Therapeutic benefit to immune checkpoint blockade (ICB) is currently limited to the subset of cancers thought to possess a sufficient tumor mutational burden (TMB) to allow for the spontaneous recognition of neoantigens (NeoAg) by autologous T cells. We explored whether the response to ICB of an aggressive low-TMB squamous cell tumor could be improved through combination immunotherapy using functionally defined NeoAg as targets for endogenous CD4+ and CD8+ T cells. We found that, whereas vaccination with CD4+ or CD8+ NeoAg alone did not offer prophylactic or therapeutic immunity, vaccines containing NeoAg recognized by both subsets overcame ICB resistance and led to the eradication of large established tumors that contained a subset of PD-L1+ tumor-initiating cancer stem cells (tCSC), provided the relevant epitopes were physically linked. Therapeutic CD4+/CD8+ T cell NeoAg vaccination produced a modified tumor microenvironment (TME) with increased numbers of NeoAg-specific CD8+ T cells existing in progenitor and intermediate exhausted states enabled by combination ICB-mediated intermolecular epitope spreading. We believe that the concepts explored herein should be exploited for the development of more potent personalized cancer vaccines that can expand the range of tumors treatable with ICB.

CD4+ T cells play key roles in a range of immune responses, either as direct effectors or through accessory cells, including CD8+ T lymphocytes. In cancer, neoantigen (NeoAg)-specific CD8+ T cells capable of direct tumor recognition have been extensively studied, whereas the role of NeoAg-specific CD4+ T cells is less well understood. We have characterized the murine CD4+ T cell response against a validated NeoAg (CLTCH129>Q) expressed by the MHC-II-deficient squamous cell carcinoma tumor model (SCC VII) at the level of single T cell receptor (TCR) clonotypes and in the setting of adoptive immunotherapy. We find that the natural CLTCH129>Q-specific repertoire is diverse and contains TCRs with distinct avidities as measured by tetramer-binding assays and CD4 dependence. Despite these differences, CD4+ T cells expressing high or moderate avidity TCRs undergo comparable in vivo proliferation to cross-presented antigen from growing tumors and drive similar levels of therapeutic immunity that is dependent on CD8+ T cells and CD40L signaling. Adoptive cellular therapy (ACT) with NeoAg-specific CD4+ T cells is most effective when TCR-engineered cells are differentiated ex vivo with IL-7 and IL-15 rather than IL-2 and this was associated with both increased expansion as well as the acquisition and stable maintenance of a T stem cell memory (TSCM)-like phenotype in tumor-draining lymph nodes (tdLNs). ACT with TSCM-like CD4+ T cells results in lower PD-1 expression by CD8+ T cells in the tumor microenvironment and an increased frequency of PD-1+CD8+ T cells in tdLNs. These findings illuminate the role of NeoAg-specific CD4+ T cells in mediating antitumor immunity via providing help to CD8+ T cells and highlight their therapeutic potential in ACT.

Purpose. The main purpose of the article is to develop a technology for water purification from residues of oil-fat containing drug (medicinal) cream, in particular technology and a technological scheme for purifying urban (municipal) wastewater from plastics and residues of drugs. Methodology. The studies were carried out based on the analysis of scientific sources and reporting data on the availability of drugs and plastics in the water of Ukraine, European countries, and the USA. Findings. The authors present the results of a comprehensive review of issues related to the determination of the availability of drugs in various waters, their concentrations and the most dangerous drugs-toxicants. Medicines and plastics can accumulate not only in humans and animals, but also in sea and river fish, etc. The presence of drugs and plastic in the body can have a negative impact on the health. The treatment facilities in Ukraine and the world are not sufficiently adapted for the detection of drugs and plastics and their purification. Originality. The authors of this material conducted studies to determine the content of plastic in drinking tap water in Dnipro and Zaporizhzhia cities. Drinking water from the tap contains less plastic particles than bottled water of the famous Ukrainian trade brands. The work developed technological schemes for the purification of industrial wastewater from pharmaceutical enterprises for the manufacture of creams, ointments, oils and other products for treatment and cosmetic purposes. Structures and methods for purifying water from these pollutants have been proposed, which had not been previously used at water purification stations, but successfully apply in other industries. Practical value. The use of environmentally efficient technologies in Ukraine will allow obtaining high-quality drinking water, but it will require re-equipment and reconstruction of water treatment plants. Основная цель статьи – разработать технологию очистки воды от остатков масложиросодержащих лекарственных кремов, в частности очистки городских (муниципальных) сточных вод от пластика и остатков лекарственных препаратов. Методика. Исследования выполнены на основании анализа научных источников и отчетных данных о наличии лекарств и пластика