1. Epigenetic properties of reproduction and ageing in Hymenoptera

Abstract: Epigenetic mechanisms expand the genome's regulatory flexibility. Whilst these mechanisms are conserved across eukaryotes, insects display high variation in DNA methylation profiles making them an ideal study to infer divergent function and evolution of these mechanisms. This thesis explores the role of DNA methylation and histone modifications in two species of Hymenoptera; the eusocial bumblebee Bombus terrestris and the solitary wasp Nasonia vitripennis. Previous work has demonstrated an association between epigenetic mechanisms and the reproductive plasticity found within social insects. I find, in contrast with previous studies, that chemical alteration to the methylome in B. terrestris workers does not induce reproductive morphology and that discrepancies between studies on the role of DNA methylation is due to greater inter-colony variation in DNA methylation than between castes. I also conduct the first investigation into histone modifications between reproductive castes in B. terrestris and find, like DNA methylation, they vary greatly between colonies. I show through both B. terrestris and N. vitripennis that perturbations to the epigenome via chemical modifiers can be inconsistent and highlight the need to more targeted disruption to untangle the function of these epigenetic markers. Given the confounding effect of the genetic background of colonies, I highlight the use of solitary species such as N. vitripennis to determine the functional role of these mechanisms in insects. Finally, due to the association between DNA methylation and ageing in mammals, I investigate DNA methylation and transcriptomic responses in N. vitripennis with ageing. I find hallmarks of ageing in both the methylome and transcriptome as well as a large sex-bias. Overall, this thesis enhances our understanding of epigenetics in Hymenoptera and sheds light on the need for functional validation of epigenetic modifications to further elucidate their role in regulating gene expression and how this impacts plastic phenotypes such as reproduction and ageing.

Abstract: Perennial plants create productive and biodiverse hotspots, known as fertile islands, beneath their canopies. These hotspots largely determine the structure and functioning of drylands worldwide. Despite their ubiquity, the factors controlling fertile islands under conditions of contrasting grazing by livestock, the most prevalent land use in drylands, remain virtually unknown. Here we evaluated the relative importance of grazing pressure and herbivore type, climate and plant functional traits on 24 soil physical and chemical attributes that represent proxies of key ecosystem services related to decomposition, soil fertility, and soil and water conservation. To do this, we conducted a standardized global survey of 288 plots at 88 sites in 25 countries worldwide. We show that aridity and plant traits are the major factors associated with the magnitude of plant effects on fertile islands in grazed drylands worldwide. Grazing pressure had little influence on the capacity of plants to support fertile islands. Taller and wider shrubs and grasses supported stronger island effects. Stable and functional soils tended to be linked to species-rich sites with taller plants. Together, our findings dispel the notion that grazing pressure or herbivore type are linked to the formation or intensification of fertile islands in drylands. Rather, our study suggests that changes in aridity, and processes that alter island identity and therefore plant traits, will have marked effects on how perennial plants support and maintain the functioning of drylands in a more arid and grazed world.

Abstract: This dataset details the growth of Gammarus pulex at 11°C and 15°C when exposed to imidacloprid concentrations of 0, 0.01, 0.1, and 0.3 µg/L for periods of 3 months and 6 months. The organisms were housed individually in 100 mL bottles with an adequate supply of food, and the medium was refreshed monthly. In the 6-month experiment, the ages of the test organisms were known, and their initial size was approximately 2 mm. Five specimens were sampled destructively each month to record body size, head width, and dry weight. For the 3-month experiment, organisms with an initial size of about 4 mm were used, and growth was recorded every month using non-destructive methods. At the end of the 3-month experiment, the sex of the organisms was also determined.

