Depolarizing sodium (Na+) leak currents carried by the NALCN channel regulate the resting membrane potential of many neurons to modulate respiration, circadian rhythm, locomotion and pain sensitivity1–8. NALCN requires FAM155A, UNC79 and UNC80 to function, but the role of these auxiliary subunits is not understood3,7,9–12. NALCN, UNC79 and UNC80 are essential in rodents2,9,13, and mutations in human NALCN and UNC80 cause severe developmental and neurological disease14,15. Since fundamental aspects about the composition, assembly, and gating of the NALCN channelosome remain obscure, we determined the structure of this ~1 megadalton complex. UNC79 and UNC80 are massive HEAT-repeat proteins that form an intertwined anti-parallel superhelical assembly which docks intracellularly onto the NALCN-FAM155A pore-forming subcomplex. Calmodulin copurifies bound to the carboxy-terminal domain of NALCN, identifying this region as a putative modulatory hub. Single channel analyses uncovered a low open probability for the wild-type complex, highlighting the tightly closed S6-gate in the structure, and providing a basis to interpret the altered gating properties of disease-causing variants. Key constraints between the UNC79-UNC80 subcomplex and the NALCN DI-DII and DII-DIII linkers are identified that lead to a model of channelosome gating. Our results provide a structural blueprint to understand NALCN channelosome physiology and a template for drug discovery to modulate the resting membrane potential.

While anabolic hormone deficit is a common finding in heart failure with reduced ejection fraction (HFrEF), few data are available in heart failure with preserved ejection fraction (HFpEF). Methods. Blood samples were collected for metabolic (total cholesterol, HDL cholesterol, LDL cholesterol, creatinine, and glucose) and hormonal (IGF-1, DHEA-S, TSH, fT3, fT4, and T) determination, comparing 30 patients with HFpEF and 20 patients with HFrEF. Total antioxidant capacity was evaluated by using the spectrophotometric method using the latency time in the appearance of the radical species of a chromogen (LAG, sec) as a parameter proportional to antioxidant content of the sample. Echocardiographic parameters were also assessed in the two groups. Results. A high prevalence of testosterone (32% in HFrEF and 72% in HFpEF, ) and DHEA-S deficiencies was observed in HFpEF patients. Echocardiographic parameters did not correlate with hormone values. A significant direct correlation between T (r2 = 0.25, ) and DHEA-S (r2 = 0.19, ) with LAG was observed only in HFpEF. Conclusion. Anabolic hormone deficiency is clearly shown in HFpEF, as already known in HFrEF. Although longitudinal studies are required to confirm the prognostic value of this observation, our data suggest different mechanisms in modulating antioxidants in the two conditions, with possible therapeutic implications.

Cloud computing systems handle large volumes of data by using almost unlimited computational resources, while spatial data warehouses (SDWs) are multidimensional databases that store huge volumes of both spatial data and conventional data. Cloud computing environments have been considered adequate to host voluminous databases, process analytical workloads and deliver database as a service, while spatial online analytical processing (spatial OLAP) queries issued over SDWs are intrinsically analytical. However, hosting a SDW in the cloud and processing spatial OLAP queries over such database impose novel obstacles. In this article, we introduce novel concepts as cloud SDW and spatial OLAP as a service, and afterwards detail the design of novel schemas for cloud SDW and spatial OLAP query processing over cloud SDW. Furthermore, we evaluate the performance to process spatial OLAP queries in cloud SDWs using our own query processor aided by a cloud spatial index. Moreover, we describe the cloud spatial bitmap index to improve the performance to process spatial OLAP queries in cloud SDWs, and assess it through an experimental evaluation. When high resolution is achievable, crystal structure docking and de-novo modeling allows for precise assignment of individual protein domain sequences. However, the achievable resolution may limit the ability to do so, especially when small or flexible complexes are under study. In such cases, protein labeling has emerged as an important complementary tool to characterize domain architecture and elucidate functional mechanistic details. All labeling strategies proposed to date are either focused on the identification of the position of protein termini or require multi-step labeling strategies, potentially interfering with the final labeling efficiency. Here we describe a strategy for determining the position of internal protein domains within EM maps using a recombinant one-step labeling approach named Efficient Mapping by Internal Labeling (EMIL). EMIL takes advantage of the close spatial proximity of the GFP's N- and C-termini to generate protein chimeras containing an internal GFP at desired locations along the main protein chain. We apply this method to characterize the subunit domain localization of the human Polycomb Repressive Complex 2.