в воде Украины, европейских странах, США. Результаты. Авторы представляют результаты комплексного рассмотрения вопросов, связанных с определением наличия лекарств в различных водах, их концентраций и наиболее опасных лекарственных препаратов-токсикантов. Медикаменты и пластик могут накапливаться не только в организме людей и животных, но и в морской и речной рыбе и т. д. Присутствие в организме лекарств и пластика оказывает негативное влияние на здоровье. Очистные сооружения Украины и мира недостаточно приспособлены для обнаружения в воде лекарств и пластика, ее очистки. Научная новизна. Авторы этой статьи провели исследования по определению содержания пластика в питьевой водопроводной воде городов Днепра и Запорожья. В питьевой воде из-под крана содержится меньше частиц пластика, чем в бутилированной воде известных украинских торговых брендов. Разработаны технологические схемы по очистке сточных вод фармацевтических предприятий по изготовлению кремов, мазей, масел и другой продукции для лечения и косметических целей. Предложены сооружения и методы для очистки вод от указанных загрязнений, которые не использовали ранее на станциях очистки вод, но успешно применяют в других отраслях промышленности. Практическая значимость. Применение в Украине экологически эффективных технологий позволит получать высококачественную питьевую воду, однако потребует переоборудования и реконструкции водоочистных станций. Для этого нужно предусмотреть соответствующие статьи по основному финансированию. В настоящее время в Украине (даже в Киеве) нет станций по очистке питьевой и сточных вод, на которых бы использовались современные технологии и соор, Мета. Основна мета статті – розробити технологію очищення води від залишків масложировмісних лікарських кремів, зокрема очищення міських (муніципальних) стічних вод від пластику і залишків лікарських препаратів. Автори цієї статті провели дослідження з визначення вмісту пластику в питній водопровідній воді міст Дніпра й Запоріжжя. Питна вода з-під крана містить менше частинок пластику, ніж бутильована вода відомих українських торгових брендів. Розроблені технологічні схеми з очищення стічних вод фармацевтичних підприємств із виготовлення кремів, мазей, масел та іншої продукції для лікування й косметичних цілей. Запропоновано споруди й методи для очищення вод від зазначених забруднень, які не використовували раніше на станціях очистки вод, але успішно застосовують в інших галузях промисловості. Практична значимість. Застосування в Україні екологічно ефективних технологій дозволить отримувати високоякісну питну воду, проте вимагає переобладнання й реконструкції водоочисних станцій. Для цього потрібно передбачити відповідні статті за основним фінансування. У наш час в Україні (навіть у Києві) немає станцій з очищення питної і стічних вод, на яких би використовувались сучасні технології і споруди, але ж якісна вода – це здоров’я і життя людей.

Purpose. The main purpose of the article is to analyze the presence of plastic in the water of Ukraine and the world, as well as to suggest the methods for purifying water from plastic on the basis of the world and personal experience. Methodology. The studies were carried out on the basis of the analysis of scholarly domestic and foreign literature sources on the presence of plastic in the water of Ukraine, European countries, and the USA (1999–2018). Findings. The analyzed sources indicate that one of the main environmental threats of global scale is the presence of plastic in drinking and waste waters. The wastewater treatment facilities of the world are not sufficiently adapted to detect and trap plastic. The authors present the results of a comprehensive consideration of the issues related to the determination of the presence of plastic in various waters, their size and concentrations, and manufacturing materials. Plastic, entering the body of people, especially children, animals, sea fish, etc., has a negative influence on their health. Studies have been conducted to determine the content of plastic in drinking tap water in the cities of Dnipro and Zaporozhye. For example, drinking water from the tap contains from 4 to 10 particles of plastic per liter of water, the size of which exceeded 100 microns, that is, the average thickness of a human hair. Були проведені дослідження з визначення вмісту пластика в питній водопровідній воді міст Дніпра і Запоріжжя. Наприклад, у питній воді з-під крана міститься від 4 до 10 часток пластику на літр води, розмір яких перевищував 100 мікрон, тобто середню товщину людської волосини. А в бутильованій воді відомих торгових марок «Моршинська» та «Buvette» міститься від 49 до 102 часток пластика на літр води. Наукова новизна. У роботі узагальнені наявні й представлені нові методи й технології очищення води від пластику, такі як: проціджування на різних спорудах (механічні методи), флотаційні, мембранні й зворотноосмотичні системи, мембранні біореактори (фізико-хімічні й комбіновані методи). Як другий шлях зниження кількості пластику у воді запропоновано заборону його виготовлення або створення біорозкладаного пластику, здатного біологічно руйнуватися. Практична значимість. З огляду на поширеність споживання бутильованої води по всьому світу, результати дослідження підтверджують необхідність подальших досліджень впливу мікро- і нанопластів на здоров’я людини.