12. Preparing uniform supported Pd-Ni catalysts with citrate-assisted impregnation

Abstract: Bimetallic supported catalysts can show synergistic effects compared to their monometallic constituents. However, it is challenging to produce bimetallic catalysts with a uniform nanoparticle distribution over the support, while ensuring each nanoparticle contains both metals. A promising strategy is to use impregnation precursors that facilitate uniform distribution of both metals over a support. In this work, citrate-based precursors were studied to prepare bimetallic Pd-Ni nanoparticles supported on SBA-15 mesoporous silica. Amongst others, cryo-electron microscopy demonstrated the excellent distribution of Ni citrate precursor after drying. Co-impregnation with Ni citrate precursor and Pd(OAc)2 or Pd(NH3)4(NO3)2 produced well-distributed Pd-Ni nanoparticles with a narrow particle size distribution. Extensive characterization with STEM-EDX, EXAFS and TPR showed that the type of Pd precursor controlled the Pd to Ni nanoscale intimacy. CO2 hydrogenation experiments demonstrated that increasing Pd-Ni intimacy decreased the activity. Our strategy to produce uniform Pd-Ni nanoparticles using citric acid is relevant also for other bimetallic systems.

Abstract: Population models provide insights into population dynamics under diverse and untested chemical exposure scenarios, supporting their environmental risk assessment (ERA). In this study, we investigate the interplay of temperature and imidacloprid exposure on population dynamics using an Individual-Based Model (IBM) incorporating a dynamic energy budget (DEB) model for population dynamics and toxicokinetic-toxicodynamic models of the General Unified Threshold model for Survival (GUTS) framework to predict toxicity effects. For this, we tested different model configurations, where i) only the DEB parameters are corrected for temperature, as is common practice, and ii) also the TKTD parameters of the GUTS model are corrected for temperature. In doing so, we aim to evaluate the importance of temperature corrections in the GUTS model within an IBM framework. As expected, increased temperature amplitudes increase the range of simulated population sizes, and chemical exposure reduces the maximum population size. The combined effect of correcting both the DEB and TKTD parameters, however, yield an overall strongly negative effect on population sizes, particularly at lower temperatures. These results highlight the necessity of temperature-sensitive parameterization in population models for a protective risk assessment under the projected future climate conditions with increased temperatures and variability. Future considerations include incorporating local adaptations and acclimatization, particularly in different climate zones, to accurately interpret population model outcomes in the context of evolving environmental conditions. Such insights contribute to the refinement of ecological realism in ERA, enhancing the robustness of chemical risk management strategies.

Abstract: Background: The bluetongue virus serotype 3 (BTV-3) outbreak in the Netherlands in 2023 caused severe clinical signs in ruminants. The clinical and pathological signs in ruminants and their spread during the outbreak in 2023 are described. Methods: Data from the Dutch monitoring and surveillance system were available to describe clinical signs and pathological findings related to BTV-3 in sheep, cattle and goats. During the outbreak, 13 farms (five sheep, five cattle and three dairy goats) were closely monitored. Results: In 2023, BTV-3 infections were confirmed by real-time polymerase chain reaction in sheep flocks (n = 1807), cattle herds (n = 1864), goat herds (n = 62), alpaca and/or llama herds (n = 15) and one dog. Sheep exhibited the most severe clinical signs and had the highest mortality. In other animal species, a large variation in both occurrence and severity of clinical signs was observed. Limitation: Only 13 farms were closely monitored. Conclusions: The clinical signs observed in affected animals during the 2023 BTV-3 outbreak seem to be more severe than those observed during the BTV-8 outbreak between 2006 and 2008. It seems likely that BTV-3 will overwinter, similar to BTV-8. Therefore, the availability of an effective and safe vaccine is crucial to limit the future impact of BTV-3.

