts. Part One introduces the context of the debate. Chapter One looks at relevant secondary literature relating to inter war Lancashire and four main themes are discussed, Mosley and his road to the BUF, the cotton industry in the inter war years, politics in Lancashire and finally anti Semitism and the development of the Jewish communities of Manchester and Liverpool. The second chapter presents themes linking the BUF and Lancashire, most notably cotton and Fascist plans to reinvigorate that industry. Linked to this is the BUF accusation that Jewish controlled international finance was largely responsible for the travails of the cotton industry - the question of Jewish control of international finance and the consequences of this being a constant one throughout the first two parts of the thesis. The changing attitude of the BUF to parliamentarism is also examined. Part Two begins with a case study of Manchester which notes the inclusive and diverse nature of anti BUF activism in the city and the nature of Jewish opposition to the Movement. Left characterisation of the National Government, another recurrent theme is highlighted, as are the unproductive electoral forays late in the period under review. Chapter Four examines the nature of responses to the BUF in Liverpool and the impact of a highly sectarian local politics on available political space is examined. This theme of electoral space is another constant throughout the thesis. The last local case study, of the cotton towns and in particular Nelson, notes a diversity of response to the BUF. Nelson is seen as having a radical civic culture and political allegiances characterised by youthful optimism, these factors combining to produce a confident response to the BUF. The following chapter examines the response of the Anglo Jewish elite to the threat of the BUF. Jewish opposition to the BUF was highly diverse. The response of the elite was couched in modest and traditional terms, being less concerned with Fascism or anti Semitism than with the often violent confrontational activism of parts of the Jewish community. The question as to whether elite responses meant a tacit acceptance ofBUF slanders is also discussed. The role of Neville Laski as an important conduit between Manchester Jewry, Anglo Jewry at the national level and the state is considered as is the wide intelligence gathering capacity of the Anglo Jewish elite, to the benefit of both themselves and the state. The following chapter considers the role of the state in monitoring BUF/anti Fascist activism. The links between the state at the local and national level are highlighted as is the question of the circumscription of civil liberties, notably in Manchester. The response to Cable Street in terms of the Public Order Act of 1937 is questioned as is the alleged development of a conservative authoritarian state. Chapters eight and nine offer explanations for the political stability of Britain in the inter war years, this being contrasted with regime failure in Germany and Italy. Reasons for the success of the Nazis in Germany and PNF in Italy are contrasted with the failure of the BUF in Britain, the question of political space being seen to have important explanatory relevance. The final chapter focusses on Britain and the importance of the maintenance of two party competition. The role of the Conservative Party is examined as is the National Government and the enduring strength of the political settlement post 1931. The moderation of the Labour Party and its relative weakness post 1931 is considered as is the weakness of the CPGB in comparison with its German counterpart. Factors linked to political development are highlighted as having useful explanatory worth, these themes include the contrasting abilities of the ruling elites in Germany and Britain and the legitimacy of the moderate British left.

5. Marine fouling processes upon stainless steel and elastomeric surfaces

Abstract

Marine fouling development upon stainless steel and elastomeric surfaces was studied by light and electron microscopy and the observed contributions of the various organisms involved is discussed. As an extension of the above, the bacteriology of a mature fouling film from stainless steel was examined. Characterized isolates were classified by numerical taxonomy. Species of Vibrio dominated the isolatable aerobic heterotrophic bacterial community. A number of previously undescribed species were discovered, highlighting inadequacies of current identification schemes. Isolate characteristics are discussed in relation to conditions during fouling development. Immuno- location of species within TEM sections of fouling was attempted without success. Macromolecular dissolved organic materials were isolated from seawater by ultrafiltration, and characterization showed these materials to be humic substances, predominantly fulvic acid. Concentrated solutions of these materials were used to organically pretreat stainless steel surfaces prior to marine immersion, and the fouling of these and clean surfaces was compared. It was discovered that preadsorbed material can influence both the in vivo attachment and growth of microorganisms and subsequent fouling succession. Natural molecular fouling was also studied in relation to bacterial attachment. In vitro assessment of the attachment of a marine Vibrio Sp. to pretreated and clean stainless steel after prior growth in the presence of different concentrations of different macromolecular species revealed a relationship between growth regime and adhesive tendancy. The effect of humic materials on the growth of selected bacteria was investigated in vitro. Some isolates grew with these materials acting as sole sources of carbon, nitrogen and energy. Bacterial growth on acetate was stimulated by humic materials at specific concentrations, although inhibitory effects were apparent if concentrations of the latter were increased further. The growth physiology of marine bacteria in the. presence of humic substances and the possible role of humic substances in fouling community development is discussed.