Proses pemesinan adalah sebuah proses produksi yang banyak digunakan di industri manufaktur terutama untuk komponen-komponen mesin yang membutuhkan ketelitian dan kepresisian geometrik (dimensi, bentuk dan kehalusan permukaan yang tinggi). Proses pemesinan terjadi karena adanya gerak relatif pahat dengan benda kerja. Pahat adalah suatu alat yang digunakan untuk memotong, salah satu pahat yang sering digunkan paht HSS. Semakin lama pahat digunakan dalam pemotongan maka pahat akan semakin aus. Tidak hanya waktu pemotongan saja yang mempengaruhi kausan pahat, tergantung dari pahat yang digunakan terhadap benda kerjanya. Maka untuk mengurangi keausan pada pahat dilakukan perlakukan kriogenik. Perlakuan kriogenik adalah proses dimana meningkatkan sifat mekanik dan fisik material. Benda kerja yang digunakan adalah komposit serat kelapa dimana komposit merpakan gabungan dua atau lebih material. Serat kelapa banyak terdapat dialam sehingga tidak sulit ditemukan, maka dari itu serat kelapa dijadikan penguat dalam komposit. Jadi penelitian mengetahui pengaruh perlakuan pahat HSS kriogenik dan tanpa kriogenik terhadap keausan pahat untuk pemotongan komposit serat kelapa. Pembuatan sampel dimulai dengan mendapatkan selulosa dari serat kelapa, setelah itu selulosa dihaluskan dan dilakukan pencampuran dengan Unsaturated Polyester Resin. Unuk pahat juga diberikan perlakuan krogenik dengan variasi lama waktu perendaman dengan nitrogen cair dan pemanasan. Kemudian dilakukan pengujian dengan cara mengatur parameter pada mesin freis lalu dilakukan pemotongan untuk melihat keausannya. Dari data yang didapatkan pahat HSS kriogenik variasi pertama memiliki nilai keaan terendah dari keempat pahat dan yang paling aus adalah pahat HSS tanpa kriogenik. Introduction: Hyperprolactinemia (HPRL) is known as a side effect of some antidepressants and antipsychotics. These medicines are common in treatment of schizophrenia. Thus, HPRL is often observed in schizophrenic patients. It is also known that HPRL can occur in Hashimoto' s thyroiditis due to prolactoliberin effect of thyroliberin. The clinical pathophysiology of the patients with the comorbodity of schizophrenia and Hashimoto's thyroiditis, receiving antipsychotics, is of special interest.It's fair to assume that these patients have higher risks of HPRL. To analyze risks of HPRL with antipsychotic treatment, to identify an association between the antipsychotic therapy (AT) and HPRL in Hashimoto's patiens receiving AT ,to explore the association of HPRL and other laboratory parameters in patients with Hashimoto's thyroiditis and schizophrenia during AT. Subjects and methods: We studied 17 patients with HT in comorbidity with schizophrenia receiving AT (mean age 46,5+-12,8 years), all euthyroid or with light hypothyroidism. Different laboratory parameters such as anti-thyroid peroxidase (anti-TPO) and anti-thyroglobulin (anti-TG) antibodies, blood levels of thyroid stimulating hormone (TSH), free thyroxine (FT4), free triiodothyronine (FT3) and prolactin (PRL) were analysed. Results: The study revealed the high levels of PRL, anti-TPO and anti-TG autoantibodies. Thus, patients were classified into 3 groups by the degree of expected HPRL risk from the antipsychotics used: without expected risk, with low and high expected risks. The correlation analysis detected an inverse significant correlation (R=-0.51; p=0.037) between expected level of drug-associated HPRL risk and actual PRL levels in studied group. At the same time, we detected a positive significant correlation between the levels of PRL and FT4 in the groups (R=0.53; p=0.03). Some pollutants concentration exceeding the maximum permissible concentration for discharges is observed. In order to eliminate this problem, some solutions are proposed. To increase wastewater treatment effectiveness at the plant, recirculation of wastewaters and replacement of current filling material of the tower biofilters with plastic plate blocks is proposed., Метою даної роботи є аналіз ефективності очистки стічних вод очисних споруд Хорольського заводу дитячих продуктів харчування та пошук шляхів підвищення ефективності очистки до нормативних вимог по максимально допустимій концентрації при скиді очищених стоків.