Abstract: Pyrethroid insecticides are widely detected in aquatic ecosystems due to their extensive use in agriculture and horticulture, which could pose a potential risk to aquatic non-target organisms. While previous ecotoxicological studies have been conducted mainly with standard tests and local species under temperate conditions, scarce information is available on the effects of pyrethroid insecticides on communities and ecosystems under (sub-)tropical conditions. A single application of lambda-cyhalothrin at concentrations of 0, 9, 30, and 100 ng/L was evaluated in outdoor mesocosms under sub-tropical conditions. Lambda-cyhalothrin was found to dissipate rapidly in the water column, with only 11 % and 7 % of the remaining dose measured at 1 and 3 days after application, respectively. Lambda-cyhalothrin concentrations disappeared considerably faster from the water compartment compared to temperate conditions. Consistent decreases in abundance were observed for Lecane lunaris at the medium and higher treatments (NOEC = 9 ng/L) and at the highest treatment (NOEC = 30 ng/L) for Keratella tropica. On the contrary, two taxa belonging to Cladocera (i.e., Ceriodaphnia sp. and Diaphanosoma sp.) showed the most prominent increase in abundance related to the lambda-cyhalothrin treatments. At the community level, a consistent no observed effect concentrations (NOECs) of 9 ng/L could be calculated for the zooplankton community. A marginal significant overall treatment related effect was observed for the macroinvertebrate community. The results of species sensitivity distribution (SSD) analysis based on results of acute toxicity experiments conducted alongside the mesocosm experiment and obtained from the literature indicated that macroinvertebrates from temperate regions may be generally more sensitive than their counterparts in (sub-)tropical regions. Abstract: Purpose To compare patients with and without a history of mental illness on process and outcome measures in relation to prehospital and emergency surgical care for patients with perforated ulcer. Methods A nationwide registry-based cohort study of patients undergoing emergency surgery for perforated ulcer. We used data from the Danish Prehospital Database 2016–2017 and the Danish Emergency Surgery Registry 2004–2018 combined with data from other Danish databases. Patients were categorized according to severity of mental health history. Results We identified 4.767 patients undergoing emergency surgery for perforated ulcer. Among patients calling the EMS with no history of mental illness, 51% were identified with abdominal pain when calling the EMS compared to 31% and 25% among patients with a history of moderate and major mental illness, respectively. Median time from hospital arrival to surgery was 6.0 h (IQR: 3.6;10.7). Adjusting for age, sex and comorbidity, patients with a history of major mental illness underwent surgery 46 min (95% CI: 4;88) later compared to patients with no history of mental illness. Median number of days-alive-and-out-of-hospital at 90-day follow-up was 67 days (IQR: 0;83). Adjusting for age, sex and comorbidity, patients with a history of major mental illness had 9 days (95% CI: 4;14) less alive and out-of-hospital at 90-day follow-up. Conclusion One-third of the population had a history of mental illness or vulnerability. Patients with a history of major mental illness were less likely to be identified with abdominal pain if calling the EMS prior to arrival. Abstract: Introduction: Using pre-procedure analgesia with the risk of apnoea may complicate the Less Invasive Surfactant Administration (LISA) procedure or reduce the effect of LISA. Methods: The NONA-LISA trial (ClinicalTrials.gov, NCT05609877) is a multicentre, blinded, randomised controlled trial aiming at including 324 infants born before 30 gestational weeks, meeting the criteria for surfactant treatment by LISA. Infants will be randomised to LISA after administration of fentanyl 0.5–1 mcg/kg intravenously (fentanyl group) or isotonic saline solution intravenously (saline group). All infants will receive standardised non-pharmacological comfort care before and during the LISA procedure. Additional analgesics will be provided at the clinician’s discretion. The primary outcome is the need for invasive ventilation, meaning mechanical or manual ventilation via an endotracheal tube, for at least 30 min (cumulated) within 24 h of the procedure. Secondary outcomes include the modified COMFORTneo score during the procedure, bronchopulmonary dysplasia at 36 weeks, and mortality at 36 weeks. Discussion: The NONA-LISA trial has the potential to provide evidence for a standardised approach to relief from discomfort in preterm infants during LISA and to reduce invasive ventilation. The results may affect future clinical practice. Impact: Pre-procedure analgesia is associated with apnoea and may complicate procedures that rely on regular spontaneous breathing, such as Less Invasive Surfactant Administration (LISA).This randomised controlled trial addresses the effect of analgesic premedication in LISA by comparing fentanyl with a placebo (isotonic saline) in infants undergoing the LISA procedure. Abstract: Background Atrial fibrillation (AF) is the most common sustained arrhythmia and results in a high risk of stroke. The number of immigrants is increasing globally, but little is known about potential differences in AF care across migrant populations. Aim To investigate if initiation of oral anticoagulation therapy (OAC) differs for patients with incident AF in relation to country of origin. Methods A nationwide register-based study covering 1999–2017. AF was defined as a first-time diagnosis of AF and a high risk of stroke. Stroke risk was defined according to guidelines from the European Society of Cardiology (ESC). Poisson regression adjusted for sex, age, socioeconomic position and comorbidity was made to compute incidence rate ratios (IRR) for initiation of OAC. Results The AF population included 254 586 individuals of Danish origin, 6673 of Western origin and 3757 of non-Western origin. Overall, OAC was initiated within −30/+90 days relative to the AF diagnosis in 50.3% of individuals of Danish origin initiated OAC, 49.6% of Western origin and 44.5% of non-Western origin. Immigrants from non-Western countries had significantly lower adjusted IRR of initiating OAC according to all ESC guidelines compared with patients of Danish origin. The adjusted IRRs ranged from 0.73 (95% CI: 0.66 to 0.80) following the launch of the 2010 ESC guideline to 0.89 (95% CI: 0.82 to 0.97) following the launch of the 2001 ESC guideline. Conclusion Patients with AF with a high risk of stroke of non-Western origin have persistently experienced a lower chance of initiating OAC compared with patients of Danish origin during the last decades.