6. Acoustic waves and electron wakes in a positive column plasma

Abstract

Part A. Measurements with a plane Langmuir probe near an obstacle in a positive column with a longitudinal magnetic field reveal periodic spatial variations in the number of high energy electrons reaching the probe, and in the local plasma parameters (density, potential, electron temperature). The variations are due to the helical motion of electrons along the wake after partial selection of the phase of their motion by the obstacle. The distribution of high energy electrons in the region downstream from the obstacle is similar to that calculated by Al'pert et al for the ions behind a satellite moving through the ionosphere, parallel to a magnetic field. There is strong evidence that the spatial variations of plasma parameters, as seen in the frame of reference moving with the average velocity of the anode-directed electrons, constitute an electron cyclotron wave. Part B. Externally excited waves of small amplitude in low pressure mercury, argon, neon and hydrogen positive columns have been observed at frequencies mainly below 100 kc/s. A weak longitudinal magnetic field is usually employed to facilitate propagation.' Phase sensitive detection is used to distinguish the weak wave signal from the strong background of noise. The experimental dispersion curves of frequency against wave number fall into two groups, those having positive slope (forward waves) and those having negative slope (backward waves).

Abstract

Do some patients benefit from an unrelated donor (URD) transplant because of a stronger graft-versus-leukemia (GVL) effect? We analyzed 4099 patients with acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), and chronic myeloid leukemia (CML) undergoing a myeloablative allogeneic hematopoietic cell transplantation (HCT) from an URD (8/8 human leukocyte antigen [HLA]matched, n = 941) or HLA-identical sibling donor (n = 3158) between 1995 and 2004 reported to the CIBMTR. In the Cox regression model, acute and chronic GVHD were added as time-dependent variables. In multivariate analysis, URD transplant recipients had a higher risk for transplantation-related mortality (TRM; relative risk [RR], 2.76; P < .001) and relapse (RR, 1.50; P < .002) in patients with AML, but not ALL or CML. Chronic GVHD was associated with a lower relapse risk in all diagnoses. Leukemia-free survival (LFS) was decreased in patients with AML without acute GVHD receiving a URD transplant (RR, 2.02; P < .001) but was comparable to those receiving HLA-identical sibling transplants in patients with ALL and CML. In patients without GVHD, multivariate analysis showed similar risk of relapse but decreased LFS for URD transplants for all 3 diagnoses. In conclusion, risk of relapse was the same (ALL, CML) or worse (AML) in URD transplant recipients compared with HLA-identical sibling transplant recipients, suggesting a similar GVL effect. Abstract