З огляду на результати лабораторних дослідів по визначенню концентрації забруднювачів у зразках стоків після основних етапів очистки (освітлювачі-перегнивачі, баштові біофільтри, вторинні відстійники) та у випуску очищеної води, зроблено висновок про, зокрема, незадовільну роботу баштових біофільтрів по зниженню ХСК та БПК5 (причини – зменшення подачі стічних вод, незадовільний стан розподільчої системи, замулювання заповнювача біофільтра), вторинних відстійників по зниженню завислих речовин (причини – зменшення подачі стічних вод, незадовільний стан відстійників).З метою підвищення очистки стічних вод на очисних спорудах Хорольського заводу дитячих продуктів харчування, були запропоновані наступні кроки: 1) заміна існуючого завантаження біофільтрів (щебінь з розміром фракцій 40-60 мм) на площинне полімерне завантаження, що є менш замулюваним, створює кращу циркуляцію кисню, сприяє інтенсивнішому виносу відмерлого активного мулу; 2) улаштування системи рециркуляції води для підвищення гідравлічного навантаження на споруди, покращення роботи нижніх шарів біофільтра, підвищення ефективності освітлення води у освітлювачі-перегнивачі; 3) улаштування тонкошарових блоків у вторинних відстійниках для інтенсифікації процесу осадження., Целью данной работы является анализ эффективности очистки сточных вод очистных сооружений Хорольского завода детских продуктов питания и поиск путей повышения эффективности очистки до нормативных требований по максимально допустимой концентрации при сбросе очищенных стоков.С целью повышения очистки сточных вод на очистных сооружениях Хорольского завода детских продуктов питания, были предложены следующие шаги: 1) замена существующей загрузки биофильтров (щебень с размером фракций 40-60 мм) на плоскостную полимерную загрузку, являющуюся менее заиливающейся, создает лучшую циркуляцию кислорода, способствует интенсивному выносу отмершего активного ила; 2) устройство системы рециркуляции воды для повышения гидравлической нагрузки на сооружения, улучшения работы нижних слоев биофильтра, повышение эффективности осветления воды в осветителях-перегнивателях; 3) устройство тонкослойных блоков во вторичных отстойниках для интенсификации процесса осаждения.

Immigrants from regions with a high incidence of tuberculosis (TB) are a risk group for TB in low-incidence countries such as Switzerland. In a previous analysis of a nationwide collection of 520 Mycobacterium tuberculosis isolates from 2000 to 2008, we identified 35 clusters comprising 90 patients based on standard genotyping (24-locus mycobacterial interspersed repetitive-unit-variable-number tandem-repeat [MIRU-VNTR] typing and spoligotyping). Here, we used whole-genome sequencing (WGS) to revisit these transmission clusters. Genome-based transmission clusters were defined as isolate pairs separated by ≤12 single nucleotide polymorphisms (SNPs). WGS confirmed 17/35 (49%) MIRU-VNTR typing clusters; the other 18 clusters contained pairs separated by >12 SNPs. Most transmission clusters (3/4) of Swiss-born patients were confirmed by WGS, as opposed to 25% (4/16) of the clusters involving only foreign-born patients. The overall clustering proportion was 17% (90 patients; 95% confidence interval [CI], 14 to 21%) by standard genotyping but only 8% (43 patients; 95% CI, 6 to 11%) by WGS. The clustering proportion was 17% (67/401; 95% CI, 13 to 21%) by standard genotyping and 7% (26/401; 95% CI, 4 to 9%) by WGS among foreign-born patients and 19% (23/119; 95% CI, 13 to 28%) and 14% (17/119; 95% CI, 9 to 22%), respectively, among Swiss-born patients. Using weighted logistic regression, we found weak evidence of an association between birth origin and transmission (adjusted odds ratio of 2.2 and 95% CI of 0.9 to 5.5 comparing Swiss-born patients to others). In conclusion, standard genotyping overestimated recent TB transmission in Switzerland compared to WGS, particularly among immigrants from regions with a high TB incidence, where genetically closely related strains often predominate. We recommend the use of WGS to identify transmission clusters in settings with a low incidence of TB.