Abstract: Group B Streptococcus (GBS) disease in neonates occurs in two forms: early-onset disease (EOD), (day 0-6), and late-onset disease (LOD), (day 7-90). This review investigates that risk-based intrapartum screening and antibiotics have reduced the incidence of EOD, but not LOD, in Denmark. No clinical or laboratory tests can rule out GBS disease at symptom onset. Thus, a high proportion of uninfected infants receive antibiotics, although this varies widely, and may be reduced by strategies of antibiotic stewardship. A future GBS vaccine for pregnant women may potentially reduce disease burden and antibiotic exposure.

Abstract: Introduction: The majority of patients with mycosis fungoides (MF) have an indolent disease course, but a substantial fraction (20-30%) of patients progress to advanced stages – usually with a grave prognosis. Early differentiation between indolent and aggressive types of MF is important for the choice of treatment regimen and monitoring of the individual patient. Good biomarkers are therefore desired. Methods: Here, we used spatial transcriptomics on skin samples at time-of-diagnosis to enable prediction of patients who later progressed to advanced stages of MF. Formalin-fixed, paraffin-embedded skin biopsies at time of diagnosis from six patients with MF who progressed to advanced stages of disease within 4 months to 12 years after diagnosis, and nine patients who remained in early-stage disease over 9 to 27 years were analyzed using the GeoMx Digital Spatial Profiler to capture spatially resolved high-plex RNA gene expression data. Five different regions of interest (the epidermis, the basal layer of epidermis, CD4+ T-cells and neighboring cells, and Pautrier’s microabscesses) were profiled for further assessment. Results and discussion: Interestingly, RUNX2, SHMT2, and MCM7 were upregulated in the enriched population of malignant T-cells in Pautrier’s microabscesses in patients who later developed advanced stages of disease. Expression of RUNX2, SHMT2 and MCM7 in malignant T-cells was confirmed in a subset of patients in MF skin using scRNA-seq datasets across multiple studies and correlating with stage of disease. Taken together, we provide first evidence that RUNX2 has potential as a biomarker to identify MF patients progressing to advanced stage disease. As RUNX2 has not previously been linked to MF, our data also shows the analytical strength of combining spatial transcriptomics with scRNA-seq analysis.