The lack of an appropriate conceptual model for data warehouses and OLAP systems has led to the tendency to deploy logical models (for example, star, snowflake, and constellation schemas) for them as conceptual models. ER model extensions, UML extensions, special graphical user interfaces, and dashboards have been proposed as conceptual approaches. However, they introduce their own problems, are somehow complex and difficult to understand, and are not always user-friendly. They also require a high learning curve, and most of them address only structural design, not considering associated operations. Therefore, they are not really an improvement and, in the end, only represent a reflection of the logical model. The essential drawback of offering this system-centric view as a user concept is that knowledge workers are confronted with the full and overwhelming complexity of these systems as well as complicated and user-unfriendly query languages such as SQL OLAP and MDX. In this article, the authors propose a user-centric conceptual model for data warehouses and OLAP systems, called the Cube Algebra. It takes the cube metaphor literally and provides the knowledge worker with high-level cube objects and related concepts. A novel query language leverages well known high-level operations such as roll-up, drill-down, slice, and drill-across. Polycomb Repressive Complex 2 (PRC2) is essential for gene silencing, establishing transcriptional repression of specific genes by tri-methylating Lysine 27 of histone H3, a process mediated by cofactors such as AEBP2. In spite of its biological importance, little is known about PRC2 architecture and subunit organization. Here, we present the first three-dimensional electron microscopy structure of the human PRC2 complex bound to its cofactor AEBP2. Using a novel internal protein tagging-method, in combination with isotopic chemical cross-linking and mass spectrometry, we have localized all the PRC2 subunits and their functional domains and generated a detailed map of interactions. The position and stabilization effect of AEBP2 suggests an allosteric role of this cofactor in regulating gene silencing. Varicella-zoster virus (VZV), of the family Alphaherpesvirinae, causes varicella in children and young adults, potentially leading to herpes zoster later in life on reactivation from latency. The conserved herpesvirus glycoprotein gB and the heterodimer gHgL mediate virion envelope fusion with cell membranes during virus entry. Naturally occurring neutralizing antibodies against herpesviruses target these entry proteins. To determine the molecular basis for VZV neutralization, crystal structures of gHgL were determined in complex with fragments of antigen binding (Fabs) from two human monoclonal antibodies, IgG-94 and IgG-RC, isolated from seropositive subjects. These structures reveal that the antibodies target the same site, composed of residues from both gH and gL, distinct from two other neutralizing epitopes identified by negative-stain electron microscopy and mutational analysis. Inhibition of gB/gHgL-mediated membrane fusion and structural comparisons with herpesvirus homologs suggest that the IgG-RC/94 epitope is in proximity to the site on VZV gHgL that activates gB. Immunization studies proved that the anti-gHgL IgG-RC/94 epitope is a critical target for antibodies that neutralize VZV. Thus, the gHgL/Fab structures delineate a site of herpesvirus vulnerability targeted by natural immunity.