Purpose. The paper deals with the galvanic coupling effect (time of treatment and other factors) on the removal of oil, synthetic surface active substances (SSAS), chromium, etc. from waste process solutions used in metalworking. It presents an overview of wastewater treatment technologies from heavy metal ions based on physicochemical processes. Methodology. To restore hexavalent chlorine to trivalent, authors decided to use the existing averager of acid-alkaline effluents as a kind of galvanic coagulant, for which it was filled with iron turnings and crushed coke. Findings. Authors researched wastewater of the electrotechnical industry and highlighted sorption methods for their purification with the use of coagulants, ion exchange. They considered a new galvanocoagulation method for purification from heavy metal (HM) ions (arsenic, chromium, non-ferrous metals), as well as petroleum products and organic substances dissolved in water. Permissible values of the quality indicators of wastewater and water in reservoirs are given. The classification of heavy metal groups according to the degree of toxic effect is presented. Their influence on the human body is considered. The use of the «LOTOS» reactor in the technological process of electroplatings wastes cleaning allows: to process them in a single stream, to abandon special reagents for cleaning effluents from hexavalent chromium and cyanides, to exclude the need for acidification of effluents, to reduce the use of alkaline reagents up to complete rejection of them, to reduce the equipment rate and number of production personnel (their main function is to purify solutions containing up to 1 g/dm3 of hexavalent chromium. В работе необходимо изучить влияние вида гальванопары (времени обработки и других факторов) на удаление масла, синтетических поверхностно-активных веществ (СПАВ), хрома и т. п. из отработанных технологических растворов, применяемых при металлообработке. Представлен обзор технологий очистки сточных вод от ионов тяжелых металлов, основанных на физико-химических процессах. Методика. Для восстановления шестивалентного хлора до трехвалентного было принято решение использовать существующий усреднитель кислотнощелочных стоков в качестве своеобразного гальванокоагулятора, для чего в него засыпались железная стружка и коксовая крошка. Результаты. Исследованы сточные воды электротехнической промышленности и освещены сорбционные методы их очистки с применением коагулянтов, ионного обмена. Рассмотрен новый гальванокоагуляционный способ очистки от ионов ТМ (мышьяка, хрома, цветных металлов), а также нефтепродуктов и растворенных в воде органических веществ. Приведены допустимые величины показателей качества сточных вод и воды водоемов. Дана классификация групп тяжелых металлов по степени токсичного воздействия. This paper is based on the main results of an analysis of spontaneous flora and vegetation on the small islet of Vrnik (0.281 km2), on which there are some abandoned limestone quarries, on the eastern Adriatic coast. The investigations were carried out from 2014 to 2016. Altogether, 251 vascular plant taxa (species and infraspecific units) were recorded on the islet. A total of 11 plant associations, one subassocation and two stands within 10 vegetation classes were identified. Due to high anthropogenic influences during the last centuries, quarrying in particular, the islet investigated showed a relatively low variety of vascular plant taxa. In addition, clear signs of fragmentation of the forest vegetation were observed.

This article refers to four twentieth-century Polish narrative texts that use elements of secrecy in different ways. Mrożek’s Moniza Clavier conceals the confabulatory character of the first-person statement, Miłosz’s Dolina Issy and Gombrowicz’s Kosmos conceal the deeply autobiographical character of the reflection on individual fate, while the narrative in Lem’s Solaris activates the possibility of ‘vertical reading’, referring to the concept of some absolute.