Abstract: Objective. The apparent diffusion coefficient (ADC) extracted from diffusion-weighted magnetic resonance imaging (DWI) is a potential biomarker in radiotherapy (RT). DWI is often implemented with an echo-planar imaging (EPI) read-out due to speed, but unfortunately low geometric accuracy follows. This study aimed to investigate the influence of geometric distortions on the ADCs extracted from the gross tumor volume (GTV) and on the shape of the GTV in abdominal EPI-DWI. Approach. Twenty-one patients had EPI-DWI scans on a 1.5 T MRI sim before treatment and on a 1.5 T MRI-Linac at one of the first treatment fractions. Off-resonance correction with and without eddy current correction were applied to ADC maps. The clinical GTVs were deformed based on the same (but inverted) corrections to assess the local-regional geometric influence of distortions. Mean surface distance (MSD), Hausdorff distance (HD), and Dice similarity coefficient (DSC) were calculated to compare the original and distorted GTVs, and ADC values were calculated based on a mono-exponential model. Phantom measurements were performed to validate the applied correction method. Main results. The median (range) ADC change within the GTV after full distortion correction was 1.3% (0.02%-6.9%) for MRI-Sim and 1.5% (0.1%-6.4%) for MRI-Linac. The additional effect of the eddy current correction was small in both systems. The median (range) MSD, HD, and DSC comparing the original and off-resonance distorted GTVs for all patients were 0.43 mm (0.11-0.94 mm), 4.00 mm (1.00-7.81 mm) and 0.93 (0.82-0.99), respectively. Significance. Overall effect of distortion correction was small in terms of derived ADC values, indicating that distortion correction is unimportant for prediction of outcomes based on ADC. Abstract: Increases in the abundance of woody species have been reported to affect the provisioning of ecosystem services in drylands worldwide. However, it is virtually unknown how multiple biotic and abiotic drivers, such as climate, grazing, and fire, interact to determine woody dominance across global drylands. We conducted a standardized field survey in 304 plots across 25 countries to assess how climatic features, soil properties, grazing, and fire affect woody dominance in dryland rangelands. Precipitation, temperature, and grazing were key determinants of tree and shrub dominance. The effects of grazing were determined not solely by grazing pressure but also by the dominant livestock species. Interactions between soil, climate, and grazing and differences in responses to these factors between trees and shrubs were key to understanding changes in woody dominance. Our findings suggest that projected changes in climate and grazing pressure may increase woody dominance in drylands, altering their structure and functioning.

Abstract: Earth harbours an extraordinary plant phenotypic diversity that is at risk from ongoing global changes. However, it remains unknown how increasing aridity and livestock grazing pressure—two major drivers of global change—shape the trait covariation that underlies plant phenotypic diversity. Here we assessed how covariation among 20 chemical and morphological traits responds to aridity and grazing pressure within global drylands. Our analysis involved 133,769 trait measurements spanning 1,347 observations of 301 perennial plant species surveyed across 326 plots from 6 continents. Crossing an aridity threshold of approximately 0.7 (close to the transition between semi-arid and arid zones) led to an unexpected 88% increase in trait diversity. This threshold appeared in the presence of grazers, and moved toward lower aridity levels with increasing grazing pressure. Moreover, 57% of observed trait diversity occurred only in the most arid and grazed drylands, highlighting the phenotypic uniqueness of these extreme environments. Our work indicates that drylands act as a global reservoir of plant phenotypic diversity and challenge the pervasive view that harsh environmental conditions reduce plant trait diversity. They also highlight that many alternative strategies may enable plants to cope with increases in environmental stress induced by climate change and land-use intensification.

Abstract: Background Increasing severity of chronic obstructive pulmonary disease (COPD) is associated with increasing risk of poor outcomes. Using health registry data, we aimed to assess the association between treatment intensity levels (TIL), as a proxy for underlying COPD severity, and long-term outcomes. Methods Using Danish nationwide registries, we identified patients diagnosed with COPD during 2001–2016, who were alive at index date of 1 January 2017. We stratified patients into exclusive TILs from least to most severe: no use, short term therapy, mono-, dual-, triple therapy, oral corticosteroid (OCS), and long-term oxygen treatment (LTOT). Survival analyses were used to assess 5-year outcomes by TIL. Results We identified 53,803 patients with COPD in the study period (median age: 72 years [inter quartile range, 64–80], 48 % male). The three most severe TILs were associated with a significant incremental increase in all-cause mortality with an adjusted hazard ratio (aHR) for triple therapy, OCS and LTOT of 1.44 (95 % CI: 1.38–1.51), 1.67 (95 % CI: 1.59–1.75), and 2.91 (95 % CI: 2.76–3.07) compared with those receiving no therapy as reference. The same pattern was evident for the composite outcome of 5-year mortality or COPD-related hospitalization with an aHR for triple therapy, OCS and LTOT of 2.30 (95 % CI: 2.22–2.38), 2.85 (95 % CI: 2.74–2.96), and 4.00 (95 % CI: 3.81–4.20), respectively. Abstract: Surfactant deficiency remains a significant cause of mortality and morbidity in preterm infants.1 The Less Invasive Surfactant Administration (LISA) procedure involves laryngoscopy and intratracheal surfactant administration via a thin catheter during spontaneous breathing supported by nasal CPAP or other types of non-invasive support or ventilation.2 In a recent study including 153 international LISA experts from 14 countries, 41% indicated no use of analgesic premedication for LISA, while 49% reported using fentanyl.3 On the contrary, non-pharmacological comfort care was used extensively for LISA but with no standardized approach.