To study the early stages of development from stem cells of the CD56+ cell population [which includes natural killer (NK) cells], granulocyte-colony stimulating factor-mobilized peripheral blood CD34+ cells from healthy donors were sorted to >99% purity and cultured in the presence of stem cell factor and interleukin (IL)-2. After 3 weeks in culture, the majority of cells acquired CD33, with or without human leukocyte antigen-DR and CD14. In 20 stem cell donors tested, 8.7 +/- 8.8% of cells were CD56+. Two major CD56+ subsets were identified: CD56(bright), mainly CD33- cells (7+/-10%, n=11) with large, granular lymphocyte morphology, and CD56dim, mainly CD33+ (2.5+/-2, n=11) cells with macrophage morphology. The CD56bright population had cytoplasmic granzyme A but lacked killer inhibitory receptor, suggesting they were immature NK cells. The CD56dim, CD33+, population lacked NK markers. They may represent a minor subset of normal monocytes at a developmental stage comparable with the rare CD56+ CD33+ hybrid myeloid/NK cell leukemia. Consistent with a monocyte nature, CD56dimCD33+ proliferated and produced a variety of cytokines upon lipopolysaccharide stimulation, including IL-8, IL-6, monocyte chemoattractant protein-1, and macrophage-derived chemokine but not interferon-gamma. In a short-term cytotoxicity assay, they failed to kill but powerfully inhibited the proliferation of the NK-resistant cell line P815. Abstract

Clinical observations suggest that in chronic myelogenous leukemia (CML), the Philadelphia chromosome (Ph+) clone has a growth advantage over normal hematopoiesis. Patients with CML have high levels of neutrophil elastase, which has recently been shown to antagonize the action of granulocyte-colony-stimulating factor (G-CSF) and other growth factors. We therefore compared the effect of elastase on the growth of normal and CML progenitor cells. In 10-day suspension cultures of normal or CML CD34+ cells supplemented with G-CSF, stem cell factor (SCF), and granulocyte macrophage-colony-stimulating factor (GM-CSF), CML cells had diminished sensitivity to the growth inhibitory effect of elastase. When equal numbers of CML and normal CD34+ cells were cocultured for 10 days, there was no change in the relative proportions of normal and leukemic cells (measured by fluorescence in situ hybridization [FISH] or flow cytometry). However, when elastase was added, CML cells predominated at the end of the culture period (78% vs 22% with 1 microg/mL and 80% vs 20% with 5 microg/mL elastase). CML neutrophils substituted effectively for elastase in suppressing the proliferation of normal CD34+ cells, but this effect was abrogated by serine protease inhibitors. Abstract

There is evidence that neutrophil production is a balance between the proliferative action of granulocyte-colony-stimulating factor (G-CSF) and a negative feedback from mature neutrophils (the chalone). Two neutrophil serine proteases have been implicated in granulopoietic regulation: pro-proteinase 3 inhibits granulocyte macrophage-colony-forming unit (CFU-GM) growth, and elastase mutations cause cyclic and congenital neutropenia. We further studied the action of the neutrophil serine proteases (proteinase 3, elastase, azurocidin, and cathepsin G) on granulopoiesis in vitro. Elastase inhibited CFU-GM in methylcellulose culture. In serum-free suspension cultures of CD34+ cells, elastase completely abrogated the proliferation induced by G-CSF but not that of GM-CSF or stem cell factor (SCF). The blocking effect of elastase was prevented by inhibition of its enzymatic activity with phenylmethylsulfonyl fluoride (PMSF) or heat treatment. When exposed to enzymatically active elastase, G-CSF, but not GM-CSF or SCF, was rapidly cleaved and rendered inactive. 16. Specific depletion of alloreactivity against haplotype mismatched related individuals by a recombinant immunotoxin: a new approach to graft-versus-host disease prophylaxis in haploidentical bone marrow transplantation.

Abstract

Haploidentical bone marrow transplantation (BMT) is associated with a high risk of severe graft-versus-host disease (GVHD). While pan-T cell depletion of the graft is the most effective means of preventing severe GVHD, it is associated with delayed recovery of T cell function leading to fatal infections. We used two related Pseudomonas exotoxin-based immunotoxins, anti-Tac(Fv)-PE38 and anti-Tac(Fv)-PE38KDEL, that both target the IL-2 receptor on activated T cells, to specifically deplete alloreactive lymphocytes against haploidentical stimulators. The functional capacity of the remaining lymphocytes was tested in proliferative assays against the original haploidentical stimulator and pooled cells from other mismatched donors (third party). We varied the recombinant toxin concentration and schedule to determine the optimum conditions for selective depletion. In 10 experiments, the mean residual reactivity after depletion was 7.6 +/- 1.4% against the haploidentical stimulator and 64.2 +/- 5% against the third party, expressed as a percentage of the undepleted response to the same stimulators. Depletion was shown to be specific for mixed lymphocyte culture (MLC)-activated lymphocytes. The immunotoxin did not affect CFU-GM growth of normal BM cells. Abstract