Spatial data warehouses (SDW) and spatial online analytical processing (SOLAP) enhance decision making by enabling spatial analysis combined with multidimensional analytical queries. A SDW is an integrated and voluminous multidimensional database containing both conventional and spatial data. SOLAP allows querying SDWs with multidimensional queries that select spatial data that satisfy a given topological relationship and that aggregate spatial data. Existing SDW and SOLAP applications mostly consider phenomena represented by spatial data having exact locations and sharp boundaries. They neglect the fact that spatial data may be affected by imperfections, such as spatial vagueness, which prevents distinguishing an object from its neighborhood. A vague spatial object does not have a precisely defined boundary and/or interior. Thus, it may have a broad boundary and a blurred interior, and is composed of parts that certainly belong to it and parts that possibly belong to it. Although several real-world phenomena are characterized by spatial vagueness, no approach in the literature addresses both spatial vagueness and the design of SDWs nor provides multidimensional analysis over vague spatial data. These shortcomings motivated the elaboration of this doctoral thesis, which addresses both vague spatial data warehouses (vague SDWs) and vague spatial online analytical processing (vague SOLAP). A vague SDW is a SDW that comprises vague spatial data, while vague SOLAP allows querying vague SDWs. The major contributions of this doctoral thesis are: (i) the Vague Spatial Cube (VSCube) conceptual model, which enables the creation of conceptual schemata for vague SDWs using data cubes; (ii) the Vague Spatial MultiDim (VSMultiDim) conceptual model, which enables the creation of conceptual schemata for vague SDWs using diagrams; (iii) guidelines for designing relational schemata and integrity constraints for vague SDWs, and for extending the SQL language to enable vague SOLAP; (i, Les entrepôts de données spatiales (EDS) et l'analyse en ligne spatiale (ALS) améliorent la prise de décision en permettant l'analyse spatiale combinée avec des requêtes analytiques multidimensionnelles. Ces lacunes ont motivé l'élaboration de cette thèse de doctorat, qui adresse à la fois les entrepôts de données spatiales vagues (EDS vagues) et l'analyse en ligne spatiale vague (ALS vague). Un EDS vague est un EDS qui comprend des données spatiales vagues, tandis que l'ALS vague permet d'interroger des EDS vagues. Contudo, tais aplicações negligenciam dados espaciais afetados por imperfeições, tais como a vagueza espacial, a qual interfere na identificação precisa de um objeto e de seus vizinhos. Um objeto espacial vago não tem sua fronteira ou seu interior precisamente definidos. Além disso, é composto por partes que certamente pertencem a ele e partes que possivelmente pertencem a ele. Apesar de inúmeros fenômenos do mundo real serem caracterizados pela vagueza espacial, na literatura consultada não se identificaram trabalhos que considerassem a vagueza espacial no projeto de DWE e nem para consultar o DWE. Tal limitação motivou a elaboração desta tese de doutorado, a qual introduz os conceitos de DWE vago e de SOLAP vago. Um DWE vago é um DWE que armazena dados espaciais vagos, enquanto que SOLAP vago provê os meios para consultar o DWE vago. This paper focuses on macroeconomic interdependencies between the Euro area and three transition economies (Estonia, Lithuania and Latvia), with the aim of establishing whether the latter are ready to adopt the Euro. The theoretical framework is based on the Generalised Purchasing Power Parity (GPPP) hypothesis, which is empirically tested within a Vector Error Correction (VEC) model. Using both monthly and quarterly data over the period 1993-2005, it is found that GPPP holds for the real exchange rate vis-à-vis the Euro of each Baltic country, reflecting a degree of real convergence consistent with Optimum Currency Area criteria. Further, the adopted joint modelling approach for the real exchange rates of the Baltic region outperforms a number of alternative models in terms of out-of-sample forecasts.

Polychlorotrifluoroethylene (CTFE) and CTFE copolymeric films are being used in the electronic packaging industry as insulating dielectric layers between microwave circuits. Since these films are semicrystalline and, in this application, are being used as hot melt adhesives, the cooling rate is an important processing variable, affecting the crystallinity of the CTFE films which in turn affect many properties including dielectric characteristics and mechanical properties. In this study, the crystallinity of CTFE and CTFE copolymeric films as a function of cooling rate was characterized by WAXS and FTIR. As expected, the degree of crystallinity decreased as the cooling rate increased. Analysis of mechanical properties as a function of cooling rate by tensile testing showed that the mechanical behavior of the films became more ductile with faster COOling rates. Since the cooling rate has also been shown to significantly influence adhesion in previous studies, the effect of cooling rate on the bond strength between CTFE and a glass substrate was analyzed.