40. Understanding how indoor environmental classroom conditions influence academic performance in higher education

Abstract: Purpose – This study aims to qualitatively examine the relationship between the indoor environmental quality (IEQ), lecturers’ and students’ perceived internal responses and academic performance. Design/methodology/approach – To capture user experiences with the IEQ in classrooms, semistructured interviews with 11 lecturers and three focus group discussions with 24 students were conducted, transcribed, coded and analyzed using direct content analysis. Findings – The findings show that lecturers and students experience poor thermal, lighting, acoustic and indoor air quality (IAQ) conditions that may influence their ability to teach and learn. Maintaining acceptable thermal and IAQ conditions was difficult for lecturers, as opening windows or doors caused noisedisturbances. In uncomfortable conditions, lecturers may decide to give a break earlier or shorten a lecture. When students experienced discomfort, it may affect their ability to concentrate, their emotional status and their quality of learning. Research limitations/implications – The findings originate from a relatively small sample, which might have limited the number and variety of identified associations between environment and users. Practical implications – Maintaining acceptable air and thermal conditions will mitigate the need to open windows and doors. Keeping doors and windows closed will prevent noise disturbances and related distractions. This will support the quality of learning in classrooms. This study reveals the end users’ perspectives and preferences, which can inspire designers of new school buildings in higher education. Originality/value – This study emphasizes the importance of creating and maintaining optimal IEQ conditions to support the quality of teaching and learning. These conditions are particularly relevant when classroom occupancy rates are high or outdoor conditions are unfavourable.

Abstract: Cancer vaccines are a promising strategy to increase tumor-specific immune responses in patients who do not adequately respond to checkpoint inhibitors. Cancer vaccines that contain patient-specific tumor antigens are most effective but also necessitate the production of patient-specific vaccines. This study aims to develop a versatile cancer vaccine format in which patient-specific tumor antigens can be site-specifically conjugated by a proximity-based Sortase A (SrtA)-mediated ligation (PBSL) approach to antibodies that specifically bind to antigen-presenting cells to stimulate immune responses. DEC205 and CD169 are both receptors expressed on antigen-presenting cells that can be targeted to deliver antigens and stimulate T-cell responses. We used the CRISPR/HDR platform to produce mouse heavy chain IgG2a antibodies with DEC205 or CD169 specificity containing an SrtA recognition motif followed by a SpyTag at the C-terminus. Using a recombinant protein of SrtA linked to SpyCatcher, we applied proximity-based SrtA-mediated ligation to ligate fluorescein isothiocyanate (FITC)-labeled or antigenic peptides to the antibodies. Ligated antibodies bound to DEC205-expressing dendritic cells or CD169-expressing macrophages both in vitro and in vivo. More importantly, immunization with DEC205- or CD169-specific Abs linked to T-cell epitopes efficiently stimulated T-cell responses in vivo. To conclude, we have developed a cancer vaccine format using PBSL that enables the rapid incorporation of tumor antigens and could potentially be implemented for the synthesis of personalized cancer vaccines.

Abstract: De huidige binaire en essentialistische registratie van sekse staat ter discussie. Veel binaire en non-binaire trans en intersekse mensen, maar niet alleen zij, hebben bezwaar tegen de voorwaarden voor het wijzigen van het juridisch geslacht op hun geboorteakte, tegen het ontbreken van andere opties dan man of vrouw of tegen de registratiepraktijk als zodanig. Het wettelijk kader is in de loop der jaren langzaam opgerekt maar dat heeft, ondanks enkele aarzelende aanzetten in de vorm van een (ingetrokken) amendement, voorbereidende inventarisaties en een conceptwetsvoorstel, nog niet geleid tot een doorbreking van de binariteit. Via de rechter wordt steeds vaker een alternatieve vermelding van geslacht gevraagd én toegekend. Tegelijkertijd neemt de politieke steun voor versoepeling van het systeem af. Het risico van rechtsonzekerheid blijft voor rekening van de betrokkenen.