Major histocompatibility complex (MHC) class I antigens on tumour cell surfaces have been shown to modulate target susceptibility to natural killer (NK) cell-mediated lysis in some, although not all, systems investigated. MHC class II expression may also affect NK cell function, but the mechanism by which MHC class II antigen regulates NK cell activity has not been fully examined. In this study we induced HLA-DR1 expression by gene transfection into the classic NK-sensitive K562 cell line to study the interaction of NK cells with MHC class II molecules and the effect of brefeldin-A (BFA), an endogenous antigen-processing pathway blocker, on NK-target cell interaction. We demonstrated that the expression of HLA-DR1 on the cell surface reduced K562 cell susceptibility to NK lysis by peripheral blood monuclear cells and a NK cell line. The effect was demonstrable in prolonged (8 hr) cytotoxicity assays and was blocked by pretreatment of target cells with anti-HLA-DR antibody. Treatment of K562 DR transfectant with BFA abrogated the resistance of K562 transfectant to NK-mediated cytolysis. 18. Helper and cytotoxic lymphocyte responses to chronic myeloid leukaemia: implications for adoptive immunotherapy with T cells.

Abstract

We studied 19 patients with chronic myeloid leukaemia (CML) to characterize T-cell autologous responses to leukaemia. Third party stimulated alloresponses in mixed lymphocyte reactions were normal in all patients. Using the helper T-lymphocyte precursor frequency (HTLPf) assay we demonstrated a low frequency of T helper cells recognizing autologous leukaemia cells from CML blood (1/850 000) and marrow (1/965 000). However, similar frequencies to autologous bone marrow and lymphoid cells were also found in normal individuals. In 11 patients studied, HLA-matched siblings had a higher HTLPf to leukaemia than the patient's autologous response (P < 0.004). Alloresponse in mixed lymphocyte reactions (MLR), and autologous HTLPf to leukaemia, were comparable at all stages of disease progression and time from diagnosis, and independent of treatment given. In order to generate autologous cytotoxic lymphocyte responses to CML, lymphocytes were stimulated with CML cells. Cultures were fed again with CML cells and examined for cytotoxicity after 21 d. Strong lymphokine-activated killer (LAK) cytotoxicity was found against K562 and Daudi cell lines, and to Epstein-Barr virus-transformed allogeneic and autologous lymphoblastoid cell lines. Autologous leukaemia cells were lysed to a lesser extent in only 3/13 patients tested. The findings indicate that immune reactivity in CML is normal but suggest that CML cells are relatively resistant to lysis by cytotoxic T cells. 19. Hyperfine Magnetic Fields at ¹⁸¹Ta Impurity Nuclei in Ferromagnetic ZrZn₂

Abstract

The hyperfine magnetic field at ¹⁸¹Tm nuclei substituting at the zirconium sites in ferromagnetic ZrZn₂ was measured by the method of time-differential perturbed angular correlations (TDPAC). The value obtained was -17.3 ± 0.3 k0e with 2% hafnium impurity at the zirconium sites. The hyperfine field was also measured as a function of hafnium and titanium concentration (substituting at zirconium sites) and compared with magnetization measurements on the same samples. The magnetization data agreed qualitatively with measurements by Ogawa: the magnetization decreases to zero at about 15% hafnium concentration, but increases to a maximum at about 20% titanium concentration. The hyperfine field, however, decreases less rapidly with hafnium concentration but drops off fairly suddenly near 20% hafnium.