Abstract: EU legislation has undergone profound changes in recent years. Existing scholarship has only limitedly addressed such transformation, which can be characterized as the EU legislative power reaching maturity. This has fundamentally changed both institutionally (the EU Parliament acquired prominence), substantially (the EU from a market regulator became a political legislator), and procedurally (with the introduction of impact assessment and better regulation policies). This transformation added a new layer of complexity to a persistent feature of EU legislation: the tension between EU unity and national autonomy. The core question of this volume is: How are unity and diversity accommodated, and balanced, in this new and transformed law-making process in the EU? It explores which legislative strategies have been adopted across various fields of EU law and policy to shape unity and diversity and the practical, conceptual and normative issues these bring along. Abstract: We present new spectroscopic and photometric follow-up observations of the known sample of extreme coronal line-emitting galaxies (ECLEs) identified in the Sloan Digital Sky Survey (SDSS). With these new data, observations of the ECLE sample now span a period of two decades following their initial SDSS detections. We confirm the non-recurrence of the iron coronal line signatures in five of the seven objects, further supporting their identification as the transient light echoes of tidal disruption events (TDEs). Photometric observations of these objects in optical bands show little overall evolution. In contrast, mid-infrared (MIR) observations show ongoing long-term declines consistent with power-law decay. The remaining two objects had been classified as active galactic nuclei (AGNs) with unusually strong coronal lines rather than being TDE related, given the persistence of the coronal lines in earlier follow-up spectra. We confirm this classification, with our spectra continuing to show the presence of strong, unchanged coronal line features and AGN-like MIR colours and behaviour. We have constructed spectral templates of both subtypes of ECLE to aid in distinguishing the likely origin of newly discovered ECLEs. We highlight the need for higher cadence, and more rapid, follow-up observations of such objects to better constrain their properties and evolution. We also discuss the relationships between ECLEs, TDEs, and other identified transients having significant MIR variability.

Abstract: The early evolution of a supernova (SN) can reveal information about the environment and the progenitor star. When a star explodes in vacuum, the first photons to escape from its surface appear as a brief, hours-long shock-breakout flare1,2, followed by a cooling phase of emission. However, for stars exploding within a distribution of dense, optically thick circumstellar material (CSM), the first photons escape from the material beyond the stellar edge and the duration of the initial flare can extend to several days, during which the escaping emission indicates photospheric heating3. Early serendipitous observations2,4 that lacked ultraviolet (UV) data were unable to determine whether the early emission is heating or cooling and hence the nature of the early explosion event. Here we report UV spectra of the nearby SN 2023ixf in the galaxy Messier 101 (M101). Using the UV data as well as a comprehensive set of further multiwavelength observations, we temporally resolve the emergence of the explosion shock from a thick medium heated by the SN emission. We derive a reliable bolometric light curve that indicates that the shock breaks out from a dense layer with a radius substantially larger than typical supergiants.

Abstract: For nearly 40 years, MDMA has been used as a party drug in the Netherlands. Today, more than one in 10 people have used this prohibited substance at least once. Quite separate from this recreational use, MDMA also has a medical use, like ketamine or cannabis. This medical use of MDMA has been the subject of research for nearly half a century. In sum, MDMA is definitely not a novelty in Dutch society; it has been present here for decades. Why, then, should we have a State Commission on MDMA at this specific point in time?

Abstract: The Dutch term 'regie' (loosely translated as governance or steering) is often linked to the experienced need to control the cooperation in complex horizontal networks. However, it is also visible as a steering mechanism used in more regulated tasks, as well as decentralised tasks. The Participation Act, and in particular social assistance, has detailed regulations that were not considered by the central government to be competitive with the current ideas, which calls for a more tailored approach. The article explores the government's 'regie' efforts, and makes clear that the central government has used multiple steering mechanisms to adjust the decentralised task of implementing social assistance. However, it remains unclear whether the central government has 'regie', as this implies control over those who are being steered.
